植物生物质是地球上最丰富的可再生资源,对其生物炼制可生产高附加值的生物基产品。生物炼制需要使用植物多糖降解酶(plant-polysaccharide-degrading enzymes,PPDEs),如纤维素酶、木聚糖酶和生淀粉酶。丝状真菌草酸青霉(Penicillium oxalicum)能分泌完整的具有高活力的植物多糖降解酶,但其产量低限制了大规模生产及应用。草酸青霉中植物多糖降解酶的生物合成受到多种调控因子包括转录因子的严格调控。本文主要介绍在以植物生物质甘蔗渣和木薯生淀粉为原料的生物炼制中,涉及的一些关键微生物方面的问题,如从高产植物多糖降解酶的真菌菌株的筛选、育种,到草酸青霉植物多糖降解酶合成及其基因表达的调控基因的鉴定,以及酶产量提高的工程菌株的构建等,为丝状真菌资源的开发与利用提供理论指导。  相似文献

Penicillium oxalicum strain 212 (PO212) is an effective biocontrol agent (BCA) against a large number of economically important fungal plant pathogens. For successful registration as a BCA in Europe, PO212 must be accurately identified. In this report, we describe the use of classical genetic and molecular markers to characterize and identify PO212 in order to understand its ecological role in the environment or host. We successfully generated pyrimidine (pyr‐) auxotrophic mutants. In addition we also designed specific oligonucleotides for the pyrF gene at their untranslated regions for rapid and reliable identification and classification of strains of P. oxalicum and P. rubens, formerly P. chrysogenum. Using these DNA‐based technologies, we found that PO212 is a strain of P. rubens, and is not a strain of P. oxalicum. This work presents PO212 as the unique P. rubens strain to be described as a BCA and the information contained here serves for its registration and commercialization in Europe.  相似文献

The potential of the biological control fungus Penicillium oxalicum to suppress wilt caused by Fusarium oxysporum f. sp. melonis and F. oxysporum f. sp. niveum on melon and watermelon, respectively, was tested under different growth conditions. The area under disease progress curve of F. oxysporum f. sp. melonis infected melon plants was significantly reduced in growth chamber and field experiments. In glasshouse experiments, it was necessary to apply P. oxalicum and dazomet in order to reduce Fusarium wilt severity in melons caused by F. oxysporum f. sp. melonis. For watermelons, we found that P. oxalicum alone reduced the area under the disease progress curve by 58% in the growth chamber experiments and 54% in the glasshouse experiments. From these results, we suggested that P. oxalicum may be effective for the management of Fusarium wilt in melon and watermelon plants.  相似文献

This work examines the efficacy of radiation in reducing the viability of certain contaminating bacteria of sugar-cane must and the consequential beneficial effect of lethal doses of radiation on some physiological parameters of the yeast-based ethanolic fermentation. The must from sugar-cane juice was inoculated with different bacteria that usually contaminate the must in the production facilities: Bacillus and Lactobacillus. The contaminated must was irradiated at 2.0, 4.0, 6.0, 8.0 and 10.0 kGy of gamma radiation. The population density of the bacteria in the irradiated must was recorded. Ethanolic fermentation by yeast (Saccharomyces cerevisiae) was carried out and the total acidity, the volatile acidity and the organic acids (lactic and acetic) produced during the fermentation were determined. The ethanol yield was also recorded. The treatment with radiation reduced the population of the contaminating microorganisms of the sugar-cane must. The acidity and the organic acids (lactic and acetic) produced during the fermentation decreased as the dose of radiation applied to the must increased. It is concluded that irradiation was efficient in decontaminating the sugar-cane must and improved the biochemical parameters of the ethanolic fermentation, including the ethanol yield by 2%.  相似文献

