Racemic diisophorone (500 mg) was converted by Cephalosporium aphidicola and Neurospora crassa over 10 days at 25 °C to 8β-hydroxydiisophorone in yields of 10% (52 mg) and 20% (103 mg), respectively. Double-stranded RNA (dsRNA)-mediated interference (RNAi) is a powerful tool for silencing of gene expression in many organisms. To establish a DNA vector-based method for stable RNAi in Spodoptera frugiperda cells (Sf9), we created a stably transfected Sf9 cell line to express large dsRNA fragment targeting to silence the firefly luciferase gene (luc). The luc dsRNA specifically and stably suppressed the baculovirus-mediated luciferase expression. Thus, gene silencing in Sf9 cells was achieved using DNA vectors similar to the facile design described in this study. In Escherichia coli, disruption of ptsG, which encodes the glucose-specific permease of the phosphotransferase transport system (PTS) protein EIICBGlc, is crucial for high succinate production. This mutation can, however, cause very slow growth and low glucose consumption rates. The ptsG mutant (TUQ2), from wild type E. coli W1485, and E. coli galP (encoding galactose permease) and glk (encoding glucose kinase) gene expression plasmids were constructed. TUQ2 increased the generation time to approximately 4 h and gave a higher final cell density of 0.5 g/l by expression of galP. However, glk expression had no effect on the mutant. After expression of pyruvate carboxylase (PYC) and galactose permease, the ptsG mutant showed higher succinate yield (1.2 mol/mol glucose) and the specific rate of glucose consumption from 0.33 to 0.6 g/1 h. DNA photolyase can repair UV-induced DNA damage in a light-dependent manner. A cDNA of (6-4)photolyase from Dunaliella salina (GenBank accession number: AY845324) was cloned, sequenced and its amino acid sequence was deduced. The derived amino acid sequence showed high homology with other (6-4)photolyases and a predicted 3D model was constructed by homology modeling. The life history of the burrowing mayfly Ephoron virgo (Olivier, 1791) (Ephemeroptera: Polymitarcyidae) was studied during spring and summer 2005 in the lower Ebro river (Catalonia) and compared to a previous study performed in 1987 (Ibáñez, Escosa, Muñoz and Prat 1991). The results showed an advancement of Ephoron virgo life cycle and an increase of production estimates. In 2005 larval development reached the maximum size one month earlier than in 1987, and adult emergence peak began three weeks earlier. Comparing adult sex ratios (F:M), there was a major presence of females in 2005 (1:4), while the opposite was observed in 1987 (2:1). Secondary production was higher in 2005 than in 1987, obtaining 950 mg dry weight/m2/year with the increment summation method and 1080 mg dry weight/m2/year using the removal summation method. Higher water temperatures were measured for the entire 2005 larval growth period, which were related to higher air temperatures. Therefore, that temperature increment was likely the main cause of changes observed in the Ephoron virgo life cycle.  相似文献

The expression efficiency of an insect-derived cellulase was assayed in silkworm larvae infected with recombinant Bombyx mori nucleopolyhedrovirus (BmNPV) mutants lacking the virus-encoded chitinase (chiA) and/or cathepsin (v-cath) genes. Expression was increased by approx. 10% in mutants lacking chiA or v-cath and 17% in a mutant lacking both chiA and v-cath compared with that of the unmodified recombinant BmNPV. The recombinant BmNPV lacking both chiA and v-cath can therefore be used for a large-scale production of foreign proteins in silkworms. Osteopontin is a major non-collagenous bone matrix protein secreted into the mineralizing extracellular matrix by osteoblasts during bone development. Recombinant human osteopontin (hOPN) that includes the Arg-Gly-Asp (RGD) cell recognition site was expressed in Escherichia coli and the purified osteopontin increased cell adhesion, proliferation and differentiation of osteoblast cells (p<0.05). Salmonella were isolated from black-headed gulls (Larus ridibundus) in six locations in the Czech Republic from 1984 to 2005 (Chropyně and Nymburk in 1984–1986; Nové Mlyny, Bartošovice, and Hodonín in 1991–1994; and Nové Mlyny, Bartošovice, and Ostrava in 2005). Antimicrobial susceptibility was determined in 12 antimicrobial drugs using disk diffusion. Although 95% of Salmonella isolates (197 out of 207) were pansusceptible, the prevalences of resistance increased significantly from 1 (2%) out of 59 isolates in 1984–1986 and 3 (3%) out of 100 isolates in 1991–1994 to 6 (13%) out of 48 isolates in 2005. Furthermore, in 2005, two isolates were nalidixic acid-resistant and one isolate was multidrug-resistant Salmonella Typhimurium DT 104. These findings suggest that the occurrence of salmonellae in black-headed gulls depends to a large extent on the contamination where the gulls feed and possibly reflects the dissemination of these strains among farm animals and humans. Black-headed gulls may also become infected with resistant Salmonella and thus pose a potential risk of Salmonella contamination of surface water and animal feeds, and consequently dissemination.  相似文献

