Sex determination and hatching success in sea turtles is temperature dependent and as a result global warming poses a threat to sea turtles. Warmer sand temperatures may skew sea turtle population′s sex ratios towards predominantly females and decrease hatching success. Therefore, understanding the rates at which sand temperatures are likely to increase as climate change progresses is warranted. We recorded sand temperature and used historical sea surface and air temperature to model past and to predict future sand temperature under various scenarios of global warming at key sea turtle nesting grounds (n = 7) used by the northern Great Barrier Reef (nGBR) green turtle, Chelonia mydas, population. Reconstructed temperatures from 1990 to the present suggest that sand temperatures at the nesting sites studied have not changed significantly during the last 18 years. Current thermal profile at the nesting grounds suggests a bias towards female hatchling production into this population. Inter-beach thermal variance was observed at some nesting grounds with open areas in the sand dune at northern facing beaches having the warmest incubating environments. Our model projections suggest that a near complete feminization of hatchling output into this population will occur by 2070 under an extreme scenario of climate change (A1T emission scenario). Importantly, we found that some nesting grounds will still produce male hatchlings, under the most extreme scenario of climate change, this finding differs from predictions for other locations. Information from this study provides a better understanding of possible future changes in hatching success and sex ratios at each site and identifies important male producing regions. This allowed us to suggest strategies that can be used at a local scale to offset some of the impacts of warmer incubating temperatures to sea turtles. 相似文献

Complex population structure can result from either sex-biased gene flow or population overlap during migrations. Loggerhead turtles (Caretta caretta) have both traits, providing an instructive case history for wildlife management. Based on surveys of maternally inherited mtDNA, pelagic post-hatchlings show no population structure across the northern Atlantic (phi(ST) < 0.001, P = 0.919), subadults in coastal habitat show low structure among locations (phi(ST) = 0.01, P < 0.005), and nesting colonies along the southeastern coast of the United States have strong structure (phi(ST) = 0.42, P < 0.001). Thus the level of population structure increases through progressive life history stages. In contrast, a survey of biparentally inherited microsatellite DNA shows no significant population structure: R(ST) < 0.001; F(ST) = 0.002 (P > 0.05) across the same nesting colonies. These results indicate that loggerhead females home faithfully to their natal nesting colony, but males provide an avenue of gene flow between regional nesting colonies, probably via opportunistic mating in migratory corridors. As a result, all breeding populations in the southeastern United States have similar levels of microsatellite diversity (H(E) = 0.70-0.89), whereas mtDNA haplotype diversity varies dramatically (h = 0.00-0.66). Under a conventional interpretation of the nuclear DNA data, the entire southeastern United States would be regarded as a single management unit, yet the mtDNA data indicate multiple isolated populations. This complex population structure mandates a different management strategy at each life stage. Perturbations to pelagic juveniles will have a diffuse impact on Atlantic nesting colonies, mortality of subadults will have a more focused impact on nearby breeding populations, and disturbances to adults will have pinpoint impact on corresponding breeding populations. These findings demonstrate that surveys of multiple life stages are desirable to resolve management units in migratory marine species. 相似文献

The loggerhead sea turtle (Caretta caretta), one of the three sea turtle species inhabiting the Mediterranean Sea, is an endangered species. However, although frequently
treated in marine animal rescue centers, and the subject of several studies in literature, as yet, few studies have been conducted
on large numbers of loggerhead sea turtles in order to establish physiological reference ranges that enable the identification
of pathological values. The lack of studies on reference parameters probably depends on the fact that marine turtles treated
in wildlife rescue centers are usually in a critical conditions, thus precluding the collection of data on healthy animals
and compromising the reliability of any data obtained from them. The present biological study was therefore conducted in order
to obtain a database from healthy animals in natural conditions. Serum biochemistry and serum protein electrophoresis were
performed on blood samples obtained from 65 healthy adult loggerhead sea turtles captured and delivered to the Sea Turtle
Rescue Center of Linosa (Italy); the blood samples were collected during the clinical examination of rescued animals. Laboratory
analyses of serum samples were made in order to establish reference parameters, commonly required for laboratory diagnoses
in mammals and diseased animals.

