A new isoalloxazine alkaloid, named bupleurine A ( 1 ), along with five known compounds ( 2 – 6 ), were isolated from the aerial parts of Bupleurum chinense DC. The structure elucidation of the new alkaloid ( 1 ) was employed by combining NMR and HR‐MS data with comparison of reference in the literature. Five known compounds ( 2 – 6 ) were isolated from Bupleurum genus for the first time. Additionally, their antiproliferative activities on HeLa cells were evaluated by MTT assay and IC50 of compounds 1 and 4 – 6 were below 10 μm after treatment for 24 h.  相似文献

Five new phenyl dihydroisocoumarin glycosides ( 1 – 5 ) and two known compounds ( 6 – 7 ) were identified from the butanol fraction of Scorzonera longiana. The structures of 1 – 7 were elucidated based on spectroscopic methods. Antimicrobial, antitubercular, and antifungal evaluation of compounds 1 – 7 were carried out using the microdilution method against nine microorganisms. Compound 1 was active only against Mycobacterium smegmatis (Ms) with a MIC value of 14.84 μg/mL. All tested compounds ( 1 – 7 ) were active against Ms but only compounds 3–7 were active against fungi (C. albicans, S. cerevisiae) with MIC values of 25.0–125 μg/mL. In addition, molecular docking studies were conducted against Ms DprE1 (PDB ID: 4F4Q), Mycobacterium tuberculosis (Mbt) DprE1 (PDB ID: 6HEZ), and arabinosyltransferase C (EmbC, PDB ID: 7BVE) enzymes. Compounds 2 , 5 , and 7 are the most effective Ms 4F4Q inhibitors. Compound 4 was the most promising inhibitory activity on Mbt DprE with the lowest binding energy of −9,9 kcal/mol.  相似文献

In this study, a new phthalonitrile derivative namely 4-[(2,4-difluorophenyl)ethynyl]phthalonitrile ( 1 ) and its metal phthalocyanines ( 2 and 3 ) were synthesized. The resultant compounds were conjugated to silver nanoparticles and characterized using transmission electron microscopy (TEM) images. The biological properties of compounds ( 1 – 3 ), their nanoconjugates ( 4 – 6 ), and silver nanoparticles ( 7 ) were examined for the first time in this study. The antioxidant activities of biological candidates ( 1 – 7 ) were studied by applying the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. The highest antioxidant activity was obtained 97.47 % for 200 mg/L manganese phthalocyanine-silver nanoconjugates ( 6 ). The antimicrobial and antimicrobial photodynamic therapy (APDT) activities of biological candidates ( 1 – 7 ) were examined using a micro-dilution assay. The highest MIC value was obtained 8 mg/L for nanoconjugate 6 against E. hirae. The studied compounds and their silver nanoconjugates exhibited high APDT activities against all the studied microorganisms. The most effective APDT activities were obtained 4 mg/L for nanoconjugates ( 5 and 6 ) against L. pneumophila and E. hirae, respectively. All the studied biological candidates displayed high cell viability inhibition activities against E. coli cell growth. The biofilm inhibition activities of the tested biological candidates were also investigated against S. aureus and P. Aeruginosa. Biological candidates ( 1 – 6 ) can be considered efficient metal nanoparticle-based materials for multi-disciplinary biological applications.  相似文献

Six new nonactic and homononactic acid derivatives, ethyl homononactate ( 1 ), ethyl nonactate ( 2 ), homononactyl homononactate ( 6 ), ethyl homononactyl nonactate ( 7 ), ethyl homononactyl homononactate ( 8 ), and ethyl nonactyl nonactate ( 9 ), as well as four known compounds, homononactic acid ( 3 ), nonactic acid ( 4 ), homononactyl nonactate ( 5 ), and bishomononactic acid ( 10 ), were isolated from culture broth of Bacillus pumilus derived from Breynia fruticosa. The structures of new compounds were elucidated by spectroscopic analysis and chemical methods. The optical purities of 1 – 6 were determined by HPLC/MS after treatment with L ‐phenylalanine methyl ester. The dimeric compounds 5 – 9 showed weak cytotoxic activities against five human cancer cell lines (IC50 19–100 μg/ml).  相似文献

