Preleptotene chromosome contraction was observed in several clones of Lilium longiflorum cultivars Ace, Nellie White and Croft. The degree of chromosome contraction was variable, among microsporocytes of single anthers, buds of individual plants, and clones of single cultivars. It is suggested that one of the sequence of genetically controlled events that determine the orderly development of meiosis provides for the temporary inhibition of mitotic coiling, thus allowing the development of leptotene; and if this gene action is delayed or out of sequence, a period of mitotic-like chromosome contraction precedes leptotene. These cultivars appear to share an inherited predisposition for such an irregularity in sequence of gene action. Differences between clones may be attributed to differences in other genes affecting these earliest stages of meiosis. There is some evidence of external environmental influence upon this genetic predisposition for preleptotene chromosome contraction, that is, there appears to be an inverse correlation between degree of preleptotene contraction and temperature. The degree of chromosome contraction may also be influenced by different metabolic conditions affecting microsporocytes developing sequentially in the anther. Experiments are presently underway to determine whether the degree of preleptotene chromosome contraction is related to chiasma frequency.  相似文献

A period of chromosome contraction between premeiotic interphase and leptotene was regularly observed in three samples of Lilium longiflorum Croft. Extensive preleptotene chromosome contraction was also observed in L. longiflorum Ace and in the Lilium hybrid Enchantment. Although the stage resembles late mitotic prophase, microsporocytes never develop to metaphase, but despiralize to leptotene, and regular alignment, pairing and chiasma formation follow. As preleptotene chromosome contraction is discovered in an increasing number of organisms it becomes less likely that it represents a true reversion to mitosis. However its absence in many organisms and its extreme variability in others do not support the concept of preleptotene chromosome contraction as a regular meiotic stage. It is suggested that the line of demarcation between mitosis and meiosis is often imprecise, and meiocytes may fluctuate to some extent between these states before a final transition to meiosis is made. The occurrence and extent of this fluctuation may possibly be related to some externally produced substances required for the orderly development of meiotic prophase.  相似文献

Various aspects of premeiosis and meiosis in Lilium Enchantment are described. There was evidence of a premeiotic slow-down but no cells in premeiotic despiralization were observed. A relationship was found between sequence of bud development, or reproductive age of the individual, and degree of preleptotene chromosome contraction. The sequence of development of microsporocytes in the anther differed from the apex-to-base order previously reported in Lilium and, in contrast to observations in L. longiflorum cultivars, the maximum degree of preleptotene contraction was found in basal, last developing microsporocytes. Delayed despiralization of telophase nuclei was observed. There were extremely rare cells in meiotic division in anthers in which all other archesporial cells had not yet reached premeiotic interphase. Extreme variation was observed among anthers in proportions of microsporocytes in mid-stages of meiosis as well as in preleptotene contraction. These observations are discussed in relation to meiosis readiness, meiotic behavior in early and late developing regions of reproductive organs and in aging individuals, synchrony of meiotic development and rates of meiotic division.  相似文献

Summary The seasonal progression of phenophases in 21 shrub species of the Chilean matorral was analyzed. Five modules or basic units that are responsible for the aboveground architecture of the plants were characterized. These modules appear to be organized in seven different spatial arrangements. In drought-deciduous species a module type with an absolute short shoot with limited apical growth, leafy or spiny, predominated. In evergreen species long shoot and temporal leafy short shoot module types were more frequent. The spatial arrangement of morphologically different modules and the temporal sequence of their formation allow a dynamic interpretation of the modular architecture of the plants.  相似文献

A period of chromosome spiralization and contraction was observed between premeiotic interphase and leptotene in Lilium longiflorum Croft. There was variation in the extent of preleptotene spiralization and contraction of chromosomes among microsporocytes, anthers and buds. The chromosomes sometimes contracted sufficiently to be visible as separate entities. It could then be determined that the chromosomes were single and entirely separate; synapsis and crossing-over had not yet occurred. Furthermore there was no evidence of alignment or association of homologues during the preleptotene contraction period; the chromosomes appeared to be distributed at random. The chromosomes subsequently elongated into the leptotene stage. Wherever they were visible separately the chromosomes were single in early leptotene. These observations support the classical view that synapsis of homologous chromosomes takes place during zygotene, followed by crossing-over at pachytene.It is a pleasure to dedicate this paper to Dr. Sally Hughes-Schrader on the occasion of her seventy-fifth birthday.  相似文献

