Survival of fecal microorganisms in marine and freshwater sediments.   总被引:19,自引:9,他引:10       下载免费PDF全文
The survival of culturable fecal coliforms, fecal streptococci, and Clostridium perfringens spores in freshwater and marine sediments from sites near sewage outfalls was studied. In laboratory studies, the inhibition of protozoan predators with cycloheximide allowed the fecal coliforms to grow in the sediment whereas the presence of predators resulted in a net die-off. C. perfringens spores did not appear either to be affected by predators or to die off throughout the duration of the experiments (28 days). Studies using in situ membrane diffusion chambers showed that, with the exception of C. perfringens, die-off of the test organisms to 10% of their initial numbers occurred in both marine and freshwater sediments within 85 days. The usual exponential decay model could not be applied to the sediment survival data, with the exception of the data for fecal streptococci. It was concluded that application of the usual decay model to the fecal coliform data was confounded by the complex relationship between growth and predation. The survival of seeded Escherichia coli in marine sediment was studied by using an enumeration method which detected viable but nonculturable bacteria. Throughout the duration of the experiment (68 days), the same proportion of E. coli organisms remained culturable, suggesting that sediment provides a favorable, nonstarvation environment for the bacteria.  相似文献

Fecal bacteria can contaminate water and result in illness or death. It is often difficult to accurately determine sources of fecal bacteria contamination, but bacteria source tracking can help identify non-point sources of fecal bacteria such as livestock, humans and wildlife. The Soil and Water Assessment Tool (SWAT) microbial sub-model 2005 was used to evaluate source-specific fecal bacteria using three years (2004-2006) of observed modified deterministic probability of bacteria source tracking data, as well as measure hydrologic and water quality data. This study modeled source-specific bacteria using a model previously calibrated for flow, sediment and total fecal coliform bacteria (FCB) concentration. The SWAT model was calibrated at the Rock Creek sub-watershed, validated at the Deer Creek sub-watershed, and verified at the Auburn sub-watershed and then at the entire Upper Wakarusa watershed for predicting daily flow, sediment, nutrients, total fecal bacteria, and source-specific fecal bacteria. Watershed characteristics for livestock, humans, and wildlife fecal bacterial sources were first modeled together then with three separate sources and combinations of source-specific FCB concentration: livestock and human, livestock and wildlife and human and wildlife. Model results indicated both coefficient of determination (R(2)) and Nash-Sutcliffe Efficiency Index (E) parameters ranging from 0.52 to 0.84 for daily flow and 0.50-0.87 for sediment (good to very good agreement); 0.14-0.85 for total phosphorus (poor to very good agreement); -3.55 to 0.79 for total nitrogen (unsatisfactory to very good agreement) and -2.2 to 0.52 for total fecal bacteria (unsatisfactory to good agreement). Model results generally determined decreased agreement for each single source of bacteria (R(2) and E range from -5.03 to 0.39), potentially due to bacteria source tracking (BST) uncertainty and spatial variability. This study contributes to new knowledge in bacteria modeling and will help further understanding of uncertainty that exists in source-specific bacteria modeling.  相似文献

Fecal coliforms and enterococci are indicator organisms used worldwide to monitor water quality. These bacteria are used in microbial source tracking (MST) studies, which attempt to assess the contribution of various host species to fecal pollution in water. Ideally, all strains of a given indicator organism (IO) would experience equal persistence (maintenance of culturable populations) in water; however, some strains may have comparatively extended persistence outside the host, while others may persist very poorly in environmental waters. Assessment of the relative contribution of host species to fecal pollution would be confounded by differential persistence of strains. Here, freshwater and saltwater mesocosms, including sediments, were inoculated with dog feces, sewage, or contaminated soil and were incubated under conditions that included natural stressors such as microbial predators, radiation, and temperature fluctuations. Persistence of IOs was measured by decay rates (change in culturable counts over time). Decay rates were influenced by IO, inoculum, water type, sediment versus water column location, and Escherichia coli strain. Fecal coliform decay rates were significantly lower than those of enterococci in freshwater but were not significantly different in saltwater. IO persistence according to mesocosm treatment followed the trend: contaminated soil > wastewater > dog feces. E. coli ribotyping demonstrated that certain strains were more persistent than others in freshwater mesocosms, and the distribution of ribotypes sampled from mesocosm waters was dissimilar from the distribution in fecal material. These results have implications for the accuracy of MST methods, modeling of microbial populations in water, and efficacy of regulatory standards for protection of water quality.  相似文献