A species ofMycosphaerella with aPseudocercospora anamorph was collected on overwintered fallen leaves of Japanese beech,Fagus crenata. Based on comparison of morphology withMycosphaerella species on Fagaceae, the fungus was newly described asMycosphaerella buna. ThePseudocercospora anamorph derived from a single ascospore of the fungus was morphologically identical to an endophytic anamorph isolated from asymptomatic living leaves of Japanese beech. We studied (a) the extent adhesion of Penicillium oxalicum conidia to tomato roots after application of P. oxalicum conidial formulations with or without stickers, (b) the relationship between the extent of conidial adhesion to roots and biocontrol of the conidial formulations against tomato wilt, and (c) colonisation of roots by P. oxalicum. Adhesion of P. oxalicum conidia to tomato roots occurred within the first minute of contact between the root and the conidial formulation and the bonding strength was sufficiently strong to prevent conidial removal from the roots. In addition, some formulations with stickers that increased conidial adhesion to roots improved the biocontrol of tomato wilt, when compared to that of formulations without stickers. A “dried conidia without stickers” with 0.025% Nu-Film 17 had no effect on the biocontrol of tomato wilt, despite good adherence of the conidia to the roots. The numbers of P. oxalicum conidia that adhered to the roots was constant for 60 days after application of a “dried conidia without stickers” conidial formulation. The significance of these results (speed of adhesion, number of adhered conidia, and variability of conidial external surface) are discussed in relation to the biocontrol success of tomato wilt using different types of conidial formulations with and without stickers.  相似文献

There is growing interest in research and development to develop novel tools to study, detect, and characterize microbes and their communities in industrial environments. However, knowledge about their validity in practical industrial use is still scarce. This review describes the advantages and limitations of traditional and molecular methods used for biofilm and/or planktonic cell studies, especially those performed with Listeria monocytogenes, Bacillus cereus, and/or Clostridium perfringens. In addition, the review addresses the importance of isolating the microorganisms from the industrial environment and the possibilities and future prospects for exploiting the described methods in the industrial environment.  相似文献

Summary The utility of spraying some known N2-fixing microorganisms on rice leaves grown both in N-less sand culture and under field conditions was examined. The effect was compared with that of spraying a phyllosphere N2-fixing isolate of Klebsiella, KUPBR2, and application of nitrogenous fertilizers. All the growth parameters studied including dry weight and N-content were enhanced. Under field conditions number of tillers was increased by 26% withKlebsiella pneumoniae M5al and by 65% with Aphanothece. The dry weight of the plants was enhanced by 61–119%. The yield per 10 m2 was almost doubled with Aphanothece, Beijerinckia 8007,Mycobacterium flavum, K. pneumoniae M5al and KUPBR2. The increases observed withStreptomyces sp. G12 though less spectacular was significant at 1% level with respect to several growth parameters.K. pneumoniae M5al,M. flavum andStreptomyces sp. G12 exhibited nitrogenase activity both in laboratory culture and in association with rice plants.  相似文献

The pathway of photosynthesis in sugar-cane, which gives most of the radio-activity fixed during short periods in 14CO2 in C-4 of oxaloacetate, malate and aspartate, was examined under varied conditions. 2. The pattern of labelling was essentially the same with leaves of different ages and with leaves equilibrated at carbon dioxide concentrations in the range 0–3·8% (v/v) and light-intensities in the range 1400–9000ft.-candles before adding 14CO2. 3. Radioactive products were examined after exposing leaves of 33 different plant species to 14CO2 for 4sec. under standard conditions. 4. A labelling pattern typical of sugar-cane was found in several species of Gramineae but not in others. Of 16 species from other Families only a species of Cyperaceae contained a large proportion of the fixed radioactivity in oxaloacetate, malate and aspartate.  相似文献

Dominant phylloplane fungi of guava (Psidium guajava L.) were screened for their antagonistic activities against the two pathogens,Colletotrichum gloeosporioides andPestalotia psidii, bothin vitro andin vivo. Culture filtrates ofAspergillus niger, Fusarium oxysporum andPenicillium citrinum caused more than 50% growth inhibition ofC. gloeosporioides. Filtrates ofCephalosporium roseo-griseum andF. oxysporum were most effective in reducing the growth ofP. psidii. Volatiles produced from the cultures ofA. niger, F. oxysporum, P. citrinum andP. oxalicum inhibited the growth ofC. gloeosporioides, whereas volatiles fromC. roseo-griseum, F. oxysporum andTrichoderma harzianum inhibited the growth ofP. psidii. The inhibitory effect of volatiles decreased with increase in incubation time. In general, the maximum effect of volatiles was noticed after 48 h incubation. Different grades of colony interactions in dual cultures were recognised between the two pathogens and the phylloplane fungi examined. Maximum inhibition ofC. gloeosporioides was caused byAureobasidium pullulans, Cladosporium cladosporioides, epicoccum purpurascens, F. oxysporum andMyrothecium roridum, whereasAspergillus terreus, C. roseo-griseum andP. oxalicum significantly reduced the growth ofP. psidii. Application of a spore suspension of each test fungus inhibited lesion development of guava leaves caused by the test pathogensin vitro. Inhibition was more pronounced when the spore concentration was increased.A. pullulans, C. cladosporioides, E. purpurascens, F. oxysporum, andT. harzianum were found to be strongly antagonistic toC. gloeosporioides. Summary The principles of a new process for the continuous manufacture of yoghurt withLactobacillus bulgaricus andStreptococcus thermophilus entrapped in Ca-alginate beads were given. Characteristics of non-optimized pH-stat continuous stirred tank reactors withL. bulgaricus andS. thermophilus entrapped in the same or separate particles were examined. In these reactors the highest production rate were 9.4 g.l–1.h–1 for lactic acid and 3.4×1011 C.F.U. 1–1.h–1 for inoculation when microorganisms were entrapped separately. A stable balance of the yoghurt bacteria liberated in continuously prefermented milk was observed.  相似文献

