Summary The osmotic effect of Polyethylene glycol (PEG) has been shown to be sufficient to induce the germination of Pistacia vera L. pollen in liquid medium. The prehydration of the pollen in a saturated atmosphere for approximately 10 h was necessary to obtain maximum in vitro germination. Imbibition of the pollen in water resulted in the rapid leakage of solutes into the medium. These solutes consisted of approximately 50% carbohydrates, of which sucrose (0.65 mol/mg), glucose (0.77 mol/mg) and fructose (0.78 mol/mg) were the major sugars; the remaining 50% comprised proteins with the following major molecular weights 63 kDa, 60 kDa, 59 kDa, 40 kDa, 36 kDa, 35.5 kDa, 31 kDa, other organic matter and minerals.  相似文献

The nucellar beak is a proboscis-like outgrowth of the nucellus at the micropylar end, being the obligatory path for the pollen tube entering the ovule. Among the few angiosperm families with nucellar beak, Cucurbitaceae is remarkable because the pollen tube may develop at least two types of growth within the nucellar beak: tubular and ampulliform. Wondering about the possibility that Cucurbitaceae ovules may express some histological variation that could be related to pollen tube growth within the nucellar beak, we performed a compared anatomical and histochemical study of the nucellar beak and the pollen tube growth of ten species of Cucurbitaceae. Results show that Cucurbitaceae ovules are diverse in size and proportions (of integuments, nucellar body, and nucellar beak), and they have at least four types of nucellar beak histology: pectic-tracked, secretory-like, amylaceous, and mixed. Amylaceous and mixed nucellar beaks are related to the ampulliform growth of the pollen tube, which could have appeared independently in most derived tribes of Cucurbitaceae, although information about nucellar beak structure in the basal tribes is still needed. In addition, the understanding of the relation between amylaceous nucellar beaks and the ampulliform growth of the pollen tube, whose function is still to be discovered, might open the possibility of a unique model of pollen tube-ovule co-evolution in angiosperms.  相似文献

In the present study, antibacterial, antifungal, and antiviral properties of 15 lipohylic extracts obtained from different parts (leaf, branch, stem, kernel, shell skins, seeds) of Pistacia vera were screened against both standard and the isolated strains of Escherichia coli, Pseudomonas aeruginosa, Enterococcus faecalis, Staphylococcus aureus, Candida albicans and C. parapsilosis by microdilution method. Both Herpes simplex (DNA) and Parainfluenza viruses (RNA) were used for the determination of antiviral activity of the P. vera extracts by using Vero cell line. Ampicilline, ofloxocine, ketoconazole, fluconazole, acyclovir and oseltamivir were used as the control agents. The extracts showed little antibacterial activity between the range of 128-256 microg/ml concentrations whereas they had noticeable antifungal activity at the same concentrations. Kernel and seed extracts showed significant antiviral activity compared to the rest of the extracts as well as the controls.  相似文献

We used Randomly Amplified Polymorphic DNA (RAPD) markers to examine patterns of relatedness among 29 pistachio (Pistacia vera L.) cultivars and accessions. These included 13 cultivars that we had previously described, and an additional 16 items from the USDA National Clonal Germplasm Repository/Davis comprising cultivars and land races originating further east of the cultivars described previously, and material from wild P. vera stands in or near the putative center of origin for pistachio in South Central Asia. The results show high levels of polymorphism in the species emphasizing the importance of preservation of the remaining wild stands of P. vera. Analyses support the concept that cultivars in use west of the Zagros-Caucasus ranges likely originate from a limited germplasm base. The newly examined cultivated material shows greater genetic diversity, consistent with the hypothesis that pistachio cultivation originated in or near South Central Asia. Results also indicate that for at least two cases, material identified differently in two collections are the same clones, thus illustrating the value of molecular marker techniques in describing and maintaining germplasm collections for clonally propagated species.
Résumé  En este trabajo se ha empleado la técnica del ADN polimórfico amplificado al azar (RAPD) para examinar la similitud genética entre 29 cultivares y accesiones de pistachero (Pistacia vera L). Este material incluye 13 cultivares considerados en un trabajo anterior y 16 nuevas accesiones del Banco Nacional de Germoplasma Clonal del USDA en Davis que incluyen cultivares y razas locales de regiones más orientales que las consideradas anteriormente así como material procedente de basques naturales de P. vera situados en las proximidades del presunto centro de origen del pistachero. Los resultados obtenidos muestran un alto grado de polimorfismo, lo que indica la necesidad de conservar el germoplasma de P. vera todavía existente en estado natural. Además se confirma la hipótesis de que los cultivares procedentes del oeste de la zona Caucásica y del Zagros se originaron a partir de una base genética limitada. El nuevo material estudiado muestra una mayor diversidad genética lo que corrobora la idea de que el cultivo del pistachero se inició en Asia Central. Al menos en dos de los casos estudiados, el material identificado en dos colecciones diferentes como distintos genotipos, en realidad se trata del mismo clon, lo que demuestra la utilidad de los marcadores moleculares en la descripción y mantenimiento de colecciones de germoplasma en especies de reproducción vegetativa.

