The fungus associated with the Japanese horntail,Urocerus japonicus, in Kochi, Kagawa and Ehime Prefectures was studied. Cultures isolated from the mycangia of 113 adult females of the horntail showed the same cultural characteristics. Four of basidiocarps found on felled logs ofCryptomeria japonica were identifieds asAmylostereum laevigatum based on morphological characteristics. This was the first record ofA. laevigatum from Japan. The cultures isolated from the basidiocarps had the same cultural characteristics as those from the mycangia ofU. japonicus. One mycangial isolate produced basidiocarps on artificially inoculated stem segments ofCr. japonica after a 6-mo incubation and was identified asA. laevigatum. One isolate from the basidiocarps ofA. laevigatum and one from the mycangium ofU. japonicus were artificially inoculated into five trees each ofChamaecyparis obtusa andCr. japonica. The wood of all inoculated trees showed discoloration, with no difference in shape and pattern of discoloration between the two isolates. The inoculated fungi were reisolated from the areas of discoloration in the inoculated trees.  相似文献

Analyses of DNA sequences from the internal transcribed spacer (ITS) region of the nuclear rDNA and from a portion of a manganese-dependent peroxidase gene were used to assess the species inAmylostereum, including isolates from the mycangia of horntails, decay, and basidiomes. Four species are recognized:A. areolatum, A. chailletii, A. laevigatum, andA. ferreum. An unidentifiedAmylosterum isolate from the mycangium ofXoanon matsumurae had an ITS sequence identical to that ofA. areolatum. Another unidentifiedAmylostereum isolate from the mycangium ofSirex areolatus was nearA. laevigatum, which appears to be the mycangial symbiont for those horntails attacking cedar-like trees. The other horntail isolates, primarily from Pinaceae, proved to be eitherA. areolatum orA. chailletii. The DNA sequences ofEchinodontium tinctorium, E. tsugicola andE. japonicum were similar to those of theAmylostereum species, andAmylostereum species are now recognized as members of the family Echinodontiaceae rather than the family Stereaceae.Echinodontium taxodii was found to be distinct from the Echinodontiacaea andStereum, andE. taxodii is recognized as aLaurilia species.  相似文献

A fungus was found to be stored in the mycangia of a horntail,Tremex longicollis, as hyphal fragments. All fungal isolates from the mycangia of 31 adult females of the horntail produced the same colonies on PDA. Basidiocarps ofCerrena unicolor occurred near the emergence hole of the horntail on a dead hackberry tree (Celtis sinensis). The cultures of this fungus were similar to those from the mycangia of the horntail in cultural characteristics. Mating between single-basidiospore mycelia ofC. unicolor and single-arthrospore mycelia from the mycangia of the horntail showed that they were compatible. These results revealed that the fungus isolated from the mycangia ofT. longicollis wasC. unicolor.  相似文献

The dynamics of the fungal symbionts in the gallery system and the mycangia of the ambrosia beetle,Xylosandrus mutilatus, were studied in relation to its life history using both isolation experiments and scanning electron microscopy (SEM). In the galleries,Ambrosiella sp. was predominant during the larval stages but its relative dominance gradually decreased during the development of the larvae. In contrast, yeasts (mainlyCandida sp.) andPaecilomyces sp. dominated continuously in the galleries after eclosion.Ambrosiella sp. was consistently stored in the mycangia in all adult stages, except in the teneral and overwintering adults when the other fungi were dominant. No fungal spores occurred in the mycangia of the adult beetles reared under aseptic conditions from the pupal stage, while onlyAmbrosiella sp. was stored in those reared from the teneral-adult stage. These results suggest that: (i) Xmutilatus is associated with at least three fungal species, among whichAmbrosiella sp. is the most essential food resource for development of the broods; (ii) immediately after eclosion, new female adults may take at least four associated fungal species, with no or incomplete selection, into their mycangia from the walls of the cradles; and (iii) conditions may well be produced in the mycangia of both matured and dispersing beetles whereby only the spores ofAmbrosiella sp. can proliferate.  相似文献

