The formation of aerial tubers in Begonia plants, an SD response,was inhibited by IAA, NAA and IBA applied to their leaves duringthe dark periods. The effectiveness of IAA differed accordingto the time of application during the dark periods, and themost sensitive time varied with daylengths employed. In order to inhibit the tuberization under optimal photoperiods(8-hr SDs), IAA had to be applied during the first 2 days orso of the SDs. Under non-optimal photoperiods, however, IAAwas effective even when applied somewhat later. The auxin activity of leaf extracts from the plants subjectedto 8-hr SDs decreased during the first 2 or 3 days to a minimum,and then increased until finally began to decrease, again; undernon-optimal photoperiods, the minimum of auxin activity wasattained more slowly. The paper-chromatographic study suggestedthat the change in auxin activity was mainly due to the changein IAA content. The number of SDs making the auxin content minimal agreed withthe minimum number of SDs required for tuberization. Gibberellin-like substances and an auxin similar to IAA weredetected by bioassays in extracts of flowers of Chrysanthemummorifolium. The activity of these substances was shown to reacha maximum early in the development of the flower when its relativegrowth-rate was at a maximum, and then to decline with the relativegrowth-rate. The leaves of lateral flowering shoots were found to containgibberellins similar to those detected in the flowers whilea different gibberellin, which appeared to decrease in activitywith the age of the shoot, was detected in the stem. An auxinsimilar to indol-3yl-acetic acid (IAA) was also detected inthese stems. Growth-promoting substances were not detected inthe old stems and leaves from the main shoot. Gas-liquid chromatographyrevealed the presence of a number of additional gibberellinsin the flowers. The chemical nature of the growth substances is discussed inrelation to their biological and chromatographic behaviour.  相似文献

The exceptionally high capacity for transformation to autotrophyfor hormones (habituation) discovered in Nicotiana bigeloviiled us to a comparison of the responses of 2 varieties (quadrivalvisand bigelovii) to different hormone treatments applied to theirseeds. Hormones used were: kinetin (0.8, 1.6, 3.2 ppm), 2,4-D(0.1, 0.4 ppm), IAA (2 ppm) and NAA (1, 2, 4 ppm). Callus formation and habituation were scored after transferon minimal medium (without hormones) 3 months after sowing.Var. quadrivalvis showed a much higher ability to form callusbut the callus formed was autonomous in a lower percentage ofcases when IAA or kinetin treatment was considered. Analysisof auxin-like substances in leaves of the 2 varieties showeda higher content in N. bigelovii var. quadrivalvis. Changes in free IAA level were studied in the leaves of the central stem zone of short-day tobacco (Mcotianatabacum, cv. Maryland Mammoth) and long-day tobacco(Nicotiana silvestris) in inductive photoperiodic regime after 10, 20, 30 and 40 d, respectively. The leaves of SD tobacco Mammoth showed a high free IAA level in vegetative plants kept under long days but it significantly decreased (by ca. 50 %) after 10, 30 and 40 short days, respectively. After 20 short days the IAA level was as high as in the leaves of plants at the beginning of inductive treatment. The changes of freeIAA level in the leaves of LD tobacco N.silvestris were similar to those of SD Mammoth, but the IAA level in this species was significantly lower than that of Mammoth throughout the investigated period. Consequently, the changes observed in N. silvestris were much less pronounced. Plants of both tobacco species were fully induced to flowering by 30 inductive days and this was associated with differentiation of the flower organs. Application of 10 -4 M IAA during the last 10 d of the inductive treatment of 30 d significantly reduced flowering in SD tobacco Mammoth without changing the stem length and apex width. Apex length was slightly reduced. IAA application elicited almost no effect inN. silvestris. The results are discussed with respect to the possible role of IAA in flower induction in SD and LD plants.  相似文献

