A portable, immersible incubator to measure the primary productivity of phytoplankton is described. Battery operated, inexpensive,
it is designed to overcome the problem of changes in natural illumination. Tested over three years, using the 14C-technique, it is now successfully used in research work on South Wales rivers. This incubator is not submersible. 相似文献

利用2000-2006年MOD17A3数据集的年均NPP数据和GIS技术定量分析了陕西省植被NPP的时空变化特征,结果表明:陕西省年NPP变化范围为340 ~434 g C·m-2·a-1,平均值为383 g C·m-2·a-1;年均NPP分布全省呈现北低南高,关中、陕南呈现西高东低的趋势.长城沿线风沙区年平均NPP在0~200gC·m-2·a-1,黄土高原丘陵沟壑区年平均NPP在200~ 300 g C·m-2·a-1,中部林区年平均NPP在400~500 g C·m-2·a-1,渭北旱作农业区年平均NPP在300~400 g C·m-2·a-1,关中灌溉农作区年平均NPP大部在400~500 g C·m-2·a-1,秦巴山地林区年平均NPP >400 gC·m-2·a-1.与2000年相比,2006年陕西省年NPP大部分地区是增加的,年NPP增加的面积占国土面积的90.5%.陕西省NPP线性变化趋势以增加为主,NPP变化百分率增加10%以上的面积所占陕西省国土面积的比例为50.6%;植被NPP的变化百分率>10%的植被主要分布在延安市以北地区,说明通过实施退耕还林等生态建设工程,这些地区植被状况得到较好的改善. 相似文献

The impact of navigationally induced suspended sediments from the Upper Great Lakes Connecting Channels on the size-fractionated primary productivity was evaluated by the Carbon-14 technique. The method applied was on-site, rapid, sensitive, inexpensive, and provided dynamic-toxicological information essential for hazard assessment. Both enhancement and inhibition of the primary productivity was observed in various parts of the Upper Great Lakes Connecting Channels. These responses appear to depend on the type of natural plankton and their exposure to various contaminant/nutrient complexes generated by the disturbance of the bottom sediments during the passage of ships. Traditionally, only the inhibition of primary productivity has been monitored from a toxicity point of view, but it is important also to evaluate the implications of enhancement since it may result in increased eutrophication, propagation of nuisance blooms, and change of intricate food-web interactions. The procedure adopted in this study for the first time appears to possess considerable potential for a simple and rapid screening of environmental perturbations resulting from navigational activities. 相似文献

Ministik Lake (longitude 113°01; latitude 53°21), a well-mixed, shallow (mean depth 1.83 m), eutrophic lake is characterized by eutrophic chlorococcalean and cyanophycean phytoplankton associations, and little change in standing crop size with increasing depth. Standing crops and primary productivity are low during the winter but pronounced spring and late summer/early autumn maxima occur. Mean yearly areal standing crop (B) and primary productivity (A) were 199.2 mg m–2 chlorophyll a and 319.5 mg C hr–1 m–2 respectively. Annual productivity was estimated at 1399,6 g C m–2yr–1. The mean increase in the extinction coefficient () per unit increase in standing crop (B) was 0.03 In units m–1. High non-algal light attenuation (q) occurred averaging 46% which prevented the ratio B from attaining more than 74.2% of the theoretical maximum except once when selfshading occurred. Insignificant relationships existed between B (mg m–3 chlorophyll a) and Amax (mg C hr–1 m–3), A and B and A and B. Close correlations existed between A and Amax/ and A and I0 (W m–2). The depth of the euphotic zone (Zeu) varied between 0.6 and 2.0 m; the average relationship between zeu and was zeu = 3.78/te, and the mean standing crop in the euphotic zone represented 58.3% of the theoritical maximum. The high q values made the model of Talling (1957) inapplicable to this lake. The Q10 value for the lake was 1.20. max (mg C chlorophyll a
–1 hr–1) was closely related to both temperature and irradiance, and depressed by high pH values.Growth of the cyanophycean algae was correlated with temperature, chlorophycean algae with phosphate-phosphorus and temperature and the diatoms with dissolved silica and phosphate-phosphorus, but only in the case of Chaetoceros elmorii did any nutrient appear limiting. Carbon dioxide and oxygen exchange procedures for measuring community metabolism (two open stream methods and three chamber methods) were compared on the same reach of a third-order stream. Open stream methods were complicated by high diffusion rates and yielded net community primary productivity estimates lower than those obtained with chamber methods. Chamber methods yielded variable productivity and respiration data. However, when normalized for chlorophyll a, productivity estimates from the chamber methods were within an expected range for the system. Balances of photosynthesis and respiration from the chamber methods were similar between methods and indicated that autotrophic or heterotrophic processes could dominate the system. Considerations in applying the various procedures are discussed. 相似文献

