Ant-dispersed plants usually produce seeds with appendages (elaiosomes) as reward for ants. Plants that produce high-quality elaiosomes benefit because ants preferentially disperse their diaspores. We therefore hypothesized that seeds and elaiosomes differ in chemical composition in ways that make elaiosomes of high nutritional quality for ants, capable of providing essential dietary components that explain the increased fitness and higher gyne production documented for colonies with elaiosome consumption. To test the hypothesis we analysed the content and composition of lipids, amino acids, soluble carbohydrates, proteins and starch in seeds and elaiosomes of 15 central European ant-dispersed plants. After separating the different fractions, total lipids were determined gravimetrically, fatty acids and soluble carbohydrates were detected by gas chromatography (GC) and GC–mass spectrometry, free amino acids by an amino acid analyser while starch and protein were analysed photometrically. Seeds accumulated high molecular weight compounds such as proteins and starch, whereas elaiosomes accumulated more easily digestible low molecular weight compounds such as amino acids and monosaccharides. Analysis of similarities and similarity percentages analysis demonstrated that the composition of fatty acids, free amino acids and carbohydrates differed markedly between elaiosomes and seeds. The most important difference was in total amino acid content, which was on average 7.5 times higher in elaiosomes than in seeds. The difference was especially marked for the nitrogen-rich amino acid histidine. The availability of essential nutrients and, in some species, the higher nitrogen content in elaiosomes suggest that their nutritional value for larvae plays a key role in this interaction. Two breeding lines of “zero-erucic-acid” rapeseed (Brassica napus) were grown in climate chambers at a constant night temperature (12°C) and constant photoperiod (16 hours) but with different day temperatures (15, 20 and 25°C). Samples of leaves, siliques and immature seeds were analysed for total fatty acid pattern. The content of different acyl lipids and the fatty acid pattern of these lipids were also determined in some of the samples by use of preparative TLC followed by GLC of the fatty acids. The mature seeds produced by ten plants of each selection in each climate were analysed separately for total fatty acid composition. Mono- and digalactosyl diglycerides (MGDG, DGDG) were the predominant acyl lipids in leaves and siliques. In developing seeds they also were more abundant than the phospholipids, but in this case the neutral lipids, mainly triacylglycerols, contained about 95% of the total fatty acids. Large variations were found in the fatty acid composition of monogalactosyl diglyceride and digalactosyl diglyceride, isolated from leaves, siliques and immature seeds. The palmitic acid content of leaf MGDG was about 15 %, atypically high for MGDG from photosynthetic tissue. The linolenic acid content of the MGDG was about 45 %, 30 % and 10 % in the leaf, silique and seed tissues respectively. A hexadecatrienoic acid (16: 3) was found almost exclusively in the MGDG samples of leaves, siliques and immature seeds (about 25 %, 10 % and 3 % 16:3 respectively). The lipids of siliques — mainly photosynthetising tissue — were different from those of leaves and had especially high contents of stearic acid (6–12 % in the different lipids). For all lipid classes studied, leaves grown at the lowest day temperature had a slightly lower oleic and higher linolenic acid content than those grown at the highest temperature. On the other hand, increasing the day temperature caused a decreased level of oleic, an increased level of linoleic and an essentially unchanged level of linolenic acids in the mature seeds from both selections.  相似文献

The moisture, lipids and fatty acid composition of developing winged bean (Psophocarpus tetragonolobus) seeds were studied. The moisture content decreased steadily as the seeds matured. The lipid content increased gradually and reached a maximum ca 6 weeks after flowering (WAF). In the early stage (2 WAF) of the developing seeds there were more polar lipids (glycolipids and phospholipids) than neutral lipids but, as the seeds developed, neutral lipids gradually accumulated while the polar lipids decreased until 6 WAF. Thereafter, both the neutral lipid and polar lipid levels remained little changed. The amounts of palmitic and stearic acids decreased, but the level of behenic acid increased as the seeds matured. On the other hand, the oleic acid content increased while that of linolenic acid decreased rapidly as the seeds matured. The concentration of linoleic acid, however, fluctuated during the development of the seeds.  相似文献