Factors affecting fecal microorganism survival and distribution in the Antarctic marine environment include solar radiation, water salinity, temperature, sea ice conditions, and fecal input by humans and local wildlife populations. This study assessed the influence of these factors on the distribution of presumptive fecal coliforms around Rothera Point, Adelaide Island, Antarctic Peninsula during the austral summer and winter of February 1999 to September 1999. Each factor had a different degree of influence depending on the time of year. In summer (February), although the station population was high, presumptive fecal coliform concentrations were low, probably due to the biologically damaging effects of solar radiation. However, summer algal blooms reduced penetration of solar radiation into the water column. By early winter (April), fecal coliform concentrations were high, due to increased fecal input by migrant wildlife, while solar radiation doses were low. By late winter (September), fecal coliform concentrations were high near the station sewage outfall, as sea ice formation limited solar radiation penetration into the sea and prevented wind-driven water circulation near the outfall. During this study, environmental factors masked the effect of station population numbers on sewage plume size. If sewage production increases throughout the Antarctic, environmental factors may become less significant and effective sewage waste management will become increasingly important. These findings highlight the need for year-round monitoring of fecal coliform distribution in Antarctic waters near research stations to produce realistic evaluations of sewage pollution persistence and dispersal.  相似文献

The fecal bacteria in natural waters may pose serious risks on human health. Although many source tracking methods have been developed and used to determine the possible sources of the fecal pollution, little is known about the overall diversity and abundance of fecal bacterial community in natural waters. In this study, a method based on fecal bacterial sequence library was introduced to evaluate the fecal bacterial profile in the Yangtze River (Nanjing section). Our results suggested that the Yangtze River water harbors diverse fecal bacteria. Fifty-eight fecal operational taxonomic units (97% identity level) were detected in the Yangtze River water samples and the relative abundance of fecal bacteria in these samples ranged from 0.1 to 8%. It was also found that the relative abundances of the fecal bacteria in locations near to the downstream of wastewater treatment plants were obviously higher than those in other locations. However, the high abundance of fecal bacteria could decrease to the normal level in 2~4 km in the river due to degradation or dilution, and the overall fecal bacteria level changed little when the Yangtze River flew through the Nanjing City. Moreover, the fecal bacteria in the Yangtze River water were found to be highly associated (Spearman rho = 0.804, P < 0.001) with the potential pathogenic bacteria. Collectively, the findings in this study reveal the diversity, abundance, and possible sources of fecal bacteria in the Yangtze River and advance our understandings of the fecal bacteria community in the natural waters.

The determination of bacterial densities in aquatic sediments generally requires that a dilution-mixing treatment be used before enumeration of organisms by the most-probable-number fermentation tube method can be done. Differential sediment and organism settling rates may, however, influence the distribution of the microbial population after the dilution-mixing process, resulting in biased bacterial density estimates. For standardization of sample preparation procedures, the influence of settling by suspended sediments on the fecal coliform distribution in a mixing vessel was examined. This was accomplished with both inoculated (Escherichia coli) and raw, uninoculated freshwater sediments from Saguaro Lake, Ariz. Both test sediments were coarse (greater than 90% gravel and sand). Coarse sediments are typical of southwestern U.S. lakes. The distribution of fecal coliforms, as determined by the most-probable-number method, was not significantly influenced by sediment settling and remained homogenous over a 16-min postmix period. The technique developed for coarse sediments may be useful for standardizing sample preparation techniques for other sediment types.  相似文献

