为了评价利用不同冷冻保护剂冷冻早期卵裂期胚胎的效果,用小鼠为实验动物,采用慢速冷冻、快速融解的冷冻技术,比较丙二醇、二甲基亚砜和甘油作冷冻保护剂对小鼠2-细胞、4-细胞、8-细胞胚胎冷冻后胚胎存活率和囊胚形成率的影响。发现以丙二醇和蔗糖为冷冻保护剂冷冻4-细胞、8-细胞胚胎,解冻后胚胎成活率和囊胚形成率显著高于以二甲基亚砜或甘油为冷冻保护剂。结果表明,丙二醇是一种冷冻早期卵裂期小鼠胚胎有效的冷冻保护剂。  相似文献

目的研究胚胎冷冻在抗菌肽转基因FVB小鼠保种传代中的应用。方法对6~8周正常雌性FVB小鼠进行超排分别与雄性杂合子抗菌肽转基因FVB小鼠交配,收集2-cell胚胎,进行胚胎冷冻。1周后进行胚胎复苏移植,通过PCR方法对仔代鉴定。结果冻存胚胎140枚,复苏获得存活胚胎98枚,移植85枚,产仔38只,获得阳性后代12只。结论通过胚胎冷冻技术保种及复苏移植技术可对抗菌肽转基因小鼠进行传代。  相似文献

目的探讨EFS和DAP两种玻璃化冷冻方法对不同品系小鼠胚胎冷冻的效果。方法6个品系小鼠(KM、ICR、BALB/c、C57BL/6J、OB/OB、LAP/~TAOF59)的2-cell胚胎分别用EFS和DAP两种玻璃化冷冻方法进行冷冻和复苏,比较两种冷冻方法的胚胎复苏率和着床率。结果6个品系小鼠冷冻胚胎EFS方法的平均复苏率为69.97%(47.9%~83.6%),DAP方法的平均复苏率47.23%(26.3%-76.7%),EFS方法明显优于DAP方法。其中KM、ICR和BALB/c小鼠EFS方法的冷冻复苏率显著高于DAP方法(P〈0.01);冻融胚胎移植后EFS方法的平均着床率27.23%(1.75%一45.0%),DAP方法的平均着床率31.43%(7.0%一46.3%)。除KM、ICR小鼠外,其他4个品系小鼠的着床率DAP方法高于EFS方法。结论KM和ICR远交群小鼠胚胎适合用EFS方法冷冻保存;C57BL/6J、OB/OB、LAP/aTAOF59三个品系小鼠DAP方法优于EFS方法,但差异不大;BALB/c小鼠两种玻璃化冷冻方法的冻融胚胎着床率均较低,需进一步研究。  相似文献

目的建立绿色荧光蛋白转基因小鼠模型,并采取胚胎冷冻的方法进行保种。方法通过原核显微注射法,把线性化、纯化后的外源基因pEGFP注射入BDF1小鼠受精卵中,胚胎移植给同期发情的假孕受体母鼠,获得子代小鼠。经鉴定对有表达的转基因鼠进行胚胎冷冻保种。结果移植注射胚胎385枚给30只假孕小鼠共出生了306只后代鼠,经PCR和southern blot检测得到5只阳性小鼠。F2代转基因鼠胚胎冷冻240枚胚胎。结论通过显微注射法使外源基因pEGFP在小鼠基因组中得到整合,建立了转pEGFP的转基因小鼠模型。  相似文献

经程序化冷冻的小鼠休眠胚胎的基因表达谱差异分析   总被引:1,自引:0,他引:1  
目的探讨小鼠休眠胚胎经程序化冷冻后基因表达谱的变化及相关信号通路的改变趋势。方法采用Affymetrix基因芯片检测小鼠正常休眠胚胎和经程序化冷冻后的休眠胚胎的差异表达基因;采用GO分析和Pathway分析等生物信息学方法进一步了解相关信号通路的改变。结果经程序化冷冻后的小鼠休眠胚胎与正常休眠胚胎相比,存在228个差异表达基因,其中50个基因表达上调,178个基因表达下调。Pathway分析显示黏着斑通路、细胞外基质受体相互作用通路、肌动蛋白细胞骨架调节通路、细胞凋亡通路、细胞通讯通路、泛素介导的蛋白质水解通路、甘油磷脂代谢通路、小细胞肺癌通路、TGF-β信号通路、MAPK信号通路等基因差异表达变化趋势明显。结论小鼠休眠胚胎经程序化冷冻后会导致一系列基因调控变化,并可能影响多条信号通路的协同变化。  相似文献

