Twenty-one X-linked recessive lethal and sterile mutations balanced by an unlinked X-chromosome duplication have been identified following EMS treatment of the small nematode, Caenorhabditis elegans. The mutations have been assigned by complementation analysis to 14 genes, four of which have more than one mutant allele. Four mutants, all alleles, are temperature-sensitive embryonic lethals. Twelve mutants, in ten genes, are early larval lethals. Two mutants are late larval lethals, and the expression of one of these is influenced by the number of X chromosomes in the genotype. Two mutants are maternal-effect lethals; for both, oocytes made by mutant hermaphrodites are rescuable by wild-type sperm. One of the maternal-effect lethals and two larval lethals are allelic. One mutant makes defective sperm. The lethals and steriles have been mapped by recombination and by complementation testing against 19 deficiencies identified after X-ray treatment. The deficiencies divide the region, about 15% of the X-chromosome linkage map, into at least nine segments. The deficiencies have also been used to check the phenotypes of hemizygous lethal and sterile hermaphrodites.  相似文献

Spore formation is a sophisticated mechanism by which some bacteria survive conditions of stress and starvation by producing a multilayered protective capsule enclosing their condensed DNA. Spores are highly resistant to damage by heat, radiation, and commonly employed antibacterial agents. Previously, spores have also been shown to be resistant to photodynamic inactivation using dyes and light that easily destroy the corresponding vegetative bacteria. We have discovered that Bacillus spores are susceptible to photoinactivation by phenothiazinium dyes and low doses of red light. Dimethylmethylene blue, methylene blue, new methylene blue, and toluidine blue O are all effective, while alternative photosensitizers such as Rose Bengal, polylysine chlorin(e6) conjugate, a tricationic porphyrin, and a benzoporphyrin derivative, which easily kill vegetative cells, are ineffective. Spores of Bacillus cereus and B. thuringiensis are most susceptible, B. subtilis and B. atrophaeus are also killed, and B. megaterium is resistant. Photoinactivation is most effective when excess dye is washed from the spores, showing that the dye binds to the spores and that excess dye in solution can quench light delivery. The relatively mild conditions needed for spore killing could have applications for treating wounds contaminated by anthrax spores, for which conventional sporicides would have unacceptable tissue toxicity.  相似文献

Molecular phylogenetic analyses have revealed many relationships in Veronica (Plantaginaceae) never anticipated before. However, phytochemical characters show good congruence with DNA-based analyses. We have analysed a combined data set of 49 species and subspecies derived from the nuclear ribosomal ITS-region (18 new sequences) and the plastid trnL-F region (12 new sequences) of Veronica emphasizing subgenera Chamaedrys and Pocilla and separate analyses of subgenera Pentasepalae (ITS only) and Pseudolysimachium. Results for subgenus Chamaedrys show that European and Asian perennial species are monophyletic sister groups with the annual species consecutive sisters to them. All species of Veronica that contain cornoside are found in this subgenus, although some species seem to have secondarily lost the ability to produce this compound. Subgenera Pocilla and Pentasepalae are well supported sister groups characterized by the occurrence of 8-hydroxyflavones. The traditional subsection Biloba of subgenus Pocilla is biphyletic with Veronica intercedens being clearly separate from the rest of the group. This result is mirrored by the unusual phytochemical arsenal of V. intercedens, which is the only species in the genus analysed to date to contain melittoside and globularifolin. Subgenus Pentasepalae appears to be a clade of diverse lineages from southwestern Asia and a single European clade. Species shown to have 6-hydroxyflavones do not form a monophyletic group. Subgenus Pseudolysimachium seems to have originated in Eastern Asia. 6-Hydroxyflavones acylated with phenolic acids are common in this subgenus but may have originated only later in the evolution of the group. Possible chemotaxonomic markers for other groups are discussed.  相似文献

Fully subtituted di-O-acetyl-N-acetylchitosan (chitin diacetate) has been prepared by a route in which the hydroxyl groups are acetylated prior to N-acetylation. This overcomes the previously reported intramolecular steric hindrance to esterification caused by the N-acetamido group. The resultant products were of high viscosity but had a limited solubility range. Di-O-acrylcarbamate derivatives of N-acetylchitosan (chitin) have been produced by a similar route, whilst di-O-arylcarbamate-N-arylureidochitosans have been prepared directly from chitosan. These products also have limited solubility ranges and have inherent viscosities similar to that of di-O-acetylchitosan prepared from the same batch of chitosan.  相似文献