Long-term monitoring data (1968–2008) were used to investigate internal phosphorus (P) loading following external P loading reduction in shallow Loch Leven, Scotland. A whole-lake sediment P inventory (upper 3 cm of sediment; 2005) suggested a release-potential of 29.7 tonnes (t) from the release sensitive sediment P pools. 18.5 t was contained within shallow water sediments (<4.5 m water depth) with 7.6 t in deeper water sediments below the photic zone (>5 m water depth). The “observed” release (<5.1 t), estimated using a water column P mass balance approach (1989–2008), was <5.1 t, indicating the presence of regulating mechanisms. Observed P release declined between 1989 and 2008, with the exception of 2003–2006. Observed P release estimates were positively correlated with annual average water column P concentration after 1989, highlighting the role of internal loading in maintaining poor water quality conditions after management intervention. Multiple regression analysis suggested that internal loading was driven by the wave mixed depth in spring (positive driver), summer water temperature (positive driver) and spring water clarity transparency (negative driver). The potential importance of biological and physico-chemical feedback mechanisms in the regulation of benthic–pelagic coupling and water quality in Loch Leven are discussed.  相似文献

Water suppliers have often implicated roosting birds for fecal contamination of their surface waters. Geese and gulls have been the primary targets of this blame although literature documenting the fecal coliform content of these birds is quite limited. To determine the actual fecal coliform concentrations of these birds, fecal samples from 249 ring-billed gulls and 236 Canada geese in Westchester County, N.Y., were analyzed over a 2-year period. Results indicate that gull feces contain a greater average concentration of fecal coliform bacteria per gram (3.68 x 10(8)) than do goose feces (1.53 x 10(4)); however, average fecal sample weights of the geese were more than 15 times higher than those of the gulls.  相似文献

Salmonella spp. were not, however, present in any sample containing less than 230 fecal coliforms per 100 g. Analysis of the data suggests that low fecal coliform levels in both fresh and stored oysters are good indicators of the absence of Salmonella spp., but that high levels of fecal coliforms are somewhat limited in predicting the presence of Salmonella spp. E. coli levels correlated very strongly with fecal coliform levels in both fresh and stored oysters and clams, suggesting that there is no advantage in replacing fecal coliforms with E. coli as an indicator of shellfish quality.  相似文献

River watersheds in the western part of Turkey (Aegean Sea) are anthropogenically impacted areas, due to the high population density, intensive agricultural and/or industrial activities. The aim of the present survey was to assess the microbiological quality of river waters using indicators of fecal contamination. Five rivers (Meric, Bakircay, Gediz, Kucuk Menderes and Buyuk Menderes) were sampled as seasonally from 2006 to 2008 for fecal coliforms, and fecal streptococci. In order to determine the number of fecal indicator bacteria, membrane filtration method was used. The minimum fecal coliforms and fecal streptococci were counted from the samples taken from the Buyuk Menderes River (5 × 101–3 × 101 CFU/100 ml) especially in the spring and autumn. The highest fecal coliform result was obtained in winters (1.3 × 106 CFU/100 ml) while maximum fecal streptococci value was detected in autumns (6.3 × 104 CFU/100 ml) in the Kucuk Menderes River during 2006–2008. Additionally, the relationships between the fecal indicator bacteria and physico-chemical parameters (temperature, conductivity and pH) were also evaluated, but no significant relationships were observed between the fecal indicator bacteria and environmental parameters. Considering the findings of these studies, it can be said that there is fecal pollution in the rivers reaching the Aegean Sea. The results show that there are a great number of microbial pollution sources in the areas where the river passes through and thus, in order to implement strategies to improve water quality in these rivers, monitoring of these rivers should continue.  相似文献

Concentrations of hypophosphite and phosphite oxidizing bacteria were found to be high, relative to bacterial concentrations growing on phosphate, in sediment and soil during winter and summer seasons from 12 common terrestrial and aquatic sites using a most probable number method. The percent of total culturable bacterial concentrations that could use these reduced phosphorus compounds as a sole source of phosphorus were as follows: hypophosphite, 7–100%; phosphite, 10–67%; aminoethylphosphonate, 34–270%. The average MPN/g (±SEM) was as follows: phosphate, 6.19 × 106 (±2.40 × 106); hypophosphite, 2.61 × 106 (±1.35 × 106) phosphite, 1.91 × 106 (±1.02 × 106); aminoethylphosphonate, 3.90 × 106 (± 1.95 × 106). Relatively high concentrations of reduced phosphorus oxidizing bacteria were found in both pristine sites and sites with urban and agricultural disturbance. Concentrations of reduced phosphorus oxidizing bacteria in anoxic sediments and soil were equivalent. Our data indicate that reduced phosphorus oxidizing bacteria are abundant in the environment and provide strong evidence for the importance of bacterial P oxidation in nature.  相似文献

