Postprandial glycemic responses to meals are inhibited by polyphenol-rich plant foods. Combinations of polyphenols may be particularly effective through complementary mechanisms. A randomized, controlled, double-blinded cross-over trial was conducted in healthy volunteers to test the hypothesis that apple and blackcurrant polyphenol-rich drinks would reduce postprandial blood glucose concentrations. Secondary outcomes included insulin and glucose-dependent insulinotropic polypeptide (GIP) secretion. Twenty men (mean age 26 y, SD 8) and 5 postmenopausal women (mean age 57 y, SD 3) consumed a placebo drink (CON) and 2 polyphenol-rich drinks containing fruit extracts: either 1200 mg apple polyphenols (AE), or 600 mg apple polyphenols+600 mg blackcurrant anthocyanins (AE+BE), in random order with a starch and sucrose meal. Incremental areas under the curve (iAUC) for plasma glucose concentrations were lower following AE+BE over 0–30 and 0–120 min compared with CON; mean differences (95% CI) −32 mmol/L·min (−41, −22, P<.0005) and −52 mmol/L min (−94, −9, P<.05), respectively. AE significantly reduced iAUC 0–30 min (mean difference −26 mmol/L min, −35, −18, P<.0005) compared with CON, but the difference over 120 min was not significant. Postprandial insulin, C-peptide and GIP concentrations were significantly reduced relative to CON. A dose response inhibition of glucose transport was demonstrated in Caco-2 cells, including total and GLUT-mediated transport, and SGLT1-mediated glucose transport was strongly inhibited at all doses in Xenopus oocytes, following 10 min incubation with 0.125–4 mg apple polyphenols/ml. In conclusion, ingestion of apple and blackcurrant polyphenols decreased postprandial glycemia, which may be partly related to inhibition of intestinal glucose transport.  相似文献

The objective of this study was to evaluate the effect of supplementing a CP-reduced diet with rumen-protected methionine on growth performance of Fleckvieh bulls. A total of 69 bulls (367 ± 25 kg BW) were assigned to three feeding groups (n = 23 per group). The control (CON) diet contained 13.7% CP and 2.11 g methionine/kg diet (both DM basis) and was set as positive control. The diet reduced in CP (nitrogen) (RED) diet as negative control and the experimental RED + rumen-protected methionine (MET) diet were characterised by deficient CP concentrations (both 9.04% CP). The RED + MET diet differed from the RED diet in methionine concentration (2.54 g/kg DM vs. 1.56 g/kg DM, respectively) due to supplementation of rumen-protected methionine. Rumen-protected lysine was added to both RED and RED + MET at 2.7 g/kg DM to ensure a sufficient lysine supply relative to total and metabolisable protein intake. Metabolisable energy (ME) and nutrient composition were similar for CON, RED, and RED + MET. Bulls were fed for 105 days (d) on average. Individual feed intake was recorded daily; individual BW was recorded at the beginning of the experiment, once per month, and directly before slaughter. At slaughter, blood samples were collected and carcass traits were assessed. Reduction in dietary CP concentration reduced feed intake, and in combination with lower dietary CP concentration, daily intake of CP for RED and RED + MET was lower compared with CON (P < 0.01). Daily ME intake was reduced in RED and RED + MET compared with CON (P < 0.01). Consequently growth performance and carcass weights were reduced (both P < 0.01) in both RED and RED + MET compared with CON. Supplemental rumen-protected methionine was reflected in increased serum methionine concentration in RED + MET (P < 0.05) as compared to RED but it did not affect growth performance, carcass traits and serum amino acid (AA) concentrations, except for lysine which was reduced (P < 0.01) compared to CON and RED. In conclusion, bulls fed RED or RED + MET diets were exposed to a ruminal CP deficit and subsequently a deficit of prececal digestible protein, but methionine did not appear to be the first-limiting essential AA for growth under the respective experimental conditions.  相似文献

