Polyribosomes have been isolated from tobacco leaves. Upon infection with TMV, a new polyribosome has appeared which is specific for infection, and TMV-antigenic protein is formed on this polyribosome. This new polyribosome has an S-value of 360–380. From these results, it is suggested that this TMV specific polyribosome is an aggregate of 60–80 ribosomes which is bound by TMV-RNA as messenger, and that TMV-RNA is translated polycistronically.  相似文献

烟草花叶病毒对烟草叶片光合特征和POD表达的影响   总被引:2,自引:0,他引:2  
以烤烟(Nicotiana tabacum L.)品种'中烟5号'为实验材料,对烟草健康株与感染烟草花叶病毒(TMV)株的叶绿素、光合速率、光合速率对光强的响应曲线、光暗反应荧光特征、POD活性及其表达等进行研究,以探讨TMV感染对烟草植株生理生态特征的影响.结果显示:病株的叶绿素a(Chl a)和叶绿素b(Chl b)含量显著低于健康株,但Chl a/Chl b值基本相同;病株暗中初始荧光(F0)、暗中最大荧光(Fm)、暗中可变荧光(Fv)、光下初始荧光(F0′)、光下最大荧光(Fm′)、光下可变荧光(Fv′)、非光化学猝灭系数(NPQ)、PSⅡ捕光效率(Fv′/Fm′)、PSⅡ实际光化学效率(ФPSⅡ)及光饱和点显著低于健康株;净光合速率在光强较大(>1 500 μmol·m-2·s-1)时病株比健康株低,光强适中(1 500 μmol·m-2·s-1左右)时两者相差不大,光强较弱(<1 500 μmol·m-2·s-1左右)时病株比健康株高;病株叶片的过氧化物酶(POD)活性显著升高,POD同工酶中一些大分子量蛋白分子表达量加大.研究表明,感染TMV使烟草植株对光抑制更为敏感,叶片的荧光激发能力和热耗散能力下降,PSⅡ反应中心捕光效率和光化学反应效率降低,光合电子传递能力和碳同化能力受到抑制;POD活性提高和表达量增加可能是诱导烟草抗病性的一个关键生理过程.  相似文献

The low molecular weight tobacco mosaic virus (TMV)-specific RNA component (LMC) was demonstrated in tobacco mesophyll protoplasts by polyacrylamide gel electrophoresis of 14C-uridine-labelled RNA from infected protoplasts. Free and membrane-bound polysomes were isolated from infected protoplasts, and RNA extracted from them was analyzed. TMV-specific RNA species including full-length viral RNA, its replicative intermediate, and LMC were found in both free and membrane-bound polysomes, but were present in free polysomes in much larger amounts. In particular, as much as 37 % of total LMC in protoplasts was found in free polysomes. Fractionation of polysomes by sedimentation in sucrose gradients showed that LMC is associated with small-sized polysomes (mono- to tetrasomes). Polysomes of this size class produced viral coat protein in a cell-free protein synthesizing system from rabbit reticulocytes. On the other hand, full-length TMV-RNA was associated predominantly with larger polysomes which produced in the cell-free system TMV-specific high molecular weight polypeptides but no coat protein. These results indicated that LMC, a subgenomic RNA of TMV, in fact functions in vivo as messenger RNA for viral coat protein, as has been postulated on the basis of in vitro studies.  相似文献

Synthesis of tobacco mosaic virus (TMV)-specific low molecular weight component RNA (LMC) was investigated in relation to that of other TMV-related RNAs and proteins, and formation of progeny virus particles using synchronously infected tobacco mesophyll protoplasts. Timing of LMC synthesis was shown to be almost the same as, but somewhat earlier than that of TMV-RNA synthesis. In contrast, synthesis of TMV-specific double-stranded RNAs (replicative intermediate, RI and replicative form, RF) as well as a high molecular weight virus-induced protein (140 K protein) showed the maximum incorporation rate 4–6 h earlier than LMC synthesis. While, synthesis of coat protein and formation of progeny virus particles lagged behind LMC synthesis for 6–8 h. LMC occurring in polysomes was also investigated during the course of virus replication. The amount of produced coat protein calculated theoretically from the amount of LMC in polysomes of infected protoplasts was shown to be well agreed with the experimental results, indicating that LMC in polysomes is actively functioning as messenger for coat protien synthesis in vivo.  相似文献

用原生质体法制备出高纯度的完整叶绿体经SDS-PAGE电泳,银染后,发现黄瓜花叶病毒(CMV)侵染的烟草病叶叶绿体蛋白质图谱和健叶叶绿体相比,多出一条染色较弱的迁移率与CMV衣壳蛋白质相同的带,经Western转移,用CMV游离衣壳蛋白亚基抗血清进行斑点酶联(Immunoblot)检测,证明这条带就是CMV衣壳蛋白质。健康叶绿体中加入去掉叶绿体的病叶汁液而制备出的叶绿体中无CMV衣壳蛋白质,说明这不是在叶绿体提纯过程中得到的假象,即衣壳蛋白质存在于被CMV侵染的完整叶片叶绿体中。这个结果否认了以往报道的CMV衣壳蛋白质不存在于烟草叶绿体中的结论。另外还发现,叶绿体中的衣壳蛋白质浓度与叶片症状严重程度呈正相关。  相似文献

