The aim of the investigation was to verify our hypothesis that extreme tolerance of newborn rodents to anoxia is determined by their ability to maintain reduced body temperature and to keep on gasping.Newborn Wistar rats were used. In separate experiments we checked (1) effect of extreme thermal conditions on rectal temperature (Tre) of the newborns in their nests; (2) effect of ambient temperature (Ta) on oxygen consumption; (3) effects of controlled changes in Tre on thermoregulatory and respiratory responses to anoxia and on anoxia tolerance.In their nests rat pups controlled Tre at 32–36 °C while the TreTa difference changed within a range of 1–20 °C. The lowest oxygen consumption of ∼24 ml O2 kg−1 min−1 was recorded at Ta of 32 °C. Pups, exposed to anoxia at their normal Tre of 33 °C, were able to decrease Tre by another 1.7 °C and they kept on extremely slow and quiescent gasping for scheduled 25 min. In contrast, rats at Tre of 37 °C and 39 °C reached a critical phase of accelerated and shallow gasping after 14.95±0.40 min and 9.25±0.30 min, respectively.In conclusion, reduced Tre and unique gasping ability make newborn rats extremely tolerant to asphyxia.  相似文献

Laboratory mice housed under standard vivarium conditions with an ambient temperature (Ta) of ~22 °C are likely to be cold stressed because this Ta is below their thermoneutral zone (TNZ). Mice raised at Tas within the TNZ adapt to the warmer temperatures, developing smaller internal organs and longer tails compared to mice raised at 22 °C. Since mice prefer Tas equal to their TNZ when housed in a thermocline, we hypothesized that mice reared for long periods (e.g., months) in a thermocline would undergo significant changes in organ development and tail length as a result of their thermoregulatory behavior. Groups of three female BALB/c mice at an age of 37 days were housed together in a thermocline consisting of a 90 cm long aluminum runway with a floor temperature ranging from 23 to 39 °C. Two side-by-side thermoclines allowed for a total of 6 mice to be tested simultaneously. Control mice were tested in isothermal runways maintained at a Ta of 22 °C. All groups were given cotton pads for bedding/nest building. Mass of heart, lung, liver, kidney, brain, and tail length were assessed after 73 days of treatment. Mice in the thermocline and control (isothermal) runways were compared to cage control mice housed 3/cage with bedding under standard vivarium conditions. Mice in the thermocline generally remained in the warm end throughout the daytime with little evidence of nest building, suggesting a state of thermal comfort. Mice in the isothermal runway built elaborate nests and huddled together in the daytime. Mice housed in the thermocline had significantly smaller livers and kidneys and an increase in tail length compared to mice in the isothermal runway as well as when compared to the cage controls. These patterns of organ growth and tail length of mice in the thermocline are akin to warm adaptation. Thus, thermoregulatory behavior altered organ development, a process we term behaviorally mediated, warm adaptation. Moreover, the data suggest that the standard vivarium conditions are likely a cold stress that alters normal organ development relative to mice allowed to select their thermal preferendum.  相似文献

Body temperature (Tb) represents one of the key parameters in ecophysiological studies with focus on energy saving strategies. In this study we therefore comparatively evaluated the usefulness of two types of temperature-sensitive passive transponders (LifeChips and IPTT-300) and one data logger (iButton, DS1922L) mounted onto a collar to measure Tb in the field. First we tested the accuracy of all three devices in a water bath with water temperature ranging from 0 to 40 °C. Second, we evaluated the usefulness of the LifeChips and the modified iButtons for measuring Tb of small heterothermic mammals under field conditions. For this work we subcutaneously implanted 14 male edible dormice (Glis glis) with transponders, and equipped another 14 males with data loggers to simultaneously record Tb and oxygen consumption with a portable oxygen analyzer (Oxbox). In one individual we recorded Tb with both devices and analyzed recorded Tb patterns.LifeChips are able to measure temperature within the smallest range from 25 to 40 °C with an accuracy of 0.07±0.12 °C. IPTT-300 transponders measured temperature between 10 and 40 °C, but accuracy decreased considerably at values below 30 °C, with maximal deviations of nearly 7 °C. An individual calibration of each transponder is therefore needed, before using it at low Tbs. The accuracy of the data logger was comparatively good (0.12±0.25 °C) and stable over the whole temperature range tested (0–40 °C). In all three devices, the repeatability of measurements was high.LifeChip transponders as well as modified iButtons measured Tb reliably under field conditions. Simultaneous Tb-recordings in one edible dormouse with an implanted LifeChip and a collar-mounted iButton revealed that values of both measurements were closely correlated. Taken together, we conclude that implanted temperature-sensitive transponders represent an appropriate and largely non-invasive method to measure Tb also under field conditions.  相似文献

