The effects of temperature and light on the growth and geosmin production of Lyngbya kuetzingii were determined. Of the three temperatures tested, 10, 25 and 35°C, the maximal geosmin concentration and geosmin productivity were yielded at 10°C, while the highest chl a production was observed at 25°C. In the studies on light intensity, the maximal geosmin concentration and geosmin productivity were observed at 10 μmol m−2 s−1, while the highest chl a production was at 20 μmol m−2 s−1. It was suggested that more geosmin was synthesized with lower chl a demand. Meanwhile, the relative amounts of extra- and intracellular geosmin were investigated. Under optimum growth conditions (20 μmol m−2 s−1, 25°C; BG-11 medium), the amounts of extracellular geosmin increased as the growth progressed and reached the maximum in the stationary phase, while the intracellular geosmin reached its maximum value in the late exponential phase, and then began to decline. However, under the low temperature (10°C) or light (10 μmol m−2 s−1) conditions, more intracellular geosmin was synthesized and mainly accumulated in the cells. The proportions of extracellular geosmin were high, to 33.33 and 32.27%, respectively, during the stationary phase at 35°C and 20 μmol m−2 s−1. It was indicated that low temperature or light could stimulate geosmin production and favor the accumulation of geosmin in cells, while more intracellular geosmin may be released into the medium at higher temperatures or optimum light intensity.  相似文献

A volatile metabolite, 2-methylisoborneol (2-MIB), causes an unpleasant taste and odor in tap water. Some filamentous cyanobacteria produce 2-MIB via a two-step biosynthetic pathway: methylation of geranyl diphosphate (GPP) by methyl transferase (GPPMT), followed by the cyclization of methyl-GPP by monoterpene cyclase (MIBS). We isolated the genes encoding GPPMT and MIBS from Pseudanabaena galeata, a filamentous cyanobacterium known to be a major causal organism of 2-MIB production in Japanese lakes. The predicted amino acid sequence showed high similarity with that of Pseudanabaena limnetica (96% identity in GPPMT and 97% identity in MIBS). P. galeata was cultured at different temperatures to examine the effect of growth conditions on the production of 2-MIB and major metabolites. Gas chromatograph–mass spectrometry (GC–MS) measurements showed higher accumulation of 2-MIB at 30 °C than at 4 °C or 20 °C after 24 h of culture. Real-time-RT PCR analysis showed that the expression levels of the genes encoding GPPMT and MIBS decreased at 4 °C and increased at 30 °C, compared with at 20 °C. Furthermore, metabolite analysis showed dramatic changes in primary metabolite concentrations in cyanobacteria grown at different temperatures. The data indicate that changes in carbon flow in the TCA cycle affect 2-MIB biosynthesis at higher temperatures.  相似文献

Taste and odor (T & O) episodes always cause strong effects on drinking water supply system. Luanhe River diversion into Tianjin City in China is an important drinking water resource. Massive growth of a benthic filamentous cyanobacterium with geosmin production in the open canal caused a strong earthy odor episode in Tianjin. On the basis of the morphological and molecular identification of this cyanobacterium as Oscillatoria limosa Agardh ex Gomont, the genetic basis for geosmin biosynthesis and factors influencing growth and geosmin production of O. limosa CHAB 7000 were studied in this work. A 2268-bp open reading frame, encoding 755 amino acids, was amplified and characterized as the geosmin synthase gene (geo), followed by a cyclic nucleotide-binding protein gene (cnb). Phylogenetic analysis implied that the evolution of the geosmin genes in O. limosa CHAB 7000 might involve a horizontal gene transfer event. Examination on the growth and geosmin production of O. limosa CHAB 7000 at different light intensities showed that the maximum geosmin production was observed at 10 μmol photons m−2 s−1, while the optimum growth was at 60 μmol photons m−2 s−1. Under three temperature conditions (15 °C, 25 °C, and 35 °C), the maximum growth and geosmin production were observed at 25 °C. Most amounts of geosmin were retained in cells during the growth phase, but high temperature and low light intensity increased the release of geosmin into the medium, implying that O. limosa CHAB 7000 had a high potential harm for the release of geosmin from its cells at these adverse conditions.  相似文献