We have expanded our collection of recessive lethal and sterile mutants in the region of the X chromosome balanced by mnDp1(X;V), about 15% of the X linkage map, to a total of 54 mutants. The mutations have been mapped with respect to 20 overlapping deficiencies and five X duplications, and they have been assigned to 24 genes by complementation testing. Nine mutants are hermaphrodite-sterile: one of these is a sperm-defect mutant, two have abnormal gonadogeneses and six, in five genes, are maternally influenced mutants, producing inviable zygote progeny. One of the gonadogenesis mutants and two of the maternally influenced mutants are male fertile. All but one of the maternally influenced mutants give cross progeny when mated with wild-type males. Forty-three mutants were tested for suppression by homozygous sup-5(e1464), which is believed to be specific for null alleles. Ten mutants that were judged by independent criteria not to be null mutants are not suppressed. Nine of the other 33 mutants, in nine genes, are suppressed, five in both heterozygous and homozygous suppressor stocks and four only in homozygous suppressor stocks.  相似文献

Flagellate species have been shown to survive transocean passage by ballast water and the large dinoflagellateGymnodinium catenatum was introduced from Japanese to Tasmanian waters in this way.Gymnodinium mikimotoi—better known asGyrodinium aureolum—andFibrocapsa japonica as well asAlexandrium leeii are good candidates to have been introduced recently. Species which seem to have been introduced recently into the North Sea but apparently are transported from adjacent seas by currents into the region areGymnodinium chlorophorum andAlexandrium minutum. Species reported as introduced due to misidentifications areGymnodinium catenatum andLepidodinium viride. Under other names the speciesProrocentrum minimum, Prorocentrum redfieldii, andHeterosigma akashiwo have been known for a long time in the North Sea. The recent reports of threeChattonella species may be either due to introduction or they have been overlooked. The reasons why the introduction of flagellates into coastal North Sea waters is difficult to prove will be discussed.  相似文献

Cattle are the only non-primate species to have an expanded KIR gene family. In addition they have a single Ly49 gene, thought to be monomorphic. We have identified two additional Ly49 cDNA sequences in cattle that encode molecules predicted to differ by 14 and 16 amino acids from the original sequence. All available data indicate the presence of only one Ly49 gene in cattle, thus the divergent sequences reported here may represent ancient allelic lineages, a high level of polymorphism in an ancestral gene or a previously expanded and subsequently contracted Ly49 gene family. Cattle are the only species known to have an expanded polymorphic KIR gene family and a polymorphic Ly49 gene.  相似文献

The Collembola fauna of Iran is little known and no comprehensive examination of this group of Hexapoda is available for this region. The only notable work on Collembola was carried out by Cox (1982). Recently, studies on the Collembola fauna have started in several regions. In this paper, publications by different researchers are documented and the species that have been found in different regions of Iran until January 2013 are listed. At present, 112 species, belonging to 18 families and 57 genera are known from Iran.  相似文献

Tree invasions have substantial impacts on biodiversity and ecosystem functioning, and trees that are dispersed by animals are more likely to become invasive. In addition, hybridisation between plants is well documented as a source of new weeds, as hybrids gain new characteristics that allow them to become invasive. Corymbia torelliana is an invasive tree with an unusual animal dispersal mechanism: seed dispersal by stingless bees, that hybridizes readily with other species. We examined hybrids between C. torelliana and C. citriodora subsp. citriodora to determine whether hybrids have inherited the seed dispersal characteristics of C. torelliana that allow bee dispersal. Some hybrid fruits displayed the characteristic hollowness, resin production and resin chemistry associated with seed dispersal by bees. However, we did not observe bees foraging on any hybrid fruits until they had been damaged. We conclude that C. torelliana and C. citriodora subsp. citriodora hybrids can inherit some fruit characters that are associated with dispersal by bees, but we did not find a hybrid with the complete set of characters that would enable bee dispersal. However, around 20,000 hybrids have been planted in Australia, and ongoing monitoring is necessary to identify any hybrids that may become invasive.  相似文献

Fifteen species of Heterostropha are described, 12 of them for the first time. All are newly interpreted with regard to their taxonomic relation to fossil and living gastropods. The Streptacidoidea with long Paleozoic history are represented in the Late Triassic St. Cassian Formation by several genera that can be differentiated into four families. The Ebalidae are represented byEbala, with smooth protoconch, Cassianebalidae byCassianebala andLoxebala with axially ornamented protoconch. The Donaldinidae of St. Cassian are represented by one species ofDonaldina and two ofNeodonaldina that stand in the continuation of Paleozoic species ofDonaldina. Architectonicoidea with shells coiled in a plane and Valvatoidea appear in the St. Cassian fauna without known Paleozoic relation. In the former superfamily the Architectonicidae can be recognized in the genusRinaldoconchus with two species. Cassianaxidae withCassianaxis, Amphitomariidae withAmphitomaria, Stuoraxidae withStuoraxis andAmpezzogyra have a sinistral protoconch and planispirally coiled dextral teleconchs. They all resemble different modern species that have similarly small shells. Modern Hyalogyrinidae have withAlexogyra a new representative from the Triassic. The Valvatoidea are represented with the generaCarboninia andBandellina of the Cornirostridae in the Triassic representatives. The relation of described species in the system of the Heterostropha is discussed.  相似文献

