Microtiter plate colorimetric assays are widely used for analysis of carbohydrates and glycoconjugates. However, mucins are often not easily detected, as they have low neutral sugar content. We have adapted and optimised the periodic acid–Schiff’s reagent (PAS) staining for microtiter plate assay by examining five factors: concentration and volume of periodic acid, oxidation time, volume of Schiff’s reagent, and color development time. This assay requires just 25 μl of sample, utilises standardised Schiff’s reagent, and has decreased assay time (140 min to completion). Seventeen monosaccharides (acidic, neutral, basic, phosphorylated, and deoxy) and four disaccharides were assessed. PAS-positive carbohydrates (amino, N-acetylamino, deoxy, and certain neutral monosaccharides, and sialic acids) responded linearly within a 10–100 nmol range approximately, which varied for each carbohydrate. The assay response for fetuin and porcine gastric mucin (PGM) was linear up to 150 μg (highest concentration tested), with no response from nonglycosylated protein. A lower response for asialofetuin was observed, but desialylated PGM preparations were similar or higher in response than their sialylated counterparts. The simplicity and low sample consumption of this method make it an excellent choice for screening or quantitation of chromatographic fractions containing carbohydrates and glycoconjugates, especially in the case of mucins.  相似文献

The photometric assay of reducing sugars with 2,2'-bicinchoninate was improved and its conditions were optimized. Optical density is linear between 1 and 25 nmol sugar/sample, and the assay is not affected by borate, phosphate, or other buffer anions. The molar extinction coefficients produced by the standard procedure with the 12 most common monosaccharides occurring in carbohydrates and conjugated glycocompounds are listed.  相似文献

This Viewpoint article addresses comments made on our original article describing a symbolic system for the depiction of N- and O-linked carbohydrate structures and proposes a method for extending the symbol set to include monosaccharides commonly found in carbohydrates present in bacteria and plants. As before, basic monosaccharides are shown by shape with one or more additions such as solid fill or additions of lines, crosses or dots to represent functional groups. The use of colour to differentiate constituent monosaccharides is avoided, thus enabling the system to be used in a variety of formats. Linkage and anomericity are shown by the angle and type of line connecting the symbols. In this extended version, new symbols are proposed for additional hexoses and it is proposed that pyranose and furanose forms of the monosaccharides could be shown by solid or broken outlines to the symbols. Conventions for depicting the presence of multiple functional groups such as deoxy-(NH(2))(2) are also discussed. It is hoped that these proposals will stimulate discussion so that a consensus can be reached as to how the glycobiology community can best convey complex information in a simple manner.  相似文献

Pollen grains of Helleborus foetidus and H. bocconei were exposed to low temperature treatments to simulate the natural events in pollen presentation of these two winter flowering species and to analyze the pollen carbohydrate content (glucose, fructose, sucrose and starch). In both species, cytoplasmic polysaccharides, monosaccharides and sucrose were found, while only Helleborus foetidus contained starch. Polysaccharide, sucrose and monosaccharide content varied as low temperature exposure time varied, a decrease in temperature decreases polysaccharide content and increases sucrose and monosaccharides. The relative quantities of the various types of carbohydrates were not affected by variations in the naturally occurring thermal cycles. Treatments did not greatly affect pollen viability. Although the occurrence of carbohydrates in pollen is known since many years, their function is still unclear. The findings of this research suggest a role of cytoplasmic pollen carbohydrates in resistance to low temperature exposure. The inter-conversion of carbohydrate type may be an adaptation for sustaining viability during pollen presentation that is particularly important for a winter flowering species such as Helleborus foetidus and H. bocconei .  相似文献

Considerable evidence implicates gp90MEL as a lymphocyte homing receptor mediating lymphocyte attachment to high endothelial venules of lymph nodes in mouse. The protein appears to function as a calcium-dependent, lectin-like receptor as inferred primarily by the ability of specific carbohydrates to block its function and by the presence of a calcium-type lectin domain in its primary sequence. An ELISA assay is described which provides the first demonstration that the isolated protein has lectin activity and allows a further definition of its carbohydrate specificity. In addition to the monosaccharides mannose-6-phosphate and fructose-1-phosphate, ligand activity is shown for the sulfated glycolipid, sulfatide, and for two sulfated fucose-containing polysaccharides (fucoidin and egg jelly coat) from nonmammalian sources.  相似文献

