Synthesis of amphiphilic amylose and starch derivatives   总被引:2,自引:0,他引:2  
For non-food uses starch generally is modified in order to obtain products with properties suitable for various applications. In the present work, starch and amylose were hydrophobically modified through reactions with long-chain -alkyl epoxides (C6 and C12) in DMSO solution, in the presence of NaH as a catalyst. The molar substitution (MS) was calculated from NMR spectra. Derivatives with high as well as low MS values were obtained. In order to reach MS values above 1.5, the reaction had to be run for 150–300 h. Viscosity and GPC measurements indicated that the polysaccharides were degraded in DMSO under the influence of methyl sulfinyl anion, which presumably is the active catalyst.

The derivatives were also characterized by FTIR. The ratio between the peak areas for OH stretching and alkyl stretching vibrations, respectively, in the FTIR spectra, was found to be proportional to MS values determined from NMR spectra.

The solubility of the hydrophobically modified polysaccharide in various solvents was tested. Samples having C12-alkyl side chains and MS>1 were soluble in toluene. The C6 derivatives were water soluble up to a MS value of 0.3.  相似文献

Mucoadhesive properties of cross-linked high amylose starch derivatives   总被引:2,自引:0,他引:2  
Acetate (Ac-), aminoethyl (AE-) and carboxymethyl (CM-)derivatives of cross-linked high amylose starch (HASCL-6) were previously shown to control, over more than 20h, the release of drugs from highly loaded (up to 60% drug) monolithic tablets. It was now of interest to evaluate their mucoadhesive characteristics in view of further utilization in buccal or vaginal transmucosal delivery. The present study shows that ionic AE-HASCL-6 and CM-HASCL-6 derivatives exhibit higher mucoadhesive properties than neutral HASCL-6 and Ac-HASCL-6, suggesting that the ionic groups introduced on cross-linked starch chains play a role in the bioadhesion process. The adhesiveness seemed related to capillary attraction forces. Surface adhesion parameters were calculated for slabs based on the mentioned polymers and corroborated with their swelling behavior at various pH changes. The positively charged AE-derivatives presented a higher adhesion at acidic pH, being thus recommended for vaginal delivery, whereas the negatively charged derivatives (CM-HASCL-6) exhibited a better adhesion at neutral pH, being thus more appropriate for buccal delivery.  相似文献

O-aminopropyl starch was prepared by Michael addition of acrylonitrile and subsequent reduction with freshly prepared cobalt boride and sodium borohydride. In a second approach, the aminopropyl group was introduced via Williamson etherification with N-phthalyl-protected 3-bromo-1-propylamine. The protecting group was removed by borohydride reduction and subsequent hydrolysis in acetic acid. The DS of all samples and the degree of reduction of the cyanoethyl groups were estimated from the 1H NMR spectra. Total monomer composition was determined after methanolysis or hydrolysis and trimethylsilylation by GLC and GCMS. While the regioselectivity in the thermodynamically controlled reaction was O-6 > O-2 > O-3 (50:37:13), the kinetically controlled process showed strongly preferred O-2-etherification (up to 94%) followed by O-6- and O-3-substitution. It could be influenced by choice of solvent (water, Me(2)SO) and base (NaOH, Li-dimsyl).  相似文献

A gelatinized cross-linked high amylose starch matrix with magnetic properties was synthesized via in situ formation of iron oxides inside the polymer matrix. Precipitation and multiple oxidation of ferrous ions were performed. The samples were observed using transmission and scanning electron microscopy, showing morphological changes in the magnetic and polymer phases. The iron content analysis revealed a decay from one oxidation cycle to the next one if no fresh ferrous solutions are added before the multiple oxidation. X-ray diffractograms, magnetization curves and Mössbauer spectra were also recorded for the characterization of the magnetic phase. The products exhibit superparamagnetic properties due to the presence of ferrimagnetic nanoparticles, although some other iron compounds are also present.  相似文献

Small-angle X-ray scattering (SAXS) together with several complementary techniques, such as differential scanning calorimetry and X-ray diffraction, have been employed to investigate the structural features that give diverse functional properties to wheat starches (Triticum aestivum L.) within a narrow range of enriched amylose content (36–43%). For these starches, which come from a heterogeneous genetic background, SAXS analysis of duplicate samples enabled structural information to be obtained about their lamellar architecture where differences in lamellar spacing among samples were only several tenths of nanometer. The SAXS analysis of these wheat starches with increased amylose content has shown that amylose accumulates in both crystalline and amorphous parts of the lamella. Using waxy starch as a distinctive comparison with the other samples confirmed a general trend of increasing amylose content being linked with the accumulation of defects within crystalline lamellae. We conclude that amylose content directly influences the architecture of semi-crystalline lamellae, whereas thermodynamic and functional properties are brought about by the interplay of amylose content and amylopectin architecture.  相似文献

