Collagen and elastin synthesis in the developing chick aorta   总被引:3,自引:0,他引:3  
Thoracic aortas from 8- to 18-day embryonic chicks were incubated in vitro for 30 min with [3H]glycine and the newly synthesized, labeled proteins were subjected to polyacrylamide gel electrophoresis in sodium dodecyl sulfate. The gels were fractionated and the incorporation of label into procollagen (125,000 Mr) and tropoelastin (70,000 Mr) was estimated by summation of the radioactivity found in the appropriate regions of the gel. The analyses showed that at Day 8 approximately 14% of the incorporated [3H]glycine was found in procollagen and 22% in tropoelastin. In the following 6 days of development, there was a significant decline in the relative incorporation into procollagen and an increase into tropoelastin so that at Days 14–18 less than 10% of the label was found in collagen and 40% was now found in tropoelastin. Since glucocorticoids have been shown to alter the rate of synthesis of other proteins in the developing chick, 150 μg of hydrocortisone was injected into 8-day eggs and 24 h later the aortas were incubated and treated as described above. The pattern of protein synthesis exhibited by the hormone-treated aortas resembled that of 14- to 18-day embryos. Furthermore, incubation of 8-day aortas with 10?8m hydrocortisone for 24 h produced a significant increase in the rate of elastin synthesis relative to that of other proteins. These results demonstrate that collagen and elastin synthesis vary during development of the chick aorta and they suggest that glucocorticoids may be involved in the control of their synthesis.  相似文献

Hydroxynorvaline (DL-alpha-amino-beta-hydroxyvaleric acid) was shown to competitively inhibit the activation of threonine and valine when tested with tRNA and synthetases prepared from whole chick embryos. However, the hydroxynorvaline was transferred only to threonyl-tRNA and not valyl-tRNA. The hydroxynorvaline had no effect when tested with other amino acids. The Km for threonine was 25 muM and the Ki for hydroxynorvaline was 181 muM. When fibroblasts from embryonic chick tendons were incubated with [3H]threonine and increasing concentrations of hydroxynorvaline, there was a progressive decrease in the incorporation of [3H]threonine so that 1 mM hydroxynorvaline the incorporation into nondialyzable protein was 26% of the control value. A much smaller decrease in the incorporation of other radioactive amino acids was observed. When the cells were incubated hith [14C]proline and 1 mM hydroxynorvaline, the labeled procollagen containing hydroxynorvaline accumulated intracellularly and very little was secreted. Control experiments demonstrated that free hydroxynorvaline did not inhibit the secretion of unsubstituted procollagen. Although the individual pro alpha chains containing hydroxynorvaline were of normal molecular weight (125,000) and hydroxyproline content, only about 50% of this intracellularly retained procollagen was triple helical within the cell as 37 degrees as measured by sensitivity to pepsin digestion. Also only approximately 50% of the pro alpha chains were disulfide-linked to form triple stranded molecules as compared to greater than 85% linkage in unsubstituted procollagen. We postulate that incorporation of hydroxynorvaline alters the conformation of the propeptide extension sufficiently so that: (a) normal assembly of disulfide-linked, triple helical molecules is reduced and (b) assembled triple helical molecules are not properly recognized by the secretory mechanism.  相似文献

