For freshwater cyanobacteria, the autofluorescence of phycocyanin is quite high while the in vivo fluorescence (IVF) yield of chlorophyll-a is relatively low, apparently because of low chlorophyll concentrations associated with photosystem II. In eucaryotic phytoplankton, even those with phycobili-protein accessory pigments (e.g. some cryptophytes), the opposite is true. Thus, an IVF ratio which relates phycocyanin to chlorophyll-a signals could be a good index of relative cyanobacterial abundance in the field. Spectrofluorometric scans of whole cells from laboratory cultures indicated that the ratio Em660 @ Ex630/Em680 @ Ex430 could be a very sensitive cyanobacterial indicator. Tandem flowthrough fluorometers were then fitted with the appropriate interference filters and their discriminatory power was evaluated with mixtures of cyanobacterial and eucaryotic phytoplankton. Although subject to many of the constraints of other IVF assays, tandem fluorometry should be particularly appropriate for real-time mapping of the relative spatial and temporal distributions of broad phytoplankton taxa in continuous vertical of horizontal profiles in lakes.  相似文献

Cyanobacterial metabolites have proven to be invaluable as tools in thedissection of signal transduction pathways in mammalian cells and some arecurrently under clinical evaluation as drug candidates. It is now also realizedthat cyanobacteria are the true biosynthetic origin of many bioactive moleculesisolated from marine invertebrates; marine invertebrates may sequestercyanobacteria through diet or by symbiosis. This review discusses thedietary-derived cyanobacterial origin of the dolastatins, potent cytotoxiccompounds, originally isolated from the Indian Ocean sea hare,Dolabella auricularia. A discussion on the dietarydissemination of cyanobacterial metabolites through the marine food chain isalso presented. Reference to the metabolites isolated fromDysidea sponges is given to illustrate their origin fromsymbiotic cyanobacteria associated with this organism.  相似文献

Cyanobacterial biovolume is used as a guide to the public health risk from these organisms for users of potable and recreational waters. Most routine surveillance programs preserve phytoplankton samples before analysis. We tested the effect of Lugol's Iodine, a common preservative, on the cell biovolume of four common freshwater cyanobacteria, Microcystis aeruginosa, Anabaena circinalis, Cylindrospermopsis raciborskii and Aphanocapsa incerta. Linear dimensions and cell area were measured with an image analyser. All four species shrank after preservation. The magnitude of shrinkage varied with species and preservation time but was not affected by Lugol's concentration. The maximum shrinkage in each species was a 30–40% reduction compared to the live cell biovolume. These results suggest shrinkage can be a greater source of uncertainty in estimating the biovolume of toxigenic cyanobacteria in aquatic environments than natural variability in the cell dimensions, instrument precision or cell counting. Standardised cyanobacterial biovolume lists based on agreed geometric shapes and formulae would improve the value of this information for public health risk assessment.  相似文献

Biomanipulation, through the reduction of fish abundance resulting in an increase of large filter feeders and a stronger top-down control on algae, is commonly used as a lake restoration tool in eutrophic lakes. However, cyanobacteria, often found in eutrophic ponds, can influence the grazing capacity of filter feeding zooplankton. We performed grazing experiments in hypertrophic Lake Blankaart during two consecutive summers (1998, with and 1999, without cyanobacteria) to elucidate the influence of cyanobacteria on the grazing pressure of zooplankton communities. We compared the grazing pressure of the natural macrozooplankton community (mainly small to medium-sized cladocerans and copepods) with that of large Daphnia magna on the natural bacterioplankton and phytoplankton prey communities. Our results showed that in the absence of cyanobacteria, Daphnia magna grazing pressure on bacteria was higher compared to the grazing pressure of the natural zooplankton community. However, Daphnia grazing rates on phytoplankton were not significantly different compared to the grazing rates of the natural zooplankton community. When cyanobacteria were abundant, grazing pressure of Daphnia magnaseemed to be inhibited, and the grazing pressure on bacteria and phytoplankton was similar to that of the natural macrozooplankton community. Our results suggest that biomanipulation may not always result in a more effective top-down control of the algal biomass.  相似文献

Cyanobacteria Spirulina platensis and Nostoc linckia were grown in the presence of 5 mM and 50 mM glucose or 5 mM mannose, non-metabolisable glucose analogue that effectively triggers the repression of photosynthesis. Glucose evoked active cyanobacterial growth but chlorophyll (Chl) content decreased to some extent and porphyrins were excreted. The content of monogalactosyldiacylglycerol decreased in glucose-grown cyanobacteria and that of phosphatidylglycerol increased substantially. Mannose inhibited cyanobacteria growth as well as Chl synthesis, however, phosphatidylglycerol contents were higher than in respective control samples. In cyanobacterial cells glucose may not only inhibit photosynthetic processes, but also cause structural transformations of membranes which may be necessary for the activity of respiratory electron transport chain components under heterotrophic conditions.  相似文献