The mechanisms underlying physical exercise-induced hyperthermia may be species specific. Therefore, the present study aimed to investigate the effects of exercise intensity and ambient temperature on the core body temperature (T core) of running mice, which provide an important experimental model for advancing the understanding of thermal physiology. We evaluated the influence of different protocols (constant- or incremental-speed exercises), treadmill speeds and ambient temperatures (T a) on the magnitude of exercise-induced hyperthermia. To measure T core, a telemetric sensor was implanted in the abdominal cavity of male adult Swiss mice under anesthesia. After recovering from the surgery, the animals were familiarized to running on a treadmill and then subjected to the different running protocols and speeds at two T a: 24 °C or 34 °C. All of the experimental trials resulted in marked increases in T core. As expected, the higher-temperature environment increased the magnitude of running-induced hyperthermia. For example, during incremental exercise at 34 °C, the maximal T core achieved was increased by 1.2 °C relative to the value reached at 24 °C. However, at the same T a, neither treadmill speed nor exercise protocol altered the magnitude of exercise-induced hyperthermia. We conclude that T core of running mice is influenced greatly by T a, but not by the exercise protocols or intensities examined in the present report. These findings suggest that the magnitude of hyperthermia in running mice may be regulated centrally, independently of exercise intensity.  相似文献

Temperature-dependent development of Spodoptera exigua (Hübner) were evaluated at eight constant temperatures of 12, 15, 20, 25, 30, 33, 34 and 36 °C with a variation of 0.5 °C on sugar beet leaves. No development occurred at 12 °C and 36 °C. Total developmental time varied from 120.50 days at 15 °C to 14.50 days at 33 °C. As temperature increased from 15 °C to 33 °C, developmental rate (1/developmental time) of S. exigua increased but declined at 34 °C. The lower temperature threshold (Tmin) was estimated to be 12.98 °C and 12.45 °C, and the thermal constant (K) was 294.99 DD and 311.76 DD, using the traditional and Ikemoto–Takai linear models, respectively. The slopes of the Ikemoto–Takai linear model for different immature stages were different, violating the assumption of rate isomorphy. Data were fitted to three nonlinear models to predict the developmental rate and estimate the critical temperatures. The Tmin values estimated by Lactin-2 (12.90 °C) and SSI (13.35 °C) were higher than the value estimated by Briere-2 (8.67 °C). The estimated fastest development temperatures (Tfast) by the Briere-2, Lactin-2 and SSI models for overall immature stages development of S. exigua were 33.4 °C, 33.9 °C and 32.4 °C, respectively. The intrinsic optimum temperature (TΦ) estimated from the SSI model was 28.5 °C, in which the probability of enzyme being in its native state is maximal. The upper temperature threshold (Tmax) values estimated by these three nonlinear models varied from 34.00 °C to 34.69 °C. These findings on thermal requirements can be used to predict the occurrence, number of generations and population dynamics of S. exigua.  相似文献