This investigation was conducted to observe changes in the fatty acid distributions of glycolipids (GL) and phospholipids (PL) in cotyledons of soybean seeds which were germinated either in the dark or the light at 28°C for 8 days. The GL isolated from the total lipids of cotyledons at different germinating stages were : acyl sterylglycoside (ASG), monogalactosyl diglyceride (MGD), digalactosyl diglyceride (DGD) and sulfolipid (SL). The PL isolated from the same total lipids as described above were : diphosphatidyl glycerol (DPG), phosphatidic acid (PA), phosphatidyl ethanolamine (PE), phosphatidyl glycerol (PG), phosphatidyl choline (PC) and phosphatidyl inositol (PI).

During germination of soybean seeds, the content of linoleic and linolenic acids in MGD or DGD was markedly higher than that of the other GL. The positional distribution of fatty acids in PE, PC and PI was shown in all PL, in which saturated fatty acids, especially palmitic acid, were highly concentrated in position 1 and unsaturated fatty acids, especially linoleic acid, mainly occupied position 2. A remarkable difference in the changing patterns of fatty acid composition, which depended on the germinating conditions tested, was observed between GL and PL. The changes in fatty acid composition of GL were more marked in the light-grown seedlings than in the dark-grown, whereas those of PL were more remarkable in the latter than in the former. Therefore, the positional distribution of fatty acids in PL was more evident in the light-grown seedlings than in the dark-grown ones.

These results suggest the metabolic fate of GL and PL in cotyledons of soybean seeds, probably owing to the differences in the two germinating conditions tested.  相似文献

Germinated oil palm seeds were placed in special trays in the field and at different intervals the haustoria were harvested. Changes in haustorial lipids were followed until the eighth week after germination. The moisture content decreased while the lipids increased. The haustorial fatty acid profile was similar to that of the palm kernel. Changes in fatty acid composition at different weeks after germination were observed. Background

In recent years, the desire to adopt a healthy diet has drawn attention to legume seeds and food products derived from them. Mash bean is an important legume crop used in Pakistan however a systematic mapping of the chemical composition of mash bean seeds is lacking. Therefore seeds of four mash bean (Vigna mungo (L.) Hepper, family Leguminoseae) cultivars (NARC-Mash-1, NARC-Mash-2, NARC-Mash-3, NARC-Mash-97) commonly consumed in Pakistan have been analyzed for their chemical composition, antioxidant potential and biological activities like inhibition of formation of advanced glycation end products (AGE) activity and tyrosinase inhibition activity.

Results

The investigated cultivars varied in terms of biochemical composition to various extents. Mineral composition indicated potassium and zinc in highest and lowest amounts respectively, in all cultivars. The amino acid profile in protein of these cultivars suggested cysteine is present in lowest quantity in all cultivars while fatty acid distribution pattern indicated unsaturated fatty acids as major fatty acids in all cultivars. All cultivars were found to be rich source of tocopherols and sterols. Fourier transform infrared spectroscopy (FTIR) fingerprints of seed flour and extracts indicated major functional groups such as polysaccharides, lipids, amides, amines and amino acids. Results indicated that all investigated cultivars possessed appreciable antioxidant potential.