目的检验小鼠休眠胚胎冻融后的质量及体内外发育潜力,为胚胎休眠技术的生产应用提供必要的参考。方法采用常规冷冻方法将正常孵化期胚胎和休眠胚胎进行冷冻,之后分别进行体外复苏培养实验和胚胎移植实验。随后利用双重荧光染色的方法分别对冻融前后的小鼠休眠胚胎与正常孵化期胚胎进行细胞计数,观察两种胚胎冻融前后的质量变化。结果休眠胚胎的冷冻解冻回收率、发育率均极显著高于孵化期胚胎(72.1%vs 50.2%,P<0.01;94.2%vs 73.9%,P<0.01)。休眠胚胎的移植妊娠率显著高于孵化期胚胎(40.8%vs 30.1%,P<0.05)。休眠胚胎的内细胞团细胞数显著高于孵化期胚胎(27.83 vs 19.53,P<0.05),滋养层细胞数差异不显著。冻融培养后休眠胚胎的内细胞团数,滋养层细胞数均显著高于孵化期胚胎(25.18 vs 14.68,P<0.05;114.09 vs 73.88,P<0.05)。结论小鼠休眠胚胎冻融后胚胎质量及体内外发育潜力均优于小鼠正常孵化期胚胎。  相似文献

牛血清白蛋白对小鼠原核期胚胎玻璃化冷冻的影响   总被引:1,自引:0,他引:1  
以小鼠原核期胚胎为对象,以胚胎的存活率、卵裂率、囊胚率以及囊胚细胞数作为检测指标,在M2液的基础上添加8种浓度(0,2,4,8,16,32,64,96mg/mL)牛血清白蛋白(BSA)配置防冻液,探讨防冻液和玻璃化冷冻后对胚胎发育的影响。BSA防冻液对胚胎发育影响的实验结果表明,8个浓度组间以及与对照组间胚胎的卵裂率、囊胚率以及囊胚细胞数无显著差异(P>0.05),说明在防冻液中加入一定浓度的BSA对小鼠胚胎无毒性作用。防冻液经玻璃化冷冻后对胚胎发育影响的实验表明,8个浓度组间冷冻胚胎复苏后的存活率、卵裂率、囊胚率及囊胚细胞数无显著差异(P>0.05)。表明BSA在这种防冻液中没有明显的保护作用。从经济、实用、生物安全角度考虑,不支持在玻璃化防冻液中添加BSA。  相似文献

目的 为扩大胚胎来源并获取特定胚龄胚胎 ,建立小鼠冷冻胚胎库。方法 运用超数排卵、体外受精与胚胎培养及胚胎冷冻技术系统研究了小鼠受精卵的体内发育与运行规律。卵母细胞的体外成熟与受精、单细胞胚胎培养及胚胎快速冷冻。结果  (1)注射hCG后 12~ 2 0h受精卵发育至原核期 ,4 2~ 4 8h为 2 细胞期 ,4 8~ 6 0h为 4 细胞期 ,6 0~ 6 8h为 8 细胞期 ,以上各期受精卵均处于输卵管中 ;75~ 78h为桑椹胚 ,78~ 80h为致密桑椹胚 ,90~ 92h为早期囊胚 ,92~ 96h为囊胚 ,以上各期均处于子宫角中。 (2 )培养液中添加促性腺激素 (FSH与hCG) ,能显著提高卵母细胞的体外受精率 ,添加FCS和激素组的体外受精率又显著高于单独添加激素组 ,FCS还能显著提高胚胎发育。 (3)在培养液中添加EDTA ,能有效克服小鼠胚胎的 2 细胞阻断 ,其 2 细胞胚的发育率达 10 0 % ,8 细胞胚发育率达 5 5 %以上 ;牛、羊上皮细胞培养液上清也能有效克服 2 细胞阻断。添加乳酸钠和丙酮酸钠可使 2细胞与 8细胞期胚的发育率显著提高。 (4)以D PBS +甘油 +蔗糖为冷冻液 ,以D PBS +蔗糖为稀释液 ,对小鼠胚胎进行快速冷冻 ,桑椹胚的存活率为 6 9 3% ,早期囊胚的存活率为 6 0 4 %。结论 研究为将生物技术应用于小鼠 ,扩大卵子和胚胎来源  相似文献