Protocorms or protocorms with roots of an achlorophyllous orchidGaleola septentrionalis were inoculated with isolates ofRhizoctonia repens, R. solani, andRhizoctonia spp. The seedlings were infected with eight of twelve isolates ofR. repens. Fungal coils were formed in the cells, which was suggestive of a symbiotic association. The other isolates caused soft rot or no infection to the protocorms or the protocorms with a root. A study of the type specimen ofUstilago jehudana resulted in the correction of the diagnosis. The sori are localized in the host gynoecium but not in the anthers. Morphological characters of the sori and ustospores of the later describedU. moenchiae-manticae are identical with these ofU. jehudana. Ustilago moenchiae-manticae is reduced here to a synonym ofU. jehudana. The smut is reported as new to Bulgaria on a new host, viz.,Moenchia erecta. Two naturalized dandelions,Taraxacum officinale andTaraxacum laevigatum, were grown in mixed stands at various plant densities and mixing ratios with various levels of fertilizer supply until seeds were produced. The effect of fertilizer supply on the competitive relationship for seed production between the species was analyzed. In pure stands, the simultaneous effects of plant density and fertilizer supply on mean plant weight were formulated for each species. By introducing the density conversion factor, the results of plant weight growth in mixed stands were formulated successfully.T. laevigatum produced more seeds thanT. officinale at the same plant weight. The relationship between mean plant weight and mean number of mature seeds produced per plant was also formulated for each species. Ratio diagrams were predicted with these formulas. The density conversion factors showed thatT. officinale was always dominant overT. laevigatum for plant weight growth within a generation. The ratio diagrams showed thatT. officinale became dominant overT. laevigatum for seed production with repeating generations at higher levels of fertilizer supply, but that the species with a larger ratio of plant density became dominant over the other species at lower levels of fertilizer supply.  相似文献

The status ofLymantria obfuscata Walker in India is discussed. Notes on its taxonomy, distribution, life-history and economic importance are given for comparison with those of the gypsy moth,Lymantria dispar (L.). Parasites ofL. obfuscata in India are reviewed. A survey in 1961–66 revealed an additional 50 species of natural enemies ofL. obfuscata in India. The incidence and biology of some of the important ones are presented. A comparison is made between these parasites and those ofL. dispar. Some of the natural enemies ofL. obfuscata already attackL. dispar in America. Others are closely related to the parasites of the gypsy moth. The scope for utilisation of parasites ofL. obfuscata in other countries is discussed. Uranoscopus flavipinnis sp. nov. is described based on 39 specimens from the coasts of southern Japan and Taiwan. It differs from otherUranoscopus species in having the following combination of external characters: posterior nasal valve tubular, as long as anterior one; nape naked between the lateral lines; body reddish brown with irregular yellow spots. Previously,U. flavipinnis was wrongly identified asU. japonicus Houttuyn, 1782 (=U. asper Temminck et Schlegel, 1843). This new species occurs from the South China Sea northward to Ibaraki and Niigata Prefectures, Japan. Since the type specimen ofU. japonicus has been lost, one of the present specimens, HUMZ 109237, is designated as the neotype ofU. japonicus to stabilize the nomenclature.Uranoscopus japonicus is redescribed and compared withU. flavipinnis.  相似文献

Some phytophagous insects have been known to inoculate certain fungi on plant substrates. In many cases of such insect–fungi relationships it has been considered that fungi contribute to insects by decomposing lignin or polysaccharides, and that the insects feed on the decomposition products or fungi themselves. Females of the leaf-rolling weevil in the genus Euops (Attelabidae) store spores of symbiotic fungi in the mycangia and inoculate them on leaf rolls. To determine the effect of mycangial fungi on larval nutrition in E. lespedezae, the nutritional value was compared between leaves with and without mycangial fungi. Two Penicillium species were isolated from the mycangia. These mycangial fungi showed little effect on the decomposition of lignin and polysaccharides, and showed little effect on enhancement of soluble sugars within leaves. Thus, the mutualism between Euops and its mycangial fungi contrasts with the mainly nutritional mutualisms between wood-infesting insects (termites, bark/ambrosia beetles, and wood wasps) and lignin/polysaccharide-decomposing fungi.  相似文献

The present communication deals with the observations made on the morphology, reproduction and cytology ofUlothrix zonata (Weber & Mohr)Kütz. in culture. The alga displays a remarkable phenotypic plasticity in nature as well as in culture. The present study provides additional evidence of karyology in support ofLokhorst andVroman's treatment ofU. zonata which merges a number of earlier described species in it. Sclerotium rolfsii isolates from peanut fields in Ibaraki were classified into mycelial compatibility groups (MCGs) based on the barrage zone formation. A total of 132 isolates collected from four fields within a 120 m radius in 1994 comprised four MCGs; MCG A (71 isolates), B (34 isolates), C (26 isolates) and D (one isolate). Fields 1 and 2 were occupied exclusively by MCG A. MCG A also predominated in field 3. In field 4, MCGs A, B and C were dominant. Population structure in 3 additional fields was determined in 1997. All 11 isolates from Field 5, which was 400 m distant from field 1, belonged to MCG C. A total of 42 isolates from fields 6 and 7, 2.5 km distant from other fields and 100 m distant from each other, were all MCG A. These results suggested that the population structure ofS. rolfsii was simple. RAPD fingerprintings showed that most isolates of the same MCG were clonal.  相似文献