In decades, Pseudanabaena species have been frequently reported as cyanobacterial bloom components. However, these species are always overlooked because they cannot be observed easily owing to their small sizes. This study aimed to characterize Pseudanabaena species and detect them in field waters by developing an effective DNA marker. A region with approximately 600 bp in the rbcLX sequences was specific to Pseudanabaena species alone compared with other cyanobacterial genera. Therefore, new primers specific for Pseudanabaena were generated. By using this molecular tool, seasonal and spatial dynamics of Pseudanabaena populations of six sites in Lushui Reservoir, China were measured from July 2010 to August 2011. Pseudanabaena species existed in the reservoir throughout the year, and cell abundance ranged between 104 and 106 equivalent cells/L, with the peak at 8.07 × 106 equivalent cells/L. The seasonal dynamics pattern of Pseudanabaena was similar to that of Microcystis but showed a slight lag at the transitional points of water temperature in the reservoir.  相似文献

Anabaena azotica FACHB-118 and Anabaena sp. CH1, heterocystous cyanobacteria isolated from Chinese and Taiwanese rice fields, expressed vanadium-containing nitrogenase when under molybdenum deficiency. This is the second direct observation of an alternative nitrogenase in cyanobacteria. The vanadium nitrogenase-specific genes vnfDG are fused and clustered in a phylogenetic tree next to the corresponding genes of Methanosarcina. The expression of vnfH in cells cultured in Mo-free medium and of nifH in Mo-grown cells was shown for the first time by sequencing cDNA derived from cultures of A. azotica and Anabaena sp. CH1. The vnfH sequences clustered with that of Anabaena variabilis. The vnf genes were strongly transcribed only in cultures grown either in Mo-free medium, or in W-containing medium, but also weakly in Mo-containing medium. NifH was transcribed in all media. On-line measurements of acetylene reduction by Mo-free A. azotica cultures demonstrated that the V-nitrogenase was active. Ethane was formed continuously at a rate of 2.1% of that of ethylene. Acetylene reduction of cultures grown either with or without Mo had a high temperature optimum of 42.5°C. The uptake hydrogenase gene hupL was expressed in Mo-free medium concomitantly with vnfDG in A. azotica, Anabaena sp. CH1, and A. variabilis.  相似文献

Cyanobacteria are the major producers of geosmin in natural waters. To identify a gene involved in geosmin biosynthesis in cyanobacteria, the polymerase chain reaction (PCR) was used to amplify a 2298-bp open reading frame (ORF) from the geosmin-producing cyanobacterium Lyngbya kuetzingii UTEX 1547. This ORF encoded a protein of 765 amino acids. Alignment of the deduced amino acid sequence demonstrated that geoL had high similarity to the corresponding genes of Oscillatoria sp. PCC 6506 (100% identity), Calothrix sp. PCC 7507 (89%), Anabaena ucrainica CHAB 1432 (88%), A. ucrainica CHAB 2155 (87%), Nostoc punctiforme PCC 73102 (87%), Phormidium sp. P2r (84%) and Cylindrospermum stagnale PCC 7417 (83%), and modest similarity to myxobacteria (61–73%). It also indicated geoL with low similarity to the corresponding genes of actinomycetes (<60%). The encoded protein GEOL was estimated to have two geosmin synthase domains, and each contained two strictly conserved Mg2+-binding motifs (aspartate-rich motif and NSE triad). The geoL gene was shown to be responsible for geosmin biosynthesis in L. kuetzingii UTEX 1547. Then, geoL had been cloned into pET21a(+) vector and expressed in Escherichia coli BL21(DE3) with the isopropyl-β-d-thiogalactoside (IPTG) induction. The recombinant GEOL protein was purified and exhibited a single band (MW  90 kDa) on the sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), which was consistent with the predicted molecular weight (MW) of 87,046 Da. In conclusion, this study has confirmed that geosmin synthase gene and its expression product can be identified and characterized from cyanobacteria, which will help understand the fundamental biological mechanism of geosmin biosynthesis in cyanobacteria.  相似文献

The isolated cyanobacterium containing biopigments like chlorophyll-a, phycoerythrin, phycocyanin, and carotenoid was cultured under different quality of light modes to ascertain biomass and pigment productivity. On the basis of 16S rRNA gene sequence, the isolate was identified as Pseudanabaena sp. Maximum biomass concentration obtained in white-, blue-, and green-light was 0.82, 0.94, and 0.89 g/L, respectively. It was observed that maximum phycoerythrin production was in green light (39.2 mg/L), ensued by blue light (32.2 mg/L), while phycocyanin production was maximum in red light (10.9 mg/L). In yellow light, pigment production as well as the growth rate gradually declined after 12 days. Carotenoid production decreased in blue-, white-, and red-light after 15 days, while in green light it had increased gradually. The present communication suggests that Pseudanabaena sp. can be used for commercial production of phycoerythrin when grown under green light.  相似文献

