A technique is described which indicates by u.v. fluorescence the concentration of caffeoylquinic, chlorogenic and isochlorogenic acids in roots of germinating lettuce and carrot. The surface fluorescence of radicles from lettuce root aphid (Pemphigus bursarius) resistant seedlings was more intense than for susceptible seedlings, attributable to the higher concentration of isochlorogenic acid. In contrast, radicles of carrot seedlings resistant to carrot fly (Psila rosae) were less fluorescent than susceptible seedlings, corresponding to the lower concentration of chlorogenic acid in resistant seedlings. The technique is non-destructive and has been developed to distinguish between cultivars or breeding lines potentially resistant to these insect pests.  相似文献

The inheritance of resistance in two lettuce cultivars to lettuce root aphid, Pemphigus bursarius, was studied in a series of laboratory and field experiments at Wellesbourne between 1989 and 1992. A source of total resistance in the cv. ‘Avoncrisp’ which is linked to the downy mildew resistance gene Dm6, was shown to be governed by a single dominant gene. There were no maternal effects evident in the inheritance of this resistance. The basis of the high level of resistance which exists in the cv. ‘Lakeland’ (formerly known as ‘Jubilee’) was also shown to be controlled by the same dominant gene. The linkage between Dm6 and root aphid resistance was broken in ‘Lakeland’ as this cultivar does not possess the Dm6 gene. The linkage was presumably broken when the original cross between the parents of cv. ‘Lakeland’, ‘Calmar’ and ‘Avoncrisp’ was made. Under laboratory conditions small numbers of aphids commence development on cv. ‘Lakeland’ but colonies fail to develop and under field conditions the resistance provides adequate Protectión against the pest. The resistance in both ‘Avoncrisp’ and ‘Lakeland’ was effective against a population of lettuce root aphid collected from an endive crop in southern France as well as being effective against the Wellesbourne population of this aphid.  相似文献

该文以14个扦插培育的甜叶菊品种叶为材料,从8种不同型号的树脂中筛选出一种合适的大孔吸附树脂对甜叶菊叶中绿原酸类成分进行纯化前处理,采用HPLC法对不同甜叶菊品种叶中所含绿原酸类成分进行比较分析。结果表明:(1)在8种不同型号的树脂中,XAD~(-1)6对甜叶菊叶中绿原酸类成分吸附-解析性能最佳。(2)经优化,上样液浓度1.20 mg·mL~(-1)、样品溶液pH 3、解析液乙醇体积分数70%时XAD~(-1)6树脂对甜叶菊叶中绿原酸类成分具有较好的纯化效果。(3) HPLC检测分析表明,在14个品种中共检测出新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C六种绿原酸类成分,其中主要成分均为异绿原酸A、绿原酸、异绿原酸C,而在品种3、5、13、14中没有检测出异绿原酸B。(4) 14个品种中6个绿原酸类成分的含量分别为异绿原酸A 20.55~54.3 mg·g~(-1)、绿原酸17.96~32.93 mg·g~(-1)、异绿原酸C 4.15~19.49 mg·g~(-1)、新原酸0.61~4.61 mg·g~(-1)、隐绿原酸0.52~3.11 mg·g~(-1)、异绿原酸B 0.0~3.17 mg·g~(-1),6种绿原酸类成分总量为43.9~97.8 mg·g~(-1)。可见,不同品种甜叶菊叶中绿原酸类成分含量有明显差异,富含绿原酸类成分的甜叶菊品种可用于开发获取绿原酸类物质。  相似文献

Ethylene stimulated the activity of l -phenylalanine ammonia-lyase (PAL) in carrot (Daucus carota L.) tissues. It is also known to induce the formation of isochlorogenic acid. However, the induction patterns were different for isochlorogenic acid and PAL activity. Ethylene action seems to be indirect, as it did not affect the PAL activity of cell-free extracts.  相似文献

Striking varietal differences in susceptibility to attack by the lettuce root aphid, Pemphigus bursarius L., were first found in lettuces grown at Wellesbourne in 1955. Subsequent work has confirmed that White Favourite and Imperials E-4 and 19551-M are highly resistant; Imperial 45634-M, Continuity and Iceberg are markedly resistant. Midas, Salad Bowl and Imperial 4164 each appear to be a mixture of susceptible and resistant plants.
Immigrant winged forms of P. bursarius showed no preference for colonizing any particular variety of lettuce, and it seems that resistance to attack results from antibiosis.
It is suggested that varietal differences in the composition of root sap may account for the differences in susceptibility to lettuce root aphid, but analyses of water and alcohol extracts of root sap from resistant and susceptible lettuce varieties have not shown consistent differences.  相似文献