The levels of endogenous IAA and the production of ethylenehave been followed during maturation and senescence in herbaceous(Phaseolus vulgaris, Ecballium elaterium) and deciduous (Prunusserrulata) leaves. Comparisons have been made with similar estimationsduring ripening of a herbaceous fruit. Ecballium elaterium.Whereas a correlation can be made between auxin content andethylene production in immature tissues, no such relationshipexists in senescing or ripening tissues where ethylene productionappears to be independent of the total endogenous auxin content.Both IAA and fusicoccin enhance ethylene production in developingleaves but fail to do so in senescent tissues. A mechanism forthe regulation of the rate of ethylene biosynthesis is described.This involves modifications in the release from a membrane-boundor membrane-enclosed compartment of cofactor(s) essential toone or more steps in the pathway. The mechanism accommodatesobserved normal and senescence-related rates of ethylene production.  相似文献

IR68 and Dular rice cultivars were grown under ambient, 13.0 (simulating 20% ozone depletion) and 19.1 (simulating 40% ozone depletion) kJ m-2 day-1 of biologically effective ultraviolet-B (UV-BBE) for 4 weeks. Plant height and leaf area were significantly reduced by supplemental UV-BBE radiation. Greater reduction in leaf area than of plant height was observed. A decrease in indole-3-acetic acid (IAA) content and increase in peroxidase and IAA oxidase activities of UV-B treated plants in both cultivars were observed compared with ambient control. Calmodulin content also decreased after plants were treated with high supplemental UV-B for two weeks and medium UV-B treatment for four weeks. The results indicated that peroxidase and IAA oxidase activities in rice leaves were stimulated by supplemental UV-B, resulting in the destruction of IAA which in turn may cause inhibition of rice leaf growth. Although the mechanism is unclear, calmodulin is most likely involved in leaf growth.  相似文献

Plants of Lycoperstcon esculentum Mill. cv. Ailsa Craig wildtype and flacca (flc) were sprayed daily with H2O or 2?10–2mol m–3 abscisic acid (ABA). ABA treatment effected apartial phenotypic reversion of flc shoots; leaf areas wereincreased and transpiration rates decreased. Leaf expansionof wild type shoots was inhibited by ABA. Indoleacetic acid (IAA), ABA and l-aminocyclopropane-l-carboxylicacid (ACC) concentrations were determined by combined gas chromatography-massspectrometry using deuterium-labelled internal standards ABAtreatment for 30 d resulted in greatly elevated internal ABAlevels, increasing from 1?0 to 4?3 and from 0?45 to 4?9 nmolg–1 fr. wt. in wild type and flc leaves respectively.Endogenous IAA and ACC concentrations were much lower than thoseof ABA. IAA content ranged from 0?05 to 0?1 nmol g–1 andACC content from 0?07 to 0?24 nmol g–1 Ethylene emanationrates were similar for wild type and flc shoots. Wilting of detached leaves induced a substantial increase inethylene and ACC accumulation in all plants, regardless of treatmentor type. Ethylene and ACC levels were no greater in flc leavescompared to the wild type. ABA pretreatment did not preventthe wilting-induced increase in ACC and ethylene synthesis. The control of tiller bud growth during reproductive developmentwas investigated in experimental plants ofLolium multiflorumLam. cv. Westerwoldicum that were reduced to a main axis havinga developing but unemerged ear, elongating stem internodes,a series of expanded leaves, slow-growing tiller buds and aroot system. Isolation of the ear by excision of its base, ordecapitation so as to remove the ear together with the upperleaves, promoted the movement of 14C-assimilates to tiller buds,decapitation being the more effective treatment. Applicationof 0.1 per cent indol–3yl-acetic acid (IAA) to cut tissuesof decapitated plants diverted 14C-assimilates to upper internodesbut did not reduce import by buds, whereas application of 1.0per cent IAA both diverted labelled assimilates to upper internodesand reduced bud import. Radioactivity from [14C] IAA appliedto the upper leaves or to the ear base was recovered from budsin very small amounts; larger amounts were recovered from budsfollowing the application of labelled IAA to an elongating internode,especially from the bud at the base of the treated internode.It is suggested that tiller bud suppression may be influencedby the movement of inhibitory levels of auxin into buds fromnearby elongating stem internodes, whose activity in turn maybe controlled by the developing inflorescence and upper leaves.  相似文献