Hydrogenases are important enzymes in the energy metabolism of microorganisms. Therefore, they are widespread in prokaryotes. We analyzed the occurrence of hydrogenases in cyanobacteria and deduced a FeFe-hydrogenase in three different heliobacterial strains. This allowed the first phylogenetic analysis of the hydrogenases of all five major groups of photosynthetic bacteria (heliobacteria, green nonsulfur bacteria, green sulfur bacteria, photosynthetic proteobacteria, and cyanobacteria). In the case of both hydrogenases found in cyanobacteria (uptake and bidirectional), the green nonsulfur bacterium Chloroflexus aurantiacus was found to be the closest ancestor. Apart from a close relation between the archaebacterial and the green sulfur bacterial sulfhydrogenase, we could not find any evidence for horizontal gene transfer. Therefore, it would be most parsimonious if a Chloroflexus-like bacterium was the ancestor of Chloroflexus aurantiacus and cyanobacteria. After having transmitted both hydrogenase genes vertically to the different cyanobacterial species, either no, one, or both enzymes were lost, thus producing the current distribution. Our data and the available data from the literature on the occurrence of cyanobacterial hydrogenases show that the cyanobacterial uptake hydrogenase is strictly linked to the occurrence of the nitrogenase. Nevertheless, we did identify a nitrogen-fixing Synechococcus strain without an uptake hydrogenase. Since we could not find genes of a FeFe-hydrogenase in any of the tested cyanobacteria, although strains performing anoxygenic photosynthesis were also included in the analysis, a cyanobacterial origin of the contemporary FeFe-hydrogenase of algal plastids seems unlikely. DNA was prepared from cyanobacteria freshly isolated from coralloid roots of natural populations of five cycad species: Ceratozamia mexicana mexicana (Mexico), C. mexicana robusta (Mexico), Dioon spinulosum (Mexico), Zamia furfuraceae (Mexico) and Z. skinneri (Costa Rica). Using the Southern blot technique and cloned Anabaena PCC 7120 nifK and glnA genes as probes, restriction fragment length polymorphisms of these cyanobacterial symbionts were compared. The five cyanobacterial preparations showed differences in the sizes of their DNA fragments hybridizing with both probes, indicating that different cyanobacterial species and/or strains were in the symbiotic associations. On the other hand, a similar comparison of cyanobacteria freshly collected from a single Encephalartos altensteinii coralloid root and from three independently subcultured isolates from the same coralloid root revealed that these were likely to be one and the same organism. Moreover, the complexity of restriction patterns shows that a mixture of Nostoc strains can associate with a single cycad species although a single cyanobacterial strain can predominate in the root of a single cycad plant. A submersible in situ spectrofluorometer, which permits the differentiation of four algal groups (green algae, diatoms, cryptophytes and cyanobacteria), was used for phytoplankton monitoring in five reservoirs with varying levels of eutrophication and composition of their phytoplankton communities. Data obtained in situ were compared to standard laboratory methods for phytoplankton quantification; concentration of chlorophyll a and microscopy analysis. A high correlation (r = 0.95, n = 96) between chlorophyll a levels using different methods was found in all types of phytoplankton community. Taxonomic analyses and cell counts were closely related to the ratio of algal classes measured by the in situ spectrofluorometer. The submersible device used in the study measures in a continuous mode, which is advantageous in comparison with discrete sampling. This method appears to be a good tool for water quality management and can be used in the detection of natural horizontal and vertical variability in phytoplankton communities or for the early detection of cyanobacterial blooms. The device used in this study is recommended as a screening tool that enables more effective sampling that can be focused on the localities and depths where changes in phytoplankton composition occur.  相似文献

