Variations of copper (Cu) concentration and ceruloplasmin (Cp) activity in serum and plasma of dairy cows at different stages of lactation were assessed in 240 Holstein dairy cows. Furthermore, ceruloplasmin to copper ratios were also investigated. The cows were classified according to their lactation stages into four different groups as close-up stage (3–1 week(s) antepartum), fresh lactation stage (0–1 week postpartum), early lactation stage (3–5 weeks postpartum), and mid-lactation stage (15–18 weeks postpartum). Each group consisted of 60 multiparous cows. Serum and heparinised plasma samples were obtained from each cow. Concentrations of serum copper (sCu) and plasma copper (pCu) as well as activities of plasma Cp (pCp) were higher in the group of fresh lactation stage than other groups (P < 0.05). Serum Cp (sCp) exhibited no significant difference between fresh lactation and early lactation groups (P > 0.05). Low correlations were obtained between sCp and pCu, sCp and sCu, and sCu and pCu. Plasma copper concentration and plasma ceruloplasmin activity were higher than serum copper concentration and serum ceruloplasmin activity, respectively (P < 0.001). The ratios of Cp activity to Cu concentration (Cp/Cu) were not significantly changed in the different lactation stages of dairy cows (P > 0.05). Use of sCp/pCu and sCp/sCu rather than pCp/pCu will reduce the calculated value of Cp/Cu. Furthermore, for evaluation of copper status, use of sCp/sCu or sCp/pCu identified more animals as ‘low’ and ‘marginal’ than using pCp/pCu (P < 0.001). It can be concluded that ceruloplasmin and copper undergo a physiological increase just after calving; thus, their values should be interpreted with caution during assessment of copper status. Plasma measurements should be used for calculation of Cp/Cu, and further research is required to refine diagnosis criteria for use of such ratio in determining copper status in dairy cows.  相似文献

Previous studies showed that responses to chronic administration of copper were significantly associated with gender, raising the need to better characterize the relation between the effects observed and stradiols. The objective of this study was to measure copper and liver function indicators and the sex hormone binding globulin (SHBG) serum concentrations in healthy adults exposed to copper, grouped by sex and phase of the female hormonal cycle. Healthy females on day 7 (follicular phase, Group 1, n = 39), on day 21 (secretory phase, Group 2, n = 34) and males (comparison group, Group 3, n = 34) received 8 mg Cu/day (as copper sulfate), orally, for 6 months. On days 0, 30, 60, 120, and 180, the serum concentration of copper, ceruloplasmin, liver aminotransferases, and SHBG were measured. Analysis of results included analysis of variance (ANOVA; repeated measures) and the post hoc Bonferroni correction. Participants remained healthy throughout the study period, including aminotransferases below the cut off in all measures. GGT, AST, and ALT activities were significantly different by group and by time (ANOVA repeated measures P < 0.05). Six-month curves of serum copper and ceruloplasmin concentrations were different by group, by time and interaction group × time (all P < 0.001). SHBG curves were different by group and time (P < 0.01), and interaction group × time (P < 0.009). Serum copper, ceruloplasmin, and liver aminotranferases are influenced by estrogens/progesterone, something that should be considered when these indicators are used as outcomes of effects. Time of sampling was also significantly associated with the indicators and deserves further study.  相似文献

Iron (Fe), copper (Cu), and zinc (Zn) fulfill various essential biological functions and are vital for all living organisms. They play important roles in oxygen transport, cell growth and differentiation, neurotransmitter synthesis, myelination, and synaptic transmission. Because of their role in many critical functions, they are commonly used in food fortification and supplementation strategies globally. To determine the involvement of divalent metal transporter 1 (DMT1) and human copper transporter 1 (hCTR1) on Fe, Cu, and Zn uptake, Caco-2 cells were transfected with four different shRNA plasmids to selectively inhibit DMT1 or hCTR1 transporter expression. Fe and Cu uptake and total Zn content measurements were performed in shRNA-DMT1 and shRNA-hCTR1 cells. Both shRNA-DMT1 and shRNA-hCTR1 cells had lower apical Fe uptake (a decrease of 51% and 41%, respectively), Cu uptake (a decrease of 25.8% and 38.5%, respectively), and Zn content (a decrease of 23.1% and 22.7%, respectively) compared to control cells. These results confirm that DMT1 is involved in active transport of Fe, Cu, and Zn although Zn showed a different relative capacity. These results also show that hCTR1 is able to transport Fe and Zn.  相似文献

