Because of its stringent sequence specificity, tobacco etch virus (TEV) protease emerges as a useful reagent with wide application in the cleavage of recombinant fusion proteins. However, the solubility of TEV protease expressed in Escherichia coli is extremely low. In the present study, we introduced a more efficient system to improve and facilitate the soluble production of TEV protease in E. coli. Optimal expression of soluble His6-TEV was achieved by examining the contribution of chaperone co-expression and lower temperature fermentation. When further purified by Ni(2+) affinity chromatography, 65mg of His6-TEV was isolated with purity over 95% from 1L of culture. The enzyme activity of His6-TEV was generally characterized by using GST-EGFP and His6-L-TNF fusion protein as substrates, which contained a TEV cleavage site between two moieties.  相似文献

DNA错配修复蛋白MutL和其它的修复蛋白相互作用来共同完成大肠杆菌甲基介导的错配修复过程 .为研究修复蛋白MutL的体外生物功能构建了融合蛋白Trx His6 Linkerpeptide MutL(THLL)的表达载体并使其高效表达及易于纯化 .mutL基因片段是以E .coliK 12基因组为模板经PCR扩增获得 ,并通过基因的体外拼接成功构建了融合蛋白THLL表达载体pET32a linkerpeptide mutL .重组菌株E .coliAD4 94 (DE3) pET32a linkerpeptide mutL经过IPTG的诱导表达了融合蛋白THLL .收集菌体细胞、超声波破碎后离心取上清进行SDS PAGE分析 ,结果表明有一与预期分子量(84kD)相应的诱导表达条带出现 ,其表达量约占全细胞蛋白的 30 %且以可溶形式存在 .利用固定化金属离子 (Ni2 +)配体亲和层析柱纯化融合蛋白THLL ,其纯度达到 90 % .通过非变性凝胶电泳分析 ,对融合蛋白THLL在DNA错配修复过程中的分子伴侣生物功能进行了系统研究 .结果表明 ,THLL能增加融合蛋白Trx His6 Linkerpeptide MutS (THLS)与含有错配碱基DNA双链的结合 ,但受ATP的浓度变化影响很大  相似文献

LECT2 is a chemotactic protein to neutrophils. A recombinant six-histidine-tagged human LECT2, (His)6-LECT2, was expressed in E. coli using a pET21a(+) vector. The (His)6-LECT2 was purified from the soluble fraction in E. coli as a single band in sodium dodesyl sulfate/polyacrylamide gel electrophoresis using three steps of column chromatography with Ni2+-charged nitrilo-triacetic acid (Ni-NTA) agarose, DEAE-Sepharose, and CM-Sepharose. The purified (His)6-LECT2 was yielded with 96 μg from the soluble fraction of 1,500 ml culture of E. coli. The circular dichroism spectrum of (His)6-LECT2 showed the folded structure, which is rich in β-sheet structure and rare in α-helix. We report the results on the expression in Escherichia coli of a functional neurotoxin LqqV from the scorpion Leiurus quinquestriatus quinquestriatus. The gene for LqqV was synthesized using recursive PCR and expressed as a poly-histidine-tagged fusion protein in thioredoxin mutant E. coli strain [AD494(DE3)pLysS], thus permitting disulfide-bond formation. When cultured at 37 degrees C, about 50% of the expressed protein is contained as a monomer in the soluble fraction of the E. coli extract. The fusion protein from the soluble fraction was purified and the His-tag was cleaved by thrombin, resulting in a yield of about 1.5 mg/liter. The globular structure of the purified protein was confirmed by NMR and CD spectroscopy. Patch-clamp measurements using native sodium channels in guinea pig ventricular myocytes reveal (1) a slowing of inactivation and (2) a decrease in peak current upon application of toxin, thus confirming the alpha-toxin activity of the purified recombinant protein.  相似文献

