稻米直链淀粉含量测定方法研讨   总被引:3,自引:0,他引:3  
通过标准碘化色法与改良碘比色法的比较认为改良碘比色法的优点是:既可测定精米粉或糙米粉等粉体材料也可测定精米粒和糙米粒等粒体材料,能将样品的直链淀粉更完全地分离出来,测试误差小,但需时间较长。相同品种不同类型样品直链淀粉的含量测定结果:精米粒>精米粒和糙米粒>糙米粉。测试精米粒或精米粉时宜选择强碱KOH,而测试糙米粒或糙米粉时宜选择强碱NaOH。样品量以100mg为宜,但不是100mg时进行品种间的比较仍可考虑采用。  相似文献

通过对多个NaOH浓度、糊化时间等不同处理参数的筛选,提用0.6-0.8mol.L^-1的冷藏液直接糊化脱糙后的精米,然后于37℃糊化21h,继而煮沸10min测定小样品直链淀粉含量的实验方法,具有前处理前便、重复间误差小、用样量少等优点,测得的直链淀粉含量经t检验,与常规法(NY147-88)差异不显著。该方法适用于分离群体或人工气候箱处理等小样品直链淀粉含量的分析测定,同时可作为普通实验室对众多育种种材料及婀交后代直链淀粉含量的快速鉴定。  相似文献

稻米直链淀粉含量的形成及其与灌浆结实期温度的关系   总被引:27,自引:1,他引:27  
通过多类型品种的田间分期番种和人工气候箱控温处理等试验,对稻米直链淀粉含量(Amylose content,简称AC)的形成动态变化特征及灌浆结实期温度的影响效应进行了分析。结果表明:水稻灌浆结实过程中稻米AC的形成动态变化过程叮嘱一次或二次曲线进行模拟,灌浆初期籽粒中的AC值很低,随之急剧上升,至20~30d左右达到最大值,然后AC的变化基本趋于稳定;稻米AC的形成变化与籽粒于物质积累过程喑存在  相似文献

杂交水稻稻谷是从F1代杂合体收获的F2代种子,由于F2代属分离群体,直链淀粉含量等稻米品质会发生分离现象.本研究选择直链淀粉含量低、中、高的5个亲本配组的5个杂交组合,按单粒分析法分析双亲亲本、F1和F2代的种子的直链淀粉含量.结果表明,F2代呈现1(低):3(中+高)孟德尔分离规律.在5个组合的单粒分析中发现分离的F2代群体呈双峰的连续分布,该现象表明高直链淀粉含量对低直链淀粉含量为不完全显性,除单一基因控制外,还受微效多基因修饰.因此杂交水稻育种工作中,尤其是优质杂交稻新组合选育时,必须注意选择双亲的直链淀粉含量基本一致,以防分离现象影响米质.  相似文献

将含单拷贝Anti-Waxy基因的纯合植株分别作为父本或母本进行正反交,获得两组含双拷贝Anti-Waxy基因的杂交后代,分析了未转基因对照、转基因亲本及两组杂交后代糙米直链淀粉含量以揭示不同Anti-Waxy基因拷贝数对降低稻米直链淀粉含量程度的影响.结果显示,2个单拷贝转基因水稻糙米直链淀粉平均含量分别为10.72%和11.13%,比对照分别降低17.98%和14.84%;两组正交和反交杂交后代糙米直链淀粉平均含量分别为8.96%和8.23%,其平均值为8.60%,比2个转基因杂交亲本糙米直链淀粉含量分别降低19.78%和22.73%,比对照降低了34.20%.结果表明,增加转基因水稻基因组中Anti-Waxy基因拷贝数在一定程度上能够进一步降低稻米直链淀粉含量,通过将独立转化获得同品种转基因植株之间杂交可以成为获得高表达转基因植物的途径.  相似文献

FT-NIRS技术应用于稻米直链淀粉含量分析研究   总被引:8,自引:0,他引:8  
运用近红外光谱快速分析技术,使用偏最小二乘法建立了近红外光谱和水稻糙米直链淀粉含量的数学模型,并进行糙米直链淀粉含量预测.结果表明糙米近红外光谱与其直链淀粉含量具有良好的相关性,决定系数r2=0.8429,最大绝对误差4.82%,平均误差2.30%.该方法在不破坏样品的前提下快速分析水稻直链淀粉含量,可用于稻种资源的快速鉴定,对于水稻优质育种及其相关研究具有重要意义.  相似文献