The toxicity and growth of Microcystis aeruginosa (UV-006) from the Hartbeespoort Dam, South Africa were investigated at different temperatures and photon fluence rates under laboratory conditions. Cells harvested in late logarithmic growth phase were most toxic when grown at 20°C (LD50) median lethal dose [IP, mouse]=25.4 mg kg-1). Toxicity was markedly reduced at growth temperatures above 28° C. Fluence rate had a smaller effect on the toxicity of the cells, but toxicity tended to be less at the very low and high light fluences. Optimal conditions for growth did not coincide with those for toxin production. Well-aerated cultures of this isolate kept at pH 9.5 by CO2 addition, a temperature of 20–24° C, a fluence rate of 145 mol photons m-2 s-1 and harvested in the late logarithmic growth phase yielded the maximum quantity of toxin.Abbreviation LD50 median lethal dose An abstract of this work, presented as a poster at the IUBS symposium on toxins and lectins, held at the CSIR, Pretoria, South Africa during 1982 was published in S. Incubation in the dark of photoautotrophically grown N2-fixing heterocystous cyanobacteria leads to a loss of nitrogenase activity. Original levels of nitrogenase activity are rapidly regained upon re-illumination of the filaments, in a process dependent on de novo protein synthesis. Ammonia, acting indirectly through some of its metabolic derivatives, inhibits the light-promoted development of nitrogenase activity in filaments of Anabaena sp. ATCC 33047 and several other cyanobacteria containing mature heterocysts. The ammonia-mediated control system is also operative in N2-fixing filaments in the absence of any added source of combined nitrogen, with the ammonia resulting from N2-fixation already partially inhibiting full expression of nitrogenase. High nitrogenase levels, about two-fold higher than those in normal N2-fixing Anabaena sp. ATCC 33047, are found in cell suspensions which have been treated with the glutamine synthetase inhibitor l-methionine-d,l-sulfoximine or subjected to nitrogen starvation. Populations of Limnothrix redekei, Oscillatoria lanceaeformis, Planktothrix agardhii and Pseudanabaena limnetica were found in a hypertrophic, gravel-pit lake near Madrid (Spain), throughout a one year sampling at weekly intervals. Physico-chemical factors, phytoplankton biomass and net growth rates were measured. Oscillatoria lanceaeformis was only observed a few weeks, probably being related to phosphorus limitation. Planktothrix agardhii biomass was related to PhAR irradiances, light attenuation coefficient in the water, Brunt-Väisäla frequency and decrease of soluble reactive phosphorus. Limnothrix redekei and Pseudanabaena limnetica biomass values were related to a decrease of inorganic nitrogen and temperature. A different lag response of populations was observed in relation to the environmental features.  相似文献

Sporangiophore initiation in Pilobolus crystallinus grown in white light was induced by either a dark or a low-temperature treatment. The period of darkness necessary to induce sporangiophore initiation was shortened by lowering the temperature. Arrhenius plots for the sporangiophore-suppressing reaction in both light and darkness consisted of two straight lines with a Q10 of about 2 at lower temperatures and 8–11 at higher temperatures. The temperature at which the Q10 changed was the lower, the higher the fluence rate: 14° C at 8 W/m2, 19.5° C at 0.24 W/m2 and 24.5° C in darkness. Light effect on cultures of microalgae has been studied mainly on single species cultures. Cyanobacteria have photosynthetic pigments that can capture photons of wavelengths not available to chlorophylls. A native Louisiana microalgae (Chlorella vulgaris ) and cyanobacteria (Leptolyngbya sp.) co‐culture was used to study the effects of light quality (blue–467 nm, green–522 nm, red–640 nm and white–narrow peak at 450 nm and a broad range with a peak at 550 nm) at two irradiance levels (80 and 400 μmol m?2 s?1) on the growth, species composition, biomass productivity, lipid content and chlorophyll‐a production. The co‐culture shifted from a microalgae dominant culture to a cyanobacteria culture at 80 μmol m?2 s?1. The highest growth for the cyanobacteria was observed at 80 μmol μmol m?2 s?1 and for the microalgae at 400 μmol m?2 s?1. Red light at 400 μmol m?2 s?1 had the highest growth rate (0.41 d?1), biomass (913 mg L?1) and biomass productivity (95 mg L?1 d?1). Lipid content was similar between all light colors. Green light had the highest chlorophyll‐a content (1649 μg/L). These results can be used to control the species composition of mixed cultures while maintaining their productivity.  相似文献

