The influence of stonewool substrate on the exudation of the major soluble carbon nutrients and of the auxin precursor tryptophane for Pseudomonas biocontrol agents was studied. To this end, the composition of the organic acids and sugars, as well that of tryptophane, of axenically collected exudates of seed, seedlings, and roots of tomato, cucumber, and sweet pepper was determined. The major results were as follows. i) The total amount of organic acid is much higher than that of total sugar. ii) Exudation of both organic acids and sugars increases during plant growth. iii) Citric, succinic, and malic acids represent the major organic acids, whereas fructose and glucose are the major sugars. iv) Compared with glass beads as a neutral substrate, stonewool substantially stimulates exudation of organic acids and sugars. v) It appeared that enhanced root-tip-colonizing bacteria isolated previously from the rhizosphere of tomato and cucumber grow much better in minimal medium with citrate as the sole carbon source than other, randomly selected rhizobacteria do. This indicates that the procedure which selects for excellent root-tip colonizers enriches for strains which utilize the major exudate carbon source citrate. vi) The content of L-tryptophane, the direct precursor of auxin, is approximately 60-fold higher in seedling exudates of tomato and sweet pepper than in cucumber seedling exudates, indicating a higher possibility of plant growth stimulation after inoculation with auxin-producing rhizobacteria for tomato and sweet pepper crops than for cucumber. However, the biocontrol strain Pseudomonas fluorescens WCS365, which is able to convert tryptophane into auxin, did not stimulate growth of these three crops. In contrast, this strain did stimulate growth of roots of radish, a plant which exudes nine times more tryptophane than tomato does.  相似文献

The aim of this study was to determine the temporal release of fatty acids and sugars from corn and cucumber seeds during the early stages of seed germination in order to establish whether sugars found in exudate can prevent exudate fatty acid degradation by Enterobacter cloacae. Both saturated (long-chain saturated fatty acids [LCSFA]) and unsaturated (long-chain unsaturated fatty acids [LCUFA]) fatty acids were detected in corn and cucumber seed exudates within 15 min after seed sowing. LCSFA and LCUFA were released at a rate of 26.1 and 6.44 ng/min/seed by corn and cucumber seeds, respectively. The unsaturated portion of the total fatty acid pool from both plant species contained primarily oleic and linoleic acids, and these fatty acids were released at a combined rate of 6.6 and 0.67 ng/min/seed from corn and cucumber, respectively. In the absence of seed exudate sugars, E. cloacae degraded linoleic acid at rates of 29 to 39 ng/min, exceeding the rate of total fatty acid release from seeds. Sugars constituted a significant percentage of corn seed exudate, accounting for 41% of the total dry seed weight. Only 5% of cucumber seed exudate was comprised of sugars. Glucose, fructose, and sucrose were the most abundant sugars present in seed exudate from both plant species. Corn seeds released a total of 137 microg/seed of these three sugars within 30 min of sowing, whereas cucumber seeds released 0.83 microg/seed within the same time frame. Levels of glucose, fructose, and sucrose found in corn seed exudate (90 to 342 microg) reduced the rate of linoleic acid degradation by E. cloacae to 7.5 to 8.8 ng/min in the presence of either sugar, leaving sufficient concentrations of linoleic acid to activate Pythium ultimum sporangia Our results demonstrate that elevated levels of sugars in the corn spermosphere can prevent the degradation of LCUFA by E. cloacae, leading to its failure to suppress P. ultimum sporangial activation, germination, and subsequent disease development.  相似文献

