The co-transport of sodium and glucose is the first step for intestinal glucose absorption. Dietary glucose and sodium chloride (NaCl) may facilitate this physiological process in common carp (Cyprinus carpio L.). To test this hypothesis, we first investigated the feeding rhythm of intestinal glucose absorption. Carps were fed to satiety once a day (09:00 a.m.) for 1 month. Intestinal samples were collected at 01:00, 05:00, 09:00, 13:00, 17:00 and 21:00. Result showed that food intake greatly enhanced sodium/glucose cotransporter 1 (SGLT1) and glucose transporter type 2 (GLUT2) expressions, and improved glucose absorption, with highest levels at 09:00 a.m.. Then we designed iso-nitrogenous and iso-energetic diets with graded levels of glucose (10%, 20%, 30%, 40% and 50%) and NaCl (0%, 1%, 3% and 5%), and submitted to feeding trial for 10 weeks. The expressions of SGLT1 and GLUT2, brush border membrane vesicles (BBMVs) glucose transport and intestinal villus height were determined after the feeding trial. Increasing levels of dietary glucose and NaCl up-regulated mRNA and protein levels of SGLT1 and GLUT2, enhanced BBMVs glucose transport in the proximal, mid and distal intestine. As for histological adaptive response, however, high-glucose diet prolonged while high-NaCl diet shrank intestinal villus height. Furthermore, we also found that higher mRNA levels of SGLT1 and GLUT2, higher glucose transport capacity of BBMVs, and higher intestinal villus were detected in the proximal and mid intestine, compared to the distal part. Taken together, our study indicated that intestinal glucose absorption in carp was primarily occurred in the proximal and mid intestine, and increasing levels of dietary glucose and NaCl enhanced intestinal glucose absorption in carp.  相似文献

The homoeostatic regulation of bilayer order is a property of functional importance. Arguably, it is best studied in those organisms which experience and must overcome disturbances in bilayer order which may be imposed by variations in temperature of hydrostatic pressure. This article reviews our recent work on the adaptations of order in brain membranes of those fish which acclimate to seasonal changes in temperature or which have evolved in extreme thermal or abyssal habitats. The effects of temperature and pressure upon hydrocarbon order and phase state are reviewed to indicate the magnitude of the disturbances experienced by animals in their environments over the seasonal or evolutionary timescale. Acclimation of fish to altered temperature leads to a partial correction of order, while comparison of fish from extreme cold environments with those from temperate or tropical waters reveals a more complete adaptation. Fish from the deep sea also display adaptations of bilayer order which largely overcome the ordering effects of pressure.  相似文献

The crucian carp (Carassius carassius, L.) and the brown trout (Salmo trutta, L.) are vanishing from freshwater ecosystems of central Europe. To conserve both species, tailor‐made conservation management of habitats and populations of both species was implemented and tested in the Czech Republic (central Europe). This management was adjusted to reflect the ecological needs of both species. This study aimed to describe the results of a tested in situ management and to analyze the population growth of brown trout and crucian carp under ideal conditions. An experiment was performed at 14 small gravel pit lakes. Seven of them were adjusted to fit the crucian carp habitat requirements while the other seven were treated as a control group. The same experiment was done on 14 smaller streams and with brown trout. The occurrence and growth of crucian carp and brown trout were surveyed over 2 years. A significantly faster growth of both crucian carp and brown trout was observed on the adjusted lakes and streams in comparison to the control group ones. Trout and carp prospered on small streams and gravel pit lakes (respectively) that were free of angling, fish stocking, pollution, piscivorous predators, and competition with hybridizing species like Prussian carp (Carassius auratus, L.) or hatchery‐reared brown trout.  相似文献

Groups of common carp (Cyprinus carpio) were trained for 33 days to discriminate between two acoustic signals differing in frequency and temporal pattern. One signal (positive stimulus) was reinforced by food, while the other (negative stimulus) was not rewarded. When exposed to 3 positive and 3 negative stimuli per day (training Paradigm 1) the fish responded similarly to the two signals. When daily negative stimuli outnumbered positive stimuli (3 positive and 33 negative, training Paradigm 2) the fish responded similarly to the two signals for the first 2 weeks and then began to discriminate between them. Discriminatory response was statistically significant from Day 21 of training. After 33 days, training was switched from Paradigm 2 to Paradigm 1 and the fish maintained their discriminatory capability. The results are interpreted in terms of anticipatory frustration and risk management.  相似文献

