Seasonal fluctuations in immune status have been documented for avian and mammalian populations. During the late summer and early fall, immune function is bolstered to help animals cope with the more physiologically demanding winter. The environmental cue for these seasonal changes is apparently decreasing photoperiod. In the present study, we determined the potential role of leptin in mediating the effect of photoperiod on cell-mediated immune responses in male mice. Leptin-deficient (ob/ob) and littermate control mice were housed for 10 wk in either a short (8L:16D) or a long (16L:8D) photoperiod beginning at 6 wk of age. After the mice were killed, immune and reproductive organs were weighed and splenocytes isolated. The proliferative and cytokine responses (interleukin [IL]-2 and IL-4) of splenocytes to the T-cell mitogen, concanavalin A (Con A; 0-40 microg/ml), were determined. Body weights were elevated and both testes and seminal vesicle weights subnormal in ob/ob mice (by ANOVA, main effect of leptin deficiency), but thymuses and spleens were of normal size. Serum leptin levels were at minimum detection limits in ob/ob mice, but leptin levels in control mice housed at 8L:16D were higher than in control mice housed at 16L:8D. The proliferative response of splenocytes from ob/ob mice to Con A was subnormal (by ANOVA, main effect of leptin deficiency), but photoperiod had no effect on this response. Production of IL-2 in splenocytes of ob/ob mice was subnormal (by ANOVA, main effect of leptin deficiency) irrespective of photoperiod, but cells from mice housed at 8L:16D (by ANOVA, main effect of photoperiod) produced more IL-2 than cells from animals housed at 16L:8D. In contrast, a leptin deficiency did not alter IL-4 production, but cells from animals (ob/ob and controls) housed at 16L:8D produced less IL-4 than cells from animals housed at 8L:16D (by ANOVA, main effect of photoperiod). The present study suggests that both photoperiod and leptin have mutually independent effects on the proliferation of lymphocytes and cytokine production profiles. The data do not provide definitive support for the hypothesis that photoperiod-induced changes in leptin secretion mediate the effects of season on immune status.

The concept of critical day length is well established among rodents; reproductive function is maintained when day lengths are greater than some specific threshold. In addition to day length cues, seasonal breeding in deer mice can also be regulated by food availability. The caloric threshold necessary to support reproduction remains unspecified for seasonally breeding rodents. The present study examined the interaction between photoperiod and food availability on reproductive function in adult male deer mice (Peromyscus maniculatus). A critical caloric intake profile was constructed in long (16L:8D) and short (8L:16D) photoperiods; groups of deer mice in both photoperiods either received food ad libitum or 90, 80, or 70% of their individual ad libitum food intake for 10 wk. At autopsy, paired testes, epididymides, and seminal vesicles were removed and weighed. Body mass, total body fat, and total body water contents were also obtained. Short, as compared to long, day lengths inhibited the reproductive systems of male deer mice. However, food consumption interacted with photoperiod to affect reproductive function. Significant reductions in reproductive organ size as well as spermatogenic activity were observed among short-day mice after a 10% reduction in ad libitum food intake. Long-day animals required a 20% reduction in caloric intake to depress reproductive function. Body mass and total body water content were generally unaffected by either photoperiod or food consumption. Total body fat content was reduced in short- as compared to long-day mice. Individual reproductive responsiveness to short days increased as food availability decreased.(ABSTRACT TRUNCATED AT 250 WORDS)