While continuous cooling strategies may induce some ergonomic problems to occupational workers, cooling between work bouts may be an alternative for cooling them down in hot environments. The purpose of this study was to assess the effects of wearing a newly designed hybrid cooling vest (HCV) between two bouts of exercise. Inside a climatic chamber set at an air temperature of 37 °C and a relative humidity of 60%, twelve male participants underwent two bouts of intermittent exercise interspersed with a 30 min between-bout recovery session, during which HCV or a passive rest without any cooling (PAS) was administered. The results indicated that thermoregulatory, physiological, and perceptual strains were significantly lower in HCV than those in PAS during the recovery session (p≤0.022), which were accompanied with a large effect of cooling (Cohen's d=0.84–2.11). For the second exercise bout, the exercise time following HCV (22.13±12.27 min) was significantly longer than that following PAS (11.04±3.40 min, p=0.005, d=1.23) During this period, core temperature Tc was significantly lower by 0.14±0.0.15 °C in HCV than that in PAS. The heart rate drift over time was declined by 2±2 bpm min−1 (p=0.001, d=1.00) and the rise in physiological strain index was reduced by 0.11±0.12 unit min−1 (p=0.010, d=0.96) following the use of HCV. These findings suggested that using HCV could accelerate between-bout recovery and improve subsequent exercise performance by the enlarged body core temperature margin and blunted cardiovascular drift.  相似文献

The effect of four cooling strategies on cooling performance of a hybrid personal cooling system (HPCS) incorporated with phase change materials (PCMs) and electric fans in a hot environment (i.e., Tair = 36 ± 0.5 °C, RH = 59 ± 5%) was investigated. Twelve healthy young male participants underwent four 90-min trials comprising 70 min walking and 20 min resting periods. Cooling strategies adopted in this work were CON (control), PCM-control (PCMs were removed at the end of exercise), Fan-control (fans were switched OFF during the initial 20 min) and PCM&Fan-control (fans were turned ON after 20 min exercising and PCMs were removed after the 70-min exercise). Results demonstrated that the control of electric fans could suppress the mean skin temperature rise to 34.0 °C by over 15 min and also cut down the energy consumption of the HPCS from 15.6 W h to 12.1 W h over the entire 90-min trials. Thus, it is recommended that fans should be turned off at the beginning of hot exposure and switched on once participants felt warm. Our findings also showed that the removal of fully melted PCM packs from the HPCS could enhance the evaporative cooling effect brought about by air circulation. The removal of melted PCMs significantly reduced the physical load by 37.3% and ratings of perceived exertion (RPE) were decreased by 3.5–4.2 RPE units. This could also help quickly restore the PCM energy for future usage. In summary, cooling strategies demonstrated in this work could improve HPCS's overall cooling performance on workers while working in the studied hot environment.  相似文献

Productive developmental cycle of the obligate intracellular bacterial pathogen Chlamydia trachomatis depends on the interaction of the replicative vacuole, named the inclusion, with cellular organelles. We have recently reported the formation of ER-Inclusion membrane contact sites (MCSs), where the endoplasmic reticulum (ER) is in apposition to the inclusion membrane. These platforms contain the C. trachomatis inclusion membrane protein IncD, the mammalian ceramide transfer protein CERT and the ER resident proteins VAPA/B and were proposed to play a role in the non-vesicular trafficking of lipids to the inclusion. Here, we identify STIM1 as a novel component of ER-Inclusion MCSs. STIM1, an ER calcium (Ca2+) sensor that relocate to ER-Plasma Membrane (PM) MCSs upon Ca2+ store depletion, associated with C. trachomatis inclusion. STIM1, but not the general ER markers Rtn3C and Sec61ß, was enriched at the inclusion membrane. Ultra-structural studies demonstrated that STIM1 localized to ER-Inclusion MCSs. Time-course experiments showed that STIM1, CERT and VAPB co-localized throughout the developmental cycle. By contrast, Orai1, the PM Ca2+ channel that interacts with STIM1 at ER-PM MCSs, did not associate with C. trachomatis inclusion. Upon ER Ca2+ store depletion, a pool of STIM1 relocated to ER-PM MCSs, while the existing ER-Inclusion MCSs remained enriched in STIM1. Finally, we have identified the CAD domain, which mediates STIM1-Orai1 interaction, as the minimal domain required for STIM1 enrichment at ER-Inclusion MCSs. Altogether this study identifies STIM1 as a novel component of ER-C. trachomatis inclusion MCSs. We discuss the potential role(s) of STIM1 during the infection process.  相似文献

