Research on drunken horse grass (Achnatherum inebrians)-endophytic fungi (Epichloë gansuensis, E. inebrians) represented an important area of endophyte research in China. Compared with tall fescue (Festuca arundinacea)-endophyte (E. coenophiala) in the United States and perennial ryegrass (Lolium perenne)-endophyte (E. festucae var. lolii) in New Zealand, A. inebrians-endophyte research in China is becoming a leading branch of endophyte research. The studies of A. inebrians-endophyte symbionts over the past three decades were reviewed, mainly including endophyte distribution, detection methods, infection rate, fungal diversity, stress tolerances of drought, cold, salt, heavy metal, pest, disease and their mechanisms, secondary metabolite of alkaloids, feeding experiments and its ecological functions etc. Research reveals that drunken horse grass itself has no toxicity, the grazed animals will be intoxicated if the grass bears infection by Epichloë endophytes and produces ergot alkaloids. Future research will focus on E. gansuensis and E. inebrians genomics and endophyte functions. In other research we will create new grass cultivars that are 1) sanitized of the endophyte and non-toxic, 2) utilizing novel endophytes that deter birds feeding and 3) utilizing endophytes that are modified to produce novel fungicides, pesticides or medicines. © 2018 Editorial Office of Chinese Journal of Plant Ecology. It is well known that the earthworm's activities can increase the availability of soil nutrients, improve soil structure, and enhance the biomass of plants in uncontaminated soil. Recently, many researchers found that some metal-tolerant earthworms can survive and even change the fractional distribution of heavy metals in contaminated soil. Furthermore, it has been revealed that earthworms are able to increase metal availability, and therefore, accumulate more metals in plants through their burrowing and casting activity. It is clear that the influence of soil animals is an important factor for phyto-remedation that must be taken into account. ~In this article, the authors studied some effects of addition of earthworms (Metaphire guillelmi), corn straw, and in combinations of earthworms and corn straw on the growth and Cu uptake by ryegrass in Cu contaminated pot soils. The experiment consisted of four levels of Cu addition (0, 100, 200, 400 mg·kg?1) and four treatments. The treatments were 1. control (CK); 2.straw mulching only (M); 3. earthworm additions to soil only (E); and 4.straw mulching plus earthworm additions (ME). Each treatment had three replicates. 10 seeds of ryegrass (Lolium multiflorum) were sowed in each pot and harvested after 30 days. After 30 days of incubation, all earthworms were found to be alive and the pot soils were burrowed through by earthworms. Results showed that the biomass of earthworm declined with the increase of the dosage of Cu additions. The biomass of earthworm increased significantly in treatment 4 (ME) as compared with treatment 3 (E). Not only the earthworms could get more food from the straw, but also could counteract some negative effects of Cu on the earthworms. The rates of straw decomposition in ME treatment increased by about 58.11% ?77.32%. The earthworm activities increased root biomass of ryegrass significantly, but did not show the effect on plant root growth. On the contrary, straw enhanced roots biomass significantly instead of shoots biomass. It was also found that the concentration of Cu in the plant shoot and the plant root, as well as plant Cu uptake were enhanced by earthworm's activities and straw mulching. The increased amount by straw mulching was lower than that of earthworms (E). The treatment of the earthworm–straw mulching combinations enhanced plant Cu concentration, and the amount increased by it was lower than that of the earthworm treatment (E) but higher than that of straw mulching treatment (M). The accumulation factors of copper in the shoots of ryegrass were increased by 31.22% ?121.07%, 2.12% ?61.28% and 25.56% ?132.64%, respectively, in treatment 3(E), 2(M), and 4(ME), respectively. In conclusion, the earthworm activities, straw-mulching and their interactions may have potential roles in elevating phyto-extraction efficiency in low to medium level Cu contaminated soil.  相似文献

