In this study, multiple paternity (MP) was investigated in three commercially important shark species, common smoothhound Mustelus mustelus, dusky shark Carcharhinus obscurus and scalloped hammerhead Sphyrna lewini occurring in southern Africa. Reduced marker panels of between five and six microsatellite loci were constructed for each species and used to genotype and assess the presence of MP in a total of 60 M. mustelus individuals from six litters, 90 C. obscurus individuals from 14 litters and 54 S. lewini individuals from 13 litters. Analysis in GERUD and COLONY revealed the presence of MP in all three species. Multiple paternities were observed in 67, 35 and 46% of the litters of M. mustelus, C. obscurus and S. lewini, with corresponding average sire size of 1·6, 1·4 and 2·0, respectively. The variation in the rate of MP among the three species is in accordance with previous studies whilst the comparatively high frequency of MP observed for M. mustelus, matches what has previously been reported for shark species demonstrating aggregation behaviour.  相似文献

In this study, life-history traits (maximum and average size, size at maturity and fecundity) of two congeneric smooth-hounds, Mustelus mustelus and Mustelus punctulatus, which share a geographical distribution and experience a similar fishing exploitation, were estimated and compared between species. The results indicated a lower maximum and average size, a lower size at maturity and a higher fecundity in M. punctulatus compared with those in M. mustelus. Considering that these two species co-occur in the same areas and are caught by the same fishing gears, the results indicate a higher vulnerability to exploitation of M. mustelus compared with that of M. punctulatus.  相似文献

The common smoothhound, Mustelus mustelus, is an epibenthic species targeted by fisheries around the world driven by the increasing demand for shark products. Given the wide-spread occurrence of this species and corresponding lack of molecular data in many areas of said distribution, baseline molecular assessments of this commercially important shark may contribute to finer-scale analyses in areas in which this species is targeted. Therefore, population genetic analyses were conducted along the East Atlantic, from the Mediterranean Sea to the south-east coast of Africa, using microsatellite markers and the mitochondrial control region (mtCR). Overall, M. mustelus displayed low to moderate genetic diversity, with the Mediterranean populations appearing to exhibit the lowest mitochondrial diversity, and the west African populations displaying the lowest nuclear diversity. Microsatellite analysis indicated strong genetic differentiation between the three regions, with finer-scale population structure in each region, without correlation between genetic and geographical distance. For the mtCR sequences, a total of 18 haplotypes were identified, with a high degree of divergence discernable between the regions, largely in accordance with the microsatellite data. The study documents a remarkable level of population isolation across a vast area, suggesting little or no present-day connectivity among extant populations. The findings may serve as an essential baseline for global population management and commercial traceability of this threatened shark.

Small‐scale fisheries are generally promoted as a sustainable alternative to large‐scale industrial fisheries. However, there is recent growing evidence that small‐scale fisheries may be the largest threat to marine species of conservation concern. The objective of this study was to evaluate the potential impact of the trammel net fishery on elasmobranchs in the Gulf of Gabès, Southern Tunisia. Data are based on 191 shrimp trammel net set (40 mm stretched mesh size) surveys conducted aboard commercial fishing vessels from May to July 2009. Five species of the small coastal elasmobranchs (Mustelus mustelus (Linnaeus, 1758), Mustelus punctulatus Risso 1827, Dasyatis pastinaca (Linnaeus, 1758), Dasyatis marmorata (Steindachner, 1892) and Torpedo torpedo (Linnaeus, 1758)) and two species from the large coastal shark (Carcharhinus plumbeus (Nardo, 1827) and Carcharhinus brevipinna (Müller & Henle, 1839)) were recognized as by‐catch in this fishery. Elasmobranch by‐catch was dominated by sharks (90.3%), smoothhound sharks Mustelus sp. being by far the most important (88.9%) and reflecting their abundance in the area; 58% of the sets caught at least one specimen, with 4.8 ± 1.3 caught per set. Captures were composed essentially of neonate and juvenile sharks, while the batoids were dominated by mature individuals. This study shows that shrimp trammel nets represent a considerable source of mortality for early life stages of elasmobranch species in the Gulf of Gabès. Additionally, there was a high density of neonates and small juvenile M. mustelus in the Sfax zone, suggesting that these nearshore waters are a nursery grounds for smoothhound sharks. Further research should focus on the incidents of by‐catch and evaluate the potential solutions to allow trammel net fisheries to coexist alongside the elasmobranch species.  相似文献