Thermal behavior at subzero temperatures has been investigated for aqueous solutions of various monosaccharides. The heat of fusion of ice measured with differential scanning calorimetry has given linear plots against sugar concentration (wt.%), from which the amount of unfrozen water, Uw, has been determined for each monosaccharide. The results for Uw are analyzed by employing, as a measure of hydration characteristics, known physico-chemical properties of aqueous monosaccharides, such as partial molar compressibilities, etc. It was revealed that the anti-freeze characteristics of carbohydrates depend on their stereochemistry. More water remains unfrozen in the aqueous solutions of carbohydrates having poorer compatibility with the three-dimensional hydrogen-bond network of water. Monosaccharides studied can be subdivided into three groups according to the extent of the anti-freeze effect. These results are rationalized in terms of a modified stereospecific hydration model.  相似文献

It was shown that the content of carbohydrates and their composition in embryo axes of horse chestnut seeds changed as seeds acquired a capability of dormancy release and germination. Sucrose prevailed among carbohydrates, comprising to 150–160 mg/g dry wt. During the first half of the seed imbibition time, oligosaccharides, namely raffinose and stachyose, degraded, whereas the contents of glucose and fructose were very low. The second half of the imbibition period (until radicle protrusion) was characterized by a cessation of oligosaccharide breakdown and accumulation of monosaccharides. Carbohydrate balance showed that the contribution of oligosaccharide breakdown to sucrose and monosaccharide accumulation was rather small, and monosaccharides accumulated mostly at the expense of sucrose gradually coming from cotyledons during imbibition. The trend of carbohydrate metabolism in imbibing axial organs was similar during the entire period of a seed dormancy release in the course of stratification. A readiness for the commencement of these processes during the entire dormancy period implies that carbohydrate conversions in embryo axes are not a trigger for a dormancy release. Monosaccharide accumulation in embryo axes before radicle protrusion produces an increase in the osmotic pressure, as compared to that provided by sucrose, by approximately 20%. Recalcitrance of the horse chestnut seeds is discussed in relation to the role of carbohydrates and other endogenous osmotica in the establishment of osmotic properties.  相似文献

Endogenous carbohydrates released from the intestinal mucus represent a constant source of nutrients to the intestinal microbiota. Mucus‐derived carbohydrates can also be used as building blocks in the biosynthesis of bacterial cell wall components, thereby influencing host mucosal immunity. To assess the uptake of endogenous carbohydrates by gut microbes in healthy mice and during intestinal inflammation, we applied azido‐monosaccharides that can be tracked on bacterial cell walls after conjugation with fluorophores. In interleukin‐10 deficient mice, changes in the gut microbiota were accompanied by decreased carbohydrate hydrolase activities and increased lumenal concentrations of host glycan‐derived monosaccharides. Tracking of the monosaccharide N‐azidoacetylglucosamine (GlcNAz) in caecum bacteria revealed a preferential incorporation of this carbohydrate by Xanthomonadaceae in healthy mice and by Bacteroidaceae in interleukin‐10 deficient mice. These GlcNAz‐positive Bacteroidaceae fractions mainly belonged to the species B. acidifaciens and B. vulgatus. Growth of Bacteroides species in the presence of specific monosaccharides changed their stimulatory activity toward CD11c+ dendritic cells. Expression of activation markers and cytokine production was highest after stimulation of dendritic cells with B. vulgatus. The variable incorporation of monosaccharides by related Bacteroides species underline the necessity to investigate intestinal bacteria down to the species level when addressing microbiota‐host interactions.  相似文献

The utilization of mono-, di-, and oligosaccharides by Bifidobacterium adolescentis MB 239 was investigated. Raffinose, fructooligosaccharides (FOS), lactose, and the monomeric moieties glucose and fructose were used. To establish a hierarchy of sugars preference, the kinetics of growth and sugar consumption were determined on individual and mixed carbohydrates. On single carbon sources, higher specific growth rates and cell yields were attained on di- and oligosaccharides compared to monosaccharides. Analysis of the carbohydrates in steady-state chemostat cultures, growing at the same dilution rate on FOS, lactose, or raffinose, showed that monomeric units and hydrolysis products were present. In chemostat cultures on individual carbohydrates, B. adolescentis MB 239 simultaneously displayed α-galactosidase, β-galactosidase, and β-fructofuranosidase activities on all the sugars, including monosaccharides. Glycosyl hydrolytic activities were found in cytosol, cell surface, and growth medium. Batch experiments on mixtures of carbohydrates showed that they were co-metabolized by B. adolescentis MB 239, even if different disappearance kinetics were registered. When mono-, di-, and oligosaccharides were simultaneously present in the medium, no precedence for monosaccharides utilization was observed, and di- and oligosaccharides were consumed before their constitutive moieties.  相似文献

