We previously reported that broad band UV radiation or narrow bands of UV (Hbw 3 nm) of wavelengths 250 to 320 nm cause a systemic suppression of contact hypersensitivity (CHS) in mice, observed when the contact sensitizer is applied to a nonirradiated site. To determine if this effect is associated with UV-induced alterations in epidermal Langerhans cell (LC) numbers and morphology, we performed the following study. LC were identified by ATPase staining of EDTA-separated epidermal sheets. Electron microscope studies confirmed that this method was a satisfactory indicator of the presence of LC; we found no evidence for LC which did not stain for ATPase in either irradiated or unirradiated epidermis. Mice were irradiated on the back with narrow band UV of peak wavelength 270, 290, or 320 nm. The irradiated skin was excised 24 hr later and was stained as described. The number of LC with ATPase staining dendrites and the number of nondendritic LC were enumerated. We found that UV radiation of 270 or 290 nm caused 1) an alteration in LC morphology (loss of dendrites) and 2) a decrease in the total number of epidermal LC. Both effects occurred in a dose-dependent fashion. Previously, these same wavelengths of narrow band UV, but at higher doses, had been shown to cause systemic suppression of CHS. In this study, the doses of 270 or 290 nm UV that resulted in the decreased LC numbers and alterations in LC morphology described above were insufficient to cause systemic suppression of CHS. The converse was found if the irradiating waveband of UV had a peak at 320 nm. A dose of 320 nm UV that caused 50% systemic suppression of CHS had no effect on either the number or the morphology of LC at the site of irradiation. In addition, the number and morphology of LC were unaffected in the ventral epidermis (site of contact sensitization) of mice that had been previously irradiated on the back with a systemically suppressive dose of UV. We conclude: (a) UV-induced alterations in the number and morphology of LC at the site of irradiation are not necessary for the generation of systemic suppression of CHS by UV radiation; this indicates that the initial UV-absorbing event triggering systemic suppression is neither a loss of, nor morphologic alterations to, LC at the irradiation site. (b) A systemic effect of UV radiation on the number and morphology of LC at the unirradiated site of contact sensitization does not occur, and thus is not responsible for the UV-induced systemic suppression of CHS by UV radiation.  相似文献

Abstract: The effects of solar ultraviolet radiation (UV) on carbon uptake, oxygen evolution and motility of marine phytoplankton were investigated in coastal waters at Kristineberg Marine Research Station on the west coast of Sweden (58° 30'N, 11° 30'E). The mean irradiances at noon above the water surface during the investigation period were: photosynthetic active radiation (PAR, 400–700 nm) 1670 μmol m−2 s−1; ultraviolet-A radiation (UV-A, 320–400 nm) 35.9 W m−2 and ultraviolet-B radiation (UV-B, 280–320 nm) 1.7 W m−2. UV-B radiation was much more attenuated with depth in the water column than were PAR and UV-A radiation. UV-B radiation could not be detected at depths greater than 100–150 cm. Inhibition of carbon uptake by UV-A and UV-B in natural phytoplankton populations was greatest at 50 cm depth and the effects of UV-B were greater than those of UV-A. At depths greater than 50 cm there was almost no effect of ultraviolet radiation on carbon uptake. PAR, UV-A and UV-B decreased oxygen evolution by the dinoflagellate Prorocentrum minimum . Inhibition of oxygen evolution was greater after 4 h than 2 h but it was not possible to distinguish the negative effects of the different light regimes. The motility of P. minimum was not affected by PAR, UV-A and UV-B. The importance of exposure of phytoplankton to different light regimes before being exposed to natural solar radiation is discussed.  相似文献

Reflectance characteristics of the epidermal coverings (hair, feathers) of several white birds and mammals were examined in the visible and ultraviolet regions of the solar spectrum. Non-white phases of the same species, and other non-white animals were examined for comparison. As expected, non-white animals exhibited lower reflectance values than white animals in the visible spectrum. Most species examined demonstrated reduced reflectance in the ultraviolet, reaching minimum values between 290 and 310 nm. In white animals, significant differences were found in the reflectance of UV-A (320–400 nm) and UV-B (280–320 nm) radiation. This accounts for the apparent differences in ultraviolet reflectance among various arctic mammals detected previously with ultraviolet photography. Reflectance patterns in the visible and ultraviolet were not obviously correlated with phylogenetic relationship, nor with the gross structure of hair or feathers.

