AIM: To determine how eugenol affects fermentation parameters and faecal coliforms in cattle and swine waste slurries stored anaerobically. METHODS AND RESULTS: Waste slurries (faeces:urine:water, 50:35:15) were blended with and without additives and aliquoted to triplicate 1-l flasks. Faecal coliforms were eliminated in cattle and swine waste slurries within 1 or 2 days with additions of eugenol at 10.05 mm (0.15%) and 16.75 mm (0.25%). At these concentrations volatile fatty acids (VFA) were reduced ca 70 and 50% in cattle and swine waste, respectively, over 6-8 weeks. Additionally, in cattle waste, eugenol stimulated the accumulation of lactate (>180 mm) when compared with thymol treatment (20 mm lactate). In swine waste, lactate accumulation did not occur without additives; eugenol and thymol stimulated lactate accumulation to concentrations of 22 and 32 mm, respectively. CONCLUSIONS: Eugenol added to cattle waste may be more beneficial than thymol because not only does it effectively control faecal coliforms and odour (VFA production), it also stimulates lactate accumulation. This in turn, causes the pH to drop more rapidly, further inhibiting microbial activity and nutrient emissions. SIGNIFICANCE AND IMPACT OF THE STUDY: Plant essential oils have the potential to solve some of the environmental problems associated with consolidated animal feeding operations. Thymol and eugenol reduce fermentative activity, thus, have the potential to reduce emissions of greenhouse gases and odour, and curtail transmission of pathogens in cattle and swine wastes.  相似文献

Aims: To determine the effects of urine, a urease inhibitor and/or an odour‐reducing antimicrobial compound, on zoonotic pathogens in swine manure slurries. Methods and Results: Swine faeces were collected and blended with different amounts of urine. Marker strains of Salmonella enterica subsp. enterica serovar Typhimurium and Escherichia coli O157:H7 were inoculated into the manure slurries with or without the urease inhibitor, N‐(n‐butyl) thiophosphoric triamide, or the antimicrobial compound thymol. In slurries containing approximately 1 : 1 or 2 : 1 of urine and faeces, the decreases in colony forming units (CFU) of Salm. Typhimurium and E. coli were similar and few counts were observed after 14 days. When the urine content of the slurry was increased to 5 : 1, both strains died off rapidly. When NBPT was added to the slurries, pathogen CFU was not affected in the 1 : 1 slurry. The 2 : 1 and 5 : 1 slurries had higher urea content and NBPT increased CFU of both pathogens. Thymol addition rapidly killed the pathogens and few CFU of Salmonella or E. coli were observed after day 1. Conclusions: These experiments demonstrate that urea hydrolysis in swine manure affects pathogens. Inhibition of urea hydrolysis may promote pathogen viability. Significance and Impact of the Study: Manure amendments that prevent urea hydrolysis may promote pathogen persistence. Additional treatments with antimicrobials may be required to kill pathogens.  相似文献

Aims: This study evaluated the effect of anaerobic digestion at 22, 38 and 55°C on odour, coliforms and chlortetracycline (CTC) in swine manure or monensin (MON) in cattle manure. Methods and Results: Swine or cattle were fed the respective growth promotant, manure was collected, and 2‐l laboratory methane digesters were established at the various temperatures and sampled over 25 or 28 days. After 21 days, the concentration of CTC in the 22, 38 and 55°C swine digester slurries decreased 7, 80 and 98%, respectively. Coliforms in the 22°C digester slurries were still viable after 25 days; however, they were not detectable in the 38 and 55°C slurries after 3 and 1 days, respectively. After 28 days, the concentration of MON in the 22, 38 and 55°C cattle digester slurries decreased 3, 8 and 27%, respectively. Coliforms in the 22°C cattle digester slurries were still viable after 28 days; however, they were not detectable in the 38 and 55°C slurries after 14 and 1 days, respectively. Conclusions: These studies indicate that anaerobic digestion at 38 or 55°C may be an effective treatment to reduce coliforms and CTC; however, it is not an effective treatment to reduce MON. Significance and Impact of the Study: More studies are needed to determine which pharmaceuticals are susceptible to degradation by a specific manure treatment to prevent negative environmental consequences.  相似文献