To measure the effect of acute copper (Cu) administration, given as an aqueous solution, on the absorption of iron (Fe), 29 healthy adult women participated in two iron absorption studies. Subjects received 0.5 mg of Fe, as ferrous sulfate, alone or with Cu, as copper sulfate, at 0.5:1, 1:1, or 2:1 Cu/Fe molar ratios (study I) or at 4:1, 6:1, or 8:1 Cu/Fe molar ratios (study II) as an aqueous solution on days 1, 2, 14, and 15 of the study. Fe absorption was assessed by erythrocyte incorporation of iron radioisotopes 55Fe and 59Fe. Geometric mean (range ± SD) absorption of Fe alone or at 0.5:1, 1:1, 2:1 Cu/Fe molar ratios were 34.4% (17.3–68.5%), 40.9% (24.9–67.2%), 48.3% (24.8–94.1%), and 50.2% (25.3–99.5%), respectively (ANOVA, p = 0.12). Geometric mean (range ± SD) absorption of Fe alone or at 4:1, 6:1, 8:1 Cu/Fe molar ratios were 28.7% (12.1–67.9%), 21.5% (6.5–71.5%), 29.6% (10.3–85.4%), and 36.5% (18.3–73.1%), respectively (ANOVA, p = 0.16). In conclusion, combined Cu and Fe administration in an aqueous solution does not inhibit Fe bioavailability. This information could help in the design of rational guidelines for copper and iron supplementation programs. Our results support the hypothesis that divalent metal transporter 1 is not physiologically relevant for copper absorption in humans.  相似文献

Males (n=34) and females (n=23) of long-tailed duck (Clangula hyemalis) collected on the southern coast of the Baltic Sea in winter were studied to determine brain tissue concentration of iron (Fe), zinc (Zn), copper (Cu), manganese (Mn), and cadmium (Cd). There were no significant differences in the concentration of metals between males and females, except for Cd, the concentration of which in male brains was twice that of the females. In the brains of 20 birds (41% male and 26% female), the lead (Pb) concentration exceeded 1 μg/g wet wt. In all the duck brains examined, Spearman correlation coefficients were significant and positive for Zn−Mn, Cu−Mn, Cu−Fe, and Pb−Fe; a negative correlation was found for Pb−Zn. Additionally, female brains showed a positive correlation between Pb and Fe. The results obtained were compared with literature data for diving and nondiving anseriforms and birds of other taxa. It seems that concentration of heavy metals in the brain tissue of birds reflect natural adaptations (e.g., for diving) as well as effects of environmental pollution.  相似文献

An attempt has been made to analyze some trace elements and electrolytes like Zn, Cu, Mn, Fe, Co, Na, K, Ca, and Li present in the Swertia chirayita roots and leaves. The objective of the present study was to determine the effects of exercise and zinc deficiency on some elements in rats. Forty adult male Sprague–Dawley species male rats were allocated to four groups as follows: Group 1: control, Group 2: zinc-deficient, Group 3: exercise in which exercise group fed with a normal diet, Group 4: zinc-deficient exercise, exercise group fed by a zinc-deficient diet for 15 days. After the procedure ended, rats in groups 3 and 4 were exercised on the treadmill for 60 min at a speed of 6 m/min until the exhaustion. The rats were decapitated 48 h after exercise together with their controls, and blood samples were collected to determine copper (Cu), iron (Fe), magnesium (Mg), calcium (Ca), and phosphorus (P) levels. The highest Cu and Fe values in the serum were obtained in group 2 (p < 0.01). The levels of these elements in group 4 were lower than those in group 2 and higher than the levels in groups 1 and 3 (p < 0.01). Serum Mg levels did not differ significantly between groups. Group 4 had the lowest serum Ca and P levels (p < 0.01). These same parameters in Group 2 were higher than those in group 4 but significantly lower than those in groups 1 and 3 (p < 0.01). There was no significant difference between Ca and P levels of groups 1 and 3. The results of the study indicate that zinc deficiency adversely affects copper, iron, calcium, and phosphorus mechanisms and that these adverse effects much more marked after an effort exercise.  相似文献

