Two bacteriophages isolated onEnterobacter cloacae (C2, C2F) and three isolated onErwinia herbicola (E3, E16P, E16B) were purified by D2O gradient centrifugation. Phage-containing fractions were negatively stained and examined by electron microscopy. Phages C2, C2F, E3, and E16P showed an elongated head 153×51 nm and a short noncontractile tail 12 nm long terminated by at least two short fibers. These phages correspond to the rate taxonomic group C3. Big capsomeres composing the phage head were evidenced when phage suspensions in D2O were stained. Phage E16B showed an elongated head 97×40.5 nm, and a contractile tail 89 nm long. This phage corresponds to the extremely rate group A3.  相似文献

We here characterize five globular endolysins, encoded by a set of Gram-negative infecting bacteriophages: BcepC6gp22 (Burkholderia cepacia phage BcepC6B), P2gp09 (Escherichia coli phage P2), PsP3gp10 (Salmonella enterica phage PsP3), K11gp3.5 and KP32gp15 (Klebsiella pneumoniae phages K11 and KP32, respectively). In silico, BcepC6gp22, P2gp10 and PsP3gp10 are predicted to possess lytic transglycosylase activity, whereas K11gp3.5 and KP32gp15 have putative amidase activity. All five endolysins show muralytic activity on the peptidoglycan of several Gram-negative bacterial species. In vitro, Pseudomonas aeruginosa PAO1 is clearly sensitive for the antibacterial action of the five endolysins in the presence of the outer membrane permeabilizer EDTA: reductions are ranging from 1.89 to 3.08 log units dependent on the endolysin. The predicted transglycosylases BcepC6gp22, P2gp10 and PsP3gp10 have a substantially higher muralytic and in vitro antibacterial activity compared to the predicted amidases K11gp3.5 and KP32gp15, highlighting the impact of the catalytic specificity on endolysin activity. Furthermore, initial data exclude the synergistic lethal effect of a combination of the predicted transglycosylase PsP3gp10 and the predicted amidase K11gp3.5 on PAO1. As these globular endolysins show a lower enzymatic and antibacterial activity, in comparison to modular endolysins, we suggest that the latter should be favored for antibacterial applications.  相似文献

Bacteriol. 92:148-154. 1966.-A medium for the growth of group A streptococcal phages is described, consisting of Brain Heart Infusion broth supplemented with 0.2% yeast extract, 10(-4)m CaCl(2), and 10 mug/ml of dl-tryptophan. Cell and phage growth in this medium was excellent, and did not require the addition of serum or other proteins as indicated by other workers. Growth of one phage has also been achieved in a completely synthetic medium. The adsorption characteristics of two group A phages in protein broth and synthetic broth were studied, and the initial adsorption of phage was found to be more extensive in synthetic broth. However, the final amounts of adsorption in both were similar. The addition of purified group A carbohydrate antigen to the adsorption mixture in synthetic broth had no effect on the adsorption, and cells containing type-specific M protein adsorbed phage at the same rate as those lacking M protein. It was concluded that neither the group antigen nor the type antigen was the primary site of phage adsorption. One-step growth curves of the two phages showed a second step or burst occurring. Sonic oscillation of the bacterial cultures, which broke up the chains to single cells, abolished the second step of the growth curve. It appears that the second step is a function of the chain formation of streptococcal cells.  相似文献

Species composition and a number of persistence characteristics enterobacteria isolated from urine of 42 pregnant and 22 nonpregnant women with pyelonephritis (relapse, remission), from prostatic fluid of 225 males and secretions of cervical canal of 124 women with urogenital pathology (prostatitis, salpingo-oophoritis) were studied. The study revealed that enterobacteria, including Escherichia coli, prevailed in the structure of uromicroflora (66.7-83.3%) and constituted a relatively small proportion among "genital" isolates of microorganisms (19.9-22.2%). Male and female sterility and the presence of enterobacteria in the reproductive tract of patients were found to be directly correlated. Clinical isolates of enterobacteria were shown to possess pronounced seroresistance and the complex of persistence characteristics, including antilysozyme, anti-intercidal and anticomplementary activity.  相似文献

