Three groups of six calves each were fed a milk replacer at 0.8 kg and a starter concentrate ad libitum. Calves of the control group received the basal diet supplemented with rapeseed oil at 10 g per kg of feed solids. Calves of treatment groups were fed diets supplemented with a synthetically produced oil containing 62.3% methyl esters of CLA. The CLA-oil was added to milk at expense of rapeseed oil and fed at 5 and 10 g x kg(-1) feed solids for 63 days. Calves were slaughtered at 115 days of age. There was no significant effect of CLA on growth, intake of starter, feed conversion, chemical composition of meat and its oxidative stability. Dietary supplementation with CLA at 10 g x kg(-1) significantly increased CLA content in m. longissimus dorsi (MLD) from 5.6 to 19.3 mg x 100 g(-1), in liver from 13.1 to 68.8 mg x 100 g(-1), and in perirenal fat from 0.37 to 3.17 g x 100 g(-1). Dietary CLA decreased the ratio of cis-9, trans-11 and trans-10, cis-12 isomers of CLA in tissues, concentration of monounsaturated fatty acids in the MLD and fat, as well as the concentration of fatty acids with 20 and 22 carbon atoms. It can be concluded that in veal calves unprotected CLA apparently escaped ruminal hydrogenation, but was preferentially incorporated into depot fat. 相似文献

The heart phospholipid content and fatty acid composition were examined in adult rats after four weeks of feeding lipid-supplemented diets (20 g % w/w) containing sunflower oil-lard (1:1) mixture (SL group) or margarine (M group). Our results showed a decreased cardiolipin content and distribution in both experimental groups and an increased lysophosphatidylcholine and phosphatidylcholine content and distribution in the SL group with a tendency to lower phosphatidylcholine/phospatidylethanolamine ratio in both experimental groups. In the SL group, the content of saturated fatty acids was higher and that of monounsaturated fatty acids was lower than in the control group. The M group showed inverse results. The content of saturated fatty acids was lower and that of monounsaturated was higher than in the control group. Polyunsaturated n-6 fatty acids were decreased in both experimental groups and n-3 fatty acids were increased in the M group. Feeding lipid-supplemented diets reduced n-6/n-3 and 20:4/22:6 ratios in the M group. The polyunsaturated/saturated fatty acid ratio was lower in the SL and higher in indicating the M group than in the control group. Our results are in agreement with the other reports indicating that the heart is sensitive to diet-induced lipid alterations. 相似文献

The purpose of this study was to explore the effect of dietary fats on the hepatic fatty acid profile of mice liver after
partial hepatectomy and to observe a correlation with changes in element content. Male Balb/C mice were divided into three
groups: the control animals fed a standard diet (FSD), the FOO group fed a diet enriched with 5% olive oil, and the FCO group
fed a diet enriched with 5% corn oil. Hepatic fatty acid and element content were analyzed within each group in intact animals
and on d 1, 2, and 7 after partial hepatectomy. During the regenerative process, proportions of 18∶1n−9 and 18∶3n−6 substantially
increased in the FSD diet, correlating with the Zn level. On the other hand, 20∶4n−6 and 22∶6n−3 decreased on d 1 and 2. Consequently,
a significant increase in the n−6: n−3 ratio was found in these animals. In the FOO diet, a decreased polyunsaturated fatty
acid/monounsaturated fatty acid (PUFA/MUFA) ratio was related to a significant decrease in PUFA content, mainly the results
of decreased 30∶3n−6 and 20∶4n−6. The proportion of 18∶1n−9 was highly increased when compared with other diets and remained
high during the regeneration. Furthermore, the n−6∶n−3 ratio was increased on d 2 and 7. Zn increased on d 1 and 2, and Fe
increased on d 2. Feeding with corn oil generally induced an increase in the PUFA n−6 series, compared with other diets. The
PUFA n−3 series decreased and the 18∶1n−9 increased on d 1, compared to intact animals. Consequently, the n−6∶n−3 ratio was
elevated during the regeneration. Zn increased on d 1 and 2, whereas Fe remained unchanged until d 7, when it decreased. Decreased
20∶4n−6 on d 1 and 2, as well as Cu on d 7, and increased Zn in the first 2 d were common to all three diets. These findings
suggest that some significant signals transmitted during the regenerative process have induced alterations in the fatty acid

