Fatty acid composition of the seed oil of Allium tuberosum   总被引:3,自引:0,他引:3

The fatty acid compositions of seed oil from ten kinds of Allium tuberosum (Chinese chive, Liliaceae) were determined by Gas Chromatography-Mass Spectroscopy (GC-MS). The seed oil of Allium tuberosum contained much important fatty acids with linoleic (57.0-71.6%) and palmitic (6.6-9.7%), the most abundant unsaturated and saturated fatty acids, respectively. The total unsaturation for the oil was 79.2-93.0%. It is concluded that Allium tuberosum oil is of important nutritional value.  相似文献

The whole-cell phospholipid composition of the six known polymorphic species of Tetrahymena has been examined by [(3)H]acetate and [(3)H]myristic acid radiolabeling, and by gas-liquid chromatography of total phospholipid-bound fatty acids. Five of the polymorphic species contained similar phospholipid profiles following radiolabeling in that phosphatidylethanolamine (PE) was the predominant phospholipid; however, in cells of Tetrahymena patula LFF, aminoethylphosphonolipid was present in amounts nearly equal to PE. Tetrahymena patula LFF contained an unusually large percentage of sphingolipid (16.2% by [(3)H]acetate radiolabeling). Substantial differences were found in the fatty acid profiles of the polymorphic species, which included the degree of fatty acid unsaturation and relative weight percentages of odd-chain fatty acids. Tetrahymena vorax contained a low ratio of unsaturated C(18) fatty acids to saturated C(18) fatty acids as compared with all other species examined. The differentiating species generally contained a lesser percentage of monoenoic fatty acids and a lower ratio of unsaturated C(16) fatty acids to saturated C(16) fatty acids as compared with the two monomorphic species examined.  相似文献

Cells of Tetrahymena mimbres (formerly T. pyriformis NT-1) in midlogarithmic growth under isothermal conditions (at 39 degrees C) contained a very small, compositionally discrete pool of free fatty acids, principally (60.6% of the total free fatty acid mass) palmitic and stearic acids. The composition, degree of unsaturation, and size of this free fatty acid pool were rapidly (15 min or less) altered in response to chilling. During the acclimation period following chilling to 15 degrees C, the size of the free fatty acid pool increased from a mean value of 15.5 nmol free fatty acid/mumol lipid phosphorus in 39 degrees C cells to 24.2 nmol free fatty acid/mumol lipid phosphorus. The degree of free fatty acid saturation rapidly increased over the initial hour following the onset of hypothermal conditions, but by 24 h the unsaturated free fatty acid/saturated free fatty acid ratio was 0.35 (equivalent to a 2.7-fold increase in unsaturation relative to 39 degrees C controls (unsaturated/saturated ratio = 0.13) and 4.4-fold greater than cells acclimated for 1 h (unsaturated/saturated ratio = 0.08)). By 24 h the percentage of palmitic and stearic acids had decreased to 45.6%. Similar, and in some instances more pronounced, changes were observed to occur in triacylglycerol-bound fatty acids. Modulation of steady-state free fatty acid composition could also be achieved by the addition of exogenous fatty acids to the growth medium. The ability to manipulate the level of intracellular free fatty acids should prove to be a valuable experimental tool in determining how specific fatty acids regulate various lipid-modifying enzymes.  相似文献

Cold acclimation and overwintering state can affect fatty acid compositions of insects. To determine compositional change of fatty acids during nondiapause and diapause stages, an experiment was conducted to investigate fatty acid constituents from whole body of C. suppressalis larvae. Five most abundant fatty acids were found to be palmitoleic (35–58%), palmitic (18–44%), oleic (14–23%), stearic (0.5–2.5%) and linoleic acid (0.4–2%). However, linolenic, erucic, lauric and myristic acid were found at lower level. Saturated fatty acids significantly decreased and conversely unsaturated fatty acids increased from August (pre-diapause) to October (initiation of diapause). The increase in seasonal cold hardiness during cold acclimation, exposed at −15°C for 24 h, was related to degree of fatty acid unsaturation. The elevation of palmitoleic acid content at low temperature resulted in an increase in the overall degree of unsaturation in the whole body. These results indicated the importance of unsaturated fatty acids composition to prepare larvae entering diapause phase.  相似文献