以大白菜、萝卜、番茄和黄瓜种子为受体,采用实验室培养皿种子发芽生物测试法研究了黄瓜种子浸提液、种子萌发、胚根和芽苗分泌物、芽苗腐解物和芽苗浸提液的化感效应。结果表明:(1)黄瓜种子浸提液对大白菜、萝卜、番茄和黄瓜种子萌发均有化感抑制作用,即黄瓜种子内含有某些化感抑制物质。(2)在水浸提过的黄瓜种子萌发过程中,它不仅对其近邻套种的大白菜、萝卜和番茄种子萌发产生化感抑制作用,而且其胚根和芽苗分泌物对后茬播种的4种蔬菜种子发芽也表现出不同程度的化感抑制作用;黄瓜芽苗腐解物和芽苗水浸提液也对各受体蔬菜种子发芽与生长产生不同程度的化感抑制作用,且随着腐解芽苗量的增加或浸提液浓度的升高,各受体蔬菜种子的发芽指标值、化感效应指数值和综合效应值随之降低。(3)黄瓜种子浸提液及芽苗各器官的化感物质对黄瓜种子的萌发与生长产生了自毒作用,且黄瓜芽苗腐解物、芽苗浸提液、胚根及芽苗分泌物对受体黄瓜的自毒作用均为最大。研究发现,黄瓜种子浸提液、种子萌发时期以及芽苗各器官的化感物质主要是通过抑制受体胚根的生长而起化感抑制作用,即受体蔬菜种子胚根对化感效应最为敏感;因黄瓜种子及萌发期释放化感物质的途径有所不同,导致受体大白菜、萝卜、黄瓜和番茄的化感响应也不相同;在黄瓜种子萌发和芽苗生长的早期,化感物质即开始在芽苗体内进行合成与积累,一部分可通过胚根和芽苗分泌途径释放到环境中,另一部分可通过芽苗腐解途径释放化感物质,并对受体蔬菜种子萌发与生长表现出较强的化感抑制作用。  相似文献

Summary We detected aromatic and aliphatic acids in root and seed exudates of aseptic cultures of pea, cotton and barley plants by thin-layer and gas-liquid chromatography. There were traces of p-hydroxybenzoic acid in the root and seed exudates of all three plant species. Acid hydrolysis of pea and barley seed exudates yielded p-hydroxybenzoic, and of cotton seed exudates yielded p-coumaric acid, as the predominant aromatic acid constituents of materials exuded by the germinating seeds. Lactic was the predominant aliphatic acid detected in pea and barley root exudates whereas malic acid was predominant in cotton exudates. With the exception of citric acid in peas, malic acid was the predominant acid found in pea, cotton and barley seed exudates. The germinating seed was responsible for a large portion of the total aliphatic and aromatic acid exudation of the seedling plant grown aseptically for 14 days. Trade names are used in this publication only to provide scientific information. Summary Seed and root exudates from three varieties of both broad bean and cotton grown aseptically were analyzed for amino acids and sugars. Broad bean varieties were found to excrete considerable amounts of amino compounds with 15 different amino acids, while cotton varieties excreted less consisting of 12 amino acids. Five sugars were identified in the seed and root exudates from broad bean varieties, while three were present in the seed and root exudates from cotton varieties. Seed and root exudates stimulated spore germination and growth of fungi isolated from the rhizosphere.  相似文献

Abstract Intact seeds and seed and seedling root exudates of velvetleaf ( Abutilon theophrasti Medik.) were used as chemoattractants in experiments to determine the relative importance of chemotaxis in spermosphere and rhizosphere colonization by selected rhizobacteria. Results for soft-agar, capillary tube and soil chemotaxis assays indicated that selected deleterious rhizobacteria were specifically attracted to seed and seedling root exudates. Several amino acids and sugars detected in exudates were chemoattractants for these rhizobacteria. Using soil-chemotaxis assemblies, migration of rhizobacterial isolates through 2-cm distances of soil toward velvetleaf seeds or exudates was detected within 24 h. Isolates were not detected at the same site in soils without seeds or exudates until 72 h after inoculation. These results suggest that attraction of delecterious rhizobacteria toward seeds and seedling roots mediated by exudates (chemotaxis) might be the first step in establishment and subsequent colonization of biological control bacteria on weed seeds and seedling roots in soil.  相似文献