The growth response of Tapinanthus bangwensis (Engler and K.Krause) Danser to exogenous growth substances, indole-3yl-aceticacid (IAA), kinetin, and gibberellic acid (GA3), not only varieswith the concentrations of each of these substances in the mediumbut follows a similar trend in each case. Kinetin, IAA, andGA3 were found to enhance growth but not by very appreciableamounts, kinetin being the most effective growth promoter. Highconcentrations of these substances (5.0 and 10.0 ppm) exceptGA3 were toxic to the parasite. Gibberellic acid induced theproduction of accessory leaves at all the concentrations used.Kinetin was also effective in inducing accessory leaves butonly at a concentration of 1.0 ppm. Some of the lower concentrationsof the growth substances affected significantly some aspectsof the parasite's growth and development. Nevertheless, thegrowth in vitro was still slower than that in vivo.  相似文献

The activity of IAA-oxidase increased in the leaves of Impatiensbalsamina plants receiving inductive photoperiodic cycles andin plants receiving treatments with gibberellic acid (GA3) and/ortannic acid (TA), even under non-inductive photoperiods; theactivity also increased in the stem receiving inductive photoperiodiccycles (8 h). Treatment with GA3 and TA mimics the effect ofSD cycles in the development of some isoenzymes of IAA-oxidase.Thus a new isoenzyme at Rf 0.48 developed in the leaves andone at Rf 0.82 developed in both the stem and the leaves ofall plants receiving inductive treatments – photoperiodicor chemical – but not in water-treated controls undernon-inductive photoperiods. Partial inhibition of extension growth of the primary leaf occurswhen whole Triticum seedlings are immersed in aerated solutionsof IAA but is replaced by growth promotion when sucrose is addedto the external solution. In seedlings in which the coleoptilehas been excised, IAA increases the growth of the leaf bothwith and without additional sucrose. Inhibition of the leaf by moderate concentrations of IAA nolonger occurs when the seedling is detached from the endosperm.Sucrose added to the external solution raised the percentageelongation of the coleoptile almost to the level of that attainedin intact seedlings without additional carbohydrate. It alsoenabled the leaf to show a positive growth response with IAA. The results indicate that in intact seedlings treated with IAAthe growth of the primary leaf is markedly diminished owingto diversion of carbohydrate to the coleoptile if the growthof the latter is promoted as a result of the treatment. Whenthe competition of the coleoptile for carbohydrate is diminishedor eliminated, acceleration of the growth of the primary leafby IAA becomes apparent. In addition to the endogenous rhythm, with a period close to24 hours, induced in the growth-rate of the coleoptile whenseedlings of Avena are transferred from red light to darkness,a similar rhythm, with a slightly longer period, is inducedin the growth-rate of the primary leaf. This rhythm persistsin elongating leaves so long as they remain within the coleoptile.It can be recorded for at least 100 hours in deseeded seedlings. When intact seedlings of Avena are immersed for one hour inrelatively high concentrations of IAA and then transferred todistilled water for 18 hours, the elongation of the coleoptileis greater and the inhibition of the leaf is less than whenthey are transferred to humid air. Sections of the leaf of Triticum showed a slight increase inelongation in concentrations of IAA up to 5 mg./l., but no evidencewas obtained that sections of leaf and coleoptile exert any.influenceon each other's elongation when floated together on solutionsof IAA.  相似文献