采用索氏提取法提取木通科植物猫儿屎和三叶木通种子的脂溶性成分,甲酯化处理后用气相色谱-质谱联用技术(GC-MS)分离和鉴定其组成和含量。从猫儿屎种子油中鉴定出9种脂肪酸,占检出物总质量分数的94.67%(其中饱和脂肪酸占12.63%,不饱和脂肪酸占82.04%),主要成分为9-十六烯酸(47.22%)、9-油酸(27.13%)、棕榈酸(10.75%)、亚油酸(7.47%)和硬脂酸(1.61%)。从三叶木通种子油中鉴定出10种脂肪酸,占检出物总质量分数的99.75%(其中饱和脂肪酸占23.39%,不饱和脂肪酸占76.36%),主要成分为11-油酸(47.63%)、亚油酸(27.05%)、棕榈酸(20.14%)、16-甲基-十七烷酸(3.03%)和8-油酸(1.07%)。结果表明,猫儿屎和三叶木通种子油中脂肪酸含量丰富,在食用、医疗保健等方面具有较高的应用潜力和综合开发前景。  相似文献

The effect of change in unsaturated fatty acid composition on ethanol tolerance in Saccharomyces cerevisiae overexpressing ScOLE1 (?9 fatty acid desaturase gene of S. cerevisiae), CaFAD2 (?12 fatty acid desaturase gene of Candida albicans), or CaFAD3 (ω3 fatty acid desaturase gene of C. albicans) was examined. ScOLE1 over-expression increased the total unsaturated fatty acid content and enhanced ethanol tolerance, compared with a control strain. In contrast, overexpression of CaFAD2 and CaFAD3, which led to production of linoleic acid (18:2) and α-linolenic acid (18:3), respectively, neither changed total unsaturated fatty acids nor enhanced ethanol tolerance. The total unsaturated fatty acid content rather than the degree of unsaturation is thus an important factor for ethanol tolerance.  相似文献

The seed oils of three species (Litsea cubeba (Lour.) Pers, L. auriculata Chien et Cheng, L. subcoriacea Yang et P. H. Huang) were examined and the fatty acid composition of these oils was determined by GLC. Their major fatty acid was identified as lauric acid, Its amount ranged from 34.6%–75.4%. The major acid of the fruit coat oil from L. subcoriacea Y. H. Huang was different from that of the seed oil. The former contained 50% linoleic acid. The unsaturated C10, C12, C14 acids of the seed oil from L. cubeba (Lour.) Pers were separated by distillation, column chromatography and were identified by Periodate-Perman-ganate oxidation, IR, NMR and MS. They are cis-4-decenoic, cis-4-dodecenoic and cis-4-tetradecenoic acids respectively.  相似文献

该研究以细叶小羽藓为材料,测定了基本营养成分总糖、粗脂肪、粗蛋白、灰分的含量,采用气相色谱、氨基酸自动分析仪分别检测了脂肪酸和氨基酸的组分含量,并通过化学分析法和扫描电镜明确了细叶小羽藓矿质元素含量及分布情况。结果表明:细叶小羽藓中主要成分是糖类,占16.11%,粗蛋白质、灰分含量分别为11.20%、23.34%。氨基酸组成均衡,必需氨基酸(EAA)占氨基酸总量(TAA)的27.70%,必需氨基酸与非必需氨基酸的比值(EAA/NEAA)为0.38;限制性氨基酸为组氨酸,比值系数分(SRC)为12.96;药用氨基酸占总氨基酸的62.12%;天冬氨酸、谷氨酸、精氨酸、亮氨酸等含量相对较高。不饱和脂肪酸与饱和脂肪酸的比值为0.74,不饱和脂肪酸花生四烯酸(AA)和二十二碳六烯酸(DHA)相对含量较高。富含矿物质,茎叶元素均匀分布,其中Al、Ca、Fe、K、Mg等元素含量很高。综上认为,细叶小羽藓是一种含糖量较多、矿质丰富、低脂、低蛋白的苔藓,作为生物资源具有开发潜力。  相似文献

Cyanidium caldarium was cultured at 20 and 55 C and harvested during exponential growth phase. Comparative lipid studies on each cell type show a decrease by one-half of the total lipid in cells grown at 55 C over cells grown at 20 C. While the distribution of lipid into each of five lipid classes was not influenced by high temperature (55 C), the degree of unsaturation was greatly affected. Ratios of unsaturated to saturated fatty acids in these cells decreased 3-fold with increased temperature in the growth environment. Cells cultured at 20 C contained 30% of their fatty acids as linolenic while this fatty acid could not be detected in cells cultured at 55 C.  相似文献

