Biomass production by the green algae Scenedesmus obliquus and Chlorella vulgaris in intensive laboratory continuous cultures was considerably affected by the pH at which the cultures were maintained. Carbon photoassimilation experiments revealed that pH values in the range of 8 to 9 were important for determining the free CO2 concentrations in the medium. With higher pH values, additional pH effects were observed involving a decrease in the relative high affinity of low CO2-adapted algae to free CO2. The carbon uptake rate by high CO2-adapted algae after transfer to low free CO2 medium was characterized by a lag period of about 30 min, after which the affinity of the algae to CO2 increased considerably. Both continuous growth and carbon uptake experiments indicated that artificially maintained high free CO2 concentrations are recommended for maximal production in intensive outdoor algal cultures.  相似文献

An on-line technique, based on measuring the increase in pressure due to CO2 release in a closed air-tight reactor, was used to evaluate the fermentation of lactate by propionibacteria. The method was applied to batch cultures of Propionibacterium shermanii grown in yeast extract/sodium lactate medium containing lactate as a carbon source under micro-aerophilic conditions. Gas pressure evolution was compared both with substrate consumption and metabolites production and with acidification and growth. Linear relationships were found between gas pressure variation, lactate consumption and propionate and acetate production. The technique also enabled the evaluation of total CO2 produced, by taking account of pressure, oxygen and pH measurements. These results tend to show that this simple and rapid method could be useful to monitor propionic acid bacteria growth.  相似文献

Electrodialysis culture of Clostridium thermoaceticum increased the yield of acetate by its continuous removal. In normal batch cultures without pH control the yield was 4.2 g acetic acid/800 ml, while in pH-controlled culture it was 16.8 g/800 ml. Although electrodialysis cultures gave almost the same yield (15.4 g/800 ml) as that in pH-controlled cultures, sparging CO2 into the broth in electrodialysis culture increased the amount of acetic acid to 22.3 g/800 ml. CO2 sparging into normal cultures with or without pH control did not significantly increase the amount of acetate produced but yields, in terms of amounts of glucose consumed, were higher than without sparging. Abstract The experimental determination of cytoplasmic and vacuolar pH values is discussed. Despite variation in these values evidence indicates that intracellular pH values are normally regulated within narrow limits. The regulatory mechanisms proposed involve the metabolic consumption of OH& and the active efflux of H +. The evidence for intracellular pH modification in response to CO2 hydration and the production of HCO?3 and H+ is examined. Theoretical calculations and experimental data indicate that CO2 concentrations as high as 5% will lower intracellular pH. Conversely, variation in CO2 levels around atmospheric concentrations is unlikely to perturb intracellular pH. High CO2 levels are found in bulky tissues, and flooded root systems. Evidence is presented that the slow diffusion of dissolved CO2 compared to gaseous CO2 results in its accumulation. It is proposed that the accumulation of respiratory CO2 may reduce intracellular pH values when plant tissues, cells or protoplasts are maintained in a liquid culture medium. Finally, the possible role of dark CO2 fixation and organic acid synthesis in the regulation of intracellular pH is examined.  相似文献

pH control is critical in bioreactor operations, typically realized through a two-sided control loop, where CO2 sparging and base addition are used in bicarbonate-buffered media. Though a common approach, base addition could compromise culture performance due to the potential impact from pH excursions and osmolality increase in large-scale bioreactors. In this study, the feasibility of utilizing control of sparge gas composition as part of the pH control loop was assessed in Chinese hamster ovary (CHO) fed-batch cultures. Fine pH control was evaluated in multiple processes at different setpoints in small-scale ambr®250 bioreactors. Desired culture pH setpoints were successfully maintained via air sparge feedback control. As part of the pH control loop, air sparging was increased to improve CO2 removal automatically, hence increase culture pH, and vice versa. The effectiveness of this pH control strategy was seamlessly transferred from ambr®250 to 200 L scale, demonstrating scalability of the proposed methodology. © 2018 American Institute of Chemical Engineers Biotechnol. A 3-liter culture vessel has been developed for the growth of animal cells in suspension at controlled pH and dissolved oxygen partial pressure (pO2). The culture technique allows metabolically produced CO2 to be measured; provision can be made to control the dissolved CO2 partial pressure. In cultures containing a low serum concentration, gas sparging to control pO2 was found to cause cell damage. This could be prevented by increasing the serum concentration to 10%, or by adding 0.02% of the surface-active polymer Pluronic F68. The growth of mouse LS cells in batch culture without pO2 control was found to be limited by the availability of oxygen. Maximum viable cell populations were obtained when dissolved pO2 was controlled at values within the range 40–100 mm Hg.  相似文献

