Serum and milk Immunoglobulin M (IgM) concentrations in 11 mother-pup pairs were measured in southern elephant seals (Mirounga leonina) throughout lactation during 2 breeding seasons at King George Island. Samples were obtained sequentially throughout the suckling period (approximately 23 days). The IgM concentration was measured by single radial immunodiffusion on agarose plates. Milk IgM concentrations showed significant differences throughout lactation, with the highest concentrations on the 1st day (x=989.7 mg/dL skimmed milk; SD=433.2) followed by a sharp fall during the next 3–6 days of the suckling period. The ratio of milk IgM/serum IgM concentrations from mothers ranged from 0.21 to 21.92, with highest values in the 1st day of lactation (x=8.25, SD=5.4) and a decrease in concentration as lactation progressed. This was due to the fact that, throughout lactation, milk IgM concentrations fell while serum IgM values showed an increasing trend. Pups showed the lowest serum IgM values in the 1st day of the suckling period (x=13.0 mg/dL, SD=4.3) with an increasing trend as lactation progressed. Despite the high IgM concentrations of milk at day 1 of lactation, by 1 week of age pups had serum IgM concentrations only slightly greater than at birth. This suggests that much of this Ig was newly formed and little or no milk IgM was absorbed during the 1st week. Possibly, the function of ingested IgM is to provide local immunity in the pup's gut, during the first few days of postnatal life. Accepted: 26 March 2000  相似文献

We quantified immunoglobulins (Ig) in mammary secretions of 12 female southern elephant seals ( Mirounga leonina) at King George Island, Antarctica, using single radial immunodiffusion on agarose plates. Seals were chemically immobilized for milk sample collection at four time points during the 3- to 4-week suckling period. All three major mammalian immunoglobulins (IgG, IgM, IgA) were detected in southern-elephant seal milk. Total immunoglobulin levels and the ratio of Ig subclasses varied throughout the suckling period. Total immunoglobulin levels were highest on the 1st day of lactation, when they represented over 57% of the mean total protein concentration of the milk, and declined steadily throughout lactation, representing approximately 40% of the mean total protein concentration at the end of the suckling period. IgG was the most abundant immunoglobulin class (85.93-93.78% of total milk immunoglobulins), followed by IgM (6.06-13.93%), and IgA (0.14-0.23%). There was no significant difference in IgA levels throughout the suckling period. IgG levels were significantly lower during the second stage (3-6 days post-parturition) than during the first, third or fourth stages (1, 13-15, and 19-25 days post-parturition, respectively). IgM levels were highest during the first stage of lactation; these values were significantly higher than levels measured during the second, third and fourth stages of lactation. Transfer of passive immunity from female to offspring in other mammalian species is correlated with the subclass of immunoglobulin secreted in the milk; species acquiring passive immunity in utero, via the placenta, secrete a preponderance of IgA, whereas species acquiring immunity post-partum, via lacteal secretions and gut resorption, secrete a preponderance of IgG. The Ig patterns and concentrations observed in our study of southern elephant seals are consistent with an important role of post-partum transmission of passive immunity during the pinniped lactation period.  相似文献

1. The neuraminidase activity of homogenates of the mucosa of the middle and distal thirds of the small intestine of rats increased about 5-fold between birth and 4 to 8 days of age, and then gradually declined to the much lower adult activity by 24 days. No comparable changes occurred in the proximal third. 2. In 8-day-old rats, the neuraminidase activity of the middle and distal thirds of the small intestine was about 10 times greater than that of the proximal third, 20 times greater than that of the colon and at least 100 times greater than that of the liver, brain, gastric mucosa or pancreas. 3. In all other species investigated (mice, rabbits, cats and guinea pigs), the neuraminidase activity of the middle and distal thirds of the small intestine was greater in suckling animals than in adults. 4. The sialic acid content of rat milk increased about 2-fold between birth and 8 days post partum and then declined. 5. There was a highly significant positive correlation between the intestinal neuraminidase activity of suckling animals of various species and ages and the sialic acid content of milk obtained from the corresponding species and stage of lactation. 6. It is suggested that the intestinal neuraminidase of suckling mammals functions primarily to remove sialic acid from various components of milk, thus providing sialic acid for the synthesis of sialoglycoproteins and gangliosides by the young.  相似文献