Individuals of many nontropical rodent species display reproductive, immunological, and somatic responses to day length. In general, short day (SD) lengths inhibit reproduction and enhance immune function in the laboratory when all other conditions are held constant. Most studies to date have focused on seasonal variation in immune function in adulthood. However, perinatal photoperiods also communicate critical day length information and serve to establish a developmental trajectory appropriate for the time of year. Nontropical rodents born early in the breeding season undergo rapid reproductive development, presumably to promote mating success during their first reproductive season. Rodents born late in the breeding season suspend somatic growth and puberty until the following vernal breeding season. We tested the hypothesis that perinatal day lengths have similar enduring effects on the immune system of rodents. Siberian hamsters (Phodopus sungorus) were maintained prenatally and until weaning (21 days) in either SDs (8 h light:16 h dark) or long days (LD) (16 h light:8 h dark), then they were weaned into either the opposite photoperiod or maintained in their natal photoperiod, forming four groups (LD-LD, LD-SD, SD-LD, and SD-SD). After 8-wk in these conditions, cell-mediated immune activity was compared among groups. SD-SD hamsters of both sexes enhanced immune function relative to all other groups. The reproductive effects of perinatal photoperiod were not evident by the end of the experiment; circulating testosterone and cortisol sampled at the end of the experiment reflected the postweaning, but not the perinatal photoperiod. This experiment demonstrates long-lasting organizational effects of perinatal photoperiod on the rodent immune system and indicates that photoperiod-induced changes in the immune system are dissociable from changes in the reproductive system.

Photoperiod is a significant modulator of behavior and physiology for many organisms. In rodents changes in photoperiod are associated with changes in circadian period and photic resetting of circadian pacemakers. Utilizing rhythms of in vivo behavior and in vitro mPer2::luc expression, we investigated whether different entrainment photoperiods [light:dark (L:D) 16:8 and L:D 8:16] alter the period or phase relationships between these rhythms and the entraining light cycle in Per2::luc C57BL/6J mice. We also tested whether mPer2::luc rhythms differs in anterior and posterior suprachiasmatic nucleus (SCN) slices. Our results demonstrate that photoperiod significantly changes the timing of the mPer2::luc peak relative to the time of light offset and the activity onset in vivo. In both L:D 8:16 and L:D 16:8 the mPer2::luc peak maintained a more stable phase relationship to activity offset, while altering the phase relationship to activity onset. After the initial cycle in culture, the period, phase, and peaks per cycle were not significantly different for anterior vs. posterior SCN slices taken from animals within one photoperiod. After short-photoperiod treatment, anterior SCN slices showed increased-amplitude Per2::luc waveforms and posterior SCN slices showed shorter-duration peak width. Finally, the SCN tissue in vitro did not demonstrate differences in period attributable to photoperiod pretreatment, indicating that period aftereffects observed in behavioral rhythms after long- and short-day photoperiods are not sustained in Per2::luc rhythms in vitro. The change in phase relationship to activity onset suggests that Per2::luc rhythms in the SCN may track activity offset rather than activity onset. The reduced amplitude rhythms following long-photoperiod treatment may represent a loss of coupling of component oscillators.

Daylengths during the spring are repeated in reverse order in the autumn. For some photoperiodic species, a given photoperiod may be stimulatory for reproduction in the spring and inhibitory in the autumn. The mechanisms regulating this type of seasonal response have, until recently, remained a mystery. Horton (1984a) showed in Microtus montanus that the photoperiod experienced by the mother influences the gonadal development of her young after weaning. To determine if this phenomenon is characteristic of other photoperiodic rodents, adult Djungarian hamsters were paired on 16L:8D, 14L:10D, or 12L:12D. Young males born from these pairings were killed at 15, 28, and 34 days of age to assess gonadal development (testes weight). At 15 days testicular development was identical in all groups; by 28 days, however, males raised in 16L:8D or 14L:10D exhibited a greater degree of testicular development than those raised in 12L:12D. Next, females maintained on each of the three photoperiods throughout gestation were transferred, with their offspring, to the other two photoperiods at birth. Postnatal exposure to 14L:10D or 12L:12D inhibited testicular development in young that had been gestated on 16L:8D. Both 16L:8D and 14L:10D stimulated testicular growth in animals that had been gestated on 12L:12D or 14L:10D. Therefore, a) 16L:8D stimulates testicular growth in all animals, b) 12L:12D inhibits testicular growth in all animals, and c) the testicular response to 14L:10D depends on the photoperiod experienced by the mother during pregnancy.(ABSTRACT TRUNCATED AT 250 WORDS)