Abstract— A 100,000 g supernatant fraction from rat brain that was passed through a column of Sephadex G-25-40 was able, after addition of some factors, to incorporate [I4C]arginine (apparent Km= 5 μM) and [14C]tyrosine (apparent Km= 20 μM) into its own proteins. The factors required for the incorporation of [14C]arginine were: ATP (optimal concentration = 0-25-2 μM) and Mg2+ (optimal concentration 5 mM). For the incorporation of [I4C]tyrosine the required factors were: ATP (apparent Km= 0-75 μM), Mg2+ (optimalconcentration 8-16 mM) and K+ (apparent Km= 16 mM). Addition of 19 amino acids did not enhance these incorporations. Optimal pHs were: for [14C]arginine and [14C]tyrosine, respectively, 7-4 and 7-0 in phosphate buffer and 7–9 and 7-3-8-1 in tris-HCl buffer. Pancreatic ribonuclease abolished the incorporation of [14C]arginine but had practically no effect in the incorporation of [14C]tyrosine. Furthermore, [14C]arginyl-tRNA was a more effective donor of arginyl groups than [14C]arginine, whereas [14C]tyrosyl-tRNA was considerably less effective than [14C]tyrosine. The incorporations of [14C]arginine and [14C]tyrosine into brain proteins were from 25- to 2000-fold higher than for any other amino acid tested (12 in total). In brain [14C]arginine incorporation was higher than in liver and thyroid but somewhat lower than in kidney. In comparison to brain, the incorporation of [14C]tyrosine was negligible in liver, thyroid or kidney. Kinetic studies showed that the macromolecular factor in the brain preparation was complex. The protein nature of the products was inferred from their insolubilities in hot TCA and from the action of pronase that rendered them soluble. [14C]Arginine was bound so that its a-amino group remained free. Maximal incorporation of [14C]tyrosine in brain of 30-day-old rats was about one-third of that in the 5-day-old rat. The changes with postnatal age in the incorporation of [14C]arginine were not statistically significant.  相似文献

Rat brain membranes were incubated with bee venom phospholipase A2 (PLA2) or phospholipase C (PLC) from Clostridium perfringens. PLA2 caused a significant increase in free polyunsaturated fatty acids concomitant with membrane phospholipid degradation as monitored by HPLC and by gas chromatography. Equal concentrations of PLC had a much lesser effect than PLA2. Divergent and differential effects were shown on deacylation and incorporation of [3H]arachidonic acid in membrane phospholipids. The incorporation of [3H]arachidonic acid into various phospholipids was greatly reduced by PLA2 (0.018 units/ml) whereas PLC at identical concentration was not effective. PLA2 inhibited (Na+ + K+)-ATPase but was not effective on p-nitrophenyl-phosphatase activity whereas PLC stimulated both enzymes. PLA2 induced swelling of cortical brain slices whereas PLC was not effective. Thus, the severity of the perturbation of membrane integrity, and the inhibition of (Na+ + K+)-ATPase in brain membranes may play an important role in cellular swelling of brain slices induced by PLA2.  相似文献

The effects of glucose (10 mm), glycerol (3 mm), and lactate/pyruvate (10 mm) on the incorporation of 3H from 3H2O into fatty acids were studied in isolated hepatocytes prepared from chow-fed female rats. Lactate/pyruvate markedly increased lipogenic rates, while glucose and glycerol did not significantly affect rates of lipogenesis. In cells incubated with lactate/pyruvate plus glycerol, the increase in 3H incorporation was greater than observed with lactate/pyruvate alone. In hepatocytes isolated from 24-h starved rats, lactate/pyruvate again increased de novo fatty acid synthesis to a greater extent than either glucose or glycerol. Glycerol significantly increased lipogenesis compared to the endogenous rates and when incubated with lactate/pyruvate produced an increase above lactate/pyruvate alone. (?)-Hydroxycitrate, a potent inhibitor of ATP-citrate lyase (EC 4.1.3.8), and agaric acid, an inhibitor of tricarboxylate anion translocation, were studied in hepatocytes to determine their effects on lipogenesis by measuring 3H2O, [1-14C]acetate, and [2-14C]lactate incorporation into fatty acids. 3H incorporation into fatty acids was markedly inhibited by both inhibitors with agaric acid (60 μm) producing the greater inhibition. (?)-Hydroxycitrate (2 mm) increased acetate incorporation into fatty acids from [1-14C]acetate and agaric acid produced a strong inhibitory effect. Combined effects of (?)-hydroxycitrate and agaric acid on lipogenesis from [1-14C]acetate showed an inhibitory response to a lesser extent than with agaric acid alone. With substrate concentrations of acetate present, there was no significant increase in rates of lipogenesis from [1-14C]acetate and the increase previously observed with (?)-hydroxycitrate alone was minimized. Agaric acid significantly inhibited fatty acid synthesis from acetate in the presence of exogenous substrate, but the effect was decreased in comparison to rates with only endogenous substrate present. With [2-14C]lactate as the lipogenic precursor, agaric acid and (?)-hydroxycitrate strongly inhibited fatty acid synthesis. However, agaric acid despite its lower concentration (60 μm vs 2 mm) was twice as effective as (?)-hydroxycitrate. A similar pattern was observed when substrate concentrations of lactate/pyruvate (10 mm) were added to the incubations. When (?)-hydroxycitrate and agaric acid were simultaneously incubated in the presence of endogenous substrate, there was an additive effect of the inhibitors on decreasing fatty acid synthesis. Results are discussed in relation to the origin of substrate for hepatic lipogenesis and whether specific metabolites increase lipogenic rates.  相似文献

