不饱和脂肪酸在低渗牵张加强豚鼠胃窦平滑肌细胞毒蕈碱电流中的作用 总被引:1,自引:0,他引:1
利用全细胞膜片钳技术在急性分离的胃窦平滑肌细胞上记录离子电流的方法 ,探讨外源性不饱和脂肪酸是否参与低渗牵张加强毒蕈碱电流的过程。在豚鼠胃窦平滑肌细胞上膜电位被钳制在 - 2 0 0mV等渗状态时 ,5 0 μmol/L卡巴胆碱 (carbachol,CCh)引起的毒蕈碱电流 (ICCh)作为对照 ,发现低渗牵张可以使ICCh明显增加到对照的 2 2 6 0±2 1 0 %。当用含 5 μmol花生四烯酸 (arachidacid ,AA)、亚麻酸 (linoleicacid ,LA)或亚油酸 (oleicacid,OA)细胞外液灌流时 ,ICCh分别被抑制在对照的 3 8± 0 6%、3 5 2± 0 8%和 66 6± 0 6%。在这种情况下 ,低渗牵张刺激可以使ICCh分别增加到 10 6 0± 2 5 %、173 2± 6 8%和 2 2 2 1± 11 0 %。 5 μmol/LAA抑制低渗牵张增加的毒蕈碱电流 5 1 2± 3 8% ,而在等渗状态下抑制ICCh为 96 2± 1 6%。上述结果提示 ,不饱和脂肪酸中双键数目越多 ,抑制效应越强 ;但不饱和脂肪酸不参与低渗刺激加强毒蕈碱电流的过程。 相似文献

Three ruminally cannulated and multicatheterised lactating dairy cows were used to investigate the effect of different supplement strategies to fresh clover grass on urea and short-chain fatty acid (SCFA) metabolism in a zero-grazing experiment with 24-h blood and ruminal samplings. Fresh clover grass was cut every morning and offered from 0800 to 1500 h. Maize silage was fed at 1530 h. The three treatments, arranged in a Latin square, differed by timing of feeding rolled barley and soya-bean hulls relative to fresh clover grass. All diets had the same overall composition. Treatments were soya-bean hulls fed at 0700 h and barley fed at 1530 h (SAM), barley fed at 0700 h and soya-bean hulls fed at 1530 h (BAM), and both soya-bean hulls and barley fed at 1530 h (SBPM). The grass had an unexpectedly low content of crude protein (12.7%) and the cows were severely undersupplied with rumen degradable protein. The treatment effects were numerically small; greater arterial ammonia concentration, net portal flux of ammonia and net hepatic flux of urea during part of the day were observed when no supplementary carbohydrate was fed before grass feeding. A marked diurnal variation in ruminal fermentation was observed and grass feeding increased ruminal concentrations of propionate and butyrate. The net portal fluxes of propionate, butyrate, isovalerate and valerate as well as the net hepatic uptake of propionate, butyrate, valerate and caproate increased after feeding at 0700 h. The hepatic extraction of butyrate showed a relatively large depression with grass feeding with nadir at 1200 to 1330 h. The increased net portal absorption and the decreased hepatic extraction resulted in an approximately six-fold increase in the arterial blood concentration of butyrate. The gut entry rate of urea accounted for 70 ± 10% of the net hepatic production of urea. Saliva contributed to 14% of the total amount of urea recycled to the gut. Urea recycling to the gut was equivalent to 58% of the dietary nitrogen intake. Despite the severe undersupply of rumen degradable protein, the portal-drained viscera did not extract more than 4.3% of the urea supplied with arterial blood. This value is in line with the literature values for cows fed diets only moderately deficient in rumen degradable protein and indicates that cows maximise urea transfer across gut epithelia even when the diet is moderately deficient in rumen degradable protein. 相似文献

Unidirectional fluxes of short-chain fatty acids across pig, sheep and pony caecum, proximal and distal colon were studied
under short-circuit current conditions in Ussing chambers. Findings are compared with results from guinea pig. Marked species
differences are apparent; highest mucosal-to-serosal fluxes of acetate, propionate and butyrate were seen in guinea pig, lower
values in pig and smallest fluxes in sheep and pony. Segmental differences between caecum, proximal and distal colon exist
mainly in guinea pig and are less developed in pig, sheep and pony. Inhibition of Na+/H+ exchange by amiloride added to the mucosal solution decreased the mucosal-to-serosal fluxes of short-chain fatty acids clearly
in guinea pig caecum and proximal colon, and very little in distal colon. This effect was somewhat less pronounced in pig
caecum and distal colon, in caecum and distal colon of sheep and caecum of the pony. In pig, sheep and pony proximal colon
and pony distal colon no significant inhibition was observed. Inhibition of the K+-H+ ATPase by addition of ouabain to the mucosal solution diminished mucosal-to-serosal fluxes of short-chain fatty acids in