Descendants of a sample of cane mice (Zygodontomys brevicauda) trapped at 8 degrees latitude in Venezuela were tested for reproductive photoresponsiveness. This species breeds continuously, year around, despite living in a seasonally harsh habitat. At 50 days of age there were no differences in the weights of the testes or seminal vesicles or in sperm counts of males born and reared on 16L:8D, 13L:11D, 11L:13D, or 8L:16D photoperiods, although there were small differences in body weight. Females born and reared on 16L:8D vs. 8L:16D cycles became pregnant at the same rates and ages when paired with males at 21 or 31 days of age. The daily duration of melatonin secretion depended on the length of the dark phase of the cycle in both sexes. Circulating levels of melatonin were elevated for 8 h on a 16L:8D cycle and for between 9 and 16 h on an 8L:16D cycle. In this tropical species, the neuroendocrine pathway that links photoperiod to reproduction apparently is disconnected somewhere between melatonin and gonadotropin secretion, causing cane mice to be reproductively unresponsive to variation in photoperiod.

Energetic demands are high while energy availability is minimum during winter. To cope with this energetic bottleneck, animals exhibit numerous energy-conserving adaptations during winter, including changes in immune and reproductive functions. A majority of individual rodents within a population inhibits reproductive function (responders) as winter approaches. A substantial proportion of small rodents within a species, however, fails to inhibit reproduction (nonresponders) during winter in the field or in the laboratory when maintained in winter-simulated day lengths. In contrast, immune function is bolstered by short day lengths in some species. The specific mechanisms that link reproductive and immune functions remain unspecified. Leptin is a hormone produced by adipose tissue, and several studies suggest that leptin modulates reproductive and immune functions. The present study sought to determine if photoperiodic alterations in reproductive function and leptin concentrations are linked to photoperiod-modulated changes in immune function. Siberian hamsters (Phodopus sungorus) were housed in either long (LD 16:8) or short (LD 8:16) day lengths for 9 wk. After 9 wk, blood samples were collected during the middle of the light and dark phase to assess leptin concentrations. One week later, animals were injected with keyhole limpet hemocyanin to evaluate humoral immunity. Body mass, body fat content, and serum leptin concentrations were correlated with reproductive responsiveness to photoperiod; short-day animals with regressed gonads exhibited a reduction in these measures, whereas short-day nonresponders resembled long-day animals. In contrast, immune function was influenced by photoperiod but not reproductive status. Taken together, these data suggest that humoral immune function in Siberian hamsters is independent of photoperiod-mediated changes in leptin concentrations.

Changes in photoperiod can significantly impact the physiology of many species. For example, we have observed an improvement in cellular immune function in cattle on short-day photoperiod (SDPP) relative to long-day photoperiod (LDPP). In addition, prolactin (PRL) and PRL receptor (PRL-R) are affected by photoperiod management. Our hypothesis is that the inverse relationship observed between PRL and PRL-R mRNA expression during photoperiod treatment alters the sensitivity of the animal to PRL, thereby affecting the changes in their cellular immune function. The objective of this study was to determine the effects of exogenous PRL on photoperiodic-mediated immune responses. Eight Holstein steers received each of four treatments: LDPP (16L:8D), SDPP (8L:D), SDom (SDPP plus PRL via osmotic minipump for 10 days), and SDinj (SDPP plus PRL via 3x daily injections for 10 days). Steers on SDPP had decreased PRL relative to the other treatments. Expression of PRL-R mRNA was increased in SDPP animals relative to LDPP, SDom, and SDinj. Prior to PRL treatment, SDPP animals had greater lymphocyte proliferation and neutrophil chemotaxis relative to LDPP animals. Following PRL treatment, cellular immune function of SDom and SDinj animals was reduced to the level of LDPP animals. Addition of PRL to the in vitro lymphocyte proliferation did not alter response of LDPP animals but increased proliferation of lymphocytes from SDPP animals. The results of these experiments suggest that an animal's responsiveness to PRL correlate to changes in cellular immune function that occur with photoperiod manipulation.