An analysis of the phospholipid ester-linked and the lipopolysaccharide (LPS) fatty acids and hydroxy fatty acids of six lactate-utilizing Desulfovibrio-type sulfate-reducing bacteria (SRB) has been performed using capillary gas-liquid chromatography-mass spectrometry (GLC-MS). The concentrations of normal fatty acids were essentially similar, with the possible exception of a high content of normal fatty acids in the LPS of Desulfovibrio gigas. Determination of monounsaturated acid double bond configuration was performed by GLC-MS analysis of the derivatized fatty acids. A total of nine branched chain and eight straight chain monounsaturated fatty acids was detected in the Desulfovibrio species analyzed. The major component detected in five Desulfovibrio was the 17-carbon iso-branched monoenoic acid which showed cis unsaturation [i17:1(n-7)c] seven carbons from the terminal methyl group of the fatty acid chain. D. gigas, in contrast, contained almost no unsaturated fatty acids and was greatly enriched in iso-branched 15:0. Major differences between strains were found in the phospholipid and LPS hydroxy fatty acids. These components, in addition to the i17:1(n-7)c and other characteristic branched chain unsaturated acids, can possibly be utilized as signatures of the lactate-utilizing SRB.  相似文献

NaCl induced changes in fatty acid composition and nitrogenase, glutamine synthetase (GS) and nitrate reductase (NR) activities have been studied in a diazotrophic cyanobacterium Anabaena cylindrica. GC-MS analysis revealed that the cellular fatty acid composition of NaCl untreated cells of A. cylindrica contained saturated and unsaturated fatty acids in high (85.15%) and low (13.17%) proportions, respectively. In contrast, NaCl adapted cells of A. cylindrica had reduced and increased levels of saturated (45.2%) and unsaturated (40%) fatty acids, respectively. It had a higher overall level of fatty acid unsaturation under NaCl stress mainly due to increase in C12:4, C10:1, C16:1 and C18:2 constituents. The activities of nitrogenase, GS and NR were reduced significantly in NaCl adapted cells as compared to its NaCl untreated counterparts.  相似文献

Four subfractions of phosphatidycholine and phosphyatidylethanolamine according to the degree of unsaturation of their fatty acids have been separated from lipid extracts of microsomes, and inner and outer mitochondrial membranes. The predominant species found in the three membranes contained one saturated and one unsaturated fatty acid. In microsomes completely saturated species of both phosphatidylcholine and phosphatideylethanolamine were practically nonexistent. In outer mitochondrial membranes species with two unsaturated fatty acids were absent. In the inner mitochondrial membranes, however, disaturated species and those with two unsaturated fatty acids were found.  相似文献

The aim of this study was to develop a nondestructive method to quantitate relative amounts of n-3 and n-6 polyunsaturated fatty acid (PUFA) species in vegetable oils and oil seeds using Fourier transform IR spectroscopy (FTIR). The alkene Cbond;H stretching vibrations of unsaturated fatty acids in oils showed IR absorption bands with various peak positions and intensities at around 3010 cm(-1), depending on the extent of unsaturation and PUFA species. With the aid of partial least-squares regression analysis, the FTIR measurement could practically predict the content of each PUFA species in the oil to be tested. A calculation method was also presented to directly find PUFA species in oils from the FTIR spectra. This technique was applied to dried soybean seeds to demonstrate a nonhomogenous distribution of saturated fatty acids and PUFAs, as well as glycans, in soybean cross sections.  相似文献

The chilling sensitivity of several plant species is closely correlated with the levels of unsaturation of fatty acids in the phosphatidylglycerol (PG) of chloroplast membranes. Plants with a high proportion of unsaturated fatty acids, such as Arabidopsis thaliana, are resistant to chilling, whereas species like squash with only a low proportion are rather sensitive to chilling. The glycerol-3-phosphate O-acyltransferase (GPAT) enzyme of chloroplasts plays an important role in determining the levels of PG fatty acid desaturation.A cDNA for oleate-selective GPAT of Arabidopsis under the control of a maize Ubiquitin promoter was introduced into rice (Oryza sativa L.) using the Agrobacterium-mediated gene transfer method. The levels of unsaturated fatty acids in the phosphatidylglycerol of transformed rice leaves were found to be 28% higher than that of untransformed controls. The net photosynthetic rate of leaves of transformed rice plants was 20% higher than that of the wild type at 17°C. Thus, introduction of cDNA for the Arabidopsis GPAT causes greater unsaturation of fatty acids and confers chilling tolerance of photosynthesis on rice.  相似文献