Summary The seven possible primary trisomics of Petunia (2 n= 14) located in the progenies of triploid, hypertriploid and hypotriploid plants were distinguished from one another and from diploid on the basis of cytological and morphological criteria. They were provisionally named as Oval, Semi, Slender, Pseudonormal, Arrow, Narrow and Giant. In three of the trisomics, the extra chromosome was identified for the first time at pachytene stage. Postpachytene studies revealed no precise relationship between the length of extra chromosome and the frequency of multiple association.  相似文献

The endemic grasshopper Cryptobothrus chrysophorus is widely distributed throughout S.E. Australia and its populations display an extensive and spectacular pattern of autosomal variation. While the standard telocentric complement of three long (L1–3), six medium (M4–9) and two short (S10–11) autosome pairs is present throughout most of its range, two quite distinct chromosome races can be defined within this species. Populations in the northern part of its distribution (northern N.S.W. and southern Queensland-northern race) are differentiated from the remainder (southern race) by fixed blocks of distal heterochromatin on autosomes M4, 5, 6, 8 and 9 and by differences in the character of the megameric M7 chromosome. Additionally, while many populations in both races show a polymorphic system of supernumerary segments on the two smallest autosomes (S10–11), that found in the northern race is both more variable and more complex. On the other hand all the populations of the southern race we have examined are polymorphic for a series of centric shifts which convert telocentrics into acro- or meta-centrics. These occur more commonly in the megameric M7 and the two smallest autosomes (S10–11) although in one population (Forbes Creek, N.S.W.) at least 12 different shifts involving 8 of the autosomes (L3, M4, 5, 6, 7, 8, 9 and S10) are known. By contrast, in the northern erace only the small autosomes (S10–11) show centric shifts. These several floating and fixed variants thus involve all chromosomes of the standard set other than the two largest autosomes (L1–2) and the X-chromosome, which appear to be invariate. Finally, morphologically distinct supernumerary (B) chromosomes, intermediate in size between the standard S10 and the M9 elements, are found in both races but are especially common in Tasmania, the most southerly point of the species range. These B-chromosomes are partly heterochromatic and partly euchromatic so that they too add to the considerable heterochromatin variation in this species.  相似文献

Summary The mitochondrial ATPase from a PHO 1 mutant (OLI 2, PHO 1, OLI 4 region on mit DNA of S. cerevisiae) was further examined. A new purification method using Lysolecithin instead of Triton allowed us to solubilize and separate a heterogeneous ATPase population from PHO 1-mitochondria: the major abnormal fraction had extremely low oligomycin-sensitivity (but normal specific immunological reactivity), while a minor normal fraction (representing about 20% of the initial mitochondrial ATPase activity) had high sensitivity and affinity for oligomycin.Moderate urea treatment of PHO 1-mitochondria leads to partial loss of ATPase activity and a concomitant increase of oligomycin-sensitivity, suggesting that a heterogeneous ATPase population exists in situ in the mitochondrial membrane: part of the major abnormal ATPase fraction is selectively inactivated by urea, producing a concomitant enrichment in the initially minor normal ATPase fraction.If the minor normal ATPase fraction is the only one capable of in vivo ATP synthesis, the deficient but oligomycin-sensitive cell growth and oxidative phosphorylation in vitro are readily explained.Further structural studies are under way to ascertain whether the minor normal ATPase fraction is strictly identical to the wild type, in which case PHO 1 is a regulatory gene, or not, in which case PHO 1 is a structural gene.  相似文献

Summary A single injection of -interferon (-IFN) (30 000 units/mouse), a major biological modifier of natural killer (NK) cytolytic activity, strongly stimulated NK activity in normal mice, as expected, while the same treatment did not statistically alter the NK response in cyclophosphamide (CY)-suppressed animals.We investigated the possibility of thymosin 1 cooperating with -IFN in boosting NK activity in CY-suppressed animals.The results show that treatment with thymosin 1 (200 g/kg) for 4 days, followed by a single injection of -IFN 24 h before testing, strongly restored NK activity in CY-suppressed mice. Thymosin 1 was, moreover, able to accelerate the recovery rate of NK activity in bone marrow reconstituted murine chimeras.Taken together the data support the concept that the synergic effect between thymosin 1 and -IFN could be the result of effects on differentiation of the NK lineage at different levels.  相似文献

