Chinese sea bass (Lateolabrax maculatus), an important commercial and recreational fishery species in China, whose natural resources have decreased dramatically for decades due to overfishing and environmental changes. To provide guidelines for the source conservation and management, genetic diversity and population structure of L. maculatus were analyzed based on the mitochondrial cytochrome oxidase subunit I (COI) gene. Totally 192 individuals were sampled from five locations: Qingdao (QD), Chongming (CM), Dongtou (DT), Lieyu (LY) and Fangcheng (FC). After sequencing of a 586 bp fragment of COI gene, 20 haplotypes were defined. H4 (haplotype 4) and H8 were the dominant haplotypes and existed in all populations. Haplotype diversity (Hd) and nucleotide diversity (π) of each population ranged from 0.572 to 0.721 and from 0.00129 to 0.00271, respectively. The highest Hd and π were found in DT and LY populations, while the lowest value of Hd and π were in the QD population. Genetic distance ranged from 0.0031 to 0.0027 within populations and from 0.0018 to 0.0035 between populations. Analysis of molecular variance (AMOVA) revealed that significant genetic divergence was found in QD and FC populations. In addition, neutrality tests and mismatch distribution analysis indicated that this species experienced potential population expansion events.  相似文献

为准确掌握中国沿海口虾蛄(Oratosquilla oratoria)种群遗传结构、合理开发利用其资源,采用线粒体DNA(mt DNA)细胞色素氧化酶Ⅰ(COⅠ)序列分析方法检测东海海域(庙子湖岛、南韭山、大陈岛、南麂岛)口虾蛄种群遗传多样性,并与黄渤海群体和南海群体进行比较分析(基因序列来源于Gen Bank)。经PCR扩增与测序获得100条658 bp的东海海域口虾蛄COⅠ基因序列,基于这些序列分析得到的变异位点数、单倍型数、单倍型多样性指数与核苷酸多样性指数分别为60、60、0.963±0.011和0.005 94±0.000 44,分析认为东海海域口虾蛄具有较高的单倍型多样性和较高的核苷酸多样性。单倍型分子系统树、分子方差分析及两两群体间的遗传分化系数(Fst)分析结果表明,东海海域口虾蛄遗传变异主要来自于群体内(Fst=﹣0.007 78,P0.05),各地理群体间遗传分化不显著,Fst值范围为﹣0.016 53~﹣0.009 08(P0.05),它们可能进行了一定程度的基因交流;通过与黄渤海群体及南海群体基因序列比较分析,口虾蛄东海群体、黄渤海群体与南海群体遗传变异主要来自于群体间(Fst=0.849 71,P0.01),且单倍型分子系统树存在2个显著分化的单倍型类群。东海群体与黄渤海群体间存在显著的遗传分化(Fst=0.884 58,P0.01),而与南海群体间不存在显著的遗传分化(Fst=0.020 44,P0.05),这种遗传结构模式可能与历史上的气候变化及所处海域海洋环境条件相关。建议今后对中国沿海口虾蛄资源进行开发利用时,将黄渤海群体看作一个管理单元,东海群体与南海群体看作一个管理单元。  相似文献

The miiuy croaker (Miichthys miiuy) is an economically important fish species in China. However, information about population structure in this species is limited. In this study, the genetic diversity and population structure of four populations of Miichthys miiuy were investigated using a mitochondrial DNA (mtDNA) cytochrome c oxidase subunit I (COI) segment of 599 bp. A total of 35 polymorphic sites, and 31 haplotypes were defined in 109 individuals. High levels of haplotype diversity (0.731 ± 0.047) and low nucleotide diversity (0.00278 ± 0.0019) were observed in miiuy croaker populations. The results of AMOVA and pairwise fixation index (FST) analysis did not indicate any significant genetic differentiation among populations obtained from four sites (P > 0.05). In addition, no significant genealogical branches or clusters were recognized in the NJ tree. Analysis of neutral evolution and mismatch distribution suggested that the miiuy croaker might have experienced a recent population expansion.  相似文献

