With repetitive stimulation of two sensory pathways which are intact within the isolated nervous system of Hermissenda, features of a cellular conditioning paradigm were identified. Type A photoreceptors, unlike type B photoreceptors, produce fewer impulses in response to light following temporally specific pairing of light stimuli with rotation stimuli. Type A photoreceptor impulse wave-forms are also specifically changed by such stimulus regimens. These findings can be explained, at least in part, by increased inhibition of type A cells by type B cells after stimulus pairing.  相似文献

Type B photoreceptors of the nudibranch mollusc Hermissenda crassicornis receive excitatory synaptic potentials (EPSPs) whose frequency is controlled by potential changes of a neighboring cell known as the S optic ganglion cell which is thought to be electrically coupled to the presynaptic source of these EPSPs, the E optic ganglion cell. The frequency of the EPSPs increases when a conditioned stimulus (light) is paired with an unconditioned stimulus (rotation) during acquisition of a Pavlovian conditioned response. The results of the present study are consistent with an adrenergic origin for these EPSPs. Noradrenergic agonists (greater than 100 microM), norepinephrine and clonidine, only slightly depolarize the type B cell but clearly prolong its depolarizing response to light. Serotonin, by contrast, causes hyperpolarization of the type B cell's resting potential as well as after a light step. Clonidine reduces voltage-dependent outward K+ currents (IA, an early current, ICa2+-K+, a late Ca2+-dependent current) that control the type B cell's excitability (and thus its light response and membrane potential). These effects of clonidine are reduced or blocked by the alpha 2-receptor antagonist, yohimbine (0.5 microM), but not the alpha 1-blocker, prazosin. The same yohimbine concentration also blocked depolarizing synaptic excitation of the type B cell in response to depolarization of a simultaneously impaled S optic ganglion cell. Histochemical techniques (both the glyoxylic acid method of de la Torre and Surgeon and the formaldehyde-induced fluorescence or Falck-Hillarp method) demonstrated the presence of a biogenic amine(s) within a single neuron in each optic ganglion as well as three or four cells within the vicinity of previously identified visual interneurons. No serotonergic neurons were found within the optic ganglion or in proximity to visual interneurons. A clonidine-like synaptic effect on type B cells, therefore, could amplify conditioning-specific changes of membrane currents by increasing type B depolarization and possibly, as well, by elevating intracellular second messengers.  相似文献

The number of postsynaptic gamma-aminobutyric acid type A (GABAA) receptors is a fundamental determinant of the variability of inhibitory synaptic responses in the central nervous system. In rat visual cortex, [3H]SR-95531 binding assays revealed that brain-derived neurotrophic factor (BDNF), one of the neurotrophins, induced a rapid increase in the total number of cell surface GABAA receptors, through the activation of Trk B receptor tyrosine kinases. We also demonstrated that BDNF rapidly induced a sustained potentiation of GABAA receptor-mediated currents, using nystatin-perforated patch clamp recordings, in visual cortical layer 5 pyramidal neurons freshly isolated from P14 rats. The potentiation was caused by the activation of Trk B receptor tyrosine kinase and phospholipase C-gamma. In addition, intracellular Ca2+ was important for the potentiation of GABAA responses induced by BDNF. The selective increase in mean miniature inhibitory postsynaptic (mIPSC) current amplitude without effects on mIPSC time courses supports the idea that BDNF rapidly induces an increase in the total number of cell surface functional GABAA receptors in visual cortical pyramidal neurons. These results suggest that BDNF could alter the number of cell surface GABAA receptors in a region-specific manner.  相似文献

Focal and impulse activity of neurones, recorded in pairs with one microelectrode, was studied in the field 18 of cat visual cortex, during learning of visual differentiation of shaped stimuli--light strips with different orientation one towards the other. Differences were found between functional properties of neighbouring visual neurones before learning and in learning dynamics. During learning, independently of visual signal stimuli orientation, reaction patterns of the same type were formed in neighbouring visual neurones.  相似文献

Responses of caudate neurons to two kinds of visual stimuli, namely diffuse light and a more local stimulus (a slit of light), oriented in different directions on a screen, were studied in alert cats during natural fixation of the gaze. The number of neurons which responded to local stimulation was several times greater than the number responding to diffuse light. Besides on-responses to local stimulation, a more distinct phase of inhibition of activity during presentation of the stimulus and off-responses also appeared. The latent periods of responses to both kinds of stimulation were commensurate at 40–90 msec for most neurons. Differences in neuronal responses also were found on a change in orientation of the slit. The results are discussed from the standpoint of participation of the caudate nucleus in visual information analysis.  相似文献