Goat keeping is feasible for smallholder farmers in many world regions especially those best suited for extensive management. However, summertime grazing in arid zones entails major challenges to animal thermoregulation and well-being. An experiment was conducted to evaluate the thermoregulatory performance and selected hemogramic parameters in intensively (INT) or extensively (EXT) managed goat kids (N = 14). We applied a previously established technique to evaluate body thermal state of freely ranging animals, in which contemporaneous temperatures of the core (Tc) and periphery (Tp) are chronically recorded. Animals were initially kept for 12 days under INT management. Subsequently, seven animals were transferred to a grazing pasture and gradually transitioned over a four-day acclimatization period, then kept for the last 22 days under EXT conditions. Water drinking was limited to twice daily in both groups. Excessive solar radiation-induced heat load – with daytime black globe temperatures (Tbg) often exceeding 40 °C – under EXT was primarily responsible (r2 = 0.49; P < 0.05) for 0.57 and 1.72 °C rises in Tc and Tp, respectively, over INT kids. Unlike the typically biphasic pattern noticed for daily temperatures of both body sites in INT goats, that of EXT counterparts became rather polyphasic, whereby water drinking had drastic and prolonged thermolytic effect, inducing 0.40–0.41 and 0.79–1.45 °C declines in Tc and Tp, on midday and afternoon watering bouts, respectively. Despite indication for added daytime heat load, EXT goats displayed lower early morning Tc than INT. All animals exhibited hypohydration, as reflected by rises in hematocrit, serum osmolality, albumin, potassium, and sodium, being more pronounced in EXT conditions. Results emphasize the excessive thermophysiological strain facing grazing animals in arid zones during the summer.  相似文献

The purpose of this study was to investigate the effect of active pre-warming combined with three regimens of fluid ingestion: (1) fluid replacement equal to sweat rate (FF), (2) fluid replacement equal to half the sweat rate (HF), and (3) no fluid replacement (NF). Eight males cycled to voluntary fatigue at 70% of peak power output (PPO) in 31.3±0.4°C, 63.3±1.2% relative humidity in a randomised fashion in either of FF, HF or NF conditions. For each trial the time to fatigue test was preceded by 2×20 min active pre-warming periods where subjects also cycled at 70% PPO. Subjects commenced each exercise period with identical rectal temperatures (Tre). The rate of increase in Tre for each condition during the first 20 min of active pre-warming was not different. However, the rate of increase in Tre was significantly reduced in the second active pre-warming period for all fluid conditions but no differences between conditions were noted. During the fatigue test, the rate of increase in Tre for FF was 0.29°C h−1 and 0.58°C h−1 for HF but were not significantly different. The rate of increase in Tre for the NF trial was 0.92°C h−1 and was significantly higher compared to the FF trial. Overall mean skin temperatures and mean body temperatures were higher for NF compared to FF and HF. The rate of heat storage during the fatigue test was similar for FF (80.1±11.7 W m−2) and HF (73.0±13.7 W m−2) conditions but increased to 155.8±31.2 W m−2 (P<0.05) in the NF trial. The results indicate that fluid ingestion equal to sweat rate has no added benefit over fluid ingestion equal to half the sweat rate in determining time to fatigue over 40 min of sub-maximal exercise in warm humid conditions. Fluid restriction accelerates the rate of increase in Tre after 40 min of exercise, thereby reducing the time to fatigue. The data support the model that anticipation of impending thermal limits reduces efferent command to working skeletal muscle ensuring cellular preservation.  相似文献

Bats are among the most heterothermic mammals, with nearly all species investigated under free-ranging conditions to date exhibiting some degree of daily torpor and/or hibernation. We investigated thermoregulation during late winter by seven Nycteris thebaica in a warm, semi-arid habitat in northern South Africa, using temperature-sensitive transmitters to measure skin temperature (Tskin). Unexpectedly, we found no evidence for any expression of daily torpor or hibernation based on a total of 86 days of data from 7 bats (one male and six females), despite air temperatures as low as ~10 °C. Instead, daytime Tskin was distributed unimodally with most values in the 33–35 °C range, and a minimum Tskin of 28.4 °C at a roost temperature of 24.6 °C. There are several possible reasons why N. thebaica may avoid torpor, including predation in roosts, and the long nightly foraging periods of this species compared to many others.  相似文献