Nine members of the genus Taenia (Taenia taeniaeformis, Taenia hydatigena, Taenia pisiformis, Taenia ovis, Taenia multiceps, Taenia serialis, Taenia saginata, Taenia solium and the Asian Taenia) were characterised by their mitochondrial NADH dehydrogenase subunit 1 gene sequences and their genetic relationships were compared with those derived from the cytochrome c oxidase subunit I sequence data. The extent of inter-taxon sequence difference in NADH dehydrogenase subunit 1 (5.9–30.8%) was usually greater than in cytochrome c oxidase subunit I (2.5–18%). Although topology of the phenograms derived from NADH dehydrogenase subunit 1 and cytochrome c oxidase subunit I sequence data differed, there was concordance in that T. multiceps, T. serialis (of canids), T. saginata and the Asian Taenia (of humans) were genetically most similar, and those four members were genetically more similar to T. ovis and T. solium than they were to T. hydatigena and T. pisiformis (of canids) or T. taeniaeformis (of cats). The NADH dehydrogenase subunit 1 sequence data may prove useful in studies of the systematics and population genetic structure of the Taeniidae.  相似文献

Eleutheronema tetradactylum is an economically important fish species in China water. To investigate the genetic diversity and describe population structure of it, an 1151 base pair (bp) fragment of the mitochondrial DNA Cytb sequence was analyzed in 120 individuals from four populations in the East China Sea and the South China Sea. A total of 16 haplotypes were defined by 24 variable nucleotide sites. High level of haplotype diversity and low nucleotide diversity were observed in all populations. The results of AMOVA detected that 89.44% of the genetic variation occurred within populations. Significant genetic differentiations were detected among populations (0.05097, P < 0.05), but no large-scale regional differences were detected. Analysis of neutral evolution and mismatch distribution suggested no recent population expansion happened. The present results provided new information for genetic assessment, fishery management and conservation of this species.  相似文献

Partial sequences of mitochondrial DNA 16S rDNA and COI genes (395 bp and 498 bp respectively) were sequenced from samples of ten cultured populations of Macrobrachium rosenbergii (Giant Freshwater Prawn – GFP) in Zhejiang, Guangdong and Guangxi Provinces in China and two wild populations of GFP from Mekong River and Dongnai River in Viet Nam. Five haplotypes of 16S rDNA were identified in the 360 samples. The wild populations displayed nucleotide diversity (π) of 0.0008 and 0.0003, and genetic diversity (h) of 0.3030 and 0.1310 in the Mekong River and Dongnai River respectively. The cultured populations displayed no significant genetic diversity. COI sequences identified 17 haplotypes based on 21 polymorphic sites. At this marker, the 12 populations showed a range of h from 0.1290 to 0.6940 and π from 0.0003 to 0.0073. The largest genetic distance (Da) among the 12 populations was 0.0065 (between ZJB and BT/DN populations) and the lowest Da was 0.0003 (between GDD and GDA populations). The wild populations had higher genetic diversity than the cultured populations, but three of cultured populations from Zhejiang (ZJA, ZJB and ZJC) had π higher than wild populations, because they originated from Thailand, Bangladesh and the Mekong River in Viet Nam.  相似文献

The first hypervariable (HV1) region of mitochondrial DNA (mtDNA) of two popular Nigerian goat breeds: West African Dwarf (WAD) (n = 35) and Red Sokoto (RS) (n = 37) and one exotic breed: Kalahari Red (KR) (n = 38) imported from South Africa were sequenced to investigate sequence diversity, genetic structure, origin, and demographic history of the populations. A total of 68 polymorphic sites were found in 110 sequences that grouped into 68 haplotypes. Average haplotype and nucleotide diversities for all breeds were 0.982 ± 0.005 and 0.02350 ± 0.00213, respectively. Phylogenetic analysis revealed two mtDNA lineages (A and B). Lineage A was predominant and included all haplotypes from WAD and RS and 5 out of 11 haplotypes of KR goats. The remaining haplotypes (6 Amills M, Ramírez O, Tomàs A, et al. Mitochondrial DNA diversity and origins of South and Central American goats. Animal Genetics 2009; 40(3): 315322.[Crossref], [PubMed], [Web of Science ®] [Google Scholar]) of KR belong to lineage B. The analysis of molecular variance revealed a high-within breed genetic variance of 82.4% and a low-between breed genetic variance of 17.6%. The three breeds clustered with Capra aegagrus as their wild ancestor. Mismatch distribution analysis showed that WAD, RS and haplogroup A have experienced population expansion events. The study has revealed very high diversity within the three breeds which are not strongly separated from each other based on mtDNA analysis. The information obtained on the genetic structure of the breeds will be useful in planning improvement and conservation programs for the local populations.  相似文献