A general and simple equation is presented to account for the effect of CO2 absorption and the dissociation of carbonic acid in liquid on the measurement of the CO2 evolution rate (QCO2) of both anaerobic and aerobic continuous cultures. For aerobic cultures the same equation applies to the measurement of the respiratory quotient (RQ). The deviation of QCO2 and RQ, calculated from gas-phase measurements, from their true values can be assessed with two parameters: one accounts for the influence of pH, resulting from dissociation of carbonic acid, the other for the influence of operating conditions. Carbon dioxide (CO2) efflux from the soil surface, which is a major source of CO2 from terrestrial ecosystems, represents the total CO2 production at all soil depths. Although many studies have estimated the vertical profile of the CO2 production rate, one of the difficulties in estimating the vertical profile is measuring diffusion coefficients of CO2 at all soil depths in a nondestructive manner. In this study, we estimated the temporal variation in the vertical profile of the CO2 production rate using a data assimilation method, the particle filtering method, in which the diffusion coefficients of CO2 were simultaneously estimated. The CO2 concentrations at several soil depths and CO2 efflux from the soil surface (only during the snow-free period) were measured at two points in a broadleaf forest in Japan, and the data were assimilated into a simple model including a diffusion equation. We found that there were large variations in the pattern of the vertical profile of the CO2 production rate between experiment sites: the peak CO2 production rate was at soil depths around 10 cm during the snow-free period at one site, but the peak was at the soil surface at the other site. Using this method to estimate the CO2 production rate during snow-cover periods allowed us to estimate CO2 efflux during that period as well. We estimated that the CO2 efflux during the snow-cover period (about half the year) accounted for around 13% of the annual CO2 efflux at this site. Although the method proposed in this study does not ensure the validity of the estimated diffusion coefficients and CO2 production rates, the method enables us to more closely approach the “actual” values by decreasing the variance of the posterior distribution of the values.  相似文献

The effects of changes in CO2 and pH on biomass productivity and carbon uptake of Pleurochrysis carterae and Emiliania huxleyi in open raceway ponds and a plate photobioreactor were studied. The pH of P. carterae cultures increased during day and decreased at night, whereas the pH of E. huxleyi cultures showed no significant diurnal changes. P. carterae coccolith production occurs during the dark period, whereas in E. huxleyi, coccolith production is mainly during the day. Addition of CO2 at constant pH (pH-stat) resulted in an increase in P. carterae biomass and coccolith productivity, while CO2 addition lowered E. huxleyi biomass and coccolith production. Neither of these algae could grow at less than pH 7.5. Species-specific diurnal pH and pCO2 variations could be indicative of significant differences in carbon uptake between these two species. While E. huxleyi has been suggested to be predominantly a bicarbonate user, our results indicate that P. carterae may be using CO2 as the main C source for photosynthesis and calcification.  相似文献

Under selected conditions, nonlinear dynamical systems, which can be described by deterministic models, are able to generate so-called deterministic chaos. In this case the dynamics show a sensitive dependence on initial conditions, which means that different states of a system, being arbitrarily close initially, will become macroscopically separated for sufficiently long times. In this sense, the unpredictability of the EEG might be a basic phenomenon of its chaotic character. Recent investigations of the dimensionality of EEG attractors in phase space have led to the assumption that the EEG can be regarded as a deterministic process which should not be mistaken for simple noise. The calculation of dimensionality estimates the degrees of freedom of a signal. Nevertheless, it is difficult to decide from this kind of analysis whether a process is quasiperiodic or chaotic. Therefore, we performed a new analysis by calculating the first positive Lyapunov exponent L 1 from sleep EEG data. Lyapunov exponents measure the mean exponential expansion or contraction of a flow in phase space. L 1 is zero for periodic as well as quasiperiodic processes, but positive in the case of chaotic processes expressing the sensitive dependence on initial conditions. We calculated L 1 for sleep EEG segments of 15 healthy men corresponding to the sleep stages I, II, III, IV, and REM (according to Rechtschaffen and Kales). Our investigations support the assumption that EEG signals are neither quasiperiodic waves nor a simple noise. Moreover, we found statistically significant differences between the values of L 1 for different sleep stages. All together, this kind of analysis yields a useful extension of the characterization of EEG signals in terms of nonlinear dynamical system theory.  相似文献