No differences have been observed in vivo between Lycopersicum peruvianum compatible and incompatible pollen during activation and pollen tube emission and organization, that is until 4 h and 30 min after pollination. During pollen activation the main events are the setting free of rough endoplasmic reticulum (RER) cisterns which were stacked in the mature pollen, the increase in the number of polysomes, and a great activity of the dictyosomes. Immediately after germination of the vegetative nucleus and the generative cell move into the tube, the generative cell diviting to form the male gametes; the tube then becomes organized in four zones. This series of changes is similar to what has already been observed in vitro except that in vitro the generative cell remains undivided and the whole process from seeding to tube organization takes 3 h instead of 4 h and 30 min after pollination, as it does in vivo. Pollen tube dynamics following different competition regimes were studied in sweet cherry (Prunus avium L.). In the process from pollination to fertilization, a constant reduction in the number of pollen tubes that travel along the style is observed. There could be two main causes of this reduction. One is a physical or physiological constraint consisting of the progressive decrease in the reserves and space available for pollen tube growth along the transmitting tissue of the style, and the other is genetic interaction both among the male gametophytes and between the male gametophytes and the female tissues of the flower. To evaluate the roles that these two forces play in reducing the number of pollen tubes that travel along the style, pistils were subjected to various pollen competition regimes by applying different mixtures of live and dead pollen onto the stigmata. The results obtained were similar when the experiment was repeated with different genotypes over 2 years, both in the laboratory and in the field. The role of stylar constriction is important, but it is not the only cause of pollen tube attrition because with low pollen loads fewer pollen tubes reach the different parts of the style than could fit therein. The fact that under different pollen competition regimes the number of pollen tubes is reduced by the same proportion in each stylar level indicates that genetic interactions play an important role in the control of pollen tube attrition.  相似文献

The class III pistil-specific extensin-like proteins (PELPIII) of Nicotiana tabacum accumulate in the intercellular matrix (IM) of the style transmitting tissue (TT). After pollination, the 110–140 kDa PELPIII is translocated from the IM into the pollen tube walls. PELPIII-like sequences have been found in several solanaceous species. These sequences are expressed in mature non-pollinated styles at both RNA and protein level. Of the genus Nicotiana, the species N. alata, N. x sanderae and N. sylvestris (section Alatae), and N. tomentosiformis and N. otophora (section Tomentosae) showed an expression level of PELPIII homologues similar to that in mature styles of N. tabacum. PELPIII genes were absent in the most ancient species studied, namely N. trigonophylla (section Trigonophyllae). To study the species dependence of the translocation of PELPIII into the pollen tube wall in tobacco, interspecific pollinations on N. tabacum pistils were carried out with pollen from the incongruous species N. rustica, N. trigonophylla and Petunia hybrida, where PELPIII homologues are absent in the style. Immunocytological tests showed that the N. tabacum PELPIII is translocated into the pollen tube walls of all three species. Thus, the pollen tube walls of these species do not form a barrier for IM compounds such as the 110–140 kDa PELPIII and the absence of any possible effect of PELPIII on pollen tube growth cannot be due to failure of PELPIII transport through the wall. The importance of these findings is discussed with respect to the evolutionary origin of PELPIII, the pollen pistil interaction, the function of style TT-specific proteins and the physical properties of pollen tube walls.  相似文献

