Oxidation is a leading cause for quality deterioration during processing and storage of food. The objective of the present study was to examine the sensitivity of common carp (Cyprinus carpio) myofibrillar protein (MP) to oxidising radicals produced by a hydroxyl radical-generating system. Both structural and functional changes of common carp MP were evaluated. With increasing H2O2 concentrations and oxidation time, the protein carbonyl content, surface hydrophobicity and turbidity of MP increased (P < 0.05), while total sulfhydryl groups decreased (P < 0.05). Sodium dodecyl sulphate-polyacrylamide gel electrophoresis revealed protein polymerisation in oxidised MP. The oxidative process destroyed (P < 0.05) the texture (springiness and hardness) of MP gels and decreased their water-binding capacity and whiteness. The thermal gelation profile analysis indicated that oxidation led to a great reduction in the elasticity of samples. Taken together, proteins are susceptible to free radical attack, and oxidative stress had a detrimental effect on protein structure and the general functionality of MP.  相似文献

The effects of hydroxyl radicals induced oxidation on water distribution of porcine myofibrillar protein (MP) gels were investigated using nuclear magnetic resonance (NMR) and magnetic resonance imaging (MRI), and relationship between MP gel properties and water distribution was analyzed with Pearson’s correlation. Results of NMR spin-spin relaxation time (T2) and MRI suggested that, comparing with unoxidized MP gel, the population of immobile water of gel (P22) decreased by 18.81 %, while that of free water (P23) increased by 85.56 %, and grey scale value decreased by 18.52 %, after being oxidized with 20 mM H2O2, respectively. Images of scanning electron microscope (SEM) showed the highest degree of oxidation led to the most coarse and porous gel-network. Gel properties including WHC, whiteness and strength of MP gels decreased by 14.65 %, 2.83 % and 52.74 % after being oxidized with 20 mM H2O2 (p?<?0.05), respectively. Pearson’s correlation analysis showed oxidation, gel-properties and NMR results were highly correlated (p?<?0.05). Therefore, we hypothesized that hydroxyl radicals induced oxidation resulted in immobile water shifting to free water partly, thus reduced the abundance of water and gel properties of gels.  相似文献

The objective of the present study was to investigate the effects of a cryoprotectant (a mixture of sucrose and sorbitol) on protein and lipid oxidation and structural changes in common carp (Cyprinus carpio) surimi subjected to different numbers of freeze-thaw (FT) cycles. Increased numbers of FT cycles were accompanied by increases in carbonyl content and thiobarbituric acid-reactive substances values and concomitant decreases in sulphydryl group (P<0.05) content, revealing that multiple FT cycles could cause protein and lipid oxidation in the carp tissue. Myofibrillar protein isolated from FT surimi also showed a reduced ATPase activity (P<0.05), increased hydrophobicity (P<0.05), and enhanced susceptibility to thermal aggregation. The addition of cryoprotectants significantly inhibited carbonyl formation and lipid oxidation (P<0.05), and its good protective effect on protein denaturation was demonstrated by its efficacy in maintaining Ca-ATPase activity, reducing the rapid exposure of hydrophobic and sulphydryl groups on the protein surface, and decreasing the degree of protein aggregation. Overall, the use of cryoprotectants reduced protein oxidation and structural deterioration induced by freeze-thaw in common carp surimi.  相似文献

Effects of protein isolate from bambara groundnut (BGPI) at different levels (0–6 %, w/w) in combination with microbial transglutaminase (MTGase) at different concentrations (0, 0.3 and 0.6 U g?1surimi) on gels properties of sardine (Sardinella albella) surimi were investigated. In the absence of MTGase, the increases in breaking force and deformation of gels were obtained when BGPI at levels of 1.5–3 % was incorporated (P?<?0.05). The further increases in BGPI levels (4.5–6 %) resulted in the decrease in breaking force and deformation (P?<?0.05). When MTGase (0.3 and 0.6 U g?1surimi) was added, the increase in breaking force and deformation were noticed, regardless of BGPI levels, and the strengthening effect was in dose-dependent manner. The increases in hardness, gumminess and chewiness were also observed when surimi gel was added with BGPI and MTGase (P?<?0.05). Water-holding capacity of gels was improved with increasing level of BGPI, and MTGase incorporated (P?