水稻籽粒直链淀粉含量的生态模型研究   总被引:6,自引:0,他引:6  
在中国、日本和泰国不同生态环境下进行了多品种籼稻和粳稻的种植试验,通过分析水稻籽粒直链淀粉含量与纬度、海拔、温度、太阳辐射等气候生态因子的相互关系,确立了影响水稻籽粒直链淀粉积累的主要气候因子函数,并使用权重系数进一步修订各个气候生态因子对籽粒直链淀粉的作用,构建出基于生态效应(综合气候因子函数)的水稻籽粒直链淀粉含量预测模型.利用不同年份、不同生态点和不同品种类型的试验资料对所建模型进行了检验,籼稻和粳稻籽粒直链淀粉含量的预测误差(RMSE)平均分别为0.4%和0.5%;籼稻和粳稻种植区的预测误差(RMSE)平均为0.39%和0.50%,表明模型具有较好的预测性和实用性.  相似文献

测定了48个不同马铃薯品种表观淀粉含量以及块茎中和淀粉中直链淀粉含量,对表观淀粉含量和块茎中直链淀粉含量间,表观淀粉含量和淀粉粒直链淀粉含量间进行了相关分析。结果表明:表观淀粉含量和块茎中直链淀粉含量间相关显著,表观淀粉含量和淀粉粒直链淀粉含量间相关不显著,且中熟和晚熟基因型表观淀粉含量和淀粉粒直链淀粉含量间相关也不显著,这些结论将为淀粉生物合成的理论研究和淀粉品质改良提供基本的表型数据。  相似文献

研究利用重离子辐照杂交籼稻9311创建农艺性状突变体库,使用化学方法筛选直链淀粉、蛋白质突变材料,并分析籽粒品质性状与农艺性状之间的相关性,为后续筛选直链淀粉、蛋白质突变体工作奠定基础。结果显示:在169份直链淀粉、蛋白质突变体中,直链淀粉含量变幅范围是7.64%~32.37%,其中高含量直链淀粉突变体材料有11份,低含量直链淀粉突变体材料有5份;蛋白质含量变幅范围是7.05%~13.79%,其中高蛋白突变体有34份,低蛋白突变体有3份。从169份突变体中筛选出直链淀粉、蛋白质含量都有梯度差异的突变体材料,筛选出材料的农艺性状分析结果表明:在直链淀粉、蛋白质突变体材料的农艺性状中,每穗实粒数、有效穗数、结实率和株高这四个农艺性状与籽粒品质性状间有关联,变异系数分别为37.53%、30.72%、24.70%、15.38%。相关性结果表明:直链淀粉含量和蛋白质含量呈负相关,直链淀粉含量与农艺性状呈正相关,蛋白质与农艺性状呈负相关,其中蛋白质含量和每穗实粒数、结实率呈极显著负相关,相关系数分别为-0.504、-0.592。  相似文献

分析了蜡质基因引导区的两个简单重复序列 (SSR) (CT) n 和 (AATT) n 在 74份水稻材料中的多态性及其与直链淀粉含量 (AC)的关系。这些材料包括了籼稻 (OryzasativaL .ssp .indica)、粳稻 (O .sativassp .japonica)和普通野生稻 (O .rufipogon) ,其AC值覆盖了栽培稻AC分布的整个范围。以 (CT) n 作标记检测到 8个等位基因 ,粳稻品种趋于含有重复数目较多 (n≥ 16 )的等位基因 ,重复次数较少 (n≤ 14)的等位基因只出现在籼稻中。 (AATT)n检测到 2个等位基因 ,野生稻中少数植株表现出杂合性。分析表明AC与这两个SSR序列基因型高度相关 ,高AC (>2 2 .0 % )品种具有 (CT)重复次数较少 (n≤ 14)的等位基因 ;相反 ,除了糯米外 ,所有低或者中等AC的品种都有 (CT)重复数较多 (n≥ 16 )的等位基因。具有重复次数较多的 (AATT) 6等位基因的品种多为高AC ,具有重复次数较少的(AATT) 5等位基因的品种多为低或中等AC。不同SSR基因型品种间AC差异极显著。虽然目前还不能确定这两个SSR序列在直链淀粉合成中的直接功能 ,SSR变异与AC间近乎完全的相关性可作为分子标记直接用于水稻的品质改良。  相似文献

The waxy gene encodes a granule-bound starch synthase. A 1.0-kb portion of the sequence of the rice waxy gene, which includes the region between exon 4 and exon 9, was inserted in an antisense orientation between the 35 S promoter and the GUS gene of pBI221. The resultant plasmid, pWXA23, was introduced into rice protoplasts by electroporation. GUS activity was clearly detected in derived callus lines, suggesting that the antisense component of the fusion gene was also expressed. Transgenic rice plants were regenerated from these callus lines and their GUS activity was confirmed. Some of the rice seeds from these transformants showed a significant reduction in the amylose content of grain starch, even though they had become polyploid. These results suggest that even when intron sequences are included, antisense constructs can bring about a reduced level of expression of a target gene. The utility of GUS, included as a reporter gene, for the simple detection of expression of an antisense gene, was apparent from these results.  相似文献