The aim of the present study was to determine the effect of environmental stresses (temperature, pH, osmotic pressure, high hydrostatic pressure (HHP)) on the antibiotic susceptibility of 12 different pathogenic Vibrio parahaemolyticus isolates. Isolates were subjected to growth at (30 ℃and 37 ℃), an osmotic pressure of (1% and 6% NaCl), a media at pH (6.0 and 9.0) and three different HHP treatment (180, 250, 300 MPa). The minimal inhibitory concentrations (MICs) of tested antibiotics used against unstressed (control), stressed or post-stressed isolates were determined using the broth microdilution method. The study found that incubation under increased salt (6%), reduced salt (1%) and increased pH (9.0) conditions were commonly associated with increased antibiotic resistance. Incubation at 30 ℃ temperature, reduced pH (6.0) and HHP treatment were commonly associated with decreased antibiotic resistance. Besides, both ciprofloxacin and cefotaxime kept a constant MIC during almost all stress challenges. Thus our data demonstrate that exposure of V. parahaemolyticus to some stress conditions may contribute on the rapid development of antibiotic resistance in this food borne pathogen.  相似文献

An experiment was designed to determine the effect of the fungal endophyte Neotyphodium Iolii on the growth, physiological parameters and mineral element content of perennial ryegrass (Lolium perennel L.), when growing at two N supply levels. Endophyte infection had a significant positive effect on both shoot and root growth of ryegrass, but this difference was only significant in the high N supply treatment. At high N supply, endophyte-infected (El) plants accumulated more soluble sugar in the sheath and the root than endophyte-free (EF) plants. Endophyte infection affected mineral element concentrations in the root more than in the shoot. We found a significant effect of endophyte infection on B, Mn and Mg in the root, but significant effect was only found on B in the shoot. El plants tended to accumulate less B in the shoot at both N levels, but accumulated more B, Mn and Mg in the root at low N levels. The difference of growth parameters in different periods was significant. The content of soluble sugar and crude protein in the sheath were also dependent on the growth stages of both El and EF plants.  相似文献

Epichloё内生真菌(epichloё endophytes)是禾本科植物的共生真菌,开展其多样性研究对菌株的生物学特性和进化规律的了解以及开发利用都有着十分重要的意义。本研究筛选了12条随机引物对24株epichloё内生真菌进行RAPD扩增,探讨真菌种类、宿主种类、采集地和菌株间的遗传多样性。结果表明,宿主为拂子茅Calamagrostis sp.、雀麦Bromus sp.、披碱草Elymus sp.、小颖羊茅Festuca parvigluma Steud.的菌株分别独立聚为一个分枝;而宿主为鹅观草Roegneria spp.的Neotyphodium属菌株聚类情况较为复杂;宿主为早熟禾Poas pp.的菌株分别在三个不同的分枝,表现出丰富的遗传多样性。以上结果反应了我国具有丰富的epichloё内生真菌资源,它们和其宿主之间有着比较明显的相互关系。本研究还显示,采集于南京的拂子茅与其内生真菌共生可能发生在较久远的年代。  相似文献

作者首先研究Epichloё yangzii在植株上的人工杂交,明确了供试菌株的交配型。然后将分离自无子座鹅观草属植物的23株"Neotyphodium属"真菌孢子分别与E.yangzii的子座杂交,其中发现有21株与E.yangzii(mat-1,mat-2)杂交不亲和,有2株与E.yangzii(mat-1)杂交亲和。利用tubB基因片段对8株"Neotyphodium属"真菌菌株进行系统发育分析,结果表明与E.yangzii杂交亲和的2个菌株和E.yangzii聚为一枝,而其它6个菌株形成独立的分枝,进一步证实了这2个菌株是有时在宿主植物上不形成子座的E.yangzii。这说明了在宿主植物上的人工杂交是区别有时不产子座的E.yangzii和Neotyphodium属真菌的有效手段。  相似文献