River regulation can advantage non-native aquatic biota at the expense of native species. Nevertheless, flow regulating structures are sometimes used with the aim of achieving positive environmental outcomes in aquatic ecosystems. In the lower River Murray, Australia, drought-induced water level recession and acid sulfate soil exposure prompted the construction of an earthen levee, isolating a section of river channel (the Goolwa weir pool (GWP)) within which water levels were managed to mitigate a risk of water body acidification. The present study aimed to determine the impact of water level management on the fish community by investigating variation in species abundance and recruitment between sites subject to water level management in the GWP and unmanaged sites in Lake Alexandrina. Prior to levee construction, in August 2009, the abundance of the non-native common carp was similar in the GWP and Lake Alexandrina. Following water level management, in December 2009 and April 2010, the abundance of common carp in the GWP was approximately 1000 and 250 times greater than abundance in Lake Alexandrina, as a result of recruitment of young-of-year fish. No native freshwater species were significantly more abundant in the GWP in August 2009, December 2009 or April 2010. The results of this study suggest that the isolation of a river reach and a managed rise in water level facilitated spawning and recruitment of a non-native fish species. As such, the ecological benefits and risks of restoration and mitigation projects that involve the construction of flow regulating structures and water level management should be carefully considered.  相似文献

Cryopreservation of sperm is common procedures in aquaculture, particularly used for routine in artificial insemination. However, these application cause damages and adversely affected sperm motility, viability and consequently lower hatching rates. The objective of this study is to determine whether propolis has an effect on cryopreservation and fertilization ability and to investigate the potential protective effect of propolis on spermatozoa of Cyprinus carpio. Many studies have been done in cryopreservation offish spermatozoa, but none of them contain propolis in extender composition. The extenders were prepared by using modified Kurokura Solution to which 10% Me2SO added with different levels of propolis (0.2, 0.4, 0.6, 0.8 and 1 mg ml−1) and 10% egg yolk (as a control without propolis). The pooled semen samples diluted at the ratio of 1:9 by the extenders were subjected to cryopreservation. The percentage and duration of motility and fertilization tests of cryopreserved sperm samples have been done immediately after thawing and compared with control and fresh semen. The extenders containing propolis exhibited higher percentage motility and motility duration than control group (P < 0.05). Especially the group IV (0.8 mg ml−1 propolis) and the group V (1 mg ml−1 propolis) showed significant positive effects on both post thaw motility and hatching ability. The propolis maintained the integrity of the spermatozoa during the cryopreservation process. Evaluating with its contents, it has been shown that propolis is an appropriate cryoprotective agent in fish semen.  相似文献

The effects of temperature and pH on the phosphohydrolase activity of carp hepatic glucose-6-phosphatase (EC 3.1.3.9) have been investigated. The enzyme activity was maximum at about 308 K and in the pH range 5-6.5. The apparent Michaelis constant (KM) and Vmax of the reaction with glucose-6-phosphate were found to be 14.8 mM and 2.27 nmol/min/mg protein. The enzyme activity was partly inhibited by EDTA, while in the presence of sufficient PCMB virtually total inhibition was observed.  相似文献

CYP19 aromatase is the key enzyme in vertebrate steroidogenesis, catalyzing the conversion of C19 androgens to 17β-estradiol (E2). The objective of the present study was to assess the effect of the CYP19 inhibitors (AIs) fadrozole and anastrozole on gonadal development and sex differentiation in Cyprinus carpio and investigate the possible involvement of in vivo melatonin (MLT) production during sex differentiation. The CYP19 activity in 30 day-post fertilized (30 dpf) fingerlings was inhibited by treating with fadrozole and anastrozole in doses of 100 mg/kg and 200 mg/kg of feed. Gonado-somatic-index (GSI) of fish decreased (P < 0.005) and the changes in GSI was dose dependent. Serum testosterone (T) concentration increased (P < 0.001) after AI treatments and was negatively correlated with serum E2 concentration which decreased (P < 0.005). Morning serum MLT concentration decreased during the period of inhibited CYP19 activity with a positive correlation with E2 concentration. Sex-ratio in anastrazole (200 mg/kg) treated fish were 98.1% males while with fadrozole treatment at the same dose resulted in a 97.1% masculinization. Histological examination of fadrozole-treated fish gonads resulted in detection of atretic follicles and intensified spermiation. The protein and lipid production was depressed in AIs-treated fish. The results suggested that fadrozole and anastrozole both effectively inhibited oogenesis and ovarian development in C. carpio accelerating testicular formation. There was a physiological correlation between CYP19 activity, E2 and MLT synthesis during gonadal development and sex differentiation.  相似文献