Heat shock proteins (HSPs) constitute a superfamily of molecular chaperones that are rapidly biosynthesized in response to various biotic and abiotic factors. In this study, we first cloned the full-length HSP70 gene of the Eastern honeybee Apis cerana. Then, using real-time quantitative PCR, we explored HSP70 expression profiles in drones at different developmental stages, ages, and reproductive statuses (with and without semen). The full-length HSP70 cDNA is 2421 bp, including a 1953-bp open reading frame (ORF) that encodes a polypeptide of 650 amino acids. The HSP70 gene consists of one intron and two exons. The phylogenetic analysis revealed that the HSP70 genes of A. cerana and Apis mellifera are the most closely related. We observed HSP70 expression at all selected developmental stages and detected the highest expression in pupae with an unpigmented body cuticle and brown eyes (Pb) and much lower expression in larvae hatched within 72 h. In adult drones of different ages, the highest expression level of HSP70 was observed in 16-day-old drones; significantly lower accumulation of HSP70 mRNA was detected in 4-day-old drones. There was no significant difference in HSP70 expression between drones with and without semen captured at the entrance, while the HSP70 gene expression level strikingly differed between drones captured at the entrance and the drones collected within the hive. Our study suggests that HSP70 might play a critical role in drone development and during reproductive mating events. 相似文献

Marine turtles are large reptiles that compensate for high juvenile mortality by producing hundreds of hatchlings during a long reproductive lifespan. Most hatchlings are taken by predators during their migration to, and while resident in, the open ocean. Their survival depends upon crypticity, minimizing movement to avoid detection, and foraging efficiently to grow to a size too difficult for predators to either handle or swallow. While these behavioral antipredator tactics are known, changes in morphology accompanying growth may also improve survival prospects. These have been only superficially described in the literature. Here, we compare the similarities and differences in presumed morphological defenses of growing loggerhead (Caretta caretta) and Kemp's ridley (Lepidochelys kempii) posthatchlings, related species that differ in growth rate, timing of habitat shift (the return from oceanic to neritic locations), and size at maturity. In both species, vertebral spination and carapace widening increase disproportionally as small turtles grow, but later in ontogeny, the spines regress, sooner in ridley than in loggerhead turtles. Carapace widening occurs in both species but loggerheads are always longer than they are wide whereas in Kemp's ridley turtles, the carapace becomes as wide as long. Our analysis indicates that these changes are unrelated to when each species shifts habitat but are related to turtle size. We hypothesize that the spines function in small turtles as an early defense against gape‐limited predators, but changes in body shape function throughout ontogeny—initially to make small turtles too wide to swallow and later by presenting an almost flat and hardened surface that large predators (such as a sharks) are unable to grasp. The extremely wide carapace of the Kemp's ridley may compensate for its smaller adult size (and presumed greater vulnerability) than the loggerhead. J. Heat shock protein 70 (HSP70) is one of the most abundant and best characterized heat shock protein family that consists of highly conserved stress proteins, expressed in response to stress, and plays crucial roles in environmental stress tolerance and adaptation. The present study was conducted to identify major types of genes under the HSP70 family and to quantify their expression pattern in heat- and cold-adapted Indian goats (Capra hircus) with respect to different seasons. Five HSP70 gene homologues to HSPA8, HSPA6, HSPA1A, HSPA1L, and HSPA2 were identified by gene-specific primers. The cDNA sequences showed high similarity to other mammals, and proteins have an estimated molecular weight of around 70 kDa. The expression of HSP70 genes was observed during summer and winter. During summer, the higher expression of HSPA8, HSPA6, and HSPA1A was observed, whereas the expression levels of HSPA1L and HSPA2 were found to be lower. It was also observed that the expression of HSPA1A and HSPA8 was higher during winter in both heat- and cold-adapted goats but downregulates in case of other HSPs. Therefore, both heat and cold stress induced the overexpression of HSP70 genes. An interesting finding that emerged from the study is the higher expression of HSP70 genes in cold-adapted goats during summer and in heat-adapted goats during winter. Altogether, the results indicate that the expression pattern of HSP70 genes is species- and breed-specific, most likely due to variations in thermal tolerance and adaptation to different climatic conditions. 相似文献