Two experiments were conducted to evaluate the nutritional quality of cattle manure silage for ruminants. In experiment 1 a silage containing 50% cattle manure, 20% cane molasses (containing 0.4% urea) mixed with water in a 1:1.5 ratio, and 30% corn stover replaced 0, 50, 75 and 100% of corn stover in isonitrogenous diets (1.92% N) containing 70 or 80% corn stover (dry matter (DM) basis) in a 4×2 factorial arrangement. Forty lambs (34.5±3.2 kg body weight (BW)) were used to measure apparent digestibility and nitrogen balance in a completely randomized design with factorial arrangement (4×2) with five lambs per treatment. The cattle manure silage was also evaluated in growing Holstein heifers (212±32 kg BW) replacing 0, 25 or 37.5% of corn stover with cattle manure silage in isonitrogenous diets (2.24% N). Apparent digestibility of DM, organic matter (OM), neutral detergent fibre (NDF) and acid detergent fibre (ADF) were not affected. According to these results, digestibility is not affected by the level of cattle manure silage; however, intake was depressed at the highest levels of silage in lambs. Cattle manure silage may be considered as a potential by-product to be included in heifer's diet up to 37.5%, apparently without affecting performance. An incomplete anoxic fermentation of livestock waste results in offensive odor emissions. Antimicrobial additives may be useful in controlling odor emissions and pathogens. Natural antimicrobial compounds, carvacrol or thymol at 16.75 mM (2.5 g/l) completely inhibited the production of the offensive odor compounds, isobutyrate, valerate, isovalerate, and cresol, and significantly reduced other short-chain volatile fatty acids and gas emissions from swine waste. Fecal coliforms were reduced from 6.3 × 106 to 1.0 × 103 cells per ml 2 days after treatment with carvacrol (13.3 mM) and were not detectable within 14 days. Total culturable anaerobic bacteria were reduced from 12.4 × 1010 to 7.2 × 108 cells per ml after 2 days and were suppressed below this level for 28 days. Lactate production was not prevalent in untreated swine waste indicating that the microbial populations differ from those in cattle waste. Carvacrol and thymol were stable in swine waste under anoxic conditions for 62 days with 90 to 95% of the additive being recovered in the waste solids. In conclusion, carvacrol and thymol are not metabolized in anoxic swine waste and they are potentially useful in controlling odor emissions and pathogens in swine waste. Distillers' grains (DG), a by-product of ethanol production, are fed to cattle. Associations between Escherichia coli O157 prevalence and feeding of DG were investigated in feedlot cattle (n = 379) given one of three diets: steam-flaked corn (SFC) and 15% corn silage with 0 or 25% dried distillers' grains (DDG) or SFC with 5% corn silage and 25% DDG. Ten fecal samples were collected from each pen weekly for 12 weeks to isolate E. coli O157. Cattle fed 25% DDG with 5 or 15% silage had a higher (P = 0.01) prevalence of E. coli O157 than cattle fed a diet without DDG. Batch culture ruminal or fecal microbial fermentations were conducted to evaluate the effect of DDG on E. coli O157 growth. The first study utilized microbial inocula from steers fed SFC or dry-rolled corn with 0 or 25% DDG and included their diet as the substrate. Ruminal microbial fermentations from steers fed DDG had higher E. coli O157 contents than ruminal microbial fermentations from steers fed no DDG (P < 0.05) when no substrate was included. Fecal fermentations showed no DDG effect on E. coli O157 growth. In the second study with DDG as a substrate, ruminal fermentations with 0.5 g DDG had higher (P < 0.01) E. coli O157 concentrations at 24 h than ruminal fermentations with 0, 1, or 2 g DDG. In fecal fermentations, 2 g DDG resulted in a higher concentration (P < 0.05) at 24 h than 0, 0.5, or 1 g DDG. The results indicate that there is a positive association between DDG and E. coli O157 in cattle, and the findings should have important ramifications for food safety.  相似文献

Carvacrol and thymol in combination at 6.7 mM each completely inhibited the production of short-chain volatile fatty acids and lactate from cattle waste in anoxic flasks over 23 days. Fecal coliforms were reduced from 4.6 x 10(6) to 2.0 x 10(3) cells per ml 2 days after treatment and were nondetectable within 4 days. Total anaerobic bacteria were reduced from 8.4 x 10(10) to 1.5 x 10(7) cells per ml after 2 days and continued to be suppressed to that level after 14 days. If the concentration of carvacrol or thymol were doubled (13.3 mM), either could be used to obtain the same inhibitory fermentation effect. We conclude that carvacrol or thymol may be useful as an antimicrobial chemical to control pathogens and odor in stored livestock waste.  相似文献