Spatial and temporal variation in phytoplankton community structure within a large flood-control reservoir (Sardis Reservoir, MS, USA) was investigated in relation to variation in physicochemical properties, location within the reservoir, hydraulic residence time (HRT), nutrient concentrations, temperature, and light conditions over a 14-month period. During periods of short HRT, phytoplankton communities throughout the reservoir were homogeneous in biomass, composition, and production. With a gradual increase in HRT from spring to summer, spatially heterogeneous phytoplankton communities developed along the longitudinal axis of the reservoir. During this period of longer HRT, diatoms and chlorophytes were a larger proportion of total phytoplankton biomass at shallow and more turbid locations near the head of the reservoir, whereas cyanobacteria were a larger proportion of the community at deeper and less turbid locations closer to the outflow. Seasonal succession of the phytoplankton community was represented by high abundance of diatoms in spring, increasing biomass of cyanobacteria through summer, and a secondary bloom of diatoms in fall. Species of Cyclotella, Asterionella, Nitzschia, and Ankistrodesmus were among the first colonizers in the early growing season, closely followed by Aulacoseira, whereas species of Staurastrum and Tetraedron appeared later in the spring. Species of Synedra, Crucigenia, Selenastrum, Scenedesmus, and Merismopedia occurred throughout the sampling period. As the diatoms started to decrease during mid-spring, cryptophytes increased, prior to dominance of species of Pseudanabaena in summer. Reservoir management of HRT, in combination with spatial variation in reservoir morphology and seasonal variation in temperature and riverine nutrient inputs, creates seasonally variable yet distinct spatial patterns in phytoplankton community biomass, composition, and production. Two strains of Myxococcus xanthus, and a strain of Myxococcus fulvus were compared with respect to their ability to entrap and lyse trichomes of the cyanobacterium Phormidium luridum var. olivaceae. All of these isolates form colonial aggregates and spherules in either axenic culture with a tryptone-salts medium or in a mixed culture with viable cyanobacterial cells as the sole source of nutrients. Light microscopy showed evidence of swarming activity on the surface of all three myxococci with the accompanying formation of fruiting structures. Extended incubation of mixed cultures showed the myxococci to be capable of long-term control of the cyanobacterial population with predator-prey population cycling occurring on average every 9 days. Serial transfer of mixed cultures into either fresh autotrophic medium or cyanobacterial cultures of 107 per ml showed the persistence of predatory activity. Myxococcal densities were shown to return repeatedly to initial virulent levels. Predator inoculum levels could be reduced to 50 cells per 100 ml in a cyanobacterial culture of 107 per ml. These in vitro data enhance the potential of the myxococcus predatory colony as a biological control agent for in situ cyanobacteria.  相似文献

A Myxococcus xanthus isolate from a farm drainage ditch, designated strain PCO2, is capable of rapidly inducing lysis of both agar and liquid-grown cultures of the cyanobacterium, Phormidium luridum, var. olivacea. Microscopic studies of the predator-prey interaction demonstrate that lysis of the cyanobacterium occurs within clumps and spherules formed by the cells of M. xanthus PCO2. In the earliest stage, one sees the formation of irregular microclumps of bacteria and cyanobacterial filaments. As these clumps mature, colonies 1 to 6 mm in diameter develops. The center of these densely green colonies contains cyanohacteria in various stages of degradation, while the periphery is almost exclusively a tightly woven mass of myxobacterial cells. Electron microscopy shows that long extrusions from the outer membrane of the M. xanthus PCO2 cells are involved in the formation both of initial clumps and of mature colonial spherules. These extrusions appear to efficiently entangle the cyanobacterial filaments in the culture environment. Predator-to-prey ratios of 1/10, 1/100 and 1/1,000 have resulted in cyanobacterial lysis. Because the entrapment and lysis of P. luridum filaments by M. xanthus PCO2 appears to be independent of any other heterotrophic nutritional requirement, as well as of environmental agitation, this system has potential as a biological control technique for undesirable aquatic cyanobacteria.Abbreviations TEM transmission electron microscopy - SEM scanning electron microscopy - AB algae broth - ABT algae broth plus 0.2% tryptone  相似文献

Nitrogen-fixing cyanobacteria inhabit the zone between the inner and outer cortex of cycad coralloid roots. In the growing tip of such roots the cyanobacterial heterocyst frequency, nitrogenase activity (C2H2-reduction) and glutamine synthetase activity (both transferase and biosynthetic) were comparable to those found in freeliving cyanobacteria. The relative level of glutamine synthetase protein and its pattern of cellular/subcellular localization in heterocysts and vegetative cells were also similar to those of free-living cyanobacteria. However, there was a progressive decline in nitrogenase activity along the coralloid root with maximum reduction occurring in the regions farthest from the growing tip. A similar but less pronounced pattern was observed for glutamine synthetase activity. Distribution of glutamine synthetase protein in cyanobacteria in the first 2–3 mm of the root tip indicated a slight decrease in the heterocysts and vegetative cells. However, the overall level of cyanobacterial glutamine synthetase protein did not change because of a drastic increase in the numbers of heterocysts, which contain a proportionally higher level of glutamine synthetase than the vegetative cells.Abbreviation GS glutamine synthetase  相似文献