The formation of synaptic connections requires the coordination of specific guidance molecules and spontaneous neuronal activity. The visual system has provided a useful model for understanding the role of these cues in shaping the precise connections from the neural retina to the brain. Here, we demonstrate that two essential genes in the Reelin signaling pathway function during the patterning of synaptic connectivity in the retina. Physiological studies of mice deficient in either reelin or disabled-1 reveal an attenuation of rod-driven retinal responses. This defect is associated with a decrease in rod bipolar cell density and an abnormal distribution of processes in the inner plexiform layer. These results imply that, in addition to its essential role during neuronal migration, the Reelin pathway contributes to the formation of neuronal circuits in the central nervous system.  相似文献

Summary Descending deviation detector neurons (DDNs) of Locusta migratoria are characterized physiologically by their responses to light on/off stimuli, simulated course deviation (rotation of an artificial horizon), passive rotation of the head, frontal wind, and flight activity. The investigation emphasises on the co-processing of exteroceptive input signalling course deviation (mainly movement of the retinal image, but also wind), and proprioceptive input signalling head movement and position. Stimuli were presented in combinations as expected during natural behavior. Eight DDNs are described for the first time, and 3 previously described DDNs are characterized further. Responses to horizon rotation and imposed head movements are assigned to one of 4 response types: (1) the horizon-only type codes retinal slip and/or the position of the horizon in the visual field but ignores cervical proprioception; (2) the head-only type ignores visually simulated course deviation but codes for movement or position of the head; (3) in the compensating type, head rolling causes visual input and cervical proprioceptive input of opposite signs, so that head movements themselves are ignored, whereas course deviations are recognized; (4) in the amplifying type, head rolling causes visual input and cervical proprioceptive input of the same sign, i.e. one input amplifies the other. This classification does not take the various responses to wind into account. In several DDNs, responses to phasic and tonic stimuli of the same modality, and/or responses to deviations about different axes could be assigned to different response types. Activity in DDNs has been shown previously to result in steering responses of wings, legs, abdomen and/or the head. Several aspects of behavior and physiology, such as sleep and wakefulness, blood pressure, body temperature, and hormone secretion exhibit daily oscillations known as circadian rhythms. These circadian rhythms are orchestrated by an intrinsic biological clock in the suprachiasmatic nuclei (SCN) of the hypothalamus which is adjusted to the daily environmental cycles of day and night by the process of photoentrainment. In mammals, the neuronal signal for photoentrainment arises from a small subset of intrinsically photosensitive retinal ganglion cells (ipRGCs) that send a direct projection to the SCN. ipRGCs also mediate other non-image-forming (NIF) visual responses such as negative masking of locomotor activity by light, and the pupillary light reflex (PLR) via co-release of neurotransmitters glutamate and pituitary adenylate cyclase-activating peptide (PACAP) from their synaptic terminals. The relative contribution of each neurotransmitter system for the circadian photoentrainment and other NIF visual responses is still unresolved. We investigated the role of glutamatergic neurotransmission for circadian photoentrainment and NIF behaviors by selective ablation of ipRGC glutamatergic synaptic transmission in mice. Mutant mice displayed delayed re-entrainment to a 6 h phase shift (advance or delay) in the light cycle and incomplete photoentrainment in a symmetrical skeleton photoperiod regimen (1 h light pulses between 11 h dark periods). Circadian rhythmicity in constant darkness also was reduced in some mutant mice. Other NIF responses such as the PLR and negative masking responses to light were also partially attenuated. Overall, these results suggest that glutamate from ipRGCs drives circadian photoentrainment and negative masking responses to light.  相似文献

The receptive field of a visual neurone is classically defined as the region of space (or retina) where a visual stimulus evokes a change in its firing activity. Intracellular recordings in cat area 17 show that the visually evoked synaptic integration field extends over a much larger area than that established on the basis of spike activity. Synaptic depolarizing (dominant excitation) responses decrease in strength for stimuli that are flashed at increasing distances away from the centre of the discharge field, while their onset latency increases. A detailed spatio-temporal analysis of these electrophysiological data shows that subthreshold synaptic responses observed in the 'silent' surround of cortical receptive fields result from the intracortical spread of activation waves carried by slowly conducting horizontal axons within primary visual cortex. They also predict that a perceptual facilitation may occur when feedforward activation produced by the motion signal in the retina travels in phase in the primary visual cortex with the visually induced spread of horizontal activation. A psychophysical correlate has been obtained in humans, showing that apparent motion produced by a sequence of co-linear Gabor patches, known to preferentially activate V1 orientation selective cells, are perceived by human observers as much faster than non co-linear sequences of the same physical speed.  相似文献