This study investigates how microtubules and microfilaments control organelle motility within the tips of conifer pollen tubes. Organelles in the 30-m-long clear zone at the tip of Picea abies (L.) Karst. (Pinaceae) pollen tubes move in a fountain pattern. Within the center of the tube, organelles move into the tip along clearly defined paths, move randomly at the apex, and then move away from the tip beneath the plasma membrane. This pattern coincides with microtubule and microfilament organization and is the opposite of the reverse fountain seen in angiosperm pollen tubes. Application of latrunculin B, which disrupts microfilaments, completely stops growth and reduces organelle motility to Brownian motion. The clear zone at the tip remains intact but fills with thin tubules of endoplasmic reticulum. Applications of amiprophosmethyl, propyzamide or oryzalin, which all disrupt microtubules, stop growth, alter organelle motility within the tip, and alter the organization of actin microfilaments. Amiprophosmethyl inhibits organelle streaming and collapses the clear zone of vesicles at the extreme tip together with the disruption of microfilaments leading into the tip, leaving the plasma membrane intact. Propyzamide and oryzalin cause the accumulation of membrane tubules or vacuoles in the tip that reverse direction and stream in a reverse fountain. The microtubule disruption caused by propyzamide and oryzalin also reorganizes microfilaments from a fibrillar network into pronounced bundles in the tip cytoplasm. Summary Cytochalasins B and D in the concentration range 0.5–10 g ml–1 produced similar effects on growth, movement and cytoplasmic structure in the pollen tubes of Iris spp. cultured in vitro. Continuous video recording showed that at 5 g ml–1, CB was capable of stopping organelle circulation in as short a period as 20 s. The usually elongated vegetative nuclei were also arrested, and subsequently contracted irregularly. Generative cells were not radically changed in shape, but occasionally moved erratically before being halted. Detailed examination of CD- and CB-treated tubes regarded as being capable of recovering growth upon transfer to normal medium revealed several characteristic effects on cytoplasmic structure. Fibrils presumed to consist of, or contain, microfilament bundles are readily visible in the older parts of the living tube where they form the pathways of organelle movement; these were either condensed into amorphous columns or fragmented by treatment. In the distal parts of the tube, the cytoplasm had contracted into amorphous masses which continued to show very slow shape changes. With the arrest of extension growth, pectin accumulated over the tube tip and in patches along the flanks. In a medium containing 1 mM ATP, recovery from treatment was achieved in some instances within l min. Organelle circulation in the younger tubes was resumed, and fresh adventive tube tips were formed. The fibrillar system of the older tubes was not restored, however; instead, the cytoplasm in these zones formed aggregates which underwent continuous amoeboid movement, the organelles within moving rapidly in irregular trajectories with no indication of the resumption of the original long-range cyclotic flux. Summary The addition of DAPI particles to the stigma exudate of Gasteria results in the labelling of the pollen nuclei. By means of epifluorescence microscopy and clearing of the ovule, the labelled nuclei of the sperm cells and, subsequently, the zygotic nucleus can be observed. The method was used in a cross-pollination with low seed setting to examine different types of penetration and transport of the pollen tube nucleus and sperm cell nuclei. More than one pollen tube can penetrate, but generally only one set of sperm cell nuclei is accepted.  相似文献