利用滇重楼(Paris polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.)种子外种皮和胚乳的水浸液对白菜(Brassica pekinensis(Lour.)Rupr.)、绿豆(Vigna radiata(Linn.)Wilczak)、小麦(Triticum aestivum L.)种子进行处理,研究滇重楼种子水浸液对3种植物种子萌发、幼苗生长和保护酶活性的影响,并利用GC-MS方法对滇重楼种子内源抑制物的成分进行分析。结果显示,不同浓度滇重楼外种皮、胚乳水浸液对上述3种受体植物的发芽率、苗高、根长及鲜重均产生影响,其作用强度和水浸液的浓度有关,总体上表现出低促高抑的双重浓度效应。滇重楼种子水浸液对白菜的影响作用最强,对绿豆的影响作用最弱,且胚乳水浸液的影响较外种皮强。不同浓度滇重楼种子外种皮和胚乳水浸液均能影响3种植物幼苗体内保护酶的活性,随着水浸液浓度的升高,叶片中超氧化物歧化酶(SOD)、过氧化物酶(POD)活性总体增加,与对照相比差异显著。白菜、小麦过氧化氢酶(CAT)活性减少,与对照相比差异显著;绿豆过氧化氢酶(CAT)活性增加,但与对照相比无显著差异。利用GC-MS方法从胚乳和外种皮水浸液中分别检出8种和2种物质。研究结果表明滇重楼种子中存在内源抑制物质,可能是导致种子休眠的原因;种子水浸液可能通过影响植物幼苗保护酶的活性进而影响其正常生长;有机酸类物质可能是滇重楼种子内源抑制物之一。  相似文献

Summary Studies conducted to examine the exudation pattern of amino acids and sugars in four crop plants,viz sorghum, sunnhemp, ragi, and tomato indicated that in all, 17 known amino acids and 4 sugars were exuded and that the number and nature of the exuded amino acids and sugars differed with the plant species and with the age of plant. Glutamic and aspartic acids were found to be present in the exudates of all the plant species at all stages of plant growth examined. The quantities of amino acids and sugars differed with plant species and the maximum quantity of the chemicals was exuded during the early stages of plant growth. Glutamic acid among amino acids, and glucose among sugars, were always present in higher concentrations than the others, in the exudates in all the four crop plants.Foliar application of nitrogen in the form of NaNO3 and phosphorus as Na2HPO4, was found to alter the exudation pattern of amino acids and sugars and such influence differed in different plant species. There was a general increase in the total concentration of amino acids and a decrease in sugar content in the exudates after treatment of the foliage with N, while a decrease in the amino acid content and increase in total sugars with P-treatment was observed.  相似文献

以黄瓜为受体,以不同化感效应(促进/抑制)小麦品种为供体,采用PCR-DGGE技术,研究了小麦根系分泌物及伴生小麦对黄瓜生长及土壤真菌群落结构的影响.结果表明: 在处理第6天和第12天,化感促进效应小麦根系分泌物分别显著提高了黄瓜幼苗株高和茎粗;在处理第18天,化感促进和抑制效应小麦根系分泌物均显著提高了黄瓜幼苗株高;在处理第6天,不同化感效应小麦根系分泌物均显著降低了黄瓜幼苗根际土壤真菌群落条带数、Shannon指数及均匀度指数,有苗对照(W)显著高于无苗对照(Wn);在处理第18天,各处理的真菌群落结构条带数、Shannon指数及均匀度指数均显著高于无苗对照(Wn).伴生化感抑制效应小麦显著降低了黄瓜根际土壤真菌群落Shannon指数和均匀度指数,说明小麦根系分泌物及伴生小麦改变了土壤真菌群落结构.DGGE图谱及其主成分分析结果表明,伴生不同化感效应小麦对土壤真菌群落结构影响较大.  相似文献

Summary The amounts of amino acids in seed exudates were generally higher than in root exudates of the same plant. The spectra and relative abundance of amino acids in both plants were similar but they were generally more abundant in cowpea exudates than in sorghum. Glutamic acid and alanine were the most abundant amino acids in the seed and root exudates of both plants. The proportions of the amino acids in the seed exudates were comparable to that stored in the seeds. Many of the major amino acids identified in the exudates were also found to support thein vitro growth ofFusarium spp. isolated from the rhizosphere and rhizoplane. This suggests that the amino acids exuded might contribute signficantly to Fusaria nutrition and its consequent predominance around the root. The significance of this pathogenesis is also discussed.  相似文献