Glial cells isolated from the nervous system are sensitive to neurotransmitters and may therefore be involved in synaptic transmission. The sensitivity of individual perisynaptic Schwann cells to activity of a single synapse was investigated, in situ, at the frog neuromuscular junction by monitoring changes in intracellular Ca2+ in the Schwann cells. Motor nerve stimulation induced an increase in intracellular Ca2+ in these Schwann cells; this increase was greatly reduced when transmitter release was blocked. Furthermore, local application of the cotransmitters acetylcholine and ATP evoked Ca2+ responses even in the absence of extracellular Ca2+. Successive trains of nerve stimuli or applications of transmitters resulted in progressively smaller Ca2+ responses. We conclude that transmitter released during synaptic activity can evoke release of intracellular Ca2+ in perisynaptic Schwann cells. This Ca2+ signal may play a role in the maintenance or modulation of a synapse. These data show that synaptic transmission involves three cellular components with both postsynaptic and glial components responding to transmitter secretion.  相似文献

The individual life history of the desert ant Cataglyphis fortis is characterized by a fast transition from interior tasks to mainly visually guided foraging. Previous studies revealed a remarkable structural synaptic plasticity in visual and olfactory input regions within the mushroom bodies of the ants' brain centers involved in learning and memory. Reorganization of synaptic complexes (microglomeruli) was shown to be triggered by sensory exposure rather than an internal program. Using video analyses at the natural nest site and activity recordings after artificial light treatments we investigated whether the ants get exposed to light before onset of foraging and whether this changes the ants' activity levels. We asked whether synaptic reorganization occurs in a similar time window by immunolabeling and quantification of pre- and postsynaptic compartments of visual and olfactory microglomeruli after periods of light-exposure. Ants reverted back to dark nest conditions were used to investigate whether synaptic reorganization is reversible. The behavior analyses revealed that late-interior ants (diggers) are exposed to light and perform exploration runs up to 2 days before they start foraging. This corresponds well with the result that artificial light treatment over more than 2-3 days significantly increased the ants' locomotor activities. At the neuronal level, visual exposure of more than 1 day was necessary to trigger reorganization of microglomeruli, and light-induced changes were only partly reversible in the dark. We conclude that visual preexposure is an important and flexible means to prepare the ants' visual pathway for orientation capabilities essential during foraging.  相似文献

Extraocular photosensitivity in Aplysia fasciata was studied in the skin and in the central nervous system (CNS). Local illumination causes contractions of the muscles of the body wall, which are obviously mediated by the peripheral nervous system (PNS). Afferent sensory activity is supposedly mainly dependent on stretch reception. Light-induced peripheral reflexes habituate after repetitive stimulation in preparations in which the CNS is present. In preparations without CNS light-induced contractions are remarkably stronger and do not habituate after repititive stimulation. Central responses to peripheral stimulation could be evoked by both "light on" and "light off" stimulation, indicating that 2 types of photosensitive elements are present in the periphery. Observations on isolated CNS-preparations revealed that in the central ganglia photoreceptive elements are also present. Here, too, elements responding to the onset as well as elements responding to the offset of light have been detected.  相似文献

The magnetic compass of migratory birds has been suggested to be light-dependent. Retinal cryptochrome-expressing neurons and a forebrain region, "Cluster N", show high neuronal activity when night-migratory songbirds perform magnetic compass orientation. By combining neuronal tracing with behavioral experiments leading to sensory-driven gene expression of the neuronal activity marker ZENK during magnetic compass orientation, we demonstrate a functional neuronal connection between the retinal neurons and Cluster N via the visual thalamus. Thus, the two areas of the central nervous system being most active during magnetic compass orientation are part of an ascending visual processing stream, the thalamofugal pathway. Furthermore, Cluster N seems to be a specialized part of the visual wulst. These findings strongly support the hypothesis that migratory birds use their visual system to perceive the reference compass direction of the geomagnetic field and that migratory birds "see" the reference compass direction provided by the geomagnetic field.  相似文献

Changes in synaptic responses of identified command neurones of avoidance behaviour to the electric nerve stimulation were investigated in the isolated nervous system of the snail during bath application of serotonin or noradrenaline. Serotonin (10(-5) M) elicited an increase of summary EPSP amplitude in the cells without changes of input resistance and resting potential. Noradrenaline (10(-5) M) application evoked an increase of EPSP amplitude, accompanied by an increase of the input resistance. Mechanisms of serotonin and noradrenaline influence on synaptic responses are discussed.  相似文献

Optomotor flight control in houseflies shows bandwidth fractionation such that steering responses to an oscillating large-field rotating panorama peak at low frequency, whereas responses to small-field objects peak at high frequency. In fruit flies, steady-state large-field translation generates steering responses that are three times larger than large-field rotation. Here, we examine the optomotor steering reactions to dynamically oscillating visual stimuli consisting of large-field rotation, large-field expansion, and small-field motion. The results show that, like in larger flies, large-field optomotor steering responses peak at low frequency, whereas small-field responses persist under high frequency conditions. However, in fruit flies large-field expansion elicits higher magnitude and tighter phase-locked optomotor responses than rotation throughout the frequency spectrum, which may suggest a further segregation within the large-field pathway. An analysis of wing beat frequency and amplitude reveals that mechanical power output during flight varies according to the spatial organization and motion dynamics of the visual scene. These results suggest that, like in larger flies, the optomotor control system is organized into parallel large-field and small-field pathways, and extends previous analyses to quantify expansion-sensitivity for steering reflexes and flight power output across the frequency spectrum.  相似文献