Age and growth are important parameters for better understanding of life history and population dynamics of animal species, as well as for formulating management strategies. However, these data are difficult to obtain for sea turtles because of overall slow growth, delayed maturation and highly migratory behaviour. The loggerhead sea turtle, Caretta caretta, is a widely distributed species, globally listed as endangered. Although the species has been well‐studied in some regions, little is known about various aspects of its biology in other populations, such as those in the waters of the western South Atlantic Ocean, especially outside nesting areas. To address age and growth, loggerhead turtles found dead stranded on the northern coast of the state of Rio Grande do Sul during a period of 16 years (1994–2010) were utilized for estimation of age and growth rates using skeletochronology. The individuals analyzed were predominantly neritic juveniles, ranging from 53 to 101 cm curved carapace length (CCL; mean = 71 cm), with estimated ages between 10 and 29 years (mean = 15 years). Mean estimated annual growth rate was 2.1 cm CCL year?1 (1.9 cm SCL yr?1), showing large variation among individuals and between successive years in the same individual. Generalized additive mixed models analysis indicated that growth response was influenced by age, CCL and year. The results demonstrated that the southern coast of Brazil is an important area for the development of neritic juveniles of this species, which appear to recruit to this region beginning at about 12 years of age and sizes greater than 55 cm CCL. 相似文献

Thermal stress has been shown to result in decreased egg production, decreased eggshell quality, and ultimately millions of dollars in losses to the industry. Therefore, there are many factors to consider when implementing genetic selection programs aimed at improving egg production under tropical conditions. So, trial is trying to improve the productivity and eggshell quality traits of the Fayoumi chicken under high ambient temperatures via selection programs and gene expression. In the present study, day-old Fayoumi chicks were raised either under normal temperature (control) or conditions of thermal stress (the heated group). At 35 weeks, male and female chickens from the control group were mated randomly and females selected for higher egg production and eggshell strength were mated to male siblings to obtain the progeny of the first generation (F1). F1 birds were further selected and mated to obtain the progeny of the second generation. Our results show that egg production and eggshell strength traits improved over successive generations via selection under conditions of heat stress. Furthermore, the reduction in egg production and eggshell strength as a result of heat stress declined from one generation to the next in birds selected for good heat tolerance, and an inverse relationship was observed between the OC-17 and eggshell strength. Additionally, levels of HSP90 and gene expression increased in the two successive generations, indicating that both productivity and heat tolerance were enhanced due to selection in birds raised under conditions of thermal stress. Moreover, generation exerted an important effect on this trait. Thus, desirable traits such as improved heat tolerance in producing lines were observed in Fayoumi chickens exposed to conditions of thermal stress via selection. Therefore, modern advances in studies of poultry breeding and genetics, such as gene expression studies, should be examined. 相似文献

Electroretinographic recordings were made from hatchling loggerhead and leatherback sea turtle eyecup preparations to generate dark-adapted spectral sensitivity curves. Both species were maximally sensitive to wavelengths between 500 and 540 nm, with a secondary peak near 380 nm. The spectral sensitivity curve for leatherbacks was attenuated at the long wavelength end of the spectrum relative to that of the loggerheads. This difference may reflect adaptations to lighting available at the relatively shallow (loggerhead) versus deeper (leatherback) sites where each species forages. The broad spectrum of wavelengths detected by both species (near UV to yellow–orange) indicates that vision is likely mediated by more than one photopigment, potentially rendering these turtles capable of color vision. 相似文献

polymerase chain reaction (PCR) conditions to amplify 11 novel tetranucleotide microsatellite loci from the loggerhead sea
turtle. We tested primers using samples from 22 females that nested at Melbourne Beach, Florida (USA). Primer pairs yielded
an average of 11.2 alleles per locus (range of 4–24), an average observed heterozygosity of 0.83 (range 0.59–0.96), and an
average polymorphic information content of 0.80 (range 0.62–0.94). We also demonstrate the utility of these primers, in addition
to primers for 15 loci previously described, for amplifying microsatellite loci in four additional species representing the

为了检测B型烟粉虱和温室白粉虱在受到温度胁迫时热激基因Hsp70的差异表达谱,克隆了2种粉虱Hsp70基因的全长cDNA序列,并用实时定量PCR的方法分析了在不同温度条件下该基因的表达谱。结果表明:B型烟粉虱和温室白粉虱的Hsp70基因BTH-sp70和TVHsp70(GenBank登录号分别为DQ093385和EU861391),在5’-非编码区(UTR)中有类似TATA-box样元件;在3’-UTR中有poly(A)信号AATAA和富含A-T区;根据基因开放阅读框序列(ORF)推导的氨基酸序列均含有全部3个Hsp70蛋白家族高度保守的基序。在整个检测的温度范围内(-19℃~46℃),诱导温室白粉虱Hsp70表达的起始温度(Ton)或最大温度(Tmax)要比B型烟粉虱低2.5℃~7.5℃。这些结果表明,所克隆的基因属于有功能的温度诱导型Hsp70基因;在基因表达水平上,温室白粉虱比B型烟粉虱更耐冷,而后者更耐热;Hsp70的Ton(或者Tmax)能代表这2种粉虱的温度耐受能力。本研究结果在一定程度上解释了自然界中这2种粉虱种群地理分布和季节发生差异的原因。 相似文献