4- Hydroxybenzoylhydrazine ( PAHBAH ) reacts with glucose in hot aqueous solution when alkali exceeds aroylhydrazine concentration. The related 2- furoylhydrazine ( FAH ) reacts at lower alkali concentrations, making this an attractive alternative carbohydrate reagent since it is (unlike PAHBAH ) freely water soluble. FAH reacts with monosaccharides more slowly than does PAHBAH , giving about half the color. Its specificity and behavior with a bismuth catalyst parallel those of PAHBAH . Solutions which contain 0.05 mol/liter PAHBAH with 1.5 mmol/liter bismuth III (as tartrate complex) and 0.5 mol/liter sodium hydroxide, or 0.01 mol/liter FAH with 1.5 mmol/liter bismuth III and 0.1 mol/liter sodium hydroxide are sensitive reagents for quantitative analysis, giving stable colors with many carbohydrates within 10 min at 75 degrees C. Ketosamines react more rapidly than glucose at lower temperatures and undergo similar reactions in less alkaline solutions. At pH less than 8, the reaction is specific for these 1- aminohexoses , and FAH can be used as a reagent for their assay.  相似文献

During pollen development, soluble carbohydrates of sporophytic origin may be consumed immediately, polymerized to form starch reserves or intine, or transformed into other molecules. Disregarding intine, in mature pollen there are three different types of carbohydrates: (1) polysaccharides such as starch in amyloplasts or polysaccharides in cytoplasmic vesicles, (2) disaccharides such as sucrose and (3) monosaccharides such as glucose and fructose. At dispersal, pollen may be partly or slightly dehydrated, or not dehydrated at all. Partly dehydrated pollen has the capacity to lose or acquire water within limits without detriment to its viability. Slightly and non-dehydrated pollen is vulnerable to water loss and quickly becomes inviable. In partly dehydrated of pollen the carbohydrates consist of cytoplasmic polysacharides and sucrose; in slightly and non-dehydrated pollen these are absent or in low concentrations but there may be reserves of cytoplasmic callose. Starch, glucose and fructose are found in both types. It is postulated that cytoplasmic carbohydrates and sucrose are involved in protecting pollen viability during exposure and dispersal.  相似文献

Gas chromatography—mass spectrometry with an on-line data system was used to identify monosaccharides and monosaccharide derivatives in human seminal plasma. The carbohydrates were converted into the methoxime—trimethylsilyl derivatives before separation in open tubular glass capillary columns coated with SE-30. Twenty-one different compounds were detected in the seminal fluid, of which twelve have not been recognized before. Seventeen of the monosaccharides have previously been identified in urine. Similar patterns of sugars were found both in fertile and infertile individuals, including one with azoospermia. The compounds identified are, with the possible exception of -ribose, present as free monosaccharides at the time of ejaculation, and they do not seem to be preformed by spermatozoa.  相似文献

The three-dimensional structures of the 2-, 3-, 4- and 6-monosulphatesof methyl -D-galactopyranoside have been determined by X-raycrystallography; the first two as the sodium salt, the thirdas both the sodium and potassium salts, and the fourth as apotassium salt. These represent the principal sulphated monomersof the carrageenan polysaccharides. The results extend our knowledgeof the stereochemical features, such as ring conformation, sulphategeometry, hydrogen bonding and cation co-ordination, which characterizesulphated monosaccharides. The stereochemical data have beenused to derive a mean geometry of the O-sulphate group and aset of force constants for use in molecular mechanics calculationson sulphated monosaccharides. 采集贵州省茂兰喀斯特原始红楼梦 林中森林土壤和相邻农田土壤,系统分析其中碳水化合物总量和各单糖的含量。并以此来查明由森林生态系统向农业生态系统转换的过程对土壤碳水化合物的影响,结果表明:相对于森林土壤,农业土壤中碳水化合物总量明显降低。在农田土壤中六糖/五糖比值有升高的趋势,其中以M/X比值最为明显,这说明,在该转换过程中植物来源的单糖组分有所降低,微生物来源的则相对增加。  相似文献

