The metabolism of [U–14C] 1-aminocyclopropane-1-carboxylicacid (ACC) supplied to whole fruits of apple (Malus domesticaBorkh., cv. Cox's Orange Pippin) was investigated. Radioactiveethylene was recovered in mercuric acetate traps and an acidicmetabolite was formed in proportions which varied little withthe absolute amount of substrate supplied. The amount of ACCusually supplied did not cause immediate, rapid ethylene productionby mature, pre-climacteric fruit but the onset of productionwas earlier than in untreated fruit. The radioactive acidic metabolite was purified by four chromatographicprocedures and activity was coincident with authentic 1-malonylamino)cyclopropane-1-carboxylic acid (MACC). The presence of thiscompound was confirmed by gas chromatography linked to massspectrometry. MACC was a major metabolite of [14C] ACC supplied to applesthroughout fruit development. The proportion converted to ethylenewas low but increased with endogenous ethylene production inthe final samples. MACC was shown to be a natural constituent of apple fruits andto accumulate to the amol kg–1 level. The influence of storage temperature on the onset of rapid ethyleneproduction was investigated for fruits of Conference pear (Pyruscommunis L.) and five cultivars of apple (Malus domestica Borkh.).The time taken from harvest to rapid ethylene production wasshorter and more uniform at 3 ?C than at 18–20 ?C forConference pears and Golden Delicious apples. Increases in internalethylene concentration, 1-amino cyclopropane-1-carboxylic acidconcentration and ethylene production were simultaneous in GoldenDelicious apples at 3 ?C. When Golden Delicious apples wereheld at 3 ?C for 48 h and then kept at 20 ?C the mean time ofonset of ethylene production was similar to that for applesheld continuously at 20 ?C. However, two periods of 48 h at3 ?C caused earlier ethylene production. Conversely, ethyleneproduction at 3 ?C was delayed by transfer to 20 ?C for twoperiods of 48 h. Cox's Orange Pippin and other apple cultivarstended to show more synchronous ethylene production at 3 ?Cthan at higher temperatures but the mean time of onset was eitherunaffected by temperature or slighdy delayed at lower temperature.Acceleration of the onset of ethylene production by low temperaturewas never observed in Cox's Orange Pippin apples harvested atweekly intervals from 10 August to 17 September. A system was developed to test the effects of floodwater O2concentration on ethylene evolution and stem lenticel hypertrophy,and the effects of exogenous ethylene on stem lenticel hypertrophyin mango (Mangifera indica L.) trees. Dissolved O2 concentrationsof 1–7x10–9 m3 m–3 generally resulted in hypertrophyof stem lenticels within about 6 d of flooding, whereas floodwaterO2 concentrations of 13–15 x 10–9 m3 m–3 delayedhypertrophy until about day 9. After 14d of flooding, therewere more than twice the number of hypertrophied lenticels pertree with floodwater O2 concentrations of 1–7 x 10–9m3 m–3 than with floodwater O2 concentrations of 15 x10–9 m3 m–3. Ethylene evolution from stem tissueimmediately above the floodline increased 4- to 8-fold in treesexposed to floodwater O2 concentrations of 1–2 x 10–9m3 m–3, increased 2-fold for trees exposed to floodwaterO2 concentrations of 6–7 x 10–9 m3 m–3, butremained constant with floodwater O2 concentrations of 13–15x 10–9 m3 m–3. Plants maintained in highly oxygenatedfloodwater (13–15 x 10–9 m3 m–3), and givenexogenous ethylene developed many hypertrophied lenticels, whereasplants in highly oxygenated water and not given ethylene developedfewer or nohypertrophied lenticels. These data suggest thatethylene plays a role in promotion of stem lenticel hypertrophyin flooded mango trees, and that floodwater dissolved oxygenconcentration can regulate stem lenticel hypertrophy and ethyleneevolution in this species. Epidermal strips and leaf fragments of Commelina and leaf fragmentsof maize were incubated on solutions containing naturally-occurringor synthetic cytokinins and/or ABA. The effects of these treatmentson stomatal behaviour were assessed. Cytokinins alone did notpromote stomatal opening in either species but concentrationsof both zeatin and kinetin from 10–3 to 10–1 molm–3 caused some reversal of ABA-stimulated closure ofmaize stomata. The reversal of the ABA effect increased withincreasing cytokinin concentration. Cytokinins had no effecton ABA-stimulated closure of Commelina stomata. When appliedalone, at high concentration (10–1 mol m–3), toCommelina epidermis or leaf pieces both zeatin and kinetin