The histone patterns in Xenopus laevis and X. borealis have been examined at various developmental stages using acid-urea polyacrylamide electrophoresis. Qualitative differences between the two species have been demonstrated in the H1 fraction. These differences are not affected by alkaline phosphatase digestion. Quantitative changes during development in both H1 and H4 fractions have also been observed. In addition, histones from hybrids of the two species have been examined. H1 histones characteristic of both species are present by the neurula stage in laevis ♀ × borealis ♂ hybrids. Quantitative changes observed in borealis development are mimicked in laevis ♀ × borealis ♂ hybrid development.  相似文献

The positions of the metBJF and the argECBH sequences on F14 have been mapped by studying heteroduplexes of F14 with φ80dmet and φ80darg transducing phage DNAs. The structures of the DNAs of the transducing phage φ80d-metB isolated by Konrad (1969), of two φ80dmetB phages isolated by Press et al. (1971), and of some derived φ80darg phages, have been determined. They all have complex structures. In addition to the bacterial chromosome sequences corresponding to the met and arg genes, they contain certain F sequences, which have been recognized as active in F-related recombination events. Plausible models for the integration and excision events leading to the formation of the phage DNA molecules are proposed.  相似文献

During the recent years extensive efforts have been made to find out bacteriocins from lactic acid bacteria (LAB) active against various food spoilage and pathogenic bacteria, and superior stabilities against heat treatments and pH variations. Bacteriocins isolated from LAB have been grouped into four classes. Circular bacteriocins which were earlier grouped among the four groups of bacteriocins, have recently been proposed to be classified into a different class, making it class V bacteriocins. Circular bacteriocins are special molecules, whose precursors must be post translationally modified to join the N to C termini with a head-to-tail peptide bond. Cyclization appears to make them less susceptible to proteolytic cleavage, high temperature and pH, and, therefore, provides enhanced stability as compared to linear bacteriocins. The advantages of circularization are also reflected by the fact that a significant number of macrocyclic natural products have found pharmaceutical applications. Circular bacteriocins were unknown two decades ago, and even to date, only a few circular bacteriocins from a diverse group of Gram positive organisms have been reported. The first example of a circular bacteriocin was enterocin AS-48, produced by Enterococcus faecalis AS-48. Gassereccin A, produced by Lactobacillus gasseri LA39, Reutericin 6 produced by Lactobacillus reuteri LA6 and Circularin A, produced by Clostridium beijerinickii ATCC 25,752, are further examples of this group of antimicrobial peptides. In the present scenario, Gassericin A can be an important tool in the food preservation owing to its properties of high pH and temperature tolerance and the fact that it is produced by LAB L. gasseri, whose many strains are proven probiotic.  相似文献

The mating-type locus of Podospora anserina controls fusion of sexual cells as well as subsequent stages of development of the fruiting bodies. The two alleles at the locus are defined by specific DNA regions comprising 3.8 kb for mat+ and 4.7 kb for mat?, which have identical flanking sequences. Here we present the characterization of several mutants that have lost mat+-specific sequences. One mutant was obtained fortuitously and the other two were constructed by gene replacement. The mutants are deficient in mating with strains of either mat genotype but are still able to differentiate sexual reproductive structures. The loss of the mating type does not lead to any discernible phenotype during vegetative growth: in particular it does not change the life span of the strain. The mutants can recover mating ability if they are transformed with DNA containing the complete mat+ or mat? information. The transformants behave in crosses as do the reference mat+ or mat? strains, thus indicating that the transgenic mat+ and mat? are fully functional even when they have integrated at ectopic sites.  相似文献

Genetic studies of plants and their pathogens indicate that dominant alleles for resistance in hosts are complemented by corresponding dominant alleles for avirulence in pathogens. Products of these genes have not yet been identified. We have produced murine monoclonal antibodies (mAbs) to extracellular antigens of the fungal soybean pathogen Phytophthora megasperma f. sp. glycinea (Pmg, race 1) as part of a larger effort to identify antigenic determinants associated with particular avirulence genes. Thirty-six independent mAbs have been characterized by binding to Western blots of Pmg extracellular glycoproteins and by enzyme-linked immunosorbent assay with glycoproteins modified by treatment with periodate, α-mannosidase, and endo-β-N-acetylglucosaminidase H. The mAbs are predominantly carbohydrate-specific and can be placed in six groups based on interactions with Pmg glycoproteins. Binding patterns of various mAbs to Western blots indicate that Pmg proteins may have single or multiple types of attached carbohydrate antigens. Races of Pmg with differing avirulence genes exhibit more characteristic differences by Western blot analysis than by protein staining of glycoprotein profiles. Several of the mAbs show much higher reaction levels to glycoproteins from race 1 than from two other races. All of the glycoprotein-specific mAbs cross-react with purified mycelial walls.  相似文献