1. 1.|The migratroy hamster Cricetulus migratorius, a small nocturnal rodent, inhabits ecosystems characterized by dramatic seasonal fluctuations of ambient temperatures. The aim of this study was to assess seasonal acclimatization of its thermoregulatory system.

2. 2.|Heat production by means of oxygen consumption and body temperature in various ambient temperatures as well as non-shivering thermogenesis were measured in C. migratorius. The hamsters were acclimated to two different photoperiod regimes (16L:8D and 8L:16D) at a constant ambient temprature of 24°C. Overall thermal conductance was calculated for such hamsters.

Many nontropical rodent species experience predictable annual variation in resource availability and environmental conditions. Individuals of many animal species engage in energetically expensive processes such as breeding during the spring and summer but bias investment toward processes that promote survival such as immune function during the winter. Generally, the suite of responses associated with the changing seasons can be induced by manipulating day length (photoperiod). Collared lemmings (Dicrostonyx groenlandicus) are arvicoline rodents that inhabit parts of northern Canada and Greenland. Despite the extreme conditions of winter in their native habitat, these lemmings routinely breed during the winter. In the laboratory, collared lemmings have divergent responses to photoperiod relative to other seasonally breeding rodents; short day lengths can stimulate, rather than inhibit, the reproductive system. Male and female collared lemmings were maintained for 11 weeks in 1 of 3 photoperiods (LD 22:2, LD 16:8, or LD 8:16) that induce markedly different phenotypes. Following photoperiod treatment, cell-mediated immune function as assessed by delayed-type hypersensitivity reactions was elevated in lemmings housed in LD 16:8 and LD 8:16 relative to LD 22:2. However, antibody production to a novel antigen was unaffected by photoperiod. Exposure to LD 8:16 induced weight gain, molt to a winter pelage, and in contrast to previous studies, regression of the male, but not the female, reproductive tract. In conclusion, these data indicate that components of immune function among collared lemmings are responsive to changes in day length.

Environmental factors play an important role in the seasonal adaptation of body mass and thermogenesis in wild small mammals. In this study, we performed a factorial experiment (temperature x photoperiod) in which Brandt's voles and Mongolian gerbils were acclimated to different photoperiods (long photoperiod, 16L : 8D; short photoperiod, 8L : 16D) and temperatures (warm, 23 degrees C; cold, 5 degrees C) to test the hypothesis that photoperiod, temperature, or both together can trigger seasonal changes in serum leptin level, body mass, thermogenesis, and energy intake. Our data demonstrate that Brandt's voles showed a remarkable decrease in body mass in both the cold and a short photoperiod. However, no significant changes in body mass were found for gerbils exposed to similar conditions. The short photoperiod induced a decrease in serum leptin levels for both voles and gerbils that might contribute to an increase in energy intake. Furthermore, the short photoperiod induced an increase of uncoupling protein 1 (UCP1) content for both voles and gerbils, and cold can further enhance the increase in voles. No interactions between photoperiod and temperature were detected for the two species. Brandt's voles can decrease their body mass through changes in energy intake and expenditure, while Mongolian gerbils can keep body mass relatively stable by balancing energy metabolism under winterlike conditions. Leptin was potentially involved in the regulation of body mass and thermogenic capacity for the two species.