The Fo membrane domain of FoF1-ATPase complex had been purified from porcine heart mitochondria. SDS-PAGE with silver staining indicated that the purity of Fo was about 85% and the sample contained no subunits of F1-ATPase. The purified Fo was reconstituted into liposomes with different phospholipid composition, and the effect of CL (cardiolipin), PA (phosphatidic acid), PI (phosphatidylinositol) and PS (phosphatidylserine) on the H+ translocation activity of Fo was investigated. The results demonstrated that CL, PA and PI could promote the proton translocation of Fo with the order of CL>PA>>PI, while PS inhibited it. Meanwhile ADM (adriamycin) severely impaired the proton translocation activity of Fo vesicles containing CL, which suggested that CL's stimulation of the activity of reconstituted Fo might correlate with its non-bilayer propensity. 内生真菌与丛枝菌根(AM)真菌是构成草原生态系统的重要组成部分.内生真菌会抑制其宿主植物的AM真菌侵染率.本研究以感染2种香柱菌属内生真菌[Epichloё gansuensis(Eg)和E. sibirica(Es)]的天然禾草羽茅为供试材料,进行体外纯培养的内生真菌培养滤液、感染内生真菌的羽茅叶片(包括鲜叶和枯叶)浸提液,以及根系分泌物对摩西球囊霉(Gm)和幼套球囊霉(Ge)2种AM真菌孢子萌发影响试验.结果表明: 香柱菌属内生真菌的培养滤液会显著抑制2种AM真菌孢子的萌发,而感染香柱菌属内生真菌的羽茅根系分泌物只对Ge孢子萌发有显著抑制作用,且上述抑制作用与内生真菌种类无关;鲜叶浸提液对Gm和Ge的孢子萌发率均无显著影响,而枯叶浸提液对Ge的孢子萌发有显著抑制作用.在自然生态系统中,香柱菌属内生真菌通常存在于宿主植物体内,可能通过影响宿主植物的根系分泌物来影响AM真菌孢子的萌发.  相似文献

Using nystatin-perforated whole-cell recording configuration, the modulatory effect of N-methyl-D-aspartate (NMDA) on γ-aminobutyric acid (GABA)-activated whole-cell currents was investigated in neurons freshly dissociated from the rat sacral dorsal commissural nucleus (SDCN). The results showed that: (i) NMDA suppressed GABA- and muscimol (Mus)-activated currents (IGABA and IMUS), respectively in the Mg2 -free external solution containing 1 μmol/L glycine at a holding potential (VH) of -40 mV in SDCN neurons. The selective NMDA receptor antagonist, D-2-amino-5-phosphonovaleric acid (APV, 100 μmol/L), inhibited the NMDA-evoked currents and blocked the NMDA-induced suppression of IGABA; (ii) when the neurons were incubated in a Ca2 -free bath or pre-loaded with a membrane-permeable Ca2 chelator, BAPTA AM (10 nmol/L), the inhibitory effect of NMDA on IGABA disappeared. In order to verify the effect of social behavior and geographical isolation on the genetic structure of the Himalayan marmot (Marmota himalayana) population, we examined the genetic diversity of Himalayan marmots alongside the Qinghai–Tibet Railway using microsatellite markers. Eight microsatellite loci were used to examine 120 animals of 4 populations: Ulan (U), Delhi (D), Tuotuohe (T) and Ando (A). The results show that: (1) Himalayan marmots alongside the Qinghai–Tibet Railway are highly genetically diversified. The allele number (Na), effective allele number (Ne), observed heterozygosity (Ho), Nei’s expected heterozygosity (He) and polymorphism information content (PIC) of the total Himalayan marmot population were 4.75, 3.0332, 0.6990, 0.6672, 0.6102, respectively. (2) Himalayan marmots may be able to avoid inbreeding by a mechanism that will prevent the genetic diversity reduction caused by their social lifestyle. Heterozygote excess was observed at most loci. The inbreeding coefficients within the subpopulation (FIS), in the total population (FIT), the differentiation index of population (FST), and the gene flow (Nm) were ?0.2265, ?0.0477, 0.1458, and 1.4646, respectively. (3) The genetic differentiation of the Himalayan marmot population was in accordance with Wright’s “isolation by distance” theory. The Mantel test indicates that the correlation between genetic distance and geographic distance was significant (P < 0.05, r = 0.698). (4) Each of the four geographical populations had moderate differentiation. Both geographic distance and isolation could affect the population genetic structure of the Himalayan marmot. The maximum gene flow (3.5915), the smallest genetic differentiation index (0.0651), the lowest genetic distance (0.0700) and the highest genetic identity (0.9526) were all between the Ulan population and Delhi populations. (5) The cluster analysis, based on Nei’s standard genetic distance, showed that the populations of Delhi and Ulan were first merged in a cluster, and then Tuotuohe population was merged in the clustering.