We measured the soil surface CO2 efflux (R S) from January 2005 to December 2006 in two neighboring stands in Gwangneung Forest, central Korea: evergreen coniferous forest (Abies holophylla, stand A) and broad-leaved deciduous forest (Quercus-dominated, stand Q). Regarding seasonal variation, R S rate was low during the winter and early spring months in each stand and peaked in late July [1170 (stand A) and 1130 (stand Q) in 2005, and 1000 (stand A) and 740 (stand Q) mg CO2 m−2 h−1 in 2006]. R S rate was higher in stand A than in stand Q during most of the growing season. The pattern of summer rainfall differed between 2005 and 2006. R S rate for both stands was suppressed significantly by the droughts in June 2005 and September 2006. After the heavy rainfall of July 2006, R S rate was lower than in July 2005 in both stands, but this decrement was much greater in stand Q than in stand A. In midsummer (August) 2006, under higher soil temperature (ST) and lower soil water content (SWC) conditions than in August 2005, R S rate of stand A was lower than that in August 2005, whereas stand Q showed no marked change. The exponential relationship between ST and R S accounted for approximately 91–97% of the R S variability in each stand and in each year. In stand A, the application of a second-order polynomial function indicated a significant correlation between SWC and R S when the soil was warm (ST > 15°C). Our results suggest that the seasonality of R S is strongly affected by the pattern of summer rainfall even in an Asia monsoon climate regime. In addition, the vegetation type (i.e., evergreen coniferous forest vs. broad-leaved deciduous forest) plays a significant role in response of R S to various environmental fluctuations such as drought, heavy rainfall, and hot-dry condition.  相似文献

Colloidal lithography was used to make a novel array (2-D) of micro-rings, dots, and interconnected-honeycomb structures. These geometries are controlled using the curing temperature-dependent rheological properties of the siloxane elastomer precursor. Serratia marcescens was patterned on the interconnected honeycomb microstructure demonstrating a potential application for microbioanalytical devices, microfluidics, and bio-micro-electromechanical systems. A linear relationship between total solid concentration (TSC), δ-endotoxin production [Cry = 0.2795(TSC)−0.2472, R2 = 0.8644] and poly-β-hydroxybutyrate (PHB) accumulation [PHB = 0.1327(TSC) + 0.3974, R2 = 0.9877] in Bacillus thuringiensis var. kurstaki HD-73 was observed. A similar correlation between δ-endotoxin and PHB accumulation [Cry = 2.1573(PHB)−1.1248, R2 = 0.9181] was found. A minimum PHB accumulation of 0.52 mg l−1 was required before the onset of δ-endotoxin production. The paper by Sand‐Jensen (2003 , Freshwater Biology, 48 , 271), and the subsequent comment and reply ( Sukhodolov, 2005 , Freshwater Biology, 50 , 194; Sand‐Jensen, 2005 , Freshwater Biology, 50 , 196), are discussed. I present partial theory describing how the macrophyte bending angle changes with increased flow velocity, demonstrating a nonlinear relation. 2. While Sand‐Jensen (2005) is correct that plant wetted area is the only characteristic area that can be measured easily, some other measure of the exposure area of the plant stand to the flow may be preferable. The relationship in Sand‐Jensen (2003) between the dimensionless drag coefficient and flow velocity depends on the definition of characteristic area. Hence, the analysis involving the drag parameter in Sand‐Jensen (2003) should be interpreted with care.  相似文献