In a woman suckling twins it became apparent that both suckling-induced and precisely timed, spontaneous bursts of milk ejection were occurring. Observations on days 14, 28, 56, and 112 of lactation disclosed highly significnat increases in intervals between episodes of spontaneous milk ejection. Furthermore, at all stages of lactation the interval between a feed and the next episode of spontaneous ejection was significantly longer than the interval between spontaneous ejections. The decrease in frequency of episodes of spontaneous milk ejection during lactation may be related to the decreasing release of prolactin in response to suckling. Spontaneous milk-ejection episodes are felt only when the breast is full and may signal its readiness for a further suckling episode. Such bursts of milk ejection may stimulate the suckling response in babies, suggesting that rigid three- or four-hour feeding regimens may be unphysiological and pose a threat to the success of breast-feeding in the early postnatal period.  相似文献

Polyamines play a critical role in the development of intestinal and immune systems during the infant breastfeeding period, but the effect of polyamines on the microbiota has not been reported. The aim of our study was to characterize the impact on the colonization pattern in neonatal BALB/cOlaHsd mice after supplementing an infant formula (IF) with a mixture of putrescine (PUT), spermidine (SPD) and spermine (SPM). A total of 48 pups (14 days old) were randomly assigned to 4-day intervention groups as follows: breast-fed (unweaned) pups (n=12); weaned pups (n=12) fed an infant formula (IF); weaned pups (n=12) fed an IF enriched with a low concentration of PUT, SPD and SPM (2.10, 22.05 and 38.00 μg/day, respectively); and weaned pups (n=12) fed with IF enriched with a high concentration of PUT, SPD and SPM (8.40, 88.20 and 152.00 μg/day, respectively) of polyamines in accordance with normal proportions found in human milk. Microbiota composition was analyzed by fluorescent in situ hybridization (FISH) with flow cytometry detection. Microbiota changes in formula-fed mice were significantly greater following supplementation with polyamines (P<.01). Bifidobacterium group bacteria, Akkermansia-like bacteria and LactobacillusEnterococcus group levels were higher in the groups fed infant formula supplemented with polyamines, resulting in even higher numbers of bacteria than in the breastfed pups. Our findings indicate that infant formulas enriched with polyamines may interact with gut microbiota, suggesting that further studies in human infants are required to assess the impact of polyamines on both growth and microbiota levels.  相似文献

Effects of a high bromide intake in lactating rats on the performance of the dams and on the prosperity of their young were studied. In the dams, two marked consequences undoubtedly caused by high bromide intake were observed: stagnation in the extent of diet and water consumption in the course of the lactation period, and a conspicuous drop in the production rate of mother's milk. A very high intake of bromide in the mothers in the course of the nursing period (about 220 mg Br/d per dam) also caused a marked decrease in the body weight increments in their suckling young. Only about one-half of these young survived and their general condition was very poor. It is suggested that one of the possible reasons for the observed marked decrease in the production of mother's milk in dams with high bromide intake could be a decreased stimulation of the mammary glands as a consequence of reduced consumption of mother's milk by the suckling. Bromide ions ingested by the dams easily moved into the rat milk. Via mother's milk, bromide was transferred in a large extent to the suckling. The amount of bromide in mother's milk depended on the bromide concentration in the drinking water taken by the dams. With the addition of 5 g bromide per liter (providing the mean daily bromide dose of 220 mg), bromide ions replaced about 54% of the chloride in the milk. A rise in the concentration of both halogens caused also an increase in the concentration of sodium in mother's milk. The exact mechanism(s) of bromide interference with postnatal developmental processes in the young remain(s) unclear. This article discusses the consequences of different suckling systems in the industrial countries for the milk production, udder health, reproduction and behaviour of high producing dairy cows and the effects on the gain, health and behaviour of the calves. The suckling systems are divided into three different categories depending on the purpose and duration of the suckling period. Long-term suckling with or without additional milking covering the period where the calf has a nutritional need for milk, and short-term suckling, where cow and calf are kept together in the colostrum period only.Long-term suckling without additional milking in early lactation can in some situations stimulate the subsequent milk production to a greater extent than milking alone. No clear or significant differences can be found between restricted and free suckling systems. Most experiments show that suckling decreases the risk of mastitis in the suckling period and in some cases even for some time after the suckling has been terminated. Suckling and milking during the same period is not advantageous in production turns because of a very poor ejection of milk. Long-term suckling can increase the post-partum interval until first heat, in some cases until the end of the suckling period. However, as the cows appear to be more fertile, the net effect on reproduction is small. The suckled calves are usually healthy with a high daily gain. Short-term suckling have more advantages than disadvantages on production, health and behaviour of both the cow and the calf compared to an immediate separation after birth.  相似文献