Plant Growth‐promoting Fungus (PGPF) Penicillium oxalicum was isolated from rhizosphere soil of pearl millet and was tested for its ability to promote growth and induce systemic resistance in pearl millet against downy mildew disease. The fungal isolate P. oxalicum UOM PGPF 16 was identified as P. oxalicum using ITS sequencing and morphological analysis and sequence was deposited at NCBI with accession number KF150220. Pearl millet susceptible seeds were treated with three different inducers (CS, CF and LCF) of PGPF P. oxalicum and all the inducers significantly reduced the downy mildew disease and enhanced plant growth. Among the inducers tested, CS treatment recorded highest seed germination of 91% and 1427 seedling vigour followed by LCF and CF treatments. The vegetative growth parameter and NPK uptake studies under greenhouse conditions revealed that the CS treatment of P. oxalicum remarkably enhanced the parameters tested when compared to control plants. A significant disease protection of 62% and 58% against downy mildew disease was observed in plants pretreated with CS of P. oxalicum under greenhouse and field conditions, respectively. The spatio‐temporal studies revealed that inducers P. oxalicum required a minimum of 3 days for developing maximum disease resistance which was maintained thereafter. The maximum Peroxidase (POX) activity (62.7 U) was observed at 24 h in seedlings treated with CS of PGPF P. oxalicum and the activity gradually reduced at later time points after pathogen inoculation. Chitinase (CHT) activity was significantly higher in inducer treated seedlings when compared to control seedlings inoculated with pathogen after 48 h and remained constant at all time points.  相似文献

Spirochetal microorganisms were isolated from femaleIxodes ricinus in Slovakia. Morphological, immunochemical and molecular biological analysis showed that the microorganism shared several common antigens withBorrelia species while other genetic traits were distinct and not related toBorrelia burgdorferi sensu lato. Lyme disease patient’s serum contained antibodies reacting with antigens of this microorganism. On the one hand the cross-reacting antigens represent a risk of false positive results in laboratory diagnostics, while on the other hand they have a certain potential for vaccine development against Lyme disease.  相似文献

Use of stream-centered dielectrophoresis (1–4) produced continuous separations on three cell mixtures (1)Chlorella vulgaris withNetrium digitus, (2)Ankistrodesmus falcatus withStaurastrum gracile, and (3)Saccharomyces cerevisiae withNetrium digitus. Maximal separations were obtained for these mixtures of live cells at 100 kHz, 600 kHz, and 2.0 MHz, respectively. The technique was restricted to a frequency range of 0.01–32 MHz, and to suspensions of low conductivity in which microorganisms such as these algae and yeast are tolerant. Extension, however, to cellular organisms requiring higher osmolarity is readily feasible through the use of nonionic solutes such as sucrose, mannose, glycine, etc.  相似文献

Progression in the understanding of the microecology of ambrosia beetles and their associated microorganisms is briefly reviewed. Between the 1840s and the early 1960s the concept of one ambrosial fungus per ambrosia beetle was emphasized. Some subsequent research has supported the view that each ambrosia beetle plus several associated microorganisms constitute a highly co-evolved symbiotic community. It was hypothesized in this study that such a community of symbiotic microbial species, not just one ambrosial fungus, is actively cultivated and perpetuated by the ambrosia beetleXyloterinus politus. Experimental results indicated that bacteria, yeasts, a yeastlike fungus, and ambrosial fungi compose such a symbiotic microbial complex in association withX. politus. The microecology of the ectosymbiotic microorganisms in relation to this insect is discussed.  相似文献