Male rats were fed diets containing olive or marine fish oils (10% w/w) with or without added cholesterol (1% w/w). After six weeks of feeding, the major fatty acid composition, fluidity, fatty acid desaturating and cholesterol biosynthesis/esterification related enzymes of liver microsomes were determined. Both olive oil and marine fish oil diets, without added cholesterol, enriched content of oleic and docosahexaenoic acids, respectively, of rat liver microsomes. The results were consistent with reduction in delta 6 and delta 5 desaturation of n-6 essential fatty acids and higher fluidity in the marine origin oil group. Inclusion of cholesterol into diets resulted in decreased membrane arachidonic acid content, with concomitant increase in linoleic acid content. Cholesterol feeding also decreased delta 6 and delta 5 desaturase activities, as well as membrane fluidity. Furthermore, the activity of acyl-CoA:cholesterol acyltransferase decreased, whereas the activity of hydroxymethylglutaryl-CoA reductase increased, in liver microsomes from both cholesterol-fat groups. 相似文献

Dietary fatty acid incorporation and changes in various lipid and phospholipid classes in the mussel Mytilus galloprovincialis subjected to three different dietary regimens were analysed and compared. Group A was unfed; group B received a diet consisting of 100% Thalassiosira weissflogii, exhibiting the typical fatty acid composition of diatoms, and group C received a diet consisting of 100% wheat germ conferring a 18:2:n-6 abundance. Biochemical analyses of diets and mussels were carried out at the beginning and at the end of the 30-day experimental period. Starvation and T. weissflogii based diet poorly affected mussel growth and fatty acid composition which remained unchanged. On the contrary, the wheat germ-based diet increased the condition index and deeply affected the fatty acid profile of all lipid and phospholipid classes. The high dietary 18:2n-6 level drastically reduced tissue content of 20:4n-6, 20:5n-3 and 22:6n-3. The biosynthesis of Non Methylene Interrupted (NMI) dienoic fatty acid appeared to be insensitive to the high input of 16:1n-7 and 18:1n-9 respectively from diet B and C, and to the PUFA shortage of diet C. Nevertheless the two NMI trienoic derivatives, 20:3Δ5,11,14 and 22:3Δ7,13 16, were found higher in C with respect to other groups, presumably due to the high 18:2n-6 content of this diet. 相似文献

Rotifers (Brachionus plicatilis), maintained on baker's yeast, were fed for 24h upon two algal diets, Isochrysis galbana (diet A) and Isochrysis galbana + Nannochloropsis gaditana (diet B). (These algal diets were selected for their potential use as essential fatty acid (EFA) boosters, taking into account the requirements of fish larvae). The effect of these algal diets on total lipid content, lipid classes and fatty acid composition was studied. The total lipid content increased after feeding upon both diets but no significant differences were found between the two types. Neutral lipid and polar lipid contents increased and a positive correlation was observed between the neutral lipids content of rotifers and that of the food supplied. However, the content of polar lipids in rotifers did not depend upon that of the diet. The increase in neutral lipid content was found to be higher in rotifers fed upon diet B, compared to diet A which increased the phospholipid content. Non-enriched rotifers contained only small amounts of polyenoic fatty acids, i.e. 18:3n-6, 18:3n-3, 20:4n-6, 20:5n-3 and 22:6n-3, the contents of which increased significantly by feeding both diets. The EFA composition (20:4n-6, 20:5n-3 and 22:6n-3) of neutral lipids and phopholipids in rotifers reflected the EFA composition of each diet. Diet B-fed rotifers had the highest content in 20:4n-6 and 20:5n-3, whereas rotifers fed diet A and the highest 22:6n-3 content. Both animal and epidemiological studies support an effect of fatty acid composition in the diet on cancer development, in particular on colon cancer. We investigated the modulating effect of supplementation of the diet of female F344 rats with sunflower-, rapeseed-, olive-, or coconut oil on the formation of the promutagenic, exocyclic DNA adducts in the liver, an organ where major metabolism of fatty acids takes place. 1,N(6)-ethenodeoxyadenosine (etheno-dA), 3,N(4)-ethenodeoxycytidine (etheno-dC) and 1,N(2)-propandodeoxyguanosine from 4-hydroxy-2-nonenal (HNE-dGp) were determined as markers for DNA-damage derived from lipid peroxidation products and markers for oxidative stress. 8-Oxo-deoxyguanosine (8-Oxo-dG) was also measured as direct oxidative stress marker. The body weight of the rats was not influenced by the four diets containing the different vegetable oils during the 4-week feeding period. Highest adduct levels of etheno-dC (430 +/- 181 adducts/10(9) parent bases), HNE-dGp (617 +/- 96 adducts/10(9) parent bases) and 8-Oxo-dG (37,400 +/- 12,200 adducts/10(9) parent bases) were seen in rats on sunflower oil diet (highest linoleic acid content). Highest adducts levels of etheno-dA (133 +/- 113 adducts/10(9) parent bases) were found in coconut oil diet (lowest content of linoleic acid). Weakly positive correlations between linoleic acid content in the four diet groups were only observed for levels of HNE-dGp and 8-Oxo-dG. Neither the diet based on olive oil (which contains mainly oleic acid) nor the diet based on rapeseed oil (containing alpha-linolenic acid) exerted any significant protective effect against oxidative DNA damage. Our results indicate that a high linoleic acid diet may contribute to oxidative stress in the liver of female rats leading to a marginal increase in oxidative DNA-damage. 相似文献