Rapidly cooling pigs after heat stress (HS) results in a pathophysiological condition, and because rapid temperature fluctuations may be associated with reduced reproductive success in sows, it lends itself to the hypothesis that these conditions may be linked. Objectives were to determine the effects of rapid cooling on thermal response and future reproductive success in pigs. Thirty-six replacement gilts (137.8±0.9 kg BW) were estrus synchronized and then 14.1±0.4 d after estrus confirmation, pigs were exposed to thermoneutral conditions (TN; n=12; 19.7±0.9°C) for 6 h, or HS (36.3±0.5°C) for 3 h, followed by 3 h of rapid cooling (HSRC; n=12; immediate TN exposure and water dousing) or gradual cooling (HSGC; n=12; gradual decrease to TN conditions) repeated over 2 d. Vaginal (TV) and gastrointestinal tract temperatures (TGI) were obtained every 15 min, and blood was collected on d 1 and d 2 during the HS and recovery periods at 180 and 60 min, respectively. Pigs were bred 8.3±0.8 d after thermal treatments over 2 d. Reproductive tracts were collected and total fetus number and viability were recorded 28.0±0.8 d after insemination. HS increased TV and TGI (P=0.01; 0.98 °C) in HSRC and HSGC compared to TN pigs. During recovery, TV was reduced from 15 to 105 min (P=0.01; 0.33 °C) in HSRC compared to HSGC pigs, but no overall differences in TGI were detected (P<0.05; 39.67 °C). Rapid cooling increased (P<0.05) TNFα compared to HSGC and TN pigs during recovery-d 1 (55.2%), HS-d 2 (35.1%), and recovery-d 2 (64.9%). Viable fetuses tended to be reduced (P=0.08; 10.5%) and moribund fetuses tended to be increased (P=0.09; 159.3%) in HSRC compared to HSGC and TN pigs. In summary, rapid cooling prior to breeding may contribute to reduced fetal viability and reproductive success in pigs.  相似文献

Eggs of Muscovy duck and domestic fowl were incubated at 37.5°C and relative air humidity of 70%. After 60 min the influence of lowered (31.5°C, 34.5°C and 28.5°C) Ta on heat production (HP) and temperature of the allantoic fluid (Taf) as a measure of core temperature was estimated for 3 h. The Q10 was calculated using the relationships between Taf and HP. HP dropped with decreasing Taf. The Q10 crossed the 2.0 threshold mostly between 34°C and 36°C Taf in dependence of Ta. Precocial birds are endothermic in the last third of incubation but these reactions have only an ultimate but not a proximate influence on efficiency of thermoregulation.  相似文献

Medicinal leeches (Hirudo verbana) thermoregulate with respect to their sanguivorous feeding behavior. Immediate postprandial preferences are for warmer than their initial acclimation temperature (Ta, 21 °C, Petersen et al. 2011), while unfed leeches have a lower preferred temperature (Tpref, 12.5 °C). This may reduce energy expenditure and defer starvation if feeding opportunities are limited. Energetic benefits may have an associated cost if low temperatures reduce mobility and the ability to locate further hosts. These costs could be limited if mobility is unimpaired at low temperatures, or if acclimation can restore locomotor performance to the levels at Ta. The transition from Ta to the unfed Tpref significantly reduced speed and propulsive cycle frequency during swimming, and extension and retraction rates during crawling. Aerobic metabolic rate was also reduced from 0.20±0.03 W kg−1 at Ta to 0.10±0.03 W kg−1 at Tpref. The Q10 values of 1.7–2.9 for energetic and swimming parameters indicate a substantial temperature effect, although part of the decline in swimming performance can be attributed to temperature-related changes in water viscosity. 6 weeks at Ta resulted in no detectable acclimation in locomotor performance or aerobic metabolism. The energetic savings associated with a lower Tpref in unfed leeches effectively doubled the estimated time until depletion of energy reserves. Given that some mobility is still retained at Tpref, and that acclimation is in itself costly, the energetic benefits of selecting cooler temperatures between feedings may outweigh the costs associated with reduced locomotor performance.  相似文献