Summary Motor nerve terminals on white and intermediate muscle fibers of the Atlantic hagfish (Myxine glutinosa, L.) contain translucent synpatic vesicles and about 1–2% dense-core vesicles. Terminals on red muscle fibers contain up to 40% dense-core vesicles with diameter 800–1100 Å. Examinations for formaldehyde-induced fluorescence indicate yellow fluorescence (5-HT ?) apparently corresponding with terminal axons on red muscle fibers in craniovelar muscles. Summary We used a cloned human cDNA probe homologous to the placenta chorionic gonadotropin subunit (CGB) and to the pituitary luteinizing hormone subunit (LHB) and Southern blotting techniques to analyse DNA from a series of rodent x human somatic cell hybrids for the presence of specific gonadotropin subunit related sequences. Our results provide evidence for the assignment and linkage of the eight genes (or pseudogenes) coding for the subunit of these glycoprotein hormones to chromosome 19. Moreover, we observed a strict concordance between the permissivity of mouse x man hybrid cells to enteroviruses (which is linked to the presence of specific cell receptors encoded by human chromosome 19) and the presence of CGB and LHB related sequences, thus confirming the localization of the structural genes for the subunits on chromosome 19.This work was supported in part by INSERM grants CRL 81 1041 and by MRC grant MT 4860  相似文献

Alterations of receptor-G-protein-regulated adenylyl cyclase activity have been suggested to represent an important alteration leading to contractile dysfunction in the failing human heart. Recent experiments suggest that the 1-adrenoceptor(1AR) density and mRNA levels are reduced, while 2-adrenoceptors and stimulatory G-proteins are unchanged (mRNA and protein level). Functional assays demonstrated that the catalyst of the adenylyl cyclase is not different between failing and nonfailing myocardium. Inhibitory G-proteins are increased (pertussis toxin substrates, protein and mRNA) and correlate to the reduced inotropic effects of -adrenoceptor agonists and of CAMP-PDE inhibitors. Gi-coupled m-cholinoceptors and A1-adrenergic receptors are unchanged in density and affinity. Stimulation of these receptors resulted in an unchanged antiadrenergic effect on force of contraction. In conclusion, a downregulation of 1-AR and an increase of Gi have been observed as signal transduction alteration in failing human myocardium. These alterations are due to alterations of gene expression in the failing heart and are related to a defective regulation of force of contraction in heart failure.  相似文献

Axillary shoot induction and plant regeneration were obtained in Plantago ovata. The optimum medium for inducing axillary shoots was Murashige & Skoog (MS) medium [5] supplemented with 4.6 M kinetin and 0.05 M NAA. Rooting of shoots was best on half-strength MS medium containing 5.0 M IBA and 0.05 M kinetin. The regenerated plants were similar to the control plants in karyotypic and phenotypic details.  相似文献