Polyamines, putrescine (PUT), spermidine (SPD), spermine (SPM), and agmatine (AGM), are polycationic amines related to multiple cell functions found in high concentrations during the development of hypothalamus and pituitary. In previous works, we demonstrated that alpha-difluoromethylornithine (DFMO), an inhibitor of polyamines biosynthesis, induced a delay in puberty of female rats, accompanied by high, sustained follicle-stimulating hormone (FSH) levels during the infantile period. Also, DFMO treatment induced changes in polyamine concentration both in hypothalamus and pituitary of rats, mainly a decrease of PUT and SPD, an increase in SPM, and no change in AGM. In the present work, we investigated the direct effects of polyamines on the secretion of hypothalamic GnRH and pituitary gonadotropins in 6- and 15-day-old female rats. In 6-day-old animals, in vitro incubations with PUT, SPD, and AGM of hypothalami or anterior pituitaries were able to inhibit GnRH, FSH, and leutinizing hormone (LH) secretion, respectively. SPM showed a nonspecific transient inhibitory effect on FSH. When challenged with either high K(+) (hypothami) or GnRH (pituitaries), the tissues incubated in the presence of polyamines showed no differences when compared with their controls. No effects of polyamines in 15-day-old rats in either tissue were observed. Pituitary cell cultures of 6-day-old animals incubated with DFMO for 4 days showed a significant increase in FSH, but not in LH. We conclude that high PUT, SPD, and AGM levels during the first 10 days of life are important for the development of the hypothalamic-hypophyseal unit, probably related to an inhibitory effect on GnRH and gonadotropins. Therefore, polyamine participation, especially PUT and SPD, is of importance in the regulation of GnRH and gonadotropin secretion in the neonatal and infantile periods, critical stages in the establishment of sexual differentiation.  相似文献

Unlike most mammals, hooded seal (Cystophora cristata) pups are born with a substantial layer of adipose tissue. Subsequently, during the brief lactation period of only 4 days, fasting mothers mobilize enormous amounts of lipid from blubber and secrete milk (60% fat) at rates of 10 kg·day-1. Pups gain 7 kg·day-1 due primarily to the deposition of fat in blubber. We measured blubber content and fatty acid composition of blubber and milk in hooded seal mother-pup pairs at birth and over the 4-day lactation period to examine the nature and source of fetal lipids, the incorporation of maternal blubber fatty acids into milk lipid, and patterns of fatty acid deposition in suckling young. The fatty acid composition of the blubber of the newborn was notably different from that of its mother. Fetal deposition was likely due to a combination of both fetal synthesis and direct placental transfer of maternal circulating fatty acids. The blubber of the newborn was characterized by high levels (>90% of total fatty acids) of saturated and monounsaturated fatty acids of primarily endogenous origin. In particular, the fetus appeared to have high Δ-9 desaturase activity as evidenced by the large amounts of 14:1n-5 (4.2%) and 16:1n-7 (37.0%) in newborn blubber compared to maternal blubber (0.2% and 14.1%, respectively). Nevertheless, essential and long-chain polyunsaturated fatty acids of the n-3 and n-6 families, which could only have originated by direct transfer from the mother, comprised>7% of pup blubber fatty acids and indicated greater rates of placental transfer than found in humans. In hooded seal mothers, rapid lipid transfer during the brief lactation period appeared to be facilitated by direct incorporation of mobilized fatty acids into milk. Although some differences in proportions of specific fatty acids were found between milk and maternal blubber, most of these differences declined over the course of lactation. However, selective mobilization of 20:5n-3 from maternal blubber into milk was apparent throughout lactation and resulted in elevated levels in pup blubber at weaning compared to maternal blubber. Ingested fatty acids were deposited directly and without modification into the blubber of pups, and by 4 days the fatty acid composition of pup blubber was virtually identical to that of the milk consumed.  相似文献