Many documented studies have demonstrated the human mortality rate increases during severe heatwaves. There remains a need for further explore ecologically valid cooling strategies to alleviate body heat strain during extreme heatwaves. The main aim of this work was to explore whether intermittent wetting clothing can be served as an ecologically valid cooling strategy to mitigate heat stress on inactive vulnerable populations not having access to air-conditioning during a severe heatwave. Ten young male subjects underwent two 90-min separate trials: a dry clothing trial (i.e., CON) and a wetted clothing cooling trial (i.e., WEC). A set of light summer wear was chosen and intermittently wetted by tap water at intervals of every 30 min. Physiological and perceptual responses of subjects were examined and compared. All trials were performed in a chamber with an air temperature of 43 ± 0.5 °C, RH= 57 ± 5% and an air velocity of 0.15 ± 0.05 m/s (WBGT=37.35 °C). Results demonstrated that WEC, compared with CON, could significantly reduce both the mean skin temperature and the core temperature throughout the 5–90th min and 25–90th min of the trial, respectively (p < 0.05). Besides, WEC could also remarkable reduce local skin temperatures at those body sites covered by wet clothing (p < 0.05). In comparison, no significant difference was found between WEC and CON on perceptual responses. Further, it was also found from PHS simulations that conditions with a partial water vapour pressure ≤ 3.1–3.5 kPa would not induce pronounced core temperature rises at 43 °C. Finally, it may be concluded that intermittent wetting clothing could be served as an ecologically valid cooling strategy to reduce thermophysiological strain of vulnerable populations while seating during humid heatwaves and thereby improve their health and safety.  相似文献

Several studies on semen physiology and sperm fertilizing capacity have shown a beneficial effect of antioxidants. Procyanidine is a natural antioxidant, more efficient compared with vitamin C and E, with many applications in the food, agriculture, pharmaceutical and cosmetic industry. Thus, we tested whether the addition of procyanidine to the semen of infertile men has a beneficial effect on spermatozoa during their in vitro incubation and during the cryopreservation process. Semen samples of 25 infertile men were divided in to two aliquots, in which procyanidine was added or not. Semen analysis, measurement of sperm DNA fragmentation index (DFI) and measurement of reactive oxygen species (ROS) were performed 3 h after incubation at 37 °C and after sperm cryopreservation and thawing. In-vitro addition of procyanidine to semen of infertile men resulted in a lesser decrease in progressive motility [−4 (−31:+6) vs. −6 (−31:+5), p < 0.001] and total motility [−5 (−29:+3) vs. −9 (−32:+2), p < 0.001] after 3 h of incubation compared with no addition of procyanidine. Sperm morphology was decreased only in the control group after 3 h of incubation [2 (0:+6) vs. 1 (0:+4), p = 0.009]. Furthermore, a larger increase in sperm DFI was observed in the control compared with the procyanidine group [9 (−7:+27) vs. 3 (−3:+18), p = 0.005] after thawing of cryopreserved semen samples. In conclusion, in-vitro addition of procyanidine to the semen of infertile men exerts a protective effect on progressive motility during handling and after 3 h of incubation as well as on sperm DFI during the process of cryopreservation.  相似文献