施用脲酶抑制剂是降低尿素水解、减少氨气挥发损失、提高作物氮(N)肥利用率的重要途径之一.采用室内恒温、恒湿模拟试验方法,在25 ℃黑暗条件下培养,研究新型磷酰胺类脲酶抑制剂N-丙基磷酰三胺(NPPT)的脲酶抑制效果,比较其与N-丁基磷酰三胺(NBPT)在不同尿素用量条件下不同质地土壤中对脲酶的抑制差异.结果表明: 在壤土和黏土中,尿素作用时间≤9 d,添加抑制剂可以将尿素水解时间延长3 d以上.砂土中,尿素分解过程相对缓慢,添加抑制剂显著降低土壤脲酶活性,抑制NH4+-N生成.在培养期间,不同尿素用量条件下,脲酶抑制剂在不同质地土壤中的抑制效果表现为高施N量优于低施N量.培养第6天,在尿素用量250 mg N·kg-1条件下,NBPT和NPPT在砂土中脲酶抑制率分别为56.3%和53.0%,在壤土中分别为0.04%和0.3%,在黏土中分别为4.1%和6.2%;尿素用量500 mg N·kg-1,NBPT和NPPT在砂土中脲酶抑制率分别为59.4%和65.8%,在壤土中分别为14.5%和15.1%,在黏土中分别为49.1%和48.1%.不同质地土壤中脲酶抑制效果表现为砂土>黏土>壤土.不同抑制剂处理在培养期间土壤NH4+-N含量呈现先上升后下降的趋势,而NO3--N含量和表观硝化率均呈现逐渐上升的趋势.与单施尿素处理相比,添加脲酶抑制剂NBPT和NPPT显著增加土壤中的残留尿素态N,降低NH4+-N生成.新型脲酶抑制剂NPPT在不同质地土壤中的抑制效果与NBPT相似,是一款有效的脲酶抑制剂.  相似文献

本试验研究脲酶/硝化抑制剂不同组合在黑土和褐土中对尿素水解和硝化作用的调控效果,旨在筛选出适合东北黑土、褐土的高效抑制剂组合。采用室内恒温、恒湿培养试验,以不施氮肥(CK)和施用普通尿素肥料(U)为对照,研究分别添加脲酶抑制剂N-丁基硫代磷酰三胺(NBPT)及其与硝化抑制剂双氰胺(DCD)、3,4-二甲基吡唑磷酸盐(DMPP)、2-氯-6(三氯甲基)-吡啶(CP)、2-氨基-4-氯-6-甲基嘧啶(AM)、3-甲基吡唑(MP)组合制成的6种高效稳定性尿素在黑土和褐土中的尿素水解和氨氧化特征。在培养125 d内分别取土壤样品15次,通过测定2种土壤中尿素态氮、铵态氮和硝态氮含量,及氨氧化作用强度,计算硝化抑制率,确定最适合2种土壤的抑制剂或组合。结果表明: 尿素在黑土和褐土中水解时间约7 d,添加NBPT以及其与不同硝化抑制剂组合均能将尿素水解时间延长21 d以上。与U处理相比,添加抑制剂可显著增加土壤NH4+-N含量,降低NO3--N生成量,维持土壤中高NH4+-N含量的时间更久。黑土中,添加硝化抑制剂的处理均能显著抑制土壤硝化作用,有效硝化抑制时间超过125 d;DMPP、CP与NBPT配施使黑土NH4+-N含量提高1.6~1.8倍,培养125 d时其硝化抑制率分别为47.9%和24.1%。褐土中,U处理培养80 d左右基本完成硝化过程,而添加硝化抑制剂使硝化过程延长至少30 d;DCD、DMPP与NBPT配施使土壤NH4+-N含量提高2.1~3.4倍,培养125 d时其硝化抑制率分别为25.3%和23.2%。因此,尿素与NBPT+DMPP和NBPT+DCD制成的高效稳定性尿素分别在黑土和褐土中施用效果最好,其次分别是NBPT+CP和NBPT+DMPP。  相似文献