One new fawcettimine-type alkaloid ( 1 ), one new miscellaneous-type alkaloid ( 2 ), four new lycodine-type alkaloids ( 3 – 6 ), and eight known ones ( 7 – 14 ) were isolated from the whole plants of Huperzia serrata. Their structures and absolute configurations were elucidated based on spectroscopic data, X-ray diffraction, ECD calculation and Mosher's method. Compound 1 was a rare C18N2-type Lycopodium alkaloid, possessing serratinine skeleton with an amide side chain in C-5. The absolute configuration of the 18-OH of compounds 4 – 6 were first determined by Mosher's method. Moreover, compounds 1 – 14 were assayed anti-acetylcholinesterase effect in vitro, and compound 7 showed significant anti-acetylcholinesterase activity with an IC50 value of 16.18±1.64 μM.  相似文献

New naphthalene derivatives ( 1 and 2 ) and a new isomer ( 3 ) of ventilagolin, together with known anthraquinones, chrysophanol ( 4 ), physcion or emodin 3‐methyl ether ( 5 ), and emodin ( 6 ), were isolated from vines of Ventilago denticulata. The isolated compounds exhibited cytotoxic activity with IC50 values of 1.15 – 40.54 μg/ml. Compounds 1 – 3 selectively exhibited weak antibacterial activity (MIC values of 200.0 – 400.0 μg/ml), while emodin ( 6 ) displayed moderate antibacterial activity with MIC value of 25.0 μg/ml. The isolated compounds showed nitric oxide and DPPH radical scavenging activities. Compounds 1 – 3 and 6 exhibited weak xanthine oxidase inhibitory activity, while emodin ( 6 ) acted as an aromatase inhibitor with the IC50 value of 10.1 μm . Compounds 1 and 2 exhibited phosphodiesterase 5 inhibitory activity with IC50 values of 8.28 μm and 6.48 μm , respectively.  相似文献

Four new ( 1 – 4 ) and one known ( 5 ) acylated iridoid glycosides were isolated from the aerial parts of Veronicastrum sibiricum (L.) Pennell . The chemical structures of the isolated compounds were determined to be 3″,4″‐dicinnamoyl‐6‐O‐rhamnopyranosyl‐10‐O‐bergaptol‐5,7‐bisdeoxycynanchoside ( 1 ), 3″,4″‐dicinnamoyl‐6‐O‐rhamnopyranosylpaulownioside ( 2 ), 2″,4″‐dicinnamoyl‐6‐O‐rhamnopyranosylcatalpol ( 3 ), 3″,4″‐dicinnamoyl‐6‐O‐rhamnopyranosylaucubin ( 4 ), and 3″,4″‐dicinnamoyl‐6‐O‐rhamnopyranosylcatalpol ( 5 ) using spectroscopic techniques. Among these compounds, compound 5 increased antioxidant response element (ARE) luciferase activity.  相似文献

