The purpose of this investigation was to analyse the macrophage subpopulations involved in the uptake of endotoxin in the liver. The results show that in normal B10.D2 mice the liver macrophages constitute a heterogeneous population of cells which, depending on their state of differentiation, are distinguished by their differential distribution in the liver acinus and by their ability to phagocytose latex.  相似文献

For the first time V. fluvialis strains were detected on the territory of the USSR. The taxonomic position of these vibrios was determined by their nucleotide DNA composition (the content of guanine + cytosine was 49.3-51.0 mole%) and the characteristic features of their phenotype. The individual features of the strains consisted in their capacity for agglutination with cholera antisera, groups 01 and Inaba, in diagnostic dilutions in the presence of differences in genomes and phenotypes with cholera vibrios. Molecular hybridization DNA-DNA also gave no confirmation of their relationship to cholera vibrios (23-26% homology). The comparative study of V. fluvialis strains from the USSR and other countries by a broader set of their phenotypical signs confirmed their identity.  相似文献

The life spans of individual Saccharomyces cerevisiae cells were determined microscopically by counting the number of buds produced by each cell to provide a measure of the number of cell generations (age) before death. As the cells aged, their generation times increased five- to sixfold. The generation times of daughter cells were virtually identical to those of their mothers throughout the life spans of the mothers. However, within two to three cell divisions after the daughters were detached from their mothers by micromanipulation, their generation times reverted to that characteristic of their own age. Recovery from the mother cell effect was also observed when the daughters were left attached to their mothers. The results suggest that senescence, as manifested by the increase in generation time, is a phenotypically dominant feature in yeast cells and that it is determined by a diffusible cytoplasmic factor(s) that undergoes turnover. This factor(s) appeared to be transmitted by a cell not only to its daughter, but also indirectly to its granddaughter. In separate studies, it was determined that the induced deposition of chitin, the major component of the bud scar, in the yeast cell wall had no appreciable effect on life span. We raise the possibility that the cytoplasmic factor(s) that appears to mediate the "senescent phenotype" is a major determinant of yeast life span. This factor(s) may be the product of age-specific gene expression.  相似文献

While only about ten percent of the databank entries are defined as glycoproteins, it has been estimated recently that more than half of all proteins are glycoproteins. Mucin-type O-glycosylation is a widespread post-translational modification of proteins found in the entire animal kingdom, but also in higher plants. The structural complexity of the chains initiated by O-linked GalNAc exceeds that of N-linked chains by far. The process during which serine and threonine residues of proteins become modified is confined to the cis to trans Golgi compartments. The initiation of this process by peptidyl GalNAc-transferases is ruled by the sequence context of putative O-glycosylation sites, but also by epigenetic regulatory mechanisms, which can be mediated by enzyme competition. The cellular repertoir of glycosyltransferases with their distinct donor sugar and acceptor sugar specificities, their sequential action at highly-ordered surfaces, and their localizations in subcompartments of the Golgi finally determine the cell-specific O-glycosylation profile. Dramatic alterations of the glycosylation machinery are observed in cancer cells, resulting in aberrantly O-glycosylated proteins that expose previously masked peptide motifs and new antigenic targets. The functional aspects of O-linked glycans, which comprise among many others their potential role in sorting and secretion of glycoproteins, their influence on protein conformation, and their multifarious involvement in cell adhesion and immunological processes, appear as complex as their structures.  相似文献

Adults provisioning dependent young are in conflict with their partners, who would prefer a greater level of effort, and with their offspring, who would prefer a greater supply of food. To what extent, then should adults negotiate their provisioning behaviour with other family members? We used experimental manipulations of brood size, and targeted playback of begging calls to determine the extent to which adult great tits Parus major adjust their provisioning rates in response to the behaviour of their partner and their brood. We found that males and females behaved similarly, both responding more to each other's behaviour than to chick calling. We also found that the degree to which adults negotiated their provisioning rates with each other varied between years. A review of the literature suggests that the extent of negotiation over provisioning is likely to vary not only between species of diverse taxa, but also between and within (this study) populations of the same species. We suggest that provisioning behaviour lies on a 'negotiation continuum', which describes the extent to which parents respond to the actions of other family members. We argue that an individual's location on the 'negotiation continuum' is determined partly by the extent to which it can physically respond to the behaviour shown by other members of the family and partly by the quality of information on offer.  相似文献

Using chimeras and more discrete exchange mutations of the rat (r) and human (h) gonadotropin receptors, we had previously identified multiple noncontiguous residues of the lutropin (LHR) and follitropin (FSHR) receptors that dictate their rates of internalization. Since the internalization of the LHR and the FSHR is driven by their abilities to associate with the nonvisual arrestins, we hypothesized that one or more of the residues previously identified by the internalization assays are involved in the formation of the receptor/nonvisual arrestin complex. In the studies reported herein, we tested this hypothesis by measuring the association of arrestin-3 with a large number of rLHR/hLHR and rFSHR/hFSHR exchange mutants that affect internalization. The results presented show that the same residues that dictate the rate of internalization of these two receptor pairs affect their ability to associate with arrestin-3. Although these residues are located in distinct topological domains, our analyses show that threonine residues in the third intracellular loop of both receptor pairs are particularly important for the formation of the receptor/arrestin-3 complexes and internalization. We conclude that the different rates of internalization of the gonadotropin receptors are dictated by their different abilities to associate with the nonvisual arrestins and that this association is, in turn, largely dictated by the presence of threonine residues in their third intracellular loops.  相似文献