作为胚胎冷冻保存的基础性研究,冷冻保护剂的渗透性和毒性研究非常重要.本试验选用1,2-丙二醇、甘油、乙二醇和二甲基亚砜4种常用冷冻保护剂,对小鼠2-细胞胚胎进行渗透性和毒性研究.结果显示:1.5 mol/L的1,2-丙二醇、乙二醇和二甲基亚砜冷冻保护剂对2-细胞胚胎的渗透性显著高于甘油保护剂;4种冷冻保护剂对细胞膜的完整性没有影响;1.5 mol/L的乙二醇、1,2-丙二醇和甘油保护剂处理后的2-细胞胚胎的囊胚发育率和孵化率与对照组胚胎比较差异不显著(P>0.05),但显著高于二甲基亚砜处理后的2-细胞囊胚发育率和孵化率(P<0.01).结果表明:在4种冷冻保护剂中,乙二醇和1,2-丙二醇适合于小鼠2-细胞胚胎冷冻保存  相似文献

The cryopreservation of different embryo stages collected from ICR, C57BL/6 and F1 of DBA*C57BL/6 was carried out by using vitrification method. The morphology, in vitro development and birth rates of these embryos were compared after frozen-thawed. The results showed that more than 75% of the morphology from 2-cell embryos to morula stages from different strains was normal, the normal morphology rates of 8-cell embryos being the highest, while those of blastulas being the lowest. The in vitro development rates became higher as the embryos developed. The morphology of in vivo and in vitro fertilized frozen 2-cell embryos showed no difference, but the development rate of in vivo fertilized frozen 2-cell embryos was significantly higher than that of in vitro ones. Embryos that underwent 3 times frozen-thawing remained normal morphology. The pregnant rate and birth rate of frozen 2-cell embryos after embryo transfer were 64% and 40% respectively, but lower than those of fresh 2-cell embryo transfer.  相似文献

Cryopreservation of Mouse Embryos by Ethylene Glycol-Based Vitrification   总被引:1,自引:0,他引:1  
Cryopreservation of mouse embryos is a technological basis that supports biomedical sciences, because many strains of mice have been produced by genetic modifications and the number is consistently increasing year by year. Its technical development started with slow freezing methods in the 1970s1, then followed by vitrification methods developed in the late 1980s2. Generally, the latter technique is advantageous in its quickness, simplicity, and high survivability of recovered embryos. However, the cryoprotectants contained are highly toxic and may affect subsequent embryo development. Therefore, the technique was not applicable to certain strains of mice, even when the solutions are cooled to 4°C to mitigate the toxic effect during embryo handling. At the RIKEN BioResource Center, more than 5000 mouse strains with different genetic backgrounds and phenotypes are maintained3, and therefore we have optimized a vitrification technique with which we can cryopreserve embryos from many different strains of mice, with the benefits of high embryo survival after vitrifying and thawing (or liquefying, more precisely) at the ambient temperature4.Here, we present a vitrification method for mouse embryos that has been successfully used at our center. The cryopreservation solution contains ethylene glycol instead of DMSO to minimize the toxicity to embryos5. It also contains Ficoll and sucrose for prevention of devitrification and osmotic adjustment, respectively. Embryos can be handled at room temperature and transferred into liquid nitrogen within 5 min. Because the original method was optimized for plastic straws as containers, we have slightly modified the protocol for cryotubes, which are more easily accessible in laboratories and more resistant to physical damages. We also describe the procedure of thawing vitrified embryos in detail because it is a critical step for efficient recovery of live mice. These methodologies would be helpful to researchers and technicians who need preservation of mouse strains for later use in a safe and cost-effective manner.  相似文献

The present study describes the H(2)O-NaCl-ethylene glycol ternary system by using a differential scanning calorimeter to measure melting points (T(m)) of four different ratios (R) of ethylene glycol to NaCl and then devising equations to fit the experimental measurements. Ultimately an equation is derived which characterizes the liquidus surface above the eutectic for any R value in the system. This study focuses on ethylene glycol in part because of recent evidence indicating it may be less toxic to pancreatic islets than Me(2)SO, which is currently used routinely for islet cryopreservation. The resulting physical data and previously determined information regarding the osmotic characteristics of canine pancreatic islets are combined in a mathematical model to describe the volumetric response to equilibrium-rate freezing in varying initial concentrations of ethylene glycol.  相似文献