Abstract The effects of light and gibberellic acid (GA3) on growth and phenylalanine ammonia-lyase (PAL) activity were studied in seedlings of lettuce (Lactuca sativa L.). Using an in vivo assay for PAL it was shown that wounding caused by excising hypocotyls results in an increase in PAL activity with time that can mask the effect of light on the activity of this enzyme. When hypocotyl sections were excised from light-treated seedlings immediately prior to the in vivo assay of PAL, light was shown to cause a marked increase in PAL activity. Experiments with an inhibitor of PAL activity, α-aminooxy-β-phenylpropionic acid (AOPP), confirmed that the volatile radioactive products measured in the in vivo assay resulted from the activity of PAL. Gibberellic acid suppresses the light-induced increase in PAL activity and there is an inverse relationship between GA3-induced growth and the activity of PAL. Over a wide range of GA3 concentrations, the activity of PAL is also inversely correlated with growth rate along the length of the hypocotyl section; the upper halves of sections elongate more rapidly and have lower levels of PAL than the lower halves. Despite the strong correlation between growth and PAL activity, experiments with AOPP and t-cinnamic acid show that it is unlikely that elongation is regulated directly by products of PAL activity.  相似文献

Russet spotting (RS), consisting of numerous small brown spots on the midrib of head lettuce (Lactuca sativa), is a physiological disorder induced by exposure to ethylene. In leaves suffering RS, the increase in spotting was accompanied by a parallel increase in the amount of phenolic compounds. Of these, chlorogenic acid and isochlorogenic acid were identified. Ethylene induced high phenylalanine ammonia-lyase (PAL) activity and RS formation in the susceptible cultivar Salinas, but not in the resistant cultivar Calmar. In the absence of ethylene neither significant PAL induction nor RS occurred. No correlation was found between the increase in polyphenol oxidase or peroxidase and the development of RS. The increase in PAL activity, however, was closely correlated with the development of RS. The increase in PAL activity preceded the development of RS, and the extent of RS was directly related to the level of PAL. Three temperatures (0.5, 5.5, and 12.5 C) were compared on the basis of their influence on both RS and PAL induction. At the lowest temperature (0.5 C) neither PAL induction nor RS occurred to a significant extent. At the highest temperature (12.5 C) an initial rapid increase in PAL activity and an earlier development of spotting were observed, but subsequently there was a decrease in both PAL activity and the rate of development of RS. At the medium temperature (5.5 C) both PAL activity and RS increased progresively with time. The decline of PAL activity at a higher temperature might be attributed to inactivation of the enzyme. Thus, a temperature favorable for induction of PAL activity by ethylene was also favorable for RS. These observations indicate a close interrelationship between the induction of PAL activity and the development of RS in response to ethylene, and suggest a causal relationship between the two events. PAL serves as a useful biochemical marker for the RS reaction.  相似文献

The inheritance of resistance to lettuce root aphid, Pemphigus bursarius, was studied in lettuce using the Wellesbourne cultivars Avondefiance and Avoncrisp as resistant parents and Borough Wonder and Webb's Wonderful as aphid-susceptible parents. All four cultivars were crossed in all possible combinations including reciprocals and the response to root aphid of plants in the P1F1F2 and BC generations was assessed using apterae of P. bursarius from the lettuce cv. Iceberg. Resistance to attack was clearly inherited and the parents appeared to be homozygous for their resistance or susceptibility. In the F1 generation, however, in all crosses between resistant and susceptible parents, segregation into susceptible, resistant and some slightly less resistant plants occurred. This and the highly significant differences in segregation between pairs of reciprocal crosses in the F1 and other generations indicate that the inheritance of resistance to root aphid is controlled by extra-nuclear factors. Modifying genes might also be involved but there appears to be no linkage of root aphid resistance with resistance to downy mildew, for which the Wellesbourne lettuces were bred.  相似文献

The purpose of this study was to investigate the effect of repeated harvesting on the content of caffeic acid (CA) and seven species of caffeoylquinic acids (CQAs) in sweet potato leaves using a newly developed high-performance liquid chromatography method. Six cultivars and two breeding lines were used in this study. Leaves were collected at monthly intervals from 1st harvest (May) to 4th harvest (August) in 2011 and 2012. ANOVA analysis revealed that the contents of CQAs were significantly different among all cultivars and breeding lines, but no significant differences were found for CA. No annual variation was confirmed in CA and CQAs. Repeated harvest of sweet potato leaves affected the content of only 4-CQA and 5-CQA. Post-hoc comparisons using Tukey’s method indicated that the contents of 4-CQA and 5-CQA in sweet potato leaves harvested at first time were significantly higher compared to those at the other harvest times.  相似文献