曾以水稻蜡质基因5’调控区内一段31 bp 片段为探针,用酵母单杂交法从水稻cDNA 文库中筛选出若干个其编码的蛋白可能与此31 bp 片段结合的cDNA克隆,现将其中的pC73 克隆中的插入片段c73 连接到含His6 的表达载体pET28c( + ) 上,在大肠杆菌BL21(DE3) 中进行诱导表达,并用NiNTA 树脂纯化得到预期的融合表达产物。在合适的诱导表达条件下,融合表达产物主要以可溶形式存在于大肠杆菌细胞内;表达量占到大肠杆菌总蛋白的10 % 左右;经NiNTA 树脂亲和层析纯化得到的产物纯度达95 % ,可供进一步研究之用。  相似文献

The hybrid antibacterial peptide CA-MA [cecropinA(1-8)-magainin2(1-12)] with potent antimicrobial properties but no hemolytic activity is a potential alternative antibiotic. To explore a new approach for high-level expression of the hybrid peptide CA-MA in Escherichia coli, the sequence of ubiquitin (UBI) from housefly was inserted into the plasmid pQE30 to construct the vector pQEUBI. The cDNA fragment encoding CA-MA with preferred codons of E. coli was obtained by recursive PCR (rPCR) and cloned into the vector pQEUBI to express the fusion protein (His)(6)-UBI-CA-MA. The fusion protein was expressed in soluble form under the optimized conditions at high level (more than 36% of the total proteins). With (His)(6)-tag, the fusion protein was easily purified by Ni-NTA chromatography and 36 mg of fusion protein was purified from 1L of culture medium. The fusion protein was efficiently cleaved by ubiquitin C-terminal hydrolase (UCH), yielding recombinant CA-MA with high antimicrobial activity. After removing the contaminants by Ni-NTA chromatography, recombinant CA-MA was purified to homogeneity by reversed-phase HPLC and 6.8mg of pure active CA-MA was obtained from 1L culture medium. Analysis of recombinant CA-MA by MALDI-TOF-MS showed that the molecular weight of the purified recombinant CA-MA was 2559Da, which perfectly matches the mass (2559Da) calculated from the amino acid sequence. Analysis of CA-MA by circular dichroism (CD) revealed that the secondary structures of CA-MA in water solution were 17.4% alpha-helix and 82.6% random coil but no beta-sheet. Our results demonstrated that functional CA-MA can be produced in sufficient quantities using the ubiquitin fusion technique. This is the first report on the heterologous expression of a hybrid antibacterial peptide fused to ubiquitin in E. coli.  相似文献

死亡素是由21个氨基酸残基组成的广谱抗菌肽。为了高效表达可溶性的死亡素,本研究利用递归式PCR(recursive PCR, rPCR)扩增了死亡素基因thanatin,并将其和家蝇Musca domestica泛素基因ubiquitin构成嵌合基因,克隆到表达载体pET-32a,再与硫氧还蛋白融合后构建表达载体pET-TRX-UBI-THA。将酶切和测序鉴定正确的质粒转化表达宿主菌BL21,经0.6 mmol/L IPTG诱导,TRX-UBI-THA融合蛋白得到了高效可溶性表达。SDS-PAGE和Western blot检测结果表明融合蛋白的分子量为28.9 kD,与预期的结果一致,表达量占菌体总蛋白的46%。Western blot分析结果显示融合蛋白能与Ni-NTA鏊合物特异性的结合,表明在融合蛋白的N-端带有6×His标签。利用C-端带有6×His标签的泛素C-端水解酶对融合蛋白进行切割,切割产物经Ni2+-NTA亲和柱和HPLC纯化(纯化量为5.4 mg/L),Tricince-SDS-PAGE电泳得到单一的泛素蛋白条带。电喷雾质谱(ESI-MS)分析表明,纯化的泛素分子量为2.57 kD,与通过氨基酸预测的分子量完全一致。利用琼脂孔穴扩散法对泛素活性进行检测,结果显示纯化的泛素对大肠杆菌K12D31和金黄色葡萄球菌Staphylococcus aureus具有较强的活性抑制。本研究表明,利用泛素融合技术可以高效表达可溶性的死亡素。  相似文献