Mouse L-M fibroblasts, grown in a serum-free medium, were supplemented with fatty acids of 16 and 18 carbon chain lengths that contain a cyclopentene ring in the ω position. These fatty acids, unnatural to mammalian systems, were incorporated into the major lipid classes of L-M fibroblasts. Supplementation with the cyclopentenyl fatty acids caused an accumulation of neutral glycerolipids and marked inhibition of cell growth. Following the addition of supplement, the cells became more rounded. Of particular interest was the fact that the phospholipid fraction isolated from treated cells contained cyclic fatty acids that accounted for as much as 24% of the total phospholipid acyl groups. Unlike the pattern of distribution displayed by endogenous natural monoenes, the majority of the cyclic acid present was esterified in the sn-1 position of both phosphatidylcholine and phosphatidylethanolamine. The 18-carbon cyclic fatty acid [chaulmoogric acid, 13-(2-cyclopenten-1-yl)tridecanoic acid] was incorporated at the expense of the endogenous C-16:0, C-18:0, and C-18:1 fatty acids of the glycerophospholipids. The esterification altered the ratio of saturated to unsaturated acyl groups in the cellular phospholipids. No biochemical modification of chaulmoogric acid was detected.Our results imply that incorporation of unnatural fatty acid analogs, such as chaulmoogric acid, into cellular membranes would alter the functional properties of biological membranes that are dependent on membrane fluidity and structural organization. 相似文献

Dietary conditioning of juvenile trout changed the acyl chain composition of mitochondrial phospholipids and the oxidative
capacities of muscle mitochondria. Trout were fed three diets differing only in fatty acid (FA) composition. The highly unsaturated
22:6 n-3 (DHA) accounted for 0.4, 14, and 30% of fatty acids in Diets 1, 2 and 3. After 10 weeks of growth, the dietary groups
differed markedly in FA composition of mitochondrial phospholipids, with significant dietary effects for virtually all FA.
Mean mitochondrial DHA levels were 19, 40 and 33% in trout fed Diets 1, 2 and 3. Mitochondrial oxidative capacities changed
with diet, while mitochondrial concentrations of cytochromes and of the adenylate nucleotide translocase (nmol mg1 protein) did not. Mitochondria from fish fed Diet 1 had higher non-phosphorylating (state 4) rates at 5°C than those fed
other diets. When phosphorylating (state 3) rates differed between dietary groups, rates at 5 and 15°C were higher for fish
fed the more unsaturated diets. Stepwise multiple regressions indicated that FA composition could explain much (42–70%) of
the variability of state 4 rates, particularly at 5°C. At 15°C, FA composition explained 16–42% of the variability of states
3 and 4 rates. Similar conclusions were obtained for the complete data set (trout fed diets 1, 2 and 3) and for the data from
trout achieving similar growth rates (e.g. those fed Diets 1 and 2). Neither general characteristics of membrane FA, such
as % saturates, unsaturation index, n-3, n-6 or n-3/n-6 nor levels of abundant unsaturated FA such as DHA or 18:1(n-9 + n-7),
were systematically correlated with mitochondrial capacities even though they differed considerably between trout fed the
different diets. Relatively minor FA (20:5n-3, 20:0, 18:2n-6, 18:3n-3, 18:0 and 15:0) showed better correlations with mitochondrial
oxidative capacities. This supports the concept that acyl chain composition modulates mitochondrial capacities via interactions
between membrane proteins and specific FA of particular phospholipid classes in their microenvironment. 相似文献