Pubertal development in prairie deer mice (Peromyscus maniculatus bairdii) is accelerated by exposure of juveniles to a long-day photoperiod, and, conversely, retarded by exposure to short days. The purpose of the present study was to evaluate the possible involvement of the circadian system in the photoperiodic regulation of puberty. Weanling males, previously housed on a short-day light cycle of 6L:18D, were subjected to a "resonance" protocol in which they received one of the following light cycles: 6L:18D, 6L:30D, 6L:42D, 6L:54D, or 16L:8D. Post-weaning exposure to cycles of 16L:8D, 6L:30D, and 6L:54D stimulated reproductive organ growth as measured at 6 weeks of age. Exposure to cycles of 6L:18D and 6L:42D failed to stimulate reproductive development. These data support the hypothesis that young male deer mice use a circadian rhythm of responsiveness to light to measure photoperiodic time and, consequently, regulate pubertal development.

Effects of short-day photoperiod, pinealectomy, and melatonin on sexual maturation were tested in Peromyscus leucopus from either Connecticut (CT) or Georgia (GA). Laboratory reared-stocks from CT and GA were exposed to short daylength (photoperiod) from birth or 25 days of age. At 12 wk of age, delay in sexual maturation was indicated in most CT mice by decreased testis length, combined testes weight, and seminal vesicle weight. Conversely, GA animals did not delay sexual maturation when exposed to short-day photoperiod from either birth or 25 days of age. These results indicate that responses to short daylengths differ for juvenile CT and GA populations. In a second experiment, pinealectomized or sham-operated CT males were exposed to short-day (9L:15D) or long-day (16L:8D) photoperiod from birth. Pinealectomy blocked the effect of short daylength on reproduction. Therefore, the pineal must be involved in the delay of sexual maturation observed for short-day CT mice. The effects of melatonin, a pineal gland hormone, were tested with chronic s.c. implants or daily injections. In CT mice given either melatonin implants or afternoon injections, sexual maturation was delayed. GA mice were insensitive to all melatonin treatments. Further, no differences in circadian organization (phase angle, duration of activity, period under constant dark) between GA and CT animals were apparent. Collectively, these studies indicate that melatonin is involved in the mechanism responsible for delay of sexual maturation in CT mice. Short-day insensitivity of GA Peromyscus leucopus probably results from a deficiency in the melatonin effector pathway and is not due to a disruption of circadian organization.

Two groups of ovary-intact ewes were placed in separate photochambers on the day of the vernal equinox (VE). One group was exposed to a 16 h light:8 h dark (16L:8D) photoperiod and the other to 8L:16D. On the day of the summer solstice (SS) and at 90-91-day intervals thereafter [autumnal equinox (AE), winter solstice (WS), VE and SS], each group was changed to the opposite photoperiod. The latent period between each change and either onset or cessation of cycles, as determined by measuring blood progesterone concentrations, was recorded. The latent period between change to 8L:16D and onset of cycles was shortest after the exposure at AE and longest after exposure at WS (P less than 0.001). The latent period after AE was shorter (P less than 0.001) than after VE. The correlations were small between ambient temperature and interval to onset of cycles. The latent period to cessation of cycles in response to 16L:8D was shorter after SS exposure than after WS exposure (P less than 0.01), but other differences were not significant. There was a strong (r = -0.94, P less than 0.05) negative correlation between interval to cessation of cycles and ambient temperature. Cessation of cycles in response to 16L:8D occurred more rapidly (P less than 0.001) than onset in response to 8L:16D. These results show that responsiveness to the inductive effects of photoperiod varies significantly with time of the sidereal year.