Before switching totally to alternative fuel stage, CO2 mitigation process has considered a transitional strategy for combustion of fossil fuels inevitably. In comparison to other CO2 mitigation options, such as oceanic or geologic injection, the biological photosynthetic process would present a far superior and sustainable solution under both environmental and social considerations. The utilization of the cyanobacteria Anabaena sp. CH1 in carbon dioxide mitigation processes is analyzed in our research. It was found that an original developed photobioreactor with internal light source exhibits high light utilization. Anabaena sp. CH1 demonstrates excellent CO2 tolerance even at 15% CO2 level. This enables flue gas from power plant to be directly introduced to Anabaena sp. CH1 culture. Double light intensity and increased 47% CO2 bubble retention time could enhance CO2 removal efficiencies by 79% and 67%, respectively. A maximum CO2 fixation rate of 1.01 g CO2 L−1 day−1 was measured experimentally.  相似文献

In this study, we conducted experiments to accumulate practical information on the propagation and establishment of a population of Cardiocrinum cordatum var. glehnii by seed sowing. C. cordatum var. glehnii seeds require approximately 19 months from seed dispersal to cotyledon emergence in the field. However, the period from seed dispersal to radicle emergence was shortened to approximately 7–8 months by the temperature transition of 25/15°C (60 days) → 15/5°C (30 days) → 0°C (120 days) → 15/5°C (i.e., 15/5°C represents alternating temperature treatment wherein the seeds were placed at 15°C for 12 h during the day and then at 5°C for 12 h during the night). More than 90% of the seeds, which were stored dry at 5°C for 12 months and sown in pots in the field, showed cotyledon emergence, whereas in seeds stored dry at 25°C, dry at room temperature, and non-dry at room temperature, cotyledon emergence was decreased by less than 1%. More than 88% of the seeds that were stored dry at 5°C and sown in the field in October 2002 immediately after collecting, November, and from April to July 2003 showed cotyledon emergence in spring 2004. However, seeds sown in August, September, and October 2003 showed cotyledon emergences of 57.6%, 0%, and 0% in spring 2004, respectively. Seeds collected in October 2002 and sown until July 2003 in the field received adequate high temperature in summer, moderate temperature in autumn, and cold temperature in winter; therefore, the percentage of cotyledon emergence was high in spring 2004. On the other hand, seeds sown in August 2003 or later could not receive enough high temperature; thus, cotyledons emerged from only a few seeds.  相似文献

Ferredoxin-nitrite reductase (EC 1.7.7.1.) from the cyanobacteria Anabaena sp. 7119 has been purified 763-fold with a specific activity of 21.5 units/nig protein (0.358 μkatals/mg). The enzyme has a molecular mass of 52,000 daltons with a Stokes radius of 3.09 nm and a sedimentation coefficient of 4.07 S. The cellular level of nitrite reductase activity gradually increases in response to the addition of increasing amounts of iron to the culture medium.
When partially purified nitrite reductase preparations are subjected to sucrose-density-gradient centrifugation there is a dose correspondence between nitrite reductase activity and absorbance at 400 nm. This suggests the association of a heme chromophore with the enzyme. Furthermore, the presence of an iron-sulfur center is suggested by a close association of acid-labile sulfide with nitrite reductase activity. Carbon monoxide inhibits nitrite reductase activity. The nature and kinetics of this reaction are comparable to other siroheme-containing nitrite reductases.  相似文献

The influence of light and temperature on the cylindrospermopsin (CYN) production of two Aphanizomenon flos-aquae strains, isolated from North-eastern German lakes, was investigated with semi-continuously growing cultures. A light gradient from 10 to 60 μE m−2 s−1 in combination with temperatures of 16, 20, and 25 °C was tested.CYN concentrations varied by a maximum factor of 2.7 in strain 10E9 with a significant decrease with increasing temperature. Strain 22D11 showed less pronounced changes, i.e. by a factor of 1.6, and without clear relationship to temperature.Reaction patterns of CYN production to changing light intensities are different at different temperatures. In both strains CYN concentrations increase significantly at 20 °C between 10 and 60 μE m−2 s−1, whereas they decrease significantly at 25 °C in the same light gradient. The amount of synthesised CYN is not reflected by growth rates of the strains in a uniform manner. Nonetheless several temperature–light combinations which constitute physiological stress seem to trigger CYN production and particularly CYN release from cells. The lowest growth rate observed at 16 °C and 60 μE m−2 s−1 of strain 22D11 may reflect photoinhibition due to the lower temperature and related limited CO2-fixation. Under these conditions, extracellular CYN concentrations increased to 58% of total CYN, while the share of extracellular CYN of all other light and temperature regimes was 11–26%. From the results and the experimental design we conclude an active release of the toxin into medium to be more likely than mere leakage from cells.  相似文献