This work aimed to evaluate the effects of tannin-containing heather (Calluna vulgaris, Erica spp.) and energy (oats, Avena sativa) supplementation, combined or not, on feed intake, gastrointestinal nematode infections and performance of goats grazing mountain grasslands. Two successive experiments were established across one grazing season on four paddocks. The first (late April to early August, Period 1) involved two treatments, i.e., supplementation with heather (+H) vs. non-supplementation (−H), each randomly allocated to two paddocks. The second (mid-August to mid-November, Period 2) consisted on four treatments in a 2 × 2 factorial design, i.e., supplementation with heather and oats (+H+O), only heather (+H−O), only oats (−H+O), and no supplementation (−H−O). Results from Period 1 indicated that faecal nematode egg counts (FEC) were lower in +H goats (50% less in August), achieving more favourable live weight (LW) and body condition changes than −H goats. Total dry matter intake (DMI) in June was similar between treatments, with heather accounting for 0.21 of +H goat diets. Kids FEC and LW gains were not affected by heather supplementation. In Period 2, both heather and oat supplementation reduced FEC (45 and 61%, respectively) and improved goat performance, without an interaction between both factors. Total DMI in October was higher in +H+O compared to other treatments (48 vs. 30 g DM kg LW−0.75 d−1; P < 0.01). Rumen ammonia concentration was lower (P < 0.001) in +H than in −H goats, and in +O than in −O goats, whereas that of volatile fatty acids (VFA) was not affected by treatments, though the molar proportions of some VFA were modified. It is concluded that the combination of both supplements (tannins and energy) contributes to reduce gastrointestinal nematode parasitism and increases goat performance, allowing a lower dependence on conventional chemotherapy.  相似文献

Previously we showed that feeding polyphenol-rich wild blueberries to hypertensive rats lowered systolic blood pressure. Since probiotic bacteria produce bioactive metabolites from berry polyphenols that enhance the health benefits of berry consumption, we hypothesized that adding probiotics to a blueberry-enriched diet would augment the anti-hypertensive effects of blueberry consumption. Groups (n = 8) of male spontaneously hypertensive rats were fed one of four AIN ‘93G-based diets for 8 weeks: Control (CON); 3% freeze-dried wild blueberry (BB); 1% probiotic bacteria (PRO); or 3% BB + 1% PRO (BB+PRO). Blood pressure was measured at weeks 0, 2, 4, 6, and 8 by the tail-cuff method, and urine was collected at weeks 4 and 8 to determine markers of oxidative stress (F2-isoprostanes), nitric oxide synthesis (nitrites), and polyphenol metabolism (hippuric acid). Data were analyzed using mixed models ANOVA with repeated measures. Diet had a significant main effect on diastolic blood pressure (p = 0.046), with significantly lower measurements in the BB- vs. CON-fed rats (p = 0.035). Systolic blood pressure showed a similar but less pronounced response to diet (p = 0.220), again with the largest difference between the BB and CON groups. Absolute increase in blood pressure between weeks 0 and 8 tended to be smaller in the BB and PRO vs. CON and BB+PRO groups (systolic increase, p = 0.074; diastolic increase, p = 0.185). Diet had a significant main effect on hippuric acid excretion (p<0.0001), with 2- and ~1.5-fold higher levels at weeks 4 and 8, respectively, in the BB and BB+PRO vs. PRO and CON groups. Diet did not have a significant main effect on F2-isoprostane (p = 0.159) or nitrite excretion (p = 0.670). Our findings show that adding probiotics to a blueberry-enriched diet does not enhance and actually may impair the anti-hypertensive effect of blueberry consumption. However, probiotic bacteria are not interfering with blueberry polyphenol metabolism into hippuric acid.  相似文献