Fifteen lettuce cultivars representing three different morphological types were grown in a sand alumina system under conditions of low (deficient) and high (sufficient) P supply. An efficient plant was defined as one that produced a large shoot fresh weight under low P conditions. Cultivars within their respective groups varied significantly for some traits that appeared to be important in determining adaptation to P. This led to the conclusion that accumulation of P in shoot tissue or the total plant was the main difference between efficient and inefficient cultivars. Accumlation of P seemed to be due to a greater absorption capability of roots or greater root mass (weight), depending on the different lettuce groups. Differences in internal use of P did not contribute to differences in shoot fresh weight.The butterhead cultivars were the least efficient plants when grown under low P. Compared to the other groups, plants has lower translocation efficiency and a greater root: shoot ratio. Never-the-less, butterhead cultivars as efficient as the best cultivars of other groups were found. There were no differences between Brazilian and American cultivars for any of the traits analysed, probably due to the fact that in both countries vegetables are bred under high fertility levels and grown with heavy applications of fertilizers. The results of this study demonstrate that there were genotypic variability and/or genotype×environment interaction effects for shoot weight (yield) among the lettuce cultivars grown under low P conditions imposed in the sand-alumina system.  相似文献

Root architecture in cultivated and wild lettuce (Lactuca spp.)   总被引:2,自引:1,他引:1  
Root architecture is described for intact root systems of cultivated (Lactuca sativa L.) and wild (L. serriola L.) lettuce, grown for 5 to 6 weeks in greenhouse pot and cylinder experiments in coarse-textured soil. L. sativa cv. Salinas and a sclinas line of L. serriola attained the same biomass at 4 to 6 weeks after planting. Root biomass allocation was also similar, but root architecture differed. In the top zone along the tap root (0 to 5 cm), Salinas tended to produce more laterals, a greater total root length, and more external links (segments that originate at a branch point and end in a meristem) than wild lettuce. In the 5 to 55cm zone of the tap root, these measures were greater in the wild species. These patterns of root structure were generally corroborated by a second cylinder study with a different pair of L. sativa and L. serriola. Regressions of root structural traits were made against total root dry weight as a means to compare root architecture independently of the size of the root system. Regressions demonstrated that production of root segments differed between the two species; for example the slope for the regression of summed external link length in the top 0 to 5 cm with total root dry weight was significantly higher for Salinas, indicating that the rate of construction in the top 0 to 5cm was greater for cultivated than for wild lettuce. Yet, from 5 cm depth to the tap root tip, the rate of construction of external link length was greater in L. serriola. For many of these types of regression, r2 and mean slope ± SE suggested that more variation occurred in cultivated than wild lettuce, yet genetic heterogeneity was probably low within the studied taxa. Inadvertent selection may have occurred in the breeding of cultivated lettuce varieties for increased root growth in the surface zone where water and fertilizer are applied, and for greater plasticity in construction of root segments, which might maximize the efficiency of exploitation of soil moisture and nutrients.  相似文献

The genetics of partial resistance (PR) of lettuce to the leaf aphid species Macrosiphum euphorbiae and Uroleucon sonchi was studied in F1 and F2 generations of six crosses between four cultivars. Partial resistance inherited mainly additively. With M. euphorbiae the estimates of broad-sense F2 heritability were low and non-significant in crosses between parents of the same resistance level (S x S and PR x PR), and high (0.69-0.76) in crosses between parents of different resistance level (PR x S). The level of resistance in lettuce cultivars ‘Charan’ and ‘Marbello’ led to a marked reduction in percentage of plants with colonies of M. euphorbiae in a field experiment. With U. sonchi the highest estimates of F2 heritability were found in ‘Charan x ‘Avoncrisp’ (0.51) and ‘Charan’ x ‘Chou de Naples' (0.48). In the cross ‘Charan’ x ‘Marbello’ (PR x PR) no significant segregation for resistance to M. euphorbiae was found, indicating that these cultivars may have the same genetic constitution for resistance to M. euphorbiae.‘Charan’ has additional genes for resistance to U. sonchi, that are not, or only weakly linked to the gene(s) for resistance to M. euphorbiae. The heritability estimates for the partial resistance to M. euphorbiae and U. sonchi indicate that it is possible to effectively select for these traits in plant breeding programmes.  相似文献

