Summary Cytoplasmic vacuoles and bodies in the osteoclast (rat) were studied by electron microscopy. The vacuole-like structures (0.03–5 in diameter) may be classed as a) vacuoles b) coated vacuoles and c) invaginations. The cytoplasmic bodies vary in size from 0.02–3 in diameter and these may similarly be classed as a) light cytoplasmic bodies, b) dense cytoplasmic bodies, c) coated cytoplasmic bodies and d) cytoplasmic bodies containing inclusions. Both the cytoplasmic vacuoles and the bodies are limited by a triple layered membrane of about 91 Å in thickness. Their relationship to the lysosomal system and the role of this system in the osteoclast is discussed.This research was supported by the Danish Research Council. Summary New methods of fixation and embedding have revealed in plants infected withPetunia ringspot some structural features not described before. These include the X-bodies, 80 per cent of which are formed by tubular elements which are responsible for the positive staining specific for Golgi apparatus. The tubular elements are morphologically similar to agranular endoplasmic reticulum as described in some animal cells. The rest of the inclusion is formed by normal cytoplasmic elements in which are embedded long rod-shaped tubules 600 Å wide and more than 7,000 Å long, cross sections of which are formed by 10 subunits. These subunits are arranged in a helical way to form the large tubules. These subunits are probably the actual virus particles, which would be icosahedral and would form tubular crystals. Icosahedral virus particles would also form true crystalline inclusions.It is not known what the role of the agranular endoplasmic reticulum and of some dense osmiophilic bodies found in it may be in the multiplication of the virus. However, these components induced by the virus infection probably result in the manufacture of some proteins or other substances necessary for virus multiplication.  相似文献

Summary The effects of acute injections of synthetic opiate peptides into the lateral cerebral ventricle of young adult male rats on cells of the intermediate lobe of the pituitary were studied. Met-enkephalin (100/g) injected into anesthetized rats, or 20 g beta-endorphin administered via a previously implanted cannula to unanesthetized animals, will lead to cell degranulation and often to expanded Golgi zones and prominent regions of rough endoplasmic reticulum in secretory cells when tissue is fixed 45–60 min after peptide administration. Treatment of animals with the opiate antagonist naloxone hydrochloride prior to enkephalin injection appeared to prevent the cellular changes elicited with peptide alone. Observations suggest that opiate peptides administered to the cerebrospinal fluid may stimulate release of pro-opiomelanocortin-peptide from pituitary cells.  相似文献