目的构建PHD2基因原核表达载体pET-43.1b(+)-PHD2,实现Nus-PHD2融合蛋白在大肠埃希菌中的可溶性表达。方法用SacⅠ酶切pET-43.1b(+)制备线性化载体,设计与线性化载体两端具有至少15个同源序列的特异性引物,以真核重组质粒pCMV6-Entry-EGLN1为模板,PCR法扩增PHD2目的基因。采用In-Fusion技术构建原核表达载体pET-43.1b(+)-PHD2,并将其导入大肠埃希菌BL21(DE3)中诱导表达。用SDS-PAGE和Western blot分析并鉴定表达出的融合蛋白。用Ni-NTA亲和层析法纯化目的蛋白。结果成功构建了PHD2原核表达载体;SDS-PAGE结果显示融合蛋白以可溶性形式表达;Western blot鉴定表明融合蛋白可以与PHD2单克隆抗体特异性结合。结论实现了Nus-PHD2融合蛋白在大肠埃希菌中的可溶性表达,为PHD2生物学功能的研究奠定了基础。  相似文献

目的:构建PET-28a-SPA原核表达载体,在大肠杆菌BL21(DE3)中实现其高效可溶性表达,测定对肿瘤细胞的凋亡效果。方法:本实验在获得凋亡蛋白融合基因的基础上,成功地构建了重组表达质粒PET-28a-SPA,将阳性重组质粒转化表达受体菌BL21(DE3)感受态细胞中,经IPTG诱导表达,表达产物经聚丙烯酰胺凝胶电泳检测和Western blot检测,并采用MTT法检测其对肿瘤细胞的增殖抑制。结果:表达产物经聚丙烯酰胺凝胶电泳检测,凋亡蛋白融合基因获得高效表达,软件分析表明表达蛋白占菌体蛋白20%左右。上清表达量约为10%。上清蛋白经纯化后,Western blot结果显示,利用凋亡蛋白单克隆抗体可以很好地和所表达的蛋白带特异性结合,并且对A549肺癌细胞及Hela细胞具有一定的凋亡作用。结论:所获凋亡蛋白以高效胞质可溶形式表达,为其研制有效的肿瘤免疫治疗靶向药物提供一定的基础。  相似文献

Three native E. coli proteins-NusA, GrpE, and bacterioferritin (BFR)-were studied in fusion proteins expressed in E. coli for their ability to confer solubility on a target insoluble protein at the C-terminus of the fusion protein. These three proteins were chosen based on their favorable cytoplasmic solubility characteristics as predicted by a statistical solubility model for recombinant proteins in E. coli. Modeling predicted the probability of soluble fusion protein expression for the target insoluble protein human interleukin-3 (hIL-3) in the following order: NusA (most soluble), GrpE, BFR, and thioredoxin (least soluble). Expression experiments at 37 degrees C showed that the NusA/hIL-3 fusion protein was expressed almost completely in the soluble fraction, while GrpE/hIL-3 and BFR/hIL-3 exhibited partial solubility at 37 degrees C. Thioredoxin/hIL-3 was expressed almost completely in the insoluble fraction. Fusion proteins consisting of NusA and either bovine growth hormone or human interferon-gamma were also expressed in E. coli at 37 degrees C and again showed that the fusion protein was almost completely soluble. Starting with the NusA/hIL-3 fusion protein with an N-terminal histidine tag, purified hIL-3 with full biological activity was obtained using immobilized metal affinity chromatography, factor Xa protease cleavage, and anion exchange chromatography.  相似文献

目的:获得纯化抗原用于制备ZNF268的多克隆抗体。方法:PCR扩增目的基因片Spacer区序列,亚克隆入融合蛋白表达载体pET28a+,构建了重组质粒pET28a+/SPA。然后将该重组质粒转化E.coliDE3(Rosseta),IPTG诱导表达,SDS-PAGE分离,获纯化融合蛋白6His-SPA。用6His-SPA免疫家兔,颈动脉取血,分离血清,从血清中获得ZNF268特异的多克隆抗体。结论:通过构建融合蛋白重组表达质粒pET28a+/SPA,用获得的初步纯化融合蛋白6His-SPA,制备了特异的ZNF268的多克隆抗体6His-SPA Ab。  相似文献