对北京地区蕨类植物区系进行了初步分析,研究表明,北京共有蕨类植物77种,隶属于19科34属,以蹄盖蕨科、岩蕨科、鳞毛蕨科、卷柏科等为优势科,以岩蕨属、卷柏属、铁角蕨属等为优势属,科内及属内分化程度较低,种类组成贫乏;地理成分以温带成分占优势,有11属为温带分布属,占非世界分布属总数的61.11%,除世界种和中国特有种外,全为温带性质的种;生境特点以石生为主,大多数种类分布在北京西部和北部的山区;与河北的关系最为密切,其次为山西、内蒙、辽宁、山东,与秦岭也有一定的联系,与西藏的关系疏远。  相似文献

Glycolipids constitute a complex family of amphipathic molecules structurally characterized by a hydrophilic mono- or oligo-saccharide moiety linked to a hydrophobic ceramide moiety. Due to their asymmetric distribution in cell membranes, exposing the saccharide moiety to the extracytoplasmic side of the cell, glycolipids participate in a variety of cell-cell and cell-ligand interactions. Here we summarize aspects of the cell biology of the stepwise synthesis of the saccharide moiety in the Golgi complex of cells from vertebrates. In particular we refer to the participant glycosyltransferases, with emphasis on their trafficking along the secretory pathway, their retention and organization in the Golgi complex membranes and their dependence on the Golgi complex ultra structural organization for proper function.  相似文献

The inhibition of two-dimensional growth in the gametophytesof Asplenium nidus induced by purine and pyrimidine analoguesand the reversal of inhibition by natural purine and pyrimidinebases and their derivatives have been studied. Adenine and guanineand their ribosides and ribotides were more effective than cytosine,uracil, thymine, and their derivatives in preventing the inhibitiondue to 8-azaadenine and 8-azaguanine. Likewise, the inhibitoryeffects of 2-thiocytosine, 2-thiouracil,6-azauracil, and 5-fluorouracilwere overcome by the pyrimidines and their derivatives, butnot usually by the purines.Combinations of two purine analoguesor two pyrimidine analogues or one purine analogue and one pyrimidineanalogue inhibited growth more effectively than single compounds.The combined inhibitions were maximally reversed when both naturalbases or their derivatives were added to the medium. It is concludedthat there is a requirement for both purines and pyrimidinesof ribonucleic acid in the induction of two-dimensional growthin the gametophytes of Asplenium nidus.  相似文献

Primoridal germ cells (PGCs) from the circulating blood in 2-day chick embryos were observed by histochemical techniques in smear preparation and by phase contrast microscopy in fresh samples. In the blood smear, PGCs were readily distinguished from blood cells by their large size (15–20 μm in diameter) and by the round and large nuclei (8–10 μm in diameter) occupying eccentrically the greater parts of the cells. Histochemically, they were demonstrated to contain abundant glycogen, a lesser amount of yolk granules (by the PAS reaction), and a large amount of lipid droplets (by the Oil red O and Sudan black B stains) in their cytoplasm. Alkaline phosphatase activity was proved slightly in their cytoplasm by the simultaneous diazo coupling method. In phase contrast microscopy, living PGCs in the blood and from the primitive gonad of 3-day embryos in parts of the examinations were observed to change their shape in somewhat amoeboid fashion, suggesting their capability for locomotion as a possible mechanism of the migration of PGCs in vivo.  相似文献

We used the comparative method to examine the evolutionary causesof group-living in the New World cursorial hystricognath rodents.To do so, we used the available literature to collect informationon behavioral (group size, burrow digging), ecological (amountof plant cover in the habitat), and life history (body mass,time to sexual maturity) variables, along with phylogeneticrelationships of these rodents. We analyzed these variablesin the context of three major hypotheses. A first explanationposes that rodents live in groups to reduce the energy neededin the construction of their burrows. A second hypothesis suggeststhat grouped rodents increase their ability to detect and escapefrom predators. A third possibility states that group-livingis adopted by rodents to provide extra parental care to their offspring. Our comparative analysis revealed that across speciesvariation of group size is, to some extent, influenced by bodysize, and by the habit of burrow digging. Thus, large sizedrodent species that actively dig their own burrows form largergroup sizes than small sized species that do not dig burrows.In contrast, across species variation of group size was not influenced by differences in the amount of plant cover in thehabitat (an indirect measure of predatory risk), or by differencesin the time to first reproduction (a measure of parental caregiven). Therefore, group-living among the New World histricognathrodents seems more linked to a strategy aimed to reduce theirburrowing cost than to a strategy aimed to reduce their predatory risk, or to extend their parental investment.  相似文献