Summary In tip-growingChara rhizoids, the in-vivo saltatory movements of Golgi vesicles were recorded. The movements in radial direction back and forth between the ER aggregate and the plasma membrane occurred three times more often than movements passing the ER aggregate tangentially. The mean velocity of the class of Golgi vesicles observed (0.4–1 m in diameter) was approx. 0.3 m/s. Higher speed of 1–1.5 m/s occurred only in radial directions. Summary Root tips of radish,Raphanus sativus, were fixed in glutaraldehyde followed by osmium tetroxide. The fine structure of young root hairs, not exceeding about 130, in length, was studied to relate their apical growth pattern to their cytoplasmic organization. The cytoplasm in the terminal 3–5 it of the root hair is characterized by an electron dense matrix in which lie numerous smooth-surfaced vesicles, large irregularly-shaped fibrous inclusions, and clusters of ribosomes. Other organelles are largely or entirely excluded from this region. Farther than about 5, from the tip, the hair cytoplasm is filled with plastids, rough endoplasmic reticulum, mitochondria, and dictyosomes. The latter produce smooth vesicles similar in size and morphology to those present in the apical dome. Vesicles of a different kind appear in the peripheral cytoplasm along the entire length of the hair. These vesicles possess an alveolate or chambered coat about 20 m thick and have a diameter of about 85 m, including coat. They originate by evagination from the large, smooth-surfaced vesicles in the vicinity of dictyosomes. It is suggested that proteins and carbohydrates are concentrated in the dictyosomes and then segregated in the smooth vesicles released from the dictyosome cisternae. The coated vesicles which bud from the smooth vesicles may serve to isolate the proteins and transport them to the hair surface for participation in wall synthesis. The smooth vesicles are believed to convey carbohydrates to the region of active wall extension at the hair apex.This work was supported in part by grant GM-10493 from the National Institutes of Health. Summary Subcellular compartments comprising the endomembrane system in filamentous fungi are poorly characterized with most showing significant morphological differences from eukaryotic cells. For example, many filamentous fungi lack stacked Golgi-body cisternae, but contain Golgi equivalents — single cisternae or tubules which appear to serve the same functions. To help identify fungal endomembrane compartments and interrelationships between them we used a pharmacological agent, brefeldin A, known to affect specific endomembrane organelles in other organisms, most prominently the Golgi apparatus. At 10 g/ml brefeldin A, radial hyphal growth of the rice blast pathogenMagnaporthe grisea on solid agar medium was reduced by 96% over an initial 48 h, but recovered and was reduced by only 20% over a subsequent 72 h exposure. Light microscopic examination of individual living hyphae showed that apical elongation generally halted within 1 min after exposure to brefeldin A. Acute effects of 14 g/ml brefeldin A were characterized ultrasiructurally in cells prepared by freeze substitution. These included the appearance of two types of cisternae with unusual morphology, associated with ca. 45 nm diameter vesicles, as well as the unexpected persistence and increase in complexity of the Golgi equivalents. Also observed were (1) reduced numbers of apicale vesicles and disruption of Spitzenkörper organization, (2) apical clusters of 30–35 nm diameter microvesicles and associated tubular arrays, (3) dilation of rough endoplasmic reticulum, (4) packets of membrane-bounded electron-opaque cell wall inclusions, and (5) altered morphology of some vacuolar compartments. The distribution of concanavalin A binding sites, previously mapped to particular endomembrane compartments, was documented to aid the interpretation of these results. Summary The pars distalis of the adenohypophysis of normal (78), thyroideotomized (6), adrenalectomized (6), and castrated (14) White-crowned Sparrows were observed with the electron microscope. Six types of glandular cells were identified and the ultrastructural characteristics of each have been described. To each has been assigned tentatively an endocrine function.STH cells are characterized by the presence of large, dense secretory granules ranging from 220–280 m, a poorly developed endoplasmic reticulum, and a fragmented Golgi apparatus; they occur only in the caudal lobe. They show no remarkable changes after adrenalectomy, castration, and thyroidectomy.Prolactin cells, whose identity is suggested by their responses to photostimulation and surgical experiments, are characterized by large, polymorphic, dense secretory granules; they have been found mainly in the cephalic lobe.ACTH cells, whose function is confirmed by their cytological responses to adrenalectomy, have a peculiar type of secretory granule (220 m) with high and low phases of electron density. They occur exclusively in the cephalic lobe and are transformed, after adrenalectomy to large, vacuolated adrenalectomy cells.TSH cells are so designated by their response to thyroidectomy. After thyroidectomy, they lose their specific fine secretory granules and are transformed into large, vacuolated thyroidectomy cells. We have found TSH cells and thyroidectomy cells only in the cephalic lobe.Two types considered to be gonadotropic cells from their responses to gonadectomy, occur in both the cephalic and caudal lobes. One of them contains spherical, dense secretory granules (180–220 m), prominent rough endoplasmic reticulum and a well developed Golgi apparatus; the other type contains dense secretory granules of variable size (150–350 m), a less extensively developed Golgi apparatus, and sac-like endoplasmic reticulum. Both types of gonadotropic cells show extreme enlargement and vacuolization after castration. However, they retain differences in appearance in the structure of cytoplasmic organelles and vacuolization.Dedicated to Professor Dr. H. Summary The ultrastructure of corpora lutea obtained during the preimplantation, implantation and early postimplantation periods has been studied in 20 western spotted skunks. Fine structure of granulosa lutein cells was correlated with progesterone levels. The corpus luteum of the prolonged (7 month) preimplantation period contained undifferentiated small granulosa cells and differentiated large granulosa lutein cells. The former ranged in size between 12 and 20 and the latter between 20 and 45 . The ratio of small and large cells was about equal in an animal 2 days prior to nidation whereas only few small cells and numerous large cells were observed in an animal estimated to be 8 to 12 hours from nidation. Occasionally small cells were observed amidst large ones during the 24 hour nidation period, i.e. adhesion of trophoblast with the luminal uterine epithelium, but small cells were absent in animals after this period. Small cells had some smooth and rough endoplasmic reticulum, rod-shaped mitochondria with platelike cristae, small Golgi complex, and relatively smooth plasma membranes. Large lutein cells had abundant smooth endoplasmic reticulum, membranous whorls of smooth endoplasmic reticulum, usually round mitochondria with tubular and lamellar cristae, a well developed Golgi complex, variable amounts of lipid droplets, and highly plicated and ruffled plasma membranes. Peripheral plasma progesterone levels during the prolonged preimplantation period ranged between 1.1 and 7.9 ng/ml, but during implantation it was between 8 and 16.6 ng/ml. It is suggested that plasma progesterone levels fluctuate during the time of implantation and should not be regarded as a basis to predict actual nidation in the western spotted skunk.This research was supported in part by Grant Number HD06556 from the National Institute of Child Health and Human Development.  相似文献