ABSTRACT

The aim of the study was to investigate if dietary alpha-ketoglutarate (AKG) supplementation may improve the performance of lactating sows and their suckling piglets. After farrowing, 24 lactating sows (Large White × Landrace) with similar body weight (BW) were assigned to the control and AKG groups based on parity, and their lactation diets were supplemented with 0.00 or 0.25% AKG, respectively. It was found that supplementing the diet of lactating sows with 0.25% AKG enhanced growth performance of the suckling piglets from d 7 to d 21 of the lactation period, improved villus height of ileum and tended (p = 0.085) to increase mean volumetric bone mineral density of femur in the weanling piglets. In the lactating sows, dietary supplementation of AKG decreased plasma urea level on d 14 of lactation, decreased plasma calcium (Ca) concentrations from d 7 to d 21 of lactation and increased lactose and Ca levels in ordinary milk. Thus, it was proposed that AKG supplementation stimulates the capacity for lactose synthesis and Ca uptake in the mammary gland, thereby altering the composition of the ordinary milk which might be associated with the enhanced performance of piglets during the suckling period. These findings could lead to a better application of AKG in lactating nutrition, and therefore, promoting pork production.  相似文献

An understanding of the mechanisms regulating milk yield in sows is crucial for producers to make the best management decisions during lactation. Suckling of mammary glands by piglets is one factor that is essential for development of these glands during lactation and for the maintenance of lactation in sows. The process of mammary development is not static as the majority of it takes place in the last third of gestation, continues during lactation, is followed by involution at weaning and starts over again in the next gestation. During involution, the mammary glands undergo a rapid and drastic regression in parenchymal tissue, and this can also occur during lactation if a gland is not suckled regularly. Indeed, the pattern of regression is similar for glands that involute at weaning or during lactation. Suckling during 12 to 14 h postpartum is insufficient to maintain lactation and the process of involution that occurs in early lactation is reversible within 1 day of farrowing but is irreversible if a gland is not used for 3 days. However, milk yield from a gland which is ‘rescued’ within the first 24 h remains lower throughout lactation. Suckling does not only affect milk yield in the ongoing lactation, but it also seems to affect that of the next lactation. Indeed, non-suckling of a mammary gland in first-parity sows decreased development and milk yield of that gland in second parity. Nursing behaviour of piglets in early lactation was also affected, where changes were indicative of piglets in second parity being hungrier when suckling glands that were not previously used. It is not known, however, if the same effects would be seen between the second and third lactation. Furthermore, the minimum suckling period required to ensure maximal milk yield from a gland in the next lactation is not known. This review provides an update on our current knowledge of the importance of suckling for mammary development and milk yield in swine.  相似文献