采用CASA(Carnegie-Ames-Stanford Approach)模型,结合TERRA MODIS卫星数据和气象数据,对毛乌素沙地海流兔河流域2015年各月的植被净初级生产力(NPP)进行估算,并对植被NPP月平均值的时空分布规律及其与气象因子和地下水位埋深的关系进行了分析.结果表明:毛乌素沙地海流兔河流域2015年植被NPP总量为2.88×1011 g,生长季(4月份至10月份)的植被NPP总量达2.81×1011 g,占全年植被NPP总量的97.57%.随着时间推移,植被NPP月平均值和归一化差分植被指数(NDVI)月平均值呈"缓慢增加—急剧增加—急剧下降"的变化趋势.植被NPP月平均值季节变化明显,春季、夏季、秋季和冬季植被NPP月平均值之和分别为20.55、69.39、20.46和0.48 g·m-2.从空间分布上看,中部河谷和滩地的植被NPP月平均值总体上高于东南部、西部和西北部等沙丘荒漠区.月平均气温对植被NPP月平均值变化的影响最大,其次为平均实际日蒸散发量和地表月太阳辐射.植被NPP月平均值随着地下水位埋深的增加而减小,最大值出现在地下水位埋深1~2 m之间.上述研究结果显示:采用CASA模型可以较好地估算毛乌素沙地海流兔河流域植被NPP值,月平均气温和地下水位埋深对该流域植被NPP值的影响较大. 相似文献

Goal, Scope and Background Green Productivity (GP) is a new paradigm in sustainable manufacturing where resource conservation and waste minimization
constitute the strategy in simultaneously enhancing environmental performance and productivity. This productivity approach
to the sustainability of industries requires the adoption of clean production technology and the development of appropriate
indicators and instruments to measure environmental performance in a continuous improvement strategy that focuses on the manufacturing
stage of the product life cycle. The analysis may be expanded to include the entire life cycle with increasing details on
impacts, improvement strategies and indicators.
Methods The study proposes a methodology for GP assessment that integrates the essential components of life cycle assessment (LCA)
and multicriteria decision analysis specifically the analytic hierarchy process (AHP). LCA provides a systematic and holistic
perspective for GP analysis that spans inventory, impact and improvement assessment. The AHP is utilized as a decision framework
and valuation tool for impact and improvement assessment to come up with priority weights. Indicators are derived and measured
from a streamlined LCA focused on a number of parameters within the gate-to-gate analysis to demonstrate the GP concept in
relation to resource utilization and waste minimization. An input-output approach using a suitable material balance in a scenario
analysis provides the basis of GP performance measurement.
Results and Conclusion The diagnostic model is applied on a semiconductor assembly/packaging operation. From the streamlined life cycle inventory,
impact factors were derived for water resource depletion (WRD), energy resource depletion (ERD), human toxicity-air (HTA),
human toxicity-land (HTL), human toxicity-water (HTW), aquatic ecotoxicity (ETA) and terrestrial ecotoxicity (ETT). Valuation
of impact factors using the AHP showed the high significance of ETT, HTL, WRD and ERD. This especially reflects the impact
of the industry on the solid waste problem as a result of emissions to land associated with human toxicity and ecotoxicity
effects and the intensive use of water and energy resources. Using scenario analysis, the effect of implementing a process-based
improvement technique on a product-specific operation was determined and the highest values in GP are for energy utilization,
water utilization and terrestrial ecotoxicity.
Recommendation and Perspective Expert system technology was explored in developing a diagnostic prototype that emulates how human experts diagnose green
productivity of manufacturing processes. The aim was to investigate how such a diagnosis could be performed in an intelligent
fashion that it is also easily accessible as a decision support for industries. The expert system model will provide flexibility
in testing the relationships of environmental performance and productivity parameters as well as in preserving and disseminating
valuable human expertise in GP program implementation. This is a continuing research effort that is building the knowledge
base for GP assessment. It will include case studies over a wider range or level of detail regarding the impacts and improvement
techniques and the other stages of the product life cycle. 相似文献

Conventional gap‐filling procedures for eddy covariance (EC) data are limited to calculating ecosystem respiration (RE) and gross ecosystem productivity (PG) as well as missing values of net ecosystem productivity (FNEP). We develop additional postprocessing steps that estimate net primary productivity (PN), autotrophic (Ra), and heterotrophic respiration (Rh). This is based on conservation of mass of carbon (C), Monte Carlo (MC) simulation, and three ratios: C use efficiency (CUE, PN to PG), Ra to RE, and FNEP to RE. This procedure, along with the estimation of FNEP, RE, and PG, was applied to a Douglas‐fir dominated chronosequence on Vancouver Island, British Columbia, Canada. The EC data set consists of 17 site years from three sites: initiation (HDF00), pole/sapling (HDF88), and near mature (DF49), with stand ages from 1 to 56 years. Analysis focuses on annual C flux totals and C balance ratios as a function of stand age, assuming a rotation age of 56 years. All six C balance terms generally increased with stand age. Average annual PN by stand was 213, 750, and 1261 g C m−2 yr−1 for HDF00, HDF88, and DF49, respectively. The canopy compensation point, the year when the chronosequence switched from a source to a sink of C, occurred at stand age ca. 20 years. HDF00 and HDF88 were strong and moderate sources (FNEP=−581 and −138 g C m−2 yr−1), respectively, while DF49 was a moderate sink (FNEP=294 g C m−2 yr−1) for C. Differences between sites were greater than interannual variation (IAV) within sites and highlighted the importance of age‐related effects in C cycling. The validity of the approach is discussed using a sensitivity analysis, a comparison with growth and yield estimates from the same chronosequence, and an intercomparison with other chronosequences. 相似文献