A presumed XY chromosome pair is described fromt estis squashes from the mesopelagic deep-sea fish Bathylagus wesethi, whose 2N chromosome number was determined as 34-XY. Although the metacentric X-chromosome is the largest in the entire compliment, the Y is the smallest and only acrocentric element. The positive heteropycnosis of the sex elements was not easily distinguishable in the nuclei of first meiotic prophase. Tetraploid nuclei were observed in peripheral supporting cells of the testis. Zusammenfassung Untersuchungen zum Risikoeinsatz der Brutverteidigung durch Kohlmeisen bewogen uns, den Fortpflanzungswert vt (nachFisher) abhängig von Alter, Geschlecht, Biotop und Jahresbrutenzahl zu ermitteln (Abb. 1–3). Obwohl in die Berechnungen von vt viele Annahmen verschiedener Sicherheitsgrade eingehen, stellen die vt-Kurven die hinsichtlich der verarbeiteten demographischen Größen vollständigsten und daher genauesten uns bisher bekannter dar. Wir überprüften den vt-Verlauf mit Hilfe der Netto-Reproduktionsrate R0 unter der Annahme einer langfristig stabilen Populationsgröße und fanden ihn für einen Nadelwald vorzüglich bestätigt, für Laub(misch)wälder möglicherweise überschätzt (Tab. 3). haben stets ein niedrigeres vt als , ausgenommen 1jährige ; unter diesen haben nur die Brüter ein höheres vt.Aus den vt-Kurven ermittelten wir zu den Ausfliegezeitpunkten ein Nutzen-Kosten-Verhältnis (ausfliegende Junge mt je Elter/Rest-Fortpflanzungswert vRt) abhängig von Brutbiotop, Alter, Geschlecht und Jahresbrut (Abb. 4, 5). Dieses Maß sollte dem Elter zum Prüfstein für die Stärke seines Bruteinsatzes dienen. Aus Betrag und Verlauf des Maßes werden fünf Vorhersagen zum Bruteinsatz von Kohlmeisen hergeleitet; die am wenigsten erwartete besagt, daß einbrütige Kohlmeisen im Laubwald denselben Einsatz zeigen sollten wie im Nadelwald, obwohl sie andere Fortpflanzungsraten (mt) haben.Da der Verlauf von vt wie von vRt während eines Brutzyklus nicht näher bekannt ist, schufen wir das neue Maß des Brutwertes. Er gibt den Wert der Brut für den Elter vom Nestbau bis zum Ausfliegen unter Berücksichtigung der Sterblichkeit von , und Brut sowie der geschlechtsspezifischen Brutpflegefähigkeit der Elternvögel an (Abb. 7). Der Brutwert wächst von 0 (Nestbeginn) bis 100 % (Ausfliegen) an und ist dann identisch mit mt. haben anders als bei vt, nur während eines bestimmten Teils des Brutzyklus einen höheren Brutwert als die und sollten dann mehr an Kosten für die Brut aufwenden. Es wird erörtert, weshalb weder der geschlechtsspezifische Verlauf von vt noch jener vom Brutwert den höheren Einsatz des bei der Feindabwehr vom Nest erklären.
Reproductive value and brood value of the Great Tit(Parus major)
Summary Studies of the risk taken by great tit parents while defending their brood prompted us to computeFisher's reproductive value vt as a function of age, sex, breeding habitat, and number of broods per year (Fig. 1–3). Although marred by the speculative nature of some underlying assumptions the resulting vt curves are most accurate for any animal in terms of their demographic comprehensiveness. They were checked via the net reproductive rate Ro assuming long-term stability of population numbers. The demographic ingredients on which vt was based proved to be in excellent agreement with stability (R0=1) for a larch forest but may have been less well estimated for deciduous woods (Table 3). vt values are always lower than vt values with the exception of yearling which only when breeding exceed values throughout.vt values at fledging were used to compute a cost benefit ratio of parental effort (number of fledglings mt per parent/residual reproductive value vRt) as a function of breeding habitat, age, sex and of whether a first or second brood was at stake (Fig. 4, 5). This measure is expected to provide a yardstick determining the efficiency of parental care during breeding. The cost benefit ratio thus defined is known most precisely at fledging and is thought to deviate from this value as one works backward in time up to the start of a breeding cycle when it is zero. The amount and the change in time of the cost benefit ratio lead to five non-trivial predictions about parental effort by great tits; the least obvious says that great tits producing one brood per year should invest the same effort regardless of type of habitat (deciduous vs coniferous wood) despite their reproductive rate (mt) differing greatly between habitats.Since the precise course of both vt and vRt during a breeding cycle are unknown we conceived of a new measure termed brood value. It reflects the value of the brood for each parent separately from the start of a nest up to fledging; the brood value takes into account the mortalities of the , the , and of the brood as well as the sex-specific potential for parental care (Fig. 7). Brood value increases from 0 (start of nest) to 100 % (fledging) when it becomes identical with mt. Brood value of is higher than that of only during part of the cycle as compared to vt. During this phase should invest more into their brood than . It is discussed why neither the sex-specific course of vt nor that of brood value predict a higher male effort of brood defense as actually observed.

Immature embryos of apricot (Prunus armeniaca L.) cv. Royal with a PF index of 25–100 were used to regenerate plants in vitro using two methods. In the first case, callus was initiated on MS medium with 4.5 M 2, 4-D plus 0.44 M BA and regeneration of shoots from the callus occurred on MS medium with 4.4 M BA plus 1.0 M 2, 4-D. T-cell clones expressing the T-cell receptor (Tcr) were generated from peripheral blood lymphocytes (PBLs) and from a thymus sample. In the panel of ten thymus-derived clones, four Tcr phenotypes [as defined by the reaction of monoclonal antibodies (mAbs) directed against known V and V regions] were identified. All the clones lacked expression of the V3 V region, while seven clones were V1+ . V1 was found in combination with V9 or with undefined VVregions. In addition, two other Tcr phenotypes were identified on these clones: V9+ V1 V3 and V9 V1 V3 One of the clones expressed CD4 and another was CD8positive. The remaining clones were CD4 CD8. In the panel of 76 PBL-derived, Tcr-bearing clones, five Tcr phenotypes could be identified. In contrast to the thymus-derived clones, 30% of the clones were V3+ whereas V1 was expressed by a minority of the clones only. One clone was CD4-positive and approximately 30% of the clones were CD8-positive. Four of the five mAb-defined Tcr phenotypes could be identified on both thymus and PBL-derived T-cell clones. However, biochemical analysis of the Tcrs demonstrates differences in the usage of Ct- and C2-encoded y chains by T cells derived from the thymus and PBLs. The results therefore indicate that, at the clonal level, similarities and differences exist between the Tcr repertoires expressed in the thymus and by PBLs. Furthermore, they indicate that combinatorial Tcr heterogeneity is larger than has so far been described. The receptor diversity, combined with the potential of Tcr+ cells to express CD4 or CD8, indicates that these cells are a heterogeneous population that might mediate a number of immune functions.  相似文献