restrictedstomatal opening. Carbon dioxide efflux from 5- to 20-day-old pea fruits was measuredfor plants grown in controlled environment at 15 °C and600 µmol s–1 m–2 photon flux density in a16 h photoperiod. The rate of CO2 output per fruit increasedquickly from 0.005 to 0.018 mg CO2 min–1 during fruitelongation and subsequently more slowly to 0.030 mg CO2 min–1as the fruits inflated. On a d. wt basis the rate was highest,0.175 mg CO2 g–1 min–1, in the youngest fruits anddeclined curvilinearly with increasing fruit weight to 0.02mg CO2 g–1 min–1. Separation of maintenance andgrowth components was achieved by starvation methods and bymultiple regression analysis. From the latter method estimatesof the maintenance coefficient declined hyperbolically from150±8.7 mg carbohydrate g–1 d. wt day–1 inthe very young fruits (0.05 g) to 10.4±0.36 mg carbohydrateg–1 d. wt day–1 in older fruits (2.0 g). On a nitrogenbasis maintenance costs decreased from 2240 to 310 mg carbohydrateg–1 nitrogen day–1 while nitrogen concentrationfell from 6.7 to 3 per cent d. wt. A simple linear relationshipbetween maintenance cost per unit d. wt and nitrogen concentrationwas not observed. A growth coefficient of 50±6.7 mg carbohydrate g–1growth (equivalent to a conversion efficiency, YG, of 0.95)was estimated for all fruits examined. The overall efficiency, Y, increased from a mean of 0.70 to0.85 during fruit elongation and subsequently declined to 0.80.For a given fruit weight, efficiency increased asymptoticallywith relative growth rate; both asymptote and slope of the relationshipincreased as the fruits grew. The uptake of sorbitol into vacuoles from immature flesh ofapple fruit (Maluspumila Mill, var domestica Schneid.) was facilitatedby 10–6 M ABA, while such uptake into protoplasts wasnot stimulated. Enrichment of CO2 to 46 mmol m–3 (1 000 mm3 dm–3)at a moderate photosynthetic photon flux (PPF) of 450 µmolm–2 s–1 stimulated fresh and dry weight gain oflettuce leaves 39% to 75% relative to plants at 16 mmol m–3CO2 (350 mm3 dm–3). Relative growth rate (RGR) was stimulatedonly during the first several days of exponential growth. ElevatingCO2 above 46 mmol m–3 at moderate PPF had no further benefit.However, high PPF of 880–900 µmol m–2 s–1gave further, substantial increases in growth, RGR, net assimilationrate (NAR) and photosynthetic rate (Pn), but a decrease in leafarea ratio (LAR), at 46 or 69 mmol m–3 (1000 or 1500 mm3dm–3) CO2, the differences being greater at the higherCO2 level. Enrichment of CO2 to a supraoptimal level of 92 mmolm–3 (2000 mm3 dm–3) at high PPF increased leaf areaand LAR, decreased specific leaf weight, NAR and Pn and hadno effect on leaf, stem and root dry weight or RGR relativeto plants grown at 69 mmol m–3 CO2 after 8 d of treatment.The results of the study indicate that leaf lettuce growth ismost responsive to a combination of high PPF and CO2 enrichmentto 69 mmol m–3 for several days at the onset of exponentialgrowth, after which optimizing resources might be conserved. The internal cycling of nitrogen (N) has been studied in applerootstocks grown in sand culture and subjected to a constantN supply, or defoliation, or withholding the N supply in theautumn in order to manipulate the amount of N stored over thewinter. The trees subsequently received either no N or 8–0mol N m–3 (labelled with 15N to 498 atom%) with the irrigationthe following spring in order to determine the effect of thecurrent N supply on the remobilization of N for leaf growth. Provision of an autumnal N supply delayed leaf senescence andreduced the amount of N withdrawn from leaves from 156 mg Nplant–1 to 91 mg N plant–1. Loss of protein ribulose1,5-bisphosphate carboxylase/oxygenase (RUBISCO) accounted for83–87% of the soluble protein N lost during leaf senescence,there being a preferential loss of RUBISCO compared with othersoluble leaf proteins. Remobilization of N from perennial woody tissues (stems androots) in the spring was used predominantly for leaf growth.The amount of N remobilized depended upon the size of the Nstore, but was unaffected by the current N supply, demonstratingthat fertilization of trees does not alter the efficiency withwhich they cycle N. Degradation of RUBISCO in the autumn accountedfor between 32% and 48% of the N subsequently remobilized forleaf growth the following spring, suggesting that RUBISCO hasa role as a summer store for N. Two groups of tiger salamanders (Ambystoma tigrinum) were conditionedto respond to odorant-air mixtures of n-butyl acetate (8.9 x10–5M) or n-butyl alcohol (6.7 x l0–5M). They werethen