<?0.05). Whiteness of gels slightly decreased with increasing BGPI levels, however the addition of MTGase had no impact on whiteness (P?>?0.05). Based on electrophoretic study, myosin heavy chain decreased with addition of MTGase, indicating the formation of cross-links. This study was conducted to investigate the toxicity of cadmium and to evaluate the effectiveness of maifanite in preventing cadmium-induced adverse effects. Thirty-two crossbred pigs (Duroc × Landrace × Large white, sex balanced, 17.25?±?0.07 kg average body weight) were randomly allotted to one of four dietary treatments in a 2?×?2 factorial arrangement, with eight replicates per treatment and one pig per replicate. The dietary treatments included two cadmium (as CdCl2) doses (0.32 and 30.49 mg/kg) and two maifanite doses (0 and 1 %). The results showed that pigs treated with cadmium decreased their average daily feed intake (P?<?0.05) and increased (P?<?0.05) the feed/gain ratio. Cadmium was found in the tissues of pigs that were fed with cadmium-contaminated diets, but the level of cadmium was much lower when maifanite was added to the cadmium-contaminated diets. Ingestion of diets that were artificially contaminated with cadmium (30.49 mg/kg of cadmium) reduced (P?<?0.05) the number of lymphocytes, the total erythrocyte count, the hemoglobin level, and the hematocrit. However, the activities of serum aspartate aminotransferase and gamma glutamyltransferase were increased (P?<?0.05). The total protein level was lower (P?<?0.05) in pigs fed with cadmium-contaminated diets. The contents of malondialdehyde increased (P?<?0.05), while the total antioxidant capacity and the activities of total superoxide dismutase, glutathione peroxidase, glutathione S-transferase, and catalase decreased (P?<?0.05) in pigs fed with cadmium-contaminated diets. The aim of this study was to evaluate the in vitro cytotoxicity and the in vivo analgesic effect and local toxicity of the local anesthetic butamben (BTB) encapsulated in conventional or elastic liposomes incorporated in gel formulations. The results showed that both gel formulations of liposomal BTB reduced the cytotoxicity (p?<?0.001; one-way ANOVA/Tukey’s test) and increased the topical analgesic effect (p?<?0.05; one-way ANOVA/Tukey’s test) of butamben, compared to plain BTB gel. The gel formulations presented good rheological properties, and stability assays detected no differences in physicochemical stability up to 30 d after preparation. Nutrient nitrogen and water availability are co-limiting factors for grassland productivity in the Inner Mongolian steppe. The objective of this study was to evaluate the interactive effects of water and nitrogen (N) addition on soil abiotic factors and soil nematode community composition. A 3-year experiment with addition of water (with and without irrigation simulating wet year precipitation) and nitrogen (0, 25, and 50?kg?N?ha?1) was conducted at two sites in Inner Mongolia with histories of heavy grazing (HG) and moderate grazing (MG). The results showed that HG had several lower nematode ecological indicators; namely, the Simpson index (P?<?0.01), maturity index (MI, including free-living nematodes), and plant parasite index (PPI, including plant-parasitic nematodes). In addition, HG had a lower proportion of omnivores–predators and higher proportion of bacterivores (P?<?0.01), with reduced soil moisture, total nitrogen and total carbon (C), and increased C/N ratio. A relatively low addition of N did not influence soil total carbon and total nitrogen and had only a slight effect on the composition and structure of the soil nematode community. Water addition increased the proportion of plant parasites, reduced the proportion of bacterivores and omnivores–predators and increased the soil total carbon and total nitrogen. Redundancy analysis revealed that grazing history alone explained more variation in nematode taxon composition (17.4%, P?<?0.01) and PPI (35.6%, P?<?0.01) and MI (17.5%, P?<?0.01) and Simpson index (10.5%, P?<?0.01). Water addition explained more variation in the nematode feeding group (24.4%, P?<?0.01), total nematodes (7.9%, P?<?0.01) and PPI (78.7%, P?<?0.01). Nitrogen addition explained variation in the nematode taxon composition (6.2%, P?<?0.01). These results suggested that HG caused soil degradation, and water addition facilitated plant parasites and enhanced soil properties, whereas application of small amounts of nitrogen to N-limited semiarid grasslands did not induce clear changes in this system in the short term.  相似文献