Physically effective fiber is needed by dairy cattle to prevent ruminal acidosis. This study aimed to examine the effects of different sources of physically effective fiber on the populations of fibrolytic bacteria and methanogens. Five ruminally cannulated Holstein cows were each fed five diets differing in physically effective fiber sources over 15 weeks (21 days/period) in a Latin Square design: (1) 44.1% corn silage, (2) 34.0% corn silage plus 11.5% alfalfa hay, (3) 34.0% corn silage plus 5.1% wheat straw, (4) 36.1% corn silage plus 10.1% wheat straw, and (5) 34.0% corn silage plus 5.5% corn stover. The impact of the physically effective fiber sources on total bacteria and archaea were examined using denaturing gradient gel electrophoresis. Specific real-time PCR assays were used to quantify total bacteria, total archaea, the genus Butyrivibrio, Fibrobacter succinogenes, Ruminococcus albus, Ruminococcus flavefaciens and three uncultured rumen bacteria that were identified from adhering ruminal fractions in a previous study. No significant differences were observed among the different sources of physical effective fiber with respect to the microbial populations quantified. Any of the physically effective fiber sources may be fed to dairy cattle without negative impact on the ruminal microbial community.  相似文献

As lack of forage resource, alternative roughage sources have been developed for ruminant production and their inclusion would exert a great effect on the dietary nutrition, consequently affecting animal performance. Four silages (corn silage (CS), corn stalk silage (SS), inoculated CS and inoculated SS) were separately offered to 60 Bohai Black cattle (15 cattle/group) during a 24-week finishing period, in which the growth performance, carcass trait, beef quality and oxidative stability of steers were determined. Neither silage material nor silage inoculant exerted a significant effect on the growth performance, carcass trait and oxidative stability of beef cattle (P>0.05). As to beef quality, cattle offered CS had higher (P<0.05) contents of intramuscular fat than those offered SS along with a lower moisture content (P<0.05). The contents (mg/g muscle) of C10 : 0, C12 : 0, C14 : 1, C16 : 0, C16 : 1, C18 : 1n9c, C18 : 2n6c, C18 : 3n3, C20 : 1n9, C20 : 2, C20 : 3n6, saturated fatty acids, monounsaturated fatty acids, polyunsaturated fatty acids and n-6 fatty acids were higher (P<0.05) in the beef muscle of animals offered CS than those offered SS, whereas inoculated treatment made no difference (P>0.05) on the proximate components and fatty acids profile of beef muscle. There was neither an interaction (P>0.05) between inoculated treatment and silage material. There were no differences (P>0.05) in cholesterol content and meat quality traits in animals fed alternative silages. The collective findings suggest that it is not economical to substitute high-quality forage for relative low-quality forage in a high-concentrate finishing ration of beef cattle and silage inoculant inclusion would not exert a direct effect on animal performance.  相似文献

以持续9年施用不同缓/控释尿素的水田棕壤为试验对象,以普通大颗粒尿素为对照,研究了持续施用不同缓/控释尿素条件下水田土壤NH3挥发与N2O排放特征.结果表明: 与普通大颗粒尿素(U)相比,除1% 3,4-二甲基吡唑磷酸盐(DMPP)+U处理 NH3挥发增加了25.8%外,其他缓/控释尿素肥料处理对NH3有明显的减排效果.树脂包膜尿素(PCU)对NH3减排效果最明显,为73.4%,硫包膜尿素(SCU)为72.2%,0.5% N-丁基硫代磷酰三胺(NBPT)+1% DMPP+U为71.9%,1% 氢醌(HQ)+3% 双氰胺(DCD)+U为46.9%,0.5% NBPT+U为43.2%,1% HQ+U为40.2%,3% DCD+U为25.5%, 1% DMPP均与施用普通大颗粒尿素差异显著;所有缓/控释尿素处理与对照相比均可显著减少N2O排放.1% DMPP+U对N2O减排效果最明显,为74.9%,PCU为62.1%,1% HQ+3% DCD+U为54.7%,0.5% NBPT+1% DMPP+U为42.2%,3% DCD+U为35.9%,1% HQ+U为28.9%,0.5% NBPT+U为17.7%,SCU为14.5%,均与施用普通大颗粒尿素差异显著.比较0.5% NBPT+1% DMPP+U、SCU、PCU对NH3和N2O减排的综合效果,3种肥料作用相近,且均明显优于其他处理,但包膜材料的成本较抑制剂高数倍.因此,同时添加脲酶和硝化抑制剂的缓释尿素是减少水田氮素损失及环境污染的首选氮肥.  相似文献