Most of the nonstarch lipids in brown rice (Oryza sativa) of three rices differing in amylose content were contributed by bran, germ, polish and subaleurone layer. Nonstarch lipids consisted of 82–91% neutral lipids (of which 73–82% were triglycerides), 7–10% phospholipids and 2–8% glycolipids. Linoleic, oleic and palmitic acids were the major fatty acids. Nonwaxy (24 and 29% amylose) milled rice had proportionally more starch lipids and less nonstarch lipids than waxy (2% amylose) milled rice. Starch lipids were mainly lysophosphatidyl choline, lysophosphatidyl ethanolamine and free fatty acids. The major fatty acids were palmitic and linoleic acids, followed by oleic acid.  相似文献

Summary Inheritance of waxy locus was studied in crosses of a waxy variety with four non-waxy parents having high-, intermediate-, low- or very low-amylose content. The analysis for amylose content was done on a single grain basis in parents, F1, F2, B1F1, and B2F1 seeds. The waxy parent lacking synthesis of amylose content was found to differ from the ones having high-, intermediate-, low- or very low-amylose content by one gene with major effect. Dosage effects for amylose content were observed to have great influence on segregation pattern and efficiency of selection. Selection efficiency for amylose content can be enhanced by selecting for endosperm appearance in early segregating generations.  相似文献

 The inheritance of grain quality is more complicated than that of other agronomic traits in cereals due to epistasis, maternal and cytoplasmic effects, and the triploid nature of endosperm. In the present study, an established rice DH population derived from anther culture of an indica/japonica hybrid was used for genetic analysis of rice grain quality. A total of five parameters, amylose content (AC), alkali-spreading score (ASS), gel consistency (GC), percentage of grain with a white core (PGWC) and the square of the white core (SWC), were estimated for the DH lines and the parent varieties. For each parent, the value of each parameter was relatively stable in three locations, Beijing, Hangzhou and Chengdu, while the differences between the parents were significant for all five parameters. AC showed a bimodal distribution, and the distribution of ASS was skewed toward the value of JX17, while the other three parameters displayed continuous distributions among the DH lines with partially transgressive segregations. For AC, a minor and a major gene were found on chromosomes 5 and 6 respectively. The major gene, which should be an allele of wx, explained 91.9% of the total variation. For GC, two QTLs were identified on chromosomes 2 and 7 respectively. For ASS, a minor and a major gene were both located on chromosome 6. The major gene should be the same locus as the alkali degeneration gene (alk). Genetic linkage between alk and wx was found in QTL mapping. For PGWC, two QTLs were located on chromosomes 8 and 12. Only a minor QTL was found for SWC on chromosome 3. The results and the molecular markers presented here may be useful in rice breeding for grain quality improvement. (广东省农业科学院水稻研究所,广州 510640) 摘要 为了进一步完善广东地方稻种资源的鉴定评价,并为该资源在水稻品质育种中的有效利用提供科学依据,我们以国家作物种质库编目的2974份广东地方稻种为研究对象,其9个品质性状数据为基础,研究了广东地方稻种品质性状的区域分布特点,并对总体和不同稻作区品质性状多样性进行了比较分析.同时对部分生态因子与品质性状的相关性进行了初探。结果显示,广东地方稻种资源品质性状具有丰富的多样性。其糙、精米率较高,蛋白质含量丰富多样,胶稠度的变异幅度大,直链淀粉含量适中达国标的材料偏少;中南稻作区为广东优质地方稻种的主要集中地;不同稻作区的品质性状多样性表现各异;直链淀粉含量、碱消值及胶稠度是影响广东地方稻种品质形成的主要指标;湿度和温度的地域差异是广东地方稻种稻米品质多样性的主要成因。本研究结果充分说明,广东地方稻种中具有优异品质性状资源的利用对改良现有品质的稻米品质和拓宽遗传基础具有十分重要的意义。  相似文献

The cooking and eating quality of the rice grain is one of the most serious problems in many rice-producing areas of the world. In this study, we conducted a molecular marker-based genetic analysis of three traits, amylose content (AC), gel consistency (GC) and gelatinization temperature (GT), that are the most important constituents of the cooking and eating quality of rice grains. The materials used in the analysis included F2 seeds, an F2:3 population, and an F9 recombinant inbred-line population from a cross between the parents of ’Shanyou 63’, the most widely grown hybrid in rice production in China. Segregation analyses of these three generations showed that each of the three traits was controlled by a single Mendelian locus. Molecular marker-based QTL (quantitative trait locus) analyses, both by one-way analysis of variance using single marker genotypes and by whole-genome scanning with MAPMAKER/QTL, revealed a single locus that controls the expression of all three traits. This locus coincided with the Wx region on the short arm of chromosome 6, indicating that all three traits were either controlled by the Wx locus or by a genomic region tightly linked to this locus. This finding has provided clues to resolving the molecular bases of GC and GT in future studies. The results also have direct implications for the quality improvement of rice varieties.