In the present study, five known γ-lactones (majoranolide B – 1 , majorenolide – 2 , majorynolide – 3 , lincomolide D – 4 , and isolinderanolide E – 5 ), as well as a new one (perseanolide – 6 ), were isolated from Persea fulva and P. americana. All isolated compounds exhibited potential activity against trypomastigote forms of Trypanosoma cruzi, whereas compounds 2 (EC50 of 4.8 μM) and 6 (EC50 of 3.6 μM) displayed superior activity than the positive control benznidazole (EC50 of 16.4 μM), with selectivity index (SI) values of 17.8 and >55.6, respectively (benznidazole, SI>12.2). Molecular docking studies were performed for 1 – 6 against six T. cruzi molecular targets. Using this approach, we observed that, even though perseanolide ( 6 ) showed favorable docking to several studied targets, the results were especially promising for hypoxanthine phosphoribosyl transferase (PDB 1TC1). As PDB 1TC1 is associated to the transference of a monophosphorylated ribose from phosphoribosylpyrophosphate (PRPP) in the ribonucleotide synthesis pathway, this interaction may affect the survival of T. cruzi in mammalian cells. The data herein also indicate that possible intermolecular interactions between 6 and PDB 1TC1 derive from (i) hydrogen bonds in the α,β-unsaturated-γ-lactone unity and (ii) hydrophobic interactions in the long-chain alkyl group. Based on our results, perseanolide ( 6 ), reported for the first time in this work, can auspiciously contribute to future works regarding new trypanocidal agents.  相似文献

7α‐Hydroxyfrullanolide ( 1 ), a known sesquiterpenoid, was isolated from Sphaeranthus indicus using an antibacterial‐activity‐directed fractionation method. This compound had exhibited a significant antibacterial activity against Gram‐positive bacteria. Chemical and microbial reactions were performed to prepare eight different analogues of compound 1 in order to evaluate these newly synthesized compounds for antibacterial activity. These compounds were 1β,7α‐dihydroxyfrullanolide ( 2 ), 7α‐hydroxy‐1‐oxofrullanolide ( 3 ), 4,5‐dihydro‐7α‐hydroxyfrullanolide ( 4 ), 11,13‐dihydro‐7α‐hydroxyfrullanolide ( 5 ), 13‐acetyl‐7α‐hydroxyfrullanolide ( 6 ), 2α,7α‐dihydroxysphaerantholide ( 7 ), 4α,5α‐epoxy‐7α‐hydroxyfrullanolide ( 8 ), and 4β,5β‐epoxy‐7α‐hydroxyfrullanolide ( 9 ). Microbial reactions on 1 using whole‐cell cultures of Cunninghamella echinulata and Curvularia lunata yielded compounds 2 – 4 . Incubation of compound 1 with the liquid cultures of Apsergillus niger and Rhizopus circinans yielded metabolites 5 – 7 , while 8 and 9 were prepared by carrying out an epoxidation reaction on 1 using meta‐chloroperbenzoic acid (mCPBA). Structures of compounds 2 – 9 were elucidated with the aid of extensive NMR spectral studies. Compounds 2 – 4 were found to be new metabolites. Compounds 1 – 9 were evaluated for antibacterial activity and found to exhibit a wide range of bioactivities. Antibacterial‐activity data of 1 – 9 suggested that the bioactivity of 1 is largely due to the presence of C(4)?C(5), C(11)?C(13), and a γ‐lactone moiety.  相似文献

A new sesquiterpenoid, 1 , and three new diterpenoids, 3 – 5 , along with five known compounds, 2 and 6 – 9 , were isolated from rhizomes of Alpinia japonica. The structures of the new compounds were determined as (1R,4R,6S,7S,9S)‐4α‐hydroxy‐1,9‐peroxybisabola‐2,10‐diene ( 1 ), methyl (12E)‐16‐oxolabda‐8(17),12‐dien‐15‐oate ( 3 ), (12R)‐15‐ethoxy‐12‐hydroxylabda‐8(17),13(14)‐dien‐16,15‐olide ( 4 ), and methyl (11E)‐14,15,16‐trinorlabda‐8(17),11‐dien‐13‐oate ( 5 ) by means of spectroscopic data. The absolute configurations at C(4) in 1 and C(12) in 4 were deduced from the circular dichroism (CD) data of the in situ‐formed [Rh2(CF3COO)4] complexes. Inhibitory effects of the isolates on NO production in lipopolysaccharide‐induced RAW264.7 macrophages were evaluated, and 2 – 4, 6 , and 7 were found to exhibit inhibitory activities with IC50 values between 14.6 and 34.3 μM .  相似文献