given tests with various concentrations of the trainingodorants presented using a temporal forced-choice method ofascending limits. Results showed that reliable responses toodorant-air presentations were obtained with concentrationsof n-butyl acetate above 2.4 x l0–7M and with concentrationsof n-butyl alcohol above 8.5 x 10–8M. Growth and ripening of strawberry (Fragariaananassa Duch.)fruit harvested at immature stages of development was accomplishedby placing the peduncles of individual fruit in solutions composedof hydroxyquinoline hemisulphate (HQS) and sucrose. Fruit cultivarand developmental stage at harvest were the major determinantsof in vitro performance. ‘Pajaro’ fruit harvestedat 50 to 60% maturity exhibited the greatest and most uniformweight gain when placed in solutions containing 200 mol m–3HQS and 88 mol m–3 sucrose. Although the final fruit weightof in vitro-ripened fruit was less than that of field-ripenedfruit, colour development in vitro occurred at the same rateand to the same extent as field-grown fruit. Apparatus is described for exposing plants to low concentrationsof SO2 (50–500µg m–3 in air) and for measuringeffects on photosynthesis, dark respiration, and transpiration.Temperature, humidity, and irradiance in the chambers were controlledindependently, and fans ensured that leaf boundary layer resistanceswere low. Experiments with plants of Vicia faba in clean andpolluted air showed that: (i) a depression of net photosynthesisby 50 µg m–3 SO2 depended on boundary layer resistanceand on irradiance; (ii) stomatal resistance was increased ordecreased by 50 µg m–3 SO2 when relative humidityin the chambers was low (35% r.h., 22 °C) or high (50% r.h.,22 °C) respectively.  相似文献

Ingestion and clearance rates, feeding behaviors and life historyvariables of the marine cladoceran Penilia avirostris were evaluatedover a range of food concentrations encountered in nature (0.01–3.0mm3 1–1 of Isochrysis galbana). Ingestion rates increasedand clearance rates decreased with increasing food concentration.No maximum feeding thresholds were observed over the range ofalgal concentrations offered. Weight-specific ingestion ratesdecreased with increasing body weight. Feeding behaviors suchas mandibular activity, abdominal claw rejections of cloggedfeeding structures and feeding appendage activity decreasedat a food level of 0.3 mm3 l–1 of l. galbana. Peniliaavirostris had very poor survivorship at extremely low (0.01mm3 l–1 and high (3.00 mm3 l–1) food levels. Simultaneous measurement of export from leaves and import tofruits were made on tomato plants reduced to one fully expandedleaf and one fruit. Experimental leaves were exposed to sixlight flux densities (0.5–100 W m–2) for 24 h whilerapidly growing fruits were kept in the dark at 22 °C. The rates of export of assimilate from these leaves varied from70 to 120 mg C leaf–1 day–1 corresponding with ratesof carbon fixation from 3 to 290 mg C leaf–1 day–1.Export from leaves with the lowest carbon fixation rates weremaintained by a loss of up to one-sixth of their initial carbon.In contrast, leaves with the highest carbon fixation rates exportedonly half the newly fixed carbon. The rates of import of assimilate to similar-sized fruits (c.16 cm3) were between 80 and 110 mg C fr–1 day–1but differed from the export rates of the source leaves. Thespecific growth rates and the specific respiration rates ofthe fruits were related to their initial carbon content at thebeginning of the experiment. Thus, over 24 h, the rate of importwas predetermined by the developmental stage of the fruit unalteredby the rate of current carbon fixation in the source leaf. Translocationof assimilate was regulated by sink demand under both source-and sink-limiting conditions in this short-term situation. The dynamic relationship between assimilate production in leavesand its utilization in fruits is discussed together with therole of sucrose concentration in these organs in regulatingtransport. The ion relations of the halophytc Suaeda maritima are described.When plants grew in 340 mol m–3 sodium chloride (—1•76MPa) leaf solute potentials decreased, and were sustained around—2•5 MPa Inorganic ion concentration (mostly of sodiumchloride) accounted for this. Comparable shoot ion concentrationsof potassium, nitrate and sulphate occurred when plants grewon different salinity types characterized by these ions. Netsodium transport and shoot sodium concentration increased dramaticallywith increases in external sodium chloride concentration upto 85 mol m–3; thereafter, further