Summary At the onset of previtellogenesis, the oocytes of Acerentomon gallicum begin to grow and increase their content of organelles. The nuage material in the oocytes at first increases in amount, then declines gradually to disappear completely from oocytes that have attained 50 m in diameter. During the growth period, new dictyosomes, mitochondria and ribosomes appear. Numerous vesicles become detached from the cell membrane and subsequently fuse into cisternae, thus forming extensive complexes of the rough endoplasmic reticulum. In oocytes with diameters between 50 and 100 m the Golgi complexes show a considerable increase in activity, and many lamellar bodies arise from mitochondria that have undergone specific transformation.This work was supported by Government Problem Grant II-1.3.13  相似文献

Summary The moderator band in the heart of the ox and goat contains bundles of Purkinje fibers and nerve fibers separated by connective tissue. The axons are mostly unmyelinated and embedded in the cytoplasm of Schwann cells.Small bundles of axons run close to the Purkinje fibers. The axons dilate into varicosities 0.5 to 1.6 in diameter (mean 0.95 ), containing three types of vesicles: 1) agranular vesicles with a diameter of 400–500 Å, 2) large dense-cored vesicles with a diameter of 800–1200 Å, 3) small dense-cored vesicles with a diameter of 500 Å. Most varicosities contain agranular vesicles together with a few large dense-cored vesicles.The gap between the varicosities and the nearest Purkinje fiber is unusually wide and normally varies between 0.3 and 0.8 . No intimate nerve-Purkinje fiber contacts, with a cleft of 200 Å, were observed.  相似文献

The ultrastructural organization of actively secreting barley (Hordeum vulgare L. cv. Himalaya) aleurone cells was examined using ultrarapid-freezing (<-10 000°C s-1) followed by freeze-fracture and freeze-substitution. Our analysis indicates that much of the evidence supporting a direct pathway from the endoplasmic reticulum (ER) to the plasma membrane (i.e. bypassing the Golgi apparatus) for the secretion of -amylase (EC 3.2.1.1) may not be valid. Cryofixed ER cisternae show no sign of vesiculation during active -amylase secretion in gibberellic acid (GA3)-treated cells. At the same time, Golgi complexes are abundant and numerous small vesicles are associated with the edges of the cisternae. Vesicles appear to be involved in the delivery of secretory products to the plasma membrane since depressions containing excess membrane material appear there. Treatment with GA3 also induces changes in the composition of Golgi membranes; most notably, the density of intramembrane particles increases from 2700 m-2 to 3800 m-2 because of an increase of particles in the 3–8.5-nm size range. A slight decrease in 9–11-nm particles also occurs. These changes in membrane structure appear to occur as the Golgi complex becomes committed to the processing and packaging of secretory proteins. We suggest that secretory proteins in this tissue are synthesized in the abundant rough ER, packaged in the Golgi apparatus, and transported to the plasma membrane via Golgi-derived secretory vesicles. Mobilization of reserves is also accompanied by dynamic membrane events. Our micrographs show that the surface monolayer of the lipid bodies fuses with the outer leaflet of the bilayer of protein-body membranes during the mobilization of lipid reserves. Summary Freeze-fracture studies were conducted on the membranes of normal cockroach hemocytes. The plasmalemma is asymmetric with the A fracture face containing 80–100 Å membrane intercalated particles at a concentration of 2500/2. The B fracture face contains 120–150 Å particles with a relatively low density (800/2). The nuclear envelope displays an asymmetry with the A fracture face containing 1500 particles/2 and the B face containing 300/ 2. No significant particle size differences were observed in nuclear envelope fracture faces. Two types of symmetric membranes were also found in these cells. Both A and B fracture faces of the membrane surrounding the numerous cytoplasmic inclusion bodies contain particle sizes and concentrations similar to the B face of the plasmalemma. A second type of symmetry was observed in cells apparently engaged in exocytosis. Vesicles (0.1 D) from this process were completely particle free on both fracture faces. Summary Human and non-human primate placental villi (near term) were incubated in an alkaline 3,3 diamino benzidine (DAB) medium to identify the distribution of microperoxisomes in these tissues. Catalase-positive microperoxisomes were present in small numbers in cytotrophoblast, but were virtually absent from syncytiotrophoblast. Microperoxisomes were roughly circular in shape and about 0.2–0.3 m in diameter. They always had a close association with an element of agranular endoplasmic reticulum. The function of microperoxisomes in placenta, as in many other tissues, is uncertain. What-ever their function, they are presumably more important in cytotrophoblast metabolism than in syncytium.  相似文献