The transmission of parasites takes place under exposure to a range of fluctuating environmental factors, one being the changing levels of solar ultraviolet radiation (UVR). Here, we investigated the effects of ecologically relevant levels of UVR on the transmission of the intertidal trematode Maritrema novaezealandensis from its first intermediate snail host (Zeacumantus subcarinatus) to its second intermediate amphipod host (Paracalliope novizealandiae). We assessed the output of parasite transmission stages (cercariae) from infected snail hosts, the survival and infectivity of cercariae, the susceptibility of amphipod hosts to infection (laboratory experiments) and the survival of infected and uninfected amphipod hosts (outdoor experiment) when exposed to photo-synthetically active radiation only (PAR, 400-700 nm; no UV), PAR+UVA (320-700 nm) or PAR+UVA+UVB (280-700 nm). Survival of cercariae and susceptibility of amphipods to infection were the only two steps significantly affected by UVR. Survival of cercariae decreased strongly in a dose-dependent manner, while susceptibility of amphipods increased after exposure to UVR for a prolonged period. Exposure to UVR thus negatively affects both the parasite and its amphipod host, and should therefore be considered an influential component in parasite transmission and host-parasite interactions in intertidal ecosystems.  相似文献

Exposure of mice to UVB radiation produces a highly selective, systemic immunosuppression associated with the appearance of suppressor T lymphocytes. Suppression of delayed hypersensitivity to hapten-coupled syngeneic cells has been shown to result from an altered distribution of antigen-presenting cells. The purpose of this study was to determine whether an alteration in the activity of antigen-presenting cells could account for the systemic suppression of contact hypersensitivity (CHS) by UVB radiation. Fluorescein isothiocyanate (FITC) was used for contact sensitization because it uses different antigen-presenting cells than does oxazolone to induce CHS. Our previous studies demonstrated that CHS to oxazolone was suppressed by UVB irradiation. In these studies, we show that exposure of mice to UVB radiation before epicutaneous application of FITC onto unirradiated skin markedly decreased the CHS response to FITC painted on unexposed ears. Cyclophosphamide-sensitive suppressor T cells were detectable in the spleens of mice exhibiting decreased CHS. The antigen-presenting activity of cells in lymph nodes draining the site of epicutaneous sensitization (DLN cells) was assessed by injecting them into the hind footpads of syngeneic recipients and measuring the CHS response to FITC 6 days later. Viable DLN cells from UVB-irradiated, FITC-sensitized mice were equal to those from unirradiated, FITC-sensitized mice in their ability to induce CHS in normal recipients. No sensitization resulted when killed DLN cells were used for immunization, indicating that sensitization was not caused by reprocessing of antigen by host cells. We conclude that impairment of the CHS reaction in UVB-irradiated mice does not appear to be blocked at an initial step of antigen uptake, processing, or presentation, but must be impaired at some other step in the immunologic pathway.  相似文献

Seasonal reproduction in some Arctic Laminariales coincides with increased UV-B radiation due to stratospheric ozone depletion and relatively high water temperatures during polar spring. To find out the capacity to cope with different spectral irradiance, the kinetics of photosynthetic recovery was investigated in zoospores of four Arctic species of the order Laminariales, the kelps Saccorhiza dermatodea, Alaria esculenta, Laminaria digitata, and Laminaria saccharina. The physiology of light harvesting, changes in photosynthetic efficiency and kinetics of photosynthetic recovery were measured by in vivo fluorescence changes of Photosystem II (PSII). Saturation irradiance of freshly released spores showed minimal I k values (photon fluence rate where initial slope intersects horizontal asymptote of the curve) values ranging from 13 to 18 μmol photons m−2 s−1 among species collected at different depths, confirming that spores are low-light adapted. Exposure to different radiation spectra consisting of photosynthetically active radiation (PAR; 400–700 nm), PAR+UV-A radiation (UV-A; 320–400 nm), and PAR+ UV-A+UV-B radiation (UV-B; 280–320 nm) showed that the cumulative effects of increasing PAR fluence and the additional effect of UV-A and UV-B radiations on photoinhibition of photosynthesis are species specific. After long exposures, Laminaria saccharina was more sensitive to the different light treatments than the other three species investigated. Kinetics of recovery in zoospores showed a fast phase in S. dermatodea, which indicates a reduction of the photoprotective process while a slow phase in L. saccharina indicates recovery from severe photodamage. This first attempt to study photoinhibition and kinetics of recovery in zoospores showed that zoospores are the stage in the life history of seaweeds most susceptible to light stress and that ultraviolet radiation (UVR) effectively delays photosynthetic recovery. The viability of spores is important on the recruitment of the gametophytic and sporophytic life stages. The impact of UVR on the zoospores is related to the vertical depth distribution of the large sporophytes in the field.  相似文献