An array of maleimide-activated mono- and oligosaccharides were synthesized to permit site-specific glycosylation of cysteine-containing peptides and proteins. Maleimide-activated monosaccharides, in which the native alpha- or beta-O-glycosidic linkages found for nonreducing terminal sugars of native glycoproteins are preserved, were prepared using 2'-aminoethyl glycosides as the key intermediates. In addition, a native high-mannose type oligosaccharide, Man(9)GlcNAc(2)Asn, was converted into its maleimide-activated form by taking advantage of the existing amino group in the Asn portion. The application of these maleimide-activated carbohydrates was exemplified by the site-specific glycosylation of a 36-mer HIV-1 gp41 peptide, T20, which is a potent inhibitor against HIV infection. The chemoselective ligation was found to be rapid, highly efficient, and essentially quantitative. Tagging the biologically active peptide with a mannose and/or oligomannose moiety will be useful for targeting the drug to macrophage and dendritic cells, which are primary targets for HIV-1 infection and are expressing mannose- and oligomanose-specific receptors on their surface. In combination with site-specific mutagenesis, the maleimide-activated carbohydrates can serve as generally applicable tags for site-specific glycosylation of proteins via the highly efficient maleimide-thiol ligation reaction.  相似文献

For aqueous solutions of di- and oligosaccharides thermodynamic properties have been investigated at subzero temperatures using differential scanning calorimetry. The amount of unfrozen water observed is found to increase linearly with the glass transition temperatures of anhydrous carbohydrates. Furthermore, the amount of unfrozen water shows a linear relationship with known solution properties of aqueous carbohydrates, such as partial molar compressibility and heat of solution. The different effectiveness among various di- and oligosaccharides to avoid ice formation is associated with the combination of constitutive monosaccharides and attendant molecular structure features including the position and type of the glycosidic linkage between the constituent units. More unfrozen water is induced in the presence of a carbohydrate having a poorer compatibility with the three-dimensional hydrogen-bond network of water. A series of these results obtained imply that there is a common key of carbohydrate stereochemistry governing several different thermodynamic amounts of a given system involving carbohydrates. In this context, a modified stereospecific-hydration model can be used to interpret the present results in terms of stereochemical effects of carbohydrates.  相似文献

A procedure for rapid and sensitive analysis of carbohydrate in glycoproteins is described. After methanolysis and benzoylation of the monosaccharides and carbohydrates of a glycoprotein, the derivatized sugars were analyzed by reverse-phase high-performance chromatography using a Vydac C18 stationary phase and a mobile phase composed of a water/acetonitrile gradient. The advantages of this procedure over previously described methods are (1) the simple binary solvent system which is used requires no buffering salts and (2) separate sets of peaks from individual sugars obviate the usual need to reacetylate sugar amino groups.  相似文献

Milk was collected at various stages of lactation from a group of tammar wallabies, M. eugenii, in which parturition had been synchronized. The milk carbohydrate was determined by a phenol-sulfuric acid method which had been modified to give equal colour yields for galactose and glucose. The mean carbohydrate content increased gradually during the first 6 months of lactation to a peak of 13 g hexose/100 ml of milk, but then fell rapidly to much lower values, over the following 2 months. Throughouth lactation, galactose was the predominant monosaccharide constituent of acid hydrolysates of the milk carbohydrate. Glucose, glucosamine, galactosamine and sialic acid were the only other monosaccharides present. Qualitative changes were investigated by gel filtration and thin-layer chromatography. During the first 6 months post partum the milk carbohydrate was composed of a variety of oligosaccharides including lactose, but from 8 months onwards it consisted mainly of free monosaccharides. Between 6 and 8 months an intermediate pattern was observed, i.e. a mixture of lower oligosaccharides and free monosaccharides. In two animals which suckled both a new-born pouch young and a young at foot, the mammary gland supplying the new-born secreted milk which was rich in oligosaccharides, whereas that supplying the young at foot produced milk in which the carbohydrates were mainly free monosaccharides, and which had a much lower carbohydrate content.  相似文献