Cyanobacteria (photoautotrophic prokariota) have potential for the control of pathogenic bacteria and fungi. The effect of intra and extracellular products from cyanobacterial strains on the growth of fungi isolated from “wood blue stain,” was tested. Extracellular products were obtained by concentration and sterilization of the culture medium where cyanobacteria were grown. Cyanobacterial substances promoted or inhibited fungal growth according to the fungal and cyanobacterial strains tested. Extracellular products from Nostoc muscorum 79a and the methanolic extract from Microchaete tenera 84b biomass inhibited growth of Sphaeropsis sapinea 2157 (64.7 and 775.6%, respectively). Extracellular products of Nostoc piscinale 59 and biomass methanolic extract from N. muscorum 79a produced the highest growth promotion of Trichoderma boningii 452 (105.0%) and T. viride 993 (136.7%). Extracellular products of the heterotrophic lactic acid bacterium Streptococcus termophilus were also tested and strongly inhibited (64–92%) all the fungal strains. The tested fungi have different sensitivity to the bioactive substances present in the biomass and/or the culture medium of the studied cyanobacteria and lactic acid bacterium. N. muscorum 79a, M. tenera 84b, and S. termophilus have potential to control the wood blue stain fungi by a friendly environmental alternative.  相似文献

A phytoplankton-lytic (PL) bacterium, Bacillus cereus, capable of lysing the bloom-forming cyanobacterium Aphanizomenon flos-aquae was isolated from Lake Dianchi of Yunnan province, China. This bacterium showed lytic activities against a wide range of cyanobacteria/algae, including A. flos-aquae, Microcystis viridis, Microcystis wesenbergi, Microcystis aeruginosa, Chlorella ellipsoidea, Oscillatoria tenuis, Nostoc punctiforme, Anabaena flos-aquae, Spirulina maxima, and Selenastrum capricornutum. Chlorophyll a contents, phycocyanin contents, and photosynthetic activities of the A. flos-aquae decreased evidently in an infected culture for a period. Bacterium B. cereus attacked rapidly A. flos-aquae cells by cell-to-cell contact mechanism. It was shown that the lysis of A. flos-aquae began with the breach of the cyanobacterial cell wall, and the cyanobacterial cell appeared abnormal in the presence of the PL bacterium. Moreover, transmission electron microscope examinations revealed that a close contact between the bacterium and the cyanobacterium was necessary for lysis. Some slime extrusions produced from B. cereus assisted the bacterial cells to be in close association with and lyse the cyanobacterial cells. These findings suggested that this bacterium could play an important role in controlling the Aphanizomenon blooms in freshwaters.  相似文献

The control of nuisance species of cyanobacteria in reservoirs is a critical issue for the international water industry, as these organisms can produce toxins and compounds that taint potable water with unpleasant tastes and odours. To assist with effective management of cyanobacterial growth, numerical models that are either site specific or universally applicable can be employed. An artificially destratified reservoir was modelled with the coupled hydrodynamic-ecological numerical model DYRESM-CAEDYM. The validation site was Myponga Reservoir, South Australia, a highly managed drinking water supply reservoir. Chemical dosing (CuSO4) and artificial mixing via an aerator and two raft-mounted mechanical surface mixers (hereafter referred to as surface mixers) are used at Myponga to manage the growth of the scum-forming cyanobacterium Anabaena circinalis. The dominant phytoplankton community was adequately modelled, and combinations of the various management options were simulated whereupon informed operational strategies could be implemented. Without any intervention, permanent stratification would occur and the growth of Anabaena circinalis would peak above 3 μ g L−1, producing geosmin that would exceed the taste and odour threshold (10 ng L−1); with the individual use of the aerator or surface mixers, growth of Anabaena circinalis was significantly reduced to below 1 μ g L−1.  相似文献