Summary Physiological compartmentation in carp (Cyprinus carpio L.) gonads was investigated after intracardial injection of horseradish peroxidase (HRP) and two mouse anti-carp-sperm monoclonal antibodies.Immunohistochemistry revealed that a physiological barrier exists in carp testis for HRP and mouse IgG monoclonal antibody around the central lumina of the tubules in which the spermatozoa are located, but not around the cysts containing the precursor germ cells. The results with HRP were confirmed by electron microscopy. Mouse IgM monoclonal antibody did not penetrate the spermatogenic cysts. Probably because of its large size, it was almost exclusively located inside blood capillaries and only sparsely in the interstitial tissue.In the ovary, HRP was regularly distributed in the gonadal tissue, whereas the IgG antibody was predominantly localised on oogonia and early prophase oocytes. The results indicate that in contrast with the testis, no barrier around germ cells exists in the carp ovary.  相似文献

Damage to spermatozoa during cryopreservation is regarded as a major obstacle to the expansion of sperm storage technology. The authors used two-dimensional polyacrylamide gel electrophoresis and matrix-associated laser desorption/ionization time-of-flight mass spectrometry to explore whether the protein profile of common carp (Cyprinus carpio) spermatozoa is affected by cryopreservation. Fourteen protein spots were significantly altered following cryopreservation. Eleven of these were identified: three as specific membrane proteins (N-ethylmaleimide-sensitive fusion protein attachment protein alpha, cofilin 2, and annexin A4) involved in membrane trafficking, organization, and cell movement; six as cytoplasmic enzymes (S-Adenosylhomocysteine hydrolase, Si:dkey-180p18.9 protein, lactate dehydrogenase B, phosphoglycerate kinase 1, transaldolase 1, and esterase D/formylglutathione hydrolase) involved in cell metabolism, oxidoreductase activity, and signal transduction; and two as transferrin variant C and F. Based on these findings, the authors hypothesize that transferrin in cryopreserved sperm may protect spermatozoa against oxidative damage during the freeze-thaw process. Cryopreservation caused changes in spermatozoa protein profiles that may lead to decreased spermatozoa velocity, motility, and fertilization success, and to subsequent ova hatching rate.  相似文献

Summary Common carp (Cyprinus carpio L.) were acclimated to either 2, 5, 8, 11, 15, 20, or 23°C for 12 weeks (12 h light: 12 h dark). Fish did not feed after 6 weeks at temperatures below 8°C. Skinned fibres were prepared from fast myotomal muscle by freeze-drying. Measured at 0°C unloaded contraction velocity (Vmax) and maximum isometric tension generation (Po) were 2–3 times higher in the 11°C-than 23°C-acclimated groups, and had intermediate values in 15 °C-acclimated fish. Po and Vmax at 0°C were not significantly different for carp maintained at 2, 5, 8, or 11°C. Measured at the acclimation temperature of each group Vmax and Po were 51% and 71% lower for fibres from 2°C- than 23°C-acclimated fish. The results indicate a partial capacity adaptation of muscle power output in fish acclimated between 11°C and 23°C. At 8°C the ATPase activity of myofibrils was 2 times higher in fish acclimated to 8°C than to 20°C. The effects of temperature acclimation on the protein composition of myofibrils was investigated using one- and two-dimensional electrophoresis. Peptide maps of purified myosin heavy chains and actin prepared by proteolytic digestion with either Staphylococcus aureus V8 protease or chymotrypsin were similar for both acclimation groups. The molecular weights and isoelectric points of the major isoforms of tropomyosin, troponin C, troponin I, troponin T, and myosin light chains (MLC1, MLC2 and MLC3) were also similar in 8°C- and 20°C-acclimated carp. A 20 kDa molecular weight protein with a pI intermediate between that for MLC2 and MLC3 was found in myofibrils and single fibres from carp acclimated to 8°C but was not present in carp acclimated to 20°C. It is suggested that this band corresponds to a myosin light chain isoform unique to cold-acclimated fish. Evidence was also obtained that myofibrils from warm-acclimated fish contained a second minor isoform of troponin I.  相似文献