The effects of an aqueous plant-derived smoke extract, octanoic acid and ethylene on germination of light-sensitive Grand Rapids lettuce seeds were investigated. The smoke extract brought about a concentration dependent increase in germination and a complete inhibition of germination at high concentrations. Octanoic acid could not induce germination. Ethylene at concentrations over 5 L L–1 increased lettuce seed germination, but not to the same degree as smoke. Aqueous smoke in combination with ethylene showed a synergistic effect on germination at suboptimal smoke concentrations. At high smoke concentrations the effect of ethylene was almost completely inhibited. Octanoic acid in combination with ethylene brought about a higher level of germination than with ethylene alone, but only at the highest concentration of octanoic acid tested (1 mM). Standardized hexane and dichloromethane-partitioned smoke extracts and octanoic acid were subjected to TLC separation. The R f -fractions in the smoke lanes showing activity in the lettuce seed bioassay did not correspond to the R f -value of octanoic acid. As aqueous smoke can withstand autoclaving and can be separated by TLC and HPLC without loosing activity it is unlikely that the activity of aqueous smoke is linked to ethylene. Quantitative estimates of free fatty acids in total lipid extracts of plasma were obtained by glc on nonpolar columns following trimethylsilylation. The presence of other lipid esters in the reaction mixture had no effect upon the yield of the trimethylsilyl (TMS) esters or upon their resolution on the glc column. Routine quantitations by gas-liquid chromatography (glc) were obtained on 2 ft × 1/8 in. o.d. stainless steel columns packed with 3% OV-1 on 100–120 mesh Gas Chrom Q by means of temperature programming in the range 175–350°C with tridecanoin as internal standard. High resolution glc of the TMS esters of fatty acids was done on a 6 ft × 1/8 in. o.d. glass column packed with 1% SE-30 on Gas Chrom Q. In both instances the fatty acids were resolved on the basis of carbon number and by the presence or absence of double bonds. On gas chromatography/mass spectrometry (GC/MS), TMS esters of fatty acids were shown to yield proportionally greater amounts of high mass fragments, including the parent ions, than their methyl or ethyl esters, which has special advantages for the detection and characterization of polyunsaturated fatty acids.  相似文献

 Transgenic plants of four glasshouse-grown lettuce cultivars ('Cortina', 'Evola', 'Flora' and 'Luxor') were obtained by co-cultivating excised cotyledons with Agrobacterium tumefaciens. The Agrobacterium strain LBA4404 contained the binary vector pBCSL16, which carried a nitrate reductase (nia) cDNA linked to CaMV promoter and terminator sequences, and the neomycin phosphotransferase II (nptII) gene. Transformed shoots were selected by their ability to root on medium containing kanamycin sulphate, by a positive NPTII assay and by PCR analysis. The presence of the nia cDNA in transgenic lettuce was confirmed by nitrate reductase (NR) enzymatic assay, a reduction in the nitrate content of leaves and by Southern hybridisation. PCR analysis of cDNA fragments from transgenic plants confirmed that both nia and nptII genes were expressed in first seed-generation (T1) lettuce plants. 以富含绿原酸类成分的苦丁茶(Ilex kaushue)为材料,使用溶剂甲醇、乙醇、丙酮和水,结合超声波提取、水浴提取、回流提取等方法对绿原酸及其异构体的提取效率及提取后各异构体的变化进行分析。应用超高效液相色谱法可使苦丁茶的6种绿原酸类成分及咖啡酸在6 min内实现分离。提取结果表明,丙酮作为提取溶剂,在采用超声波和水浴提取时能获得较高的提取效率,易获得较多总量的绿原酸类成分和高含量异绿原酸A。而最常用的溶剂乙醇并未达到理想的提取效果。在不同溶剂的回流提取中,虽然提取的绿原酸类成分总量接近,但异绿原酸A和异绿原酸C含量有较大差异。醇溶液特别是乙醇溶液的回流提取使异绿原酸C的量大幅增加,而相应的异绿原酸A的量大幅减少,表明在醇加热条件下,异绿原酸A转化为异绿原酸C,这为获得抗氧化性更强的异绿原酸C提供了新思路。  相似文献

