Two barley catalase genes respond differentially to light   总被引:1,自引:0,他引:1  
Cloned catalase probes from barley ( Hordeum vulgare L.) and maize ( Zea mays L.) were used to examine catalase gene expression in greened and etiolated leaves of several barley lines. Etiolated leaves had greater levels of an mRNA detected by barley Cat1 , compared with greened leaves, in all lines. In contrast, a Cat2 -like mRNA (homologous to Cat2 of maize) was induced by light and accumulated to high levels in greened leaves, compared to the negligible levels detected in etiolated leaves. This suggests that barley contains light-inducible and light-repressible catalase genes. In the catalase-deficient barley mutant RPr 79/4, no hybridization signal was detected when RNA from greened or etiolated leaves was probed with maize Cat2 , indicating that this mutant is deficient for the light-induced Cat mRNA. In etiolated seedlings of both RPr 79/4 and its motherline, the level of the Cat1 mRNA increased coordinately with a steady increase in catalase activity. Even though the mutant RPr 79/4 was able to grow to maturity in normal air, it sustained chlorosis and significant head sterility, probably due to the lack of a light-inducible catalase. Although the mutant RPr 79/4 is not completely lacking catalase (EC 1.11.1.6), the loss of the CAT-1 isozyme is evidently harmful. A low-temperature-responsive gene, blt 801, isolated from a winter barley (Hordeum vulgare L.) cDNA library prepared from leaf meristematic tissue, was sequenced. The deduced amino acid sequence predicts a glycine-rich RNA-binding protein (GR-RNP) which was homology to stress-responsive GR-RNPs from several other plant species. BLT 801 is a two-domain protein, the amino-terminal domain comprises a consensus RNA-binding domain similar to that found in many eukaryotic genes and the carboxy-terminal domain is extremely glycine-rich (68.5% glycine). Blt 801 mRNA also accumulates in response to the phytohormone abscisic acid. The protein encoded by blt 801 has been produced as a recombinant fusion protein using a bacterial expression vector. The fusion protein, a chimaera of glutathione S-transferase and BLT 801, has been used in studies to determine nucleic acid binding and other characteristics. Binding studies with single-stranded nucleic acids show that BLT 801 has affinity for homoribopolymers G, A and U but not C, it also binds to single-stranded DNA and selects RNA molecules containing open loop structures enriched in adenine but low in cytosine. BLT 801 has a consensus motif for phosphorylation by cAMP protein kinase (PKA) at the junction between the two domains which can be phosphorylated by PKA in vitro and which, by analogy to animal studies, may have significance for controlling enzyme function.  相似文献

为寻找寒冷适应机体表达上调的相关基因,以Balb/C小鼠建立寒冷适应模型,分别提取肌肉和肝脏RNA,运用代表性差异分析(RDA)方法,寻找寒冷适应表达上调的相关基因片段,进行序列分析和同源性比较。结果显示,在肌肉和肝脏中有部分表达上调的基因片段,经狭线杂交验证,其中3条基因表达存在显著差异。这些新发现的机体寒冷适应相关基因为进一步理解寒冷适应的分子机制提供了帮助。  相似文献

The effect of cold treatment on gene expression in two different barley (Hordeum vulgare L.) cultivars has been studied. Cold stress induced a set of new mRNAs as determined by in-vitro translation of coleoptile RNA obtained from control and stressed seedlings. These mRNAs accumulated with different kinetics, and the cold-induced proteins could be grouped into five categories. The first category (a) is represented by a single protein with Mr of 75 kDa that reaches its highest level of expression after 6 h at 5°C. This polypeptide readily accumulates in the plant tissues and it can be detected when proteins separated by two-dimensional electrophoresis are stained with silver nitrate. The other polypeptides appear later during the 1- to 4-d stress period (protein groups b and c), increase (group d), or decrease during the period of treatment (group e). Only minor differences between the two cultivars with different cold-resistance capacities were found when the in-vitro translation products were compared. Salix paraplesia was used as an experimental model to investigate the effect of short day photoperiod (SD) and low temperature (LT) on development of freezing tolerance and on endogenous abscisic acid (ABA) contents. We characterized differences in SD and LT-induced cold acclimation in three ecotypes from different altitudes. The results demonstrated that cold acclimation could be triggered by exposing the plants to SD or LT alone, and that a combination of the different treatments had an additive effect on freezing tolerance in all ecotypes studied. However, the high altitudinal ecotype was more responsive to SD and LT than the low altitudinal ecotype. Development of freezing tolerance induced by SD and LT was accompanied by changes in ABA contents which were ecotype-dependent. Although the stem had higher initial freezing tolerance, the leaves developed freezing tolerance more quickly than the stem and thus leaves may provide an interesting experimental system for physiological and molecular studies of cold acclimation in woody plants.  相似文献