The kinetics of the incorporation into protein of [3H]phenylalanine, [3H]tyrosine and [3H]tryptophan were studied with homogenates prepared from whole brain of 1-, 7-, 21- and 60-day-old rats. The maximal velocities (Vmax)of incorporation of phenylalanine and tyrosine decreased and the apparent Michaelis-constants (Km) for all three amino acids increased with increasing age of the rats. Tyrosine had the smallest and tryptophan the largest Km values in all age groups. Phenylalanine competitively inhibited the incorporation of tyrosine, but tyrosine inhibited non-competitively the incorporation of phenylalanine. Tryptophan inhibited competitively the incorporation of phenylalanine, but at least partially non-competitively the incorporation of tyrosine. Phenylalanine and tyrosine did not significantly affect the incorporation of tryptophan in homogenates from 60-day-old rats. In 1-day-old rats only a very large excess of phenylalanine or tyrosine inhibited detectably. The Ki for phenylalanine in the incorporation of tyrosine was significantly smaller in 1- than in 60-day-old rats. In every case the inhibition presumably occurred at a single rate-limiting step in the complicated process of incorporation of amino acids into protein.  相似文献

The incorporation of [3H]phenylalanine, [3H]tyrosine, and [3H]tryptophan into protein and amino acyl–tRNA was studied in cell-free preparations from rat brain. Tyrosine and tryptophan inhibited the incorporation of phenylalanine into protein, and tyrosine inhibited the incorporation of phenylalanine and tryptophan into amino acyl–tRNAs. In most cases, homogentisate, phenylpyruvate, and phenyllactate inhibited the incorporation of phenylalanine, tyrosine, and tryptophan into protein and amino acyl–tRNAs, and the incorporation of phenylalanine into polyphenylalanine. All other protein amino acids, and phenylacetate, salicylate, and benzoate were wholly ineffectual. The results suggest that the formation of amino acyl–tRNAs may have been the step which was affected most by the inhibitors. The incorporation data at different concentrations of the aromatic amino acids were fitted to the simple Michaelis equation. Homogentisate and phenylpyruvate generally tended to reduce both Km and V in the incorporation of aromatic amino acids into protein and amino acyl-tRNAs, even if V decreased more than Km.  相似文献

Skeletal myotubes responded to passive stretch by increased amino acid uptake (as measured with [3H]α-aminoisobutyric acid), increased incorporation of amino acids into total cellular protein and myosin heavy chains, and increased accumulation of total cellular protein and myosin heavy chains. These alterations were preceded by an increase in the uptake of ouabain-sensitive rubidium-86 (86Rb+), a potassium tracer used to measure membrane sodium pump activity (Na+K+ATPase). This stretch-induced stimulation of 86Rb+ uptake resulted from a 60-70% increase in the Vmax of the Na pump with little change in the Km. [3H] ouabain binding studies showed no stretch-induced change in the number of membrane Na pumps, indicating that stretch activates the Na pumps that are already present on the cell surface. Since the stretch-induced increases in amino acid transport and amino acid incorporation into proteins were inhibited by ouabain, Na pump activation may be involved in stretch-induced cell growth of skeletal muscle cells by hypertrophy.  相似文献

Cells were isolated from the aortae of 17-day old chick embryos by digestion of the vessels with a combination of trypsin and collagenase. When these cells were incubated in suspension culture in Krebs-Ringer media containing pancreatic trypsin inhibitor and radioactive amino acids, they synthesized and secreted labeled proteins into the media. Polyacrylamide gel electrophoresis in sodium dodecyl sulfate of the secreted proteins labeled with [14C]proline revealed two major components. The larger component with an approximate molecular weight of 125,000 had a [14C]hydroxyproline content consistent with a form of procollagen. The molecular weight of 70,000 and [14C]hydroxyproline content of the second component was consistent with that previously reported for tropoelastin extracted from chick aortae. By following the kinetics and secretion of tropoelastin labeled with [3H]valine, we have estimated that 17 minutes are required to synthesize and secrete the molecule under these experimental conditions.  相似文献