Conclusions

All cultivars are rich source of protein and possess sufficient content of dietary fiber, a balanced amino acid profile, low saturated fatty acids and antioxidant capacity that rationalizes many traditional uses of seeds of this crop besides its nutritional importance. The collected data will be useful for academic and corporate researchers, nutritionists and clinical dieticians as well as consumers. If proper attention is paid, it may become an important export commodity and may fetch considerable foreign exchange for Pakistan.  相似文献

The content and composition of lipids were studied in the seeds of radish plants (Raphanus sativus L. var. radicula D.C., cv. Rosovo-krashyi s belym konchikom) grown from “seed to seed” in 2008 and 2009 in the greenhouse of the Institute of Plant Physiology in a permanent horizontal magnetic field (PMF) of Helmholz coils with the strength of ~400 A/m, in soil culture, at natural day length, and a temperature changing during the day. PMF suppressed all stages of radish plant development, from the appearance of alternative leaves to the formation of pods and mature seeds. In plants of the North-South magnetically oriented type (NS MOT), PMF reduced the number and weight of seeds; in the West-East magnetically oriented type (WEMOT), the number of seeds was reduced but their weights increased. In the seeds of the first generation of NS MOT, the total lipid content was higher than in the seeds of WE MOT. The amount of polar lipids in the seeds of NS MOT increased, whereas in the seeds of WE MOT it decreased or remained unchanged as compared with control. The content of neutral lipids reduced in both plant types. The strongest changes in the fatty acid composition of lipids with the highest content of unsaturated fatty acids were observed in the seeds of WE MOT in 2008. The weak PMF-induced differences in the changes of lipid composition and content in the seeds of different MOTs were evidently determined by seed sensitivity to the direction of field action. It is suggested that the occurrence of different MOTs increases the tolerance of plant population to unfavorable environmental factors, thus affecting its survival.  相似文献

Expression of a California bay lauroyl-acyl carrier protein thioesterase (MCTE) in developing seeds of transgenic oilseed rape alters the fatty acid composition of the mature seed, resulting in up to 60 mol% of laurate in triacylglycerols. In this study, we examined the metabolism of lauric acid and 14C-acetate in developing seeds of oilseed rape that express high levels of MCTE. Lauroyl-CoA oxidase activity but not palmitoyl-CoA oxidase activity was increased several-fold in developing seeds expressing MCTE. In addition, isocitrate lyase and malate synthase activities were six- and 30-fold higher, respectively, in high-laurate developing seeds. Control seeds incorporated 14C-acetate almost entirely into fatty acids, whereas in seeds expressing MCTE, only 50% of the label was recovered in lipids and the remainder was in a range of water-soluble components, including sucrose and malate. Together, these results indicate that the pathways for beta-oxidation and the glyoxylate cycle have been induced in seeds expressing high levels of MCTE. Although a substantial portion of the fatty acid produced in these seeds is recycled to acetyl-CoA and sucrose through the beta-oxidation and glyoxylate cycle pathways, total seed oil is not reduced. How is oil content maintained if lauric acid is inefficiently converted to triacylglycerol? The levels of acyl carrier protein and several enzymes of fatty acid synthesis were increased two- to threefold at midstage development in high-laurate seeds. These results indicate that a coordinate induction of the fatty acid synthesis pathway occurs, presumably to compensate for the lauric acid lost through beta-oxidation or for a shortage of long-chain fatty acids.  相似文献

We have studied the effects of a weak permanent magnetic field (PMF) with strength of 403 A/m on the composition and content of polar and neutral lipids and the composition of their fatty acids (FAs). The lipids were isolated from the third, fourth, and fifth leaves of onion (Allium cepa L., cv. Arzamasskii) plants, and their composition was determined using TLC and GLC techniques. Plants growth under the conditions of a natural geomagnetic field served as a control. Most intense changes in the lipid content induced by PMF were observed in the fourth onion leaf. The content of total lipids and that of polar lipids (glyco-and phospholipids) changed, whereas the content of neutral lipids either decreased or remained unchanged. The phospholipid/sterol ratio increased, causing an increase in the fluidity of the membrane lipid bilayer. PMF induced an increase in the concentration of linolenic acid and the relative content of total unsaturated FAs. The effects of PMF on the content and composition of lipids in the third and fifth onion leaves were less pronounced, demonstrating differences between the leaves of various ages in their sensitivity to the effects of magnetic field. It is concluded that changes in the weak PMF within the limits of changes in the strength of geomagnetic field in the course of evolution can affect biochemical and physiological processes of plants.  相似文献

Fatty acid composition of total lipids of gonads was rather stable; except for the two essential acids 20:523 and 22:6w3, their percentage depends on that of food r = 0.65 and 0.65 accordingly). Fatty acid composition of neutral lipids was more diverse (in number and degree of variety) as compared to polar lipids.