Milk yield was measured by a tritiated water dilution procedure during consecutive lactations in mice suckling four, 10 or 18 young. Analysis of variance revealed positive effects of lactation number and litter size on milk yield. There was a significant correlation between maternal body weight and parity; increased body weight accounted for some, but not all, of the parity-related increases in milk yield. Peak milk yield was reached between days 10 and 16 of lactation, but the efficiency with which the growing young utilized milk for weight gain was greatest before day 7. The aim of the present study was to investigate how suckling influences the plasma levels of oxytocin and vasoactive intestinal peptide in lactating dogs. Blood-samples were drawn from ten lactating beagles during suckling, in week one and week three of the five week long lactation period and the levels of vasoactive intestinal peptide and oxytocin were determined by radioimmunoassay. The immunoreactive levels of vasoactive intestinal peptide and oxytocin (below referred to VIP and oxytocin) increased rapidly in response to the suckling stimulus. The rise of both peptides was significant at both suckling experiments. The origin and role of suckling-released oxytocin is well established. The origin of and function of the VIP released in response to suckling remains unknown.  相似文献

A total of 36 mink dams and their litters of 3, 6 or 9 kits were used for determination of milk intake of the suckling young by means of deuterium dilution technique, and chemical composition of milk and of kit bodies. Measurements were performed during lactation weeks 1-4, each week with 3 dams with each litter size. Milk intake was determined over a 48 h measurement period, and by the end of this milk samples were collected and 2 kits (litters of 6 and 9) or 1 kit per litter (litters of 3) were killed for body chemical composition. Based on the results, different models were applied for calculation of the energetic efficiency of milk. Dam milk yield increased steadily from week 1 until week 3 but only slightly from week 3 to 4. The increase declined with increasing litter size, and for dams suckling 9 kits the increment from week 3 to week 4 was only 2 g. The dry matter content of milk increased significantly as lactation progressed, being reflected in crude protein increasing from 6.9% in lactation week 1 to 8.1% in week 4. Milk fat increased concomitantly from 5.6% to 8.0%. In kit bodies, crude protein content increased from 9.4% in week 1 to about 12% in weeks 3 and 4. Body fat content increased from week 1 (4.1%) to week 3 (8.4%) and then declined in week 4 (7.1%). Animals suckled in litters of 3 kits had the highest milk intake and live weight and kits suckled in litters of 9 had the lowest milk intake, live weight and daily gain. In terms of milk intake per g gain kits in litters of 6 were the most efficient, with 4.1 g milk per g body gain. In rodents, forced activation of hepatic peroxisome proliferator-activated receptor α (PPARα) by administration of exogenous PPARα activators during lactation leads to a reduction of milk triacylglycerol (TAG) production. Herein, we investigated whether a negative energy balance (NEB) induced by feed restriction (about 18% lower feed and energy intake) during lactation by increasing the release of fatty acids, which act as PPARα agonists, causes a disruption of hepatic lipid metabolism and thereby impairs milk TAG production in sows. Nutrient and energy content of the milk on day 20 of lactation and gains of litters during the first 14 d and the whole 21 d suckling period did not differ between Control and feed-restricted sows. The mRNA concentrations of several sterol regulatory element-binding protein target genes involved in lipid synthesis in the liver and the plasma concentration of TAG were reduced in the feed-restricted sows, whereas the mRNA concentrations of PPARα target genes involved in fatty acid oxidation in liver and skeletal muscle were not different between groups. In conclusion, it was shown that an NEB during lactation does not adversely affect milk composition and gains of litters, despite inhibiting hepatic expression of genes involved in lipid synthesis and reducing plasma TAG concentration. The finding that PPARα target genes involved in fatty acid utilisation in liver and muscle of sows are not induced by the NEB during lactation may explain that fatty acid availability in the mammary gland is sufficient to maintain milk TAG production and to allow normal litter gain.  相似文献