The pollen tube of Prunus avium (cherry) consists of a growth zone of vesicles at the tip and an assemblage of organelles typical of an actively metabolizing cell. Electron opaque globules are closely associated with the plasma membrane and fibrillar cell wall layer at the tip. Morphologic changes occurring during pollen grain activation and ultrastructural features of Lycopersicum peruvianum Mill. pollen tube during the first stages of growth in vitro have been studied. The more evident morphologic changes during activation, in comparison to those already described for mature inactive pollen, concern dictyosomes, rough endoplasmic reticulum (RER), and ribosomes. The dictyosomes are very abundant and produce large and small vesicles. Near the germinative pores both types of vesicles are present, while all along the remaining cell wall only the large type is observed. These latter react weakly to Thiéry's test and probably contain a callose precursor necessary for the deposition of a callosic layer lining at first only the inner side of the functioning pore and occasionally the other two pores, and subsequently the entire inner surface of the cell wall. The small vesicles, highly positive to Thiéry's test, are present only near the pores and could be involved in the formation of the pectocellulosic layer of the tube wall. The setting free of RER cisterns, which in the mature inactive pollen were aggregated in stacks, coinciding with polysome formation and resumption of protein synthesis, is in accord with the hypothesized role of RER cistern stacks as a reserve of synthesizing machinery. The pollen tube reaches a definitive spatial arrangement soon after the generative cell and vegetative nucleus have moved into it. At this stage four different zones that reflect a functional specialization are present. In the apical and subapical zone two types of dictysosome-originated vesicles, similar to those found in the activated pollen grain, are present. Summary Ultrastructural studies made on the micropyle of sunflower before and after pollination resulted in the following observations. (1) The micropyle is closed instead of a hole or canal. The inner epidermis of the integument on both sides of the micropyle is in close contact at the apex of the ovule. The boundary between the two sides consists of two layers of epidermal cuticle. (2) The micropyle contains a transmitting tissue. The micropyle is composed of an intercellular matrix produced by the epidermal cells of the integument. (3) The micropyle is asymmetrical, and is much wider on the side proximal to the funicle. On the funicle side the cells adjacent to the micropyle are similar to those of the transmitting tissue: they have large amounts of intercellular matrix and contain abundant dictyosomes, rough ER, and starch grains, and provide an appropriate environment for growth of the pollen tubes. The cells distal to the funicle are rich in rough ER and lipid bodies; they lack large intercellular spaces. (4) The micropyle is variable in the axial direction, i.e., it is much larger and more asymmetric at the level distal to the embryo sac than at a level close to the embryo sac. After pollination, one to four pollen tubes are seen in a micropyle. During their passage through the micropyle, most pollen tubes are restricted to the side proximal to the funicle. There is a greater tendency (81%) for the degenerate synergid to be located toward the funicle, i.e., at the same side as the pollen tube pathway. The data indicate a close relationship between micropyle organization, orientation of pollen tube growth, and synergid degeneration.  相似文献

Summary Microtubules inNicotiana tabacum pollen tube subprotoplasts reassembled in wave-like to concentric cortical arrays. Crosslinks between microtubules were either 15 or 80 nm in length. Cortical actin filaments showed different distributions; no colocalization like that in pollen tubes was observed. Degradation of actin filaments by cytochalasin D had no influence on microtubule organization. Degradation of microtubules and/or actin filaments did not affect outgrowth of the subprotoplasts. Organization of the microtubules occurred independent of the presence of the generative cell and/or the vegetative nucleus. Summary When pollen of Impatiens glandulifera was cultured in aerated liquid medium for 1 h, 70% of the pollen grains germinated; these attained an average tube length of 1 mm. Subsequently, these aerobic growth conditions were changed to anaerobic by substituting a nitrogen inlet for the air inlet. As a result, the pollen tubes stopped elongating and burst. The ultrastructural changes which occurred upon inducing anoxia were studied with samples taken at 0 s, 45 s, and 4 min after changing the gas. Anoxia caused rapid and considerable changes in the ultrastructure of the dictyosome vesicles involved in cell wall formation. There was an increase in the osmiophyly of the vesicle content, and the presence of fibrillar material became apparent. Simultaneously, the fusion behavior of the dictyosome vesicles changed. Instead of the normal fusion of the dictyosome vesicles with the plasma membrane, there was a premature fusion of the vesicles with each other inside the cytoplasm that resulted in the formation of aggregates. Furthermore, the cell wall precursors that were excreted were not incorporated in their usual configuration into the growing cell wall. Instead of a smooth inner cell wall surface, irregular thickenings were formed.  相似文献

