An ellagitannin monomer, nilotinin M1 (1), and three dimers, nilotinins D1 (2), D2 (3), and D3 (4), were isolated from leaves of Tamarix nilotica (Ehrenb.) Bunge. Structures were elucidated based on analysis of spectroscopic data and chemical correlations with known compounds. In addition, six known tannins, hirtellin A (5) (dimer), remurin A (6), remurin B (7), 1,3-di-O-galloyl-4,6-O-(S)-hexahydroxydiphenoyl-β-d-glucose (8), gemin D (9), and hippomanin A (10) (monomers), were isolated for the first time from this plant species. The reported 13C NMR assignments of the dehydrodigalloyl moiety and glucose cores of 5 are revised, and the 13C NMR spectroscopic data for 6 and 7 are also reported for the first time.  相似文献

A platform for screening drugs for their ability to protect neuronal cells against cytotoxicity was developed. Nerve growth factor (NGF) differentiates PC12 cells into nerves, and these differentiated PC12 cells enter apoptosis when challenged with 6-hydroxydopamine (6-OHDA). A screening spectrophotometer was used to assay cytotoxicity in these cells; pretreatment with test samples allowed identification of compounds that protected against this neuronal cytotoxicity. The 95% ethanol extract of Phoenix hanceana Naudin var. formosana Beccari. (PH) showed potential neuroprotective activity in these assays. The PH ethanol extract was further fractionated by sequential partitioning with n-hexane, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water. Subsequent rounds of assaying resulted in the isolation of ten constituents, and their structures were characterized by various spectroscopic techniques and identified by comparison with previous data as: isoorientin (1), isovitexin (2), veronicastroside (3), luteolin-7-O-β-d-glucopyranoside (4), isoquercitrin (5), tricin-7-neohesperidoside (6), tricin-7-O-β-d-gluco-pyranoside (7), (+)-catechin (8), (−)-epicatechin (9), and orientin 7-O-β-d-glucopyranoside (10). Among these compounds, isovitexin (2), luteolin-7-O-β-d-glucopyranoside (4) and (+)-catechin (8) showed significant neuroprotective activity in cell viability (WST-8 reduction), anti-apoptosis (Annexin V-FITC/propidium iodide double-labeled flow cytometry), and cellular ROS scavenging assays (besides isovitexin (2)), as well as a decreased caspase-8 activity in 6-OHDA-induced PC12 cells. Hence, isovitexin (2), luteolin-7-O-β-d-glucopyranoside (4), and (+)-catechin (8) protected PC12 cells from 6-OHDA-induced apoptotic neurotoxicity.  相似文献

A further phytochemical investigation on the whole plants of Ypsilandra thibetica yielded one sapogenin and 12 spirostanol saponins. Five of these are new compounds, designated as ypsilandrosides C-G (26). Their structures were determined by detailed spectroscopic analysis, including extensive 1D and 2D NMR data, and by the result of a hydrolytic reaction. Compounds 25 were rare steroidal saponins that an apiofuranosyl unit was directly linked at C-3 of the aglycone. Selected spirostanol saponins (26, 9) were tested for their cytotoxic activities against K562, SPC-A-1, BGC-823, Eca-109, and AGS cell lines. Compounds 6 and 9 showed moderate inhibitory activity against all five cell lines. The antifungal properties of the new spirostanol saponins (26) against Candida albicans were also determined.  相似文献

Chemical investigations of aerial parts of Tephrosia purpurea yielded the rare prenylated flavonoids, tephropurpulin A (1) and isoglabratephrin (2), in addition to a previously identified flavonoid, glabratephrin (3). A lignan glucoside, (+)-pinoresinol 4-O-[6″-O-galloyl]-β-d-glucopyranoside (1), and two megastigmane glucosides, named macarangiosides E and F (2, 3), together with 15 known compounds (418) were isolated from leaves of Macaranga tanarius (L.) Müll.-Arg. (Euphorbiaceae). Their structures were elucidated by spectroscopic and chemical analyses. In addition, the absolute stereochemistry of macarangiosides B and C isolated previously from the same plant was also determined for the first time. Six new phenolic compounds (16), along with five known phenolics (711) were isolated from the heartwood of Dalbergia odorifera T. Chen. (Leguminosae). Their structures were determined by spectroscopic techniques including MS, UV, IR, 1D and 2D NMR. Bioassay results showed that compound 1 exhibited cytotoxicity against SGC-7901 and BEL-7402 tumor cell lines, while compounds 3, 5 and 10 showed antibacterial activities against Ralstonia solanacearum.  相似文献

Two new alkaloids (12), together with six known compounds (38), were isolated from cultures of the endophytic fungus Stagonosporopsis oculihominis. Their structures were elucidated through extensive spectroscopic methods including 2D NMR and HRMS analyses. The absolute configurations of 1 was determined by the comparison of experimental and theoretical electronic circular dichroism spectra. Phytochemical investigation of the rhizome of Dioscorea dumetorum has led to the isolation by several chromatographic steps on normal and reversed phase silica gel of a new ecdysteroid, (20R)-5β,11α,20-trihydroxyecdysone (1), and two known ecdysteroids, ajugasterone C (2) and herkesterone (3). Their structures were determined by spectroscopic methods including 1D- and 2D-NMR (COSY, TOCSY, HSQC and HMBC). This is the first report on the occurrence of phytoecdysteroids in the Dioscoreaceae family. These compounds were devoid of antifungal activity against three Candida species (Candida albicans, Candida glabrata and Candida tropicalis, MIC > 200 μg/ml).  相似文献