Fertility losses in male mice occur approximately 18-28 d after heat stress. The objective of this study was to identify gene expression differences in males highly versus lowly fertile after heat stress. Mature male mice were exposed to heat stress (35 ± 1 °C; n = 50) or thermoneutral (21 ± 1 °C; n = 10) conditions for 24 h (Day 0) and hemicastrated (Day 1) to collect tissue for gene expression analyses. Males were subjected to a mating test from Days 18 to 26 when variation in fertility was anticipated. A fertility index was used to rank heat-stressed males and identify those males resistant and susceptible to heat stress, respectively. Microarray analyses were conducted on testis tissues from control (n = 5), heat stress resistant (n = 5), and heat stress susceptible (n = 5) males, and 225 genes were observed to be differentially expressed (P < 0.05), including genes involved in chaperone (Canx, Hspcb1, and Tcp1) and catalytic (Fkpb6, Psma7, and Idh1) activity. Expression patterns of these genes were confirmed using real-time RT-PCR. Male progeny from selected sires were similarly divergent in fertility after heat stress. Testicular expression levels of Canx, Hspcb, and Tcp1 genes were determined in these progeny. Hspcb expression was moderately heritable (0.31 ± 0.25); however, expression patterns of Canx and Tcp1 were not heritable. 相似文献

Heat stress may adversely affect physiochemical and immune responses of livestock and alter biological functions. The comfort or thermoneutral zone for livestock, which has long been a subject of research, mainly depends on species, breed, and health. Heat stress is associated with impaired livestock productivity due to reductions in feed intake, growth rates and immunity and changes in blood constituents and biological pathways. In ruminants, elevated temperatures have deleterious consequences on protein synthesis. Exposure of ruminant animals to elevated temperatures may induce release of heat shock proteins (HSPs); HSPs usually enter the blood circulation during tissue damage and causes cell necrosis or death. Additionally, hyperthermia is associated with augmented production of cellular reactive oxygen species (ROS), which cause protein degradation and further decrease protein synthesis by preventing protein translation. Moreover, it has been suggested that high environmental temperatures lead to increased inflammatory signalling in tissues via activation of the nuclear factor kappa B (NF-κB) and tumor necrosis factor alpha (TNF-α) pathways as well as via alteration of skin colour gene (melanocortin 1 receptor (MC1R) and premelanosome protein (PMEL)) expression. Previous proteomics analyses have suggested that heat stress can reduce adenosine triphosphate (ATP) synthesis, alter gluconeogenesis precursor supply, and induce lipid accumulation in the liver with subsequent disturbance of liver structure. This review focuses on the scientific evidence regarding the impact of heat stress on immune and inflammatory responses, antioxidant status, stress biomarkers, skin colour gene (PMEL and MC1R) expression and proteomic profiles in ruminants. 相似文献

Exploring a trade‐off between quantity and quality of offspring allows differences in the fitness between alternative life histories to be accurately evaluated. We addressed the mechanism that maintains alternative life histories (small oceanic planktivores vs. large neritic benthivores) observed in a loggerhead sea turtle (Caretta caretta) population, which has been suggested to be environmental, based on the lack of genetic structure and a large difference in reproductive output. We examined whether maternal foraging habitat affects offspring quality, by measuring the morphology, emergence success, and righting response of hatchlings following incubation in a common open sand area over the whole nesting season at Yakushima Island, Japan, and by recording early growth and survival of offspring that were reared in a common environment at a Japanese aquarium. Furthermore, we tested whether sea turtles adjust egg size in response to temporal shifts of the incubation environment. There were no significant differences in any hatchling traits between oceanic and neritic foragers (which were classified by stable isotope ratios), except for clutches laid during the warmest period of the nesting season. There were also no significant differences in the growth and survival of offspring originating from the two foragers. The size of eggs from both foragers significantly increased as the season progressed, even though the rookery had heavy rainfall, negating the need to counteract heat‐related reduction in hatchling morphology. In comparison, the sizes of adult body and clutches from both foragers did not vary significantly. The results further support our previous suggestions that the size‐related foraging dichotomy exhibited by adult sea turtles does not have a genetic basis, but derives from phenotypic plasticity. Adjustment in reproductive investment may be associated with: (1) predation avoidance, (2) founder effect, and/or (3) annual variation in egg size. 相似文献