Summary Continuous observation of organelles and other cytoplasmic inclusions in the older stretches of living pollen tubes of Iris pseudacorus shows that in the more attentuated parts of the protoplast they move along single, mainly longitudinally oriented fibrils, corresponding to those previously isolated from other species and shown to contain bundles of uniformly polarised actin microfilaments. The traffic associated with each fibril is unidirectional, but organelles move along them independently, sometimes with conspicuously different velocities. Larger columns of cytoplasm passing along the tube are associated with several such fibrils, as revealed in occasional discontinuities and also in columns isolated from the tube in suitable medium without fixation. The dimensions of the individual fibrils suggest that the bundles of actin microfilaments are not likely to be enclosed in a unit membrane corresponding to a tonoplast. If so, the nature of the continuous cavities traversed by numerous fibrils in the older parts of the pollen tube requires reappraisal, since these are more likely to be volumes of attentuated cytoplasm comparable with that of the central cavity of the sieve tube than vacuoles of the normal plant-cell type.  相似文献

Sperm delivery in flowering plants requires extensive pollen tube growth through the female sporophytic tissues of the pistil. The apical cell wall emerges as a central player in the control of pollen tube growth, since it provides strength to withstand the internal turgor pressure, while imparting sufficient plasticity to allow cell wall extension through the incorporation of new membrane and wall material. Within this scenario, pectin methylesterases (PMEs; EC 3.1.1.11) emerge as crucial regulators in determining the mechanical properties of pectins, the major component of the apical pollen tube wall. We previously identified NtPPME1, a pollen specific PME from Nicotiana tabacum. Here we show that silencing of NtPPME1 results in a mild but significant decrease of in vivo pollen tube growth while the overall PME activity in pollen is not significantly affected. Although the precise mechanisms responsible for the observed phenotype are not known, it seems likely that the cell must maintain a closely regulated level of PME activity in order to maintain the equilibrium between strength and plasticity in the apical cell wall. A relatively minor disturbance of this equilibrium, as caused by NtPPME1 silencing, compromises pollen tube growth.  相似文献

Background

Pistachio (Pistacia vera L.) is a dioecious species that has a long juvenility period. Therefore, development of marker-assisted selection (MAS) techniques would greatly facilitate pistachio cultivar-breeding programs. The sex determination mechanism is presently unknown in pistachio. The generation of sex-linked markers is likely to reduce time, labor, and costs associated with breeding programs, and will help to clarify the sex determination system in pistachio.

Results

Restriction site-associated DNA (RAD) markers were used to identify sex-linked markers and to elucidate the sex determination system in pistachio. Eight male and eight female F1 progenies from a Pistacia vera L. Siirt × Bağyolu cross, along with the parents, were subjected to RAD sequencing in two lanes of a Hi-Seq 2000 sequencing platform. This generated 449 million reads, comprising approximately 37.7 Gb of sequences. There were 33,757 polymorphic single nucleotide polymorphism (SNP) loci between the parents. Thirty-eight of these, from 28 RAD reads, were detected as putative sex-associated loci in pistachio. Validation was performed by SNaPshot analysis in 42 mature F1 progenies and in 124 cultivars and genotypes in a germplasm collection. Eight loci could distinguish sex with 100% accuracy in pistachio. To ascertain cost-effective application of markers in a breeding program, high-resolution melting (HRM) analysis was performed; four markers were found to perfectly separate sexes in pistachio. Because of the female heterogamety in all candidate SNP loci, we report for the first time that pistachio has a ZZ/ZW sex determination system. As the reported female-to-male segregation ratio is 1:1 in all known segregating populations and there is no previous report of super-female genotypes or female heteromorphic chromosomes in pistachio, it appears that the WW genotype is not viable.

Conclusion

Sex-linked SNP markers were identified and validated in a large germplasm and proved their suitability for MAS in pistachio. HRM analysis successfully validated the sex-linked markers for MAS. For the first time in dioecious pistachio, a female heterogamety ZW/ZZ sex determination system is suggested.  相似文献