Six novel regioselectively substituted amylose derivatives with a benzoate at 2‐position and two different phenylcarbamates at 3‐ and 6‐positions were synthesized and their structures were characterized by 1H nuclear magnetic resonance (NMR) spectroscopy. Their enantioseparation abilities were then examined as chiral stationary phases (CSPs) for high‐performance liquid chromatography (HPLC) after they were coated on 3‐aminopropyl silica gels. Investigations indicated that the substituents at the 3‐ and 6‐positions played an important role in chiral recognition of these amylose 2‐benzoate serial derivatives. The derivatives demonstrated characteristic enantioseparation and some racemates were better resolved on these derivatives than on Chiralpak AD, which is one of the most efficient CSPs, utilizing coated amylose tris(3,5‐dimethylphenylcarbamate) as the chiral selector. Among the derivatives prepared, amylose 2‐benzoate‐3‐(phenylcarbamate/4‐methylphenylcarbamate)‐6‐(3,5‐dimethylphenylcarbamate) exhibited chiral recognition abilities comparable to that of Chiralpak AD and may be useful CSPs in the future. The effect of mobile phase on chiral recognition was also studied. In general, with the decreased concentration of 2‐propanol, better resolutions were obtained with longer retention times. Moreover, when ethanol was used instead of 2‐propanol, poorer resolutions were often achieved. However, in some cases, improved enantioselectivity was achieved with ethanol rather than 2‐propanol as the mobile phase modifier.  相似文献

Reaction of NADP with 3-propiolactone at pH 6 gave new NADP derivatives carboxyethylated at the 2'-phosphate or 6-amino group, or both: 2'-O-(2-carboxyethyl)phosphono-NAD (I), N6-(2-carboxyethyl)-NADP (II), and 2'-O-(2-carboxyethyl)phosphono-N6-(2-carboxyethyl)-NAD (III). Their structures were assigned on the basis of ultraviolet, 1H-NMR and 31P-NMR spectra, and also treatment with nucleotide pyrophosphatase or alkaline phosphatase. Carbodiimide-promoted reaction of derivative I with 1,2-diaminoethane gave 2'-O-[N-(2-aminoethyl)carbamoylethyl]phosphono-NAD (IV); derivative III gave 2'-O-[N-(2-aminoethyl)carbamoylethyl]phosphono-N6-[N-(2-aminoethyl ) carbamoylethyl]-NAD (IV). The same reaction of derivative II, on the other hand, gave a mixture of N6-[N-(2-aminoethyl)carbamoylethyl]-NADP (Va) and its 3'-phosphate isomer (Vb). The mixture was converted to Va via the 2',3'-cyclic derivative (Vc). Their structures were assigned on the basis of ultraviolet and 1H-NMR spectra, and also treatment with alkaline phosphatase or 3'-nucleotidase. All the NADP derivatives obtained in this work could be reduced with yeast glucose-6-phosphate dehydrogenase.  相似文献

Influence of amylose content on starch films and foams   总被引:1,自引:0,他引:1  
After extraction of smooth pea starch and waxy maize starch from pure amylose and amylopectin fractions, films with various amylose contents were prepared by casting in the presence of water or water with glycerol. For unplasticized films, a continuous increase in tensile strength (40–70 MPa) and elongation (4–6%) was observed as amylose increased from 0 to 100%. Discrepancies with values obtained for native starches with variable amylose content and different botanical origins were attributable to variations in the molecular weights of components. Taking cell wall properties into account, the values obtained in the laboratory were used to improve the relation between the flexural behavior of extruded foams and the model of cellular solids with open cavities.

The properties of plasticized films were not improved by the presence of glycerol and remained constant when amylose content was higher than 40%. Results are interpreted on the basis of topological differences between amylose and amylopectin.  相似文献

[Purpose]

Efficacy and comparative characteristics of fine powdered whole soybean curd.

[Methods]

Ground dried soybean to a fine powder (700 mesh) containing bean components in its entirety, and then produced whole soybean curd. Analysed its nutritive components, bioactive substances, antioxidant activities and texture compared with pressed soybean curd.

[Results]

Compared with pressed soybean curd, the nutrients and isoflavone in whole soybean curd were slightly decreased, but antioxidant activities, dietary fibers and moisture content were increased. Also, the yield rate of the total process was improved 1.9 times.