The present study was designed to investigate the effect of temperature (20 °C, 24 °C, 28 °C and 32 °C) on the heamato-biochemical and histological alterations of Cyprinus carpio communis. Increase in the temperature showed significant decrease in the serum protein, while a reduced level of blood glucose at high temperature of 32 °C was observed leading to hypoglycemic conditions in the experimental fishes. A significant correlation (P<0.01) was observed between cholesterol (Cho) and triglycerides (TG) for different temperature treatments. Elevated blood urea nitrogen (BUN) at high temperatures was a good indicator of gill osmoregulatory failure. A variation of 86.40% and 38.33%, respectively, was noticed in alanine aminotransferase (ALT) and aspartate aminotransferase (AST) at 32 °C over minimum experimental temperature of 20 °C. The increase in red blood cell (RBC) and Heamoglobin (Hb) concentration is associated with the decrease of mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC), could be the reason for observed poikilo-anisocytosis. Histological studies of different organs of experimental fishes showed accumulation of MMC's (melanomacrophagic centers) and atrophy of the interrenal tissue on exposure to various levels of temperature. These changes were related to severity of thermal stress, being most marked when high temperature was prolonged during acclimatization. Some fishes were found infested by protozoan parasite at elevated temperature of 32 °C. Increased levels of certain biochemical and haemotological parameters studied were strongly correlated with disease in the Cyprinus carpio communis species.  相似文献

Starch, composed of amylose and amylopectin, greatly influences rice cooking and textural quality, which in turn is controlled by various isoforms of several enzymes. Activity of one or more isoforms of starch‐synthesizing enzymes results in various forms of starch structure based on the amylopectin chain length and average external, internal and core chain length distribution and hence results in varying physicochemical and cooking quality. Since the synthesis of starch is highly complex, it is crucial but essential to understand its biosynthetic pathway, starch structure and effects on the physicochemical properties that control eating and cooking quality, and alongside conduct research on gene/QTL mapping for use in marker-assisted selection (MAS) with a view to improve and select cultivars with most desirable range and class of rice starch properties. This article presents the updates on current understanding of the coordination among various enzymes/isoforms towards rice starch synthesis in endosperm and their effect on rice grain physicochemical, cooking and eating qualities. The efforts in identifying regions responsible for these enzymes by mapping the gene/QTLs have provided a glimpse on their association with physicochemical and cooking properties of rice and, hence, improvement is possible by modifying the allelic pattern, resulting in down or nil regulation of a particular enzyme. The clear understanding of the tissue specific coordination between enzyme isoforms and their subsequent effect in controlling eating and cooking properties will enhance the chances to manipulate them for getting desired range of amylose content (AC) and gelatinization temperature (GT) in improved cultivars through combining desired alleles through MAS.  相似文献

Three new coumarins, 5-hydroxy-6,7-methylenedioxycoumarin, 5-(2-hydroxy-3-methoxy-3-methylbutoxy)-6,7-methylenedioxycoumarin and 5-(2-hydroxy-3-methyl-3-butenyloxy)coumarin, were isolated from the petrol ether extract of the aerial parts of Pterocaulon virgatum. Brain microsome phospholipids and their acyl groups, from temperature and oxygen acclimated goldfish, were investigated. At the lower acclimation temperature (5C) the proportion of ethanolamine- to choline-glycerophosphatides (GPE/GPC) was increased, and the proportion of phosphatidal ethanolamine value decreased. A rise in the n-6/n-3 fatty acyl group also occurred in cold acclimation. Irrespective of acclimation temperature, 25 degrees C or 5 degrees C, a partial replacement of GPC by GPE occurred when the concentration of oxygen was increased; conversely the GPE/GPC ratio decreased at the hypoxic level. The plasmalogen GPE content increased as the oxygen concentration was raised. A rise in the n-6/n-3 ratio, for ethanolamine glycerophosphatides and phosphatidyl choline, occurred when the oxygen concentration was increased (hypoxia to hyperoxia). It is concluded that the lipid alterations associated with thermal acclimation are, in part, attributable to the concomitant change in oxygen concentration.  相似文献

The oxygen consumption of temperature acclimated mummichogs, Fundulus heteroclitus (L.) weighing ≈0.1–10.0 g, was measured at 5, 13, 21, and 29 C. Between 13 and 21°C and 21 and 29°C, the values of Q10 were 1.55 and 1.04, respectively, indicating relative thermal independence of respiratory metabolic rate over this 16°C range (Q10 = 1.27). This range encompasses the normal late spring, summer, and early fall range of habitat temperature in Maine estuaries, so that mummichogs are able to grow and reproduce relatively independent of environmental temperature. Between 5 and 13°C, respiratory metabolism is very temperature sensitive (Q10 = 4.42) indicating a substantial reduction of metabolic processes at low temperatures. This enables mummichogs to conserve any metabolic reserves during the coldest months. The regression of log weight-specific oxygen consumption on log body weight was determined at each experimental temperature. All had significantly negative slopes indicating the importance of body size in mummichog respiration.  相似文献
