Indirect immunofluorescent staining revealed that Prostatic Binding Protein, the major androgen-dependent protein in rat ventral prostate invivo, is associated specifically with the epithelial cells in primary cell cultures derived from rat ventral prostate. The epithelial cells release Prostatic Binding Protein into the medium during primary culture. Denovo synthesis of Prostatic Binding Protein is demonstrable during early phases of cell culture. Prostatic Binding Protein is an excellent marker for the identification of functional prostate epithelial cells and for the study of regulation at the cellular level of the synthesis and secretion of a major androgen-dependent prostate protein.  相似文献

In the presence of polyamines, the fidelity of protein synthesis in a wheat germ cell-free system was increased significantly, while it was increased slightly in an E. coli cell-free system. The effective concentration of polyamines for the increase in fidelity of protein synthesis was nearly equal to that for the stimulation of protein synthesis in a wheat germ cell-free system.  相似文献

Evidence for distinct mRNAs for ferritin subunits   总被引:5,自引:0,他引:5  
Poly A enriched RNA from iron loaded HeLa cells and rat liver were translated separately and together in wheat germ lysates to investigate the origins of the H and L subunits of ferritin. Most of the ferritin translated from the HeLa RNA was of the H type, while that from the liver RNA was mostly L type. Mixtures of these RNAs gave HL ratios which correlated with the relative amounts of added HeLa and rat RNAs. These results indicate that the H and L subunits of ferritin are not derived by post-translational modification but from distinct mRNA species.  相似文献

6-Thioguanine was administered to rats 12 hr after partial hepatectomy at a dose of 40 mg/kg of body weight; 6 hr later, polyadenylic acid-containing RNA was isolated and was used to measure initiation of protein synthesis in vitro in a wheat germ system. In vitro initiation was found to be 2.3-fold greater when 6-thioguanine-containing RNA was employed, than when polyadenylic acid-containing RNA isolated from untreated animals was used. The homopolymer, poly(TG), did not promote peptide synthesis in the wheat germ in vitro system employed.  相似文献

Messenger ribonucleic acid isolated from angler fish (Lophiusamericanus) islets of Langerhans was translated in the wheat germ cell-free protein synthesizing system containing different combinations of radioactive amino acids. Preproinsulin (~ 11,000 daltons) was identified amongst the translation products, by sodium dodecyl sulfate gel electrophoresis, and subjected to microsequencing techniques. The fish preproinsulin was found to possess an NH2-terminal signal peptide of 24 amino acids, with regions of homology to human, rat and chicken preproinsulin signal sequences.  相似文献

The saxitoxin-binding component of the excitable membrane sodium channel exhibits glycoprotein characteristics as evidenced by its specific interaction with various agarose-immobilized lectins. The detergent-solubilized saxitoxin-binding component interacts quantitatively with immobilized wheat germ agglutinin and concanavalin A and fractionally with immobilized Lens culinaris hemagglutinin and Ricinus communis agglutinin. These lectins preferentially bind N-acetylglucosamine and sialic acid (wheat germ agglutinin), mannose (concanavalin A and Lens cunilaris and galactose (Ricinus communis). Removal of terminal sialic acid residues by neuraminidase markedly decreases binding to immobilized wheat germ agglutinin but uncovers sites capable of interacting with lectins specific for galactose and N-acetylgalactosamine. β-N-acetylglucosaminidase, an exoglycosidase has no effect on the binding of the channel protein to wheat germ agglutinin. Similarly, phospholipase C has no effect on binding of the solubilized toxin binding component to this lectin. Neither wheat germ agglutinin nor concanavalin A free in solution alters the number of toxin binding sites or their affinity for toxin. The sodium channel saxitoxin-binding component appears to be a glycoprotein containing terminal sialic acid residues and internal mannose, galactose, N-acetylglucosamine, and N-acetylgalactosamine residues. The toxin binding site is spatially separated from the binding sites for the lectins studied. The effect of these sugar moieties must be considered when evaluating the biophysical parameters of the sodium channel.  相似文献

A novel protein was found in the nuclei of rat ventral prostate. This protein has a molecular weight of about 21 kDa as measured by SDS-polyacrylamide gel electrophoresis. It showed a characteristic change between 3 and 84 weeks after birth in close association with the level of testosterone in the blood. After castration, the level of the 21-kDa protein decreased to 160 of normal in 7 days, but on daily injection of testosterone the level was restored to normal in 8 days and to twice the normal level in 14 days. Unlike H1 and H10 histone and high mobility group proteins, the 21-kDa protein was not extracted with 5% HClO4, but was partially extracted with 0.35 M NaCl. The 21-kDa protein was not found in kidney, liver, or brain, suggesting that it is specific to the ventral prostate.  相似文献