Irradiation with ultraviolet B (UVB; 290–320 nm) initiates systemic immunosuppression of contact hypersensitivity (CHS). UV dose-responses for suppression of CHS to trinitrochlorobenzene were established in 18 strains of inbred mice. Three phenotypes with significantly different susceptibilities to UV suppression were identified. The phenotypes were: high (HI) susceptibility, 50% suppression with 0.7–2.3 kJ/m2 UV (C57BL/6, C57BL/10, and C57L and NZB females); low (LO) susceptibility, 50% suppression with 9.6–12.3 kJ/m2 UV (BALB/c, AKR, SJL and NZW), and intermediate (INT) susceptibility, 50% suppression with 4.7–6.9 kJ/m2 UV (DBA/2, C57BR, C3H/HeJ, C3H/HeN, CBA/N and A/J). UV suppression was not correlated with skin pigmentation or with the magnitude of the CHS response in non-irradiated animals. Major histocompatibility complex (MHC) haplotype was not correlated with UV suppression in MHC congenic strains B10.D2/oSnJ, B10.D2/nSnJ, B10.BR/SgSnJ, and A.BY/SnJ. There were no sex differences in UV suppression in BALB/c, C57BL/6, or NZW animals. In the autoimmune NZB strain, however, male mice (LO) were seven times less sensitive to UV suppression than NZB female mice (HI). Both sexes of (NZB × NZW)F1 and (NZW × NZB)F1 mice were HI, supporting dominance of HI over LO. Thus there are genetic factors and interacting sex-limited factors determining susceptibility to UV suppression. These findings may be of relevance to UV-related diseases such as photosensitive lupus and skin cancer. Correspondence to: F. P. Noonan.  相似文献

1. Lake Titicaca is a large, high altitude (3810 m a.s.l.) tropical lake (16°S, 68°W) that lies on the border of Bolivia and Perú, receiving high fluxes of ultraviolet radiation (UVR) throughout the year. Our studies were conducted during September of 1997 with the main objective of studying the impact of solar UVR upon phytoplankton photosynthesis.
2. Water samples were taken daily and incubated in situ (down to 14 m depth) under three radiation treatments to study the relative responses to PAR (Photosynthetic Available Radiation, 400–700 nm), UV-A (320–400 nm), and UV-B (280–320 nm) radiation.
3. Photosynthetic inhibition by UVR in surface waters was about 80%, with UV-A accounting for 60% and UV-B for 20%; the inhibition by high levels of PAR was less than 20%. The inhibition due to UVR decreased with depth so that there were no significant differences between treatments at 8.5 m depth.
4. The amount of inhibition per unit energy received by phytoplankton indicates that even though there was a significant inhibition of photosynthesis due to UVR, species in Lake Titicaca seem to be better adapted than species in high latitude environments.
5. The cellular concentration of UV-absorbing compounds, a possible mechanism of photoadaptation, was low in phytoplanktonic species. However, they were abundant in zooplankton, suggesting a high rate of bioaccumulation through the diet.  相似文献