Prebiotics are known for their health benefits to man, including reducing cardiovascular disease and improving gut health. This review takes a critical assessment of the impact of dietary fibres and prebiotics on the gastrointestinal microbiota in vitro. The roles of colonic organisms, slow fermentation of prebiotics, production of high butyric and propionic acids and positive modulation of the host health were taken into cognizance. Also, the short-chain fatty acids (SCFAs) molecular signalling mechanisms associated with their prebiotic substrate structural conformations and the phenotypic responses related to the gut microbes composition were discussed. Furthermore, common dietary fibres such as resistant starch, pectin, hemicelluloses, β-glucan and fructan in context of their prebiotic potentials for human health were also explained. Finally, the in vitro human colonic fermentation depends on prebiotic type and its physicochemical characteristics, which will then affect the rate of fermentation, selectivity of micro-organisms to multiply, and SCFAs concentrations and compositions. 相似文献

As an alternative to pharmacological treatment to diseases, lifestyle interventions, such as dietary changes and physical activities, can help maintain healthy metabolic conditions. Recently, the emerging analyses of volatile organic compounds (VOCs) from breath and short-chain fatty acids (SCFAs) from plasma/feces have been considered as useful tools for the diagnosis and mechanistic understanding of metabolic diseases. Furthermore, diet-induced changes of SCFAs in individuals with diagnosed metabolic abnormalities have been correlated with the composition changes of the gut microbiome. More interestingly, the analysis of exhaled breath (breathomics) has gained attention as a useful technique to measure the human VOC profile altered as a result of dietary interventions. In this mini-review, we examined recent clinical trials that performed promising dietary interventions, SCFAs analysis in plasma/feces, and VOC profile analysis in exhaling breath to understand the relationship between dietary intervention and metabolic health. 相似文献

1. Absorption of short-chain fatty acids (SCFA), acetate, propionate, and butyrate was studied in simultaneously perfused proximal and distal segments of the colon in anaesthetized guinea pigs. 2. Acetate absorption rates increased linearly with concentration in both segments, indicating passive transport. 3. SCFA-clearance was independent of bulk luminal pH between pH 6.2 and 8.1 in the proximal and distal colon. SCFA-clearance was slightly higher in both segments at pH values less than 6. 4. The unexpected pH-independence of SCFA-absorption is attributed to the existence of a constant pH-microclimate at the surface of the colonic epithelium. 5. Relative permeabilities to acetate:propionate:butyrate were estimated as 1:1.19 +/- 0.03:1.27 +/- 0.05 in the proximal colon and 1:2.31 +/- 0.39:3.50 +/- 0.61 in the distal colon. Short-chain fatty acids (SCFAs), the end products of fermentation of dietary fibers by the anaerobic intestinal microbiota, have been shown to exert multiple beneficial effects on mammalian energy metabolism. The mechanisms underlying these effects are the subject of intensive research and encompass the complex interplay between diet, gut microbiota, and host energy metabolism. This review summarizes the role of SCFAs in host energy metabolism, starting from the production by the gut microbiota to the uptake by the host and ending with the effects on host metabolism. There are interesting leads on the underlying molecular mechanisms, but there are also many apparently contradictory results. A coherent understanding of the multilevel network in which SCFAs exert their effects is hampered by the lack of quantitative data on actual fluxes of SCFAs and metabolic processes regulated by SCFAs. In this review we address questions that, when answered, will bring us a great step forward in elucidating the role of SCFAs in mammalian energy metabolism. 相似文献