Polysaccharide-protein gums are complex polymers with many applications in emulsion and emulsion-based products. It is crucial to enclose a concept on understanding of functional properties of polysaccharide-protein biopolymer in order to select the appropriate one based on the application scope. The main objective of the current study was to investigate the effect of three extraction variables on the surface activity, particle uniformity, apparent viscosity, and protein content of a natural polysaccharide-protein biopolymer from durian fruit seed. Three extraction variables namely water: seed (W/S) ratio (20:1?C60:1), temperature (25.0?C85.0?°C) and pH (4.0?C10.0) were considered as independent variables. The results indicated that durian seed gum induced very low viscosity (0.93?C4.98?mPa.s) with pseudoplastic rheological behavior in the aqueous system. The current study revealed that the extraction variables had the most significant (p?<?0.05) effect on the emulsifying surface activity of the natural biopolymer from durian fruit seed. This might be due to the significant (p?<?0.05) effect of the proteineous constituent present in the chemical structure of the biopolymer from durian fruit seed. The aqueous extraction variables showed the most and least significant (p?<?0.05) effect on the specific surface area and protein content, respectively. The gel properties and molecular forces of the Aohan Merino lamb myofibrillar protein (MP) were investigated during heat induction at different pH levels. The effects of pH values (from 5.0 to 8.5) on the lamb MP gel textures and water-holding capacities were analyzed. The protein gels had the lowest water-holding capacity and a disordered microstructure at pH 5.5 (P < 0.05) and had the lowest level of hardness at pH 6.5 (P < 0.05). The hardness and water-holding capacity were found to be the highest at pH 8.0 (P < 0.05), which coincided with a compact and ordered microstructure. At these three representative pH values, hydrophobic interactions were the main forces that were observed during gel formation, but the pH level also influenced the formation of ionic and hydrogen bonds. Different mechanisms of gel formation were therefore observed at different pH values. In conclusion, the formation of heat-induced gels with different properties was a result of the dominant molecular forces that are regulated by pH. These findings provide a theoretical basis for the innovation of heat-induced gel lamb products.  相似文献

In previous studies, we investigated a link between high fluoride exposure and functional IQ deficits in rats. This study is an extension conducted to explore the combined influence of physical exercise and temperature stress on the learning ability and memory in rats and to assess whether any positive modulation could be attenuated due to exercise regimen subjected to F-toxicated animals at different temperatures. Accumulation of ingested fluoride resulted significant inhibition in acetylcholinesterase activity (P?<?0.05), plasma cortisol levels (P?<?0.05), and impaired the acquisition, performance, latency time, and retention in fluoride-exposed animals. Fluoride-toxicated rats took more number of sessions during the learning phase [F 5, 35?=?19.065; P?<?0.05] and post hoc analysis on the number of correct choices revealed that there was a significant effect of treatments [F 5, 30?=?15.763; P?<?0.05]; sessions [F 8, 240?=?58.698; P?<?0.05]; and also significant difference in the interactions [F 40, 240?=?1.583; P?<?0.05]. In order to ascertain if interdependence between fluoride concentrations and the foregoing free radical parameters, respective correlation coefficients were calculated and results clearly emphasize the positive role of exercise in the promotion of cognitive functions by decreasing fluoride levels in rat hippocampus. A significant recovery in cognitive function was noticed in all the exercised animals due to reduced burden of brain oxidative stress. In comparison to exercise regimens performed at different temperatures, high (35?°C) and low temperatures (20?°C) led to a slower acquisition and poor retention of the task when compared to thermo neutral temperatures (25 and 30?°C). The study was conducted to determine the effects of iron glycine chelate (Fe-Gly) on growth, tissue mineral concentrations, fecal mineral excretion, and liver antioxidant enzyme activities in broilers. A total of 360 1-day-old commercial broilers (Ross?×?Ross) were randomly allotted to six dietary treatments with six replications of ten chicks per replicate. Broilers were fed a control diet with no Fe supplementation, while five other treatments consisted of 40, 80, 120, and 160?mg Fe/kg diets from Fe-Gly, and 160?mg Fe/kg from ferrous sulfate, respectively. After a 42-day feeding trial, the results showed that 120 and 160?mg Fe/kg as Fe-Gly improved the average daily gain (P?