Clinical endometritis (CE) is a major cause in affecting the reproductive performance of dairy cows. The objectives of this study were to ascertain the prevalence of CE and to evaluate the effect of CE on reproductive performance in dairy cows using vaginal discharge score (VDS) grading system. 803 dairy cows were examined by vaginoscope with 4-point VDS at 26 ± 3 days in milk (DIM) and classified into six groups: non-endometritis with VDS 0 (control; CON), endometritis with VDS 1 (MEM), non-treated endometritis with VDS 2 (NTME), treated endometritis with VDS 2 (TME), non-treated endometritis with VDS 3 (NTPE), and treated endometritis with VDS 3 (TPE). Cows in TME and TPE groups were treated with 200 mL of 50% dextrose solution by intrauterine infusion. The prevalence of CE was 33% at 26 ± 3 DIM. Binary logistic regression analysis revealed cows in MEM, NTME and NTPE groups had a less likelihood of first artificial insemination (AI) pregnancy than those in CON group (P < 0.05). Kaplan-Meier survival curves for days open were statistically different (P = 0.004). In Cox regression model, cows in NTME and NTPE groups had a reduced pregnancy rate than those in CON group (P < 0.05). The hazard of pregnancy in NTME group was lower than that in TME group (P = 0.044). Similarly, it was lower for the hazard of pregnancy in NTPE group than in TPE group (P = 0.048). Cows in MEM, NTME, and NTPE groups required more services per pregnancy than those in CON group (P < 0.05). In conclusion, CE examined by the VDS grading system impaired reproductive performance, and mild endometritis with VDS 1 should be treated in the early postpartum period to ameliorate fertility in dairy herds.  相似文献

Toxoplasmosis, caused by the protozoan parasite Toxoplasma gondii, is an important zoonotic infection. Veterinary personnel and abattoir workers are considered to be at a high risk of T. gondii infection owing to their occupational exposure. However, the association of T. gondii infection with occupational exposure to animals has not been determined in India. Hence, we analysed 139 and 126 blood samples of veterinary personnel and abattoir workers, respectively, for anti-T. gondii antibodies using enzyme-linked immunosorbent assay (ELISA), modified agglutination test (MAT) and indirect fluorescent antibody test (IFAT). The association of seroprevalence with sociodemographic profiles, work activities and dietary habits was determined in the study population. MAT, ELISA and IFAT results demonstrated nearly 46%, 48% and 47% seropositivity, respectively. MAT (kappa = 0.924) and IFAT (kappa = 0.962) results showed good agreement with ELISA results. Of the ELISA positive samples, 46% was copositive for IgG antibody, 1.5% for IgM antibody and 1.5% for both IgG and IgM antibodies. High IgG avidity was observed only in IgG+ IgM- and IgG+ IgM+ samples and not in IgM+ IgG- samples, indicating chronic T. gondii infection in most of the cases. Furthermore, multivariate analysis revealed that T. gondii seropositivity was associated with age > 30 years (odds ration [OR] = 1.992), cat at home (OR = 1.991), not wearing gloves (OR = 1.886), not wearing safety glasses (OR = 1.985) and contact with soil (OR = 1.695). These findings support the presence of a potentially significant association between T. gondii seropositivity and occupational exposure to animals.  相似文献

A previous study showed the additive methane (CH4)-mitigating effect of nitrate and linseed fed to non-lactating cows. Before practical application, the use of this new strategy in dairy cows requires further investigation in terms of persistency of methanogenesis reduction and absence of residuals in milk products. The objective of this experiment was to study the long-term effect of linseed plus nitrate on enteric CH4 emission and performance in dairy cows. We also assessed the effect of this feeding strategy on the presence of nitrate residuals in milk products, total tract digestibility, nitrogen (N) balance and rumen fermentation. A total of 16 lactating Holstein cows were allocated to two groups in a randomised design conducted in parallel for 17 weeks. Diets were on a dry matter (DM) basis: (1) control (54% maize silage, 6% hay and 40% concentrate; CON) or (2) control plus 3.5% added fat from linseed and 1.8% nitrate (LIN+NIT). Diets were equivalent in terms of CP (16%), starch (28%) and NDF (33%), and were offered twice daily. Cows were fed ad libitum, except during weeks 5, 16 and 17 in which feed was restricted to 95% of dry matter intake (DMI) to ensure complete consumption of meals during measurement periods. Milk production and DMI were measured weekly. Nitrate and nitrite concentrations in milk and milk products were determined monthly. Daily CH4 emission was quantified in open circuit respiration chambers (weeks 5 and 16). Total tract apparent digestibility, N balance and rumen fermentation parameters were determined in week 17. Daily DMI tended to be lower with LIN+NIT from week 4 to 16 (−5.1 kg/day on average). The LIN+NIT diet decreased milk production during 6 non-consecutive weeks (−2.5 kg/day on average). Nitrate or nitrite residuals were not detected in milk and associated products. The LIN+NIT diet reduced CH4 emission to a similar extent at the beginning and end of the trial (−47%, g/day; −30%, g/kg DMI; −33%, g/kg fat- and protein-corrected milk, on average). Diets did not affect N efficiency and nutrients digestibility. In the rumen, LIN+NIT did not affect protozoa number but reduced total volatile fatty acid (−12%) and propionate (−31%) concentrations. We concluded that linseed plus nitrate may have a long-term CH4-mitigating effect in dairy cows and that consuming milk products from cows fed nitrate may be safe in terms of nitrate and nitrite residuals. Further work is required to optimise the doses of linseed plus nitrate to avoid reduced cows performance.  相似文献