Summary The influence of short-chain fatty acids (SCFA) and osmolality on mucin release in the rat colon was studied histochemically by determining number of stained mucin-containing cells. SCFA did not significantly influence the number of cells staining for mucin. Hypertonic solutions (360 mosm/l) did not affect mucin release in the proximal colon, but stimulated mucin release in the distal colon. Solutions of lower osmolality (300 or 250 mosm/l) caused a considerable release of mucin from goblet cells as well as vacuolated cells in both the proximal and the distal colon; the lower the osmolality, the more mucin was released. The mucosa of the distal colon was conspicuously affected by solutions of lower osmolality. The unidirectional transport and metabolism of 14C-labeled acetate, propionate and butyrate across the isolated bovine rumen epithelium was measured in vitro by the Ussing chamber technique. There was a significant, but relatively small, net secretion of acetate and propionate, and a large and significant net absorption of butyrate. The results demonstrate that the mucosal-serosal (MS) pathway for short-chain fatty acids (SCFA) is different from the serosal-mucosal (SM) pathway, and that butyrate is treated differently from acetate and propionate by the epithelium. The results support that the main route for epithelial SCFA transport is transcellular. The correlation between SCFA lipophility and the flux rate was positive but weak at both pH 7.3 and 6.0. Decreasing pH increased all SCFA fluxes significantly, but not proportionally to the increase of protonized SCFA in the bathing solution. There was a significant and apparently non-competitive interaction between the transport of acetate, propionate and butyrate. It seems that mediated transport mechanisms must be involved in epithelial SCFA transport in the bovine rumen, but the data do not exclude that passive diffusion could account for a significant part of the flux. The metabolism of SCFA in the Ussing chamber system was considerable, and there was a clear preference for excretion of CO2 from this metabolism to the mucosal side, while side preference for non-CO2 metabolite excretion was not studied. Of the propionate and butyrate transported in the MS direction, 78 and 95% was metabolised, while only 37 and 38% was metabolised in the SM direction (acetate metabolism could not be measured). There was, however, no simple relation between the degree of metabolism and the transport rate or the transport asymmetry of the SCFA. 相似文献

In France, animal products (dairy products, meat and eggs) are the main source of n-3 polyunsaturated fatty acids (PUFA) in the human diet; however, many individuals do not consume enough of this nutrient. The objective of this study was to increase n-3 PUFA precursor and derivative contents in tissues and test how they influence technological and sensory qualities of meat without negatively affecting growth performances of pigs. A total of 60 male pigs [(Large White×Landrace)×Pietrain] were assigned according to their initial liveweight (50.7±2.7 kg) to five experimental groups corresponding to five different diets that they received from 14 to 22 weeks of age. Dietary lipid supplements were composed of soybean and palm oil (SP), dehulled and extruded linseed (EL-), docosahexaenoic acid (DHA)-rich microalgae (MAG) or a mixture of linseed and microalgae at 75%/25% (3EL-/MAG) and 50%/50% (EL-/MAG), respectively. Diet did not influence growth performances of pigs or the technological quality of the meat. The n-3 PUFA content in the longissimus dorsi muscle, subcutaneous backfat (SCB) and liver increased with a dietary supply of linseed and microalgae and corresponded to circulating fatty acids (FA). The amount of malondialdehyde, representative of FA lipid peroxidation measured in SCB, increased significantly with the supply of microalgae, meaning that PUFA from the microalgae included in the diet increased the meat’s susceptibility to oxidation. The MAG diet scored highest for ‘abnormal’ flavor, similar to that of fish or organ meat, but the n-3 PUFA-rich diet had no effect on other sensory characteristics. Results of this study indicate benefits of enriching animal feed with n-3 PUFA, but the inclusion of long-chain n-3 PUFA such as DHA must be limited to avoid oxidation susceptibility and development of an off-odor. 相似文献

This study aims to investigate the gut microbiota and metabolites in breastfed infants with breast milk jaundice (BMJ) using gut microbiome–metabolomics. Breastfed newborns diagnosed with BMJ and those without BMJ (control group) were enrolled. Faecal samples were collected from the participants and subjected to high-throughput sequencing of the 16s rDNA V3 and V4 regions of the gut flora and metabolomics of short-chain fatty acids (SCFAs). Proteobacteria, Fimicutes and Actinobacteria were the main bacteria at the phylum level. Eshcerichia-Shigella and Enterobacteriacea were the main bacteria at the genus level. The difference between the two groups was compared. Compared to the control group, the amount of Streptococcus was significantly increased while the amount of Enterococcus was significantly decreased in the faeces from infants with BMJ. Functional prediction analysis of 16S found that biosynthesis of penicillin and cephalosporin significantly increased in the BMJ group. Gas chromatography–mass spectrometry detection of SCFAs revealed that levels of acetic acid and propionic acid were significantly lower in the BMJ group than in the control group. The reduced levels of acetic acid and propionic acid may be related to the increase in Streptococcus and decrease in Enterococcus, both of which may contribute to BMJ. 相似文献