目的:构建结核分枝杆菌融合基因esat6-rpfD的原核表达载体,表达和纯化ESAT6-RpfD融合蛋白。方法:从结核分枝杆菌H37Rv株基因组中经PCR分别扩增esat6和慢周基因,克隆入pMD19-T载体,测序后克隆入原核表达载体pProExHTB,酶切重组质粒,转化大肠杆菌DH5α,IPTG诱导表达融合蛋白,亲和层析纯化融合蛋白。结果:PCR扩增的esat6、rpfD基因序列与GenBank报道一致;诱导表达后,经SDS-PAGE和Western blot分析,在相对分子质量约30000处有目的条带,融合蛋白以包涵体形式表达。结论:构建了esat6-rpfD融合基因原核表达载体,并在大肠杆菌中表达并纯化得到ESAT6-RpfD融合蛋白。  相似文献

目的:表达和纯化人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白。方法:利用PCR搭接方法及基因合成方法获得目的基因,插入带有6×His标签的原核高效可溶性表达载体pET32a中,构建重组表达质粒pET32a-T9-ac-9,将重组表达质粒转化大肠杆菌BL21(DE3),经IPTG诱导目的基因表达;对融合蛋白进行Ni2+金属螯合柱纯化。结果:构建的重组表达质粒经PCR、内切酶鉴定及基因序列测定证实;目的蛋白在大肠杆菌中获得表达,SDS-PAGE显示相对分子质量为22.917×103;对表达产物进行了亲和层析纯化,从上清中获得了纯度较高的人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白。结论:获得了可溶性的人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白,为其生物学功能研究奠定了基础。  相似文献

目的:利用基因工程的方法在大肠杆菌中表达并纯化生物活性肽Lunasin。方法:将合成的Lunasin基因插入原核表达载体pET-32a(+)的多克隆位点Nde I和Xho I之间,然后将重组载体转化入大肠杆菌BL21(DE3)中,利用IPTG诱导表达蛋白,经SDS-PAGE和Western Blot鉴定蛋白的表达。然后利用亲和层析技术将含有6×His标签的蛋白分离纯化、脱盐、冻干。结果:①鉴定结果表明在6kDa位置出现目的条带Lunasin重组蛋白。②亲和层析在100mM咪唑时得到了洗脱的重组蛋白。结论:在大肠杆菌BL21(DE3)中成功表达并且纯化出了生物活性肽Lunasin。  相似文献

目的:克隆、表达人vasorin(VASN)蛋白。方法:利用PCR方法从HepG2细胞的cDNA中扩增获得目的基因,并插入带有6xHis标签的原核高效可溶性表达载体pET28a中,构建重组表达质粒pET28a-VASN,将重组表达质粒转化大肠杆菌BL21(DE3),经IPTG诱导后目的基因获得表达,对融合目的蛋白进行Ni^2+金属螯合柱纯化。结果:内切酶鉴定及基因序列测定证实重组表达质粒构建成功;对目的蛋白进行了原核表达,SDS-PAGE显示相对分子质量为61x10^3的特异表达条带;Western印迹证实目的蛋白为VASN,且主要以包涵体形式存在;对经尿素变性的表达产物进行了亲和层析纯化,有利于以后的变性、复性过程。结论:获得了人VASN融合蛋白,为其进一步的生物学功能研究奠定了基础。  相似文献

以p90-HCV为模板,通过PCR分别得到全长和截短(2881-3078bp)ns2基因。将截短ns2基因克隆到pET32a原核表达载体,并在大肠杆菌中获得了Trx-His-NS2C融合蛋白的可溶性高表达。通过His树脂纯化得到融合蛋白,免疫家兔获得高效价高特异性的抗体。同时构建了含有全长ns2基因的重组腺病毒,利用该重组病毒感染HepG2和293细胞成功的表达23.2kDa的NS2蛋白。本文的研究为进一步开展NS2蛋白的结构与功能研究奠定了良好的基础。  相似文献

