长白山白眉蝮蛇蛇毒磷脂酶A2的分离和初步表征   总被引:10,自引:0,他引:10  
东北长白山白眉蝮蛇(AgkistrodonblomhoffiUsurensis)蛇毒经DEAESephadexA50离子交换层析柱,连续3步SephadexG75凝胶过滤柱得到了磷脂酶A2(PLA2)的纯品。SDS聚丙烯酰胺凝胶电泳(SDSPAGE)以及基质辅助激光解析电离飞行时质谱(MALDI/TOF/MS)表征为单一蛋白,其准确分子量为(14.008±0.007)kD。最适pH范围8.0~9.0,最适的反应温度为45℃。在溶液中有多聚体的存在。  相似文献

NNK诱发BEP2D细胞产生活性氧及其对DNA的损伤   总被引:4,自引:0,他引:4  
通过测定细胞内和细胞上清中活性氧(reactive oxygen species,ROS)水平,以及DNA 加合物——8-羟基脱氧鸟嘌呤核苷(8-hydroxydeoxyguanosine,OH8dG)含量,对烟草特异亚硝胺类化合物4-甲基亚硝胺-1(3-吡啶基)-1-丁酮(4-(m ethylnitrosam ino)-1-(3-pyridyl)-1-butanone,NNK)诱发人乳头状病毒永生化的人支气管上皮细胞(hum an papillom avirus-im m ortalized hum anbronchialepithelialcellline,BEP2D)产生的ROS及其对DNA 的氧化损伤进行研究,并观察纳米硒的保护作用.结果表明,BEP2D 细胞经不同浓度的NNK 作用后,细胞内和细胞上清中ROS以及OH8dG含量均显著增加,并有较好的剂量效应关系.1 μm ol·L- 1纳米硒(nanoselenuim ,NS)能明显抑制NNK 诱发BEP2D细胞产生的ROS及OH8dG 水平.揭示NNK 能造成细胞的氧化损伤,而NS对NNK 所致细胞的氧化损伤有保护作用.  相似文献

三唑酮处理可提高离体绿豆(PhaseolusradiatusL.)幼苗叶片叶绿素和蛋白质含量。叶片衰老过程中超氧物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(AsAPOD)活性及抗坏血酸(AsA)和还原型谷胱甘肽(GSH)含量降低。20mg/L三唑酮可提高POD、AsAPOD活性和AsA、GSH含量,对SOD、CAT活性无影响。丙二醛(MDA)含量在叶片衰老过程中提高,并与POD、AsAPOD活性和AsA、GSH含量呈显著负相关,三唑酮可降低MDA含量。表明三唑酮有提高植物对膜脂过氧化作用的保护能力,延缓叶片的衰老作用。  相似文献

盐生杜氏藻甘油-3-磷酸脱氢酶的分离纯化及其特性的研究   总被引:1,自引:0,他引:1  
利用PEG分级,DEAE离子交换层析,BlueSepharose拟亲和层析,MonoQ离子交换层析等手段,分离纯化盐生杜氏藻(Dunalielasalina(Dunal)Teod.)甘油三磷酸(G3P)脱氢酶(EC1.1.1.8),得到比活为12.6U/mg的电泳纯的酶,并对此酶的生化特性进行了研究。4%~20%非变性聚丙烯酰胺梯度凝胶电泳测得全酶分子量约为270kD,SDSPAGE表明该酶只有一种分子量约为65kD的亚基,据此推测该酶应为同四聚体。酶催化磷酸二羟丙酮(DHAP)还原的最适pH值为7.5,催化G3P脱氢的最适pH值为10。该酶对4个底物还原型辅酶Ⅰ(NADH),二磷酸吡啶核苷酸(DHAP),辅酶Ⅰ(NAD),G3P的表观Km值分别为63μmol/L,272μmol/L,1.53mmol/L,6.52mmol/L。该酶在保存过程中易失活。NADH能降低酶失活的速度,而NAD则不然。低浓度NaCl对酶略有保护作用,但高浓度NaCl加快酶的失活,且浓度越高效应越明显。  相似文献