Most mammals are known to have clear circadian rhythms of body temperature (Tb) and metabolic rate. Large parts of the rhythms correspond to the oscillation of nonshivering thermogenesis (NST), dependent on visceral organ mass, and, affected by mass of brown adipose tissue (BAT). I tested whether: (1) a different levels of BMR result in respective changes of Tb values and the magnitude of daily RMR oscillations both within and below thermoneutrality; (2) the amplitude of daily variation of RMR depends on ambient temperature (Ta). I studied: (1) daily variation of body temperature at Ta of 23 °C, and (2) the rate of resting metabolism (RMR) within and below thermoneutrality at the time of minimum and increasing Tb (minimum and maximum NST capacity), in two lines of laboratory mice subjected to divergent, artificial selection toward high (HBMR) and low (LBMR) basal rate of metabolism (BMR). All mice had a clear circadian rhythm of Tb with minimum of 36.4±0.2 °C at 7:00 and maximum of 37.8±0.2 °C at 21:00. Their RMR measured below thermoneutrality exhibited significant daily variation, with the maximum between 16:00 and 19:00, when Tb was rising. Within thermoneutral zone (TNZ) I found between-line, but not between-time, differences in RMR. All between-line differences in RMR could be explained by the magnitude of BMR. I did not find any between-line differences of RMR value in temperatures below thermoneutrality. The amplitude of daily variation of RMR measured below TNZ depended neither on the Ta value nor on level of BMR (or visceral organs).  相似文献

The red-tailed phascogale is a small arboreal dasyurid marsupial that inhabits semi-arid to arid regions of Western Australia's wheat belt. Its body mass (34.7 g) is only ~15% of that predicted based on its phylogenetic position among other dasyuromorphs; we interpret this as an adaptation to its scansorial and semi-arid/arid lifestyle. The standard physiology of this species at a thermoneutral ambient temperature of 30 °C conforms to that of other dasyurid marsupials; body temperature (34.7 ± 0.37 °C), basal metabolic rate (0.83 ± 0.076 mL O2 g?1 h?1), evaporative water loss (1.68 ± 0.218 mg H2O g?1 h?1) and wet thermal conductance (3.8 ± 0.26 J g?1 h?1 °C?1) all fall within the 95% predication limits for the respective allometric relationships for other dasyurid species. Thermolability confers an energy savings at low Ta and water savings at high Ta. Torpor, observed at low Ta, was found to be more beneficial for energy savings than for water economy. The red-tailed phascogale therefore has a physiology suitable for the challenges of arid environments without any obvious requirement for adaptations to its scansorial lifestyle, other than its considerably lower-than-expected body mass.  相似文献