NDRG1是在N myc缺陷的小鼠胚胎组织中发现的一异常高表达的新基因 .在研究高同型半胱氨酸血症引起动脉粥样硬化的机制时 ,在培养的人脐静脉内皮细胞 (HUVEC)中发现了人NDRG1 .为了研究人NDRG1的功能以及结构与功能之间的关系 ,用RT PCR技术 ,从人脑总RNA中克隆NDRG1cDNA ,进行序列测定后 ,将NDRG1插入pPROEXHTb表达载体中 ,转化E .coliDH5α感受态细胞 ,并在LB培养基中获得高效可溶表达 .表达的 6His NDRG1融合蛋白经Ni2 + NTA偶联的琼脂糖珠纯化 ,通过圆二色性分析其二级结构 ,结果为 :α螺旋 :2 3 6 ,β片层 :1 8 6 ,转角 :2 5 7,无规卷曲 :32 0 .用此融合表达的蛋白免疫家兔 ,制备得到高效价的抗体 ,利用固定于硝酸纤维素膜上的NDRG1抗原亲和吸附纯化抗血清提高了抗体的特异性 ,为进一步研究NDRG1的功能打下了良好的基础  相似文献

进行人乳头瘤病毒6b型(Human papillomavirus type 6b,HPV6b)E7蛋白原核表达并制备其多克隆抗体。用已构建的pGEX-4T-2/HPV6bE7原核表达载体诱导表达大量可溶性融合蛋白GST-HPV6bE7,用Glutathione-Sepharose 4B亲和柱和凝血酶纯化获取HPV6b型E7蛋白。将纯化的E7蛋白免疫新西兰兔并纯化为多克隆抗体IgG。采用Western-Blot及免疫荧光法分析该抗体的效价及特异性。十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析显示,异丙基-β-D-硫代半乳糖苷(IPTG)诱导3~6h后pGEX-4T-2/HPV6bE7表达载体在大肠杆菌中高水平表达可溶性融合蛋白。纯化的E7蛋白免疫新西兰兔后可获得兔多克隆抗体IgG。经Western-Blot及免疫荧光鉴定,兔抗IgG具有高效价性和抗HPV6bE7蛋白特异性。获取纯化的HPV6b型E7蛋白具有较好的免疫原性,其免疫兔产生的多克隆抗体IgG效价高,特异性好,有望进一步用于HPV6b型的生物学功能研究和免疫学效应研究。  相似文献

The gene of the capsid protein of bovine immunodeficiency virus (BIV) was linked to a sequence encoding for six histidines and expressed as the (His)6p26 capsid fusion protein. The fusion protein was strongly expressed as both soluble and insoluble forms after induction by isopropylthio-β- -galactoside. Purification was based on interaction of the hexa-histidine polypeptide with metal ions. Expression could represent 11% of the total protein inEscherichia coli,allowing more than 20 mg of highly purified protein to be obtained per liter of bacterial culture. The (His)6p26 capsid fusion protein purified by immobilized metal affinity chromatography reacted specifically in Western blot with sera from cattle experimentally infected by BIV, as well as with two monoclonal antibodies directed against different epitopes of the Gag protein. The ease of expression, purification, and specificity of this fusion protein should permit a thorough study of prevalence of BIV infection in large-scale serological studies of field samples.  相似文献

目的表达并纯化产肠毒素大肠埃希菌(Enterotoxigenic Escherichia coli,ETEC)CFA/I定居因子的CfaB亚单位,分析其免疫原性。方法将不含信号肽序列的cfaB基因克隆到pQE一30上,构建重组质粒pQE-30-cfaB并转化EcoliM15,表达融合蛋白6xHis—CfaB,将表达的蛋白纯化和复性后免疫BALB/c小鼠,制备抗CfaB的抗血清,用ELISA法检测抗血清效价。结果6×His—CfaB融合蛋白高效表达,相对分子质量为15.5kD,纯化复性后免疫小鼠所得抗血清效价为1:125000。结论成功构建了高效表达6×His-CfaB融合蛋白的重组质粒,表达的融合蛋白免疫小鼠后获得了高效价的抗血清,为进一步研制以双歧杆菌为表达系统的新型ETEC亚单位口服疫苗奠定了基础。  相似文献