A new aporocotylid blood fluke is described, based on specimens from the ventricle of the Pacific bluefin tuna, Thunnus orientalis (Temminck et Schlegel), cultured in Wakayama and Nagasaki Prefectures, Japan. The new species is morphologically similar to the members of the genus Cardicola Short, 1953, but shows distinct differences in the body form, location of the testis and the orientation of the ootype. The body of the new species is long and slender, whereas other Cardicola species are small and generally lanceolate. The testis is mostly located posterior to the caeca and anterior to the ovary, occupying 31–45% of body length, in contrast to the known Cardicola species, whose testis is typically intercaecal. The ootype is oriented anteriorly, while in most congeners, it is directed posteriorly or horizontally. Phylogenetic analyses of this aporocotylid, together with Cardicola orientalis Ogawa, Tanaka, Sugihara et Takami, 2010 from the same host, were conducted based on DNA sequences of the ITS2 rDNA and the 28S region of ribosomal RNA. The analyses revealed that the new blood fluke belongs to the genus Cardicola despite the marked morphological differences. Thus, this aporocotylid is named Cardicola opisthorchis n. sp. and the generic diagnosis is emended in this paper. In addition, 100% identity among the ITS2 sequences from the present species, Cardicola sp. from T. orientalis in Mexico and Cardicola sp. from the northern bluefin tuna, Thunnus thynnus (Linnaeus) in Spain suggests that C. opisthorchis n. sp. has a broad geographical distribution and that it infects both the Pacific and northern bluefin tuna.  相似文献

The temperature-sensitiveprp24-1 mutation defines a gene product required for the first step in pre-mRNA splicing. PRP24 is probably a component of the U6 snRNP particle. We have applied genetic reversion analysis to identify proteins that interact with PRP24. Spontaneous revertants of the temperaturesensitive (ts)prp24-1 phenotype were analyzed for those that are due to extragenic suppression. We then extended our analysis to screen for suppressors that confer a distinct conditional phenotype. We have identified a temperature-sensitive extragenic suppressor, which was shown by genetic complementation analysis to be allelic toprp21-1. This suppressor,prp21-2, accumulates pre-mRNA at the non-permissive temperature, a phenotype similar to that ofprp21-1. prp21-2 completely suppresses the splicing defect and restores in vivo levels of the U6 snRNA in theprp24-1 strain. Genetic analysis of the suppressor showed thatprp21-2 is not a bypass suppressor ofprp24-1. The suppression ofprp24-1 byprp21-2 is gene specific and also allele specific with respect to both the loci. Genetic interactions with other components of the pre-spliceosome have also been studied. Our results indicate an interaction between PRP21, a component of the U2 snRNP, and PRP24, a component of the U6 snRNP. These results substantiate other data showing U2–U6 snRNA interactions.  相似文献

Twenty-two isolates ofVerticillium dahliae, which were isolated from green soybeean (Glycine max), udo (Aralia cordata), horseradish (Cochlearia armoracia), sweetpea (Lathyrus odoratus), or a weed (Chenopodium album) were used in this study. Conidia and microsclerotia of these isolates were morphologically identical with those ofV. dahliae but did not coincide withV. longisporum. Pathogenicity tests showed that these isolates were of weak pathotype. Eleven of the 22 isolates, which were obtained from green soybean and udo, were pathogenic to green soybeans. Thus pathotype E was composed of two groups: ‘soybean pathotype’ which was pathogenic to green soybeans; and isolates nonpathogenic to green soybeans. The latter were defined as isolates of pathotype E in the narrow sense. Selected representativenit1 and NitM mutants of eachV. dahliae isolate were paired with VCGJ testers. Fourteen isolates ofV. dahliae (So1, So22, So23, So27, So28, So39, So40, So41, U54, U68, U69, U90, U95, and U115) showed complementary reactions with subgroups J1 and J3 and were assigned to subgroup J3. Isolate U108 was assigned to subgroup J2. Isolate HR1 was not compatible with any testers of VCGJ. With this exception, isolates of pathotype E in the narrow sense and those of ‘soybean pathotype’ were thus assigned to known VCGJ subgroups and did not form a unique group corresponding to their pathotype. ‘Soybean pathotype’ could not be distinguished among isolates of pathotype E by vegetative compatibility.  相似文献

Rice fields are important substitute wetlands for frogs. Traditionally, rice fields in Japan were supplied with water and drained via shallow earth ditches (old-style). In the last 30 years, however, more than 80% of rice fields have been converted to a new irrigation system in which water is typically supplied through underground pipes via taps and is drained into deep, concrete-sided ditches (new-style). We compared the occurrence of frogs in paired areas of old- and new-style rice fields at six locations in Ibaraki Pref., central Japan, from May to August 1995. The Japanese tree frog (Hyla japonica) did not differ in abundance between the two types of rice fields, but the Japanese brown frog (Rana japonica) and the Tokyo daruma pond frog (Rana porosa porosa) preferred the old-style rice fields. These findings suggest that the status ofRana species has been adversely affected by the conversion of rice fields to the new irrigation system. Modification of the deep, U-shaped concrete ditches and the water management regime during the spawning season is needed to safeguard these species.  相似文献