Numerous studies have demonstrated that conjugated linoleic acid (CLA) modulates body composition, reducing body fat accumulation in various mammalian species. However, very few studies have been carried out to assess the effect of CLA on previously stored body fat. The aim of the present work was to analyse the effectiveness of trans-10,cis-12 CLA in improving alterations produced by high-fat feeding in body fat and serum parameters when it was included in an energy-restricted diet. For this purpose male Syrian Golden hamsters were fed on high-fat diet for 7 weeks in order to increase their body fat content, and a further 25% energy-restricted diet supplemented or not with 0.5% trans-10,cis-12 CLA for 3 weeks. Adipose tissues, liver and gastrocnemious muscles were dissected and weighed. Adipocyte diameter and number were assessed in epididymal adipose tissue. Total cholesterol, triacylglycerols, non-esterified fatty acids and glucose were measured in serum. Three weeks of energy restriction resulted in a reduction in body weight and white adipose tissue size in all anatomical locations, without changes in liver and gastrocnemious muscle weights. Epididymal adipocyte size was reduced, but total adipocyte number remained unchanged. Serum cholesterol, triacylglycerols and glucose were significantly reduced. No differences were observed between the restricted groups (control and CLA supplemented). In conclusion, under our experimental conditions, the addition of trans-10,cis-12 CLA to the diet does not increase the benefits produced by energy restriction. 相似文献

In a four-week experiment on 60 7-day-old BUT-9 male turkeys the effects of dietary fructooligosaccharides (pure nystose and a fructooligosaccharide mixture) supplemented at 1 and 2%, were studied on ileal and caecal metabolism. The control carbohydrate was cellulose, added also at 1 or 2%. Each dietary treatment consists of 10 birds kept individually. The average degree of polymerisation of the nystose and oligofructose preparation amounted to 2.9 and 4.1, respectively. The addition of nystose significantly decreased the pH value and viscosity in the ileal contents compared with the cellulose treatment. On the other hand, the oligofructose preparation increased the activity of sucrase and lactase in the ileal mucosal by 30-60% and 33-47%, respectively. Both fructan preparations similarly acidified the caecal and colonic digesta (by 0.2-0.4 pH units) as well as diminished the activity of bacterial harmful beta-glucuronidase (by 24-40%), but only nystose caused an enlargement of the caeca and effectively reduced caecal ammonia concentration, especially at a higher dose. Oligofructose supplementation at 2% caused a 3.5-fold increase of bacterial activity of alph- and beta-galactosidase, while 2% nystose resulted in 1.7 and 3 times higher alpha- and beta-glucosidases activities, respectively. Compared to oligofructose, dietary nystose increased propionic and decreased butyric fermentation in caeca. Nystose and oligofructose preparations added at 2% reduced the triacylglycerol concentration in the serum in comparison to the addition of 2% cellulose by 46 and 25%, respectively. Beside the fact that dietary levels of supplementation were of great importance, the results indicated that even small difference in the length of carbohydrate chain may cause different physiological responses. 相似文献

Fatty acid composition of liver and muscle tissues of immature and mature Oncorhynchus mykiss fed on two different diets were determined. Fatty acid analyses were carried out by gas chromatography. Palmitic acid (C16:0),
oleic acid (C18:1 n-9), linoleic acid (C18:2 n-6) and docosahexaenoic acid (C22:6 n-3) were the major components in both liver
and muscle tissues of immature and mature rainbow trout of both sexes. The amounts of C22:6 n-3 were higher in the liver (29.04
± 0.06 − 27.41 ± 0.17%) and muscle (13.05 ± 0.40 − 11.37 ± 0.21%) of immature fish than in mature fish and depended on the
composition of the diet. Results of this study show that fatty acid composition in fish tissues can considerably vary, depending
on the age of fish and their diet. Thus more detailed studies are necessary on the influence of diet on immature and mature