Plants typically respond to environmental stresses by inducing antioxidants as a defense mechanism. As a number of these are also phytochemicals with health-promoting qualities in the human diet, we have used mild environmental stresses to enhance the phytochemical content of lettuce, a common leafy vegetable. Five-week-old lettuce (Lactuca sativa L.) plants grown in growth chambers were exposed to mild stresses such as heat shock (40 °C for 10 min), chilling (4 °C for 1 d) or high light intensity (800 μmol m?2 s?1 for 1 d). In response to these stresses, there was a two to threefold increase in the total phenolic content and a significant increase in the antioxidant capacity. The concentrations of two major phenolic compounds in lettuce, chicoric acid and chlorogenic acid, increased significantly in response to all the stresses. Quercetin-3-O-glucoside and luteolin-7-O-glucoside were not detected in the control plants, but showed marked accumulations following the stress treatments. The results suggest that certain phenolic compounds can be induced in lettuce by environmental stresses. Of all the stress treatments, high light produced the greatest accumulation of phenolic compounds, especially following the stress treatments during the recovery. In addition, key genes such as phenylalanine ammonia-lyase (PAL), l-galactose dehydrogenase (l-GalDH), and γ-tocopherol methyltransferase (γ-TMT) involved in the biosynthesis of phenolic compounds, ascorbic acid, and α-tocopherol, respectively, were rapidly activated by chilling stress while heat shock and high light did not appear to have an effect on the expression of PAL and γ-TMT. However, l-GalDH was consistently activated in response to all the stresses. The results also show that these mild environmental stresses had no adverse effects on the overall growth of lettuce, suggesting that it is possible to use mild environmental stresses to successfully improve the phytochemical content and hence the health-promoting quality of lettuce with little or no adverse effect on its growth or yield.  相似文献

Summary At 20° C and at 30° C in darkness, concentrations of indole acetic acid (IAA) greater than 10–7 M inhibited the germination of Grand Rapids lettuce at 24 h and 48 h after the beginning of imbibition. There was no marked, readily defined period of inhibition during germination that could be associated solely with an effect of IAA on suppressing radicle extension. Gibberellin A4+7, benzyladenine and red light were capable of reversing the effects of IAA. Cutting leaves of Romaine lettuce ( Lactuca sativa L. cv. Longifolia) produces a wound signal that induces the synthesis of phenylalanine ammonia lyase (PAL, EC 4.3.1.5) and the accumulation of phenolic compounds in cells up to 2 cm from the site of injury, and tissue browning near the site of injury. The response of leaves within a head of Romaine lettuce to putative chemical wound signals [abscisic acid (ABA), jasmonate (JA) and methyl jasmonate (MeJA)] differed significantly with leaf age. Exposure of harvested heads of lettuce to ABA, JA, MeJA, or salicylic acid (SA) did not induce changes in PAL activity, the concentration of phenolic compounds or browning in mature leaf tissue that was similar to the level induced by wounding. Methyl jasmonate applied as vapour (10, 100 or 1000 µl kg−1 FW), or as an aqueous spray or dip (0.01–100 µ M ) at 5 or 10°C did not produce an effect on PAL activity or browning that differed significantly from the untreated controls. In contrast, JA, MeJA and SA did induce elevated levels of PAL activity in younger leaves. However, the levels induced were far lower than those induced by wounding. Wound induced phenolic metabolism in mature leaves appears to be induced by different signals than those functioning in young leaves.  相似文献

When crops are bred for resistance to herbivores, these herbivores are under strong selection pressure to overcome this resistance, which may result in the emergence of virulent biotypes. This is a growing problem for crop species attacked by aphids. The Nr‐gene in lettuce confers near‐complete resistance against the black currant‐lettuce aphid, Nasonovia ribisnigri (Mosely) (Hemiptera: Aphididae). Since 2007, populations of N. ribisnigri have been reported in several locations in Europe to infest resistant lettuce varieties that possess the Nr‐gene. The objective of this study was to analyse the behaviour and level of virulence of several N. ribisnigri populations observed to have colonized Nr‐locus‐containing lettuce lines. We analysed the stylet penetration and feeding behaviour, and the performance of these N. ribisnigri populations on resistant and susceptible lettuce lines. Large variation in the degree of virulence to the Nr‐locus‐containing lettuce lines was found among populations of the Nr:1 biotype. The German population was highly virulent on the Nr‐containing resistant lettuce lines, and showed similar feeding behaviour and performance on both the susceptible and resistant lettuces. The French population from Paris was the second most virulent, though reproduction on the resistant lines was reduced. The French population from Perpignan and a population from Belgium, however, showed reduced performance and feeding rate on the resistant compared to the susceptible lettuces. The lettuce background in which the Nr‐gene is expressed influences the level of resistance to the various Nr:1 aphid populations, because the performance and feeding behaviour differed between the aphids on the cultivars (romaine lettuce) compared to the near‐isogenic lines (butterhead/iceberg lettuce). This study also shows that being able to feed on a plant not automatically implies that a population can successfully develop on that plant, because aphids showed phloem ingestion during the 8‐h recording period on resistant lettuce, but were not able to survive and reproduce on the same lettuce line.  相似文献