Different antibiotic resistance (AR) genes, such as Bla, Tet and NPTII, contaminate commercially available Taq polymerases. The specificity of the AR gene PCR can be increased when using a restriction enzyme-based decontamination of polymerase. The elimination of Taq polymerase contamination allows the use of PCR tests to screen seeds (corn) and processed food for the presence of genetically modified organisms (GMO) based on the detection of AR genes. Platythyrea conradti is the only species in this genus with ergatoid (= permanently wingless) queens. Colonies lack gamergates (unlike other species in this genus), yet aggressive interactions among queen and workers define a hierarchy. A single fertile queen has the top rank and highranking workers do not lay eggs, except when the queen dies. Colonial reproduction by both alate queens (independent foundation) and gamergates (fission) seems the ancestral state in Platythyrea. Independent foundation can be selected against in some species, causing the loss of alate queens for economic reasons. Thus gamergates become the only reproductives, except in P. conradti in which queens became ergatoid. Gamergates and ergatoid queens are two mutually redundant reproductive phenotypes that allow colonial reproduction by fission. A sensitive method for quantification of S-adenosyl methionine (SAM) in microbial cell extracts was developed and applied to Corynebacterium glutamicum. The method is based on SAM being completely hydrolyzed into 18O-homoserine when extracted in boiling H218O and thus can be clearly distinguished by GC-MS analysis from naturally labeled homoserine present in the cell extract. Additional quantification of the total homoserine pool, representing both SAM and homoserine, via HPLC allows separate determination of both metabolites. A β-1,3-glucanase was detected, using laminarin as substrate, in the culture broth of Chaetomium sp. Major activity was associated with a 70 kDa protein band visualized on a polyacrylamide gel. β-1,3-Glucanase was purified by a one-step, native gel purification procedure. Optimal activity was observed at pH 6.0 and 30 °C (over 30 min). It could degrade cell walls of plant pathogens including Rhizoctonia solani, Gibberella zeae, Fusarium sp., Colletotrichum gloeosporioides and Phoma sp. The N-terminal amino acid residues of the purified β-1,3-glucanase are PYQLQTP, which do not exhibit homology to other fungal β-1,3-glucanases suggesting it may be a novel enzyme. Generally, microbial control agents such as entomopathogenic nematodes are applied in a curative manner for achieving pest suppression; prophylactic applications are rare. In this study, we determined the ability of two Steinernema carpocapsae strains (All and Hybrid) to prophylactically protect peach trees from damage caused by the peachtree borer, Synanthedon exitiosa, which is a major pest of stone fruit trees in North America. In prior studies, the entomopathogenic nematodes S. carpocapsae and Heterorhabditis bacteriophora caused field suppression when applied in a curative manner to established S. exitiosa populations. In our current study, nematodes were applied three times (at 150,000–300,000 infective juveniles/tree) during September and October of 2005, 2006, and 2007. A control (water only) and a single application of chlorpyrifos (at the labeled rate) were also made each year. The presence of S. exitiosa damage was assessed each year in the spring following the treatment applications. Following applications in 2006, we did not detect any differences among treatments or the control (possibly due to a low and variable S. exitiosa infestation of that orchard). Following applications in 2005 and 2007, however, the nematode and chemical treatments caused significant damage suppression. The percentage of trees with S. exitiosa damage in treated plots ranged from 0% damage in 2005 to 16% in plots treated with S. carpocapsae (Hybrid) in 2007. In control plots damage ranged from 25% (2005) to 41% (2007). Our results indicate that nematodes applied in a preventative manner during S. exitios’s oviposition period can reduce insect damage to levels similar to what is achieved with recommended chemical insecticide treatments.  相似文献

ABSTRACT Red‐faced Cormorants (Phalacrocorax urile) are North Pacific endemics recognized as a vulnerable species, but little is known about their breeding ecology. We studied Red‐faced Cormorants on St. Paul Island, Alaska, from 1975 to 2009, with more detailed data collected in 2004 and 2005. Mean clutch sizes in 2004 (3.2 ± 0.8 [SD] eggs) and 2005 (3.1 ± 0.8 eggs) were similar to the long‐term average (2.9 ± 0.3 eggs from 1976 to 2009). The mean laying interval in 2004 and 2005 was 2.15 ± 0.80 d (N= 407), and the mean egg period (number of days between laying of an egg and hatching) was 31.1 ± 1.4 d (N= 158). Approximately 64 ± 17% of eggs hatched during the period from 1975 to 2009. The mean number of chicks per nest in 2004 and 2005 was 2.8 ± 0.8 (N= 232), and the mean number of fledglings per initiated nest in all years was 1.22 ± 0.52. Chicks fledged 46 to 66 d posthatching. In 2004 and 2005, the primary causes of egg loss were predation by Arctic foxes (Vulpes lagopus) and destruction of eggs and abandonment of nests due to storms. Starvation was the primary cause of nestling mortality in both years. Because chicks are dependent on parents to provide food for over 45 d, consistent near‐shore foraging opportunities must be available. From 1975 to 2009, Red‐faced Cormorants experienced only 1 yr of complete reproductive failure (1984). The consistent reproductive success of Red‐faced Cormorants suggests that conditions may be relatively stable for this species on St. Paul Island, or that the variability in their breeding ecology (e.g., phenology, clutch sizes, and incubation strategies) provides the flexibility needed to successfully fledge some chicks nearly every year.  相似文献

The chemotactic responses of Bacillus thuringiensis subsp. dendrolimus (strain 49) and thuringiensis (strain 2002) and their morphological dissociants were studied by using some natural and artificial substances as effectors.