The fatty acid and cholesterol contents of tissue membranes are the determinants of membrane stability and functionality. This study was designed to evaluate the influence of a high monounsaturated fatty acid diet on the fatty acid composition of rat liver microsomes and on their cholesterol and lipid phosphorus content. Weanling animals were fed for 5 weeks with high fat diets containing olive oil or corn oil. Saturated fatty acids were increased and oleic acid decreased in microsomal total phospholipids and in the three major phosphoglycerides, phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylinositol (PI), of rats fed corn oil as compared to the olive oil group. The percentage of linoleic acid was higher in the corn oil group, but only for total phospholipids and PC. Linoleic and alpha-linolenic metabolites were significantly increased in total phospholipids of olive oil-fed animals with respect to those fed corn oil. These changes were responsible for the low unsaturation index found in microsomal phospholipids of the corn oil group. The diet did not affect the microsome cholesterol or the lipid phosphorus content. These results show that, in olive oil-fed rats, the cholesterol content and the degree of unsaturation of liver microsomes was similar to that observed in weanling animals; this probably suggests an adequate maintenance of functionality of membranes in olive oil-fed animals.  相似文献

Lipid profiles of conidia of Aspergillus niger and a fatty acid auxotroph   总被引:2,自引:0,他引:2  
Conidial lipids of the wild-type (V35) Aspergillus niger and its unsaturated fatty acid auxotroph (UFA2) were compared. The wild type contained lower levels (7.6%) of phospholipids and higher levels (28.4%) of glycolipids than the mutant (16.5 and 22.2%, respectively). Oleic (33.4%), linoleic (22.5%), palmitic (12.8%), stearic (7.4%), and linolenic (6.2%) were the main fatty acids of the wild type (V35). The mutant grew only in the presence of unsaturated fatty acid having at least one delta 9cis double bond, and its conidial fatty acid profile was influenced by the exogenous acid. Analyses of the fatty acids of UFA2 grown in the presence of different fatty acid supplements support the original view that the mutant is defective in delta 9-desaturase activity.  相似文献

The selective mobilization of fatty acids from white fat cells depends on their molecular structure, in particular the degree of unsaturation. The present study was designed to examine if the release of fatty acids by hormone-sensitive lipase (HSL) in vitro i) is influenced by the amount of unsaturation, ii) depends on the temperature, and iii) could explain the selective pattern of fatty acid mobilization and notably the preferential mobilization of certain highly unsaturated fatty acids. Recombinant rat and human HSL were incubated with a lipid emulsion. The hydrolysis of 35 individual fatty acids, ranging in chain length from 12 to 24 carbon atoms and in unsaturation from 0 to 6 double bonds was measured. Fatty acid composition of in vitro released NEFA was compared with that of fat cell triacylglycerols (TAG), the ratio % NEFA/% TAG being defined as the relative hydrolysis. The relative hydrolysis of individual fatty acids differed widely, ranging from 0.44 (24:1n-9) to 1.49 (18:1n-7) with rat HSL, and from 0.38 (24:1n-9) to 1.67 (18:1n-7) with human HSL. No major difference was observed between rat and human HSL. The relative release was dependent on the number of double bonds according to chain length. The amount of fatty acid released by recombinant rat HSL was decreased but remained robust at 4 degrees C compared with 37 degrees C, and the relative hydrolysis of some individual fatty acids was affected. The relative hydrolysis of fatty acids moderately, weakly, and highly mobilized by adipose tissue in vivo was similar and close to unity in vitro. We conclude that i) the release of fatty acids by HSL is only slightly affected by their degree of unsaturation, ii) the ability of HSL to efficiently and selectively release fatty acids at low temperature could reflect a cold adaptability for poikilotherms or hibernators when endogenous lipids are needed, and iii) the selectivity of fatty acid hydrolysis by HSL does not fully account for the selective pattern of fatty acid mobilization, but could contribute to explain the preferential mobilization of some highly unsaturated fatty acids compared with others.  相似文献