In roots of sweet potato (Ipomoea batatas Lam. cv. Kokei 14),the metabolic response to wounding was remarkable only in theproximal side. We assumed that the polarity resulted from apolar movement of indole-3-acetic acid (IAA) produced in thecut surface (8). As the metabolic response was slight in thedistal side, the effect of IAA and the other plant hormoneson the development of various enzyme activities was examinedin this side. Increases in activities of L-phenylalanine ammonia-lyase,acid invertase, NADPHa2 : cytochrome c oxidoreductase, peroxidase,cytochrome c : O2 oxidoreductase and o-diphenol oxidase, whichdeveloped in response to wounding, were stimulated by the treatmentwith IAA. Gibberellic acid had a stimulative effect on the developmentof only acid invertase activity. Abscisic acid and kinetin hadlittle effect. The influence of a water stress or foliar ABA spraying pretreatmenton stomatal responses to water loss, exogenous ABA, IAA, Ca2+,and CO2 were studied using excised leaves of Solanum melongena.Both pretreatments increased stomatal sensitivity of water loss,in the presence and absence of CO2, but decreased stomatal sensitivityto exogenous ABA. CO2 greatly reduced the effect of exogenouslyapplied ABA. IAA decreased leaf diffusion resistance for controland ABA sprayed leaves, but did not influence the LDR of previouslywater-stressed leaves. 整株干旱降低盐棉46号叶片中的IAA总量,叶龄愈小下降愈多。幼叶中IAA总量的下降主要是结合态IAA减少的结果。气干和-1.7MPa PEG溶液渗透胁迫处理也降低离体成熟叶片的IAA总量,其变化与叶片含水量呈直线相关(r=0.905)。整株干旱处理提高各叶片的过氧化物酶活性,叶龄愈小提高愈多,但IAA氧化酶活性无显著变化。离体和整株干旱时IAA总量的下降,可能是过氧化物酶活性增加所致。  相似文献

Treatment of spruce, fir and oak trees with herbicides, which may be one of the forest damage inducing agents, caused pronounced changes in the contents and distribution of indole-3-acetic acid (IAA) and cytokinins (CKs) one year after treatment, i.e. at the time of the first microscopically visible damage in treated trees. In Picea pungens IAA content increased in the terminal buds by about 105 % and in the apical buds of the first order branches by 220 %. The same was true for young sprouts of Abies nordmanniana, while in leaves of oak trees IAA content was decreased by 15 % after glyphosate treatment and by 30 % after 2,4-dichlorophenoxyacetic acid (2,4-D) treatment. Another striking feature was a significantly decreased content of IAA in the lower parts of roots in Picea pungens (50 % of the control), which is accompanied by an increase in IAA content in the middle part of the roots (130 %). On the other hand, the IAA content of both sprouts and roots of A. nordmanniana was significantly increased after herbicide treatment.In P. pungens, the content of free cytokinins (sum of zeatin, zeatin riboside, isopentenyladenine and isopentenyladenosine) decreased due to herbicide treatment. The strongest decrease was seen in roots, especially in their upper and middle parts (the average reduction of cytokinin content in roots was 63 %). In the above-ground organs the reduction was seen namely for isopentenyladenine and isopentenyladenosine, while the abundance of zeatin riboside was, on the other hand, higher in treated plants. In Quercus robur leaves, the total content of cytokinins also decreased, namely after glyphosate treatment. In consequence of these changes, CK/IAA ratio decreased pronouncedly in all organs of herbicide-treated trees, with the exception of oak leaves treated by 2,4-D.  相似文献

This study examined the impacts of elevated CO2 or O3 on indole-3-acetic acid (IAA) content, activities of IAA oxidase (IAAO) and peroxidase (POD) in Ginkgo biloba leaves. Plants grown in open-top chambers were exposed to ambient atmosphere (control; C), elevated CO2 and elevated O3 from 1 June to 30 September. An increase in IAA content and decrease in IAAO and POD activities were observed in plants exposed to elevated CO2 compared with C. Elevated O3 had no significant effect on IAA content and IAAO activity, but increased POD activity during the early days. When trees pre-exposed to elevated CO2 were transferred to elevated O3 or C, the increase in IAAO activity resulted in the decrease in IAA content. When trees pre-exposed to elevated O3 were transferred to elevated CO2 or C, IAA content, IAAO and POD activities showed no significant changes. The influence of POD activity on the IAA activity was low.  相似文献

Changes in the contents of major endogenous plant hormones intobacco crown gall cells, namely IAA and ribosyl-trans-zeatin,during cell growth were examined using HPLC and 14C-labeledplant hormones. The content of IAA was high at the early logarithmicstage, while that of ribosyl-trans-zeatin was high at the middlelogarithmic stage.