The binding sites for the lectins wheat germ agglutinin, Ricinus communis agglutinin and concanavalin A on mouse neuroblastoma cell membranes were identified using SDS-gel electrophoresis in combination with fluorescent lectins. Ricinus communis agglutinin and wheat germ agglutinin were found to bind almost exclusively to a single polypeptide with an apparent molecular weight of 30 000. Concanavalin A labeled over 20 different polypeptides, most with molecular weights greater than 50 000. However, when the neuroblastoma cells were treated with concanavalin A so as to internalize all the concanavalin A binding sites visible at the level of the fluorescent microscope and the purified plasma membranes analyzed for their concanavalin A binding polypeptides, only four of the 20 glycopolypeptides were missing or significantly reduced in amount. Thus, these four high molecular weight concanavalin A-binding polypeptides appear to be the major cell surface receptors for concanavalin A. Binding studies with iodinated concanavalin A indicated that these polypeptides represented the high affinity concanavalin A binding sites Kd = 2 · 10?7M). Low affinity concanavalin A binding sites were present on the cell surface after internalization of high affinity concanavalin A binding sites.  相似文献

The reduction of 3H-androstanolone into 3β-androstanediol was used to study the 3β-hydroxysteroid oxidoreductase activity of rat ventral prostate. This activity is present only in the cytosol and has a pH optimum of 8.5 with NADH as cofactor. The 3β-hydroxysteroid oxidoreductase activity , as compared to 3α-hydroxysteroid oxidoreductase activity, is much lower in the present study than was observed previously during prostate perfusion in vivo or prostate organ culture superfusion.  相似文献

Prostatic binding protein (PBP), a hormonally controlled oligomeric glycoprotein secreted by the rat ventral prostate, is composed of three different polypeptide chains, C1, C2, and C3. Microinjection of prostate mRNA into Xenopus laevis oocytes results in the synthesis, processing, and correct assembly of these three components, and also in the export of PBP into the medium. The glycosylation of component C3--the only glycopeptide of PBP--by the oocyte enzymes does not lead to the same result as in the native prostate tissue. The intracellular oocyte component contains an incompletely processed oligomannosyl core unit. Upon secretion this sugar core is further processed, probably at random because the carbohydrate chains attached to the exported C3 molecules are heterogeneous; they are also different from the oligosaccharide unit of authentic C3. However, tunicamycin experiments show that glycosylation is neither a prerequisite for secretion nor for the assembly of PBP, at least in oocytes.  相似文献

A specific mRNA for a structural lipoprotein of the Escherichiacoli outer membrane was translated in a wheat germ cell-free protein synthesizing system, S-adenosyl-methionine (SAM) and S-adenosyl-homocysteine (SAH) had neither stimulative nor inhibitory effect on the translation. When the products were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, two peaks appeared at the appearent molecular weights of about 15,000 and about 7,500. Inhibition of 5α-reduction of testosterone by an anti-androgen TSAA-291 (16β-ethyl-17β-hydroxy-4-estren-3-one) was studied in rat ventral prostates and the metabolic conversion of 3H-TSAA-291 was examined both in vitro and in vivo. In the in vitro experiment using nuclear 5α-reductase of the prostate, 5α-dihydrotestosterone formation from 3H-testosterone was inhibited in a competitive manner by the anti-androgen. In the in vitro experiment using 3H-TSAA-291, 5α-reduction of the anti-androgen occurred. One, 2 and 4 hr after an intravenous administration of 140 μCi/rat of 3H-TSAA-291 to castrated rats, the unchanged TSAA-291 accumulated in higher amounts in the ventral prostate than in the plasma, skeletal muscle and levator ani muscle, thereby indicating the selective uptake of the anti-androgen by the androgen target organ. No appreciable amounts of the 5α-reduced metabolite of TSAA-291 were detected in the prostate, thus suggesting that TSAA-291 itself may be responsible for the anti-androgenic properties. The inhibitory potency on the 5α-reductase activity of several other 16β-substituted androstane and estrane analogues was also examined.  相似文献