准噶尔小胸鳖甲短时低温胁迫响应的转录组分析   总被引:1,自引:0,他引:1  
【目的】拟步甲科昆虫小胸鳖甲Microdera punctipennis是分布于中国新疆古尔班通古特沙漠的特有物种,具有很强的耐寒性,但其低温响应的分子机制尚不明确。本研究旨在利用转录组测序技术丰富小胸鳖甲的已知基因信息,分析在短时低温胁迫下,上调表达基因的主要类群及参与的主要代谢通路。【方法】利用Trinity软件对分别在4℃低温和25℃(对照)下处理3 h的小胸鳖甲成虫RNA-Seq数据进行从头组装,利用Blast2go软件进行基因注释。通过DESeq和GFOLD软件分析差异表达基因,并对上调表达基因进行GO(Gene Ontology)和KEGG(Kyoto Encyclopedia of Genes and Genomes)代谢途径富集分析。【结果】测序过滤后得到68 165 001个有效读序,51 712个平均长度为984 bp的非冗余基因,其中29 925个基因(约57.87%)有同源序列(E-value10-5),与拟步甲科的赤拟谷盗Tribolium castaneum相应基因相似性最高(核苷酸序列一致性为72.75%)。低温响应上调表达基因有514个,富集到35个GO类群,18个KEGG途径。其中胁迫响应类群、生物调节类群和免疫系统过程类群都占有重要比例,嘌呤代谢、硫胺素代谢、糖酵解/糖异生等代谢通路显著富集。在胁迫响应类群中,热激蛋白Hsp90基因、超氧化物歧化酶(SOD)基因和钙联接蛋白Calnexin基因等8个非生物胁迫相关基因显著上调表达。【结论】荒漠昆虫小胸鳖甲低温转录组揭示,在4℃冷驯化过程中,代谢过程、胁迫响应、生物调节和免疫系统等生物学过程相关基因表达显著上调,其中几个非生物胁迫响应基因提示小胸鳖甲可能从分子伴侣、细胞周期阻滞、线粒体稳定性、抗氧化胁迫等多方面应对低温胁迫。KEGG富集分析所揭示的小胸鳖甲在低温下的转录组代谢通路与其他物种有较大差异。研究结果为后续生物信息学分析及小胸鳖甲耐寒关键基因的发掘及耐寒机制的揭示提供了基础数据。  相似文献

The purpose of this work was to examine environmental control of expression, at the mRNA level, of cold-inducible genes and to test the relationship of the expression of the genes to cold acclimation. Barley plants (Hordeum vulgare L. cv. Igri) at the three- to four-leaf stage were (a) grown in different temperature environments between 20/15°C and +4/-4°C or (b) transferred between 20/15°C and 6/2°C or (c) grown under drought or nutrient stress conditions. Frost hardiness (using a regrowth method) and mRNA levels for three cold-induced genes, blt4-9, blt14 and blt101, from meristematic crown tissue (vegetative shoot meristem plus subtending stem and associated root initials) were measured. Hardiness and levels of blt4-9, blt14 and blt101 mRNAs increased with lower growth temperatures, below a maximum inductive temperature. Prior temperature environment and plant age affected the rate of change in mRNA levels of these genes in response to a change of temperature environment. Hardiness was strongly correlated with mRNA levels of these genes in plants grown in different temperature environments. This correlation did not extend to plants exposed to drought or nutrient stresses. Implications are drawn for plant responses to a warmer climate.  相似文献

Heterosis is broadly defined as the increased vigour of hybrids in comparison to their parents. In the model plant Arabidopsis thaliana, a significant heterosis effect on leaf-freezing tolerance was observed in the F(1) generation of a cross between the accessions Columbia-0 (Col) and C24. Parental Col plants were significantly more freezing-tolerant than C24 plants in both the acclimated and non-acclimated (NA) states. Mid-parent heterosis was observed in the F(1) plants, both in the basic tolerance of non-adapted plants and in freezing tolerance after cold acclimation. Best-parent heterosis, on the other hand, was only found after cold acclimation. The heterosis effect was reduced in the F(2) populations such that only mid-parent heterosis was evident. The leaf content of soluble sugars (fructose (Fru), glucose (Glc), sucrose (Suc) and raffinose (Raf)) increased dramatically in the F(1) plants after cold acclimation as compared to the parental lines. The content of proline (Pro), however, was only moderately increased in the F(1) plants under the same conditions. Correlation analyses showed that only Raf content was consistently related to leaf-freezing tolerance in both the acclimated and NA states. A quantification of mRNA levels in leaves of parental and F(1) lines using quantitative real-time RT-PCR showed no clear indication for an involvement of the investigated genes (CBF (C-repeat binding factor)1, CBF2, (cold-regulated protein (COR) 6.6, COR15a, COR15b, COR47 and COR78) in the heterosis effect.  相似文献

The role of ABA in freezing tolerance and cold acclimation in barley   总被引:4,自引:0,他引:4  
The role of ABA in freezing resistance in nonacclimated and cold‐acclimated barley ( Hordeum vulgare L.) was studied. Eleven nonacclimated cultivars differed in their LT50, ranging from −10.8 to −4.8°C. Sugars, free proline, soluble proteins and ABA were analyzed in nonacclimated cultivars and during cold acclimation of one cultivar. There was an inverse correlation between LT50 and both ABA and sucrose contents. Exogenous ABA caused a decrease in the freezing point of leaf tissue in the cultivar with the lowest level of endogenous ABA, but not in the cultivar with the highest level, suggesting that ABA in the latter may be near the optimum endogenous level to induce freezing tolerance. Plants of cv. Aramir treated with ABA or allowed to acclimate to cold temperature increased their soluble sugar content to a similar level. The LT50 of leaves of cold‐acclimated cv. Aramir decreased from −5.8 to −11.4°C, with biphasic kinetics, accumulating proline and soluble sugars with similar kinetics. The biphasic profile observed during cold acclimation could be a direct consequence of cryoprotectant accumulation kinetics. ABA and soluble protein accumulation showed a single step profile, associated mainly with the second phase of the LT50 decrease. Thus, a significant increase in endogenous ABA is part of the response of barley to low temperature and may be required as a signal for the second phase of cold acclimation. Endogenous ABA contents in the nonacclimated state may determine constitutive freezing tolerance.  相似文献