Many plants exude allelochemicals--compounds that affect the growth of neighbouring plants. This study reports further studies of the reported effect of cress (Lepidium sativum) seed(ling) exudates on seedling growth in Amaranthus caudatus and Lactuca sativa. In the presence of live cress seedlings, both species grew longer hypocotyls and shorter roots than cress-free controls. The effects of cress seedlings were allelopathic and not due to competition for resources. Amaranthus seedlings grown in the presence of cress allelochemical(s) had longer, thinner hypocotyls and shorter, thicker roots--effects previously attributed to lepidimoide. The active principle was more abundant in cress seed exudate than in seedling (root) exudates. It was present in non-imbibed seeds and releasable from heat-killed seeds. Release from live seeds was biphasic, starting rapidly but then continuing gradually for 24 h. The active principle was generated by aseptic cress tissue and was not a microbial digestion product or seed-treatment chemical. Crude seed exudate affected hypocotyl and root growth at ~25 and ~450 μg ml(-1) respectively. The exudate slightly (28%) increased epidermal cell number along the length of the Amaranthus hypocotyl but increased total hypocotyl elongation by 129%; it resulted in a 26% smaller hypocotyl circumference but a 55% greater epidermal cell number counted round the circumference. Therefore, the effect of the allelochemical(s) on organ morphology was imposed primarily by regulation of cell expansion, not cell division. It is concluded that cress seeds exude endogenous substances, probably including lepidimoide, that principally regulate cell expansion in receiver plants.  相似文献

通过盆栽试验收集了不同生育期单作和间作小麦、蚕豆的根系分泌物,用HPLC分析了根系分泌物中低分子量有机酸的含量和种类.结果表明: 小麦-蚕豆间作显著提高了有机酸的分泌量,在小麦分蘖期(57 d)、孕穗期(120 d)和灌浆期(142 d),间作使小麦根系有机酸分泌量分别提高155%、35.6%和92.6%;在蚕豆分枝期(57 d)和籽粒膨大期(142 d),间作使蚕豆根系有机酸分泌量提高87.4%和38.7%.小麦-蚕豆间作改变了根系分泌物中有机酸的种类,与单作小麦相比,在分蘖期,间作小麦根系分泌物中增加了乳酸;在拔节期(98 d),间作小麦根系分泌物中增加了柠檬酸,但未检测到乙酸;在蚕豆分枝期,间作蚕豆根系分泌物中增加了乙酸,但未检测到乳酸;在蚕豆籽粒膨大期,间作蚕豆根系分泌物中增加了乳酸.小麦-蚕豆间作提高了小麦根系有机酸的分泌速率,在小麦孕穗期,间作小麦分泌柠檬酸、富马酸的速率是单作小麦的179和184倍;在小麦灌浆期,间作小麦分泌乳酸的速率是单作的2.53倍.总之,小麦-蚕豆间作增加了有机酸的分泌量,改变了根系分泌物中有机酸的种类,提高了小麦根系有机酸的分泌速率.  相似文献

Under two monoxenic culture techniques of growing plants (filter paper and silica sand cultures), sugar in root exudate from Meloidogyne incognita-infected tomato increased 133 to 836% over controls. In contrast, amino acids were moderately reduced 52 to 56%. Chromatographic analysis showed that galled root exudate contained three sugars, twelve amino acids, and three organic acids, whereas healthy root exudate contained four sugars, fifteen amino acids, and four organic acids. Polysaccharide was responsible for the large increase of sugars in galled root exudates. The concn and the absolute amount of total sugars in the infected plant xylem sap were greater than in healthy plant xylem sap up to 6 wk after inoculation, whereas amino acids were moderately lower than in controls throughout the test period. Chromatographic analysis showed that xylem sap from both healthy and infected plants at 4 wk after inoculation contained four sugars and five organic acids. We identified 18 and 17 amino acids in the healthy and infected plant xylem sap, respectively. The concn of sugar increased as the nematode inoculum increased at 2, 4 and 6 wk after inoculation. The amino acids in all samples from the infected plant moderately decreased with an increase of nematode inoculum. We suggest that changes in total sugars and amino acids, of infected plant xylem sap and root exudate are a probable mechanism by which tomato plants are predisposed to Fusarium wilt.  相似文献