Between 2018 and 2020, 696 fish belonging to two species of sharks from the Family Triakidae (Mustelus mustelus and M. punctulatus) were collected from the coasts of Tunisia and inspected for parasites. Six copepod taxa (Perissopus dentatus Steenstrup & Lütken, 1861, Eudactylinella alba Wilson, 1932, Kroyeria lineata Van Beneden, 1853, Nesippus orientalis Heller, 1865 and Lernaeopoda galei Krøyer, 1837, Kroyeria sp.), four isopod species (Anilocra physodes (Linnaeus, 1758), Emetha audouini (H. Milne Edwards, 1840), Ceratothoa parallela (Otto, 1828) and Ceratothoa oestroides (Risso, 1816)) and two monogenean species (Erpocotyle sp1. And Erpocotyle sp2) were collected. A large number of global host records was reported, including the occurrence of E. audouini on M. mustelus and of Erpocotyle sp.2 on M. punctulatus. The study of the diversity of parasites per host species revealed that M. mustelus had a higher parasitic richness compared to M. punctulatus. In this study, it was provided for the first records on ectoparasites on Triakidae sharks from Tunisian coasts and their infection indices.  相似文献

Elasmobranchs are an important catch component in Tunisian artisanal fisheries. Generally, species‐specific information is largely unavailable for artisanal fisheries; an increase in knowledge thereof is essential to ensure proper management of these species. The study analyzed the gillnet fishery elasmobranch catches in the Gulf of Gabès, whereby 45 fishing trips were conducted from April to June 2007 and 2008 for the capture of a total of six species: three Rajiformes and three Carchariniformes. Rhinobatos cemiculus was the most important species (52%; 4.588 ind km?2 net per day). Among shark species, Mustelus mustelus was the most important (66.8%; 2.21 ind km?2 net per day). Discards represented 6.87% of total catch in weight and 14.28% in number of specimens caught. Size composition of captures varied by species, but usually mature, mainly gravid females were abundant. Further investigations are needed to obtain more information on such fragile species for the development of protective measures.  相似文献

Partial large subunit 28S rDNA sequences were obtained for specimens of Calicotyle (Monogenea: Monocotylidae) from eight different host species distributed worldwide to test the validity of some species and to address the question of host-specificity in others. Sequences obtained for Calicotyle specimens identified as C. kroyeri based on morphological methods from the type-host Raja radiata (Rajidae) and an additional host R. clavata, both from the North Sea, were identical. However, `C. kroyeri' from the cloaca of R. naevus from Tunisia, Raja sp. A from Tasmania and R. radula from Tunisia differed from C. kroyeri from R. radiata by five (0.51%), 21 (2.13%) and 39 (3.96%) base pairs, respectively, over 984 sites. Therefore, it is likely that the specimens from Raja sp. A, R. radula and perhaps even from R. naevus are not C. kroyeri. Molecular results determined that the calicotylines from the cloaca of Urolophus cruciatus and U. paucimaculatus (Urolophidae) from southern Tasmania identified previously as C. urolophi are indeed identical. Large subunit 28S rDNA sequences of C. palombi and C. stossichi collected from the cloaca and rectal gland, respectively of Mustelus mustelus (Triakidae) from the coast of Tunisia differ sufficiently for these calicotylines to be considered separate and valid species. Our results indicate that some species of Calicotyle are not strictly host-specific, but that C. kroyeri may not be as widely distributed in rajids as was believed previously. Calicotyle specimens from rajids must be re-examined critically to determine whether there are morphological differences indicative of specific differences that may have been overlooked previously.  相似文献