[Conclusion]

Fine powdered whole bean curd has antioxidant effects, contains dietary fiber and possesses soft characteristics, hence has development potential in the diet market and as food for patients.  相似文献

Activity of starch synthase and the amylose content in rice endosperm   总被引:30,自引:0,他引:30  
The content of amylose in endosperm of non-waxy japonica rice (Oryza sativa cv Akitakomachi) was increased by lowering the growth temperature from 25° to 15° during the ripening period. The activities of sucrose synthase, ADPglucose pyrophosphorylase, starch branching enzyme (Q-enzyme) and soluble starch synthase in endosperm developed at 15° were lower than or similar to those at 25°, when compared on a endosperm basis at the similar ripening stage. In contrast, the activity of starch granule-bound starch synthase, which is considered to be indispensable for amylose synthesis, was higher by 3–3.5-fold in the endosperm developed at the low temperature than that at the high ambient temperature. The results suggest that the low temperature specifically accelerates the expression of the bound starch synthase gene (waxy gene) in rice endosperm, which resulted in elevated amylose biosynthesis in the endosperm when developed at lower temperatures.  相似文献

Industrial corn starches modified with an oxidant, acid or enzyme have been analyzed by a high-performance size-exclusion chromatography (HPSEC) technique. The procedure involves derivatization of the starch to the corresponding tricarbanilate with separation afforded by commercially available polystyrene-divinylbenzene (PS-DVB) gel columns. Separation time was under 30 min and UV detection at 235 nm allows the analysis of microgram quantities. The elution behavior of amylose and amylopectin tricarbanilates appears to depend on allowed conformational states. Amylopectin tricarbanilate (APTC) is hindered by branching and elutes much earlier than the exclusion limit of the column system according to linear polystyrene standards. Observation of an apparent hydrolysis resistant region of starch and the effects of the various hydrolytic treatments are discussed. Relative molecular weight data are presented utilizing the carbanilate of monodisperse pullulan polysaccharides as primary standards.  相似文献

Thermoplastic starch was prepared by mixing native high amylose potato starch and normal potato starch in a Buss co-kneading extruder at starch to glycerol ratios of 100:45 and 100:30. The materials were also conditioned to different moisture contents at different relative humidities at 23 °C. After the mixing, the compounds were extruded into sheets with a Brabender laboratory extruder. The thermoplastic high amylose materials exhibited a higher melt viscosity than the normal potato starch materials when conditioned at 53% relative humidity. Increasing the moisture content in HAP from 27% to 30% (by weight) lowered the melt viscosity to the same level as that of normal potato starch with a moisture content of 28%. In general, the high amylose materials were more difficult to extrude than the thermoplastic material based on normal starch. The main extrusion problems encountered with the high amylose starch were unstable flow, insufficient melt tenacity and clogging of the die. By increasing the moisture content, increasing the compression ratio of the screw and increasing the rotation rate of the screw, the problems were reduced or eliminated. However, only with a starch to glycerol ratio of 100:45 was an acceptable extrusion result obtained. Extruded sheets of such high amylose materials had a stress at break of about 5 MPa at room temperature and 53% relative humidity, whereas the corresponding value for normal potato (thermoplastic) starch was 3 MPa. The elongation at break was also higher in the case of the high amylose material. The results are discussed in terms of residual crystallinity of the starch materials.  相似文献

Starch granules are principally composed of the two glucose polymers amylose and amylopectin. Native starch granules typically contain around 20% amylose and 80% amylopectin. However, it is possible to breed plants that produce starch with very different amylose and amylopectin contents. At present, the precise structural roles played by these two polymers are incompletely understood. In this study, small-angle X-ray scattering techniques have been applied to investigate the effect of varying amylose content on the internal structure of maize, barley and pea starch species. The results suggest that amylose disrupts the structural order within the amylopectin crystallites.  相似文献

Maize (Zea mays spp. mays) is a staple crop for more than 900 million people. The seeds or kernels provide a rich source of calories because ~70 % of the weight is carbohydrate, mostly in the form of starch. The content and composition of starch are complex traits controlled by many genes, offering multiple potential targets for intervention. We used a multigene engineering approach combining the overexpression of Bt2, Sh2, Sh1 and GbssIIa (to enhance the activity of sucrose synthase, AGPase and granule-bound starch synthase) with the suppression of SbeI and SbeIIb by RNA interference (to reduce the activity of starch branching enzyme). Maize plants expressing all six genes plus the selectable marker showed a 2.8–7.7 % increase in the endosperm starch content and a 37.8–43.7 % increase in the proportion of amylose, which was significant compared to untransformed control plants. We also observed improvements in other agronomic traits, such as a 20.1–34.7 % increase in 100-grain weight, a 13.9–19.0 % increase in ear weight, and larger kernels with a better appearance, presumably reflecting the modified starch structure within the kernels. Our results confirm that multigene engineering applied to the starch biosynthesis pathway can not only modulate the quality and quantity of starch but can also improve starch-dependent agronomic traits.  相似文献