A theoretical study on (ZrO2) n (n = 1–5) and Cu/ZrO2 oligomers is presented, DFT/B3LYP/6-31G** calculations along with Lanl2DZ pseudopotentials on metallic centers have been used to predict ionization potentials and electron affinities, chemical potentials and bandgaps indicating that the reactivity reaches reasonably constant values at n = 5. The effect of copper atoms adsorbed on (ZrO2) n is discussed and the reactivity of oligomers of ZrO2 and Cu/ZrO2 are compared, results indicate that Cu activates the systems by localizing the specific nucleophilic and electrophilic reactivity.  相似文献

There is accumulating evidence that the metabolism of several trace elements is altered in diabetes mellitus and that these nutrients might have specific roles in the pathogenesis and progress of this disease. The aim of present study was to compare the level of essential trace elements, chromium (Cr), copper (Cu), iron (Fe), manganese (Mn), nickel (Ni), and zinc (Zn) in biological samples (whole blood, urine, and scalp hair) of patients who have diabetes mellitus type 2 (n = 257), with those of nondiabetic control subjects (n = 166), age ranged (45–75) of both genders. The element concentrations were measured by means of an atomic absorption spectrophotometer after microwave-induced acid digestion. The validity and accuracy was checked by conventional wet-acid-digestion method and using certified reference materials. The overall recoveries of all elements were found in the range of (97.60–99.49%) of certified values. The results of this study showed that the mean values of Zn, Mn, and Cr were significantly reduced in blood and scalp-hair samples of diabetic patients as compared to control subjects of both genders (p < 0.001). The urinary levels of these elements were found to be higher in the diabetic patients than in the age-matched healthy controls. In contrast, high mean values of Cu and Fe were detected in scalp hair and blood from patients versus the nondiabetic subjects, but the differences found in blood samples was not significant (p < 0.05). These results are consistent with those obtained in other studies, confirming that deficiency and efficiency of some essential trace metals may play a role in the development of diabetes mellitus.  相似文献