Acute toxicities of three naturally occurring monoterpenoid essential oil constituents and the essential oil of rosemary were tested against late instars of Agriotes obscurus (L.) (Coleoptera: Elateridae). Both contact and volatile toxicities of thymol, citronellal, eugenol, and rosemary oil were determined. Also, phytotoxicity of these compounds was evaluated on corn germination and seedling development. Thymol had the greatest contact toxicity (LD50 = 196.0 microg/larva), whereas citronellal and eugenol were less toxic (LD50 = 404.9 and 516.5 microg/larva, respectively). Rosemary oil did not show any significant contact toxicity, even at 1,600 microg/larva. In terms of volatile toxicity, citronellal was the most toxic to wireworm larvae (LC50 = 6.3 microg/cm3) followed by rosemary oil (LC50 = 15.9 microg/cm3), thymol (LC50 = 17.1 microg/cm3), and eugenol (LC50 = 20.9 microg/cm3). Thymol, eugenol, and citronellal significantly inhibited corn seed germination and development, whereas rosemary oil had only minimal phytotoxic effects.  相似文献

The effect of thymol on the ATPase activity of myosin subfragment-1 (S1) and on the contractile properties of skinned skeletal muscle fibers was studied. At concentrations of 1.5-2 mM, thymol activated the S1 ATPase substantially and the actin-activated S1 ATPase modestly. At the same concentrations, the isometric force of skinned skeletal muscle fibers was modestly suppressed (11% at 2 mM). However, the kinetic parameters of contraction were suppressed more: the velocity of shortening and the rate of force redevelopment after shortening were suppressed by 43% and 31% at 2 mM, respectively. Thus, among other small-molecule inhibitors, thymol is unique in that it has opposite effects on the enzymatic activity and kinetic parameters of contraction. Thymol may serve as a potent tool for studying the mechanism of coupling between the ATPase reaction and contraction in muscle.  相似文献

Thymol (5-methyl-2-isopropylphenol) is a phenolic compound that is used to inhibit oral bacteria. Because little is known regarding the effects of this compound on ruminal microorganisms, the objective of this study was to determine the effects of thymol on growth and lactate production by the ruminal bacteria Streptococcus bovis JB1 and Selenomonas ruminantium HD4. In addition, the effect of thymol on the in vitro fermentation of glucose by mixed ruminal microorganisms was investigated. Neither 45 nor 90 μg/ml of thymol had any significant effect on growth or lactate production by S. bovis JB1, but 180 μg/ml of thymol completely inhibited growth and lactate production. In the case of S. ruminantium HD4, 45 μg/ml of thymol had little effect on growth and lactate production; however, 90 μg/ml of thymol completely inhibited growth of S. ruminantium HD4. Thymol also decreased glucose uptake by whole cells of both bacteria. When mixed ruminal microorganisms were incubated in medium that contained glucose, 400 μg/ml of thymol increased final pH and the acetate to propionate ratio and decreased concentrations of methane, acetate, propionate, and lactate. In conclusion, thymol was a potent inhibitor of glucose fermentation by S. bovis JB1 and S. ruminantium HD4. Even though thymol treatment decreased methane and lactate concentrations and increased final pH in mixed ruminal microorganism fermentations of glucose, concentrations of acetate and propionate were also reduced. It has been suggested that the ability of live yeast to improve milk yield and weight gain in cattle is because the yeast stimulates bacterial activity within the rumen. However it remains unclear if this is a general stimulation of all species or a specific stimulation of certain species. Here we characterised the change in the bacterial population within the rumen of cattle fed supplemental live yeast. Three cannulated lactating cows received a daily ration (24 kg/d) of corn silage (61% of DM), concentrates (30% of DM), dehydrated alfalfa (9% of DM) and a minerals and vitamins mix (1% of DM). The effect of yeast (BIOSAF SC 47, Lesaffre Feed Additives, France; 0.5 or 5 g/d) was compared to a control (no additive) in a 3×3 Latin square design. The variation in the rumen bacterial community between treatments was assessed using Serial Analysis of V1 Ribosomal Sequence Tag (SARST-V1) and 454 pyrosequencing based on analysis of the 16S rRNA gene. Compared to the control diet supplementation of probiotic yeast maintained a healthy fermentation in the rumen of lactating cattle (higher VFA concentration [high yeast dose only], higher rumen pH, and lower Eh and lactate). These improvements were accompanied with a shift in the main fibrolytic group (Fibrobacter and Ruminococcus) and lactate utilising bacteria (Megasphaera and Selenomonas). In addition we have shown that the analysis of short V1 region of 16s rRNA gene (50–60 bp) could give as much phylogenetic information as a longer read (454 pyrosequencing of 250 bp). This study also highlights the difficulty of drawing conclusions on composition and diversity of complex microbiota because of the variation caused by the use of different methods (sequencing technology and/or analysis).  相似文献

