Portions of stems from the base of asparagus spears (Asparagus officinalis L. cv. Connovor Collossus) were dissected to give the following tissues: (1) pith, which was free of vascular bundles, (2) two surrounding layers, parenchyma and fibre I and II (PFI and PFII), containing parenchyma and vascular bundles, (3) sclerenchyma sheath, (4) epidermis and sub-epidermal layers and (5) asparagus vascular fibre (AVF). The alcohol-insoluble residues (AIRs) from these tissues were shown to be free of starch. They were analysed for moisture and protein, and the component sugars were released by two hydrolytic procedures, which helped to distinguish the sugars from non-cellulosic polysaccharides and cellulose. The AIRs from pith and epidermal tissues were relatively low in xylose, but were rich in cellulosic glucose, and sugars associated with pectic polysaccharides such as galacturonic acid, galactose and arabinose. Their major component polysaccharides (in decreasing amounts) were inferred to be pectic polysaccharides, cellulose, and hemicelluloses. AIR from sclerenchyma was rich in glucose and xylose, suggesting the presence of much cellulose and (acidic) xylans. The AIRs of PFI, PFII and AVF contained significant amounts of xylose in addition tn other sugars, and the major polysaccharides inferred to be present were pectic polysaccharides, cellulose and hemicelluloses, a significant proportion of which may be acidic xylans. Methylation analysis of the AIRs confirmed the above inferences. The bulk of the glucosyl residues were (1–4)-linked, and there were small but significant amounts of (1–4, 6)-linked glucosyl residues (the linkage characteristic of xyloglucans) in all the preparations. The presence of (1–4)-linked galactosyl, (1–5)-linked arabinosyl, terminal galactosyl, terminal arabinosyl, (1–2)- and (1–2, 4)-linked rhamnosyl residues in all the AIRs except that from sclerenchyma, confirmed the presence of significant levels of pectic polysaccharides in all the parenchyma tissues. All the preparations containing vascular tissues contained significant amounts of (1–4)-linked xylosyl residues, probably derived from acidic xylans. Even in the AIR of pith, a significant amount of (1–4)-linked xylosyl residues were detected. This may be due to the ability of these cells and the parenchyma cells associated with the vascular bundles, to undergo lignification in mature asparagus plants.  相似文献

芦笋皂苷是以芦笋下脚料为原料提取出来的一种糖甙化合物。目的是充分利用再生资源,变废为宝,同时解决芦笋下脚料废弃物所造成的环境污染问题。实验探讨了乙醇浓度、液料比、温度、时间等工艺条件对芦笋皂苷提取率的影响,筛选出最佳提取工艺:乙醇浓度95 %、液料比(V/W)为6∶1、温度90℃、时间4h。从新鲜芦笋下脚料、芦笋干品提取皂苷的平均得率分别为1.70 %、4.01%。芦笋皂苷经氧化铝柱层析分离,以40%乙醇的溶液为洗脱剂,洗出曲线为单一对称峰。采用UV、IR、HPLC等色谱对芦笋皂苷的结构特点、糖基组成进行初步分析。结果显示芦笋皂苷具有呋喃甾烷的结构特征,其糖基由木糖(Xyl)、岩藻糖(Fuc)、阿拉伯糖(Ara)组成,摩尔比为Xyl∶Fuc∶Ara =1 0∶0 13∶19 4 2 ,平均分子质量(Mw)为185 0 0。  相似文献

对番茄内生细菌数量动态及其对青枯病的生物防治研究结果表明:番茄内生细菌可来源于种子内部。番茄不同生育期,内生细菌数量最多在成株期,其中抗病品种根、茎分别为24.3×104CFU/g鲜重和22.9×104CFU/g鲜重,感病品种根、茎分别为9.8×104CFU/g鲜重和13.4×104CFU/g鲜重。抗病品种中具有拮抗青枯菌的内生细菌菌株为17个,感病品种中7个。部分内生细菌具促进番茄种子萌发和防治番茄青枯病的作用,其中5R和3R内生菌株的防病效果分别达91.7%和81.3%。  相似文献

本试验以芦笋新品种"津宁"为材料,探讨植物生长调节剂对不同外植体产生丛生芽的效应。试验结果表明,NAA、6-BA和KT三者配合使用有利于丛生芽的增殖:以茎尖为增殖外植体时,培养基用MS+NAA0.1 mg/L+6-BA0.2~0.3 mg/L+KT 0.1 mg/L,30 d时平均每瓶可以产生20.5个丛生芽;以茎中段为外植体时,培养基用MS+NAA 0.1 mg/L+6-BA 0.3~0.4 mg/L+KT 0.1 mg/L,30 d时平均每瓶可产生33.5个丛生芽;以茎基为外植体时,培养基用MS+NAA 0.1 mg/L+6-BA 0.3~0.5 mg/L+KT 0.1 mg/L,30 d时平均每瓶可以产生43.1个丛生芽。进行试管苗工厂化生产时,增殖外植体以茎基为最佳。  相似文献