Environmental microorganisms have been widely applied in heavy metal remediation. This study explored the mechanisms of lead tolerance of two typical filamentous fungi, Aspergillus niger and Penicillium oxalicum. It is shown that the mechanisms of reducing Pb toxicity by these two fungi have three major pathways. The secreted oxalic acid can react with Pb (II) to form insoluble Pb minerals, primarily lead oxalate. Then, the enhanced biosorption via forming new border of cell wall prevents the transportation of Pb (II) into hypha. In addition, the fungal activity could be maintained even at high Pb concentration due to the intracellular accumulation. It was confirmed that A. niger has the higher Pb tolerance (up to 1500 mg l−1 Pb level) compared with P. oxalicum (up to 1000 mg l−1). Meanwhile, Pb levels below 1000 mg l−1 partially stimulate the bioactivity of A. niger, which was confirmed by its elevated respiration (from 53 to 63 mg C l−1 medium h−1). This subsequently enhanced microbial functions of A. niger to resist Pb toxicity. A better understanding of Pb tolerance of these two fungi sheds a bright future of applying them to remediate lead-contaminated environments.  相似文献

Kokuto-shochu is a traditional Japanese distilled liquor made from brown sugar. Kokuto-shochu distillery wastewater (KDW) contains high concentrations of organic compounds and brown pigments (called molasses pigments) which are hardly decolorized by general biological wastewater treatment. A fungus, Penicillium oxalicum d, which we isolated in a previous study, decolorizes 47% of the color from KDW without the addition of any nutrients. P. oxalicum d decolorizes KDW by absorbing the pigments into its mycelia. Here we describe a KDW treatment system that combines biodecolorization and biotreatment by P. oxalicum d with treatment by activated sludge and physical decolorization by ozonation. Adding HClO to suppress bacterial growth and replacing fresh seed sludge at regular intervals helped to maintain the dominance and decolorization ability of P. oxalicum d. In a laboratory-scale demonstration, 48 cycles (12 days) achieved a decolorization ratio of 90% and removed more than 97% of dissolved organic carbon (DOC), dissolved total nitrogen (DTN) and dissolved total phosphorus (DTP). A major feature of our system is that it uses only 6% of the water used in an activated sludge-ozonation system.  相似文献

Selective removal of symbiotic hindgut microorganisms by chemical treatments reduced methane emission by the termiteZootermopsis angusticollis. Methane emission from untreated termites incubated in 25% H2 increased 123%, from 10.3 nmol/termite/hour (U) to 22.9 U. Though linear with time, methane emission was not correlated with termite mass. Hyperbaric oxygen treatments reduced methane emission to unquantifiable levels and eliminated all but the protozoaTricercomitus andHexamastix. Exogenous H2 restored 5% of methane emission to 1.3 U. 2-bromoethanesulfonic acid, fed on filter papers to termites, eliminated methane production. Epifluorescence microscopy showed that this treatment selectively removed methanogens from symbioses withTricercomitus, Hexamastix, andTrichomitopsis, but the protozoa did not appear to be affected. The insect molting hormone 20-hydroxyecdysone reduced methane production 86% to 1.6 U from an initial level of 11.4 U. Hydrogen incubation increased this rate to 77% of the initial rate, 8.8 U. Hormone treatment reduced the number ofTrichonympha in the hindgut and induced sexuality in these protozoa. A model suggests thatTrichonympha evolve most of the hydrogen and that methanogenic bacteria symbiotic withTrichomitopsis produce most of the methane in this hindgut ecosystem.  相似文献

Lactobacillus casei subsp. casei CFTRI 2022 produced a higher concentration of lactic acid (5.27 g/100 g dry sugar-cane pressmud) in a solid-state fermentation (SSF) system as compared to L. helveticus CFTRI 2026 and Streptococcus thermophilus CFTRI 2034. The lactic acid production by L. casei subsp. casei CFTRI 2022 was found to be significantly influenced by the initial moisture content, initial pH and initial sugar concentration of the medium. Studies on four inert materials to reduce the initial sugar concentration in the medium showed the high potential of microcrystalline cellulose whereas the use of diatomaceous earth, acid-washed river sand and washed pith bagasse posed problems. The data indicate the potential of lactic acid production from sugar-cane pressmud in an SSF system.  相似文献