The applicability of the plant enzyme ficin (EC 3.4.22.3) for the prediction of protein degradability of ruminal feeds was examined. Optimum pH and the effect of enzyme concentration, incubation time and amylase pretreatment for high-starch feeds on protein solubility with ficin (FS) were determined. The predictive ability of ficin assay in the conditions specified for particular groups of feeds was estimated using 61 feeds including protein meals (7), cereal grains (2), silages (9), sugar beet pulp, dried forages (29) and compound feeds (13) of known in situ effective protein degradability (ED) determined with cows.Optimum incubation time was 2–3 h, enzyme amount (per 100 mg of crude protein in feed sample) equivalent to 9 U for compound feeds and concentrate ingredients not treated with amylase, 18 U for forages. Close linear relationships between the in vitro FS (X, %) and in situ estimates of ED (Y, %) were found. The regression equation for all samples (n = 61) was Y = 1.018 X + 1.331, R2 = 0.92 (P < 0.001), SEE 4.17. After splitting to forages (FOR) and compound feeds and ingredients (CON) SEEs were 4.25 and 3.56%, respectively. FS values accounted for 85, 92 and 95% of the variation in ED measured in situ for FOR, FOR + CON and CON samples, respectively.  相似文献

Genetic parameters were estimated for cold carcase weight (CCW), carcase conformation (CON), carcase fat class (FAT), age at slaughter (AGE) and average daily carcase gain (ADCG) in 14 common UK breeds of cattle. These included crossbred animals but purebred datasets were also analysed for the most populous sire-breeds. Heritability estimates for beef breeds that were significant ranged from 0.24 to 0.44, 0.12 to 0.35, 0.12 to 0.36, 0.15 to 0.38 and 0.26 to 0.43 for CCW, CON, FAT, AGE and ADCG, respectively. For Holstein-Friesian, a dairy breed, heritability estimates were consistently lower than most beef breeds with estimates of 0.12, 0.13, 0.13, 0.06 and 0.15 for CCW, CON, FAT, AGE and ADCG, respectively. In all breed groups, genetic correlations were positive between CCW, CON and ADCG. In general, genetic correlations were moderate between CCW and CON (0.13 to 0.77), moderate to strong between CCW and ADCG (0.57 to 0.98) and weak or moderate between CON and ADCG (0.12 to 0.82). Genetic correlations for FAT with CCW (−  0.20 to −  0.42) and CON (−  0.16 to −  0.52) tended to be negative in the beef breed but were positive in the dairy breed, although not significant between CCW and FAT. For most beef breeds genetic correlations between AGE and carcase traits were not significant with the exceptions of AGE and CCW for Simmental (−  0.15) and Salers (−  0.24), AGE and CON for Limousin (0.15) and Simmental (0.14) and AGE and FAT from three sire-breeds (−  0.17 to −  0.35). However, the correlation between AGE and ADCG was negative and moderate to strong in magnitude (−  0.23 to −  0.67) in all beef breeds as expected since faster-growing animals reach slaughter age earlier. For Holstein-Friesian, all genetic correlations with AGE were negative and moderate to strong. Genetic correlations indicate that selection for increased carcase weight should simultaneously increase growth rate and improve conformation in all breeds and reduce carcase fatness in the majority of beef breeds. The results indicate that there is genetic variation in all five traits suitable for undertaking genetic improvement of carcase traits and age at slaughter; however, there are apparent breed differences. The use of abattoir-derived phenotypes for undertaking genetic improvement is an example where the supply chain can work together to share information to enable the cattle industry to move forward.  相似文献