Summary The breeding biology of Weddell seals (Leptonychotes weddellii) was studied at Drescher Inlet (Antarctica) during the austral late winter and spring 1986. The whelping season was estimated to last about 40 days, with a peak in pupping at late October. No post natal mortality was observed during the whole study. The attachment between parous females and their pups was strong during the first five weeks after parturition. From the third week on mothers spent more time in the water than the pups. Data on daily peak haul-out time of pups, post natal presence of pups on the ice and distribution of births during the whelping season, indicate that censuses should be carried out between 12.00 hours and 16.00 hours local time and during three weeks after the peak of the pupping season. Pups weaned between 6 and 7 weeks of age. Hormone concentrations (oestradiol-17 and progesterone) indicated that females did not come into oestrus during lactation. Throughout the whole lactation period the mother dominates the relationship with her pup. Growth of the pups was linear during the first 5 weeks post partum, during which period birth weight (x=29.1kg) increased more than 3 1/2 times. Until the 6th week of age the pups gained on average 2.6 kg/day, the respective weight loss in cows was 5.8 kg/day. The latter represents an average loss of nearly 38% of the initial weight at parturition. During the course of lactation, suckling frequency decreased, whereas the length of each bout increased resulting in an approximately constant total suckling duration per week. Of the energy used by females during the first 5 weeks, 52% was consumed by pups. Pups only took milk and the cows did not feed at all during the whole period of observation. Therefore the realized growth in pups, 10 g/min of suckling, was totally derived from energy stored by the cows.  相似文献

Fatty acid composition of the major lipid classes in stomach contents of suckling rats at 1, 5, 10, 17 and 20 days of lactation was compared to that of milk lipids. In milk, 98% of fatty acids were in triacylglycerols at all lactation times. Medium-chain fatty acid concentrations increased from 8% in colostrum to 26% at day 5. Fatty acid composition of stomach acylglycerols at all lactation times was different from that of milk triacylglycerols, containing less medium-chain fatty acids, 8:0 and 10:0. This preferential hydrolysis was also shown by higher concentrations of medium-chain fatty acids in the free fatty acid fraction. The lipolysis of medium-chain fatty acids from triacylglycerols resulted in the appearance of di- and monoacylglycerols with 50-100% higher amounts of 14:0 and 16:0. The similar fatty acid composition of products suggests that considerable lipolysis occurred in stomachs of suckling rats even at 1 day of age. Although there was a 10-fold increase in milk consumption, the extent of lipolysis was similar throughout the suckling period because of a parallel rise in lingual lipase levels.  相似文献

Recent studies have shown that L-carnitine supplementation of sows increases growth of their piglets during the suckling period. In this study, the composition of the milk of sows supplemented with L-carnitine was determined to find out whether an altered milk composition could account for the increased growth rates of the piglets. Milk of 13 control sows and 14 sows supplemented with L-carnitine (125 mg/d during pregnancy, 250 mg/d during lactation) was collected 5-8 h after birth (colostrum) and on days 10 and 20 of lactation. Concentrations of fat and lactose and the energy content in milk at day 10 and 20 did not differ between both groups of sows. Sows supplemented with L-carnitine had a higher concentration of protein in colostrum (p < 0.05) while concentrations of fat, lactose, immunoglobulins G, M and A as well as the energy content in colostrum did not differ between both groups of sows. These findings show that milk composition does not play a major role for the increased postnatal growth of piglets from sows supplemented with L-carnitine observed in recent studies.  相似文献

In two experiments, we compared milk intake (assessed by weighing calves before and after suckling) and milk production (by hand milking hinds) in Iberian red deer both when calves sucked their mothers together (group-suckling experiment) and when mother and offspring were isolated (isolation-suckling experiment). In both experiments, the general lactation curve for calves increased to a peak and then decreased (type I, standard lactation curve in mammals), whereas the curve for hinds decreased from the start (type II). However, in the experiment on group suckling, calves ingested 17.2% more milk than that produced by their mothers from weeks 6 to 20. In both isolation- and group-suckling experiments, hinds showed an overproduction of milk decreasing from weeks 1 to 5. This decreasing overproduction coincides with a similar trend in calf mortality reported in the literature and might thus be aimed at ensuring calves have sufficient nutrients when mortality is highest. In addition, allosuckling observations in the group-suckling experiment showed an inverse relationship between milk production and percentage of allosucking attempts. Allosucking attempts were also more frequent after the milk overproduction period. Both findings suggest that allosuckling is a response to compensate for a reduced maternal milk supply.
