IL-13是新近发现的一种新的细胞因子,由Th2细胞产生,人IL-13(hIL-13)的分子量为17kd(糖量化)和12.4kD( 非糖基化);鼠IL-13(mIL-13)的分子量为14kD;二者的氨基酸序列同源性为58%,hIL-13和mIL-13的基因定位于人第5号染色体和鼠第11号染色体上,并与IL-4基因紧密连锁,二者的cDNA同源性为66%,重组IL-13(γIL-13)具有多种生物学活性刺激前髓样细胞增殖,诱导单核细胞表达MHC II类抗原和CD23,促进B细胞增殖,分化和分泌Ig,抑制炎症因子的产生和HIV-1的复制,抗肿瘤等,对其深入研究将有助于认识IL-13在炎症反应的发生和发展以及调节免疫应答方面的作用,并为其临床应用的可能性提供理论依据。 相似文献

Summary Internal motions play an important role in the biological function of proteins and NMR relaxation studies may characterize them over a wide range of frequencies. An experimental pulse scheme is proposed for the measurement of the 13CO-13C cross-relaxation rate. For sensitivity reasons, this measurement is performed in an indirect manner through several coherence transfer steps, which should thus be calibrated independently. Contributions of other relaxation pathways can be eliminated by the determination of the initial slope of the buildup curve. The cross-relaxation rates have been determined on a 15N-/13C-labelled 116-residue protein and the significant variations along the sequence have been interpreted as evidence of an increased amount of fast local motion. 相似文献

Human studies using Abs to two different, nonoverlapping epitopes of IL-13 suggested that epitope specificity can have a clinically significant impact on clearance of IL-13. We propose that Ab modulation of IL-13 interaction with IL-13Rα2 underlies this effect. Two Abs were administered to healthy subjects and mild asthmatics in separate dose-ranging studies and allergen-challenge studies. IMA-638 allows IL-13 interaction with IL-13Rα1 or IL-13Rα2 but blocks recruitment of IL-4Rα to the IL-13/IL-13Rα1 complex, whereas IMA-026 competes with IL-13 interaction with IL-13Rα1 and IL-13Rα2. We found ~10-fold higher circulating titer of captured IL-13 in subjects treated with IMA-026 compared with those administered IMA-638. To understand how this difference could be related to epitope, we asked whether either Ab affects IL-13 internalization through cell surface IL-13Rα2. Humans inducibly express cell surface IL-13Rα2 but lack the soluble form that regulates IL-13 responses in mice. Cells with high IL-13Rα2 expression rapidly and efficiently depleted extracellular IL-13, and this activity persisted in the presence of IMA-638 but not IMA-026. The potency and efficiency of this clearance pathway suggest that cell surface IL-13Rα2 acts as a scavenger for IL-13. These findings could have important implications for the design and characterization of IL-13 antagonists. 相似文献

Summary
13C-13CO homonuclear NOE and 13CO T1 relaxation were measured for a 20 kDa protein using tripleresonance pulse sequences. The experiments were sufficiently sensitive to obtain statistically significant differences in relaxation parameters over the molecule. The 13C-13CO cross-relaxation rate, obtained from these data, is directly proportional to an order parameter describing local motion and it is largely independent of the local correlation time. It is therefore a relatively straightforward observable for the identification of local dynamics. 相似文献

HNCO-based 3D pulse schemes are presented for measuring 1HN-15N,15N-13CO, 1HN-13CO,13CO-13C and 1HN-13C dipolar couplings in 15N,13C,2-labeled proteins. The experiments are based on recently developed TROSY methodology for improving spectral resolution and sensitivity. Data sets recorded on a complex of Val, Leu, Ile (1 only) methyl protonated 15N,13C,2H-labeled maltose binding protein and -cyclodextrin as well as 15N,13C,2H-labeled human carbonic anhydrase II demonstrate that precise dipolar couplings can be obtained on proteins in the 30–40 kDa molecular weight range. These couplings will serve as powerful restraints for obtaining global folds of highly deuterated proteins. 相似文献

目的:探讨Goodpasture综合征的临床特征及诊治要点.方法:对13例Goodpasture综合征患者的临床表现、实验室检查、诊断与治疗等资料进行回顾性分析.结果:13例患者中,全部患者(100%)均有咯血,12例(92.3%)有镜下血尿,大多数患者咯血症状先于肾小球肾炎症状出现.10例(83.3%)抗GBM抗体阳性.12例接受完整治疗方案的患者,有效7例,有效率58.3%.无效死亡5例,平均生存期为(91± 100.1)月.结论:Goodpasture综合征是一组病变进展迅速,预后凶险的自身免疫性疾病,显效组在确诊天数上与有效组与死亡组间的差别具有统计学意义(P均<0.05),改善疾病预后的关键是早发现、早诊断、早治疗. 相似文献

T-钙黏蛋白(T-Cadherin/H-Cadherin,CDH13)是非典型的钙黏素家族中的一员。研究发现,CDH13广泛分布于正常细胞表面,而在肺癌、乳腺癌、宫颈癌等多种恶性肿瘤中表达下调,并证实与肿瘤的不良预后有关。在肿瘤中CDH13的再表达可抑制肿瘤细胞的增殖与侵袭。这些特性表明,CDH13可能成为恶性肿瘤风险评估和监测预后的新的分子生物学标志。现就近年来CDH13在恶性肿瘤中的研究进展进行综述。 相似文献