The effect of relative humidity (RH) and temperature on CO2 assimilation (An), stomatal conductance (Sc), transpiration rate (Tr), chlorophyll content, fresh and dry weight, leaf length, leaf area, leaf width, formation of new root and survival rate have been assayed in Doritaenopsis in growth chamber after 1 month of acclimatization. Reduced growth was observed at below and above 25 °C whereas it was increased with increasing humidity. Relative water content (RWC) was decreased at 50% and 70% humidity after second day of transfer and recovered completely with the progression of acclimatization. RWC also reduced at high temperature but recovered slowly and a gradual decrease of RWC was observed at 15 °C. A visual symptom of severe leaf tip burn was observed at 50–70% humidity and at 35 °C during acclimatization. At 15 °C and 50% humidity sudden decrease of photosynthetic efficiency (Fv/Fm) was observed, which could not recover in temperature treated plantlets during acclimatization period. Chlorophyll content increased with increasing humidity and at 15 and 35 °C chlorophyll content was decreased compared to 25 °C. Chlorophyll a/b ratio was unchanged while total chlorophyll/carotenoids ratio was increased from low to high temperature. Exposure of plantlets to high temperature led to a noticeable decrease in An, Sc and Tr, and at 15 °C they were more decreased whereas significant differences were not observed in the parameters tested under humidity after 25 days of acclimatization. During daytime at 15 °C, increase in An, Sc and Tr indicates the plantlets adaptability in the new environment. The peroxidase activity remained unaffected in all humidity stress whereas low temperature increased the peroxidase activity compared to high temperature. These finding suggests that photosynthetic properties was greatly affected by air temperature conditions with a reduction of An, Sc and Tr at 15 and 35 °C compared to humidity stress that played a greater role in limiting photosynthesis.  相似文献

Most reptiles thermoregulate to achieve body temperatures needed for biological processes, such as digestion and growth. Temperatures experienced during embryogenesis may also influence post-hatching growth rate, potentially through influencing post-hatching choice of temperatures. We investigated in laboratory settings whether embryonic temperatures (constant 18 °C, 21 °C and 22 °C) influence selected body temperatures (Tsel) of juvenile tuatara (Sphenodon punctatus), providing a possible mechanism for differences in growth rates. We found that incubation temperature does not influence Tsel. Although the average daily mean Tsel was 21.6 ± 0.3 °C, we recorded individual Tsel values up to 33.5 °C in juvenile tuatara, which is higher than expected and above the panting threshold of 31–33 °C reported for adults. We found diel patterns of Tsel of juvenile tuatara, observing a general pattern of two apparent peaks and troughs per day, with Tsel being significantly lower around dawn and at 1500 h than any other time. When comparing our results with other studies on tuatara there is a remarkable consistency in mean Tsel of ~ 21 °C across tuatara of different ages, sizes and acclimatization histories. The ability of juvenile tuatara to withstand a wide range of temperatures supports their former widespread distribution throughout New Zealand and warrants further investigation into their plasticity to withstand climate warming, particularly where they have choices of habitat and the ability to thermoregulate.  相似文献

New technologies afford convenient modalities for skin temperature (TSKIN) measurement, notably involving wireless telemetry and non-contact infrared thermometry. The purpose of this study was to investigate the validity and reliability of skin temperature measurements using a telemetry thermistor system (TT) and thermal camera (TC) during exercise in a hot environment. Each system was compared against a certified thermocouple, measuring the surface temperature of a metal block in a thermostatically controlled waterbath. Fourteen recreational athletes completed two incremental running tests, separated by one week. Skin temperatures were measured simultaneously with TT and TC compared against a hard-wired thermistor system (HW) throughout rest and exercise. Post hoc calibration based on waterbath results displayed good validity for TT (mean bias [MB]=−0.18 °C, typical error [TE]=0.18 °C) and reliability (MB=−0.05 °C, TE=0.31 °C) throughout rest and exercise. Poor validity (MB=−1.4 °C, TE=0.35 °C) and reliability (MB=−0.65 °C, TE=0.52 °C) was observed for TC, suggesting it may be best suited to controlled, static situations. These findings indicate TT systems provide a convenient, valid and reliable alternative to HW, useful for measurements in the field where traditional methods may be impractical.  相似文献