T4 bacteriophage mRNA for lysozyme was extracted from T4 phage infected E. coli cells, partially purified by column chromatography, and translated in a heterologous cell-free protein synthesizing system prepared from wheat germ. The translation product was confirmed by SDS polyacrylamide gel electrophoresis and enzymatic activity — bacteriolysis as tested with Micrococcus luteus. Cytoplasmic free and bound polysomes were isolated from bovine adrenal cortex, and used to program invitro protein synthesis in rat liver cell sap and wheat germ lysate systems. Synthesis of adrenodoxin(Ad) and adrenodoxin reductase(AdR) in the cell-free systems was determined by immunoprecipitation using monospecific antibodies, and the sizes of the invitro products were analyzed by SDS-polyacrylamide gel electrophoresis. Ad was synthesized by both free and bound polysomes as a putative large precursor having molecular weight of approximately 20,000 daltons, which was processed to mature size Ad (MW 12,000 daltons) by invitro incubation with adrenal cortex mitochondria. On the other hand, AdR was synthesized only by free polysomes apparently as the mature size product.  相似文献

1. A review is presented of recent advances in glutamate receptor research with particular emphasis on studies which show that some glutamate receptors in the central nervous systems (CNS) of Xenopus and rat contain a mixture of N-methyl -D-aspartate-sensitive and kainate-sensitive subunits.2. Protein isolated from Xenopus CNS using a domoic acid affinity column exhibits complex pharmacological properties. It binds both [3H]kainate and [3H]glycine: the binding of the latter is strychnine-insensitive.3. When reconstituted into lipid bilayers, channels gated by kainate and NMDA can be elicited and the properties of these channels are similar to those gated by kainate receptors and NMDA receptors, respectively, in studies of vertebrate central neurones in situ.4. The protein can be fractionated into two components; one of which is sensitive only to kainate and AMPA, the other exhibiting sensitivity to both kainate and NMDA.5. When RNA isolated from Xenopus and rat CNS is injected into Xenopus oocytes, responses to kainate and NMDA can be seen within 2–3 days. The responses to co-application of these agonists support the contention that some of the glutamate receptors expressed in oocytes contain both kainate-sensitive and NMDA-sensitive subunits.  相似文献

Variants of B 16-F 1 mouse melanoma cells selected for resistance to wheat germ agglutinin (WGA) toxicity invitro were found to have undergone a stable surface change correlated both to lectin resistance and reduced metastasizing potential. The surface alteration, as indicated by the increased electrophoretic mobilities of several lactoperoxidase-iodinated cell surface proteins in SDS-PAGE, was restricted to polypeptides able to interact with WGA. The availability of lectin-resistant melanoma cell variants having altered metastasizing behavior provides a promising approach to studies of the role of specific cell surface components in the metastasizing process.  相似文献

cDNA clones were obtained by transformation of E. coli x1776 with pBR322 containing insert of ds cDNA synthesized from total rat prostate poly(A) RNA. Two prostate-specific cDNA clones were isolated by colony hybridization and identified by message selection/translation as encoding polypeptides of Mr: 13,500 and 9,300. Hybridization of poly(A) RNA from normal and castrated rat prostates to the cloned cDNAs indicated that the levels of mRNAs coding for Mr: 13,500 and 9,300 polypeptides are regulated by testosterone.  相似文献

A minicell-producing strain of E.coli carrying an F′ factor, KLF10-1, forms minicells that contain plasmid but not chromosomal DNA. These minicells were found to synthesize two polypeptides corresponding precisely to the β and β′ subunits of RNA polymerase in SDS-polyacrylamide gel electrophoresis. In contrast, minicells obtained from an isogenic strain carrying F13-1 do not synthesize these proteins under similar conditions. These results indicate that the structural genes for the β′ as well as β subunits of the polymerase are located on the chromosomal segment (78 to 81 min on the standard genetic map of E.coli) carried by KLF10-1.  相似文献

α-Difluoromethylornithine (RMI 71.782), an enzyme-activated irreversible inhibitor of ornithine decarboxylase (E.C. 4.1.1.17) in vitro, causes a rapid, long-lasting, dose-dependent decrease of ornithine decarboxylase activity in prostate and, to a lesser extent, in thymus and testis of rats when injected intraperitoneally. Repeated doses (100 mg/kg or 1 g/kg) of α-difluoromethylornithine given to rats for two weeks markedly decreased polyamine concentrations in several rat tissues and selectively slowed down growth of ventral prostate and of thymus.  相似文献