 In the redox center of azurin, the Cu(II) is strongly coordinated to one thiolate S from Cys 112 and two imidazole Ns from His 46 and 117. This site yields a complex resonance Raman (RR) spectrum with >20 vibrational modes between 200 and 1500 cm–1. We have investigated the effects of ligand-selective isotope replacements on the RR spectrum of Pseudomonas aeruginosa azurin to determine the relative spectral contribution from each of the copper ligands. Growth on 34S-sulfate labels the cysteine ligand and allows the identification of a cluster of bands with Cu–S(Cys) stretching character between 370 and 430 cm–1 whose frequencies are consistent with the trigonal or distorted tetrahedral coordination in type 1 sites. In type 2 copper-cysteinate sites, the lower ν (Cu–S) frequencies between 260 and 320 cm–1 are consistent with square-planar coordination. Addition of exogenous 15N-labeled imidazole or histidine to the His117Gly mutant generates type 1 or type 2 sites, respectively. Because neither the above nor the His46Gly mutant reconstituted with 15N-imidazole exhibits significant isotope dependence, the histidine ligands can be ruled out as important contributors to the RR spectrum. Instead, a variety of evidence, including extensive isotope shifts upon global substitution with 15N, suggests that the multiple RR modes of azurin are due principally to vibrations of the cysteine ligand. These are resonance-enhanced through kinematic coupling with the Cu–S stretch in the ground state or through an excited-state A-term mechanism involving a Cu-cysteinate chromophore that extends into the peptide backbone. The trace metal levels in the tissues of two popular leafy vegetables Amaranthus caudatus and Corchrus olithorus widely consumed in Nigeria were assessed from a cultivated floodplain receiving effluents from diverse factories in Ibadan. Although the leaves are primarily consumed, the stems are usually used as a feed for farm animals while the roots are disposed by burning when dry or by composting. The objective of this work was to evaluate the level of trace elements in the tissues of these vegetables at harvest time when they become available to the human ecosystem for exposure to the accumulated trace metals, especially the leaves which are cooked and eaten as soup. The results of the mean trace metal levels in the analyses show that the leaves of A. caudatus had the highest bioconcentration in the following order Ba>Mn>Zn>Cu>Pb>Cr>Co>Ni>Cd>U>Sb, stems: Ba>Zn>Mn>Cu>Cr>Pb>Ni>Co>Cd>U>Sb, and roots: Mn>Ba>Zn>Cu>Cr>Pb>Ni>Co>U>Cd>Sb. In C. olithorus, the order was Mn>Ba>Zn>Cu>Pb>Cr>Co>Ni>Cd>Sb>U in the leaves, Mn>Zn>Ba>Pb>Cr>Cu>Co>Ni>U>Cd>Sb in the roots, and Mn>Zn>Ba>Cu>Cr>Pb>Co>Ni>Cd>U>Sb in the stems. The final result of the vegetable samples showed that the trace metal concentration was in the range of Cr (0.8–58.7), Mn (35.0–9,495.9), Co (0.3–33.6), Cu (2.3–60.3), Zn (16.0–538.2), Cd (0.000–40.53), Sb (0.000–0.037), Ba (13.0–1,175.6), Pb (0.9–39.7), and U (0.0–2.2). The bioconcentration factors (BF) for the transfer of trace metals from soil to the tissues showed a higher ease of bioaccumulation when compared to previous studies. The element with the highest BF in the study was Ba (6.45) in the leaves while the least was Co (0.09) in the roots of A. caudatus. The level of bioconcentration of trace metals in the leaves in most of the samples exceeds the recommended levels given by the World Health Organization (WHO) and Federal Environmental Protection Agency (FEPA), Nigeria and therefore constitutes a potential public health risk to the populace who consume these vegetables cultivated in effluent-impacted floodplains.  相似文献

Copper and other transition metal ions and their complexes are catalysts for the decomposition of nitrosothiols. In this way they catalyze the biological functions of nitrosothiols. The kinetics and mechanism of the reaction of two nitrosothiols, S-nitrosothiolactic acid and S-nitrosoglutathione (GSNO), with copper(I) are reported. The kinetics of the reaction of Cu(MeCN) n + (n=0–3) with the nitrosothiols were studied. The results indicate that Cu+ aq is the active species in the GSNO system, with k(Cu+ aq+GSNO)=(9.4 ±2.0)×107 dm3 mol−1 s−1 . The results also indicate that the Cu(MeCN) n + (n=0–3) complexes react with S-nitrosothiolactic acid. Transient species are formed in these processes. The results suggest that these species contain copper(I) and thiol. The results shed light on the catalytic role of copper complexes in the decomposition of S-nitrosothiols. Trace element content in hair is affected by the age of the donor. Hair samples of subjects from four counties in China where people are known to have long lifespan (“longevity counties”) were collected and the trace element content determined. Samples were subdivided into three age groups based on the age of the donors from whom these were taken: children (0–15 years); elderly (80–99 years); and centenarians (≥100 years). We compared the trace element content in hair of different age groups of subjects. Support vector machine classification results showed that a non-linear polynomial kernel function could be used to classify the three age groups of people. Age did not have a significant effect on the content of Ca and Cd in human hair. The content of Li, Mg, Mn, Zn, Cr, Cu, and Ni in human hair changed significantly with age. The magnitude of the age effect on trace element content in hair was in the order Cu > Zn > Ni > Mg > Mn > Cr > Li. Cu content in hair decreased significantly with increasing age. The hair of centenarians had higher levels of Li and Mn, and lower levels of Cr, Cu, and Ni comparing with that of the children and elderly subjects. This could be a beneficial factor of their long lifespan.  相似文献