The sizes of endonuclease digestion fragments of DNA from cyanobacteria in symbiotic association with Azolla caroliniana or Anthoceros punctatus, or in free-living culture, were compared by Southern hybridization using cloned nitrogenase (nif) genes from Anabaena sp. PCC 7120 as probes. The restriction fragment pattern produced by cyanobacteria isolated from A. caroliniana by culture through symbiotic association with Anthoceros differed from that of the major symbiotic cyanobacterium freshly separated from A. caroliniana. The results indicate that minor cyanobacterial symbionts occur in association with Azolla and that the dominant symbiont was not cultured in the free-living state. Both the absence of hybridization to an xisA gene probe and the mapping of restriction fragments indicated a contiguous nifHDK organization in all cells of the symbiont in association with Azolla. On the other hand, in the cultured isolate from Azolla and in Nostoc sp. 7801, the nifD and nifK genes are nominally separated by an interval of unknown length, compatible with the interruption of the nifHDK operon by a DNA element as observed in Anabaena sp. PCC 7120. In the above cultured strains, restriction fragments consistent with a contiguous nifHDK operon were also present at varying hybridization intensities, especially in Nostoc sp. 7801 grown in association with Anthoceros, presumably due to gene rearrangement in a fraction of the cells.Non-standard abbreviations bp base pairs - kb kilobase pairs - kd kilodaltons  相似文献

Cyanobacteria possess thylakoid membranes that differ in their protein composition from the cytoplasmic membrane. To study possible pathways of protein targeting to these membranes, we have investigated whether or not cyanobacteria have a homologue or homologues of the signal recognition particle-like chaperone Ffh. We have amplified a fragment of ffh by polymerase chain reaction and established that ffh is present as a single copy in the genomes of three cyanobacterial species. We have cloned and sequenced ffh from Synechococcus sp. PCC7942 and predict that Ffh functions as a ribonucleoprotein in cyanobacteria and chloroplasts.  相似文献

Growth and potential photosynthetic activity of phytoplankton passed through intestine of crucian carp (Carassius auratus) from a small Siberian reservoir were compared with those of phytoplankton taken the directly from the reservoir. The dominant phytoplankton species in the reservoir, Microcystis aeruginosa, showed a significant increase of growth after the passage. Subdominant Planktothrix agardhii also showed an increase in growth rate, while subdominants Anabaena flos-aquae and Aphanizomenon flos-aquae were not stimulated by the gut passage.  相似文献

Loch Leven is a shallow, eutrophic lake in the Scottish lowlands that is famous for its brown trout (Salmo trutta L.) fishery. Studies of planktonic rotifer populations began here in January 1977. Since then, samples have been collected and analysed at more or less weekly intervals. Additional information on the composition and abundance of phytoplankton and crustacean zooplankton species, and on a variety of physical and chemical determinants, has been recorded on each sampling occasion.Long-term datasets, such as that described above, are invaluable for identifying interactions between components of the plankton that only appear for short periods each year, as these interactions would probably be overlooked in data spanning a shorter period of time. This study uses the long-term data from Loch Leven to examine the food and temperature requirements of the summer rotifer species Trichocerca pusilla (Lauterborn). The results suggest that T. pusilla prefers water temperatures above 12 °C and that it feeds, primarily, on the filamentous diatom Aulacoseira spp. During the summer months, its abundance was closely related to the availability of this diatom. When filaments of Aulacoseira spp. were abundant, rotifer densities reached 1000–3000 ind. l–1 and when they were scarce (e.g. 1980, 1997 and 1998) T. pusilla densities also remained low (i.e. less than 100 ind. l–1). The reason for the success or failure of Aulacoseira during the summer months each year is unclear but, in general, its abundance was related to the availability of dissolved silica in the water.  相似文献

In order to determine the nearly complete 16S rRNA gene sequences of cyanobacteria originating from nonaxenic cultures, a cyanobacterium-specific oligonucleotide probe was developed to distinguish polymerase chain reaction (PCR) products of the cyanobacterial rRNA operons from those resulting from amplification of contaminating bacteria. Using this screening method the 16S rRNA genes of four nonaxenic filamentous cyanobacterial strains belonging to the generaLeptolyngbya andOscillatoria were cloned and sequenced. For the genusLeptolyngbya, the 16S rRNA sequence of the axenic strain PCC 73110 was also determined. Phylogenetic trees were constructed based on complete and partial sequences. The results show that the strainsLeptolyngbya foveolarum Komárek 1964/112,Leptolyngbya sp. VRUC 135 Albertano 1985/1, andLeptolyngbya boryanum PCC 73110 belong to the same cluster. StrainOscillatoria cf.corallinae SAG 8.92, which contains the rare photosynthetic pigment CU-phycoerythrin, is not closely related to other CU-phycoerythrin-containing cyanobacteria.Oscillatoria agardhii CYA 18, which is a representative of planktonicOscillatoria species that form toxic blooms in Norwegian inland waters, has no close relatives in the tree.  相似文献