Recent investigations have shown the temperate scleractinian coral Cladocora caespitosa to be a new potential climate archive for the Mediterranean Sea. Whilst earlier studies have demonstrated a seasonal variation in growth rates, they were unable to distinguish which environmental parameter (light, temperature, or food) was influencing growth. In this study, the effect of these three factors on the coral physiology and calcification rate was characterized to aid the correct interpretation of skeletal trace element variations. Two temperatures (13 and 23°C), irradiances (50 and 120 μmol m−2 s−1), and feeding regimes (unfed and fed with nauplii of Artemia salina) were tested under controlled laboratory conditions on the growth, zooxanthellae density, chlorophyll (chl) content, and asexual reproduction (budding) of C. caespitosa during a 7-week factorial experiment. Unlike irradiance, which had no effect, high temperature and food supply increased the growth rates of C. caespitosa. The effect of feeding was however higher for corals maintained at low temperature, suggesting that heterotrophy is especially important during the cold season, and that temperature is the predominant factor affecting the coral’s growth. At low temperature, fed samples had higher zooxanthellae density and chl content, possibly for maximizing photosynthetic efficiency. Sexual reproduction investment of C. caespitosa was higher during favourable conditions characterised by high temperatures and zooplankton availability.  相似文献

Occurrence of the odours geosmin and 2-methylisoborneol (MIB) in freshwater environments indicates that odour-producing organisms are commonly occurring. In the present study, we assumed actinomycetes to be a major source of the odours. Seasonal concentrations of odours and abundance of Actinobacteria, which includes actinomycetes and other G+ and high GC bacteria, were determined in one oligotrophic and two eutrophic freshwater streams, as well as in aquacultures connected to these streams, in Denmark. Concentrations of geosmin and MIB ranged from 2 to 9 ng l(-1) and were lowest in the winter. Passage of stream water in the aquacultures increased the amount of geosmin and MIB by up to 55% and 110%, respectively. Densities of actinobacteria were determined by fluorescence in situ hybridization with catalyzed reporter deposition (CARD-FISH) technique and were found to make up from 4 to 38 x 10(7) cells l(-1), corresponding to 3-9% of the total bacterial populations. The lowest densities of actinobacteria occurred in the winter. Filamentous bacteria targeted by the FISH probe made up about 2.7-38% (average was 22%) of the actinobacteria and were expected to be actinomycetes. Combined microautoradiography and CARD-FISH demonstrated that 10-38% (incorporation of 3H-thymidine) and 41-65% (incorporation of 3H-leucine) of the actinobacteria were metabolically active. The proportion of active actinobacteria increased up to 2-fold during passage of stream water in the aquacultures, and up to 98% of the cells became active. Sequencing of 16S rRNA genes in 8 bacterial isolates with typical actinomycete morphology from the streams and ponds demonstrated that most of them belonged to the genus Streptomyces. The isolated actinomycetes produced geosmin at rates from 0.1 to 35 aggeosmin bacterium(-1)h(-1). MIB was produced at similar rates in 5 isolates, whereas no MIB was produced by three of the isolates. Addition of the odours to stream water demonstrated that indigenous stream bacteria were capable of reducing the odours, and that enrichment with LB medium stimulated the degradation. Our study shows that bacterial communities in freshwater include geosmin- and MIB-producing actinobacteria. However, the mechanisms controlling production as well as degradation of the odours in natural waters appear complex and require further research.  相似文献

Cyanobacterial blooms are common in the Baltic Sea. They are dominated by Aphanizomenon flos-aquae and Nodularia spumigena and take place in July–August. Investigations of bloom development using different approaches have been carried out in the Gulf of Finland during recent years. The ship-of-opportunity technique allows to observe the upper layer dynamics from meso- to basin-wide scale with high temporal and spatial frequency at low cost. Unattended measurements on board a commercial ferry along a transect between Tallinn and Helsinki have been conducted for 3 years (1997–1999). The influence of weather conditions—temperature and wind—on the cyanobacterial bloom development was investigated. The formation of cyanobacterial blooms was favoured by warm and calm weather, while in cold and windy conditions other species formed mass occurrences. Water temperature has been found to be the main factor controlling the initiation of the bloom, in general, while vertical stratification appeared to be the critical factor determining the intensity of the bloom at species level. The spatial distribution of the cyanobacterial bloom was determined rather by the wind-forced advection than by the possible vertical transport of nutrients in the areas of the observed upwelling events.  相似文献