体外培养大鼠星形细胞的缺糖缺氧性损伤及药物的保护   总被引:4,自引:1,他引:3  
本实验用大鼠星形细胞体外培养模型,观察了细胞在不同条件下乳酸脱氨酶(LDH)的漏出。发现当去掉葡萄糖后,细胞缺氧5和7h后LDH漏出明显增加,5b为59.7±25.3U/mg蛋白(对照组24.7±16.3,P<0.05),7h为68.3±89U/mg蛋白(对照组39.9±212,P<0.01)。用3-氧-甲基-[1- ̄3H]-D-葡萄糖摄取法测量细胞体积,结果显示缺糖缺氧5h后细胞明显肿胀,从对照组的4.1±1.2增加到8.1±3.2μl/mg蛋白(P<0.01)。丹参有效成分764-3在0.5至50μmol/L时能减少缺糖缺氧细胞LDH漏出;在50μmol/L时能减小缺糖缺氧细胞的体积。谷氨酸受体拮抗剂DNQX(6,7-dinitroquinoxaline-2,3dione)50μmol/L能减轻星形细胞肿胀和LDH漏出。  相似文献

离体培养风信子(HyacinthusorientalisL.)不同年龄的花被外植体诱导花器官直接再生的实验表明:1.在MS附加6BA2mg/L、2,4D0.1mg/L的培养基上,年龄V的外植体大量衰老,基本丧失器官再生能力,处于年龄段II~IV的外植体可发生玻璃化反应。2.玻璃化反应的外植体转移至MS附加6BA0.2mg/L、NAA0.005mg/L的培养基上继续培养30d后可再生正常的花被片,表明降低培养基中外源激素浓度能够阻止玻璃化反应继续发生。3.在MS附加6BA2mg/L、2,4D0.1mg/L的培养基上,外植体形态学下部可再生雌蕊状早期结构。平均每块外植体分化雌蕊状早期结构数以年龄Ⅲ的外植体最多  相似文献

利用谷胱甘肽S-转移酶(Glutathione S-transferase,GST)融合基因表达系统,大鼠20α羟类固醇脱氢酶(20αHydroxysteroidDehydrogenase,20αHSD)在大肠杆菌中得以成功地表达。亲和层析和Thrombin消化,可从融合蛋白中回收和纯化重组20αHSDSDS-PAGE、Western印迹法和酶活性测定显示,重组20αHSD具有天然蛋白质相同分子量、相似的抗原性和酶催化活性,其对NADP的K_m值和V_(max)分别为9.5μmol/L、334nmo1/(min·mg),对底物20α羟孕酮(20αHydroxyprogesterone,20αOHP)的K_m值和V_(max)分别为5.9μmol/L和347nmol/(min·mg),利用该表达系统大量制备大鼠重组20αHSD,为深入研究20αHSD的生理活性和功能创造条件。  相似文献

用正丁醇抽提,硫酸铵分级沉淀,DEAE-纤维素和SephacrylS-200柱层析,从南方鲇(Silurus meridionalis Chen)肠粘膜中提取出碱性磷酸酶(AKP)。提纯倍数为39.50倍,比活为68.35μ/mg蛋白,提取酶液经PAGE和SDS-PAGE只呈现一条区带。该酶的分子量为132140,N末端氨基酸为门冬氨酸,最适pH为10.10,7.5>pH>11.5时不稳定,最适温度为40℃左右,对热不很稳定,以磷酸苯二钠为底物其K_m值为1.72×10~(-3)mol/L。Mg~(2+)、Mn~(2+)为该酶的激活剂,KH_2PO_4、L-CyS、ME、DFP、EDTA-Na_2为抑制剂。选用KH_2PO_4和DFP作抑制类型的判断,结果表明,KH_2PO_4属竞争性掏剂,其抑制常数为2.3mmol/L;DFP为非竞争性抑制剂,抑制常数为1.05mmol/L。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