Summary The anterior pituitaries from a series of female White-crowned Sparrows,Zonotrichia leucophrys pugetensis, in the periods of oviposition, incubation, and brooding under natural conditions, have been investigated by electron microscopy. The prolactin cells occur in cephalic lobe and are characterized by large (ca. 300–600 m), polymorphic electron-dense secretory granules and an extremely well developed, lamellated endoplasmic reticulum. During incubation and brooding it is only these prolactin cells that are in an activated secretory phase, as indicated by increase in number and size, extremely well developed endoplasmic reticulum, decrease in number of mature secretory granules, and by active formation of granules in the enlarged Golgi apparatus. In the late stages of brooding, and post-breeding, the prolactin cells regress with involution of the endoplasmic reticulum and Golgi apparatus, reaccumulation of granules, and the appearance of lysosomes.The gonadotropes of both the cephalic and caudal lobes undergo progressive morphologic changes through the course of the breeding period. They are numerous and active in the ovulating bird. Summary In meristematic cells of the gemma of Riella helicophylla and in young bud cells from the protonema of Funaria hygrometrica the cell plate is formed by fusion of small vesicles originating from the Golgi apparatus. These spherical vesicles of about 0.1 m diameter have an electron dense centre, probably consisting of pectic substances or their precursors. The endoplasmic reticulum producing multivesicular bodies participate in cell plate formation too. Another cytoplasmic component forming the cell plate are coated vesicles, the origin of which is the Golgi apparatus and perhaps also the endoplasmic reticulum. In view of these observations the question of whether the endoplasmic reticulum or the Golgi apparatus forms the cell plate must be answered in this way: both endoplasmic reticulum and Golgi apparatus supply material for growth of the cell plate. Multivesicular bodies, coated vesicles and other small vesicles of unknown nature participate in the formation of the primary wall.

This report describes the newly identified Sarcocystis inghami n. sp. from the skeletal muscles of opossums (Mammalia: Didelphidae) that were collected from south central Michigan (42° 43-42° 79N, 84° 18-84°mathtype="display">6'W), USA. The new species is distinguished from all species described from North and South American opossums by the distinctive morphology of the villar protrusions on the cyst wall. Sarcocysts of S. inghami are microscopic, up to 700 m long and 110 m wide. The sarcocyst wall is up to 7 m thick, with long, stalked protrusions which average 5.5 × 1.2 m. These are constricted at the base, expanded laterally, rounded off distally and occasionally bifid. The villar protrusions have numerous microtubules without electron–dense bodies that extend from the tips into the granular layer. Bradyzoites are 10.7 × 4.3 (8––12 × 4––5) m. This is the second species of Sarcocystis sarcocyst described from the Virginia opossum in North America.  相似文献