Summary Three gram varieties varying in susceptibility to pre-emergence damping-off caused byRhizoctonia bataticola were compared for amounts of seed exudation. The results showed that the greatest seed exudation occurred with the variety G-130 which is the most susceptible to pre-emergence damping-off compared with the resistant varieties BG-203 and C-214. Germinating gram seeds exuded greater quantities of soluble carbohydrate and amino acids when incubated at 35°C than at 15 or 25°C. Exudates from seeds germinated at 35°C stimulated more mycelial growth ofR. bataticola than did exudates from seeds incubated at 15 or 25°C. Evidence is presented suggesting that increased seed exudation is a major factor contributing to increased pre-emergence damping-off of gram seedlings byR. bataticola at high temperatures.  相似文献

This study was initiated to understand whether differential biological control efficacy of Enterobacter cloacae on various plant species is due to differences in the ability of E. cloacae to inactivate the stimulatory activity of seed exudates to Pythium ultimum sporangium germination. In biological control assays, E. cloacae was effective in controlling Pythium damping-off when placed on the seeds of carrot, cotton, cucumber, lettuce, radish, tomato, and wheat but failed to protect corn and pea from damping-off. Seeds from plants such as corn and pea had high rates of exudation, whereas cotton and cucumber seeds had much lower rates of exudation. Patterns of seed exudation and the release of P. ultimum sporangium germination stimulants varied among the plants tested. Seed exudates of plants such as carrot, corn, lettuce, pea, radish, and wheat were generally more stimulatory to P. ultimum than were the exudates of cotton, cucumber, sunflower, and tomato. However, this was not directly related to the ability of E. cloacae to inactivate the stimulatory activity of the exudate and reduce P. ultimum sporangium germination. In the spermosphere, E. cloacae readily reduced the stimulatory activity of seed exudates from all plant species except corn and pea. Our data have shown that the inability of E. cloacae to protect corn and pea seeds from Pythium damping-off is directly related to its ability to inactivate the stimulatory activity of seed exudates. On all other plants tested, E. cloacae was effective in suppressing damping-off and inactivating the stimulatory activity of seed exudates.  相似文献

During germination of the castor bean all of the contents of the endosperm are ultimately transported to the embryo through the cotyledon or respired. A net loss of nitrogen from the endosperm begins about the fourth day, i.e. at the time when embryo growth and fat breakdown are also beginning. Amino acid analysis of the exudate from the cotyledons, still enclosed in the endosperm, showed that the amounts of aspartate, glutamate, glycine, and alanine were very low and that glutamine made up 40% of the amino acids in the exudate.

Amino acids labeled with 14C were applied to intact excised endosperms to follow utilization. Aspartate, glutamate, alanine, glycine, serine, and leucine were converted to sugar to varying extents. Proline, arginine, valine, and phenylalanine were not appreciably converted to sugars. Proline and glutamate were converted to glutamine. When 14C-glutamate, aspartate, and alanine were added to the outer endosperm of intact seedlings, only sugars and glutamine contained appreciable label in the exudate. When 14C-valine was added, it was virtually the only labeled compound in the exudate.

The results show that amino acids which on deamination can give rise to intermediates in the pathway of conversion of fat to sucrose are largely converted to sucrose and the nitrogen transported as glutamine. Other amino acids released from the endosperm protein are transported intact into the seedling axis. Some carbon from the gluconeogenic amino acids is also transported as glutamine.