Sharks play a key role in the structure and functioning of marine ecosystems. More ecological information is essential to implement responsible management and conservation actions on this fauna, particularly at a regional level for threatened species. Mustelus mustelus is widely found in the eastern Atlantic Ocean and catalogued as “Vulnerable” by the IUCN European assessment. In this study, data on the distribution and population structure of this species across the islands of the Canarian archipelago, located along an east to west gradient in the north‐eastern Atlantic, were collected by taking advantage of “Local Ecological Knowledge,” in terms of sightings in coastal waters and long‐term imprints on the local gastronomic heritage, and decadal fisheries landings. Both sources of quantitative data (sightings and fisheries landings) demonstrated that adults of M. mustelus has a significantly larger presence in the eastern and central, than in the western islands of the archipelago. This is also reflected on local gastronomic legacies, with a larger number of recipes in the eastern and central islands. Adult smooth‐hound sharks were significantly more observed in sandy and sandy‐rocky bottoms, with individuals seen throughout the entire year, whereas juveniles aggregate on very shallow waters in spring and summer. Such aggregations require a special management strategy, as they play a key role in critical life stages; these sites should be protected from human perturbations. We also suggest a temporal fishing ban between April and October, when individuals tend to concentrate on nearshore waters. Because of the large differences in presence of this shark among the Canary Islands, management of the species should be adapted to the specific peculiarities of each island, rather than adopting a management policy at the entire archipelago‐scale. Overall, this study sets the basis for further investigation to promote conservation of this vulnerable shark in the study region.  相似文献

Sex differences in skews of vertebrate lifetime reproductive success are difficult to measure directly. Evolutionary histories of differential skew should be detectable in the genome. For example, male‐biased skew should reduce variation in the biparentally inherited genome relative to the maternally inherited genome. We tested this approach in lek‐breeding ruff (Class Aves, Philomachus pugnax) by comparing genetic variation of nuclear microsatellites (θn; biparental) versus mitochondrial D‐loop sequences (θm; maternal), and conversion to comparable nuclear (Ne) and female (Nef) effective population size using published ranges of mutation rates for each marker (μ). We provide a Bayesian method to calculate Ne (θn = 4Neμn) and Nef (θm = 2Nefμm) using 95% credible intervals (CI) of θn and θm as informative priors, and accounting for uncertainty in μ. In 96 male ruffs from one population, Ne was 97% (79–100%) lower than expected under random mating in an ideal population, where Ne:Nef = 2. This substantially lower autosomal variation represents the first genomic support of strong male reproductive skew in a lekking species.  相似文献

Three-hundred twenty five droppings from parrots raised in the premises of 4 breeders and in several private households were cultured for yeasts. One-hundred sixty droppings (49.2%) resulted positive. From these specimens 212 isolates belonging to 27 different species were obtained. Mainly Candida species such as C. albicans, C. catenulata, C. curvata, C. famata, C. glabrata, C. guilliermondi, C. holmii, C. intermedia, C. krusei, C. lambica, C. lusitaniae, C. membranaefaciens, C. parapsilosis, C. pelliculosa, C. sake and C. valida were isolated. Debaryomyces marama, D. polymorphus, Geotrichum sp., Pichia etchelsii, P. ohmeri, Rhodotorula glutinis, R. rubra, Rhodotorula sp., Saccharomyces cerevisiae, S. kluyiveri and Zygosaccharomyces sp. were also obtained. Dark colonies on Staib medium were never observed. The psittacine birds apparently serve as carriers for several Candida species or their perfect states and to a lesser extent for other opportunistic yeasts such as Rhodotorula, Trichosporon and Saccharomyces spp., which are considered part of the transient microbiota of the gastrointestinal tract. The most striking finding was the absence of Cryptococcus spp. among the isolates. The present survey confirms the role of pet birds in carrying potential zoonotic yeasts. Summary Five new elements of the mitotic control in the fission yeast Schizosaccharomyces pombe were isolated from gene libraries as multicopy suppressors of the conditional lethal phenotype of win1-1 weel ts cdc25ts triple mutant strains. These genes were designated wisl + –wis5+for win suppressing, and do not correspond to winl + or any of the previously characterised mitotic control genes. None of the wis genes is capable of suppressing the cdc phenotype of cdc25 ts strains, suggesting that their effect is not simply to reverse the effect of loss of cdc25 function. wisl + has been previously reported to encode a putative serine/threonine protein kinase that acts as a dosage-dependent inducer of mitosis. wis4 + appears to be a specific suppressor of the winl-1 mutation. wis2 + and wis3 + are capable of suppressing a wide range of cdc phenotypes arising from the combination of various mutations with wee1 ts and cdc25 ts, suggesting that the wis2 + and wis3 + products may interact with elements central to the mitotic control.  相似文献