increases in externalsodium chloride concentration had relatively little effect uponeither shoot sodium concentration or upon net transport of sodiumto the shoot. The net transport of sodium plus potassium onlydoubled when the external concentration of sodium plus potassiumincreased from 24 to 687 mol m–3 Shoot ion concentrationswere remarkably constant with time, external concentration andsalinity type. The membrane flux rates and symplasmic ion concentrations neededto sustain the observed net transport of sodium (of some 10mmol g–1 dry wt. of roots d–1) are calculated fromanatomical and stereological data for the root system of thisspecies. The minimum net sodium chloride flux to load the symplasmwould be 260 nmol m–2s–1 if the whole cortical andepidermal plasmalemmal surface area were used uniformly, butthe flux rate required would be 3000 nmol m–2s–1if uptake took place only at the root surface. A flux rate ofat least 1000 nmol m–2s–1 is needed between symplasmand xylem. The symplasmic concentration of NaCl would be atleast 80 mol m–3. It is argued (1), that both symplasmicand xylem loading are likely to be passive processes mediatedby ion channels rather than active carriers, (2), that net iontransport at 340 mol m–3 sodium chloride is close to themaximum which is physiologically sustainable and (3), that growthof this halophyte is limited by NaCl supply from the root. Guttation was used as a non-destructive way to study the flowof water and mineral ions from the roots and compared with parallelmeasurements of root exudation. Guttation of the leaves of barley seedlings depends on age andon the culture solution. Best rates of guttation were obtainedwith the primary leaves of 6- to 7-day-old seedlings grown onfull mineral nutrient solution. The growing leaf tissue becomessaturated with K+ below 1.5 mM K+ in the medium, whereas K+concentration in the guttated fluid still increases furtheras K+ concentration in the medium is raised. At 3 mM K+ averagevalues of guttation were 1.4–2.4 mm3 h–1 per plantwith a K+ concentration of 10–20 mM; for exuding plantsthe flow was 4.2–7.6 mm3 h–1 per plant and K+ concentration35–55 mM. Abscisic acid (ABA) at 10–6 to 10–4 M 0–2h after addition to the root medium increased volume flow ofguttation and exudation and the amount of K+ exported. Threeh after addition of ABA both volume and amount of K+ were reduced.There was an ABA-dependent increase in water permeability (Lp)of exuding roots shortly after ABA addition. Later Lp was decreasedby 35 per cent and salt export by 60 per cent suggesting aneffect of ABA on salt transport to the xylem apart from itseffect on Lp. Benzyladenine (5 x 10–8 to 10–5 M)and kinetin (5 x 10–6 M) progressively reduced volumeflow and K+ export in guttation and exudation and reduced Lp. Guttation showed a qualitatively similar response to phytohormonesas found here and elsewhere using exuding roots. Zinc-induced Vacuolation in Root Meristematic Cells of Cereals   总被引:1,自引:0,他引:1  
In the absence of Zn, vacuolar volume fractions of root meristematiccells of Secale cereale L. cv. K2, Triticum aestivum L. cv.Chinese Spring and Oryza sativa L. cv. IR34 were 5.64 x 10–2,2.17 x 10–2 and 1.63 x 10–2 µm3 vacuole µm–3tissue, respectively. A 4-d exposure to a subtoxic concentrationof zine (0.2 µg Zn cm–3) induced a 2.93-fold anda 6.78-fold increase in the total vacuolar volume fraction inOryza and Triticum, respectively, whereas no significant increasewas observed for Secale. It is proposed that this Zn-inducedvacuolation represents a compartmentalization mechanism. Theinitial total vacuolar volume fraction in Secale was greaterthan that for Oryza and Triticum and this may enable compartmentalizationof the metal soon after the onset of treatment so reducing itscytotoxic effects. The effects of light/dark on anion fluxes in isolated guardcells of Commelina communis L. have been studied, using 82Brand 36Cl. Transfer of open guard cells from light to dark hasno effect on the 82Br influx, but produces a marked transientstimulation of 82Br or 36Cl efflux, similar to the effect ofsuch transfer on the 86Rb fluxes, and to the effects on both86Rb and 82Br fluxes of adding ABA. On return of guard cellsto light, after the transient, there is a further reductionin Cl/Br efflux. It is argued that control of a specific processof ion extrusion is important in regulating the ability of guardcells to stay open. In three out of four batches of steady-statetissue labelled with 82Br, the plasmalemma fluxes were highenough, relative to the tonoplast fluxes, for the efflux