Nitrogen removal from wastewater by algae provides the potential benefit of producing lipids for biodiesel and biomass for anaerobic digestion. Further, ammonium is the renewable form of nitrogen produced during anaerobic digestion and one of the main nitrogen sources associated with wastewater. The wastewater isolates Scenedesmus sp. 131 and Monoraphidium sp. 92 were grown with ammonium, nitrate, or urea in the presence of 5 % CO2, and ammonium and nitrate in the presence of air to optimize the growth and biofuel production of these chlorophytes. Results showed that growth on ammonium, in both 5 % CO2 and air, caused a significant decrease in pH during the exponential phase causing growth inhibition due to the low buffering capacity of the medium. Therefore, biological buffers and pH controllers were utilized to prevent a decrease in pH. Growth on ammonium with pH control (synthetic buffers or KOH dosing) demonstrated that growth (rate and yield), biodiesel production, and ammonium utilization, similar to nitrate- and urea-amended treatments, can be achieved if sufficient CO2 is available. Since the use of buffers is economically limited to laboratory-scale experiments, chemical pH control could bridge the gap encountered in the scale-up to industrial processes.  相似文献

An new cascade control system is presented that reproducibly keeps the cultivation part of recombinant protein production processes on its predetermined track. While the system directly controls carbon dioxide production mass and carbon dioxide production rates along their setpoint profiles in fed-batch cultivation, it simultaneously keeps the specific biomass growth rates and the biomass profiles on their desired paths. The control scheme was designed and tuned using a virtual plant environment based on the industrial process control system SIMATIC PCS 7 (Siemens AG). It is shown by means of validation experiments that the simulations in this straightforward approach directly reflect the experimentally observed controller behaviour. Within the virtual plant environment, it was shown that the cascade control is considerably better than previously used control approaches. The controller significantly improved the batch-to-batch reproducibility of the fermentations. Experimental tests confirmed that it is particularly suited for cultivation processes suffering from long response times and delays. The performance of the new controller is demonstrated during its application in Escherichia coli fed-batch cultivations as well as in animal cell cultures with CHO cells. The technique is a simple and reliable alternative to more sophisticate model-supported controllers.  相似文献

The control of soil nitrogen (N) availability under elevated atmospheric CO2 is central to predicting changes in ecosystem carbon (C) storage and primary productivity. The effects of elevated CO2 on belowground processes have so far attracted limited research and they are assumed to be controlled by indirect effects through changes in plant physiology and chemistry. In this study, we investigated the effects of a 4‐year exposure to elevated CO2 (ambient + 400 µmol mol?1) in open top chambers under Scots pine (Pinus sylvestris L) seedlings on soil microbial processes of nitrification and denitrification. Potential denitrification (DP) and potential N2O emissions were significantly higher in soils from the elevated CO2 treatment, probably regulated indirectly by the changes in soil conditions (increased pH, C availability and NO3 production). Net N mineralization was mainly accounted for by nitrate production. Nitrate production was significantly larger for soil from the elevated CO2 treatment in the field when incubated in the laboratory under elevated CO2 (increase of 100%), but there was no effect when incubated under ambient CO2. Net nitrate production of the soil originating from the ambient CO2 treatment in the field was not influenced by laboratory incubation conditions. These results indicate that a direct effect of elevated atmospheric CO2 on soil microbial processes might take place. We hypothesize that physiological adaptation or selection of nitrifiers could occur under elevated CO2 through higher soil CO2 concentrations. Alternatively, lower microbial NH4 assimilation under elevated CO2 might explain the higher net nitrification. We conclude that elevated atmospheric CO2 has a major direct effect on the soil microbial processes of nitrification and denitrification despite generally higher soil CO2 concentrations compared to atmospheric concentrations.  相似文献

It has been shown in this paper that stimulation of the inhibitory nerves of the neurogenic heart of Limulus, which correspond to the vagus nerves of the vertebrate heart, results in a marked diminution of CO2 production in the heart ganglion, while stimulation of the ganglion, leading to increased activity of the heart, leads also to increased CO2 production by the ganglion. This shows that inhibition of the automaticity of this ganglion by the action of its inhibitory nerves consists, not in a process of blocking, but in a diminution of those chemical reactions in the ganglion cells which give rise to the production of CO2.  相似文献

The shift from lactate production to consumption in CHO cell metabolism is a key event during cell culture cultivations and is connected to increased culture longevity and final product titers. However, the mechanisms controlling this metabolic shift are not yet fully understood. Variations in lactate metabolism have been mainly reported to be induced by process pH and availability of substrates like glucose and glutamine. The aim of this study was to investigate the effects of elevated pCO2 concentrations on the lactate metabolic shift phenomena in CHO cell culture processes. In this publication, we show that at elevated pCO2 in batch and fed‐batch cultures, the lactate metabolic shift was absent in comparison to control cultures at lower pCO2 values. Furthermore, through metabolic flux analysis we found a link between the lactate metabolic shift and the ratio of NADH producing and regenerating intracellular pathways. This ratio was mainly affected by a reduced oxidative capacity of cultures at elevated pCO2. The presented results are especially interesting for large‐scale and perfusion processes where increased pCO2 concentrations are likely to occur. Our results suggest, that so far unexplained metabolic changes may be connected to increased pCO2 accumulation in larger scale fermentations. Finally, we propose several mechanisms through which increased pCO2 might affect the cell metabolism and briefly discuss methods to enable the lactate metabolic shift during cell cultivations.  相似文献