In this study, it is hypothesized that anaerobic hydrolysis of organic solid wastes (OSWs) in leachbed reactors can be enhanced by two approaches: (1) by increasing the porosity of the leachbed; and (2) by seeding the bed with brown-rot fungi or anaerobically digested residues. The hypothesis is verified using cattle manure as a model OSW and pistachios-half-shell as an inert additive to increase the porosity of the leachbed. The enhancement was quantified in terms of volatile fatty acid (VFA) generation and soluble COD generation. Results of this study showed that 15% more VFAs and 8% more soluble COD can be obtained at higher bed porosities and with brown-rot fungi or digested residues as the seed. Compared to a leachbed without any recirculation, porosity enhancement, nor seed addition, the VFA yield (g VFA/g manure) in a leachbed with pistachios-half-shell as porosity enhancers was 193% higher; that in a leachbed with leachate recirculation (of 4 L/day), pistachios-half-shell as porosity enhancers, and brown-rot fungi or digested sludge as seed was 230% higher.  相似文献

Holstein heifers were divided into three groups of 27 at least 60 days before any were inseminated to evaluate gross effects on ovarian activity when urea was fed continuously for more than 2 years. Groups were reduced to 21 heifers each before calving. In Groups 1, 2 and 3, respectively, urea nitrogen replaced 0, 22 and 45% of plant nitrogen in heifer rations from insemination to mid pregnancy, 0, 8 and 16% there-after until 2 weeks before calving, and 0, 18 and 36% precalving and during lactation. The rations were blended and fed free-choice. Dry matter in the lactation ration was 44% corn silage, 10% alfalfa bromegrass silage and 46% concentrate. Body weight increases were similar on each ration through second calving. Likewise, estrous cycles and intervals between inseminations were similar. After the second calving, groups were not different for time to first ovulation or for intervals between ovulations. In general, urea did not affect gross measures of ovarian activity. Whole crop third cut alfalfa, brown mid-rib (bmr) corn, and corn were chopped and inoculated with one of four microbial inoculants used. Uninoculated silage was the control treatment. Each crop was ensiled in four mini-silos (1 L glass jars) per treatment. All silos were fermented for 60 days at room temperature (22 °C), and then they were opened and analyzed for fermentation products, fiber constituents and N fractions. A fraction of wet silage was ground with a blender for 30 s. In vitro gas production was measured in 160 ml sealed serum vials at 3, 6, 9, 24, and 48 h using the wet ground silage. At 9 and 48 h, rumen fluid was analyzed for volatile fatty acids (VFA) and microbial biomass yield (MBY). In all the three crops, the four inoculants produced only minor changes in pH and fermentation products during ensiling. Of the variables measured, soluble nonprotein N fractions were the characteristics most often affected by some inoculants. At 9 h incubation, in vitro gas production and VFA did not differ between control and inoculated silages, but MBY did. Among crops, alfalfa and corn silages had higher MBY than did bmr corn silage. Among inoculants, three of the four inoculated silages produced more MBY than did control. At 48 h, alfalfa silage produced higher MBY than did corn or bmr silage, and two of the inoculated silages had more MBY than did the control. There was no inoculant by crop interaction. Results suggest that some silage inoculants are capable of altering rumen fermentation, even in cases where effects on silage fermentation are small, and that this effect may be linked to better preservation of crop protein during ensiling.  相似文献