The purpose of this study was to determine whether multiple types of suppressor factors play a role in the regulation of immune responses by ultraviolet radiation-induced suppressor T lymphocytes (UV Ts). The UV Ts were induced by applying contact allergens to the ventral, unirradiated skin of mice exposed 5 days earlier to UVB radiation. Previous studies indicated that supernatants from cultures containing UV Ts, normal lymphocytes, and hapten-modified cells suppressed contact hypersensitivity (CHS) in vivo and cytotoxic T lymphocyte (CTL) generation in vitro in a hapten-specific manner. In this report, cell-free lysates from sonically disrupted UV Ts were examined for their ability to suppress these responses. When lysates were injected into normal animals at the time of sensitization, they inhibited CHS in a hapten-nonspecific manner. In addition, the lysates suppressed not only the induction but also the elicitation of CHS, and they suppressed the generation of CTL. Lysates prepared from spleen cells obtained from non-UV-irradiated mice or UV-irradiated, unsensitized mice failed to inhibit either response. Moreover, in contrast to the lysates, the hapten-specific UV Ts culture supernatants inhibited the induction but not the elicitation of CHS. These results suggest that both hapten-specific and nonspecific inhibitory factors may participate in the regulation of immune responses by UV Ts.  相似文献

The hypothalamic-pituitary-adrenal (HPA) axis maintains basal and stress-related homeostasis in vertebrates. Skin expresses all elements of the HPA axis including corticotropin-releasing hormone (CRH), proopiomelanocortin (POMC), ACTH, β-endorphin (β-END) with corresponding receptors, the glucocorticoidogenic pathway, and the glucocorticoid receptor (GR). To test the hypothesis that cutaneous responses to environmental stressors follow the organizational structure of the central response to stress, the activity of the "cutaneous HPA" axis homolog was investigated after exposure to ultraviolet radiation (UVR) wavelengths of UVA (320-400 nm), UVB (280-320 nm), and UVC (100-280 nm) in human skin organ culture and in co-cultured keratinocytes/melanocytes. The level of stimulation of CRH, POMC, MC1R, MC2R, CYP11A1, and CYP11B1 genes was dependent on UV wavelengths and doses, with the highest effects observed for highly energetic UVC and UVB. ELISA and Western assays showed significant production of CRH, POMC, ACTH, and CYP11A1 proteins and of cortisol, with a decrease in GR expression only after UVB and UVC. However, β-END expression was also stimulated by UVA. Immunocytochemistry localized the deposition of the aforesaid antigens predominantly to the epidermis with additional accumulation of CRH, β-END, and ACTH in the dermis. UVR-stimulated CYP11A1 expression was seen in the basal layer of the epidermis and cells of adjacent dermis. Thus, the capacity to activate or change the spatial distribution of the cutaneous HPA axis elements is dependent on highly energetic wavelengths (UVC and UVB), implying a dependence of a local stress response on their noxious activity with overlapping or alternative mechanisms activated by UVA.  相似文献

Impact of solar ultraviolet-B radiation (290-320 nm) upon marine microalgae   总被引:1,自引:0,他引:1
For years scientists and laymen alike have casually noted the impact of solar ultraviolet radiation upon the non-human component of the biosphere. It was not until recently, when human activities were thought to threaten the protective stratospheric ozone shield, that researchers undertook intensive studies into the biological stress caused by the previously neglected short-wavelength edge of the global solar spectrum. Stratospheric ozone functions effectively as an ultraviolet screen by filtering out solar radiation in the 220–320 nm waveband as it penetrates through the atmosphere, thus allowing only small amounts of the longer wavelengths of radiation in this waveband to leak through to the surface of the earth. Although this ultraviolet radiation (UV-B radiation, 290–320 nm) comprises only a small fraction (less than 1%) of the total solar spectrum, it can have a major impact on biological systems due to its actinic nature. Many organic molecules, most notably DNA and proteins, absorb UV-B radiation which can initiate photochemical reactions. It is life's ability, or lack thereof, to cope with enhanced levels of solar UV-B radiation that has generated the concern over the potential depletion of stratospheric ozone. The defense mechanisms that serve to protect both plants and animals from current levels of UV-B radiation are quite varied. Whether these mechanisms will suffice for marine microalgae under conditions of enhanced levels of UV-B radiation is the subject of this review.  相似文献