[Purpose]

In this study, the authors have intended to investigate the effects that the exercise training and the intake of the water extract from propolis have on the activity of antioxidant enzymes.

[Methods]

For this purpose, the exercise training (70% VO2max treadmill running exercise for 60min)of 5 times per week for six weeks and the intake (50mg/kg/day) of the water extract from propolis were performed by separating the experimental animals (SD rats, n=32) into CON(n=8) group, CON+Ex(n=8), PA(n=8), and PA+Ex(n=8).

[Results]

As a result, the following conclusions were obtained: The concentration of the blood glucose and insulin of the CON+Ex group and PA+Ex group which are the exercise parallel group were significantly decreased in comparison with the control group, whereas if comparing the glycogen concentration in skeletal muscle and liver tissue between the exercise parallel group and the CON group, the former showed significantly high value in comparison with the latter (p < .05). In the case of the activity of the antioxidant enzyme in the skeletal muscle and the liver tissue, the activities of SOD, GPX and CAT in the gastrocnemius muscle tissue of the experimental animals showed significantly high value in PA+Ex group in comparison with other experimental groups (p < .05). In addition, the SOD activity in the liver tissue showed that only PA+Ex group was significantly increased, whereas GDX activity showed significantly higher value in CON+Ex group and PA group than CON group (p < .05). However, the activity of CAT in the liver tissue showed that there is no difference between the experimental groups. As a result that measured the concentration of MDA in order to evaluate the damage level of the tissue by oxygen free radicals, the difference between the groups in the liver tissue was not shown, while it was shown that only PA+Ex group in the skeletal muscle tissue was significantly decreased in comparison with other experimental groups (p < .05).

[Conclusion]

Taken together the above findings, it is considered that the parallel treatment of the exercise training and the water extract from propolis can not only increase the use of glycogen of the skeletal muscle and liver tissue, but also it can give the effect to suppress the creation of active oxygen by inducing the activity of the antioxidant enzyme in the body, and in the future, the possibility as the exercise supplements and the antioxidant of the water-soluble propolis are expected.  相似文献

The aim was to compare the antiobesity efficacy of different concentrations of a phenolic-rich water extract from purple maize pericarp (PPE) in a murine model of obesity for 12 weeks. Forty C57BL/6 mice (n=10/group) were randomized: standard diet (SD), high-fat diet (HFD), HFD+200 mg PPE/kg (200 PPE) and HFD+500 mg PPE/kg (500 PPE). PPE contained mainly ferulic acid, anthocyanins and other phenolics (total phenolics: 448.5 μg/mg dry weight, DW). Body weight (−27.9%), blood glucose (−26.5%) and blood triglycerides (−22.1%) were most attenuated (P<.05) in 500 PPE group compared to HFD group. Also, 500 PPE group had reduced (P<.05) plasma levels of TNF-α, MCP-1, resistin and leptin compared to HFD group. Fatty liver disease scores were highest for HFD (8.4), followed by 200 PPE (6.1), 500 PPE (2.7) and SD (0.4) groups. Relative adipose tissue was lower (P<.05) in 200 PPE (7.6%), 500 PPE (8.0%) and SD (0.8%) compared to HFD (12.1%) group. In 500 PPE group, compared to HFD group, important genes were modulated related to adipogenesis (Mmp3, fold-change [FC]=7.4), inflammation (Nfkb1, FC=−1.8) and glucose metabolism (Slc2a4, FC=23.6) in adipose tissue. In liver, 500 PPE group showed modulation of genes related to gluconeogenesis (Pck1, FC=−2.9), lipogenesis (Fasn, FC=−2.4) and β-oxidation (Cpt1b, FC=3.1). Maize rich in ferulic acid and anthocyanins prevented obesity through the modulation of TLR and AMPK signaling pathways reducing adipogenesis and adipose inflammation, and promoting energy expenditure.  相似文献