Concentrations of cadmium, lead, iron, zinc, copper, chromium, nickel, aluminium and arsenium were analysed in blood serum of cattle from organic (n = 20) and conventional (n = 21) farms. The elements were determined by inductively coupled plasma atomic emission spectrometry using an Optima 2000 DV instrument (Perkin Elmer Inc.). Animals from the organic farm were characterised by significantly lower (P < 0.01) serum concentrations of Pb, Zn, Fe, Cu, Cr, Ni, As and Al compared to animals from the conventional farm. The concentration of Cd was similar in animals from both organic and conventional farms. The concentration of toxic elements in cattle from organic and conventional farms studied was very low. The trace essential elements were generally within the adequate ranges except Zn and Cu, which were deficient. In organically reared animals, also serum Fe content was below the critical level for diagnosing iron deficiency. Pb was significantly correlated with Cd, Zn, Fe, Cu and Ni. A significant positive correlation between the concentration of Cd and Zn, Cu and Ni concentrations was only observed in cows from the organic farm. The present results suggest that organically raised animals are less exposed to harmful environmental influences such as the environmental pollution with heavy metals. On the other hand, these animals are at a greater risk of mineral deficiency compared to animals kept on conventional farms.  相似文献

Determination of copper in human tissues and body fluids may be crucial in the diagnosis of Wilson’s disease. In this study we evaluated urinary copper excretion and urine and blood concentration in 14 patients in whom Wilson’s disease was confirmed (group A) and in 21 subjects in whom the disease was only suspected (group B). The following values (mean ± SD) were found: 24-h urine (μg Cu/24 h), 152 ± 135 (A) and 31.8 ± 10.9 (B); urine (μg Cu/ml), 0.091 ± 0.087 (A) and 0.028 ± 0.011 (B); and blood (μg Cu/ml), 0.62 ± 0.25 (A) and 0.72 ± 0.09 (B). By comparison, urine copper concentration in the group of apparently healthy subjects was 0.035 ± 0.010 (n = 50), and blood copper concentration in autopsy cases of nonpoisoned people was 0.85 ± 0.19 (n = 73).  相似文献

The present study was conducted to compare the trace elements and oxidative status between uremic patients with and without dementia. Chronic hemodialysis patients with dementia (n = 20) and without dementia (n = 25), and age-matched healthy volunteers (n = 20) were enrolled. The nutritional status, blood levels of trace elements aluminum (Al), zinc (Zn), copper (Cu), magnesium (Mg) and iron (Fe), malondialdehyde (MDA), and protein carbonyl production, antioxidant enzymes glutathione peroxidase (GPx), and glutathione reductase (GR) activities were measured. No significant difference in nutritional status or clinical characteristics was observed between nondementia and dementia patients. However, uremic patients with dementia have significantly higher Al, Cu, and Mg and lower Zn concentrations, as well as increased Cu/Zn ratio in comparison to nondementia patients. There were statistically significant increased MDA and carbonyl production and decreased GPx and GR activities in dementia patients. Furthermore, the significant associations of Al, Mg, and Cu/Zn ratio with oxidative status in patients with dementia were noted. The dementia may initially worsen with abnormal metabolism of trace elements and oxidative stress occurrence. Our results suggest that abnormalities in trace element levels are associated with oxidative stress and may be a major risk factor in the dementia development of uremic patients.  相似文献