Strips of bovine mesenteric arteries brought to sustained contraction by the addition of 3.0 microM phenylephrine relaxed when exposed to ultraviolet radiation (366 nm). The relaxation was reversible and associated with a rapid increase in the cGMP content. After termination of the radiation the cGMP level rapidly decreased below the basal level. The crude soluble guanylate cyclase from the artery was stimulated about 8-fold by ultraviolet radiation (366 nm). Neither the cGMP-phosphodiesterase activity nor the cAMP level were found to be changed during irradiation. The ultraviolet light-induced relaxation was not dependent on an intact intimal surface. Furthermore, the relaxing effect was found to be enhanced and accompanied by a larger increase of the cGMP level in nitroglycerin-tolerant arteries. The present results show that the ultraviolet light-induced relaxation in bovine mesenteric arteries is associated with a rapid increase in the cGMP content and that ultraviolet light and nitrocompounds may exert their relaxing actions through a common substance.  相似文献

The effect of (i) aphidicolin, a specific inhibitor of delta- and epsilon-polymerases, and nucleotide excision repair; (ii) 3-aminobenzamide, an inhibitor of poly(ADP-ribose) polymerase and base excision repair; and (iii) actinomycin D and cycloheximide, inhibitors of protein and RNA synthesis, respectively, on the induction of suppression of apoptosis of rat thymocytes by different doses of short-wavelength ultraviolet radiation was studied by flow cytometry. 3-Aminobenzamide suppressed the inhibition of apoptosis induced by the doses of short-wavelength ultraviolet radiation higher than 20 J/m2, increasing the cell death to a maximum. Thus, the inhibition of apoptosis by high short-wavelength ultraviolet radiation doses depends on the status of poly(ADP-ribose) polymerase and is prevented by 3-aminobenzamide. As opposed to 3-aminobenzamide, aphidicolin did not affect the cell death at short-wavelength radiation doses higher than 10 J/m2 but induced the apoptosis of unirradiated cells and cells irradiated with short-wavelength ultraviolet radiation doses lower than 10 J/m2. The inhibitors of protein and RNA synthesis cycloheximide and actinomycin D prevented the induction of apoptosis caused by low and medium doses but did not abolish the apoptosis-inhibiting activity of high doses of short-wavelength ultraviolet radiation.  相似文献

In field studies conducted at the Kongsfjord (Spitsbergen) changes of the irradiance in the atmosphere and the sublittoral zone were monitored from the beginning of June until the end of August 1997, to register the minimum and maximum fluxes of ultraviolet and photosynthetically active radiation and to characterise the underwater light climate. Measurements of photosynthesis in three abundant brown algal species (Alaria esculenta, Laminaria saccharina, Saccorhiza dermatodea) were conducted to test whether their photosynthetic performance reflects changing light climate in accordance with depth. Plants sampled at various depths were exposed to controlled fluence rates of photosynthetically active radiation (400–700 nm), UV-A (320–400 nm) and UV-B (280–320 nm). Changes in photosynthetic performance during the treatments were monitored by measuring variable chlorophyll fluorescence of photosystem II. In each species, the degree of inhibition of photosynthesis was related to the original collection depth, i.e. shallow-water isolates were more resistant than plants from deeper waters. The results show that macroalgae acclimate effectively to increasing irradiance levels for both photosynthetically active and ultraviolet radiation. However, the kinetics of acclimation are different within the different species. It is shown that one important strategy to cope with higher irradiance levels in shallow waters is the capability for a faster recovery from high light stress compared to isolates from deeper waters. Stomatal closure by ultraviolet radiation   总被引:5,自引:0,他引:5
The effect of ultraviolet radiation (UV) (255–325 nm) on stomatal closure was investigated on tef [ Eragrostis tef (Zucc) Trotter] in the presence of white light (ca 50 ·mol m−2 s−1). The action spectrum showed that UV (ca 2 ·mol m−2 s−1, half band width about 10 nm) of 285 nm or shorter wavelengths was very efficient in causing stomatal closure. The effectiveness decreased sharply towards longer wavelengths. Radiation of 313 nm or longer wavelengths was practically without effect. Increasing UV intensity increased stomatal resistance. When stronger white light (5 to 9 times stronger than the one used during irradiation) was administered, stomates re-opened rapidly irrespective of whether the UV was on or off, although a subsequent gradual closing tendency was observed when the UV was on.  相似文献