Although various intrinsic and extrinsic risk factors for anterior cruciate ligament (ACL) rupture have been identified, the exact aetiology of the injury is not yet fully understood. Type III collagen is an important factor in the repair of connective tissue, and certain gene polymorphisms may impair the tensile strength. The aim of this study was to examine the association of the COL3A1 rs1800255 polymorphism with ACL rupture in Polish male recreational skiers. A total of 321 male Polish recreational skiers were recruited for this study; 138 had surgically diagnosed primary ACL ruptures (ACL-injured group) and 183 were apparently healthy male skiers (control group – CON) who had no self-reported history of ligament or tendon injury. Both groups had a comparable level of exposure to ACL injury. Genomic DNA was extracted from the oral epithelial cells. All samples were genotyped on a real-time polymerase chain reaction instrument. The genotype distribution in the ACL-injured group was significantly different than in CON (respectively: AA=10.1 vs 2.2%, AG=22.5 vs 36.1, GG=67.4 vs 61.8%; p=0.0087). The AA vs AG+GG genotype of COL3A1 (odds ratio (OR)=5.05; 95% confidence interval (CI), 1.62-15.71, p=0.003) was significantly overrepresented in the ACL-injured group compared with CON. The frequency of the A allele was higher in the ACL-injured group (21.4%) compared with CON (20.2%), but the difference was not statistically significant (p=0.72). This study revealed an association between the COL3A1 rs1800255 polymorphism and ACL ruptures in Polish skiers.  相似文献

BackgroundPrevious research demonstrated that a high dose of phlorizin-rich apple extract (AE) can markedly inhibit early-phase postprandial glycemia, but efficacy of lower doses of the AE is unclear.ObjectiveTo determine whether lower AE doses reduce early-phase postprandial glycemia in healthy adults and investigate mechanisms.DesignIn a randomized, controlled, double-blinded, cross-over acute trial, drinks containing 1.8 g (HIGH), 1.35 g (MED), 0.9 g (LOW), or 0 g (CON) of a phlorizin-rich AE were consumed before 75 g starch/sucrose meal. Postprandial blood glucose, insulin, C-peptide, glucose-dependent insulinotropic polypeptide (GIP) and polyphenol metabolites concentrations were measured 0–240 min, acetaminophen concentrations to assess gastric emptying rate, and 24 h urinary glucose excretion. Effects of AE on intestinal glucose transport were investigated in Caco-2/TC7 cells.ResultsAE significantly reduced plasma glucose iAUC 0–30 min at all doses: mean differences (95% CI) relative to CON were −15.6 (−23.3, −7.9), −11.3 (−19.6, −3.0) and −8.99 (−17.3, −0.7) mmol/L per minute for HIGH, MEDIUM and LOW respectively, delayed Tmax (HIGH, MEDIUM and LOW 45 min vs. CON 30 min), but did not lower Cmax. Similar dose-dependent treatment effects were observed for insulin, C-peptide, and GIP. Gastric emptying rates and urinary glucose excretion did not differ. Serum phloretin, quercetin and epicatechin metabolites were detected postprandially. A HIGH physiological AE dose equivalent decreased total glucose uptake by 48% in Caco-2/TC7 cells.ConclusionsPhlorizin-rich AE, even at a low dose, can slightly delay early-phase glycemia without affecting peak and total glycemic response.  相似文献