Opioid receptors belong to the super family of G-protein coupled receptors (GPCRs) and are the targets of numerous opioid analgesic drugs. Prolonged use of these drugs results in a reduction of their effectiveness in pain relief also called tolerance, a phenomenon well known by physicians. Opioid receptor desensitization is thought to play a major role in tolerance and a lot of work has been dedicated to elucidate the molecular basis of desensitization. As described for most of GPCRs, opioid receptor desensitization involves their phosphorylation by kinases and their uncoupling from G-proteins realized by arrestins. More recently, opioid receptor trafficking was shown to contribute to desensitization. In this review, our knowledge on the molecular mechanisms of desensitization and recent progress on the role of opioid receptor internalization, recycling or degradation in desensitization will be reported. A better understanding of these regulatory mechanisms would be helpful to develop new analgesic drugs or new strategies for pain treatment by limiting opioid receptor desensitization and tolerance.  相似文献

The preservation of historical and contemporary data safeguards our scientific legacy. Bioacoustic recordings can have historical as well as scientific value and should be assessed for their conservation requirements. Unpreserved bioacoustics recordings are generally not referenced and are frequently at high risk of loss by material degradation and/or by misplacement. In this study we investigated the preservation status of sets of natural sound recordings made in Portugal from 1983 until 2010 inclusive. We evaluated the recordings on the basis of their rate of loss, the degree to which unpreserved recordings could be preserved, and their risk of loss. Recordists of animal sounds were surveyed (by questionnaire or interview) to identify sets of recordings and to collect information on their quality and state of preservation. Of the 78 recordists identified, we found that 32% of the recordings have an unclear status and that only 9% of the recordings are lost. Of the c. 6 terabytes of unpreserved sound recordings discovered, an estimated 49% were recoverable. Moreover, 95% of the recoverable sets of recordings were at high risk of loss by their being misplaced. These risks can be minimized if recordists are persuaded to deposit their material in an institution committed to long-term curation of such data (e.g. sound archives). Overall, the study identified a considerable body of unpreserved animal sound recordings that could contribute to our scientific heritage and knowledge of the biodiversity found in Portugal. It highlights the need to implement effective policies to promote the deposit of recordings for preservation and to reverse the present scenario so that scientific material can be preserved for future generations.  相似文献

Masquerading prey closely resemble inedible objects found in the same locality. These animals gain protection from their predators by causing their predators to misclassify them as the inedible ‘models’ that they appear to resemble. We recently demonstrated that predators are more likely to misclassify masquerading prey as their models when masqueraders are viewed in isolation from their models than when they are viewed simultaneously with examples of their models. Using domestic chicks (Gallus gallus domesticus) as predators and the twig-mimicking caterpillars of the Early Thorn Moth (Selenia dentaria) as prey, we tested whether this effect was influenced by the relative orientations of models and masqueraders; and the distance from which models and masqueraders could be viewed simultaneously. We found no effect of orientation, but that the cost to masqueraders of being viewed simultaneously with an example of the model declined as the distance between the model and masquerader increased. These results are interpreted in terms of animal cognition, and their implications for the evolutionary ecology of masquerade.  相似文献

It is argued that those neural systems (such as that responsible for stereoscopic vision) that have the greatest precision of operation are the most likely, during their developmental construction, to take advantage of infromation supplied by their own input. There is evidence that binocularly driven neurons in the kittens's visual cortex do indeed become modified in their synaptic organization during early visual experience in a manner that enhances the specificity of binocular interaction and ensures that the ranges of positional and orientational disparities of the receptive fields, within limits, become matched to the nature of the actual stimulation encountered by the animal.  相似文献

Females of the hump earwig, Anechura harmandi, are completely consumed by their offspring at the end of their care (matriphagy). The effect of this matriphagy was assessed by manipulative experiments. Matriphagy led to a delay in the dispersal of the nymphs and an increase in their survival rate. The same results were obtained when mothers were removed and the nymphs were given sufficient food. Females separated from their offspring after larval hatching failed to produce a second clutch, and three-quarters of them did not develop their ovaries. These results suggest that the survival of nymphs and their stay in the nest are dependent on food availability and that A. harmandi females are strictly semelparous.  相似文献

The use of the discrete dynamic method for the treatment of data obtained in the survey of streptococcus carriers has made it possible to find out that their immune status is determined not so much by the quantitative changes in the results of individual immunological tests (for the bactericidal activity of the blood serum, lysozyme, IgG, IgM, IgA and the phagocytic activity of neutrophils), but, to a greater extent, by the interrelations of these characteristics. Significant differences in the interrelations of various humoral characteristics and in their relationship to the phagocytic process have been detected in the group of carriers as compared with the control group.  相似文献

Three pure lines of lettuce differing in their seed germinationbehaviour were crossed, and the germination of their F2 andF3 progeny was studied. The pattern of segregation of the progenywith respect to seed germination indicates that factors controllinggermination could be transmitted in a normal mendelian manner,and that the difference between these lines in their germinationbehaviour could not be caused by the seed and fruit coats.  相似文献