Autophagy is the intracellular bulk degradation process to eliminate damaged cellular machinery and to recycle building blocks, and is crucial for cell survival and cell death. Amino acids modulate autophagy in response to nutrient starvation and oxidative stress. We investigated the relevance of reactive oxygen species (ROS) production on the regulation of autophagy using amino acids, both as a mixture and individually, in rat hepatoma H4-II-E cells. Nutrient starvation elevated ROS production and stimulated autophagy. Treatment with complete (CAA), regulatory (RegAA) and non-regulatory (NonRegAA) amino acid mixtures showed significant suppression of ROS production, whereas only CAA and RegAA exhibited significant suppression of autophagy, suggesting a dissociation of the two responses. The effects of individual amino acids were examined. Leucine from RegAA decreased ROS production and suppressed autophagy. However, methionine and proline from RegAA and arginine, cystine and glutamic acid from NonRegAA suppressed autophagy with an opposite increase in ROS production. Other amino acids from the NonRegAA group showed stimulating effects on ROS production without an autophagic response. Arginine’s effect on autophagy suppression was not blocked by rapamycin, indicating an mTOR-independent pathway. Inhibitor studies on arginine-regulated autophagy may indicate the involvement of NO pathway, which is independent from ROS and mTOR pathways. 相似文献

A wide range of cell types depend on mechanically induced signals to enable appropriate physiological responses. The skeleton is particularly dependent on mechanical information to guide the resident cell population towards adaptation, maintenance and repair. Research at the organ, tissue, cell and molecular levels has improved our understanding of how the skeleton can recognize the functional environment, and how these challenges are translated into cellular information that can site-specifically alter phenotype. This review first considers those cells within the skeleton that are responsive to mechanical signals, including osteoblasts, osteoclasts, osteocytes and osteoprogenitors. This is discussed in light of a range of experimental approaches that can vary parameters such as strain, fluid shear stress, and pressure. The identity of mechanoreceptor candidates is approached, with consideration of integrins, pericellular tethers, focal adhesions, ion channels, cadherins, connexins, and the plasma membrane including caveolar and non-caveolar lipid rafts and their influence on integral signaling protein interactions. Several mechanically regulated intracellular signaling cascades are detailed including activation of kinases (Akt, MAPK, FAK), β-catenin, GTPases, and calcium signaling events. While the interaction of bone cells with their mechanical environment is complex, an understanding of mechanical regulation of bone signaling is crucial to understanding bone physiology, the etiology of diseases such as osteoporosis, and to the development of interventions to improve bone strength. 相似文献

Mutants of the plasma membrane Ca(2+) pump (human isoform 4xb) with deletions in the linker between domain A and transmembrane segment M3 (A(L) region) were constructed and expressed in Chinese hamster ovary cells. The total or partial removal of the amino acid segment 300-349 did not change the maximal Ca(2+) transport activity, but mutants with deletions involving residues 300-338 exhibited a higher apparent affinity for Ca(2+) than the wild type h4xb enzyme. Deletion of the putative acidic lipid interacting sequence (residues 339-349) had no observable functional consequences. The removal of either residues 300-314 or 313-338 resulted in a similar increase in the apparent Ca(2+) affinity of the pump although the increase was somewhat lower than that obtained by the deletion 300-349 suggesting that both deletions affected the same structural determinant. The results show that alterations in the region of the alternative splicing site A change the sensitivity to Ca(2+) of the human isoform 4 of the PMCA. 相似文献

The long-lived, light-induced radical YD• of the Tyr161 residue in the D2 protein of Photosystem II (PSII) is known to magnetically interact with the CaMn4 cluster, situated ∼ 30 Å away. In this study we report a transient step-change increase in YD• EPR intensity upon the application of a single laser flash to S1 state-synchronised PSII-enriched membranes from spinach. This transient effect was observed at room temperature and high applied microwave power (100 mW) in samples containing PpBQ, as well as those containing DCMU. The subsequent decay lifetimes were found to differ depending on the additive used. We propose that this flash-induced signal increase was caused by enhanced spin relaxation of YD• by the OEC in the S2 state, as a consequence of the single laser flash turnover. The post-flash decay reflected S2 → S1 back-turnover, as confirmed by their correlations with independent measurements of S2 multiline EPR signal and flash-induced variable fluorescence decay kinetics under corresponding experimental conditions. This flash-induced effect opens up the possibility to study the kinetic behaviour of S-state transitions at room temperature using YD• as a probe. 相似文献