Desmids from 12 freshwater resources in the northern part of Thailand were investigated during 2002 to 2003. A total of 91 taxa were found. They belonged to 17 genera: Actinotaenium, Spirotaenia, Netrium, Gonatozygon, Pleurotaenium, Closterium, Euastrum, Micrasterias, Cosmarium, Cosmocladium, Stuarastrum, Staurodesmus, Xanthidium, Teilingia, Spondylosium, Hyalotheca and Desmidium. The water qualities in all the water resources were classified as oligotrophic to meso-eutrophic by trophic status. The taxa that could possibly be used as bioindicators of trophic state were Staurastrum gutwinskii, Spondylosium pandurifoemae, Cosmarium capitulum, C. mediosrobiculatum var. egranutum, S. tortum, Closterium gracile var. elongatum, C. kuetzingii and Closterium dianae var. dianae. The most frequently found taxa were Staurastrum limneticum var. burmense, S. tetracerum var. tetraerum, Pleurotaenium trabecula, Closterium ehrenbergii var. ehrenbergii and C. kuetzingii. The rare taxa in this study were Actinotaenium sp. Spirotaenia condensata, Pleurotaenium burmense var. dacchense and Micrasterias apiculata. Leishmania donovani and Leishmaniainfantum infections cause fatal visceral leishmaniasis, and Leishmaniamajor causes self healing cutaneous lesions. It is poorly understood what genetic differences between these Leishmania species are responsible for the different pathologies of infection. To investigate whether L.donovani species-specific genes are involved in visceral Leishmania infection, we have examined a L.donovani species-specific gene Ld1590 (ortholog of LinJ15_V3.0900) that is a pseudogene in L.major. We have previously shown that transgenic expression of L.donovani Ld1590 in L.major significantly increased the liver and spleen parasite burdens in infected BALB/c mice. In this study we report that Ld1590 potentially encodes a nucleotide sugar transporter (NST) which localizes in the L.donovani Golgi apparatus. Surprisingly, although transgenic expression of the Ld1590 NST increased L.major survival in visceral organs, deletion of Ld1590 NST in L.donovani had no significant effect on L.donovani survival in mice. These observations suggest that loss of the functional Ld1590 gene in L.major may have been associated with reduced virulence in visceral organs in its animal reservoir and could have contributed to L.major’s tropism for cutaneous infections.  相似文献

Widespread and repeated use of azoles, particularly fluconazole, has led to the rapid development of azole resistance in Candida albicans. Overexpression of CDR1, CDR2, and CaMDR1 has been reported contributing to azole resistance in C. albicans. In this study, hyper-resistant C. albicans mutant, with the above three genes deleted, was obtained by exposure to fluconazole and fluphenezine for 28 passages. Thirty-five differentially expressed genes were identified in the hyper-resistant mutant by microarray analysis; among the 13 up-regulated genes, we successfully constructed the rta2 and ipf14030 null mutants in C. albicans strain with deletions of CDR1, CDR2 and CaMDR1. Using spot dilution assay, we demonstrated that the disruption of RTA2 increased the susceptibility of C. albicans to azoles while the disruption of IPF14030 did not influence the sensitivity of C. albicans to azoles. Meanwhile, we found that ectopic overexpression of RTA2 in C. albicans strain with deletions of CDR1, CDR2 and CaMDR1 conferred resistance to azoles. RTA2 expression was found elevated in clinical azole-resistant isolates of C. albicans. In conclusion, our findings suggest that RTA2 is involved in the development of azole resistance in C. albicans.  相似文献