The dielectric properties of the Tobacco Mosaic Virus (TMV) have been measured using time domain dielectric spectroscopy (TDDS) in the temperature range from 1 to 40 degrees C. A single dielectric dispersion is observed in the MHz range. The activation energy of the process is found to be in the range 1-2 kcal/mol. The experimental data could not be completely accounted for by current theoretical models, but evidence indicates that the dielectric loss arises from polarisation of charge on and around the virus.  相似文献

The ultrastructural aheration of two host plants infected with tomato mosaic virus (ToMV) were studies with transmission electron microscopy. A large number of virus particles were found being accumulated in different cells such as epidermis, parenchyma cells and vascular bundle cells of Lycopersicon esculentum Mill. grown at 25℃ Crystalline inclusions and paracrystal inclusions composed of ToMV particles were observed in the cytoplasm or vacuoles. Some muhivesicular bodies and myeloid bodies protming into the vacuole and vires-specific vesicles associated with the tonoplast were also observed. The ultrastructuml alteration of Nicotiana tabacum L. tv. Xanthinn was similar to that in tomato infected by ToMV grown at 25 cE. In addition to the aggregate inclusions described above, some cytoplasmic angularly-layered aggregates and abnormal chloroplasts with small peripheral vesicles were observed in the parenchyma cells. The densely stained amorphous material was seen in the cytoplasm of N. tabacum L. cv. Xanthiun grown at 35℃. No X- body was observed in the cytoplasm of the ToMV infected tomato and tobacco grown at 25℃ or 35℃. The authors' results suggest a significant difference between the cytopathological effects of ToMV and tobacco mosaic virus (TMV). These characteristic difference may be useful in the virus diagnosis and identification virus infections in plants.  相似文献

Effects of the superinfection with tobacco mosaic virus (TMV) on susceptible tobacco plants infected with potato virus Y (PVY) were determined. Dynamic changes in the TMV and/or PVY contents, the ribonucleases (RNases), the phosphomonoesterase (PME), the phosphodiesterase (PDE) and the glucose-6-phosphate dehydrogenase (G6P DH) activities were studied. The PVY infection caused a substantial reduction in the multiplication of TMV. The content of TMV in the PVY inoculated leaves amounts to 6 and 9 % in the PVY systemically infected leaves when compared with single TMV. Surprisingly, the challenging virus (TMV) enhanced the content of inducing virus (PVY) in the locally inoculated leaves up to 130 – 141 %. In contrast, the reduction of PVY content down to 35 – 40 % by TMV was seen in the PVY systemically infected leaves. The activities of the RNase, the PME, the PDE and the G6P DH were increased (when compared with the healthy plants) during the acute phase of single virus multiplication (PVY or TMV). The increase in the activities of the enzymes in the leaves with mixed infection was at least as high as the sum of the increases of single infections. Moreover, a higher increase than the sum was seen for G6P DH and PDE (by about 20 – 35 %). This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The effect of viral infections on the Crassulacean Acid Metabohsm (CAM) was determined in two orchid species. Plants of a Sophrolaeliocattleya (SLC) commercial hybrid were inoculated with either the orchid strain of the Tabacco Mosaic Virus (TMV-O), with Cymbidium Mosaic Virus (CybMV) or with a mixture of both viruses. Plants of Epidendrum elongatum, a wild terrestrial orchid, were inoculated either with TMV-O or with TMV-O and CybMV simultaneously. Virus infected plants started to show a systemic mosaic on the leaves 3 to 4 years after inoculation, the symptoms being more appreciable in SLC than in E. elongatum. Double inoculation with TMV-O and CybMV proved to the lethal for the SLC plants. TMV-O infection caused 58 % and 24 % reduction in nocturnal leaf titratable acidity (TA) in SLC and E. elongatum, respectively. TMV-O also changed the daily pattern of leaf nonstructural carbohydrates typical of CAM plants. A 50 % decrease in TA at dawn and a mid-morning peak of sugar accumulation were measured in leaves of doubly inoculated E. elongatum plants. In SLC CybMV infection inhibited CAM and induced an accumulation of glucans in the leaves. Cytoplasmic agglomerations of CybMV, but not of TMV-O, were detected in SLC mfected cells, whereas TMV-O particles were clearly observed in the cytoplasm of E. elongatum. Infection by TMV-O or CybMV in SLC caused an increase in chloroplasts volume and distortion of the grana due to high glucan accumulation. In contrast, TMV-O infection in E. elongatum induced a lesser degree of damage in the cell ultrastructure.  相似文献