The aim of the experiment was to compare the antioxidative potential of an oat by-product with the effect of vitamin E on the oxidative stability of pork from pigs fed a diet enriched with linseed oil. Thirty-four crossbreed barrows were fed individually from 39 to 109 kg body weight (BW) on one of four diets: a control diet based on barley-triticale-soybean (Diet C), a diet containing an oat byproduct (Diet O), and the same diets supplemented with vitamin E (100 mg/kg diet) (Diets CE and OE, respectively). The oat by-product, comprising oat hulls and bran, was included at 10 and 20% in the grower and finisher diets, respectively. To Diets O and OE, refined rapeseed oil was added to equalise their energy content to Diets C and CE. Compared to Diets C and CE, the inclusion of the oat by-product in Diets O and OE increased the antioxidative capacity of water-soluble and lipid soluble compounds in these diets. Dietary treatment did not influence growth performance, slaughter value, longissimus dorsi (LD) muscle quality measured by nutrient contents, pH, drip loss or colour. Vitamin E supplementation increased the alpha-tocopherol concentration in serum and meat (p < 0.01), and decreased the formation of thiobarbituric acid reactive substances (TBARS) in the fresh and stored LD (p < 0.01). In addition, diets with the oat by-product increased serum alpha-tocopherol concentration (p < 0.01) and decreased the TBARS levels in the fresh and stored LD (p < 0.05), without increasing muscle alpha-tocopherol concentration. The obtained results indicate that the phenolic compounds present in oat by-products have a considerable antioxidant potential and a beneficial effect on the pig organism and oxidative stability of meat. However, dietary inclusion with the oat by-product was not as efficient as supplementation with vitamin E. 相似文献

Animal and epidemiological studies confirm an impact of the fatty-acid composition in the diet on cancer development. We investigated the role of supplementation of the diet of female F344-rats with sunflower, rapeseed, olive or coconut oil on the formation of the promutagenic, exocyclic 1,N2-propanodeoxyguanosine adduct of the main lipid peroxidation product 4-hydroxy-2-nonenal in the mucosa of the glandular stomach, the small intestine, the colon, the whole kidney and the lung. This adduct is considered as the predominant DNA adduct arising from lipid peroxidation. The correlations between adduct levels and the different fatty acids were not uniform for all organs. No clear relationships between fatty acids and adduct levels were found in the colon. Significant positive correlations were observed between linoleic acid, total polyunsaturated fatty acids (PUFAs), vitamin E and DNA adduct levels in the small intestine and in the kidney. The results indicate an increasing effect on cancer risk in these organs as a result of high intake of linoleic acid. Inverse relationships between linoleic acid, PUFA and vitamin E intake and adduct levels were found in the glandular stomach and the lung. We could not confirm a chemopreventive effect of linolenic acid (C-18 omega-3 PUFA) on the formation of adducts in our animal study, as was shown in white blood cells of women in a previous study. A tendency towards a decrease in adduct levels was seen with monounsaturated fatty acids (MUFAs) in all organs except the lung. Saturated fatty acids showed a significant positive correlation with adduct levels in the mucosa of the glandular stomach and a significant inverse correlation in the small intestine. This experiment was conducted to compare the effects of graded levels of camelina meal and/or canola meal on digestibility, performance and fatty acid composition of broiler chickens. A total of 180-day-old male broiler chicks were randomly assigned to one of the six treatments. The control diet was based on wheat and soybean meal and contained 15% canola meal. The experimental diets contained 3%, 6%, 9%, 12% or 15% camelina meal added at the expense of canola meal. Chromic oxide (0.35%) was added to all diets as a digestibility marker. On the morning of day 22, birds were killed by cervical dislocation and their abdominal fat pad was obtained. The apparent total tract digestibility of dry matter and energy as well as nitrogen retention all declined linearly (p?0.01) with increasing levels of dietary camelina meal. Weight gain (p?0.01) and feed intake (p?=?0.08) were linearly reduced as the level of camelina meal in the diet increased. Feed conversion ratio was also negatively affected by camelina meal (p?0.01). Birds fed diets containing 15% camelina meal had significantly higher (p?0.01) levels of polyunsaturated fatty acids, total n-3 fatty acids, total n-6 fatty acids and a significantly lower ratio of n-6 to n-3 fatty acids (p?0.01) than birds fed canola meal. In conclusion, the inclusion of camelina meal in their diet significantly reduced the growth and feed conversion ratio of broilers compared with canola meal. However, the potential to incorporate n-3 fatty acids into carcass tissues may provide some justification for including camelina meal in poultry rations. 相似文献