Gentiana rhodantha Franch. ex Hemsl. (Gentianaceae), an annual herb widely distributed in the southwest of China, has been medicinally used for the treatment of inflammation, cholecystitis, and tuberculosis by the local people of its growing areas. Chemical investigation on the whole plants led to the identification of eight new phenolic compounds, rhodanthenones A–D ( 1 – 4 , resp.), apigenin 7‐O‐glucopyranosyl‐(1→3)‐glucopyranosyl‐(1→3)‐glucopyranoside ( 5 ), 1,2‐dihydroxy‐4‐methoxybenzene 1‐Oα‐L ‐rhamnopyranosyl‐(1→6)‐β‐D ‐glucopyranoside ( 6 ), 1,2‐dihydroxy‐4,6‐dimethoxybenzene 1‐Oα‐L ‐rhamnopyranosyl‐(1→6)‐β‐D ‐glucopyranoside ( 7 ), and methyl 2‐Oβ‐D ‐glucopyranosyl‐2,4,6‐trihydroxybenzoate ( 8 ), together with eleven known compounds, 9 – 19 . Their structures were determined on the basis of detailed spectroscopic analyses and chemical methods. Acetylcholinesterase (AChE) inhibition and cytotoxicity tests against five human cancer cell lines showed that only rhodanthenone D ( 4 ) and mangiferin ( 12 ) exhibited 18.4 and 13.4% of AChE inhibitory effects at a concentration of 10−4 M , respectively, while compounds 1 – 5 and the known xanthones lancerin ( 11 ), mangiferin ( 12 ), and neomangiferin ( 13 ) displayed no cytotoxicity at a concentration of 40 μM .  相似文献

A new phloroglucinol derivative, 5‐deprenyllupulonol C ( 1 ), along with four other phloroglucinol derivatives, 2 – 5 , five chalcones, 6 – 10 , four flavanones, 11 – 14 , two flavonol glycosides, 15 and 16 , and five triterpenoids, 17 – 21 , were isolated from the female inflorescence pellet extracts of hop (Humulus lupulus L.). Upon evaluation of these compounds against the Epstein? Barr virus early antigen (EBV‐EA) activation induced by 12‐O‐tetradecanoylphorbol 13‐acetate (TPA) in Raji cells, twelve compounds, i.e., 1 – 4, 11 – 14, 17 – 19 , and 21 , showed potent inhibitory effects on EBV‐EA induction, with IC50 values in the range of 215–393 mol ratio/32 pmol TPA. In addition, eleven compounds, i.e., 1 – 4, 6, 11, 12, 14, 17, 18 , and 20 , were found to inhibit TPA‐induced inflammation (1 μg/ear) in mice, with ID50 values in the range of 0.13–1.06 μmol per ear. Further, lupulone C ( 2 ) and 6‐prenylnaringenin ( 14 ) exhibited inhibitory effects on skin‐tumor promotion in an in vivo two‐stage mouse‐skin carcinogenesis test based on 7,12‐dimethylbenz[a]anthracene (DMBA) as initiator and with TPA as promoter.  相似文献

Six new phloroglucinol derivatives, xanchryones I−N ( 1 – 6 ), were isolated from the leaves of Xanthostemon chrysanthus. Compounds 1 – 6 are unusual phloroglucinol-amino acid hybrids constructed through C2−N and O−C1′ bonds forming a peculiar oxazole ring. The structures and absolute configurations of compounds 1 – 6 were determined by MS, NMR, and single-crystal X-ray diffraction. Moreover, the anti-inflammatory and antibacterial activities of these compounds were evaluated.  相似文献