We examined the natural population structure of the Turkish hamster (Mesocricetus brandti) by analysing partial mitochondrial sequences of the control region and the cytochrome b gene. Evolutionary lineages were defined on haplotype clusters in genetic trees and a median-joining network. Most significant divergence events in M. brandti nested in the lower Pleistocene. Gene flow prevented spatial genetic differentiation among most populations contrasting previous ideas about potential subspeciation in Anatolia. None of the mitochondrial lineages showed significant signs of recent expansion indicating relatively stable ecological conditions during recent population history. Furthermore, we discussed aspects of the evolution of M. brandti and the genus Mesocricetus in the context of available fossils.  相似文献

To understand levels of population differentiation in Pallas’s squirrel (Callosciurus erythraeus) in fragmented habitats, we collected 83 samples from three patches of artificial forest in Hongya County, Sichuan Province, China. Sample numbers from each patch were as follows: 16 from Hanwang (HW), 27 from Muchansi (MCS) and 40 from Yanyandong (YYD). The mitochondrial DNA control region was sequenced and 18 haplotypes were observed. Our results showed that haplotype diversities of the three C. erythraeus populations were similar (0.771, 0.791 and 0.733). Fixation indices (Fst) of pairwise populations were between 0.21 and 0.31, and the estimated gene flow (Nm) was between 1 and 2. Analysis of molecular variance (AMOVA) showed that most molecular variation occurred within populations (74.82%); variances among populations were small but there was significant genetic differentiation. In addition, the neighbour-joining (NJ) tree showed three clades in the phylogenetic tree for population genetic structure. This was confirmed by the median-joining haplotype network. Furthermore, analysis of isolation by distance (IBD) showed that genetic differentiation among the three populations was positively related to geographical distance. However, tests of neutrality and the observed mismatch distribution of pairwise differences between sequences indicated that C. erythraeus populations were relatively stable in the past.  相似文献

The genetic structure of marine populations is greatly influenced by the dispersal ability of the organisms, and it is intuitive that a limited dispersal capability would result in greater genetic differentiation. Octopus variabilis is a typical cephalopod species that shows limited dispersal potential, and previous genetic surveys by a mitochondrial DNA analyses revealed high levels of genetic differentiation among its populations, suggesting the existence of cryptic or subspecies. To test this hypothesis, amplified fragment length polymorphisms (AFLPs) and the cytochrome oxidase II (COII) gene sequence were used to re-evaluate the spatial genetic structure of five populations of O. variabilis along the coast of China. Four distinct groups were identified within these five populations: the Dalian + Qingdao, Zhoushan, Wenzhou, and Dongshan groups. A deep genetic break among the Dongshan, Wenzhou, and other two groups were specifically recognized by both genetic markers. A considerable number of distinct alleles for AFLP and a deep divergence of 13.9–19.4% in the COII gene sequences were found among them, suggesting the occurrence of potential cryptic or subspecies of O. variabilis. A genetic structure of isolation by distance was identified in O. variabilis by a Mantel test, with the geographic distance explaining 62% of the variation in genetic differentiation. This result suggests that gene flow is geographically restricted for this species and that limited dispersal may be the main reason for the genetic differentiation among O. variabilis populations. The present results indicate that special care should be taken in future fishery exploitation and conservation efforts for this species and that conservation management should include populations representing all lineages.  相似文献