利用 EFS40 ,二步法对近交系 C5 7BL/6、DBA/2和远交群 ICR小鼠囊胚玻璃化冷冻保存 ,并对冷冻后胚胎体内、外发育效果进行比较。结果表明 ,相同条件下鲜胚经培养 ,近交系 C5 7BL /6小鼠的囊胚发育率 ( 93% )与 ICR( 1 0 0 % )相比差异不显著 ( P>0 .0 5 ) ;而两近交系的囊胚孵化率明显低于 ICR( P<0 .0 1 )。 C5 7BL/6、DBA/2小鼠囊胚冷冻后发育率 ( 93% ,96% )和孵化率 ( 5 2 % ,46% )与各自对照组 ( 1 0 0 % ,1 0 0 %和 61 % ,62 % )相比均无显著差异 ( P>0 .0 5 ) ;并且与 ICR冷冻组发育率和孵化率 ( 94% ,5 3% )之间也无显著差异 ( P>0 .0 5 )。两近交系冻胚移植妊娠与各自对照组和 ICR冷冻组比较均无显著差异 ( P>0 .0 5 )。C5 7BL/6胚胎移植产仔率 ( 35 % )与对照组 ( 5 1 % )之间差异显著 ( P<0 .0 1 ) ,而 DBA/2胚胎移植产仔率 ( 4 7% )与对照组和 ICR冷冻组 ( 39% ,5 8% )相比差异不显著 ( P>0 .0 5 )。  相似文献

The Mongolian gerbil (Meriones unguiculatus) has been used as a laboratory species in many fields of research, including neurology, oncology, and parasitology. Although the cryopreservation of embryos has become a useful means to protect valuable genetic resources, its application to the Mongolian gerbil has not yet been reported. In this study, we investigated the in vitro and in vivo developmental competence of Mongolian gerbil embryos cryopreserved by vitrification. In vivo-fertilized embryos were vitrified on the day of collection using the ethylene glycol (EG)-based solutions EFS20 and EFS40, which contained 20% and 40% EG, respectively, in PB1 containing 30% (w/v) Ficoll 70 and 0.5 M sucrose. First, we compared one-step and two-step vitrification protocols. In the one-step method, the embryos were directly transferred into the vitrification solution (EFS40), whereas in the two-step method, the embryos were exposed serially to EFS20 and EFS40 and then vitrified. After liquefying (thawing), late two-cell embryos (collected on day 3) vitrified by the two-step method showed significantly better rates of in vitro development to the morula stage compared to those vitrified by the one-step method (65% vs. 5%, P < 0.0001). We then examined whether the same two-step method could be applied to early two-cell embryos (collected on day 2), four-cell embryos (day 4), morulae (day 5), and blastocysts (day 6). After liquefying, 87%-100% of the embryos were morphologically normal in all groups, and 23% and 96% developed to the compacted morula stage from early two- and four-cell embryos, respectively. After transfer into recipient females, 3% (4/123), 1% (1/102), 5% (4/73), and 10% (15/155) developed to full-term offspring from vitrified and liquefied early two-cell embryos, late two-cell embryos, morulae, and blastocysts, respectively. This demonstrates that Mongolian gerbil embryos can be safely cryopreserved using EG-based vitrification solutions.  相似文献

Although embryo cryopreservation has become commonplace in many species, effective methods are not available for routine freezing of unfertilized eggs. Cryopreservation-induced damage may be caused by the high concentration of sodium ions in conventional freezing media. This study investigates the effect of a newly developed low-sodium choline-based medium (CJ2) on the ability of unfertilized, metaphase II mouse eggs to survive cryopreservation and develop to the blastocyst stagein vitro.Specifically, the effects of cooling to subzero temperatures, thawing rate, LN2plunge temperature, and equilibration with a low-sodium medium prior to freezing are examined. In contrast to cooling to 23, 0, or −7.0°C in a sodium-based freezing medium (ETFM), cooling in CJ2 had no significant negative effect on oocyte survival or development. Oocytes frozen in CJ2 survived plunging into LN2from −10, −20, or −33°C at significantly higher rates than oocytes frozen in ETFM. With the protocol used (1.5 M PrOH, 0.1 M sucrose, −0.3 C/min, plunging at −33°C) rapid thawing by direct submersion in 30°C water was more detrimental to oocyte survival than holding in air for 30 or 120 s prior to transfer to water. Equilibration of unfertilized oocytes with a low-sodium medium prior to cryopreservation in CJ2 significantly increased survival and blastocyst development. These results demonstrate that the high concentration of sodium in conventional freezing media is detrimental to oocyte cryopreservation and show that choline is a promising replacement. Reducing the sodium content of the freezing medium to a very low level or eliminating sodium altogether may allow oocytes and other cells to be frozen more effectively.  相似文献