Abstract

The Asian genus Struckia Müll.Hal. is reduced to a synonym of Plagiothecium Bruch &; Schimp. according to a phylogenetic analysis involving S. argentata (Mitt.) Müll.Hal., S. enervis (Broth.) Ignatov, T.J.Kop. &; D.G.Long and 13 representative boreal species of Plagiothecium. Two nuclear regions, ITS and partial gapC, and five chloroplast regions, rbcL, rps4-trnS, psaB, trnG and trnL-F, were utilized to reconstruct the phylogenetic relationship between Struckia and Plagiothecium. Bayesian, maximum parsimony, and maximum likelihood analyses resulted in a strongly supported clade including Struckia and Plagiothecium. Plagiothecium handelii Broth. and P. paleaceum (Mitt.) A.Jaeger, which share similar geographical ranges with Struckia and intermediate morphological traits between Struckia and Plagiothecium, grouped with the Chinese Struckia, and all of them appeared as sister to the ‘core’ of Plagiothecium. The accurate position of P. piliferum (Sw.) Schimp. remains unclear due to moderate support from all analyses, and we suggest that it is retained in Plagiothecium until further evidence is available.  相似文献

We screened for mutations that resulted in lethality when the G1 cyclin Cln2p was overexpressed throughout the cell cycle in Saccharomyces cerevisiae. Mutations in five complementation groups were found to give this phenotype, and three of the mutated genes were identified as MEC1, NUP170, and CDC14. Mutations in CDC14 may have been recovered in the screen because Cdc14p may reduce the cyclin B (Clb)-associated Cdc28 kinase activity in late mitosis, and Cln2p may normally activate Clb-Cdc28 kinase activity by related mechanisms. In agreement with the idea that cdc14 mutations elevate Clb-Cdc28 kinase activity, deletion of the gene for the Clb-Cdc28 inhibitor Sic1 caused synthetic lethality with cdc14-1, as did the deletion of HCT1, which is required for proteolysis of Clb2p. Surprisingly, deletion of the gene for the major B-type cyclin, CLB2, also caused synthetic lethality with the cdc14-1 mutation. The clb2 cdc14 strains arrested with replicated but unseparated DNA and unseparated spindle pole bodies; this phenotype is distinct from the late mitotic arrest of the sic1::TRP1 cdc14-1 and the cdc14-1 hct1::LEU2 double mutants and of the cdc14 CLN2 overexpressor. We found genetic interactions between CDC14 and the replication initiator gene CDC6, extending previous observations of interactions between the late mitotic function of Cdc14p and control of DNA replication. The Mls locus was originally defined to have four alleles; three controlled products that were detectable in primary mixed leukocyte reactions (MLR), whereas one, b, was described as being null. Recently, other investigators postulated that the Mls locus is nonpolymorphic, being composed of the b null allele and of a singly expressed allele previously thought to be the a and d alleles. We previously reported that products controlled by Mls aand Mls dwere antigenically distinct and therefore are not controlled by the same allele, and the product of Mls bon cells of three different strains was easily detectable by Mls aand Mls dresponding cells. Thus the b allele is not null. In the present report evidence is presented which indicates that both Mls band Mls cencoded products were undetectable by MLR when in the presence of Mls aor Mls d. This was demonstrated by (a) the inability of Mls a/Mls cand Mls a/Mls bF1 cells to stimulate Mls aresponding cells and Mls d/Mls cand Mls d/Mls bcells to stimulate Mls dcells; (b) the positive response of Mls a/Mls band Mls d/Mls bF1-hybrid cells to Mls b-encoded products; and (c) the reactivity of Mls a/Mls cand Mls d/Mls cF1 hybrid cells to Mls c-encoded determinants.  相似文献