Phytochemical investigation of CHCl3 extract of the rhizomes of Hedychium spicatum led to the isolation of two new labdane-type diterpenes, compounds 1 and 2 along with five known compounds (37). Their structures were established on the basis of NMR (1D and 2D) and mass spectroscopic analysis. In addition, all the isolates were tested for their cytotoxicity against the Colo-205 (Colo-cancer), A-431 (skin cancer), MCF-7 (breast cancer), A-549 (lung cancer) and Chinese hamster ovary cells (CHO). Chemical investigation of the roots of Croton crassifolius led to the isolation of five pyran-2-one derivatives, including two brand new compounds (12), one new natural product (3) and two known compounds (45). Their structures and absolute configurations were established by spectroscopic analyses as well as comparison between the calculated optical rotation (OR) values with the experimental data. Interestingly, the new compound 1 showed an unusual negative chemical shift at H-11. It is well known that negative chemical shift values of 1H NMR spectrum are extremely rare in natural products. Such a negative chemical shift of 1H NMR spectrum was reproduced by density functional theory (DFT) calculations and explained by the shielding effect from the pyran-2-one ring over the hydrogen atom in the 3D conformations. Then, MTT assay was applied to evaluate the cytotoxicity of the isolated compounds (15) against two liver cancer cell lines (HepG2 and MHCC97H). The results suggested that compound 1 displayed the highest cytotoxicity with an IC50 value of 9.8 μM against HepG2 cells. Moreover, there was no obvious cytotoxicity of compounds 15 on normal liver cell line LO2. Furthermore, the mechanism of apoptosis induction in compound 1-treated HepG2 cells was investigated. The results showed that compound 1 could induce apoptosis via p53-mediated Ras/Raf/ERK suppression in HepG2 cells.  相似文献

Bioactivity-guided separation of an antihyperglycemic extract from the leaves of Dodecadenia grandiflora afforded two phenylpropanoyl esters of catechol glycosides (1 and 4) and two lignane bis(catecol glycoside)esters (2 and 3). Their structures were established on the basis of extensive spectroscopic analysis (1D and 2D-NMR, MS). Compounds 2 and 3 are believed to be derived from dimerization via the two phenylpropanoid units of 1. Compounds 1–4 showed significant antihyperglycemic activity in streptozotocin-induced (STZ) diabetic rats, which is comparable to the standard drug metformin. Our results provide support to explain the use of D. grandiflora as antihyperglycemic agent by the traditional medical practitioners.  相似文献

Six new cassane diterpenoids (1–6) and 15 known compounds were isolated and identified from the seeds of Caesalpinia sappan. The structures of the new compounds were determined based on the spectroscopic methods, including 1D and 2D NMR techniques. Compound 1 is a rare cassane diterpenoid featuring a nitrogen containing bridge between C-19 and C-20. Compounds 1 and 2 exhibited moderate cytotoxicity against HCT116, AGS, and HepG2 cell lines with IC50 values ranging from 6.36 to 27.86 μM.  相似文献

Microbial transformation of 20(R)-panaxatriol (PT) was performed by using the fungus Aspergillus niger AS 3.739. Six metabolites (16) including five new compounds were obtained. The chemical structures of these transformed products were elucidated based on various spectroscopic analyses, including 1D and 2D NMR and HRESI-MS. Metabolites 26 are new compounds. Furthermore, metabolites 2, 3, 5, and 6 exhibited significant anti-hepatic fibrosis activity, while 4 showed cytotoxicity against HSC-T6 cells.  相似文献

Phytochemical investigation of the radix of Angelica sinensis has led to the isolation and identification of a new phthalide dimer, (3Z)-(3aR,6S,3′R,8′S)-3a.8′,6.3′-diligustilide (1), along with three known phthalide dimers, including riligustilide (2), levistolide A (3), senkyunolide O (4), and three known phthalide monomers, including 3,9-dihydroxyl-ligustilide (5), (Z)-butylidene phthalide (6), (Z)-ligustilide (7). Their structures were determined by spectroscopic methods including IR, NMR (1H NMR, 13C NMR, COSY, HSQC, HMBC and NOESY) and MS. Three new flavonoids, 6,7-dimethoxy-4′-hydroxy-8-formylflavon (1), 8-formyl-4′,6,7-trimethoxyflavon (2), 4′,7-dihydroxy-8-formyl-6-methoxyflavon (3), together with fifteen known flavonoids (418) were isolated from the leaves of oriental tobacco (a variety of Nicotiana tabacum L). Their structures were determined by means of HRESIMS, extensive 1D and 2D NMR spectroscopic studies and chemical evidences. The cytotoxicity against five human tumor (NB4, A549, SHSY5Y, PC3, and MCF7) cell lines of compounds 13 were also evaluated. The results showed that compounds 1 and 3 showed high cytotoxicity against PC3 and A549 cell lines with IC50 values of 2.6 and 1.6 μM, respectively.  相似文献

Two new butanolides, licunolides A (1) and B (2), were isolated from the roots of Litsea acuminata, together with three known compounds: isolancifolide (3), longifolin (4), and sesquirose furan (5). The structures of compounds 1 and 2 were determined by spectroscopic studies (IR, MS, 1D and 2D NMR) and chemical evidence. This is the first report of 4-hydroxy-5-methylbutanolides with a C10-side chain from a natural source. Longifolin (4) and sesquirose furan (5) showed a significant cytotoxicity against HeLa cell lines in vitro.  相似文献