kineticsto be separable into two exponential components, allowing estimationof bromide contents in cytoplasm and vacuole (Qc and Qv), andfluxes at plasmalemma and tonoplast. With opening in light,Qc increased by 3.9 ± 0.4 pmol mm–2 µm–1and Qy by 5.2 ± 0.6 pmol mm–2 µm–1(change in content per mm2 of epidermis perµm change inaperture). Using rough estimates for the volumes of cytoplasmand vacuole these figures suggest that at 6.1 µm in thedark the concentrations were about 63 mol m–3 in the cytoplasmand 35 mol m–3 in the vacuole, rising to about 185 molm–3 in the cytoplasm and 125 mol m–3 in the vacuole,at 16.7 µm aperture in light. Neither increase can providean adequate increase in salt concentration to account for theosmotic change required, and some solute other than potassiumsalt must also be involved. In one experiment with 82Br andin the only experiment with 36Cl the plasmalemma flux was lower,and not high enough relative to the tonoplast flux to allowseparation of two phases in the efflux curves, and calculationof cytoplasmic and vacuolar contents and fluxes. The rate of carbon import by tomato fruits has been relatedto their carbon metabolism by examining the effects of fruittemperature on the metabolism of imported assimilates. 14C–sucrose,–glucose, –fructose, –malic acid and –citricacid were injected individually into young growing tomato fruitswhich were subsequently maintained at 25 or 5 °C for 48h. Fruit temperature greatly affected the proportions of 14Clost from the fruits by export and respiration. Only 40 percent of the injected 14C from 14C–sugars and 20 per centfrom 14C–acids was recovered from fruits at 25 °C.Less than 10 per cent of the injected 14C was exported, thebalance being respired. In contrast, more than 50 per cent ofthe injected 14C was recovered from cooled fruits, in whichthe import rate of carbon was presumably reduced, and 20–36per cent of injected 14C was exported. Cooling enhanced thesynthesis of 14C–sucrose from injected 14C–hexosesand inhibited the incorporation of 14C into starch and insolubleresidue. When 14C–sugars were injected, radioactivityexported from the cooled fruits was detected as sucrose in thephloem of the peduncles; radioactivity was also detected instems and roots when fruits were cooled. In almost fully–grownfruits injected 14C–compounds were metabolized less readilythan in smaller fruits. Conversion of 14C–hexoses to 14C–sucrosewas again enhanced by cooling (5 °C, but was less in fruitsmaintained at 35 °C than in controls. The structure of the zooplankton biotic community and of copepodpopulation in the coastal area of Terra Nova Bay (Ross Sea,Antarctica) was investigated during the 10th Italian AntarcticExpedition (1994/1995). Zooplankton biotic community consistedmainly of pteropods (Limacina helicina and Clione antarctica),Cyclopoid (Oithona similis), Poecilostomatoid (Oncaea curvata)and Calanoid (Ctenocalanus vanus, Paraeuchaeta antarctica, Metridiagerlachei and Stephos longipes) copepods, ostracods, larvalpolychaetes and larval euphausiids. Zooplankton abundance rangedfrom 48.1 ind m–3 to 5968.9 ind m–3, and copepodabundance ranged from 45.2 ind m–3 to 3965.3 ind m–3.The highest peak of zooplankton abundance was observed between25 m and the surface and was mainly due to the contributionof O. similis, O. curvata and C. vanus. Zooplankton biomassranged from 5.28 mg m–3 to 13.04 mg m–3 dry weight;the maximum value was observed between 25 m and the surface.Total lipid content varied from 216.44 to 460.73 mg g–1dry weight.  相似文献

Rapid, Reversible Inhibition of Nitrate Influx in Barley by Ammonium   总被引:18,自引:2,他引:16  
The rate of influx of nitrate into the roots of intact barleyplants was measured over a period of 3–5 min from externalnitrate concentrations of 1–150 mmol m–3, using13N-labelled nitrate as tracer. Ammonium at external concentrationsof 0.005–50 mol m–3 inhibited nitrate influx ina manner which did not conform to a simple kinetic model butincreased approximately as the logarithm of the ammonium concentration.At any particular ammonium concentration, inhibition of nitrateinflux reached its full extent within 3 min of the ammoniumbeing supplied and was not made more severe by up to 17 minpre-treatment with ammonium. On removing the external ammonium,nitrate influx returned to its original rate within about 3min. Potassium at 0.005–50 mol m–3 did not reproducethe rapid effect of ammonium on nitrate influx. Net uptake of nitrate also decreased when ammonium was supplied,over a similar timescale and to a similar extent as nitrateinflux.