Summary The structure of the triad junction was examined in thin sections of mammalian fast-twitch skeletal muscle. The aims of the experiments were twofold: first, to examine relationships between the contents of the junctional gap and the terminal cisternae that could be significant in excitation-contraction coupling and, second, to look for structures in the transverse tubules that could support a calcium buffer system. Procedures known to stabilize cytoskeletal elements were used in an attempt to retain the original structure. Feet, pillars and bridges were often seen side by side in the same junction. In one such junction, the average center-to-center spacing between four bridges was 30.9±1.7 nm and between five foot-like structures was 29.2±1.4 nm. The subunit structure of the feet could be seen in many sections. The lumen of the terminal cisternae was filled with a tetragonal network of calsequestrin which formed parallel strands near the junctional membrane, in register with the feet. The strands overlay the area occupied by rods seen in freeze-fracture replicas of terminal cisterna membrane. The contents of the transverse tubules were aggregated into bands, or tethers, which extended across the short axis of the tubule at regular intervals of about 30 nm. The tethers consisted of flattened discs, stacked across the long axis of the tubule, aligned with the junctional feet. Lanthanum staining of the tethers indicated cationic binding sites that could buffer luminal calcium ion concentration in the vicinity of the voltage sensor for contraction. It is suggested (i) that the control of calcium concentration near the voltage sensor is necessary for normal activation, (ii) that feet, pillars and bridges are different images of a spanning structure, and (iii) that the regular alignment of tethers, feet and calsequestrin is functionally significant in excitation-contraction coupling.  相似文献

Chromosomal unit fibers consisting of long, regular fibers of about 0.40 m diameter were obtained from disintegrated, isolated chromosomes of two Drosophila melanogaster cell lines. In one cell line with an essentially normal karyotype, three clearly defined size classes of 15, 13, and 11 m length were observed corresponding to the three larger chromosomes of Drosophila. In a cell line carrying an additional translocation between the two largest chromosomes a 19 m fiber derived from the translocation chromosome was observed. Direct determinations of the DNA content per m length of Drosophila unit fibers show that DNA is contracted by a factor of about 1400x in agreement with calculations based on the length of the unit fibers and the known DNA content of the individual Drosophila chromosomes. These findings support our previously proposed model for the unit fiber sub-structure of chromosomes as being derived by a hierarchy of coiling with the corresponding contraction ratios being 7 (100 Å string of nucleosomes), 5 to 6 (250–300 Å thick nucleohistone fiber), and about 40 (unit fiber), resulting in a total contraction of DNA in unit fibers in the order of 1400x.  相似文献

Nipecotic acid is one of the most potent competitive inhibitors and alternative substrates for the high-affinity -aminobutyric acid transport system in neurons, but the structural basis of this potency is unclear. Because -aminobutyrate is a highly flexible molecule in solution, it would be expected to lose rotational entropy upon binding to the transport system, a change which does not favor binding. Nipecotic acid, in contrast, is a much less flexible molecule, and one would expect the loss of conformational entropy upon binding to be smaller thus favoring the binding of nipecotic acid over -aminobutyric acid. To investigate this possibility, the thermodynamic parameters, G°, H°, and S°, were determined for the binding of -aminobutyrate and nipecotic acid to the high affinity GABA transport system in synaptosomes. In keeping with expectations, the apparent entropy change for nipecotic acid binding (112±13 J·K–1) was more favorable than the apparent entropy change for -aminobutyric acid binding (61.3±6.6 J·K–1). The results suggest that restricted conformation per se is an important contributory factor to the affinity of nipecotic acid for the high-affinity transport system for -aminobutyric acid.This work was conducted when both authors were at the Department of Chemistry, University of Maryland, College Park.Special issue dedicated to Dr. Elling Kvamme.  相似文献