In this study, we isolated 10 novel polymorphic microsatellite markers in Mytilus edulis by using the magnetic beads enrichment procedure. The characteristics of these loci were estimated by using a sample of 32 individuals of M. edulis. The number of alleles at 10 polymorphic microsatellite loci ranged from 2 to 15 with an average of 5.667. HO and HE ranged from 0.2667 to 1.0000 (0.6800 in average) and from 0.4723 to 0.9226 (0.6190 in average), The PIC value of 6 loci was more than 0.5, and that of the other 4 was between 0.25 and 0.50, Significant deviation from Hardy–Weinberg Equilibrium was observed at ME8, ME115 and ME153 after Bonferroni correction (P < 0.004, adjusted value), which possibly was due to the presence of null alleles. This study will be useful for the analysis of population genetic diversity, and the management of this important M. edulis resource.  相似文献

The lycaenid butterfly Zizina emelina emelina (de l’Orza) (previously Zizina otis emelina; Lepidoptera, Lycaenidae) has populations distributed across Japan and Korea. Zizina emelina is listed on Japan’s Red List as an endangered species, owing to the loss of its principal plant food and habitat. Researchers previously investigated the genetic diversity and structure in Japanese populations by using mitochondrial and nuclear DNA analyses alongside their Wolbachia infection status to consider the conservation strategy. In this study, we investigated the genetic variation and Wolbachia infection status of Korean populations and compared our findings with these traits in the Japanese populations. Four novel mitochondrial haplotypes were found in the Korean populations, whereas nuclear DNA analysis revealed one new haplotype and another that matched the one reported previously in Japan. The two known strains of Wolbachia in the Japanese populations were also found in the Korean ones. Mitochondrial DNA analysis demonstrated that no gene flow had occurred between the Japanese and Korean populations. The fact that they share the same nuclear haplotype and Wolbachia strains, however, indicates a low level of population differentiation between the two.  相似文献

Leptosphaeria maculans is a serious concern for canola production worldwide. For effective disease management, knowledge of the pathogen's genetic variability and population structure is a prerequisite. In this study, whole-genome sequencing was performed for 162 of 1590 L. maculans isolates collected in the years 2007–2008 and 2012–2014 in Western Canada. DNA variants in genome-wide and specific regions including avirulence (Avr) genes were characterized. A total of 31,870 high-quality polymorphic DNA variants were used to study L. maculans genetic diversity and population structure. Cluster analysis showed that 150 isolates were clustered into 2 main groups and 4 subgroups by DNA variants located in either Avr or small secreted protein-encoding genes and into 2 main groups and 6 subgroups by genome-wide variants. The analysis of nucleotide diversity and differentiation also confirmed genetic variation within a population and among populations. Principal component analysis with genome-wide variants showed that the isolates collected in 2012–2014 were more genetically diverse than those collected in 2007–2008. Population structure analysis discovered three distinct sub-populations. Although isolates from Saskatchewan and Alberta were of similar genetic composition, Manitoba isolates were highly diverse. Genome-wide association study detected DNA variants in genes AvrLm4-7, Lema_T86300, and Lema_T86310 associated with the years of collection.  相似文献

The genetic polymorphisms of Echinococcus spp. in the eastern Tibetan Plateau and the Xinjiang Uyghur Autonomous Region were evaluated by DNA sequencing analyses of genes for mitochondrial cytochrome c oxidase subunit 1 (cox1) and nuclear elongation factor-1 alpha (ef1a). We collected 68 isolates of Echinococcus granulosus sensu stricto (s.s.) from Xinjiang and 113 isolates of E. granulosus s. s., 49 isolates of Echinococcus multilocularis and 34 isolates of Echinococcus shiquicus from the Tibetan Plateau. The results of molecular identification by mitochondrial and nuclear markers were identical, suggesting the infrequency of introgressive hybridization. A considerable intraspecific variation was detected in mitochondrial cox1 sequences. The parsimonious network of cox1 haplotypes showed star-like features in E. granulosus s. s. and E. multilocularis, but a divergent feature in E. shiquicus. The cox1 neutrality indexes computed by Tajima’s D and Fu’s Fs tests showed high negative values in E. granulosus s. s. and E. multilocularis, indicating significant deviations from neutrality. In contrast, the low positive values of both tests were obtained in E. shiquicus. These results suggest the following hypotheses: (i) recent founder effects arose in E. granulosus and E. multilocularis after introducing particular individuals into the endemic areas by anthropogenic movement or natural migration of host mammals, and (ii) the ancestor of E. shiquicus was segregated into the Tibetan Plateau by colonising alpine mammals and its mitochondrial locus has evolved without bottleneck effects.  相似文献