Cultured cells and somatic embryos derived from the mesophyll tissue of asparagus (Asparagus officinalis L.) were cryopreserved by vitrification. The vitrification solution (PVS) contains (w/v) 22% glycerol, 15% ethylene glycol, 15% propylene glycol and 7% DMSO in Murashige-Skoog medium enriched with 0.5M sorbitol. After initial cryoprotection with sorbitol supplemented MS medium containing 12% ethylene glycol, cells or embryos were exposed stepwise to 85% PVS at 0°C. They were loaded into 0.5 ml transparent straws, and were then plunged directly into liquid nitrogen. After rapid warming, PVS was removed and diluted stepwise. The highest survivals of vitrified cells and embryos were about 65 and 50%, respectively. The endosperm callus has been induced from the young endosperm of Asparagus officinalis L. on the MS supplemented with auxin. The induction frequency of callus amounts to 65.9%–83.1%. When the callus was transferred to the medium supplemented with lower concentration of NAA 0.1 ppm or containing BA 1 ppm and NAA 0.5 ppm, the differentiation of shoots, roots and a few embryoids began to occur. A few calluses and embryoids can develop into plantlets. The chromosome number in the cells from the same root tip and shoot apex of endosperm plantlet is very unstable. They can be euploids (n=10, 2n=20, 3n=30, 4n=40). or aneupl0ids (n=6, 7, 17, 25, 53).  相似文献

白芦笋的不同筛选株系之间生根能力存有差异;培养温度是影响生根率的关键因素,高温[≥(25±1)℃]不利生根,发根的适宜培养温度为(20±1)℃;低光照强度(≤25mmol·m-2·s-1)对生根不利,适宜的光合光量子通量密度(PPFD)为40μmol·m-2·s-1;PP333、嘧啶醇不能提高试管苗生根率,且浓度过高还会降低生根率和抑制植株生长。  相似文献

One new (Sarsasapogenin O) and seven known steroids were isolated from the roots of Asparagus officinalis L. Their structures were elucidated on the basis of spectroscopic analysis, including various 2D-NMR techniques, hydrolysis,and by comparison of spectral data of known compounds. These compounds together with nine steroids which were previously isolated from this plant, were tested for cytotoxic activity. Among them, eight compounds displayed significant cytotoxicities against human A2780, HO-8910, Eca-109, MGC-803, CNE, LTEP-a-2, KB and mouse L1210 tumor cells.  相似文献

Changes in the cell wall composition of stems of asparagus ( Asparagus officinalis L. cv. Connovor Collossus) during maturation and during storage in modified atmospheres were investigated. Alcohol-insoluble residues from portions of stems were analysed for changes in alkali-soluble phenolics and in the constituent sugars and their glycosidic linkages. Maturation-related changes down the stem involved increased levels of (l–4)-linked xylosyl residues from xylans, cellulose and phenolics, which arise from the lignified vascular and sclerenchyma tissues, and a decrease in the levels of (l–5)-linked arabinosyl residues from pectic arabinans from the parenchy-matous tissues. Storage under aerobic conditions permitted stem elongation in the apical sections and secondary thickening in the basal sections. This was due to continued tissue maturation. Storage under anaerobic conditions inhibited such changes. In accordance with these effects of storage on stem physiology, maturation-related changes in cell wall components were enhanced by storage only under aerobic conditions. However, levels of (1–4)-linked galactosyl residues which remained relatively constant during maturation, decreased substantially (ca 50%) in all stem sections during both aerobic and anaerobic storage.  相似文献

RP-HPLC法测定芦笋中黄酮类化合物芦丁的含量   总被引:6,自引:0,他引:6  
以芦丁标准品为对照,利用反相高效液相色谱法对芦笋中黄酮类化合物芦丁的含量进行定量测定。采用Agilent Eclipse XDB-C18色谱柱,柱温25℃,流动相由甲醇-水-磷酸(55∶44.5∶0.5)组成,流速为0.7 mL/min,检测波长390 nm。结果表明黄酮类化合物中各组分基线分离良好;进样量在0.07~0.7μg/20μL范围内,峰面积A与进样浓度C呈良好的线性关系,回归方程为A=1 5176C-10.388,相关系数R2=0.999 4;加样回收率为101.051%,RSD=3.306%;以保留时间和峰面积作精密度试验,RSD分别为0.199%和1.24%。该方法样品处理简单,准确度高,精密度好,适合于芦笋中芦丁含量的测定。  相似文献

石刁柏胚性愈伤组织继代过程中,添加浓度为10 mg·L^-1的外源腐胺能有效地保持愈伤组织的胚胎发生能力,并减少愈伤组织的褐化,但不能提高愈伤组织的体细胞胚的诱导率.腐胺处理过的胚性愈伤组织的内源腐胺含量明显提高.这可能是外源腐胺保持细胞胚性、降低褐化程度的原因.  相似文献

There were two different stages during the root regeneration from the anther derived spear of Asparagus officinalis L. c v. Marry Washington 500, the root primordia differentiation, and the development stage of root primordia into young root. Differentiation of root primordia was enhanced by amplified nutrient supplement and lowered NAA concentration, both of which were favorable to callus formation and sustained decrease of endogenous ABA at the transection of the base of the spear. Moreover, further development of the root primordia was promoted by appropriate limitation of water supply and by effective aeration.  相似文献

The superoxide dismutase activities in callus and somatic embryoids at different developmental stages of Asparagus officinalis L. were determined by means light of induced oxidation-reduction of nitro blue tetrazolium. It was shown that the superoxide dismutase activity was higher in callus than in somatic embryoids at different developmental stages. The activity increased with growth, differentiation and maturation of somatic embryoids during somatic embryogenesis. The relations between superoxide dismutade activity and somatic embryogenesis were discussed.  相似文献

用高效液相色谱法分析石刁柏愈伤组织胚性细胞诱导过程中不同时期内源激素和多胺含量的结果表明,在胚性细胞诱导过程中,Put和GA3一直呈上升趋势,胚性细胞出现时,IAA、Put和GA3含量都达到最高水平,显示高含量的IAA以及高比例的Pu“(Spd+Spm)可能有利于胚性细胞的形成。  相似文献