Preimplantation embryos from cattle, sheep, and goats may be cryopreserved for short- or long-term storage. Preimplantation embryos consist predominantly of water, and the avoidance of intracellular ice crystal formation during the cryopreservation process is of paramount importance to maintain embryo viability. Embryos are placed into a hypertonic solution (1.4 – 1.5 M) of a cryoprotective agent (CPA) such as ethylene glycol (EG) or glycerol (GLYC) to create an osmotic gradient that facilitates cellular dehydration. After embryos reach osmotic equilibrium in the CPA solution, they are individually loaded in the hypertonic CPA solution into 0.25 ml plastic straws for freezing. Embryos are placed into a controlled rate freezer at a temperature of -6°C. Ice crystal formation is induced in the CPA solution surrounding the embryo, and crystallization causes an increase in the concentration of CPA outside of the embryo, causing further cellular dehydration. Embryos are cooled at a rate of 0.5°C/min, enabling further dehydration, to a temperature of -34°C before being plunged into liquid nitrogen (-196°C). Cryopreserved embryos must be thawed prior to transfer to a recipient (surrogate) female. Straws containing the embryos are removed from the liquid nitrogen dewar, held in room temperature air for 3 to 5 sec, and placed into a 37°C water bath for 25 to 30 sec. Embryos cryopreserved in GLYC are placed into a 1 M solution of sucrose for 10 min for removal of the CPA before transfer to a recipient (surrogate) female. Embryos cryopreserved in EG, however, may be directly transferred to the uterus of a recipient.  相似文献

四倍体胚胎的制备已经成为生物学家研究小鼠以及其它哺乳动物发育生物学的有力工具。目前,制备小鼠四倍体胚胎最常用的方法是电融合法,其中电融合液中的离了种类和离子浓度是影响胚胎电融合成败的关键因素。本文比较了Ca^2+、Mg^2+、Sr^+、Na^+、K^+、Li^+等阳离子对小鼠2-细胞胚胎电融合的影响。结果发现,在100V/mm电场强度、50μsec脉冲时程和2次脉冲的电融合条件下,少量二价阳离子的存在是胚胎融合所必须的,当Ca^2+为0.1mM时,可使全部胚胎发生电融合,而一价阳离子的存在不利于胚胎的电融合。随着各种离子浓度的大幅升高,胚胎的融合率急剧下降、胚胎死亡数量增加以及死亡程度加剧。  相似文献

为制作四倍体小鼠胚胎 ,用 4种脉冲电压强度 (6 0 0、 80 0、 10 0 0和 12 0 0V/cm )和 3种脉冲宽度(10、 30和 5 0 μs)组合 ,对 2 -细胞昆明小鼠胚胎做了融合实验。 6 0 0V/cm× 5 0 μs和 12 0 0V/cm× 30 μs两种组合融合率最高 ,分别为 91 6 %和 93 0 % ;而 6 0 0V/cm× 10 μs和 12 0 0V/cm× 5 0 μs时均显著低于其他各种组合 ,融合率分别为 5 8 9%和 6 0 2 %。以上结果表明 ,一定的脉冲电压强度是胚胎融合的必要条件 ,而脉冲宽度是影响胚胎融合及其后期发育的关键因素。激光共聚焦显微镜对融合后两细胞核的观察暗示 ,细胞核的迁移、靠近和融合是自发过程  相似文献

哺乳动物早期胚胎体外培养技术是研究早期胚胎发育和胚胎工程的基本手段。目前最常用的方法是采用液体石蜡覆盖的液滴培养法。该方法中所用液体石蜡和CO2质量的好坏对体外培养胚胎的发育有严重影响。本文就此对几种中国产液体石蜡和含气体乙醇的CO2作为胚胎体外培养条件对胚胎发育的影响进行了研究。取成熟小鼠受精后的2细胞和8细胞胚胎分别用于两组实验。一、不同品牌液体石蜡对胚胎发育的影响。体外培养采用液体石蜡覆盖液滴培养法。所用液体石蜡均经过水洗。CO2为经过一次水滤的工厂粗制品,其乙醇含量经气相色谱仪测定约为0.18%。二、不同浓度乙醇对胚胎发育的影响。采用试管培养法,采用乙醇浓度不同的培养液。表1为五个不同厂家的液体石蜡对早期胚胎体外培养的影响。其中,Ⅰ、Ⅱ(上海、北京)号两种产品符合胚胎体外培养的要求,可使2细胞后期胚胎发育到囊胚的比率达到92%以上,对细胞没有毒害作用(图1A和图2A、B)。其余,则不符合要求,对细胞有毒害作用(图1B),甚至用无水乙醇和水先后各洗涤3至4遍,亦无改善。由于影响液体石蜡质量的硝基萘可溶于乙醇而被清除,说明这些不合格的液体石蜡中可能还含有其它对胚胎发育有毒害作用的未知因素,有待进一步查明  相似文献