The golden takin is an endangered species, listed as a First Grade Protected animal and found only in the Qinling Mountains in China. A great deal of research on the golden takin's living habitat, population size, and home range has been conducted. Here, we employed sequence analysis of the mitochondrial DNA control region to study the genetic diversity of the golden takin from three separate nature reserve parks in the Qinling Mountains. We also compared the results of our study with previously published data on the genetic diversity of mixed takin species located in the Qinling Mountains and the Minshan area. Based on 62 sampled golden takin individuals, we found an overall mean genetic haplotype diversity of 0.687. There is no significant geographic genetic diversity across different golden takin populations within the Qinling Mountains. However, we did show significant diversity between golden takin from the Qinling Mountains and from Minshan. These original data provide a foundation for the genetic diversity of golden takin, and will yield comprehensive information for better supporting the management in the national reserve parks.  相似文献

Small yellow croaker is one of the most important fishery species in China. The mass–scale artificial propagation of this fish species was initially developed in 2015 with the aim of facilitating the fish production stock enhancement and aquaculture programs in the future. In the present study, the wild broodfish and its corresponding progeny along with three other wild populations were sampled and subjected to sequence analysis of the mitochondrial cytochrome c oxidase subunit I gene. The genetic diversity and population genetic structure were evaluated with a total sample size of 141 individuals representing the populations of the Yellow Sea (Qingdao and Lvsi populations) and the East China Sea (Xiangshan and Ningde populations). The wild populations were characterized by high haplotype diversity (0.925–0.976) and low nucleotide diversity (0.376%–0.560%). The hierarchical analysis of molecular variance (AMOVA) analysis and the values of pairwise Ф-statistics (ФST) indicated non-significant genetic differentiation among the four wild populations. The hatchery stock XSH exhibited lower indices of genetic diversity compared with the wild populations that could be attributed to the small effective population size. The findings of the present study have valuable insight to the sustainable management and utilization of this resource.  相似文献

捕食性天敌是害虫生物防治中的重要角色,如何精确评估捕食性天敌的捕食作用是生态学中尚待研究的问题之一。为探索柑橘园捕食性天敌对柑橘木虱Diaphorina citri的捕食作用,本研究根据柑橘木虱细胞色素氧化酶I(COI)的基因序列设计了PCR特异性引物,并应用DNA标记方法检测捕食性天敌对柑橘木虱的捕食作用。物种特异性检验结果发现:引物对A对柑橘木虱DNA扩增效果明显,对其他种类的害虫及其天敌不具有扩增作用。田间检测结果表明:在所检测的8类群20种捕食性天敌中,龟纹瓢虫Propylea japonica、斜纹猫蛛 Oxyopes sertatus、丽草蛉Chrysopa formosa对柑橘木虱具有明显的捕食作用,其阳性反应检出率分别达到58.06 %,57.89 %,48.00%。室内龟纹瓢虫对柑橘木虱的消化检测表明:龟纹瓢虫成虫取食1头柑橘木虱4龄若虫或成虫时,检测阳性率均随消化时间的延长而下降,其消化半衰期分别为4.93 h 和12.98 h。采用DNA标记技术探索捕食作用,为筛选柑橘木虱的优势天敌拓展了新的思路,也为进一步田间生物防治柑橘木虱奠定了基础。  相似文献

The topmouth culter (Culter alburnus) is one of the most commercially important freshwater fish species inhabiting China. However, very limited information is available regarding its genetic diversity and population structure, thus hindering the effective management of this fish stock. Understanding the genetic diversity of wild and cultured topmouth culter populations is highly relevant for successful hatchery management. This study evaluated the genetic diversity and structure of five wild and two cultured populations of topmouth culter in China by using microsatellites and mitochondrial DNA. The genetic diversity of wild populations was found to be lower than that of cultured populations. This finding indicates that wild topmouth culter resources should be protected to prevent further degeneration and extinction. Moreover, it demonstrated that cultured populations have greater breeding potential than wild ones. Subdivisions among wild populations were observed, which should be considered as different units for conservation and hatchery management.  相似文献