Larvae released from oysters with gonads rich in lipids had a higher percentage of lipids, triacylglycerols, size and a lower ash percentage and value of ratio PE/PC, as compared to larvae from gonads with low lipid content. Total lipid and triacylglycerol contents in gonads correlate rather well with those in larvae (r = 0.77 and 0.47 accordingly).
  • 7.7. Phospholipid class composition of larvae strongly depends on that of gonads. All the correlations are high and positive in character (except for phosphatidylinositol).
  • 8.8. Amino acid composition of larvae depends on that of gonads and, as in the case with gonads, the same essential acids are accumulated in the first place.
  • 9.9. Fatty acid composition of total lipids of newly released larvae was rather stable and independent on that of gonads except for total polyunsaturated acids (r = 0.70) and 20:5w3 (r = 0.65). Fatty acid composition of neutral lipids was lesser diverse (in number and degree of variation) as compared to polar lipids.

Lipids from cultured cells, leaves and seeds of two varieties each of soybean (Glycine max) and oil seed rape (Brassica napus) were separated into neutral lipids, glycolipids and phospholipids and their fatty acids were analysed. Usually, the fatty acid composition differed between corresponding fractions from cultured cells, leaves and seeds. Differences were least marked in (i) the phospholipids from cultured cells and leaves of soybean and (ii) the neutral lipids from cultured cells and seeds of rape. In the cultured cells, the fatty acid composition of the phospholipids differed from that of the glycolipids and neutral lipids, and fatty acids of chain length greater than C18 comprised a large proportion of the fatty acids of the glycolipids.  相似文献

We previously reported on Vicia narbonensis seeds with largely decreased alpha- D-glucose-1-phosphate adenyltransferase (AGP; EC 2.7.7.27) due to antisense inhibition [H. Weber et al. (2000) Plant J 24:33-43]. In an extended biochemical analysis we show here that in transgenic seeds both AGP activity and ADP-glucose levels were strongly decreased but starch was only moderately reduced and contained less amylose. The flux control coefficient of AGP to starch accumulation was as low as 0.08, i.e. AGP exerts low control on starch biosynthesis in Vicia seeds. Mature cotyledons of antisense seeds had increased contents of lipids, nitrogen and sulfur. The protein content was higher due, in particular, to increased sulfur-rich albumins. Globulin fractions of storage proteins had a lower ratio of legumin to vicilin. Isolated cotyledons partitioned less [14C]sucrose into starch and more into soluble sugars with no change in the protein fraction. Respiration of isolated cotyledons and activities of the major glycolytic and carbohydrate-metabolizing enzymes were not affected. Sucrose and the hexose-phosphate pool were increased but UDP-glucose, 3-phosphoglyceric acid, phospho enolpyruvate, pyruvate, ATP and ADP were unchanged or even lower, indicating that carbon partitioning changed from starch to sucrose without affecting the glycolytic and respiratory pathways. Soluble compounds were increased but osmolality remained unchanged, indicating compensatory water influx resulting in higher water contents. Developmental patterns of water and nitrogen accumulation suggest a coupled uptake of amino acids and water into cotyledons. We conclude that, due to higher water uptake, transgenic cotyledons take up more amino acids, which become available for protein biosynthesis leading to a higher protein content. Obviously, a substantial part of amino acid uptake into Vicia seeds occurs passively and is osmotically controlled and driven by water influx.  相似文献