<?0.05) and average daily feed intake (P?<?0.05) of broilers (4?C6?weeks). Addition with 120 and 160?mg Fe/kg from Fe-Gly and 160?mg Fe/kg from FeSO4 increased Fe concentration in serum (P?<?0.05), liver (P?<?0.05), breast muscle (P?<?0.05), tibia (P?<?0.05), and feces (P?<?0.01) at 21 and 42?days. The contribution of soybean variety and coagulant type to the textural and rheological properties of soy protein isolate (SPI) tofu-type emulsion gels was studied. SPIs from eight soybean varieties were subjected to amino acid and sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE) analysis, and results showed that the 11S fraction proteins (r?=?0.833, p?<?0.05) and the ratio of 11S to 7S (r =?0.920, p <?0.01) were positively correlated with the hardness of CaSO4-induced emulsion gels and glucono-δ-lactone (GDL)-induced gels, with the correlation coefficients of 0.827 (p <?0.05) and 0.893 (p <?0.01), respectively. In the case of microbial transglutaminase (MTGase), strong relations between the content of glutamate (r =?0.886, p?<?0.01) and lysine (r =?0.810, p <?0.05) and gel hardness were found. Rheological data demonstrated that CaSO4-induced emulsion gel was stiffer with high rigidity but gel induced by MTGase performed better elasticity. Reperfusion injury is one of the main reasons of cardiac disease morbidity. Phytopharmaceuticals are gaining importance in modern medicine of cardioprotection because of their multiplex capacity. The aim of this study was to investigate the effect of diosgenin on the inflammatory response induced by myocardial ischemia and reperfusion injury and the role of mitochondrial ATP-sensitive potassium (mitoKATP) channels in this regard. Wistar rats (250–300 g) were used in this study. The Langendorff-perfused hearts of animals were subjected to a 30-min global ischemia followed by a 90-min reperfusion. The lactate dehydrogenase (LDH) release was measured by spectrophotometry. The levels of inflammatory mediators tumor necrosis factor-alpha (TNF-α), interleukin-1beta (IL-1β), and IL-6 in the supernatant of heart’s left ventricle were measured using an enzyme-linked immunosorbent assay rat specific ELISA kit. The LDH release into the coronary effluent during reperfusion was significantly decreased, and cardiac contractility significantly improved by diosgenin preadministration as compared with those of control or Cremophor-EL (solvent of diosgenin) groups (398?±?48 vs. 665?±?65 or 650?±?73 ml/min) (P?<?0.01). Administration of diosgenin before the main ischemia significantly reduced the levels of IL-6 (P?<?0.05), IL-1β, and TNF-α (P?<?0.01) in the reperfusion phase of diosgenin-treated hearts as compared with untreated control hearts. Inhibition of mitoKATP channels by 5-hydroxydecanoate significantly reverses the cardioprotective effects of diosgenin (P?<?0.05). The study examines plasma metabolic profiles of patients with chronic obstructive pulmonary disease (COPD) to prove whether the disease influences metabolism at rest and after endurance training. This is based on the hypothesis that metabolome levels should reflect impaired skeletal muscle bioenergetics in COPD. The study aims to test this hypothesis by evaluating plasma metabolic profiles in COPD patients before and after 8?weeks of endurance exercise training. We studied blood samples from 18 COPD patients and 12 healthy subjects. Pre- and post-training blood plasma samples at rest and after constant-work rate exercise (CWRE) at 70% of pre-training Watts peak were analyzed by 1H-nuclear magnetic resonance spectroscopy to assess metabolite profiles. The two groups presented training-induced physiological changes in the VO2 peak and in blood lactate levels (P?<?0.01 each). Before training, the two groups also showed differences in metabolic profiles at rest (P?<?0.05). Levels of valine (r?=?0.51, P?<?0.01), alanine (r?=?0.45, P?<?0.05) and isoleucine (r?=?0.51, P?<?0.01) were positively associated with body composition (Fat Free Mass Index). While training showed a significant impact on the metabolic profile in healthy subjects (P?<?0.001), with changes in levels of amino acids, creatine, succinate, pyruvate, glucose and lactate (P?<?0.05 each), no equivalent training-induced effects were seen in COPD patients in whom only lactate decreased (P?