Brain slices obtained from neocortex, hypothalamus or hippocampus were incubated with [3H]histamine and subsequently superfused and exposed to different depolarizing stimuli, viz. high K+-concentrations, electrical field stimulation and veratrine. K+-induced release of tritium was completely calcium-dependent and its magnitude depended on the K+-concentration, with maximal release being reached at 56 mM K+. Electrically-evoked release of tritium increased with increasing frequencies and reached its maximum at about 20 Hz. The electrically-evoked release appeared to be totally calcium-dependent and it was strongly inhibited by tetrodotoxin. Veratrine (5–100 μM) also induced a release of tritium; maximal release was obtained at 100 μM veratrine. Veratrine-induced release was partially calcium-dependent and was strongly reduced by tetrodotoxin.Taken together the data indicate that the depolarization-induced release of tritium from brain slices pre-labelled with [3H]histamine, represents [3H]histamine release from neurons and not from either mast cells or glial cells. It remains to be established whether these neurons are specifically histaminergic.  相似文献

Endosperms of quiescent barley grains contained, on average, 54.5 μg of neutral glyceride-glycerol, equivalent to ca 480 μg glyceride. Of this probably 90% was located in the aleurone layer. During germination the level of glyceride-glycerol declined. It also declined in degermed grains and aleurone layers incubated in vitro. The fall was accelerated by GA3, but indoleacetic acid, kinetin and glutamine were without effect. Increases in the levels of malate synthase and isocitrate lyase from very low initial values, and the results of incorporation studies with [14C]-labelled substrates, indicate that the glyoxylate cycle functions to convert glycerides to sucrose in germinating grains and degermed grains incubated with GA3, but not in degermed grains without the hormone. In the absence of GA3 the glyceride could be a respiratory substrate in degermed grains. The aleurone layers converted exogenous glucose to sucrose. Little label from [14C]-amino acids appeared in sucrose but in some cases considerable incorporation occurred into glutamine.  相似文献

Slices were prepared from rat forebrains and the incorporation of [3H]mannose and [35S]methionine into proteins and glycoproteins determined. The incorporation of methionine continued to increase for up to 8 hours whereas mannose incorporation was maximal between 2 and 4 hours and declined thereafter. Glycopeptides prepared by pronase digestion of [3H]mannose-labeled glycoproteins were digested with endoglucosaminidase H (endo H) and analysed by gel filtration. The major endo H-sensitive oligosaccharide eluted in a position similar to standard Man8GlcNAc. In the presence of castanospermine, which inhibits glucosidase I, the first enzymatic step in the processing of N-linked oligosaccharides, a new endo H-sensitive glycan similar in size to standard Glc3Man9GlcNAc2 accumulated. Synaptic membranes (SMs) were isolated from slices which had been incubated with either [3H]mannose or [35S]methionine in the presence and absence of castanospermine. In the presence of inhibitor the relative incorporation of [3H]mannose into high-mannose glycans of synaptic glycoproteins was increased. The incorporation of newly synthesized, [35S] methioninelabeled, Con A-binding glycoproteins into SMs was not affected by the addition of inhibitor. Many of the glycoproteins synthesized in the presence of castanospermine exhibited a decreased electrophoretic mobility indicative of the presence of altered oligosaccharide chains. The results indicate that changes in oligosaccharide composition produced by castanospermine had little effect on the subsequent transport and incorporation of glycoproteins into synaptic membranes.To whom to address reprint requests.  相似文献

—Isolated retinas from chick embryos and mature animals were incubated in [3H]GABA at 25°C for 10 min in order to investigate kinetic properties of the amino acid uptake system. Embryo retina accumulated [3H]GABA by two distinct kinetic systems with Km values of the order 10−4m and 10−5m for the low- and high-affinity mechanisms respectively. However, as the retina matured, the high-affinity process disappeared and only the low-affinity system was detectable. No obvious explanation can be offered for this phenomenon although a similar observation has previously been made in chick brain by other workers.  相似文献