Consumption of reishi mushroom has been reported to prevent colon carcinogenesis in rodents, although the underlying mechanisms remain unclear. To investigate this effect, rats were fed a high-fat diet supplemented with 5% water extract from either the reishi mushroom (Ganoderma lingzhi) (WGL) or the auto-digested reishi G. lingzhi (AWGL) for three weeks. Both extracts markedly reduced fecal secondary bile acids, such as lithocholic acid and deoxycholic acid (colon carcinogens). These extracts reduced the numbers of Clostridium coccoides and Clostridium leptum (secondary bile acids-producing bacteria) in a per g of cecal digesta. Fecal mucins and cecal propionate were significantly elevated by both extracts, and fecal IgA was significantly elevated by WGL, but not by AWGL. These results suggest that the reishi extracts have an impact on colon luminal health by modulating secondary bile acids, microflora, mucins, and propionate that related to colon cancer. 相似文献

Retinal pigment epithelium pigment granules stimulate the photo-oxidation of unsaturated fatty acids

The cellular pigments of the retinal pigment epithelium (RPE) have been shown to catalyze free radical activity, especially when illuminated with visible or ultraviolet light. This activity is sufficient to cause photooxidation of several major cellular components. The present investigation determined the relative ability of melanin, lipofuscin, and melanolipofuscin granules isolated from human and bovine eyes to oxidize polyunsaturated fatty acids, specifically linoleic and docosahexaenoic acids. The dark reactivity as well as the light-stimulated reactions were determined. The production of hydroperoxide derivatives of the linoleic and docosahexaenoic acids were determined by NADPH oxidation coupled to the activity of glutathione peroxidase, and also by production of thiobarbituric acid reactive substances. All RPE pigment granules stimulated fatty acid oxidation when irradiated with short wavelength (< 550 nm) visible light, with the melanosomes exhibiting the greatest light-induced activity. Only lipofuscin granules, however, caused peroxidation of fatty acids in the dark. These findings provide additional support for the role of RPE pigments in "blue light toxicity" as well as indicating that accumulation of lipofuscin may contribute to increased photooxidation in the aging RPE. 相似文献

In camelids the ventral parts of compartments 1 and 2 (C1/C2) and the total surface of compartment 3 of the forestomach are

temporarily isolated, washed and filled with buffer solutions. Absorption of short-chain fatty acids (SCFA) and net absorption
of sodium and water were estimated relative to Cr-ethylenediaminetetraacetic acid as a fluid marker. SCFA were extensively
absorbed in the forestomach; clearance rates of SCFA with different chain lengths were equal. After lowering the pH of solutions
SCFA absorption rates increased, but much less than the increase of the non-ionized fraction. Absorption of propionate was
lower when acetate had been added. Findings suggest that most of the SCFA in camels are transported in the ionized form, most
likely via an anion exchange mechanism. Net water absorption is closely related to net sodium absorption. Apparently water
absorption results from an iso-osmotic process. Differences between absorption mechanisms of SCFA from the forestomach of

Recently, a “human gut microbial gene catalogue,” which ranks the dominance of microbe genus/species in human fecal samples, was published. Most of the bacteria ranked in the catalog are currently publicly available; however, the growth media recommended by the distributors vary among species, hampering physiological comparisons among the bacteria. To address this problem, we evaluated Gifu anaerobic medium (GAM) as a standard medium. Forty-four publicly available species of the top 56 species listed in the “human gut microbial gene catalogue” were cultured in GAM, and out of these, 32 (72%) were successfully cultured. Short-chain fatty acids from the bacterial culture supernatants were then quantified, and bacterial metabolic pathways were predicted based on in silico genomic sequence analysis. Our system provides a useful platform for assessing growth properties and analyzing metabolites of dominant human gut bacteria grown in GAM and supplemented with compounds of interest. 相似文献

The role of short-chain fatty acids in the host-seeking behaviour of Triatoma infestans larvae was investigated using a locomotion compensator. Several short-chain fatty acids were tested alone over a wide range of doses, or in combination with L-lactic acid (L-LA; 100 microg). Bugs showed no attractive response to single carboxylic acids, but when L-LA was added to airstreams carrying specific intensities of either propionic (C3; 100 microg), butyric (C4; 1 microg) or valeric acid (C5; 1 microg), these mixtures elicited an attractive response, evincing a synergistic effect. No orientation response was observed when caproic acid (C6) was offered with L-LA at the doses tested. Two blends were created: (1) C3, C4 and C5 combined at the effective doses when added with L-LA [C3C4C5 (1)], and (2) C3, C4 and C5 combined at a third of those intensities [C3C4C5 (2)]. Both blends were tested alone, with L-LA (100 microg), with a sub-threshold concentration of CO(2) (300 p.p.m. above the ambient level), and combined with both compounds together. Oriented responses of bugs were only observed with the blend (2) added with L-LA and with the combination of this lure with CO(2). This last combination evoked a behavioural response similar in intensity to that induced by a live mouse. 相似文献