Background  

Microbiological indicators are commonly used in the assessment of public health risks associated with fecal contamination of freshwater ecosystems. Sediments are a reservoir of microorganisms, and can thus provide information on past pollution events, not obtainable through the testing of surface water. Moreover, pathogens present in sediment may represent future threats to human health. Clostridium perfringens, a typical colonizer of sediments, has been suggested as an alternative indicator of fecal pollution. In order to be suitable for such purpose, the microorganism should be widely distributed in contaminated environments. The objective of this study was thus to determine the composition of the anaerobic community in sediment samples of the lower Tiber basin, in central Italy, through a combined approach involving granulometric analysis of sediment samples, as well as a microbiological and molecular (16S rRNA) analysis of strains.  相似文献

The repair detection procedure of Speck et al. (Appl. Microbiol. 29:549-550, 1975) was adapted for the enumeration of coliforms, fecal coliforms, and enterococci in seafood and environmental samples. Samples were pour plated with Trypticase soy agar, followed by a 1- to 2-h incubation to effect repair; the plates were then overlaid with the selective medium and incubated. Violet red bile agar and an incubation temperature of 45 degrees C were used as the selective conditions for fecal coliforms, and KF streptococcal agar was used for the enumeration of enterococci. The method was more efficient than the standard most-probable-number method for fecal coliform enumeration and also allowed enumeration of the injured cells, which might have remained undetected when selective medium in the most-probable-number method was used. The repair detection method effectively recovered the injured portion of the population of enterococci capable of growing on KF streptococcal agar. The repair enumeration method was not suitable for coliforms in marine samples because associative marine bacteria mimicked coliforms in violet red bile agar plates incubated at 35 degrees C. The marine bacteria did not grow at 45 degrees C and therefore did not interfere with fecal coliform enumeration.  相似文献

Rapid microbial metabolism and a large phosphorus uptake potential were observed in surface sediments of Lake George, New York. This sediment (termed the flocculent layer) also exhibited a phosphorus limited condition and a large reservoir of inorganic phosphorus associated with humic substances. These observations suggest that the empirically observed phosphorus retention in oligotrophic lake sediments may be promoted by a rapid cycling of phosphorus between microflora and its associated organic matter.  相似文献

McMurdo Station, Antarctica, has discharged untreated sewage into McMurdo Sound for decades. Previous studies delineated the impacted area, which included the drinking water intake, by using total coliform and Clostridium perfringens concentrations. The estimation of risk to humans in contact with the impacted and potable waters may be greater than presumed, as these microbial indicators may not be the most appropriate for this environment. To address these concerns, concentrations of these and additional indicators (fecal coliforms, Escherichia coli, enterococci, coliphage, and enteroviruses) in the untreated wastewater, water column, and sediments of the impacted area and drinking water treatment facility and distribution system at McMurdo Station were determined. Fecal samples from Weddell seals in this area were also collected and analyzed for indicators. All drinking water samples were negative for indicators except for a single total coliform-positive sample. Total coliforms were present in water column samples at higher concentrations than other indicators. Fecal coliform and enterococcus concentrations were similar to each other and greater than those of other indicators in sediment samples closer to the discharge site. C. perfringens concentrations were higher in sediments at greater distances from the discharge site. Seal fecal samples contained concentrations of fecal coliforms, E. coli, enterococci, and C. perfringens similar to those found in untreated sewage. All samples were negative for enteroviruses. A wastewater treatment facility at McMurdo Station has started operation, and these data provide a baseline data set for monitoring the recovery of the impacted area. The contribution of seal feces to indicator concentrations in this area should be considered.  相似文献