Adult male marbled newts (Triturus marmoratus) were collected at the end of the spermatogenesis period and exposed to different photoperiods (natural-daylength-simulated photoperiod, total darkness, 8L:16D, 12L:12D, 16L:8D, and continuous light) for 3 mo. Temperature was maintained at 20 degrees C. Two additional groups of newts were blinded and exposed to either the natural-simulated photoperiod and to 16 h of light per day respectively. Quantitative histologic studies on testicular development and germ cell volume per testis were performed. The newts captured in the field at the beginning (initial controls) or at the end of the experiments (final controls) were in the period of testicular quiescence. Newts kept in total darkness or exposed to a short photoperiod (8L:16D) showed germ cell development up to primary spermatocytes, whereas germ cell development in the newts exposed to long photoperiods (12L:12D or 16L:8D) progressed to elongated spermatids. The newts exposed either to intermediate photoperiods (natural-simulated photoperiod) or to constant light showed an intermediate degree of germ cell development (up to round spermatids). No significant differences between non-blinded and blinded animals were found. These results suggest that (1) mild temperature initiates testicular development in the period of testicular quiescence, (2) long photoperiods associated with mild temperatures produce spermatogenesis in this period, (3) complete darkness or constant light are less effective than some intermediate photoperiod, and (4) the effect of photoperiod on testicular function in newts is not related to ocular photoreception.

We tested the hypothesis that adult male rufous-winged sparrows, Aimophila carpalis, exhibit relative photorefractoriness. This condition results in partial loss of sensitivity to photoperiod as a reproductive stimulus after prolonged exposure to long photoperiods and is similar to the mammalian condition called photoperiodic memory. Captive birds were exposed either to 8 h of light/16 h of dark per day (8L) or to 16L for 11 weeks and were then exposed either to 8L, 13L, 14L, or 16L. Testicular diameter, plasma luteinizing hormone (LH), and plasma prolactin (PRL) were measured to assess reproductive system activity in response to photostimulation. In free-living birds, testicular diameter, plasma LH, and PRL were compared in birds caught in September in a year when birds were breeding and in a year when birds were not breeding to further evaluate the role of PRL in the termination of seasonal breeding. Testes completely developed after transfer from 8L to 14L or to 16L and partially developed after transfer from 8L to 13L. However, after 11 weeks of 16L exposure, transfer to 14L caused partial regression and transfer to 13L caused complete regression of the testes. Plasma LH increased in all birds that were transferred from 8L to a longer photoperiod. PRL showed a weak response to longer photoperiod treatment and was elevated in birds after chronic 16L exposure in comparison to birds exposed to chronic 8L. These data indicate that male rufous-winged sparrows lose sensitivity to photoperiod after long photoperiod exposure consistent with the relative photorefractoriness and photoperiodic memory models. Lower PRL in birds that developed testes on 13L and 14L compared to birds that regressed testes on 13L and 14L are consistent with the hypothesis that PRL regulates relative photorefractoriness. However, PRL does not appear to regulate interannual differences in the timing of testicular regression.

The peroxidase-antiperoxidase method was used to determine quantitatively the effect of short photoperiod-induced gonadal regression on the immunoreactive gonadotropin-releasing hormone (GnRH) neuronal system of female Peromyscus leucopus. In mice exposed to either long (16L:8D) or short (8L:16D) photoperiod, immunoreactive cell bodies were loosely organized into six groups: olfactory peduncle, diagonal band of Broca, septum, preoptic area (POA), anterior hypothalamus (AH), and basal hypothalamus. The POA and AH contain the largest number of cell bodies, which supply the major GnRH innervation to the median eminence (ME) and several extrahypothalamic brain sites. Exposure to short photoperiod increased the number of immunoreactive cell bodies within the anterior hypothalamus and preoptic area (AHPOA) and also increased the optical density for staining of immunoreactive cell bodies in the AHPOA and olfactory peduncle. The ME of mice exposed to short photoperiod had a higher density of GnRH fibers relative to that of mice exposed to long photoperiod, and the content of GnRH fibers in the rostral ME was correlated with the optical content for immunostaining of cell bodies in the AHPOA. These results are evidence that gonadal regression induced by short photoperiod (mediated by the pineal gland) involves alterations of GnRH neuronal activity. Notably, data from this study are consistent with the hypothesis that suppressed release of GnRH from neurovascular terminals in the ME, rather than lack of availability of the decapeptide, promotes gonadal regression.