Samples of (3R)- and (3S)-4′hydroxyphenyl[3-2H1, 3-3H]pyruvate were prepared by taking advantage of the known stereospecificity of phenylpyruvate keto-enol isomerase (tautomerase). 4′-Hydroxyphenyl[3-14C]pyruvate was obtained by the action of l-amino acid oxidase on dl-[3-14C]tyrosine, whereas a simple base-catalyzed exchange procedure yielded samples of 4′-hydroxyphenyl[3-3H]- and 4′-hydroxyphenyl[3-2H2]pyruvate. All labeled samples were converted in situ into the corresponding homogentisic acids on 4′-hydroxyphenyl-pyruvate dioxygenase that is known to catalyze the migration of the acetate side chain with retention of configuration. The isolated doubly labeled homogentisic acids were incubated with chloroplasts from Raphanus sativus cv. saxa Treib, and from the lipophilic products a fraction containing inter alia tocopherol, tocoquinone, and plastoquinone was obtained by chromatographic procedures. The incorporation of radioactivity was between 0.5 and 11% based on homogentisate. Reductive acetylation of the quinones yielded crystalline diacetylhydroquinones, which were submitted to Kuhn-Roth degradation. The radioactive acetate samples thus obtained were analyzed for chirality by an enzymatic procedure previously published. (2R)-[2-2H1, 2-3H]Homogentisate gave mainly (S)-acetate, whereas (2S)-[2-2H1, 2-3H]homogentisate was converted mainly into (R)-acetate. It is concluded that the decarboxylation of the side chain occurred with stereochemical retention during the biosynthetic process.  相似文献

Human placental microsomes exhibit uptake of d-[3H]glucose which is sensitive to inhibition by cytochalasin B (apparent Ki = 0.78 /gm M). Characterization of [3H]cytochalasin B binding to these membranes reveals a glucose-sensitive site, inhibited by d-glucose with an ED50 = 40 mM. The glucose-sensitive cytochalasin B binding site is found to have a Kd = 0.15μM by analysis according to Scatchard. Solubilization with octylglucoside extracts 60–70% of the glucose-sensitive binding component. Equilibrium dialysis binding of [3H]cytochalasin B to the soluble protein displays a pattern of inhibition by d-glucose similar to that observed for intact membranes, and the measurement of an ED50 = 37.5 mM d-glucose confirms the presence of the cytochalasin B binding component, putatively assigned as the glucose transporter. Further evidence is attained by photoaffinity labelling; ultraviolet-sensitive [3H]cytochalasin B incorporation into soluble protein (Mr range 42 000-68 000) is prevented by the presence of d-glucose. An identical photolabelling pattern is observed for incorporation of [3H]cytochalasin B into intact membrane protein, confirming the usefulness of this approach as a means of identifying the presence of the glucose transport protein under several conditions.  相似文献

Incorporation of [3H]glucose into macromolecular components of 12-day chick embryo sternum incubated in vitro was stimulated by both human serum and l-3,5,3′-triiodothyronine. Under all conditions, 65–70% of the radioactivity was incorporated into glycosaminoglycans. About 10% of the radioactivity was incorporated into a fraction separable by ion-exchange chromatography which was stimulated two- to sixfold by addition of 2–10 nm triiodothyronine and 5–20% (vv) human serum. Further characterization of this fraction by paper electrophoresis at pH 3.5 showed the presence of two components, one apparently anionic and one neutral. All of the increase in incorporation of [3H]glucose was into the former species. Acid hydrolysis of this material showed that it contained only glucose. Treatment with α-amylase released 78% of the label as maltotriose and maltose; digestion with crystalline β-amylase released 75% as maltose; and treatment with glucoamylase and α-amylase released 93% as glucose. There was no incorporation of any amino acid into this fraction, nor could any incorporation of [32P]phosphate, [35S]sulfate, [3H]uridine, or [3H]acetate be demonstrated. Mild acid hydrolysis (0.1 N HC1, 100 °C, 10–20 min) converted the material to a neutral species with a much lower molecular weight. The results indicate that chick embryo sternum contains a species of glycogen whose synthesis is stimulated by thyroid hormones and other serum factors.  相似文献