It is now well established that ultraviolet radiation (UVR) may have detrimental, even lethal effects on zooplankters. Unlike copepods and other holoplankters, which may avoid UVR by undergoing diel vertical migration, larvae of many decapod crustaceans and fishes recruit to adult populations by remaining in near-surface waters during the daytime. Consequently, they are exposed to biologically damaging UVR. A possible adaptation in these larvae is chromatophores, which may absorb UVR by expanding in high light environments. The supposition is that expanded chromatophores more effectively absorb UVR, but there is some fitness cost to having expanded chromatophores in low light environments. Since the ratio of visible light to UVR in the water column changes as result of season, latitude, dissolved organic carbon, and a host of other factors, the benefits of chromatophores would be maximized if they responded specifically to UVR. The purpose of this study was to determine whether the chromatophores of crab postlarvae (megalopae) could expand in response to UVR. Megalopae of two species of crabs (Cancer oregonensis, Telmessus cheiragonus) were collected from large surface-swarms during mid-day as they recruited onshore in early May 1998 at Friday Harbor, Washington, USA. Dark-adapted megalopae (held in the dark for 8 h before experiments) were exposed to UVR (UVBR+UVAR, 280-400 nm), UVAR (320-400 nm), and light (400-1700 nm) in the laboratory. Chromatophores expanded after only minutes of exposure to UVR, UVAR, and light for both species. Two alternative hypotheses may explain why both harmful and comparatively benign wavelengths stimulated chromatophores to rapidly expand. First, larvae may not distinguish among different wavelengths, which, if true, would increase the vulnerability of these larvae to intensifying UVBR due to ozone depletion. Second, chromatophores have functions other than blocking UVR, such as crypsis and thermoregulation, and must respond to light for these other functions to operate.  相似文献

A comparison has been made of sensitivity to far (254 nm), middle (300--315 nm) and near (315--400 nm) UV radiation of 12 strains of E. coli and 2 strains of B. subtilis differing in DNA dark repair (DR) capability. The mechanisms controlled by uvrA, uvrB, polA, recA, lon, and lexA genes are very effective in cells, irradiated by far and middle UV, but by 15--70% less effective in those irradiated by near UV. As the unirradiated bacteria poorly repair the near UV damaged phages (T7, lambda, SPPI), the low bacterial DR level after UV irradiation seems to be due to the unrepairable photoproduct formation in DNA.  相似文献

Senescence induced loss in pigments and proteins of detached maize (Zea mays L. cv. Col) leaves was significantly enhanced on the exposure of leaves to different ranges of ultraviolet (UV) radiation. Compared to UV-A (320-400 nm) and UV-B (280-320 nm), the UV-C (200-320 nm) was the most damaging for the pigments and macromolecules. A severe decline in photosystem (PS) 2 mediated photoreduction during senescence of detached leaves exposed to UV irradiation suggested a damage of the system. The PS1 mediated photoreduction of methylviologen with 2,6-dichlorophenol indophenol as electron donor was stimulated by UV-A and UV-B radiations, suggesting a reorganisation of the PS1 complex. These results were fortified by the values of fast and slow kinetics of chlorophyll (Chl) a fluorescence transients.  相似文献

In this study we compared the effects of subinflammatory and inflammatory doses of solar-simulated ultraviolet (UV) radiation on enhancement of skin tumor growth, sensitization to haptens and cellular changes within the epidermis of C3H/HeN mice. Tumors transplanted into mice 3 days after exposure to inflammatory, but not subinflammatory, doses of UV radiation had a higher growth rate than those tumors inoculated into unirradiated control mice. Both doses of UV radiation suppressed the induction of contact hypersensitivity and induced tolerance when hapten was painted onto the skin 3 days after irradiation. Skin exposed to the higher, but not the lower, dose of UV radiation contained significantly increased numbers of CD11b+, CD45+ MHC class II- and CD45+ MHC class II(hi) inflammatory cells 3 days post-irradiation. The immunosuppression correlated with a reduction in Langerhans cells and dendritic epidermal T cells. Collectively, this suggests that suppression to contact sensitizers is due to the UV radiation effects on Langerhans cells and dendritic epidermal T cells. While these effects may also suppress the induction of anti-tumor immunity, at higher doses of UV radiation inflammatory cells may enhance tumor growth by a non-immunological mechanism.  相似文献