Three prenylated chalcones, 4‐hydroxyderricin ( 1 ), xanthoangelol ( 2 ), and xanthoangelol F ( 3 ), isolated from Angelica keiskei, were transformed by the fungus Aspergillus saitoi. These chalcones were converted to flavanones (i.e., 4, 8 , and 12 ), and prenyl‐chain‐hydrated (i.e., 5, 7, 9 – 11 , and 13 ) and ring‐B‐hydroxylated (i.e., 6 ) chalcones. The structures of three new metabolites, 7, 9 , and 13 , were established as 2″,3″‐dihydro‐4,3″‐dihydroxyderricin, 6″,7″‐dihydro‐7″‐hydroxyxanthoangelol, and 6″,7″‐dihydro‐7″‐hydroxyxanthoangelol F, respectively. Upon evaluation of cytotoxic activities of compounds 1 – 13 , the metabolite 7 exhibited potent cytotoxicity against HL60 cells, and this cell death was revealed to be mostly due to apoptosis. In addition, compounds 1 – 4, 7 – 10, 12 , and 13 were examined for their inhibitory effects on the induction of Epstein? Barr virus early antigen (EBV‐EA) by 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA) in Raji cells. All compounds tested showed inhibitory effects against EBV‐EA activation with potencies higher than that of β‐carotene. Furthermore, the metabolite 13 exhibited inhibitory effect on skin tumor promotion in an in vivo two‐stage mouse skin carcinogenesis test based on 7,12‐dimethylbenz[a]anthracene (DMBA) as initiator, and with TPA as promoter.  相似文献

Treatment of eight C‐seco limonoids including six of salannin‐type, 1 – 6 , and two of nimbin‐type, 7 and 8 , with a combination of BF3 · Et2O and iodide ion yielded the isomeric C‐seco derivatives, i.e., six isosalannins, 1a – 6a , and two isonimbins, 7a and 8a , respectively. Ohchinin ( 1 ) was further subjected to LiAlH4 reduction which yielded a deesterified trihydroxy limonoid, nimbidinol ( 9 ). In addition, ten limonoids including seven of azadirone‐type, 10 – 16 , and three of gedunin‐type, 17 – 19 , all of which possess no ester functionality in the molecule, were obtained from the neutral fraction of Azadirachta indica seed extract after alkaline hydrolysis. Among the above, twelve compounds, i.e., 1a – 4a , 6a , 9 , 13 – 16 , 18 , and 19 , were new compounds, and their structures were elucidated on the basis of extensive spectroscopic analysis and comparison with literature data. Upon evaluation of all these limonoids for their inhibitory activities against melanogenesis in B16 melanoma cells induced with α‐melanocyte‐stimulating hormone (α‐MSH), five structurally modified limonoids, 3‐deacetyl‐28‐oxosalannin ( 6a ), 9 , 17‐epi‐17‐hydroxynimbocinol ( 14 ), 17‐epi‐17‐hydroxy‐15‐methoxynimbocinol ( 15 ), and 7‐deacetyl‐17‐epinimolicinol ( 18 ), in addition to a natural limonoid, 1 , exhibited potent inhibitory activities with 26 – 66% reduction of melanin content at 100 μm concentration with almost no or low toxicity to the B16 melanoma cells (70 – 99% cell viability at 100 μm ).  相似文献

Ten new cassane-type diterpenoids were isolated from the seeds of Caesalpinia bonduc (L.) Roxb., including 6α-hydroxycaesalpinin P ( 1 ), 14-epi-caesalpinin E1 ( 2 ), 6-deacetylcaesalmin Z ( 3 ), 14-epi-caesalmin Z ( 4 ), caesalpinolides I ( 5 ), K ( 6 ), L ( 7 ), M ( 9 ) and N ( 10 ), and 14-epi-neocaesalpin L ( 8 ). Their planar structures and absolute configurations were fully determined by comprehensive spectroscopic methods, including 2D NMR and electronic circular dichroism spectra. Compounds 1 – 4 are tetracyclic cassane diterpenoids possessing a furan ring, and compounds 5 – 10 are tetracyclic cassane diterpenoids possessing a fused butenolide moiety. The anti-inflammatory and cytotoxic activities of the isolates were evaluated, while none of them showed obvious effects. The current study identified ten new cassane-type diterpenoids from the seeds of C. bonduc (L.) Roxb., which enriched the chemical diversity of the titled herbal medicine.  相似文献