The effect of mystatin on the plasmic membranes of the rabbit liver after intravenous administration of the antibiotic to the animals in a dose of 5 mg/kg was studied. It was found that intravenous administration of nystatin had no effect on the quantitative content of protein, lipids and nucleic acids in the plasmic membranes of the liver. The method of electrophoresis in polyacrylamide gel revealed significant changes in the composition of the liver membrane protein due to the treatment with nystatin. The effect of nystatin on the composition of lipids and fatty acids contained in the membrane lipids was also investigated. The data of the thin layer chromatography showed that nystatin did not affect the qualitative composition and the content of separate lipid fractions in the lipids of the liver plasmic membranes. However, the fatty acid analysis of the membrane lipids after intravenous administration of nystatin revealed a number of qualitative and quantitative differences in the composition of the lipid fatty acids of the membranes tested. The results showed that nystatin affected the membrane structures of the rabbit liver cells.  相似文献

We studied the alterations in phenolic compounds in grape seeds during their stratification and germination under optimal conditions (+25 °C) and at low temperature (+10 °C). Biological materials in the study were seeds of Vitis riparia. Phenolic compounds were extracted from defatted seeds using 80 % methanol or 80 % acetone. The content of total phenolics was determined with the Folin-Ciocalteau reagent, while the content of tannins was determined by vanillin assay and the protein (BSA) precipitation method. The RP-HPLC method was used to determine phenolic compounds (phenolic acids, catechins) in the extracts. High amounts of tannins, catechins, gallic acid and lesser amounts of p-coumaric acid were found in the seeds. The content of total phenolics in acetone extracts was higher than that obtained using methanol. The amounts of phenolic acids and tannins found in V. riparia seeds after stratification were much lower. It may confirm a possible role of these compounds in dormancy of V. riparia seeds. After 72 h of low temperature treatment, inhibition of grape root growth and biochemical changes in seeds were detected. The chilling stimulated increased accumulation of some phenolic compounds (free gallic acid and catechins) in the seeds. These substances can protect plants against some abiotic stressors.  相似文献

Decrease in seed viability and germination rate may be caused by biochemical changes associated with seed ageing. Different biochemical assays were conducted to investigate the changes occurring at the ageing of Bambusa bambos seeds. A reduction in the total content of food reserves such as sugars, proteins and lipids were recorded. Decreased activity of peroxidase, acid phosphatase, alkaline phosphatase were also noticed during accelerated ageing. A substantial increase in total free amino acids and the activity of amylases confirms the degradation of stored biomolecules in seeds during ageing. A comparative analysis of fatty acid composition of lipid components from skeletal muscle sarcolemma and sarcoplasmic reticulum membranes of rabbits and crayfishes Astacus fluviatilis and A. leptodactylus has been made by means of gas-liquid chromatography of methyl esters of fatty acids. There are slight differences between external and internal membranes in fatty acids composition of lipids in the same animal. Considerable differences were found, however, when lipids in corresponding membranes of different species were compared. There are more unsaturated fatty acids in the crayfish than in the rabbit membranes. The most expressive differences in fatty acid composition between the two animals concern the content of linoleic acid type. Polyunsaturated acids in the crayfish are mostly of a 3 type and those in the rabbit of a 6 type. During biochemical evolution the following changes seem to take place, a decrease in the amount of unsaturated fatty acids as well as of polyunsaturated ones of a 3 type and an increase of those of the 6 type. These changes very probably reflect the transition from an aquatic to a terrestrial habitat.  相似文献