Trace elements such as Zinc (Zn), copper (Cu), iron (Fe), and selenium (Se) are essential for male fertility. The fertilizing capacity of most animals reduced with advancing age. The objective of the present work was to determine the serum and testicular levels of Zn, Cu, and Se in young (10 months old) and old (30 months old) rabbits. Blood and testicular samples were obtained from rabbits after their slaughter. All samples were digested by concentrated acids and analyzed for trace elements by flame emission atomic absorption spectrophotometer. The results showed that serum Zn and Cu were lower in old rabbits than young (P < 0.01). Serum Se was higher in old than young rabbits (P < 0.05). Testicular Cu and Se were significantly lower in old than young rabbits (P < 0.01). However, Zn level was higher in old compared to young rabbits (P < 0.01). The serum Zn and Se were higher than the testicular levels (P < 0.01). The testicular Cu in both ages was higher than the serum level (P < 0.01), which suggest a very important role for Cu in the process of spermatogenesis in rabbits.  相似文献

This study aims at determining the association between markers of hepatic injury and serum, urinary, and intra-erythrocyte magnesium concentrations and dietary magnesium intake in obese children and adolescents. In a case–control study, 42 obese children and adolescents (8–18 years) and 42 sex- and puberty-matched controls were studied. Serum, urinary, and intra-erythrocyte magnesium levels, indices of insulin sensitivity, and liver enzymes were measured. Dietary magnesium intake was assessed using a food frequency questionnaire. Obese children and adolescents exhibited insulin resistance as determined by a higher fasting insulin and the HOMA-IR (p < 0.001) and lower QUICKI indices (p = 0.001); in addition these subjects had significantly higher intra-erythrocyte magnesium (IEM) concentrations, than non-obese ones (3.99 ± 1.05 vs. 3.35 ± 1.26 mg/dL of packed cell; p = 0.015). Among liver enzymes, only gamma-glutamyl transferase (GGT) was significantly higher in obese than in non-obese subjects (22.7 ± 9.4 vs. 17.1 ± 7.9 U/l; p = 0.002). A positive association was found between GGT and IEM in both groups; however in multivariate analysis, in obese subjects, only GGT (p = 0.026) and, in non-obese subjects, only age (p = 0.006) remained as significant predictors of IEM. In conclusion, increased IEM concentration was seen in insulin-resistant obese children and adolescents; furthermore, serum GGT was associated with IEM, independently of body mass index and HOMA-IR.  相似文献

The proper concentrations of plasma bioelements may favorably reduce the incidence of metabolic disorders, which often occur during immunosuppressive therapy. Mycophenolate mofetil (MMF) is currently one of the most frequently administered immunosuppressive agents; however, MMF treatment is often related to gastrointestinal side effects. The aim of this study was thus to verify whether the MMF treatment itself, or its metabolite pharmacokinetics, has an effect on the concentrations of plasma bioelements. To determine this, the effect of MMF on the levels of both major (sodium [Na], potassium [K], calcium [Ca], magnesium [Mg]), and trace (iron [Fe], zinc [Zn], copper [Cu]) plasma bioelements in 61 renal transplant recipients was assessed in comparison to a control group (n = 45). The pharmacokinetic parameters of mycophenolic acid were determined by the high-performance liquid chromatography method. All patients filled out a 24-h diet history questionnaire. The results showed high plasma concentrations of Fe and low plasma concentrations of Mg and Zn as compared with diagnostic norms. The patients treated with MMF had significantly lower plasma Na (P < 0.001) and significantly higher plasma Zn (P = 0.030) and Cu concentrations (P < 0.001). In conclusion, MMF treatment was found to affect plasma Fe, Zn, and Cu levels by increasing their concentrations while decreasing the plasma Na concentration. Mg and Zn deficiencies, as well as excessive Fe levels, are frequently observed irrespective of the immunosuppressive regimen applied, which suggests that monitoring of these bioelements may be favorable.  相似文献