Exposure to 1 M Ca(NO3)2 inactivated both phages, and phage ?UW 51 was also susceptible to 1 M CaCl2, 0.1 M tris(hydroxymethyl)aminomethane, and 0.3% H2O2. Phage plating efficiency was highest on the cultures at logarithmic phase and sometimes much influenced by host growth. FiveRhizobium meliloti bacteriophages isolated from soil or lysogenic bacteria and belonging to Bradley's group B or theSiphoviridae family of tailed phaes were studied. They are of identical morphology, showing isometric heads and long, noncontractile tails with transverse bars. They are temperate and closely related by host range, DNA restriction endonuclease patterns and homology, DNA mass, serological properties, adsorption velocity, and latent period. Although fiveBacteroides fragilis bacteriophages isolated over a six-year period in Nebraska and Virginia had similar physical characteristics (morphology, temperature inactivation, and sensitivity to organic solvents and antisera), there were some statistically significant differences between the phages. In addition, restriction endonuclease analysis revealed that three of the five DNAs were not identical. However, the DNAs of the phages were closely related based on DNA-DNA hybridization, percent homologies, and possession of homologous regions of DNA. It appears that the five phages are strains of the same species of phage, although each phage has a unique host range spectrum.  相似文献

The closely related lipid-containing bacteriophages PRD1, PR4, PR5, PR722 and L17 isolated from different parts of the world have double-stranded DNA genomes which replicate in a linear form. The nucleotide (nt) sequences of the genome termini of these viruses reveal 110-111-bp-long inverted terminal repeats (ITRs). Both ends of the viral DNA are identical. The first 18 bp and the last 35 bp of the ITRs are totally conserved in all viruses. Between these conserved nt sequences there is a variable sequence, which enables us to divide the phages into two groups. Comparison of the virus ITRs led also to the identification of a 10-bp-long A + T stretch, where the only changes observed were transversions between A and T. The termini of the PRD1 virus family genomes exhibit sequence similarities to those of phi 29 and Cp-1 families.  相似文献

The main cytotoxic and cytotonic enterotoxins, transient pore forming toxins (RTX), cytotoxic necrotizing factor and injection toxins have been analyzed. In complex DNA bacteriophages like lambda, T4, T7, P22, P2, the DNA is packaged into a preformed precursor particle which sometimes has a smaller size and often a shape different from that of the phage head. This packaging mechanism is different from the one suggested for the RNA phages, according to which RNA nucleates the shell formation. The different mechanisms could be understood by comparing the genomes to be packaged: single stranded fII RNA has a very compact structure with high helix content. It might easily form quasispherical structures in solution (as seen in the electron microscope by Thach & Thach (1973)) around which the capsid could assemble. Double stranded phage DNA, on the other hand, is a rigid molecule which occupies a large volume in solution and has to be concentrated 15-fold during packaging into the preformed capsid, and the change in the capsid structure observed hereby might provide the necessary DNA condensation energy.  相似文献

Growth of enterobacteria on fructo-oligosaccharides   总被引:3,自引:0,他引:3  
Fructo-oligosaccharides (FOS) can be fermented by most species of enterobacteria present in the human intestine. Fermentation was confirmed by increased growth rates, low final pH and degradation patterns using high performance anion exchange chromatography (HPAEC). Growth rates were increased when FOS was added to the growth medium. Growth rates on all substrates differed widely between strains within the same species. HPAEC analysis showed that each strain degraded the oligosaccharides differently, but a preference for the smaller oligosaccharides was observed. No differences were observed between the two commercial preparations, the inulo-oligosaccharides and neosugars. Fermentation was rapid as could be determined by acidification tests using cell suspensions. It can be concluded that enterobacteria may play a role in overall fermentation of FOS in the colon and, in addition, due to competitive exclusion, may prevent survival of ingested pathogenic enterobacteria.  相似文献