The fatty acid composition of total lipids and phospholipids of duck salt gland Na,K-ATPase (outer plasma membrane) and of rabbit skeletal muscle Ca-ATPase (intracellular membrane) was investigated. The bulk of Na,K-ATPase fatty acids is represented by palmitic (16:0), oleic (18:1), stearic (18:0) and arachidonic (20:4) acids. The duck salt gland is characterized by rather a high content of unsaturated fatty acids, especially of arachidonic acid. The unsaturation index of total-lipid fatty acids increases during purification of these preparations in the following order: homogenate greater than microsomal fraction greater than purified enzyme. The fatty acid composition of Na,K-ATPase total lipids and phospholipids reveals certain differences. Phospholipids contain more stearic and liholeic (18:2) acids than total lipids, but the level of arachidonic acid in them is twice as low. Besides, phospholipids were found to contain polyunsaturated docosohexaenic (22:6) acid. The bulk of fatty acids of rabbit skeletal muscle Ca-ATPase total lipids and phospholipids is represented by 16:0, 18:0, 18:1 and 18:2 acids. The content of polyunsaturated fatty acids in this preparation is much lower than in duck salt gland Na,K-ATPase. The fatty acid composition of total lipids and phospholipids in rabbit skeletal muscle Ca-ATPase differ insignificantly. The differences in the fatty acid composition of membrane preparations under study is conditioned mainly by the fractional composition of their lipids.  相似文献

Fatty acid composition of seed lipids for 20 of the 26 genera in the Lythraceae and seed oil and protein content for nine genera are reported. The percent oil ranges from 2.7 to 34% of total weight and protein from 11.3 to 24.9%. Linoleic acid is the dominant fatty acid in seed lipids of all genera surveyed. Variations in pattern emphasize palmitic or oleic acid or both as second most abundant lipid component. There are three exceptions: in Diplusodon capric acid ranks second in abundance; in Adenaria lauric acid and oleic acid occur in approximately equal amounts as second most abundant fatty acid; in Decodon an unusual trienoic acid, previously reported only from the Compositae, is the main secondary component. Fatty acid composition of seeds in the genera is compared to that of the previously studied lythraceous genus Cuphea. Among all the genera, only Cuphea seed produces large quantities of lauric, capric, or caprylic acids, as well as a diversity of fatty acid patterns. No relationship between oil content or seed weight and habit is apparent in any genus studied, nor are differences in seed morphology reflected in composition of the seed lipids. The fatty acid patterns are judged evolutionarily conservative, with the strong exception of Cuphea, which remains unique in the Lythraceae and among all angiosperms for the diversity of patterns displayed.  相似文献

The very high density lipoprotein (VHDL) of Triatoma infestans hemolymph from adult males has been isolated and purified by two-step density gradient ultracentrifugation. It appears to be homogeneous as judged by native polyacrylamide gel electrophoresis. The content of VHDL in hemolymph was estimated to be 8 mg protein/ml. The purified protein has a molecular weight (Mr) of 450,000, is composed of six subunits of Mr approximately equal to 77,000, and possesses a high content of aromatic amino acids. This protein is glycosylated and contains 3% of lipids by weight with a remarkable amount of free fatty acids (25% of total lipids). The T. infestans VHDL has a different lipid and amino acid composition from lipophorin. The lipid composition and the spectroscopic studies using cis-parinaric acid indicated a high fatty acid binding affinity. It has nine binding sites per mol of VHDL. Competence studies revealed that VHDL has its highest affinity for the binding of palmitic acid followed by stearic and arachidonic acids.  相似文献

Total and free amino acid composition of germinating cotton seeds (Gossypium hirsutum L.) was determined. The germinating seeds were separated into cotyledon and developing axis fractions daily and the composition of each tissue was summed to get the whole seed composition. By separating the developing seeds into these two tissue fractions, and determining total and free amino acids, a balance sheet was developed for each amino acid. This technique allowed changes in distribution with time of each amino acid to be followed in each tissue. Data for total content and amount in protein of each amino acid are presented. Asparagine increased in the whole seed, and most of this increase was found in the free pool of the developing axis. Other amino acids (e.g. arginine, glutamic acid) increased in the free pool but showed an over-all decrease, indicating that they were being metabolized. Amino acid contents of storage and nonstorage protein isolates were determined.  相似文献