The purpose of this study was to evaluate the temperature response of photosynthesis in two common bean genotypes differing in crop yield when grown under warm conditions. The cultivar Nobre is sensitive to high temperatures, whereas Diplomata shows better crop yield under high temperatures. Plants were grown in a greenhouse prior to transferring to a controlled environment cabinet for the temperature treatments. In a first experiment, 30 days-old plants were subjected to a short exposure (1 day) at temperatures that varied from 9 °C to 39 °C. Diplomata had lower net CO2 assimilation rate (A) at 15 °C and 21 °C, but higher from 27 °C to 39 °C. Photosynthetic parameters calculated from modeling the response of A to the intercellular CO2 concentration suggested that the different temperature responses of the two genotypes are caused by different rates of diffusion of CO2 to the assimilation site, not by differences in biochemical limitations of photosynthesis. While stomatal conductance (gs) did not differ between the genotypes, mesophyll conductance (gm) was slightly greater for Nobre at 15 °C, but much higher in Diplomata from 21 °C to 39 °C. In a second experiment, no difference was observed in biomass accumulation between the two genotypes after growth for 24 days under a 35/20 °C (day/night) regime. Hence, the differences in photosynthesis did not cause variation in plant growth at the vegetative stage. The differential genotypic response of gm to temperature suggests that gm might be an important limitation to photosynthesis in Nobre, the common bean genotype sensitive to elevated temperature. However, more studies are needed employing other methods for gm evaluation to validate these results.  相似文献

The functional properties of extremophilic Dictyoglomus thermophilum xylanase (XYNB) and the N-terminal disulphide-bridge mutant (XYNB-DS) were studied at high pressure and temperature. The enzymes were quite stable even at the pressure of 500 MPa at 80 °C. The half-life of inactivation in these conditions was over 30 h. The inactivation at 80 °C in atmospheric pressure was only 3-times slower. The increase of pressure up to 500 MPa at 80 °C decreased only slightly the enzyme's stability, whereas in 500 MPa the increase of temperature from 22 to 80 °C decreased significantly more the enzyme's stability. While the high temperature (80–100 °C) decreased the enzyme reaction with short xylooligosaccharides (xylotetraose and xylotriose), the high pressure (100–300 MPa) had an opposite effect. Temperature is one of the most important abiotic factors affected by climate change. It determines physiological processes, ecological patterns and establishes the limits of geographic distribution of species. The induced thermal stress frequently results in physiological and behavioral responses and, in extreme cases, may lead to mortality episodes. Scrobicularia plana and Cerastoderma edule behavioral and mortality responses to temperature were evaluated. Specimens were sampled in the Mondego estuary (Portugal), acclimated and exposed to different temperature treatments (5–35 °C). Individual activity and mortality were registered during 120 h laboratory assays. Both species showed a thermal optimum for their activity (S. plana: 15–23 °C; C. edule: 20–23 °C), and survival was mainly affected by high temperature (S. plana: LC50120 h = 28.86 °C; C. edule: LC50120 h = 28.01 °C), with 100% mortality above critical values (≥32 °C). Results further indicated that both species are more affected the higher the temperature and the longer the exposure time. This study indicates that the occurrence of extreme climatic events, especially heat waves, may be particularly impairing for these species.  相似文献

The demand on thermostable d-lactate dehydrogenases (d-LDH) has been increased for d-lactic acid production but thermostable d-DLHs with industrially applicable activity were not much explored. To identify a thermostable d-LDH, three d-LDHs from different Lactobacillus jensenii strains were screened by genome mining and then expressed in Escherichia coli. One of the three d-LDHs (d-LDH3) exhibited higher optimal reaction temperature (50 °C) than the others. The T5010 value of this thermostable d-LDH3 was 48.3 °C, much higher than the T5010 values of the others (42.7 and 42.9 °C) and that of a commercial d-lactate dehydrogenase (41.2 °C). The Tm values were 48.6, 45.7 and 55.7 °C for the three d-LDHs, respectively. In addition, kinetic parameter (kcat/Km) of d-LDH3 for pyruvate reduction was estimated to be almost 150 times higher than that for lactate oxidation at pH 8.0 and 25 °C, implying that d-lactate production from pyruvate is highly favored. These superior thermal and kinetic features would make the d-LDH3 characterized in this study a good candidate for the microbial production of d-lactate at high temperature from glucose if it is genetically introduced to lactate producing microbial.  相似文献