为探讨温室蔬菜CO2施肥的根际效应,以黄瓜幼苗为试材,研究了CO2施肥(上午施肥/上、下午施肥;施肥浓度/对照浓度(950±50)/(350±50)μmol/molCO2)对根系生长及分泌物和伤流液组成的影响。结果表明,CO2施肥明显促进黄瓜幼苗根系发育,根系生物量显著增加;单株根系分泌物中氨基酸、糖、有机酸和酚酸总量增加,但单位鲜重根系分泌量却呈现增幅减少、无变化甚至降低趋势,说明单株分泌量增加主要由根系生长量的增加所引起。CO2施肥促进幼苗对养分的吸收,伤流液中矿质元素、ZT浓度增加,但GA、ABA和IAA浓度降低;与上午CO2施肥相比,上、下午均CO2施肥的效果更明显。CO2施肥促进了黄瓜幼苗根系发育及其代谢活性,为地上部的旺盛生长创造了条件。  相似文献

通过田间小区试验和水培试验,研究了3个品种小麦(云麦42、云麦47和绵阳29)与蚕豆间作对蚕豆地上部生物量、枯萎病发生、根际微生物代谢功能多样性和枯萎病病原菌尖孢镰刀菌数量的影响,分析了3个不同小麦品种低分子量根系分泌物组分(糖、氨基酸和有机酸)的差异.结果表明: 云麦42与蚕豆间作(YM42/B)和云麦47与蚕豆间作(YM47/B)使蚕豆地上部生物量分别增加16.6%和13.4%,使蚕豆枯萎病病情指数分别降低47.6%和23.3%,绵阳29与蚕豆间作(MY29/B)对蚕豆地上部生物量和枯萎病病情指数均无显著影响.与单作蚕豆相比,YM42/B和YM47/B处理均显著提高了蚕豆根际微生物的平均颜色变化率(AWCD)及对碳源的总利用强度和Biolog ECO板中各种碳源的利用,明显改变了蚕豆根际微生物群落结构,并显著降低了蚕豆根际的尖孢镰刀菌数量;MY29/B处理对AWCD值、碳源总利用强度和Biolog ECO板中各种碳源的利用及蚕豆根际尖孢镰刀菌数量均无显著影响,也未明显改变蚕豆根际的微生物群落结构.3个小麦品种根系分泌物中可溶性糖、游离氨基酸总量和有机酸总量均表现为MY29>YM47>YM42.MY29根系分泌物中丝氨酸、谷氨酸、甘氨酸、缬氨酸、蛋氨酸、苯丙氨酸、赖氨酸的含量均显著高于YM42和YM47;精氨酸为YM42、YM47所特有,而亮氨酸为MY29所特有.MY29和YM47根系分泌物中检出酒石酸、苹果酸、柠檬酸、琥珀酸、延胡索酸和顺乌头酸6种有机酸,而YM42根系分泌物中检出洒石酸、苹果酸、柠檬酸和延胡索酸4种有机酸,且MY29和YM47根系分泌物中苹果酸含量显著高于YM42.小麦与蚕豆间作提高了蚕豆根际微生物的活性和碳源利用强度,改变了根际微生物的群落结构,降低了蚕豆根际尖孢镰刀菌的数量,促进了蚕豆生长,抑制了蚕豆枯萎病的发生,但小麦与蚕豆间作控病效果受小麦品种的影响,表明间作系统中非寄主作物根系分泌物的差异是影响间作对土传病害控制的重要因素.  相似文献

In a medium containing bean, barley and wheat seed exudates,Xanthomonas phaseoli var.fuscans (Burk.) Starr et Burk. grew substantially better than in that containing root exudates of these plants. When the bacteria were cultivated in a medium containing root exudates of bean plants deprived of cotyledons after eleven days of growth, growth was slower than in the presence of root exudates of control plants. On the other hand, the growth was stimulated in a medium containing root exudates of bean plants deprived of leaves. It was found that seed exudates of these plants contained biologically active peptides stimulating the growth of the microorganism. These peptides were not found in root exudates. These findings suggest a relationship between the survival ofXanthomonas phaseoli var.fuscans in the rhizosphere of bean and the exudation of biologically active peptides originating from the stock substances of seeds and cotyledons.  相似文献