<?0.05). During biotic and abiotic stress in plants, reactive oxygen species (ROS) may play two very different roles: high ROS concentrations can exacerbate damage, whereas low concentrations can activate defense responses. The aim of this study was to investigate the relationship between ROS generation and pollen viability after cryopreservation. ROS generation was detected from ‘Siberia’ (Lilium?×?siberia) pollen using flow cytometry with 2′,7′-dichlorodihydrofluorescein diacetate as a fluorescent probe. Pollen viability was determined by 2,3,5-triphenyltetrazolium chloride staining. ROS generation was slightly increased by rapid cooling (26.13?±?4.74 vs. 15.80?±?2.30 for fresh pollen) and significantly increased by vitrification (49.74?±?1.43; P?<?0.01). Pollen viabilities after rapid cooling and vitrification were significantly increased (58.88?±?3.76% and 70.35?±?2.90%, respectively) over that of fresh pollen (46.65?±?1.61%; P?<?0.01). No significant differences in ROS generation were associated with cold acclimation at different temperatures before rapid cooling. However, sharp decreases in viability were observed with cold acclimation at 4°C and ?20°C relative to rapid cooling without acclimation (P?<?0.01). We observed nonsignificant decreases in ROS generation among vitrification treatments that omitted different steps and a significant decrease when the unloading step was omitted (P?<?0.05). Pollen viabilities were significantly reduced when the loading or dehydration steps were omitted (P?<?0.01). No significant differences were observed in ROS generation or pollen viability among the treatments when 200 U/ml catalase was added to different solutions used in the vitrification process. This study investigated the potential effect of porcine plasma protein hydrolysates (PPPH) coupled with Tween 20 on the physical and oxidative stability of canola oil-in-water emulsions (10% wt. lipid, pH 7.0). The PPPH obtained via limited alcalase-hydrolysis (60 min) possessed the highest emulsifying activity index and emulsion stability index (P?<?0.05). Emulsions stabilized with PPPH alone were less stable than those prepared with PPPH and Tween 20. However, a lower concentration of PPPH (2.5 mg/mL) combined with Tween 20 formed the most stable emulsions, which is mainly due to competitive adsorption present at the interface. Additionally, compared with PPPH-free emulsions, the addition of PPPH was able to retard lipid oxidation, showing up to an 8.51% decrease in the formation of conjugated dienes and a 22.08% decrease in thiobarbituric acid-reactive substances after 10 days of storage (P?<?0.05). This is mainly attributed to distinct antioxidant amino acid profiles and the distribution of peptides at the interface. Therefore, our results indicate that PPPH derived from limited hydrolysis could be used as both co-emulsifiers and antioxidative compounds in food emulsions.  相似文献

In the present study we have evaluated the effect of a single hemodialysis session on the brain-derived neurotrophic factor levels in plasma [BDNF]pl and in serum [BDNF]s as well as on the plasma isoprostanes concentration [F2 isoprostanes]pl, plasma total antioxidant capacity (TAC) and plasma cortisol levels in chronic kidney disease patients. Access to an evaporative cooling system can increase production in dairy cows because of improved thermal comfort. This study aimed to evaluate the impact of ambient temperature on thermoregulation, plasma cortisol, insulin-like growth factor 1 (IGF-I), and productive status, and to determine the efficiency of an evaporative cooling system on physiological responses under different weather patterns. A total of 28 Holstein cows were divided into two groups, one with and the other without access to a cooling system with fans and mist in the free stall. The parameters were analyzed during morning (0700 hours) and afternoon milking (1430 hours) under five different weather patterns throughout the year (fall, winter, spring, dry summer, and rainy summer). Rectal temperature (RT), body surface temperature (BS), base of tail temperature (TT), and respiratory frequency (RF) were lower in the morning (P?<?0.01). The cooling system did not affect RT, and both the groups had values below 38.56 over the year (P?=?0.11). Cortisol and IGF-I may have been influenced by the seasons, in opposite ways. Cortisol concentrations were higher in winter (P?<?0.05) and IGF-I was higher during spring-summer (P?<?0.05). The air temperature and the temperature humidity index showed positive moderate correlations to RT, BS, TT, and RF (P?<?0.001). The ambient temperature was found to have a positive correlation with the physiological variables, independent of the cooling system, but cooled animals exhibited higher milk production during spring and summer (P?<?0.01).  相似文献