The metabolism of [3H]formate has been examined in etiolated and greening leaves of barley (Hordeum vulgare), dwarf bean (Phaseolus vulgarls), broad bean (Vicia faba) and corn (Zea mays). Tritium was extensively incorporated by primary leaves incubated for 20-min periods in light or dark. The organic acids and free amino acids were the principal products of formate metabolism but these and other products were more heavily labelled in green tissues. Time course experiments with barley leaves revealed a rapid labelling of serine, accompanied by increasing amounts of 3H in glycine and aspartate as the feeding period was extended. These amino acid products were formed throughout a 4-day greening period with an approximate doubling in total incorporation being due to large accumulations of tritiated glycine and aspartate. The involvement of tetrahydrofolate-dependent reactions in formate metabolism was indicated by inhibition of [14C] and [3H]formate incorporation by the folate antagonist, aminopterin. Labelling of glycine and serine was also strongly inhibited (up to 90%) when the leaves were incubated with increasing concentrations of isonicotinylhydrazide.  相似文献

Polysomes were isolated from aortas of 17-day-old chick embryos, and the synthesis of the nascent polypeptide chains was completed in vitro. When a mixture of a labeled amino acids found in elastin was used, the major radioactive product obtained was of molecular weight about 70,000 and was similar to elastin by several criteria. The 70,000 molecular weight product was extractable in propanol-butanol, it was not labeled with [35S]methionine, and it was precipitated by antibodies against elastin. Polypeptides larger than 70,000 molecular weight were also synthesized but these larger polypeptides incorporated relatively small amounts of [14C]valine, and they appeared to represent proα chains of procollagen. The results suggest that the major gene product for elastin has a molecular weight of about 70,000.  相似文献

—The accumulation of [3H]choline into synaptosome-enriched homogenates of rat corpus striatum, cerebral cortex and cerebellum was studied at [3H]choline concentrations varying from 0.5 to 100 μm . The accumulation of [3H]choline in these brain regions was saturable. Kinetic analysis of the accumulation of the radiolabel was performed by double-reciprocal plots and by least squares iterative fitting of a substrate-velocity curve to the data. With both of these techniques, the data were best satisfied by two transport components, a high affinity uptake system with Km. values of 1.4 μM (corpus striatum), and 3.1 μM (ceμ(cerebral cortex) and a low affinity uptake system with respective Km. values of 93 and 33 μM for these two brain regions. In the cerebellum choline was accumulated only by the low affinity system. When striatal homogenates were fractionated further into synaptosomes and mitochondria and incubated with varying concentrations of [3H]choline, the high affinity component of choline uptake was localized to the synaptosomal fraction. The high affinity uptake system required sodium, was sensitive to various metabolic inhibitors and was associated with considerable formation of [3H]acetylcholine. The low affinity uptake system was much less dependent on sodium, and was not associated with a marked degree of [3H]acetylcholine formation. Hemicholinium-3 and acetylcholine were potent inhibitors of the high affinity uptake system. A variety of evidence suggests that the high affinity transport represents a selective accumulation of choline by cholinergic neurons, while the low affinity uptake system has some less specific function.  相似文献

The tritium-labeled selective agonist of the nonopioid β-endorphin receptor the decapeptide immunorphin ([3H]SLTCLVKGFY) with a specific activity of 24 Ci/mmol was prepared. It was shown that [3H]immunorphin binds with a high affinity to the non-opioid β-endorphin receptor of mouse peritoneal macrophages (K d 2.4 ± 0.1 nM). The specific binding of [3H]immunorphin to macrophages was inhibited by unlabeled β-endorphin (K i of the [3H]immunorphin-receptor complex 2.9 ± 0.2 nM) and was not inhibited by unlabeled naloxone, α-endorphin, γ-endorphin, and [Met5]enkephalin (K i > 10 μM). Thirty fragments of β-endorphin were synthesized, and their ability to inhibit the specific binding of [3H]immunorphin to macrophages was studied. It was found that the shortest peptide having practically the same inhibitory activity as β-endorphin is its fragment 12–19 (K i 3.1 ± 0.3 nM).  相似文献