The question of how to extrapolate point measurements of soil CO2 processes to coarser scales remains unanswered because we know little about the spatial and temporal variability in the CO2 concentration of soil air. In this work, we describe a series of simple physically-based models that simulate soil temperature, soil tension, and soil CO2 processes. We apply these models to simulate the spatial and temporal dynamics of soil CO2 concentrations throughout a small catchment in the Virginia Blue Ridge. Output from the simulations is compared with field measurements. We find that despite some model deficiencies, we are able to simulate the gross patterns through space and time of soil air CO2 concentration. During the growing season when soil temperature is high, we find that soil water status is the limiting control on soil respiration and CO2 concentration. We also find that soil CO2 concentration can be high despite low respiration values due to decreased soil diffusivity as moisture fills pore spaces.  相似文献

Some cell cycle events were compared in Chlamydomonas segnis Ettl during its development in synchronous cultures (12:12 LD) supplied with air and air enriched with 5% CO2. In cultures bubbled with air, growth resulted in production of 2 relatively small zoospores. In cultures provided with 5% CO2, 4 large zoospores were formed but not released in darkness unless the cultures were bubbled with CO2-free air and/or exposed to light. Respiration in zoospores was inhibited by high CO2 tension. In cultures maintained under continuous illumination for one cell cycle, provision of 5% CO2 led to enhanced growth, a relatively long S-phase and a 4 h delay of the second cell division. In such cultures, the DNA content of parental cells (12 h L) was insufficient to support two cell divisions. The RNA/DNA ratio of the resulting zoospores was 10 compared to 4 in air cultures. These results provided evidence that the delay of the second cell division was a consequence of the delay in DNA production.  相似文献

A key aspect of the industrial development of microalgal production processes is the excessive cost of biomass production. A solution is a combination of biodiesel production and wastewater treatment. The microalga Scenedesmus has a high lipid content and a potential extracellular phenoloxidase activity, which could improve the phycoremediation of phenolic pollutants. In this work, the most suitable growth conditions to obtain this twofold aim were analyzed. First, different strains of Scenedesmus vacuolatus microalga were tested at different pH, salinity and CO2 concentration in the gas phase. The two most promising strains were then cultivated in autotrophic and heterotrophic conditions, and were investigated in terms of efficient nitrogen removal, fatty acid profile and maximized extracellular phenoloxidase activity in the medium. The results showed two extreme conditions: (1) biomass productivity doubled when photobioreactors were sparged with 5% CO2 supplemented air with respect to cultures sparged with air (the steady state values of strain 53 were 0.138 g L?1 day?1 in the presence of air, and 0.243 in the presence of CO2 addition), and N-starvation under 5% CO2enhanced the transesterified fraction of lipids (strain 53 FAME fraction in the presence of N-starvation was 33%, in the presence of nitrogen FAME fraction was 22%); (2) phenoloxidase activity was completely suppressed by presence of 5% CO2 in the gas phase (strain 53 0.21 U mL?1), indicating clear catabolite repression for the induction of this enzyme in the algal metabolism.  相似文献

Media concentration of total soluble CO2 increases with culture age of Tetrahymena pyriformis. CO2 is a weak acid and is capable of acidifying intracellular pH (pHi). Changes in pHi have been demonstrated to affect cell metabolism and growth in many systems. For these reasons, we investigated whether the concentrations of CO2 produced in vitro were sufficient to affect cell proliferation and pHi in Tetrahymena. In this study, we used DMO to mimic the weak acid properties of CO2. DMO is freely permeable to membranes in its uncharged form and has a pKa similar to that of CO2/HCO. In addition, it has the advantages of being metabolically inert and non-volatile. At concentrations similar to endogenously produced CO2, DMO acidifies pHi and arrests culture growth. In addition, procedures are described which decrease the media CO2 concentrations in both growing and non-growing cultures. These conditions lead to increased maximum culture density at stationary phase. The data indicate that, under our conditions, accumulation of CO2 in the culture leads to cessation of growth, probably through elimination of transmembrane pH gradients, which are necessary for regulation of metabolism and growth.  相似文献