Most algorithms currently used to model synaptic plasticity in self-organizing cortical networks suppose that the change in synaptic efficacy is governed by the same structuring factor, i.e., the temporal correlation of activity between pre- and postsynaptic neurons. Functional predictions generated by such algorithms have been tested electrophysiologically in the visual cortex of anesthetized and paralyzed cats. Supervised learning procedures were applied at the cellular level to change receptive field (RF) properties during the time of recording of an individual functionally identified cell. The protocols were devised as cellular analogs of the plasticity of RF properties, which is normally expressed during a critical period of postnatal development. We summarize here evidence demonstrating that changes in covariance between afferent input and postsynaptic response imposed during extracellular and intracellular conditioning can acutely induce selective long-lasting up- and down-regulations of visual responses. The functional properties that could be modified in 40% of cells submitted to differential pairing protocols include ocular dominance, orientation selectivity and orientation preference, interocular orientation disparity, and the relative dominance of ON and OFF responses. Since changes in RF properties can be induced in the adult as well, our findings also suggest that similar activity-dependent processes may occur during development and during active phases of learning under the supervision of behavioral attention or contextual signals. Such potential for plasticity in primary visual cortical neurons suggests the existence of a hidden connectivity expressing a wider functional competence than the one revealed at the spiking level. In particular, in the spatial domain the sensory synaptic integration field is larger than the classical discharge field. It can be shaped by supervised learning and its subthreshold extent can be unmasked by the pharmacological blockade of intracortical inhibition.  相似文献

The action of disulfiram on interconnected activity of neurones in the visual and motor cortical areas was studied in cats with food-procuring conditioned responses to light. Multiunit activity was recorded from the areas and, by means of amplitude discrimination, separated into impulse flows. Crosscorrelation analysis of the impulse series was used to reveal the character and temporal parameters of interconnected activities of neurones firing in correlation within the limits both of the same cortical area and of the two different ones. A depressing action was shown of the disulfiram on the food-procuring reaction, accompanied by a decrease of the number of pairs of neurones from the visual and motor cortical areas mostly acting in interconnection, interactions with long time delays being mostly affected. The character of action of neighbouring neurones in the visual and motor cortical areas changed in the same direction, expressed in their firing by a "common source" type. The question is discussed of disulfiram influence on interneuronal connections of both types suggesting a decrease of alimentary motivation as well as disturbance of food-procuring conditioned motor coordination.  相似文献

Application of desglycine-argininvasopressin (DG-AVP) differently influenced different types of cells of snail isolated central nervous system. In neurosecretory cells an increase of spontaneous impulse activity took place and, as a rule, bursts of impulses appeared, most often of synaptic origin, excluding PPa1 neurones and one of the neurosecretory cells of the left parietal ganglion. The increase of the bursts activity in these cells was based on the increase of the amplitude of membrane potential waves. Under the influence of neurosecretory cells system activation, EPSPs frequency and amplitude in secondary-sensory neurones increased, which led to a greater probability of the action potentials appearance. At prolonged action the spontaneous EPSPs in these cells began to group in bursts. Excitability and membrane resistance of these cells remained unchanged. DG-AVP had no influence on primary-sensory neurones and motoneurones.  相似文献

A single light episode during the first larval stage can set the phase of adult Drosophila activity rhythms, showing that a light-sensitive circadian clock is functional in larvae and is capable of keeping time throughout development. These behavioral data are supported by the finding that neurons expressing clock proteins already exist in the larval brain and appear to be connected to the larval visual system. To define the photoreceptive pathways of the larval clock, the authors investigated circadian synchronization during larval stages in various visual systems and/or cryptochrome-defective strains. They show that adult activity rhythms cannot be entrained by light applied to larvae lacking both cryptochrome and the visual system, although such rhythms were entrained by larval stage-restricted temperature cycles. Larvae lacking either pathway alone were light entrainable, but the phase of the resulting adult rhythm was advanced relative to wild-type flies. Unexpectedly, adult behavioral rhythms of the glass60j and norpAP24 visual system mutants that were entrained in the same conditions were found to be severely impaired, in contrast to those of the wild type. Extension of the entrainment until the adult stage restored close to wild-type behavioral rhythms in the mutants. The results show that both cryptochrome and the larval visual system participate to circadian photoreception in larvae and that mutations affecting the visual system can impair behavioral rhythmicity.  相似文献