The hexane extract from aerial parts Baccharis sphenophylla Dusén ex Malme (Asteraceae) displayed activity against amastigote forms of Trypanossoma cruzi and was subjected to chromatographic steps to afford one unreported – 7α-hydroxy-ent-abieta-8(14),13(15)-dien-16,12β-olide ( 1 ) and three known diterpenes – ent-kaur-16-en-19-oic acid, ( 2 ), grandifloric acid ( 3 ), and 15β-tiglinoyloxy-ent-kaur-16-en-19-oic acid ( 4 ), two sesquiterpenes – spathulenol ( 5 ) and oplopanone ( 6 ) – as well as hexacosyl p-coumarate ( 7 ). Isolated compounds were characterized by NMR and ESI-HR-MS spectra and were evaluated in vitro for activity against amastigote forms of the parasite T. cruzi – the relevant clinical form in the chronic phase of Chagas disease. In addition, the activity of compounds 1 – 7 against NCTC cells was evaluated. Compounds 1 and 7 showed effectiveness with EC50 values of 21.3 and 16.9 μM, respectively. Both compounds also exhibited reduced toxicity against NCTC cells (CC50>200 μM) with SI values higher than 9.4 and 11.9. Obtained results suggest that the new ent-abietane diterpene 1 and alkyl coumarate 7 could be used as prototypes for the development of novel and selective semisynthetic derivatives against intracellular forms of T. cruzi.  相似文献

Three new lignans ( 1 – 3 ), together with four new thymoquinol glycosides ( 4 – 7 ), were isolated from 70%‐EtOH extract of the rattan stems of Schisandra chinensis. The structures of 1 – 7 were elucidated by detailed spectroscopic analyses, and these new compounds were identified as pinobatol‐9‐Oβ‐d ‐glucopyranoside ( 1 ), 1,2,13,14‐tetramethoxydibenzocyclooctadiene 3,12‐Oβ‐d ‐diglucopyranoside ( 2 ), 3,7‐dihydroxy‐1,2,13,14‐tetramethoxydibenzocyclooctadiene 12‐Oβ‐d ‐glucopyranoside ( 3 ), thymoquinol 2‐Oβ‐d ‐apiofuranosyl‐(1→6)‐β‐d ‐glucopyranoside ( 4 ), thymoquinol 2‐Oα‐d ‐arabinofuranosyl‐(1→6)‐β‐d ‐glucopyranoside ( 5 ), thymoquinol 5‐Oβ‐d ‐apiofuranosyl‐(1→6)‐β‐d ‐glucopyranoside ( 6 ), and thymoquinol 5‐Oα‐d ‐arabinofuranosyl‐(1→6)‐β‐d ‐glucopyranoside ( 7 ). The neuroprotective activity of 1 – 7 was evaluated on PC12 cells with neurotoxicity induced by amyloid‐beta 1 – 42 (Aβ1 – 42). Compounds 2 and 3 showed protecting activity against Aβ‐induced toxicity in PC12 cells.  相似文献

Nine achiral tetraprenylated alkaloids, including three new compounds, named malonganenones I–K ( 1 – 3 , resp.), together with six known analogs, 4 – 9 , were isolated from the gorgonian Euplexaura robusta collected from Weizhou Island of Guangxi Province, China. The structures of compounds 1 – 3 were elucidated by extensive spectral analyses, especially of their 1D‐ and 2D‐NMR data. Compounds 1, 4, 6 , and 7 showed moderate cytotoxicities against K562 and HeLa tumor cell lines with IC50 values ranging from 0.35 to 10.82 μM . Compound 6 also showed moderate inhibitory activity against c‐Met kinase at a concentration of 10 μM .  相似文献