Summary Two new species of Isospora (Apicomplexa: Eimeriidae) are described from the faeces of passeriform birds of South America. I. cyanocoracis n. sp. is described from Cyanocorax chrysops (Passeriformes: Corvidae) and I. paroariae n. sp. from Paroaria coronata (Passeriformes: Emberizidae). I. cyanocoracis oocysts are spherical or subspherical, 28.7×26.8 m (25.0–30.5×24.5–29.0), with bi-layered wall about 2.0 m thick. Micropyle and oocyst residuum are absent; large polar granule present. Sporocysts are ovoid, 19.3×11.4 m (17.0–21.0×10.5–12.2), with smooth, single-layered wall about 0.8 m thick. Stieda and substiedal bodies and sporocyst residuum are present. Sporozoites 12.2×4.2 m (10.5–15.0×3.5–4.5), possess spherical anterior and posterior refractile bodies. I. paroariae oocysts are spherical or subspherical, 22.3×21.4 m (19.5–25.5×18.5–24.0), and have bi-layered wall about 1.8 m thick. Micropyle, polar granule, and oocyst residuum are absent. Sporocysts ovoid, 15.2×10.0 m (14.0–16.5×8.0–11.5), possess smooth, single-layered wall about 0.7 m thick. Stieda and substiedal bodies and sporocyst residuum are present. The distribution of cysteine oxidase (CO) and cysteine sulfinate decarboxylase (CSD) was examined in 12 regions of the rat central nervous system (CNS). The distribution of CO activity, expressed as mol of cysteine sulfinate formed per h per g, was the following: hypothalamus, superior and inferior colliculi, 94–99 mol/h/g; olfactory bulbs, cerebral cortex, striatum, and hippocampus, 44–51 mol/h/g; cerebellum, 71 mol/h/g; pons-medula and spinal cord, 94 and 60 mol/h/g, respectively. The distribution of CSD activity expressed as mol of cysteine sulfinate decarboxylated per h per g was the following: hypothalamus and colliculi, 14–21 mol/h/g; olfactory bulbs, cerebral cortex, striatum, hippocampus, and cerebellum, 8–13 mol/h/g; pons-medulla, 7.3; and spinal cord, 3.6 mol/h/g. No CSD activity was detected in sciatic nerve. The subcellular distribution of CO and CSD activities was studied in hypothalamus, colliculi, and cerebral cortex. CO activity was localized in synaptosomes, mitochondria, and microsomes. CSD was primarily confined to the crude mitochondrial fraction and after subfraction, recovered mainly in the synaptosomal fraction.  相似文献

Summary The Stannius corpuscle of the eel (Anguilla japonica) consists of numerous ovoid or polymorphic lobules separated by loose connective tissue containing blood capillaries. Each lobule is composed of a number of columnar secretory cells, containing numerous secretory granules, and arranged more or less radially. Each secretory granule, spherical, 0.5–1.0 in diameter, and osmiophilic, is surrounded by a limiting membrane derived from the Golgi membrane. The well developed rough-surfaced endoplasmic reticulum is closely associated with the Golgi field. Some Golgi elements might also be intimately associated with the outer nuclear membrane. Numerous glycogen particles are distributed throughout the cytoplasm. The secretory granules which tend to accumulate near the basal part of the cell might be released into the loose connective tissue. From these facts, the corpuscles of Stannius are considered to be protein-secretory endocrine glands without any similarity to the adrenal gland.  相似文献

Summary The incidence of myosatellite cells associated with white and red muscle fibres of the parietal muscle and red fibres of the craniovelar muscle was estimated by quantitative electron microscopy in the Atlantic hagfish (Myxine glutinosa, L.). Myosatellite cell nuclei constitute 3, 11 and 23 % of the total number of nuclei inside the basal lamina of the three types of muscle fibres, respectively. However, the total number of nuclei is highest in white fibres, most of the nuclei belonging to striated muscle cells. Myosatellite cell profiles in transverse sections constitute 23, 41 and 61 % of the number of muscle fibre profiles of the three types, respectively. The intervals between adjacent myosatellite cells are 135 m in white fibres, 55 m in red parietal fibres, and only 25 m in craniovelar fibres. Since craniovelar fibres are also comparatively thin, myosatellite cells constitute a significant fraction of the volume inside the basal lamina in these fibres. The myosatellite cells are 30–50 m long and up to 5 m thick. Some myosatellite cells possess few organelles, whereas others appear to contain many free ribosomes, granular endoplasmic reticulum, prominent Golgi apparatus and lysosome-like bodies.This investigation was supported by the Norwegian Research Council for Science and the Humanities (NAVF grant No. C20.30–37).