The COT1 and ZRC1 genes of Saccharomyces cerevisiae are structurally related dosage-dependent suppressors of metal toxicity. COT1 confers increased tolerance to high levels of cobalt; ZRC1 confers increased tolerance to high levels of zinc. The two genes are not linked and have been mapped; COT1 to chromosome XV and ZRC1 to chromosome XIII. Phenotypes related to metal homeostasis have been examined in strains with varied COT1 and ZRC1 gene doses. Overexpression of COT1 confers tolerance to moderately toxic levels of zinc and ZRC1 confers tolerance to moderately toxic levels of cobalt. Strains that carry null alleles at both loci are viable. The metal-hypersensitive phenotypes of mutations in either gene are largely unaffected by changes in dosage of the other. COT1 and ZRCI function independently in conferring tolerance to their respective metals, yet the uptake of cobalt ions by yeast cells is dependent on the gene dosage of ZRC1 as well as of COT1 Strains that overexpress ZRC1 have increased uptake of cobalt ions, while ZRCI null mutants exhibit decreased cobalt uptake. The defects in cobalt uptake due to mutations at COT1 and ZRC1 are additive, suggesting that the two genes are responsible for the majority of cobalt and zinc uptake in yeast cells. The function of either gene product seems to be more important in metal homeostasis than is the GRR1 gene product, which is also involved in metal metabolism. Mutations in the GRR1 gene have no effect on the cobalt-related phenotypes of strains that have altered gene dosage of either COT1 or ZRC1.  相似文献

A number of abnormal polypeptides which are products of the lacZ gene in Escherichia coli have been characterized. A variety of experiments indicates that one class of at least nine different fragments arises from premature termination of protein synthesis. They have been detected as products of protein synthesis both in vitro and in vivo, although the percentage of fragments made in vitro is greater than that in vivo. The percentage of total Z gene-encoded protein which is found as fragments is estimated to be roughly 23% in vivo. This means that about 31% of the total number of β-galactosidase monomers synthesized are prematurely terminated molecules. The average molecular weight of the polypeptides produced is 91,000. This corresponds to a termination event occurring on the average once every 3200 codons. The sites at which termination occurs appear to be specific and are located primarily near the 3′-end of the gene.Polypeptides synthesized in vitro and in vivo from templates containing mutations in the Z gene have also been compared. Most of the mutationally generated fragments synthesized in vitro are stable, unlike their in vivo counterparts, which are often rapidly degraded. One fragment, however, generated by an early amber mutation in the Z gene, is degraded in the in vitro system. The mechanism of degradation appears to be specific for small abnormal polypeptides. Internal reinitiation polypeptides generated by nonsense mutations, which have been found in vivo, are not detected in the in vitro protein synthesis system under the conditions used here.  相似文献

Data on the morphology and behavior of five species of spionid polychaete from muddy sand habitats in both the United Kingdom and the northwest coast of North America are presented. The data are consistent with the hypothesis that browsing predation by visual predators is important now and has been important in the evolutionary past. As predicted by such a hypothesis, regenerating individuals are common in all the populations examined. Individuals belonging to species that expose their anterior ends while feeding and/or defecating have cryptic anteriors (Pygospio elegans Claparède, Rhynchospio glutaeus (Ehlers), Malacoceros fuliginosus (Claparède), and Spiophanes bombyx (Claparède)). Individuals belonging to species that do not expose their anteriors while feeding and/or defecating do not have cryptic anteriors (Pseudopolydora kempi (Southern)). Given these data it is still unclear as to how many of the geographic patterns of coloration that have been reported for infauna are associated with the presence of visual predation. Further investigations are suggested.  相似文献

Three bisexual Pratylenchus species, P. penetrans, P. vulnus and P. coffeae have n = 5, 6 and 7 chromosomes, respectively, and reproduce by cross-fertilization. The monosexual P. scribneri comprises two chromosomal and reproductive forms. One has n = 6 chromosomes and reproduces by meiotic parthenogenesis, the other has a somatic chromosome number of approximately 25 and reproduces by mitotic parthenogenesis. The monosexual species P. zeae, P. brachyurus and P. neglectus have somatic chromosome numbers of approximately 21 to 26, 30 to 32, and 20, respectively, and reproduce by mitotic parthenogenesis. All mitotic parthenogenetic forms probably are polyploid. The phyletic relationships of some species are discussed briefly.  相似文献
